Abstract The Taihangshan-Yanshan region (TYR) is an important ecological barrier area for Beijing-Tianjin-Hebei, and the effectiveness of its ecological restoration and protection is of great significance to the ecological security pattern of North China. Based on the FVC data from 2000 to 2021, residual analysis, parametric optimal geodetector technique (OPGD) and multi-scale geographically weighted regression analysis (MGWR) were used to clarify the the multivariate driving mechanism of the evolution of FVC in the TYR. Results show that: (1) FVC changes in the TYR show a slowly fluctuating upward trend, with an average growth rate of 0.02/10a, and a spatial pattern of "high in the northwest and low in the southeast"; more than half of the FVC increased during the 22-year period. (2) The results of residual analysis showed that the effects of temperature and precipitation on FVC were very limited, and a considerable proportion (80.80% and 76.78%) of the improved and degraded areas were influenced by other factors. (3) The results of OPGD showed that the main influencing factors of the spatial differentiation of FVC included evapotranspiration, surface temperature, land use type, nighttime light intensity, soil type, and vegetation type (q > 0.2); The explanatory rates of the two-factor interactions were greater than those of the single factor, which showed either nonlinear enhancement or bifactorial enhancement, among which, the interaction of evapotranspiration with mean air and surface temperature has the strongest effect on the spatial and temporal evolution of FVC (q = 0.75). Surface temperature between 4.98 and 10.4 °C, evapotranspiration between 638 and 762 mm/a, and nighttime light between 1.96 and 7.78 lm/m2 favoured an increase in vegetation cover, and vegetation developed on lysimetric soils was more inclined to be of high cover. (4) The correlation between each variable and FVC showed different performance, GDP, elevation, slope and FVC showed significant positive correlation in most regions, while population size, urban population proportion, GDP proportion of primary and secondary industries, and nighttime light intensity all showed negative correlation with FVC to different degrees. The results can provide data for formulating regional environmental protection and restoration policies.

Abstract Depression and obesity are prevalent disorders with significant public health implications. In this study, we used a high-fat diet (HFD)-induced obese mouse model to investigate the mechanism underlying HFD-induced depression-like behaviors. HFD-induced obese mice exhibited depression-like behaviors and a reduction in hippocampus volume, which were reversed by treatment with an indoleamine 2,3-dioxygenase (IDO) inhibitor 1-methyltryptophan (1-MT). Interestingly, no changes in IDO levels were observed post-1-MT treatment, suggesting that other mechanisms may be involved in the anti-depressive effect of 1-MT. We further conducted RNA sequencing analysis to clarify the potential underlying mechanism of the anti-depressive effect of 1-MT in HFD-induced depressive mice and found a significant enrichment of shared differential genes in the extracellular matrix (ECM) organization pathway between the 1-MT-treated and untreated HFD-induced depressive mice. Therefore, we hypothesized that changes in ECM play a crucial role in the anti-depressive effect of 1-MT. To this end, we investigated perineuronal nets (PNNs), which are ECM assemblies that preferentially ensheath parvalbumin (PV)-positive interneurons and are involved in many abnormalities. We found that HFD is associated with excessive accumulation of PV-positive neurons and upregulation of PNNs, affecting synaptic transmission in PV-positive neurons and leading to glutamate-gamma-aminobutyric acid imbalances in the hippocampus. The 1-MT effectively reversed these changes, highlighting a PNN-related mechanism by which 1-MT exerts its anti-depressive effect.

Abstract Type I CRISPR-Cas systems employ multi-subunit effector Cascade and helicase-nuclease Cas3 to target and degrade foreign nucleic acids, representing the most abundant RNA-guided adaptive immune systems in prokaryotes. Their ability to cause long fragment deletions have led to increasing interests in eukaryotic genome editing. While the Cascade structures of all other six type I systems have been determined, the structure of the most evolutionarily conserved type I-B Cascade is still missing. Here, we present two cryo-EM structures of the Synechocystis sp. PCC 6714 (Syn) type I-B Cascade, revealing the molecular mechanisms that underlie RNA-directed Cascade assembly, target DNA recognition, and local conformational changes of the effector complex upon R-loop formation. Remarkably, a loop of Cas5 directly intercalated into the major groove of the PAM and facilitated PAM recognition. We further characterized the genome editing profiles of this I-B Cascade-Cas3 in human CD3+ T cells using mRNA-mediated delivery, which led to unidirectional 4.5 kb deletion in TRAC locus and achieved an editing efficiency up to 41.2%. Our study provides the structural basis for understanding target DNA recognition by type I-B Cascade and lays foundation for harnessing this system for long range genome editing in human T cells.

Mesenchymal stem cells (MSCs) experience substantial viability issues in the stroke infarct region, limiting their therapeutic efficacy and clinical translation. High levels of deadly reactive oxygen radicals (ROS) and proinflammatory cytokines (PC) in the infarct milieu kill transplanted MSCs, whereas low levels of beneficial ROS and PC stimulate and improve engrafted MSCs' viability. Based on the intrinsic hormesis effects in cellular biology, we built a microglia-inspired MSC bioengineering system to transform detrimental high-level ROS and PC into vitality enhancers for strengthening MSC therapy. This system is achieved by bioorthogonally arming metabolic glycoengineered MSCs with microglial membrane-coated nanoparticles and an antioxidative extracellular protective layer. In this system, extracellular ROS-scavenging and PC-absorbing layers effectively buffer the deleterious effects and establish a micro-livable niche at the level of a single MSC for transplantation. Meanwhile, the infarct's inanimate milieu is transformed at the tissue level into a new living niche to facilitate healing. The engineered MSCs achieved viability five times higher than natural MSCs at seven days after transplantation and exhibited a superior therapeutic effect for stroke recovery up to 28 days. This vitality-augmented system demonstrates the potential to accelerate the clinical translation of MSC treatment and boost stroke recovery.

The anti-nerve growth factor antibody class of drugs interrupts signaling by blocking NGF binding to TrkA receptors for the treatment of pain; however, this target class of drugs has been associated with serious adverse effects in the joints during clinical trials. DS002 is a novel anti-nerve growth factor antibody drug independently developed by Guangdong Dashi Pharmaceuticals. The main purpose of this study is to explore the correlation between DS002 and pain as well as cartilage and bone metabolism with the help of metabolomics technology and the principle of enzyme-linked reaction, and to examine whether DS002 will produce serious adverse effects in joints caused by its same target class of drugs, in order to provide more scientific basis for the safety and efficacy of DS002. Our results showed that DS002 mainly affected the metabolism of aromatic amino acids and other metabolites, of which six metabolites, l -phenylalanine, 5-hydroxytryptophan, 5-hydroxytryptamine hydrochloride, 3-indolepropionic acid, kynuric acid, and kynurenine, were significantly altered, which may be related to the effectiveness of DS002 in treating pain. In addition, there were no significant changes in biological indicators related to cartilage and bone metabolism in vivo, suggesting that DS002 would not have a significant effect on cartilage and bone metabolism, so we hypothesize that DS002 may not produce the serious adverse effects in joints caused by its fellow target analogs. Therefore, the Anti-NGF analgesic drug DS002 has the potential to become a promising drug in the field of analgesia, providing pain patients with an efficient treatment option without adverse effects.

Objectives: To investigate cases of five Campylobacter jejuni outbreaks and describe laboratory characteristics of these infections. Methods: Whole-genome sequencing and conventional methods were combined to thoroughly investigate the outbreaks, and data of contemporaneous sporadic cases was included for comparison. Results: Seven sequence types (STs) of C. jejuni caused 83 cases, including ST9079 which recurred across 2 years. Trace-back investigation could not identify any food items of infection but detected identical campylobacters from food contacts. Phylogenetic analysis unveiled genetic closeness between outbreak strains and some concurrent sporadic strains, indicating local campylobacteriosis may not be wholly sporadic but rather a series of linked cases. Virulence genes disclosed species/case-specific signatures to differentiate outbreaks from truly non-outbreak strains. Resistance to fluoroquinolones and/or macrolides was prevalent (90.8%, 108/119), with a noteworthy portion exhibiting multidrug resistance (31.1%, 37/119). Five types of plasmids were harbored among outbreak isolates, of which one plasmid harboring anti-stress and resistant genes was rarely found in C. jejuni. Conclusions: This is the first reported sequential outbreak of C. jejuni in China. Our observations help to define the genomic landscape and antimicrobial resistance patterns of Campylobacter, emphasizing the need for a broader ‘One Health’ perspective to combat the threats posed by campylobacteriosis.

Alkali-activated fly ash (AAFA) can offer both significant performance benefits and environmental advantages in corrosive and high-temperature environments. However, the slow setting and limited early strength development of AAFA at ambient temperature strict its use to pre-cast elements that must be cured using elevated temperatures. In order to address this limitation, this experiment pursued a distinctive approach to produce a pre-polymerized suspension (PPS) using ultrafine fly ash (UFA) that was activated with NaOH, thereby generating numerous nano-scale zeolite crystals and amorphous N-A-S-H gels. PPS was used alongside UFA in conjunction to create AAFA. Combining PPS in the AAFA mixture, while cutting UFA usage by 50%, led to a 30-minute initial setting time and a 15 MPa compressive strength after 3 days, which was significantly improved compared to the nearly 3 days required by the control sample to initially set. This remarkable result was attributed to the accelerated dissolution of active Si/Al ions from UFA, as well as the nucleation template effect of nano-scale zeolite and N-A-S-H gels within the PPS that speeded up the product's polyreaction, and resulted in a dense and quickly developed hardened structure. By using PPS, the dependency on UFA and its accompanying high energy consumption was dramatically reduced. Additionally, the fast-setting and high early strength properties of the optimized AAFA paste make it ideal for in-situ construction, thereby extending the usage of fly ash resources.

Abstract Background Ischemic stroke is a serious disease leading to significant disability in humans worldwide. Increasing evidence suggests that some microRNAs (miRNAs) participate in the pathophysiology of ischemic stroke. A key role for MiR-212 has been found in neuronal function and synaptic plasticity. Ischemic stroke can be effectively treated with electroacupuncture (EA); however, there is a lack of understanding of the relevant mechanisms. In this study, we employed behavioral test and resting-state functional magnetic resonance imaging (rs-fMRI) to detect behavioral and brain function alterations in rats suffering from ischemic stroke. The efficacy of EA therapy and miR-212-5p’s role in this process were also evaluated. Methods and results Forty rats were randomly divided into the following groups: Sham, middle cerebral artery occlusion/reperfusion (MCAO/R), MCAO/R + EA, MCAO/R + EA + antagomir-negative control and MCAO/R + EA + antagomir-212-5p groups. Behavioral changes were assessed by Catwalk gait analysis prior to and after modeling. Rs-fMRI was performed at one week after EA treatment, amplitude of low-frequency fluctuations (ALFF) and regional homogeneity (ReHo) were calculated to reveal neural activity. Furthermore, neuronal apoptosis in the ischemic penumbra was analyzed using a TUNEL assay. Treatment with EA significantly improved the performance of rats in the behavioral test. The motor and cognition-related brain regions showed decreased ALFF and ReHo following focal cerebral ischemia-reperfusion, and EA treatment could reactivate these brain regions. Moreover, EA treatment significantly decreased MCAO/R-induced cell death. However, the transfection of antagomir-212-5p attenuated the therapeutic effect of EA. Conclusions In conclusion, the results suggested that EA improved the behavioral and imaging outcomes of ischemic stroke through miR-212-5p.

ObjectiveIndividuals with hypopituitarism (HPs) have an increased risk of developing non-alcoholic fatty liver disease (NAFLD)/non-alcoholic steatohepatitis (NASH) due to growth hormone deficiency (GHD). We aimed to investigate the possible mechanisms underlying the relationship between GHD and NAFLD using proteomic and metabolomic insights.MethodsSerum metabolic alternations were assessed in male HPs using untargeted metabolomics. A rat model of HP was established through hypophysectomy, followed by recombinant human growth hormone (rhGH) intervention. The mechanisms underlying GHD-mediated NAFLD were elucidated through the application of label-free proteomics and phosphorylation proteomics.ResultsMetabolomic analysis revealed that biomarkers of mitochondrial dysfunction and oxidative stress, such as alanine, lactate, and creatine, were significantly elevated in HPs compared to age-matched controls. In rats, hypophysectomy led to marked hepatic steatosis, lipid peroxidation, and reduced glutathione (GSH), which were subsequently modulated by rhGH replacement. Proteomic analysis identified cytochrome P450s, mitochondrial translation elongation, and PPARA activating genes as the major distinguishing pathways in hypophysectomized rats. The processes of fatty acid transport, synthesis, oxidation, and NADP metabolism were tightly described. An enhanced regulation of peroxisome β-oxidation and ω-oxidation, together with a decreased NADPH regeneration, may exacerbate oxidative stress. Phosphoproteome data showed downregulation of JAK2-STAT5B and upregulation of mTOR signaling pathway.ConclusionsThis study identified proteo-metabolomic signatures associated with the development of NAFLD in pituitary GHD. Evidence was found of oxidative stress imbalance resulting from abnormal fatty acid oxidation and NADPH regeneration, highlighting the role of GH deficiency in the development of NAFLD.

To enhance the durability of copper tuyeres in blast furnaces, it is necessary to apply coatings on their surfaces to protect against wear caused by high-temperature coke. In this study, a series of in-situ synthetic TiC-reinforced Stellite 6 alloy coatings containing a Ni60A interlayer were successfully produced on copper substrates using 5–45 wt% TiFe/Cr3C2 precursor powders. The coatings exhibited excellent metallurgical bonding with the Ni60A interlayer, and an elemental transition zone was formed at the interface between the coating and the interlayer. XRD Rietveld refinement analysis revealed that the T45 coating with 45 wt% of precursor powder showed the highest TiC phase content of 12.27 %, with an average hardness of 658.15 HV0.2, which was 31.2 % higher than that of the Stellite 6 alloy. The wear mechanism of the Stellite 6 coating was plastic delamination at 25 °C, and oxidative wear at 500 °C. As the TiC particle content in the coating increased, the dominant wear mechanism tended to shift towards abrasive wear. The specific wear mechanism of the coating was influenced by the content of coating phases and the operating temperature. The best wear resistance was achieved by the T45 coating, which showed an abrasive wear mechanism at both 25 °C and 500 °C, with a wear resistance of 4.47 and 5.94 times higher than that of the Stellite 6 alloy respectively.

Abstract Multi-source data fusion techniques are widely applied in dynamic target detection scenarios, such as target situational awareness, radar signal resolution, and feature fusion labeling. Currently, techniques including clustering, neural networks, Bayesian analysis, and machine learning have been applied to improve the success rate of multi-source data fusion in terms of interference data noise reduction. The research on data tampering prevention of multiple data sources is mainly based on the data distributed authentication technology. The research on performing data fusion process in a trusted execution environment is mainly based on cryptography and codec technology. This paper focuses on the technical application architecture that can effectively improve the comprehensive efficiency of multi-source data fusion processing under the constraints of business scenarios. Accordingly, this paper proposes a trusted execution environment architecture based on blockchain technology for multi-source data fusion scenarios. It integrates the strategy of trusted data source data verification in blockchain smart contracts into the typical multi-source data fusion application architecture. After comparison tests in a simulation environment, the trusted execution environment architecture based on blockchain technology has shown considerable improvements in fusion success rate with limited performance cost.

To clarify the influence of process parameters on the microstructure and wear resistance of coating, a series of Ni60 A alloy were deposited on copper surface by plasma cladding under different preheating temperatures. The obtained coatings were new alloy materials formed by the dilution of Ni60 A alloy with Cu, and their microstructure and properties were closely related to the dilution rate. With the increase of preheating temperature, the dilution rate of the coatings increased obviously, which led to a gradual increase of the copper content in the γ-(Cu, Fe, Ni) solid solution phase, thus resulting in compositional inhomogeneity and performance deterioration of the coating. From 500 °C to 800 °C (preheating temperature), the average hardness of the coating decreased from 680.3 HV0.1 to 406.6 HV0.1, and the wear resistance reduced by 2.8 times. The wear mechanism of the coating changed from abrasive wear to the combination of abrasive wear and adhesive wear. For practical applications, to ensure the excellent wear resistance of the coating, the preheating temperature should be controlled at a low level.

Abstract Accurate delineation of glioma infiltrative margins remains a challenge due to the low density of cancer cells in these regions. Here, a hierarchical imaging strategy to define glioma margins by locating the immunosuppressive tumor‐associated macrophages (TAMs) is proposed. A pH ratiometric fluorescent probe CP2‐M that targets immunosuppressive TAMs by binding to mannose receptor (CD206) is developed, and it subsequently senses the acidic phagosomal lumen, resulting in a remarkable fluorescence enhancement. With assistance of CP2‐M, glioma xenografts in mouse models with a tumor‐to‐background ratio exceeding 3.0 for up to 6 h are successfully visualized. Furthermore, by intra‐operatively mapping the pH distribution of exposed tissue after craniotomy, the glioma allograft in rat models is precisely excised. The overall survival of rat models significantly surpasses that achieved using clinically employed fluorescent probes. This work presents a novel strategy for locating glioma margins, thereby improving surgical outcomes for tumors with infiltrative characteristics.

In cross-domain hyperspectral image (HSI) classification, the labeled samples of the target domain are very limited, and it is a worthy attention to obtain sufficient class information from the source domain to categorize the target domain classes (both the same and new unseen classes). This article investigates this problem by employing few-shot learning (FSL) in a meta-learning paradigm. However, most existing cross-domain FSL methods extract statistical features based on convolutional neural networks (CNNs), which typically only consider the local spatial information among features, while ignoring the global information. To make up for these shortcomings, this article proposes novel convolutional transformer-based few-shot learning (CTFSL). Specifically, FSL is first performed in the classes of source and target domains simultaneously to build the consistent scenario. Then, a domain aligner is set up to map the source and target domains to the same dimensions. In addition, a convolutional transformer (CT) network is utilized to extract local-global features. Finally, a domain discriminator is executed subsequently that can not only reduce domain shift but also distinguish from which domain a feature originates. Experiments on three widely used hyperspectral image datasets indicate that the proposed CTFSL method is superior to the state-of-the-art cross-domain FSL methods and several typical HSI classification methods in terms of classification accuracy.

The multisource remote sensing classification task has two main challenges. 1) How to capture hyperspectral image (HSI) and light detection and ranging (LiDAR) features cooperatively to fully mine the complementary information between data. 2) How to adaptively fuse multisource features, which should not only overcome the imbalance between HSI and LiDAR data but also avoid the generation of redundant information. The local information interaction transformer (LIIT) model proposed herein can effectively address these above issues. Specifically, multibranch feature embedding is first performed to help in the fine-grained serialization of multisource features; subsequently, a local-based multisource feature interactor (L-MSFI) is designed to explore HSI and LiDAR features together. This structure provides an information transmission environment for multibranch features and further alleviates the homogenization processing mode of the self-attention process. More importantly, a multisource feature selection module (MSTSM) is developed to dynamically fuse HSI and LiDAR features to solve the problem of insufficient fusion. Experiments were carried out on three multisource remote-sensing classification datasets, the results of which show that LIIT has more performance advantages than the state-of-the-art CNN and transformer methods.

The heat alternation and bending during the service process of the pipeline structure are the key issues affecting its residual strength. The nonuniform plastic deformation of its cross-section causes nonlinear damage to the structure, which makes it difficult to calculate the residual strength of the overall structure. This study proposed a novel variable-strength material damage model with damage accumulation, which achieved the coupling analysis of structural damage and material fatigue residual strength, making the structural strength of nonlinear damage cross-section computable. Based on this, a theoretical ultimate load model for nonlinear damage cross-section and a finite element analysis model for dynamic material strength with damage accumulation were established. The analysis was conducted on the repeated banding damage to coiled tubing. The results showed that the residual yield strength of nonlinear damaged coiled tubing showed a trend of first rapid decrease and then slow decrease with damage accumulation, and the tensile displacement showed an increasing trend with damage accumulation. The ultimate internal pressure strength of nonlinear damaged structures showed a similar downward trend to the yield strength. The nonlinear damage coupling analysis model proposed in this study has significant practical engineering application value.

Tuina, a method of traditional Chinese manual manipulation, is an effective alternative therapy for neuropathic pain (NP), but its analgesic mechanism remains unclear. In this study, we used resting-state functional magnetic resonance imaging (R-fMRI) to explore the analgesic mechanism of Tuina in an NP rat model. After undergoing surgery to induce chronic compression of the dorsal root ganglion (CCD), one group of rats underwent Tuina at the ipsilateral BL40 acupoint once a day for 10 min during the 25 days following surgery while another group did not. Behavioral tests were performed at baseline, on the third day following surgery, and once a week for the next 4 weeks. R-fMRI was performed at baseline and 7 days and 28 days following surgery. Behavioral testing revealed that the Tuina group presented a significant response improvement to mechanical and thermal nociception stimuli compared to the untreated group 2 weeks following CCD surgery. Interestingly, rats submitted to Tuina presented higher measures of spontaneous neuronal activity in basal forebrain region, primary somatosensory cortex barrel field, dentate gyrus, secondary somatosensory cortex, striatum, descending corticofugal pathways, and globus pallidum of the left hemisphere 4 weeks after the CCD surgery compared to rats having undergone CCD only. In addition, on the 28th day, the ALFF signals of the left dentate gyrus, left secondary somatosensory cortex, left striatum, and bilateral primary cingulate cortex were significantly increased while those in the right dentate gyrus and bilateral periaqueductal gray were significantly decreased compared to those on the 7th day. Correlation analysis showed that the ALFF values of the left descending corticofugal pathways and globus pallidum had a positive correlation with mechanical withdrawal threshold and paw withdrawal thermal latency tests. Altogether, these results indicate that NPP induced by CCD surgery affects the plasticity of the cerebral cortex, and that Tuina alleviate pain behavior by promoting cortical remodeling.

IntroductionInborn errors of immunity (IEI) are a heterogeneous group of disorders characterized by increased risk of infections, autoimmunity, autoinflammatory diseases, malignancy and allergy. Next-generation sequencing has revolutionized the identification of genetic background of these patients and assists in diagnosis and treatment. In this study, we identified a probable unique monogenic cause of IEI, and evaluated the immunological methods and pathogenic detections.MethodsA family with a member with a clinical diagnosis of IEI was screened by whole genomic sequencing (WGS). Demographic data, clinical manifestations, medical history, physical examination, laboratory findings and imaging features of the patient were extracted from medical records. Comprehensive immune monitoring methods include a complete blood count with differential, serum levels of cytokines and autoantibodies, T-cell and B-cell subsets analysis and measurement of serum immunoglobulins. In addition, metagenomic sequencing (mNGS) of blood, cerebrospinal fluid and biopsy from small intestine were used to detect potential pathogens.ResultsThe patient manifested with recurrent infections and autoimmune disorders, who was eventually diagnosed with IEI. Repetitive mNGS tests of blood, cerebrospinal fluid and biopsy from small intestine didn’t detect pathogenic microorganism. Immunological tests showed a slightly decreased level of IgG than normal, elevated levels of tumor necrosis factor and interleukin-6. Lymphocyte flow cytometry showed elevated total B cells and natural killer cells, decreased total T cells and B-cell plasmablasts. WGS of the patient identified a novel heterozygous mutation in IRF2BP2 (c.439_450dup p. Thr147_Pro150dup), which was also confirmed in his father. The mutation was classified as variant of uncertain significance (VUS) according to the American College of Medical Genetics and Genomics guidelines.ConclusionWe identified a novel IRF2BP2 mutation in a family with a member diagnosed with IEI. Immune monitoring and WGS as auxiliary tests are helpful in identifying genetic defects and assisting diagnosis in patients with clinically highly suspected immune abnormalities and deficiencies in inflammation regulation. In addition, mNGS techniques allow a more comprehensive assessment of the pathogenic characteristics of these patients. This report further validates the association of IRF2BP2 deficiency and IEI, and expands IEI phenotypes.

Abstract Background Despite limited efficiency, modulation of microglia/macrophages has shown to attenuate neuroinflammation after intracerebral hemorrhage (ICH). In this context, we evaluated the efficacy of modified exosomal signal regulatory protein α (SIRPα) variants (SIRPα-v Exos) in microglia/macrophages and neuroinflammation-associated white matter injury after ICH. Methods SIRPα-v Exos were engineered to block CD47-SIRPα interactions. After obtaining SIRPα-v Exos from lentivirus-infected mesenchymal stem cells, C57BL/6 mice suffering from ICH underwent consecutive intravenous injections of SIRPα-v Exos (6 mg/kg) for 14 days. Afterwards, the volume of hematoma and neurological dysfunctions were assessed in mice continuously until 35 days after ICH. In addition, demyelination, electrophysiology and neuroinflammation were evaluated. Furthermore, the mechanisms of microglial regulation by SIRPα-v Exos were investigated in vitro under coculture conditions. Results The results demonstrated that the clearance of hematoma in mice suffering from ICH was accelerated after SIRPα-v Exo treatment. SIRPα-v Exos improved long-term neurological dysfunction by ameliorating white matter injury. In addition, SIRPα-v Exos recruited regulatory T cells (Tregs) to promote M2 polarization of microglia/macrophages in the peri-hematoma tissue. In vitro experiments further showed that SIRPα-v Exos regulated primary microglia in a direct and indirect manner in synergy with Tregs. Conclusion Our studies revealed that SIRPα-v Exos could accelerate the clearance of hematoma and ameliorate secondary white matter injury after ICH through regulation of microglia/macrophages. SIRPα-v Exos may become a promising treatment for ICH in clinical practice. Graphical Abstract

Repetitive TMS has been used as an alternative treatment for various neurological disorders. However, most TMS mechanism studies in rodents have been based on the whole brain stimulation, the lack of rodent-specific focal TMS coils restricts the proper translation of human TMS protocols to animal models. In this study, we designed a new shielding device, which was made of high magnetic permeability material, to enhance the spatial focus of animal-use TMS coils. With the finite element method, we analyzed the electromagnetic field of the coil with and without the shielding device. Furthermore, to assess the shielding effect in rodents, we compared the c-fos expression, the ALFF and ReHo values in different groups following a 15 min 5 Hz rTMS paradigm. We found that a smaller focality with an identical core stimulation intensity was achieved in the shielding device. The 1 T magnetic field was reduced from 19.1 mm to 13 mm in diameter, and 7.5 to 5.6 mm in depth. However, the core magnetic field over 1.5 T was almost the same. Meanwhile, the area of electric field was reduced from 4.68 cm2 to 4.19 cm2, and 3.8 mm to 2.6 mm in depth. Similar to this biomimetic data, the c-fos expression, the ALFF and ReHo values showed more limited cortex activation with the use of the shielding device. However, compared to the rTMS group without the shielding application, more subcortical regions, like the striatum (CPu), the hippocampus, the thalamus, and the hypothalamus were also activated in the shielding group. This indicated that more deep stimulation may be achieved by the shielding device. Generally, compared with the commercial rodents’ TMS coil (15 mm in diameter), TMS coils with the shielding device achieved a better focality (~6 mm in diameter) by reducing at least 30% of the magnetic and electric field. This shielding device may provide a useful tool for further TMS studies in rodents, especially for more specific brain area stimulation.

In recent times, multisource remote sensing technology [e.g., hyperspectral image (HSI) and light detection and ranging (LiDAR) data] has been widely used in urban land-use recognition owing to its high classification effectiveness compared to using only single-source data. In this study, a multiview hierarchical network (MVHN) technique is developed for HSI and LiDAR data classification, which conducts the following execution procedures. First, based on the a preset band step length, the original HSI is sampled and divided into multiple groups with exactly the same number of bands to obtain spectral features. Then, principal components analysis is performed on the raw HSI to extract the first principal components (PCs) that meet the size of the LiDAR image. The Gabor filters are applied to the PCs and LiDAR to capture spatial details (i.e., textural features) of scenes. Specifically, a stacking mechanism is employed to generate fusion features once the above features are available. Next, a three-dimensional ResNet-like deep CNN is designed to extract the spectral–spatial information of the fusion feature. Finally, majority-voting is introduced into the classification results of the network trained using each fusion feature to achieve high-confidence final results. Experiments on three well-known HSI and LiDAR datasets (i.e., Houston, MUUFL, and Trento datasets) demonstrate the effectiveness of the proposed MVHN method compared to state-of-the-art comparable classification methods.

Anther development is a programmed biological process crucial to plant male reproduction. Genome-wide analyses on the functions of transcriptional factor (TF) genes and their microRNA (miRNA) regulators contributing to anther development have not been comprehensively performed in maize. Here, using published RNA-Seq and small RNA-Seq (sRNA-Seq) data from maize anthers at ten developmental stages in three genic male-sterility (GMS) mutants (ocl4, mac1, and ms23) and wild type W23, as well as newly sequenced maize anther transcriptomes of ms7-6007 and lob30 GMS mutants and their WT lines, we analyzed and found 1079 stage-differentially expressed (stage-DE) TF genes that can be grouped into six (premeiotic, meiotic, postmeiotic, premeiotic-meiotic, premeiotic-postmeiotic, and meiotic-postmeiotic clusters) expression clusters. Functional enrichment combined with cytological and physiological analyses revealed specific functions of genes in each expression cluster. In addition, 118 stage-DE miRNAs and 99 miRNA-TF gene pairs were identified in maize anthers. Further analyses revealed the regulatory roles of zma-miR319 and zma-miR159 as well as ZmMs7 and ZmLOB30 on ZmGAMYB expression. Moreover, ZmGAMYB and its paralog ZmGAMYB-2 were demonstrated as novel maize GMS genes by CRISPR/Cas9 knockout analysis. These results extend our understanding on the functions of miRNA-TF gene regulatory pairs and GMS TF genes contributing to male fertility in plants.

The evolution of microstructure and wear resistance of Co-based alloy coating reinforced with WC phase prepared on copper substrate by plasma cladding were investigated. As the WC contents increased, several carbide phases appeared in the coating, the shape of these carbide phases grew from punctate and flower to block and dendrite. The proportion of these carbide phases such as M3W3C and M6W6C in the coating increased, which caused the average hardness to improve from 553.43 HV0.1 to 954.64 HV0.1. Compared with pure cobalt-based alloy coating, WC/cobalt-based coating had a higher oxidation degree and forms CoWO4 phase. The Co-based coating with 40% WC exhibited relatively good wear resistance at room temperature, 400 °C and 500 °C. At 600 °C, the sample with 10% WC possessed the lowest wear weight loss.

We leveraged a novel index of diffusion MRI to investigate the relationships among cortical free water, macro-organizations and gene expression in healthy adults. Few research has been conducted to investigate the role of free water in the healthy adults due to it can easily be affected also by aging diseases. High quality data of 350 subjects from Human Connectome Project were used in our study. Cortical free water was estimated by using a bi-tensor model. The free water was high in the limbic, insular and somatosensory cortex, while being lower in motor and association cortex. The negative correlation between the free water and cortical thickness has been consistently identified in almost all the cortical regions. Negative correlation between the cortical free water and structural covariance (rho=-0.38, pspin=0.005) revealed the free water was sensitive to cortical heterogeneity. Using human gene expression dataset, we found the gene expression pattern of the relationship between the free water and cortical thickness spatially coupled with primary gradient of structural covariance network (rho=0.40, pspin=0.004). Our findings indicated the free water was sensitive to the cortical cellular status. The relationship between free water and macroscale organization also reflected hierarchal structures of cerebral cortex.

Soil moisture (SM) affects ecosystem homeostasis through interactions among land use type (LUT), precipitation, heat and human activity (HA). In the core areas of China Loess Plateau, we selected three sites along gradients of annual precipitation and annual accumulated temperature ≥ 0℃ and compare SM variations among three LUTs, including cropland, woodland, and rangeland, and in three precipitation years, including dry, wet, and normal year. The SM of three LUTs were sensitive to wet years in the north, dry years in the south, and both wet and dry years in the middle, respectively. From north to south, water use (WU) increased while water use efficiency (WUE) decreased. The WU for cropland was the highest and WUE was the lowest, while rangeland had the opposite tendency. The mean annual water deficiencies for cropland, rangeland and woodland were 42.41 mm, 21.38 mm, and 15.49 mm, respectively. HA for three LUTs increased as precipitation increased and the order was cropland > woodland > rangeland. The stability of SM to precipitation was as following order: rangeland > woodland > cropland. Rangeland was insensitive to precipitation, while cropland was sensitive to precipitation. Productivity of rangeland and cropland were stabilized mainly by increasing resistance to SM and the resilience to SM, respectively. To maximize the WUE of the rain fed system and enhance the stability of SM, the area ratios of cropland, woodland, and rangeland were recommended as 4:2:19, 3:2:5 and 3:2:4 in the northern, central, and southern regions, respectively.

Rheumatoid arthritis (RA) is an autoimmune disease and is mainly characterized by abnormal proliferation of fibroblast-like synoviocytes (FLS). The up-regulated cellular membrane expression of G protein coupled receptor kinase 2 (GRK2) of FLS plays a critical role in RA progression, the increase of GRK2 translocation activity promotes dysfunctional prostaglandin E4 receptor (EP4) signaling and FLS abnormal proliferation. Recently, although our group found that paeoniflorin-6ʹ-O-benzene sulfonate (CP-25), a novel compound, could reverse FLS dysfunction via GRK2, little is known as to how GRK2 translocation activity is suppressed. Our findings revealed that GRK2 expression up-regulated and EP4 expression down-regulated in synovial tissues of RA patients and collagen-induced arthritis (CIA) rats, and prostaglandin E2 (PGE2) level increased in arthritis. CP-25 could down-regulate GRK2 expression, up-regulate EP4 expression, and improve synovitis of CIA rats. CP-25 and GRK2 inhibitors (paroxetine or GSK180736A) inhibited the abnormal proliferation of FLS in RA patients and CIA rats by down-regulating GRK2 translocation to EP4 receptor. The results of microscale thermophoresis (MST), cellular thermal shift assay, and inhibition of kinase activity assay indicated that CP-25 could directly target GRK2, increase the protein stability of GRK2 in cells, and inhibit GRK2 kinase activity. The docking of CP-25 and GRK2 suggested that the kinase domain of GRK2 might be an important active pocket for CP-25. G201, K220, K230, A321, and D335 in kinase domain of GRK2 might form hydrogen bonds with CP-25. Site-directed mutagenesis and co-immunoprecipitation assay further revealed that CP-25 down-regulated the interaction of GRK2 and EP4 via controlling the key amino acid residue of Ala321 of GRK2. Our data demonstrate that FLS proliferation is regulated by GRK2 translocation to EP4. Targeted inhibition of GRK2 kinase domain by CP-25 improves FLS function and represents an innovative drug for the treatment of RA by targeting GRK2.

Aiming to improve the surface properties of copper, a Ni-Cr-B-Si coating was fabricated by plasma transferred arc (PTA) cladding. The coating formed a good metallurgical bonding with the copper substrate, and the dilution rate was about 15.9%. The coating mainly contained γ-(Cu, Fe, Ni), Cr23C6, CrB and Ni3Si phases, which played a role of solid solution strengthening and dispersion strengthening. The hardness of the coating was significantly improved compared to the copper substrate, which was approximately 7.8 times that of the substrate. The wear resistance of the coating was 5.0 times higher than that of the copper substrate, and the wear mechanism was abrasive wear.

Abstract Blood pressure (BP) variability may have its effect on the development of vascular disease. The authors aimed to examine the association between the visit‐to‐visit variability (VVV) of BP and arterial stiffness in Chinese adults. The authors included 1407 participants from a prospective cohort study of community residents who were ≥40 years, without a history of myocardial infarction or stroke, and with data at the baseline, the second and the third visits in 2008, 2009, and 2013. The VVV of BP was defined as the standard deviation (SD), the coefficient of variation (CV), the average successive variability (ASV), and the variability independent of the mean (VIM) in BP levels at the 3 visits. Arterial stiffness was measured by brachial‐ankle pulse wave velocity (ba‐PWV) at the 2nd and the 3rd visits. Levels of ba‐PWV change and the occurrence of an elevated ba‐PWV increased significantly in the highest tertile of VVV measures of systolic BP (SBP) and pulse pressure (PP) compared with the lowest tertile, respectively. The multivariable regression analysis revealed that VVV measures of SBP and PP were significantly associated with levels of ba‐PWV change and the risks of developing an elevated ba‐PWV. The odds ratios (ORs) and 95% confidence intervals (CIs) for the risk were 2.12 (1.57–3.12) and 1.92 (1.38–2.68) in participants with the highest versus the lowest tertile of SBP‐SD and PP‐SD, respectively. No significant association was found for diastolic BP variability measures. The increased long‐term variabilities of SBP and PP were associated with an increased risk of arterial stiffness.

Microstructure evolution of the joints formed by Ag-6.6 mol% Cu brazing BaCo0.7Fe0.2Nb0.1O3-δ (BCFN) ceramic after long-term heat treatment in different atmospheres was revealed. After heat treatment in air, the layered structure in the original interfacial reaction layer disappears, accompanied by the aggregation and growth of the interfacial products. The thickness of the reaction layer increased first and then stabilized. Under the action of the concentration gradient, the residual copper oxide diffused along the pores of BCFN matrix and continued to react with the matrix until it exhausted. The types of interfacial products remained unchanged, which were Ba2Cu2O5 and Co-Cu-O (solid solution). However, serious decomposition of interfacial products (Ba2Cu2O5) occurred due to the low oxygen partial pressure after heat treatment in argon, resulting in the formation of holes and cracks. Part of cobalt precipitated from the BCFN matrix and formed a cobalt-rich phase at the junction of Ag-Cu alloy and interfacial reaction zone. Results showed that the joint microstructure is unstable at high temperature. Controlling the amount of the interfacial products formed during brazing is key to improving the stability of the joint, which provides a theoretical support for further optimizing the sealing quality of membrane components.

Au fil du développement de la crise sanitaire liée à la COVID-19, de plus en plus de pays ont adopté le dispositif de type « pass sanitaire » pour tenter d’endiguer la propagation du virus. Cet article prend le « pass sanitaire » français comme objet de recherche, en le comparant avec le QR code de santé chinois. Il affirme que, premièrement, la conception et l’usage du « pass sanitaire » favorise l’indépendance technologique et la gouvernance numérique en France. Deuxièmement, bien que l’usage technique du QR code soit différent, de StopCovid au « pass sanitaire », il construit tout de même une surveillance du grand public à travers un dispositif numérique en France comme en Chine. Enfin, cette QR code-isation sociale renforce une potentielle classification des individus reposant sur la collecte des données personnelles.

Abstract Surgeons face challenges in intraoperatively defining margin of brain tumors due to its infiltrative nature. Extracellular acidosis caused by metabolic reprogramming of cancer cells is a reliable marker for tumor infiltrative regions. Although the acidic margin‐guided surgery shows promise in improving surgical prognosis, its clinical transition is delayed by having the exogenous probes approved by the drug supervision authority. Here, an intelligent surface‐enhanced Raman scattering (SERS) navigation system delineating glioma acidic margins without administration of exogenous probes is reported. With assistance of this system, the metabolites at the tumor cutting edges can be nondestructively transferred within a water droplet to a SERS chip with pH sensitivity. Homemade deep learning model automatically processes the Raman spectra collected from the SERS chip and delineates the pH map of tumor resection bed with increased speed. Acidity correlated cancer cell density and proliferation level are demonstrated in tumor cutting edges of animal models and excised tissues from glioma patients. The overall survival of animal models post the SERS system guided surgery is significantly increased in comparison to the conventional strategy used in clinical practice. This SERS system holds the promise in accelerating clinical transition of acidic margin‐guided surgery for solid tumors with infiltrative nature.

The effect of carbon dioxide (CO2) on the desulfurization of sintering flue gas with hydrate (Ca(OH)2) as an absorbent was investigated, and the formation of calcium carbonate (CaCO3) and its effect on the desulfurization was discussed. The competitive relationship between carbon dioxide (CO2) and sulfur dioxide (SO2) with the deacidification agent in sintering flue gas is discussed thermodynamically, showing that sulfates are more likely to be generated under high oxygen potential conditions and that SO2 reacts more preferentially than CO2 under a thermodynamic standard state. The amount of produced CaCO3 increases under the condition that the CO2 concentration is absolutely dominant to SO2 in the sintering flue gas atmosphere. The effect of temperature, humidity and CO2 concentration on the desulfurization of Ca(OH)2 are discussed experimentally. The increasing temperature is not conducive to desulfurization, and the humidity can promote desulfurization, while excessive humidity could inhibit desulfurization. The suitable relative humidity is 20%. In situ generated calcium carbonate has a certain desulfurization effect, but the desulfurization effect is not as good as Ca(OH)2. However, a large proportion of CaCO3 was produced in the desulfurization ash under the condition that CO2 concentration was absolutely dominant to SO2 in the sintering flue gas atmosphere.

Abstract Background The activation of NOD-like receptor protein 3 (NLRP3) inflammasome-dependent pyroptosis has been shown to play a vital role in the pathology of manganese (Mn)-induced neurotoxicity. Sodium para-aminosalicylic acid (PAS-Na) has a positive effect on the treatment of manganism. However, the mechanism is still unclear. We hypothesized that PAS-Na might act through NLRP3. Methods The microglial cell line BV2 and male Sprague-Dawley rats were used to investigate the impacts of PAS-Na on Mn-induced NLRP3 inflammasome-dependent pyroptosis. The related protein of the NF-κB pathway and NLRP3-inflammasome-dependent pyroptosis was detected by western blot. The reactive oxygen species and mitochondrial membrane potential were detected by immunofluorescence staining and flow cytometry. The activation of microglia and the gasdermin D (GSDMD) were detected by immunofluorescence staining. Results Our results showed that Mn treatment induced oxidative stress and activated the NF-κB pathway by increasing the phosphorylation of p65 and IkB-α in BV2 cells and in the basal ganglia of rats. PAS-Na could alleviate Mn-induced oxidative stress damage by inhibiting ROS generation, increasing mitochondrial membrane potential and ATP levels, thereby reducing the phosphorylation of p65 and IkB-α. Besides, Mn treatment could activate the NLRP3 pathway and promote the secretion of IL-18 and IL-1β, mediating pyroptosis in BV2 cells and in the basal ganglia and hippocampus of rats. But an inhibitor of NF-κb (JSH-23) treatment could significantly reduce LDH release, the expression of NLRP3 and Cleaved CASP1 protein and IL-1β and IL-18 mRNA level in BV2 cells. Interestingly, the effect of PAS-Na treatment in Mn-treated BV2 cells is similar to those of JSH-23. Besides, immunofluorescence results showed that PAS-Na reduced the increase number of activated microglia, which stained positively for GSDMD. Conclusion PAS-Na antagonized Mn-induced NLRP3 inflammasome dependent pyroptosis through inhibiting NF-κB pathway activation and oxidative stress.

Purpose: Severe early childhood caries (S-ECC) is a microbe-mediated disease with tooth hard tissue destruction. However, the role of the fungal community in various ecological niches of deciduous dental caries has not been fully elucidated. This study aimed to analyze the differences of mycobiome in diverse spatial niches with and without S-ECC.Method: A total of 48 samples were obtained from 8 S-ECC children (SE group) and 8 caries-free children (CF group) aged 4–5 years. Unstimulated saliva (S), healthy supragingival plaque (FMIX), mixed plaque from decayed teeth (SMIX) and carious lesion (DMIX) samples were collected. The ITS2 region of the fungi was amplified and sequenced using the Ion S5™XL platform.Results: A total of 281 species were identified. Candida albicans showed relatively higher abundance in S-ECC children, while Alternaria alternata and Bipolaris sorokiniana were more enriched in CF group. In this study, the relative abundance of C. albicans in CF.FMIX (0.4%), SE.FMIX (12.5%), SE.SMIX (24.0%), and SE.DMIX (37.2%) increased successively. Significant differences of fungal species richness and diversity were observed between SE.FMIX-SE.SMIX, SE.FMIX-SE.DMIX (P < 0.05).Conclusions: The diversity of fungal communities in S-ECC children showed significant differences in various spatial niches of primary teeth. The richness of C. albicans was closely related to the caries states and depth, suggesting that it may play a crucial role in caries pathogenicity.

ObjectiveThe aim of the study was to investigate the association between the visit-to-visit variability (VVV) of fasting plasma glucose (FPG) and arterial stiffness in Chinese adults.MethodsWe performed a cohort study involving 2002 Chinese adults with no history of myocardial infarction or stroke. All the participants attended three visits (the baseline visit in 2008, the 2nd visit in 2009 and the 3rd visit in 2013). We used four measures to define the VVV of FPG across the three visits: the standard deviation (SD), the coefficient of variation (CV), the average successive variability (ASV) and the variability independent of the mean (VIM). We used brachial-ankle pulse wave velocity (ba-PWV) to measure arterial stiffness at the 2nd and the 3rd visits.ResultsCompared with the lowest tertile of all the four measurements of VVV of FPG, significantly increased levels of ba-PWV change, ratio of ba-PWV change and the occurrence of the elevated ba-PWV were found in the highest tertile. The odds ratio (OR) and 95% confidence interval (CI) comparing participants in the highest tertile vs. the lowest tertile of FPG-SD was 1.37 (1.01-1.86) for risks of having elevated ba-PWV, even after adjustment for covariates including the mean FPG. Similar results were found for FPG-CV and FPG-VIM.ConclusionGreater long-term variability of FPG was associated with an increased risk of arterial stiffness, suggesting that the VVV of FPG could be used for an early detection of subclinical atherosclerosis.

Objective: Hypopituitarism (Hypo-Pit) is partial or complete insufficiency of anterior pituitary hormones. Besides hormone metabolism, the global metabolomics in Hypo-Pit are largely unknown. We aimed to explore potential biomarkers to aid in diagnosis and personalized treatment.Methods: Using both univariate and multivariate statistical methods, we identified 72 differentially abundant features through liquid chromatography coupled to high-resolution mass spectrometry, obtained in 134 males with Hypo-Pit and 90 age matched healthy controls.Results: Hypopituitarism exhibits an increased abundance of metabolites involved in amino acid degradation and glycerophospholipid synthesis, but decreased content of metabolites in steroid hormone synthesis and fatty acid beta-oxidation. Significantly changed metabolites included creatine, creatinine, L-alanine, phosphocholines, androstenedione, hydroprenenolone, and acylcarnitines. In Hypo-Pit patients, the increased ratio of creatine/creatinine suggested reduced creatine uptake and impaired creatine utilization, whereas the decreased level of beta-hydroxybutyrate, acetylcarnitine (C2) and a significantly decreased ratio of decanoylcarnitine (C10) to free carnitine suggested an impaired beta-oxidation. Furthermore, the creatine/creatinine and decanoylcarnitine/carnitine ratio were identified as diagnostic biomarkers for Hypo-Pit with AUCs of 0.976 and 0.988, respectively. Finally, we found that the creatinine and decanoylcarnitine/carnitine ratio could distinguish cases that were sensitive vs. resistant to human chorionic gonadotropin therapy.Conclusion: We provided a global picture of altered metabolic pathways in Hypo-Pit, and the identified biomarkers in creatine metabolism and beta-oxidation might be useful for the preliminary screening and diagnosis of Hypo-Pit.

G protein-coupled receptor kinase 2 (GRK2), as a vital Ser/Thr kinase, is an important regulatory protein in the inflammatory immune response (IIR) by maintaining the balance between the function of inflammatory immune cells and non-conventional inflammatory immune cells and regulating inflammatory immune cell infiltration, inflammatory cytokine secretion, and the signaling associated with endothelial function. However, the imbalance of GRK2 expression and activity plays an important role in the development of IIR-related diseases, such as hypertension, heart failure, Alzheimer’s disease, type 2 diabetes mellitus, insulin resistance, rheumatoid arthritis, thyroid cancer, multiple sclerosis, and liver cancer. Small molecule GRK2 inhibitors, including balanol, Takeda inhibitors, paroxetine and derivatives, M119 and gallein, peptides, RNA aptamers, Raf kinase inhibitory protein, and microRNAs, that can directly inhibit GRK2 kinase activity have been identified by different strategies. This review discusses recent progress in one of the hallmark molecular abnormalities of GRK2 in IIR-related diseases and explores the soft regulation of IIR by innovative drugs reducing the excessive activity of GRK2 to basal levels, without damaging normal physiological function, to ameliorate inflammatory disorders.

Objective: To explore the most suitable calculation method for insulin dosage in an insulin tolerance test (ITT) and to evaluate the clinical application value of the optimization coefficient (γ).Methods: In this study, 140 adult patients with congenital growth hormone deficiency (GHD) or acquired hypopituitarism were randomized into the following two groups: the conventional group (n = 70) and the optimized group (n = 70). Oral glucose tolerance tests (OGTTs), insulin release tests (IRTs), and ITTs were conducted. For ITTs, insulin doses were the product of body weight (kg) and related coefficient (0.15 IU/kg for the control group and γ IU/kg for the optimized group, respectively). Notably, γ was defined as −0.034 + 0.000176 × AUCINS + 0.009846 × BMI, which was based on our previous study.Results: In the ITTs, the rate of achieving adequate hypoglycemia with a single insulin dose was significantly higher for the optimized group compared with the conventional group (92.9 vs. 60.0%, P < 0.001). The optimized group required higher initial doses of insulin (0.23 IU/kg). Meanwhile, the two groups did not differ significantly in their nadir blood glucose (1.9 vs. 1.9 mmol/L, P = 0.828).Conclusion: This study confirmed that the proposed optimized calculation method for insulin dosage in ITTs led to more efficient hypoglycemia achievement, without increasing the incidence of serious adverse events.

Distinct brain remodeling has been found after different nerve reconstruction strategies, including motor representation of the affected limb. However, differences among reconstruction strategies at the brain network level have not been elucidated. This study aimed to explore intra-network changes related to altered peripheral neural pathways after different nerve reconstruction surgeries, including nerve repair, end-to-end nerve transfer, and end-to-side nerve transfer. Sprague–Dawley rats underwent complete left brachial plexus transection and were divided into four equal groups of eight: no nerve repair, grafted nerve repair, phrenic nerve end-to-end transfer, and end-to-side transfer with a graft sutured to the anterior upper trunk. Resting-state brain functional magnetic resonance imaging was obtained 7 months after surgery. The independent component analysis algorithm was utilized to identify group-level network components of interest and extract resting-state functional connectivity values of each voxel within the component. Alterations in intra-network resting-state functional connectivity were compared among the groups. Target muscle reinnervation was assessed by behavioral observation (elbow flexion) and electromyography. The results showed that alterations in the sensorimotor and interoception networks were mostly related to changes in the peripheral neural pathway. Nerve repair was related to enhanced connectivity within the sensorimotor network, while end-to-side nerve transfer might be more beneficial for restoring control over the affected limb by the original motor representation. The thalamic-cortical pathway was enhanced within the interoception network after nerve repair and end-to-end nerve transfer. Brain areas related to cognition and emotion were enhanced after end-to-side nerve transfer. Our study revealed important brain networks related to different nerve reconstructions. These networks may be potential targets for enhancing motor recovery.

Few studies have examined the association between Asian dietary pattern and prediabetes, in particular, the Chinese diet. We conducted a cross-sectional study to identify dietary patterns associated with impaired fasting glucose (IFG) which considered a state of prediabetes in Chinese men. The study included 1495 Chinese men aged 20 to 75 years. Information about diet was obtained using an 81-item food frequency questionnaire (FFQ), and 21 predefined food groups were considered in a factor analysis. Three dietary patterns were generated by factor analysis: (1) a vegetables-fruits pattern; (2) an animal offal-dessert pattern; and (3) a white rice-red meat pattern. The multivariate-adjusted odds ratio (OR) of IFG for the highest tertile of the animal offal-dessert pattern in comparison with the lowest tertile was 3.15 (95% confidence intervals (CI): 1.87–5.30). The vegetables-fruits dietary pattern was negatively associated with the risk of IFG, but a significant association was observed only in the third tertile. There was no significant association between IFG and the white rice-red meat pattern. Our findings indicated that the vegetables-fruits dietary pattern was inversely associated with IFG, whereas the animal offal-dessert pattern was associated with an increased risk of IFG in Chinese men. Further prospective studies are needed to elucidate the diet-prediabetes relationships.

Main rotor actuator failure leads to catastrophic accidents for single main rotor helicopters. This paper focuses on safe landing trajectories after an actuator is locked in place by the remaining actuators, without introducing other control inputs. A general swashplate geometry is described, and new reconfiguration solutions for the control mixer are presented. The safe landing trajectories are obtained by formulating a nonlinear optimal control problem based on a nonlinear helicopter dynamic model and geometry constraints due to actuator failure. Safe landing trajectory results are shown with various initial forward velocities of all actuator failure cases. The safe initial speed boundaries are also explored by employing speed sweeps.

Lateral organ boundaries (LOB) domain (LBD) genes, a gene family encoding plant-specific transcription factors, play important roles in plant growth and development. At present, though there have been a number of genome-wide analyses on LBD gene families and functional studies on individual LBD proteins, the diverse functions of LBD family members still confuse researchers and an effective strategy is required to summarize their functional diversity. To further integrate and improve our understanding of the phylogenetic classification, functional characteristics and regulatory mechanisms of LBD proteins, we review and discuss the functional characteristics of LBD proteins according to their classifications under a phylogenetic framework. It is proved that this strategy is effective in the anatomy of diverse functions of LBD family members. Additionally, by phylogenetic analysis, one monocot-specific and one eudicot-specific subclade of LBD proteins were found and their biological significance in monocot and eudicot development were also discussed separately. The review will help us better understand the functional diversity of LBD proteins and facilitate further studies on this plant-specific transcription factor family.

Lanxangia tsaoko is a famous Chinese traditional medicine with long history. In this study, the complete chloroplast (cp) genome of L. tsaoko was characterized and assembled using a high-throughput sequencing method. The complete cp genome is 164,008 bp in length, including a large single copy (LSC) region of 89,134 bp and a small single copy (SSC) region of 15,372 bp separated by two inverted repeat (IR) regions of 29,751 bp. A total of 137 genes were identified, including 88 protein-coding genes (PCG), 38 transfer RNA genes, 8 ribosomal RNA genes, and 3 pseudogenes. The phylogenetic analysis revealed that L. tsaoko is located in Zingiberaceace, closely related to Amomum and Alpinia with a 100% bootstrap support.

Experiments were conducted to investigate forced convective cooling performance of an air cooled parallel plate fin heat sink with and without circular pin fins between the plate fins. The original parallel plate heat sink was fabricated consist of 9 parallel plates of length 53 mm with cross-sectional area of 1.4 mm in width by 20 mm height for each plate. The second heat sink has the same geometry of original one but with some circular pins between the plate fins. Thermal and hydrodynamics performances of the heat sinks have been assessed from the results obtained for the pressure drop, thermal resistance and overall performance with the free stream air velocity ranging from 4.7 to 12.5 m/s. Results show that the free stream air velocity has a significant effect on the thermal and hydrodynamics performance of the system. With increasing free stream velocity, the heat transfer coefficient increases and consequently the thermal resistance decreases while pressure drop increases due to higher inertial of fluid at higher velocities. Furthermore, at the same free stream air velocity the thermal resistance for the heat sink with circular pin is about 37.7% lower than that of the original heat sink.

Compressed air energy storage (CAES) is a commercial, utility-scale technology that provides long-duration energy storage with fast ramp rates and good part-load operation. It is a promising storage technology for balancing the large-scale penetration of renewable energies, such as wind and solar power, into electric grids. This study proposes a CAES-CC system, which is based on a conventional CAES combined with a steam turbine cycle by waste heat boiler. Simulation and thermodynamic analysis are carried out on the proposed CAES-CC system. The electricity and heating rates of the proposed CAES-CC system are lower than those of the conventional CAES by 0.127 kWh/kWh and 0.338 kWh/kWh, respectively, because the CAES-CC system recycles high-temperature turbine-exhausting air. The overall efficiency of the CAES-CC system is improved by approximately 10% compared with that of the conventional CAES. In the CAES-CC system, compressing intercooler heat can keep the steam turbine on hot standby, thus improving the flexibility of CAES-CC. This study brought about a new method for improving the efficiency of CAES and provided new thoughts for integrating CAES with other electricity-generating modes.

An improved flexible solar-aided power generation system (SAPG) for enhancing both selective catalytic reduction (SCR) de-NOx efficiency and coal-based energy efficiency of coal-fired power plants is proposed. In the proposed concept, the solar energy injection point is changed for different power plant loads, bringing about different benefits for coal-fired power generation. For partial/low load, solar energy is beneficially used to increase the flue gas temperature to guarantee the SCR de-NOx effectiveness as well as increase the boiler energy input by reheating the combustion air. For high power load, solar energy is used for saving steam bleeds from turbines by heating the feed water. A case study for a typical 1000 MW coal-fired power plant using the proposed concept has been performed and the results showed that, the SCR de-NOx efficiency of proposed SAPG could increase by 3.1% and 7.9% under medium load and low load conditions, respectively, as compared with the reference plant. The standard coal consumption rate of the proposed SAPG could decrease by 2.68 g/kWh, 4.05 g/kWh and 6.31 g/kWh for high, medium and low loads, respectively, with 0.040 USD/kWh of solar generated electricity cost. The proposed concept opens up a novel solar energy integration pattern in coal-fired power plants to improve the pollutant removal effectiveness and decrease the coal consumption of the power plant.

The expression of Adenovirus serotype 2 or serotype 5 (Ad2/5) E1A in tumor cells reduces their tumorigenicity in vivo by enhancing the NK cell mediated and T cell mediated anti-tumor immune response, an activity that correlates with the ability of E1A to bind p300. We determined if E1A could be used as a molecular adjuvant to enhance antigen-specific T cell responses to a model tumor antigen, ovalbumin (OVA). To achieve this goal, we stably expressed a fusion protein of E1A and OVA (MCA-205-E1A-OVA), OVA (MCA-205-OVA) or a mutant version of E1A unable to bind p300 and OVA (E1A-Δp300-OVA) in the B6-derived, highly tumorigenic MCA-205 tumor cell line. MCA-205-E1A-OVA tumor cells were over 10,000 fold less tumorigenic than MCA-205-OVA, MCA-205-E1A-Δp300-OVA, or MCA-205 in B6 mice. However, immunization of B6 mice with live MCA-205-OVA, MCA-205-E1A-Δp300-OVA and MCA-E1A-OVA tumor cells induced nearly equivalent OVA-specific CD4 T cells and CD8 CTL responses. Further studies revealed that mice with primary, enlarging MCA-205-OVA or MCA-205-E1A-Δp300-OVA tumors on one flank exhibited OVA-specific anti-tumor T cell responses that rejected a tumorigenic dose of MCA-205-OVA cells on the contralateral flank (concomitant tumor immunity). Next we found that tumor associated macrophages (TAMs) in progressive MCA-205-OVA tumors, but not MCA-205-E1A-OVA tumors that expressed high levels of arginase-1, which is known to have local immunosuppressive activities. In summary, immunization of mice with MCA-205 cells expressing OVA, E1A-Δp300-OVA or E1A-OVA induced equivalent OVA-specific CD4 and CD8 anti-tumor responses. TAMs found in MCA-205-OVA, but not MCA-205-E1A-OVA, tumors expressed high levels of arginase-1. We hypothesize that the production of arginase-1 by TAMs in MCA-205-OVA or MCA-205-E1A-Δp300-OVA tumor cells leads to an ineffective anti-tumor immune response in the tumor microenvironment, but does not result in inhibition of a systemic anti-tumor immunity.
