The Moon, as the closest celestial body to the Earth, plays a pivotal role in the progression of deep space exploration, and the establishment of research outposts on its surface represents a crucial step in this mission. Lunar lava tubes are special underground caves formed by volcanic eruptions and are considered as ideal natural shelters and scientific laboratories for lunar base construction. This paper begins with an in-depth overview of the geological origins, exploration history, and distribution locations of lunar lava tubes. Subsequently, it delves into the presentation of four distinctive advantages and typical concepts for constructing bases within lava tubes, summarizing the ground-based attempts made thus far in lunar lava tube base construction. Field studies conducted on a lava tube in Hainan revealed rock compositions similar to those found during the Apollo missions and clear lava tube structures, making it a promising analog site. Lastly, the challenges and opportunities encountered in the field of geotechnical engineering regarding the establishment of lunar lava tube bases are discussed, encompassing cave exploration technologies, in-situ testing methods, geomechanical properties under lunar extreme environments, base design and structural stability assessment, excavation and reinforcement techniques, and simulated Earth-based lava tube base.

Abstract The concept of the gut-kidney axis is gaining significant attention due to the close relationship between gut microbiota and kidney disease. Peritoneal dialysis is recognized as a crucial renal replacement therapy for end-stage renal disease (ESRD). The alterations in gut microbiota and related mechanisms after receiving this dialysis method are not fully understood. This study conducted shotgun metagenomic sequencing on fecal samples from 11 end-stage renal disease patients who did not receive dialysis (ESRD_N) and 7 patients who received peritoneal dialysis (ESRD_P). After quality control and correlation analysis of the data, our study is aimed at exploring the impact of peritoneal dialysis on the gut microbiota and health of ESRD patients. Our research findings indicate that the complexity and aggregation characteristics of gut microbiota interactions increase in ESRD_P. In addition, the gut microbiota drives the biosynthesis pathways of sesquiterpenes and triterpenes in ESRD_P patients, which may contribute to blood purification and improve circulation. Therefore, our research will lay the foundation for the prevention and treatment of ESRD.

Black soldier fly larvae (BSFL) treatment offers a promising avenue for manure valorization. However, there is a lack of larval density studies and ton-scale exploration in swine manure bioconversion. This study delves into the efficiency of larval fatty acid (FA) bioconversion, examining the impact of larval density on a kilogram scale and extending the analysis to a ton scale. Across a range of 50 to 600 larvae/kg, the larval FA content decreased from 15.3% to 7.85%. The peak larval FA yield, at 3.04% (based on manure dry matter), occurred at a density of 200 larvae/kg. Both low (50 larvae/kg) and high (600 larvae/kg) densities adversely affected BSFL bioconversion performance. Dominant larval FAs included C12:0 (39.7%), C16:1 (24.2%), C18:1 (17.5%), and C16:0 (8.3%). The scaling-up process maintained a consistent larval FA content and composition but resulted in decreased larvae FA yield due to increased larval mortality. Ultimately, each ton of swine manure yielded 12.4 kg of fresh larvae or 0.71 kg of larval FA components, corresponding to a 1.14% larval FA yield. This study underscores the feasibility of upscaling swine manure treatment using BSFL for FA bioconversion and emphasizes the necessity for large-scale studies to enhance larval survivorship and bioconversion efficiency.

In this paper, alkali protease was used to enzymatically hydrolyze wheat germ proteins grown in the Hetao region. A univariate analysis and an orthogonal experiment were conducted to optimize the enzymatically hydrolyze process conditions of wheat germ proteins, with the degree of hydrolysis taken as the indicator. Moreover, the in vitro hypolipidemic effect of wheat germ polypeptides was investigated. The results showed that the degree of hydrolysis of wheat germ proteins was 37.18%±0.25% achieved by the following conditions, substrate concentration of 6.0%, temperature at 50 ℃, pH of 10.5 and protease amount of 12000 U/g. When the concentration of wheat germ polypeptides was 5 mg/mL, the rate of inhibiting cholesterol solubilisation into micelles was 27.11%±0.36%, the pancreatic lipase activity inhibition rate was 33.54%±0.57%, and cholesterol esterase activity inhibition rate was 38.13%±0.29%. Moreover, after ultrafiltration, the relative molecule weight of polypeptides less than 1 kDa accounted for 54.30%±0.32% of all polypeptides, indicating that low-molecular-weight wheat germ polypeptides might yield superior hypolipidemic effects. The work provides a theoretical foundation for further exploring the mechanism between Hetao wheat germ polypeptides and hypolipidemic effect.

In 3D printing, especially direct ink writing (DIW), different materials are used to create complex 3D shapes by formulating inks with controlled rheological properties. Herein, a printable MXene ink is developed, exhibiting good rheological properties, to print different complex shapes, and potentially manufacture electronic devices such as sensors. Abstract Large language models (LLMs), such as ChatGPT, have demonstrated impressive capabilities in various tasks and attracted increasing interest as a natural language interface across many domains. Recently, large vision-language models (VLMs) that learn rich vision–language correlation from image–text pairs, like BLIP-2 and GPT-4, have been intensively investigated. However, despite these developments, the application of LLMs and VLMs in image quality assessment (IQA), particularly in medical imaging, remains unexplored. This is valuable for objective performance evaluation and potential supplement or even replacement of radiologists’ opinions. To this end, this study introduces IQAGPT, an innovative computed tomography (CT) IQA system that integrates image-quality captioning VLM with ChatGPT to generate quality scores and textual reports. First, a CT-IQA dataset comprising 1,000 CT slices with diverse quality levels is professionally annotated and compiled for training and evaluation. To better leverage the capabilities of LLMs, the annotated quality scores are converted into semantically rich text descriptions using a prompt template. Second, the image-quality captioning VLM is fine-tuned on the CT-IQA dataset to generate quality descriptions. The captioning model fuses image and text features through cross-modal attention. Third, based on the quality descriptions, users verbally request ChatGPT to rate image-quality scores or produce radiological quality reports. Results demonstrate the feasibility of assessing image quality using LLMs. The proposed IQAGPT outperformed GPT-4 and CLIP-IQA, as well as multitask classification and regression models that solely rely on images.

Abstract Background The time‐series data of the Normalized Difference Vegetation Index (NDVI) is a crucial indicator for global and regional vegetation monitoring. However, the current assessment of global and regional long‐term vegetation changes is subject to large uncertainties due to the lack of spatiotemporally continuous time‐series data sets. Methods In this study, a long time‐series monthly NDVI data set with a spatial resolution of 250 m from 1982 to 2020 was developed by combining Moderate Resolution Imaging Spectroradiometer (MODIS) and AVHRR (Advanced Very High‐Resolution Radiometer) time‐series NDVI products using the Random Forest (RF) downscaling model. Results Compared to the MODIS NDVI product, the fused product shows RMSE and mean absolute error ranging from 0 to 0.075 and from 0 to 0.05, respectively, with R2 values mostly above 0.7. Conclusions The long time‐series NDVI products generated in this study are reliable in terms of accuracy and have great potential for long‐term dynamic monitoring of terrestrial ecosystems on the Qinghai–Tibet Plateau.

Abstract Background Perioperative management and cardiac surgery in pregnant women with anti-phospholipid syndrome combined with heart valve disease have been rarely reported. Case presentation We describe a case of transcatheter mitral valve-in-valve replacement in a pregnant woman with bioprosthetic valve failure and anti-phospholipid syndrome at 18 weeks’ gestation. The patient underwent a cesarean section delivery at 34 weeks of gestation, resulting in the birth of a healthy baby. Conclusions Transapical mitral valve-in-valve surgery resulted in safe maternal and infant outcomes in a pregnant woman with anti-phospholipid syndrome combined with mitral bioprosthetic valve failure. The success of this procedure underscored the importance of multidisciplinary teamwork.

Abstract Background Anesthetic drugs may alter exosomal microRNA (miRNA) contents and mediate cancer progression and tumor microenvironment remodeling. Our study aims to explore how the anesthetics (sevoflurane and propofol) impact the miRNA makeup within exosomes in hepatocellular carcinoma (HCC), alongside the interconnected signaling pathways linked to the tumor immune microenvironment. Methods In this prospective study, we collected plasma exosomes from two groups of HCC patients (n = 5 each) treated with either propofol or sevoflurane, both before anesthesia and after hepatectomy. Exosomal miRNA profiles were assessed using next-generation sequencing (NGS). Furthermore, the expression data from The Cancer Genome Atlas-Liver Hepatocellular Carcinoma (TCGA-LIHC) was used to pinpoint the differentially expressed exosomal miRNAs (DEmiRNAs) attributed to the influence of propofol or sevoflurane in the context of HCC. Gene set enrichment analysis (GSEA) and gene set variation analysis (GSVA) were used to dissect the signaling pathways and biological activities associated with the identified DEmiRNAs and their corresponding target genes. Results A total of 35 distinct DEmiRNAs were exclusively regulated by either propofol (n = 9) or sevoflurane (n = 26). Through TCGA-LIHC database analysis, 8 DEmiRNAs were associated with HCC. These included propofol-triggered miR-452-5p and let-7c-5p, as well as sevoflurane-induced miR-24-1-5p, miR-122-5p, miR-200a-3p, miR-4686, miR-214-3p, and miR-511-5p. Bovine respiratory disease complex (BRDC) represents a global acute respiratory condition that imposes substantial economic burdens on the cattle industry due to its high morbidity and mortality rates. Various factors contribute to the development of BRDC, including pathogen infections, environmental stresses, weaning of calves, and herd relocation. Viral pathogens, notably bovine respiratory syncytial virus (BRSV) and bovine viral diarrhea virus (BVDV), play a critical role in the etiology of BRDC, with single or combined viral infections being particularly clinically significant. In this study, we developed a duplex TaqMan-based real-time RT-PCR assay targeting the conserved regions of the F gene of BRSV and the 5′ UTR sequence of BVDV. The limits of detection for BRSV and BVDV were 6.83 copies/μL and 5.24 copies/μL, respectively. Our validation data suggest the assay has excellent sensitivity, specificity and reproducibility. Testing of clinical samples revealed prevalence of BRSV and BVDV in local farms in Jiangsu Province, China. This study provides an efficient diagnostic tool for the epidemiological investigation of BRDC.

It is indisputable that microplastics (MPs) can profoundly alter nitrogen transformation in soil. However, it remains poorly understood how MPs impact soil nitrogen processes. This review systematically analyzed literature published in recent years related to the impact of MPs on nitrogen transformation. After reviewing the environmental behavior of MPs in soil media, the mechanisms of action and key factors of MPs’ effects on soil nitrogen transformation are elucidated. The size, shape, concentration, and type of MPs significantly alter nitrogen transformation. When MPs enter the soil, they can significantly affect the habitat and diversity of soil microorganisms and the transformation of soil nitrogen by adsorbing pollutants, releasing additives, and altering the physicochemical characteristics of the soil. As organic substrates, MPs can directly affect microbial community structure by promoting microbial colonization. Besides, MPs can also be toxic to soil microorganisms by coming into direct contact with cell surfaces. Microorganisms, key enzymes, and functional genes associated with nitrogen transformation respond to the presence of MPs, thereby affecting the nitrogen conversion process. At the last, measures to mitigate soil MPs contamination are suggested. The article highlights the effects of MPs on soil nitrogen transformation factors, leading to valuable insights into microbially-mediated nitrogen transformation processes in MP-contaminated soils. It offers useful information for determining nitrogen regulation and assessing ecological risks in soils contaminated by MPs.

In the context of a warming climate, glacial debris flows will inevitably increase under the coupled effect of increased temperature and precipitation. However, the extent of future losses is unclear due to the complexity of modelling glacial debris flow processes. This study integrated the degree-day model, the TRIGRS model, and the LS-RAPID model to assess increasing risk in the future period (2071–2100) under the SSP2-4.5 scenario compared to the present. A fatal glacial debris flow in the Tianmo Gully that occurred on September 4, 2007 (hereafter referred to as “the event”) was taken as an example. By the 2090s, compared to the event, the contribution of temperature to the occurrence of glacial debris flows was projected to increase significantly, from 77.05 % to 89.68 % under the SSP2-4.5 scenario. Combining meltwater and precipitation, the study reproduced the propagation of the event and simulated the future extreme debris flow event. Although the section of National Highway 318 (G318) below Tianmo Gully was relocated and reconstructed 150 m away from the historic sedimentary fan, a section of 88.46 m remained at risk of inundation in the future. The maximum height of the debris flow dam in the Parlung Zangbo River could reach up to 34.50 m, doubling the height of the event. Debris removal from the road is estimated to take 1 day, resulting in a projected loss of tourism revenue amounting to RMB 17.82 million. Without relocation, the clearing time would take up to 3 days. The economic benefit of rerouting the section was projected at RMB 13.81 million. The mechanisms of glacial debris flow in Parlung region are similar. Both the concept of model integration and the comparison of the simulation results are of great importance for risk assessments under global warming.

Objective: This study aimed to evaluate the efficacy of acupuncture in treating allergic rhinitis (AR) in animal models of the disease, and to explore the underlying mechanism of acupuncture in AR. Methods: Related literature was retrieved from multiple databases, including China National Knowledge Infrastructure (CNKI), Wan Fang databases, SinoMed databases, VIP database, PubMed, EMBASE, the Cochrane Library, up to September 2023. The inclusion criteria were focused on animal experiments that investigated the effect of acupuncture therapy on animal models of AR. Studies combining acupuncture with other Chinese medicine therapies were excluded. Data were extracted independently by two reviewers using standardized forms. A total of 75 studies were finally included. The risk of bias in individual studies was evaluated using the Systematic Review Centre for Laboratory animal Experimentation (SYRCLE) tool, and the quality of reporting was evaluated according to Animal Research: Reporting of In Vivo Experiments (ARRIVE) guidelines. Meta-analyses and plotting were conducted using STATA 17.0 and RevMan 5.4. When heterogeneity was present, a random effects model was applied. Subgroup, meta-regression and sensitivity analyses were also performed. Results: The SYRCLE scores ranged from 3 to 7, and the ARRIVE scores ranged from 6.5 to 11 points. Meta-analysis results demonstrated that acupuncture could significantly down-regulate EOS counts in both blood and nasal mucosa, and reduce the serum levels of IL-5, compared to the AR model group. The results of the qualitative analysis showed that acupuncture could reduce the behavior scores of AR, down-regulate serum levels of IL-4, IgE and sIgE, as well as up-regulate IFN-γ levels. Subgroup analysis results suggested that the different interventions might contribute to the observed heterogeneity. Conclusion: Our results demonstrate that acupuncture effectively alleviates the nasal allergic symptoms in animal models, inhibits immune and inflammatory signaling transduction, and reduces the release of inflammatory mediators. This study highlights the potential of acupuncture as a promising therapeutic option for AR, however, further studies are required to fully understand its mechanisms of action.

ObjectiveStudies have shown the adverse psychological impact of polycystic ovary syndrome (PCOS), but the state of mental health in adolescents with PCOS remains unclear. Thus, we performed a systematic review and meta-analysis to investigate the prevalence and severity of depression and anxiety, as well as potential effects on self-esteem and quality of life (QoL) in this specific population.MethodsWe systematically searched four electronic databases: PubMed, Embase, Web of Science, and the Cochrane Reviews database for articles published until 25/8/2024. We considered observational studies in which the subjects were adolescent girls with PCOS who had reported symptoms including anxiety, depression, self-esteem, and QoL. The Review Manager version 5.4 was used to analyze the available data extracted. We used the Newcastle-Ottawa Quality Assessment Scale (NOS) to evaluate the quality of selected studies. A funnel plot was utilized to assess the risk of literature bias, and a forest plot was used to represent the combined outcomes. This systematic review was previously registered in PROSPERO with the registration number CRD42022382036.ResultsWe included 11 studies in the systematic review and conducted meta-analyses on 10 of them. Adolescents with PCOS reported a higher risk of depression (OR = 2.21, 95% CI: 1.23 to 4.00, p = 0.008) and a higher level of depression scores (SMD = 0.43, 95% CI: 0.16 to 0.71, p = 0.002) than controls. There were no significant differences in anxiety (OR = 1.90, 95% CI: 0.52 to 6.96, p = 0.33; SMD = 0.19, 95% CI: -0.21 to 0.59, p = 0.36), self-esteem (SMD = -0.17, 95% CI: -0.85 to 0.52, p = 0.64), and QoL (SMD = -0.15, 95% CI: -0.42 to 0.11, p = 0.26) between the two groups.ConclusionsOur research indicated that adolescents with PCOS experienced more severe depressive symptoms than those without PCOS. This highlights the importance of evaluation and early treatment of mental health in PCOS. Ethnopharmacological relevance: Yiai Fuzheng formula (YAFZF), as a Traditional Chinese Medicine (TCM) prescription, has been used widely at Zhongnan Hospital of Wuhan University for its therapeutic effects and high safety on triple-negative breast cancer (TNBC). Objective: In this study, we employed ultra-high-performance liquid chromatography-quadrupole/orbitrap high-resolution mass spectrometry (UPLC-Q-Orbitrap-HRMS), network pharmacology, and experimental validation to elucidate the underlying action mechanism of YAFZF in the treatment of TNBC. Methods: The key active ingredients in YAFZF were analyzed using UPLC-Q-Orbitrap-HRMS, and then the potential components, target genes and signalling pathways of YAFZF were predicted using the network pharmacological method. We then used molecular docking to visualize the combination characteristics between major active components and macromolecules in the crucial pathway. In vitro experiments were conducted to investigate the inhibitory effects of YAFZF treatment on the cell viability, invasion, and migration of 4T1 and MDA-MB-231 cells. The xenograft TNBC models were constructed using female Balb/c mice, and their body weights, tumour volumes, and weights were monitored during YAFZF treatment. Quantitative real-time PCR (qRT-PCR), Hematoxylin-eosin (HE), immunohistochemistry (IHC) staining, Western blot (WB), and terminal deoxynucleotidyl transferase (TdT)-dUTP nick-end labeling (TUNEL) staining were used for further experimental validation. Results: Based on UPLC-Q-Orbitrap-HRMS and network pharmacology analysis, 6 major bioactive components and 153 intersecting genes were obtained for YAFZF against TNBC. Functional enrichment analysis identified that the phosphatidylinositol 3-kinase (PI3K)-protein kinase B (Akt) signalling pathway might be the mechanism of action of YAFZF in the treatment of TNBC. Molecular docking results suggested that the main active compounds in YAFZF had strong binding energies with the proteins in the PI3K/Akt pathway. In vitro experiments showed that YAFZF inhibited the cell viability, invasion, and migration abilities of TNBC cells. Animal experiments confirmed that YAFZF treatment suppressed tumour cell proliferation and increased apoptotic cells. PCR, HE, WB, and IHC results indicated that YAFZF could suppress xenograft tumour metastases by inhibiting the PI3K/AKT/mTOR pathway regulating the epithelial-mesenchymal transition (EMT) process. Conclusion: YAFZF therapy showed its potential for reducing proliferation, invasion, and migration abilities, increasing apoptosis of TNBC cells. Furthermore, YAFZF treated TNBC by inhibiting xenograft tumour distant metastases via the regulation of EMT by the PI3K/Akt/mTOR pathway, suggesting that it may be useful as an adjuvant treatment.

ABSTRACT: This study aimed to investigate how various selenium sources affect the intestinal health of broiler chickens. A total of 384, one-day-old Arbor Acres broilers were weighed and randomly allocated to four treatment groups. The control diet was a basal diet added with: 0.2 mg/kg Sodium Selenite (SS-control), 0.2 mg/kg Selenium nano-particles (Nano-Se), 0.2 mg/kg Selenomethionine (SeMet), and 0.2 mg/kg Selenocysteine (Sec) as the treatments. The results indicated that Nano-Se and SeMet were effective in enhancing the villus height (VH) and the villus height/crypt depth ratio (VH/CD) in the jejunum compared to (SS) (P < 0.05). The inclusion of Nano-Se into the diets increased the mRNA levels of zonula occluden-1 (ZO-1), ZO-2, Occludin, Claudin-1, and Claudin-3 compared to the SS diet (P < 0.05). The SeMet increased the levels of ZO-1 and Claudin-3 compared to the SS (P < 0.05). Moreover, SeMet upregulated the marker genes of intestinal enteroendocrine cells, stem cells, and epithelial cells compared to the SS diet (P < 0.05). However, supplementation of Nano-Se reduced the mRNA levels of interleukin 1β (IL-1β), and IL-8 and the concentration of reactive oxygen species (ROS) in the jejunum compared to the SS (P < 0.05). The Nano-Se and SeMet also increased the protein levels of CAT and SOD compared to the SS and Sec diet (P < 0.05). The number of the goblet cells and Mucin-2 (Muc2) levels were the highest in the Nano-Se group (P < 0.05). The protein expression levels of goblet cell differentiation regulator (v-myc avian myelocytomatosis viral oncogene homolog, c-Myc) were highest in the Nano-Se compared to the SS diet (P < 0.05). The Nano-Se decreased the mRNA and protein levels of NLRP3 signaling pathway-related genes compared to the SS diet (P < 0.05). In conclusion, our study demonstrated that Nano-Se and SeMet are better at improving the intestinal health of 21-day-old broilers. Additionally, Nano-Se demonstrated superior antioxidant and anti-inflammatory effects, promoting the development of intestinal goblet cells by modifying the NLRP3 signaling pathway.

Abstract To construct a prediction model of olfactory dysfunction after transnasal sellar pituitary tumor resection based on machine learning algorithms. A cross-sectional study was conducted. From January to December 2022, 158 patients underwent transnasal sellar pituitary tumor resection in three tertiary hospitals in Sichuan Province were selected as the research objects. The olfactory status was evaluated one week after surgery. They were randomly divided into a training set and a test set according to the ratio of 8:2. The training set was used to construct the prediction model, and the test set was used to evaluate the effect of the model. Based on different machine learning algorithms, BP neural network, logistic regression, decision tree, support vector machine, random forest, LightGBM, XGBoost, and AdaBoost were established to construct olfactory dysfunction risk prediction models. The accuracy, precision, recall, F1 score, and area under the ROC curve (AUC) were used to evaluate the model's prediction performance, the optimal prediction model algorithm was selected, and the model was verified in the test set of patients. Of the 158 patients, 116 (73.42%) had postoperative olfactory dysfunction. After missing value processing and feature screening, an essential order of influencing factors of olfactory dysfunction was obtained. Among them, the duration of operation, gender, type of pituitary tumor, pituitary tumor apoplexy, nasal adhesion, age, cerebrospinal fluid leakage, blood scar formation, and smoking history became the risk factors of olfactory dysfunction, which were the key indicators of the construction of the model. Among them, the random forest model had the highest AUC of 0.846, and the accuracy, precision, recall, and F1 score were 0.750, 0.870, 0.947, and 0.833, respectively. Compared with the BP neural network, logistic regression, decision tree, support vector machine, LightGBM, XGBoost, and AdaBoost, the random forest model has more advantages in predicting olfactory dysfunction in patients after transnasal sellar pituitary tumor resection, which is helpful for early identification and intervention of high-risk clinical population, and has good clinical application prospects.

Abstract Luminescent materials with narrowband emission show great potential for diverse applications in optoelectronics. Purely organic phosphors with room-temperature phosphorescence (RTP) have made significant success in rationally manipulating quantum efficiency, lifetimes, and colour gamut in the past years, but there is limited attention on the purity of the RTP colours. Herein we report a series of closed-loop molecules with narrowband phosphorescence by multiple resonance effect, which significantly improves the colour purity of RTP. Phosphors show narrowband phosphorescence with full width at half maxima (FWHM) of 30 nm after doping into a rigid benzophenone matrix under ambient conditions, of which the RTP efficiency reaches 51.8%. At 77 K, the FWHM of phosphorescence is only 11 nm. Meanwhile, the colour of narrowband RTP can be tuned from sky blue to green with the modification of methyl groups. Additionally, the potential applications in X-ray imaging and display are demonstrated. This work not only outlines a design principle for developing narrowband RTP materials but also makes a major step forward extending the potential applications of narrowband luminescent materials in optoelectronics.

Background:Cordyceps sinensis is a traditional Chinese medicine that has shown promise for the management of chronic bronchitis (CB). We aim to assess the efficacy and safety of a preparation of C sinensis named Bailing capsule (Hirsutella sinensis, Cs-C-Q80) compared with a placebo in patients with CB.Methods: This randomized, double-blind, placebo-controlled, parallel-group clinical trial (Chinese Clinical Trial Registry; registration number: ChiCTR1900025707) recruited patients with CB from eight hospitals in China between May 2019 and December 2020. Patients were randomized 2:1 to receive Bailing capsule or a placebo orally for 48 weeks (2.0 g, three times a day).Results: Among 240 patients who were randomized, 238 (Bailing capsule: 159, placebo: 79) were included in the primary analysis. Bailing capsule significantly reduced the frequency of acute exacerbation of CB (AECB) compared with the placebo during treatment (0.43 ± 0.82 vs. 1.56 ± 1.34; P < 0.001) and follow-up (0.21 ± 0.64 vs. 0.45 ± 0.93; P = 0.026). Bailing capsule improved the severity of expectoration (P = 0.046) and wheezing (P = 0.010) in AECB during follow-up. The severity of CB after treatment was significantly improved in the Bailing capsule group compared with the placebo group (P = 0.035), particularly in terms of expectoration (P = 0.012) and wheezing (P = 0.003). The risk of adverse events, mainly including infectious and invasive diseases and gastrointestinal symptoms, did not significantly differ between the two groups (29.6% vs. 30.4%).Conclusion: In patients with CB, Bailing capsule significantly reduces the frequency of AECB and ameliorates the severity of AECB and CB symptoms.Clinical Trail Registration:https://www.chictr.org.cn, identifer ChiCTR1900025707.

BackgroundThis study aims to prioritize genes potentially involved in multifactorial or causal relationships with gout.MethodsUsing the Summary Data-based Mendelian Randomization (SMR) approach, this research analyzed expression quantitative trait loci (eQTL) data from blood and renal tissues and genome-wide association study (GWAS) data related to gout. It sought to identify genetic loci potentially involved in gout. Heterogeneity testing was conducted with the HEIDI test, and results were adjusted for the False Discovery Rate (FDR). Blood cis-eQTL data were sourced from the eQTLGen Consortium’s summary-level data, and renal tissue data came from the V8 release of the GTEx eQTL summary data. Gout GWAS data was sourced from the FinnGen Documentation of the R10 release.ResultSMR analysis identified 14 gene probes in the eQTLGen blood summary-level data significantly associated with gout. The top five ranked genes are: ENSG00000169231 (labeled THBS3, PSMR = 4.16 × 10−13), ENSG00000231064 (labeled THBS3-AS1, PSMR = 1.88 × 10−8), ENSG00000163463 (labeled KRTCAP2, PSMR = 3.88 × 10−6), ENSG00000172977 (labeled KAT5, PSMR = 1.70 × 10−5), and ENSG00000161395 (labeled PGAP3, PSMR = 3.24 × 10−5). Notably, increased expression of KRTCAP2 and PGAP3 is associated with an increased risk of gout, whereas increased expression of THBS3, THBS3-AS1, and KAT5 is associated with a reduced gout risk. No significant gene associations with gout were observed in renal tissue, likely due to the limited sample size of kidney tissue.ConclusionOur findings have highlighted several genes potentially involved in the pathogenesis of gout. These results offer valuable insights into the mechanisms of gout and identify potential therapeutic targets for its treatment.

The restoration of ecological functions in abandoned Moso bamboo forests facilitates positive succession in evergreen vegetation. Nevertheless, there is a limited understanding regarding the leaf functional traits and nutrient utilization strategies of understory vegetation after abandonment. This study investigated the impact of abandonment duration (9 and 21 years) on leaf morphological traits, soluble sugars characteristics, carbon (C), nitrogen (N), phosphorus (P), and their ratios in dominant tree seedlings (Lithocarpus brevicaudatus and Schima superba) within Moso bamboo forests. Leaf sizes increased significantly with prolonged abandonment in both species, accompanied by a consistent decrease in leaf thickness and leaf tissue density. These morphological indices were more pronounced in L. brevicaudatus compared to S. superba (P < 0.05), and they showed a positive correlation with tree sizes. Moreover, leaf soluble sugars, C, and C: N: P ratios of both tree species decreased over duration abandonment, while N and P contents increased. Specifically, L. brevicaudatus exhibited elevated levels of fructose, glucose, N, and P, whereas S. superba displayed higher sucrose, C, and C: N: P ratios. Our findings reveal that both species, with extended abandonment, enlarged their leaf area and improved carbon utilization, facilitating adaptation to competitive resource habitats. Particularly, L. brevicaudatus exhibited a propensity to augment leaf sizes, as well as N, and P contents, suggesting a strategy to enhance carbon assimilation through additional N and P nutrient supplementation. Conversely, S. superba primarily focused on bolstering carbohydrate reserves, indicating that moderate thinning or pruning of bamboo forests could boost sunlight penetration, along with the appropriate nutrient supplementation. These insights significantly contribute to the scientific understanding necessary for the classification and management of mixed stands in abandoned bamboo forests.

Objectives To quantify the economic investment required to increase bariatric surgery (BaS) capacity in National Health Service (NHS) England considering the growing obesity prevalence and low provision of BaS in England despite its high clinical effectiveness.Design Data were included for the patients with obesity who were eligible for BaS. We used a decision-tree approach including four distinct steps of the patient pathway to capture all associated resource use. We estimated total costs according to the current capacity (current scenario) and three BaS scaling up strategies over a time horizon of 20 years (projected scenario): maximising NHS capacity (strategy 1), maximising NHS and private sector capacity (strategy 2) and adding infrastructure to NHS capacity to cover the entire prevalent and incident obesity populations (strategy 3).Setting BaS centres based in NHS and private sector hospitals in England.Main outcome measures Number of BaS procedures (including revision surgery), cost (GBP) and resource utilisation over 20 years.Results At current capacity, the number of BaS procedures and the total cost over 20 years were estimated to be 140 220 and £1.4 billion, respectively. For strategy 1, these values were projected to increase to 157 760 and £1.7 billion, respectively. For strategy 2, the values were projected to increase to 232 760 and £2.5 billion, respectively. Strategy 3 showed the highest increase to 564 784 and £6.4 billion, respectively, with an additional 4081 personnel and 49 facilities required over 20 years.Conclusions The expansion of BaS capacity in England beyond a small proportion of the eligible population will likely be challenging given the significant upfront economic investment and additional requirement of personnel and infrastructure.

Plant height (PH) is a crucial trait for strengthening lodging resistance and boosting yield in foxtail millet. To identify quantitative trait loci (QTL) and candidate genes associated with PH, we first developed a genetic map using a recombinant inbred line (RIL) population derived from a cross between Aininghuang and Jingu 21. Then, PH phenotyping data and four variations of best linear unbiased prediction (BLUP) were collected from nine environments and three development stages. Next, QTL mapping was conducted using both unconditional and conditional QTL methods. Subsequently, candidate genes were predicted via transcriptome analysis of parental samples at three developmental stages. The results revealed that the genetic map, based on re-sequencing, consisted of 4,360 bin markers spanning 1,016.06 cM with an average genetic distance of 0.23 cM. A total of 19 unconditional QTL, accounting for 5.23%–35.36% of the phenotypic variation explained (PVE), which included 7 major and 4 stable QTL, were identified. Meanwhile, 13 conditional QTL, explaining 5.88%–40.35% of PVE, including 5 major and 3 stable QTL, were discovered. Furthermore, four consistent and stable QTL were identified. Finally, eight candidate genes were predicted through RNA-seq and weighted gene co-expression network analysis (WGCNA). Those findings provide a crucial foundation for understanding the genetic mechanisms underlying PH development and facilitate molecular marker-assisted breeding of ideal plant types in foxtail millet.

BackgroundWhile some studies have suggested a link between gut microbiota (GM) and abortion, the causal relationship remains unclear.MethodsTo explore the causal relationship between GM and abortion, including spontaneous abortion (SA) and habitual abortion (HA), we performed a two-sample Mendelian randomization (MR) analysis. We used summary statistics data from MiBioGen and FinnGen for genome-wide association studies (GWAS), with GM data as the exposure variable and abortion data as the outcome variable.ResultsIn the absence of heterogeneity and horizontal pleiotropy, the inverse-variance weighted (IVW) method identified five genetically predicted GM genera linked to the risk of abortions. Lactococcus was negatively correlated with the risk of SA, whereas the Eubacterium fissicatena group was positively correlated with the risk of SA. Genetic predictions of Coprococcus3 and Odoribacter were linked to a reduced risk of HA, while the Eubacterium ruminantium group was associated with an increased risk of HA.ConclusionOur study suggests a genetic causal relationship between specific GM and two types of abortions, improving our understanding of the pathological relationship between GM and abortion.

The purpose of this study was to investigate the regulatory mechanism of Bacillus tequilensis (GCB-3) in improving the growth performance of grass carp by microbiome and targeted metabolomics analysis. Fish were fed with different amounts of GCB-3 including B0 (0 CFU/g), B6 (1×106 CFU/g), B7 (1×107 CFU/g), B8 (1×108 CFU/g), and B9 (1×109 CFU/g) for 8 weeks. The results indicate that with the increase of dietary GCB-3 concentration, grass carp showed a significant linear trend in growth performance, and appetite factors showed a significant linear and quadratic trend, the growth performance and appetite-promoting factors in the B8 group reach their maximum value in the B8 group. Microbiome analysis found that Proteobacteria and Actinobacteria were the most abundant phylum in the gut microbiota of grass carp. The functional potential predicted that GCB-3 affected the synthesis and degradation of carbohydrates, amino acids, fatty acids and lipids. It found that GCB-3 also affected the tricarboxylic acid (TCA) cycle VII (acetate-producers) pathway. The results of targeted metabolomics showed a significant linear and quadratic increase in intestinal acetate with GCB-3 concentration and a maximum in the B8 group. The results of the correlation analysis showed a significant positive correlation between Actinobacteria abundance and acetic acid content, and also between TCA cycle VII (acetate-producers) pathway abundance and Actinobacteria abundance and acetic acid content. In B8 group, grass carp showed a faster regulation speed of blood glucose levels after glucose injection, and the GLP-1 content remained stable. The activities of glycolytic enzymes, lipolytic enzymes, protein metabolism enzymes, and their related mRNA expression levels in grass carp showed a linear or quadratic increasing trend. However, the expression levels of gluconeogenic enzymes and fatty acid synthesis enzyme related mRNA showed an opposite trend. In summary, the addition of 1×108 CFU/g GCB-3 in grass carp feed can improve the abundance of dominant bacterial phyla in the intestine, especially Actinobacteria, increase the concentration of intestinal acetate, provide substrate for the TCA cycle VII (acetate-producers) pathway, thereby regulating the nutrient metabolism of grass carp and ultimately promoting growth.

BackgroundImatinib is the most widely used tyrosine kinase inhibitor (TKI) in patients with newly diagnosed chronic-phase chronic myeloid leukemia(CML-CP). However, failure to achieve optimal response after imatinib administration, and subsequent switch to second-generation TKI therapy results in poor efficacy and induces drug resistance. In the present study, we developed and validated a nomogram to predict the efficacy of imatinib in the treatment of patients newly diagnosed with CML-CP in order to help clinicians truly select patients who need 2nd generation TKI during initial therapy and to supplement the risk score system.MethodsWe retrospectively analyzed 156 patients newly diagnosed with CML-CP who met the inclusion criteria and were treated with imatinib at the Second Affiliated Hospital of Xi’an Jiao Tong University from January 2012 to June 2022. The patients were divided into a poor-response cohort (N = 60)and an optimal-response cohort (N = 43) based on whether they achieved major molecular remission (MMR) after 12 months of imatinib treatment. Using univariate and multivariate logistic regression analyses, we developed a chronic myeloid leukemia imatinib-poor treatment (CML-IMP) prognostic model using a nomogram considering characteristics like age, sex, HBG, splenic size, and ALP. The CML-IMP model was internally validated and compared with Sokal, Euro, EUTOS, and ELTS scores.ResultsThe area under the curve of the receiver operator characteristic curve (AUC)of 0.851 (95% CI 0.778–0.925) indicated satisfactory discriminatory ability of the nomogram. The calibration plot shows good consistency between the predicted and actual observations. The net reclassification index (NRI), continuous NRI value, and the integrated discrimination improvement (IDI) showed that the nomogram exhibited superior predictive performance compared to the Sokal, EUTOS, Euro, and ELTS scores (P < 0.05). In addition, the clinical decision curve analysis (DCA) showed that the nomogram was useful for clinical decision-making. BackgroundMeaning in life is a crucial aspect of psychological well-being, often overlooked despite its clinical significance. This warrants further investigation, especially regarding its relationship with frailty and psychological resilience.ObjectiveThis study aims to assess the status and relevance of frailty, psychological resilience, and meaning in life among older adults in Chinese nursing homes. Additionally, it explores the mediating role of psychological resilience between frailty and meaning in life, providing insights to improve the meaning in life for older adults in nursing homes.MethodsBetween August 2022 and November 2022, 302 older adults in Chinese nursing homes were selected using convenience sampling. The study utilized the Socio-demographic Characteristics Questionnaire, Tilburg Frailty Indicator, Connor-Davidson Resilience Scale, and the Source of Meaning Scale for Older Adults. A face-to-face questionnaire survey was conducted, and SPSS 27.0 was employed for analyzing correlations between frailty, psychological resilience, and meaning in life. The mediating effect of psychological resilience was assessed using Model 4 in the Process plug-in.ResultsOlder adults in nursing homes exhibited a frailty total score of 4.00 (2.00, 5.00), with a prevalence of 28.5%. Psychological resilience scored 66.00 (51.75, 76.00), and meaning in life scored 149.00 (132.00, 158.25). Frailty showed a negative correlation with both meaning in life and psychological resilience, while meaning in life demonstrated a positive correlation with psychological resilience. Psychological resilience exhibited a partial mediating effect, accounting for 51.04% of the total effect between frailty and meaning in life.ConclusionFrailty incidence is high among older adults in nursing homes, with psychological resilience at a general level and meaning in life in the upper middle level. Psychological resilience plays a crucial role as a partial mediator between frailty and meaning in life. Timely assessment of frailty, targeted interventions, and improvements in psychological resilience are essential for enhancing the meaning in life and promoting successful aging.

Abstract Neuron localization is a fundamental step in the neuron morphology reconstruction and quantitative analysis of cell counting and spatial distribution. The recent development of labelling and imaging techniques has resulted in growing demand for automatic neuron localization methods of 3D neuronal microscopy images. However, accurately localizing touching neurons remains a challenge. To address this issue, a novel method utilizing a super‐resolution segmentation network based on 2D Conv‐LSTM and a region growing method are proposed. This network can map and detect individual neurons in higher resolution space, allowing for the separation of closely touching neurons with reduced resource consumption. Subsequently, a region growing method is employed to localize neurons accurately. This method is evaluated using neuronal images generated by TDI‐fMOST. This method achieved neuron localization with an F1 score of 0.91. In comparison, other automatic localization methods achieved F1 scores lower than 0.85. It is also demonstrated that our network has fewer computational requirements compared to 3D neural networks. This method is promising for accurately localizing neurons in large‐scale neuron images.

Abstract Monitoring the position of orthopedic implants in vivo is paramount for enhancing postoperative rehabilitation. Traditional radiographic methods, although effective, pose inconveniences to patients in terms of specialized equipment requirements and delays in rehabilitation adjustment. Here, a nonradiographic design concept for real‐time and precisely monitoring the position of in vivo orthopedic implants is presented. The monitoring system encompasses an external magnetic field, a three‐dimensional (3D)‐printed superparamagnetic intervertebral body fusion cage (SIBFC), and a magnetometer. The SIBFC with a polyetheretherketone framework and a superparamagnetic Fe3O4 component was integrally fabricated by the high‐temperature selective laser sintering technology. Owing to the superparamagnetic component, the minor migration of SIBFC within the spine would cause the distribution change of the magnetic induction intensities, which can be monitored in real‐time by the magnetometer no matter in the static states or dynamic bending motions. Besides horizontal migration, occurrences of intervertebral subsidence in the vertical plane of the vertebrae can also be effectively distinguished based on the obtained characteristic variations of magnetic induction intensities. This strategy exemplifies the potential of superparamagnetic Fe3O4 particles in equipping 3D‐printed orthopedic implants with wireless monitoring capabilities, holding promise for aiding patients' rehabilitation.

This study presents a snow drought assessment method using snow water equivalent products to examine the patterns and differences in snow drought events in China from 1980 to 2020. The findings indicate that snow drought changes over the past 40 years can be categorized into three stages: The most severe snow drought occurred in the 1980s, followed by alleviation until 2009, and a subsequent aggravation after 2010. Light snow drought has the widest distribution and shows an increasing trend, whereas medium, heavy, and extreme snow droughts decrease gradually but show a decrease followed by an increasing trend over time. The distribution of snow drought in China displays significant spatial heterogeneity, with areas such as Alxa League in Inner Mongolia, Hami, Turpan, Bayingol, Xinjiang, and the Tibetan Plateau hinterland experiencing frequent snow drought events. Moreover, the low-altitude region has the largest average annual proportion of extreme drought, whereas the high-altitude region has the highest proportion of heavy drought. Among the three snow-dominant areas, Northern Xinjiang has the largest proportion of snow drought areas. The Northeast-Inner Mongolia has the highest proportion of extreme drought while the Tibetan Plateau exhibited abrupt changes in snow drought occurrence. These results contribute to a better understanding of the underlying mechanisms of snow drought changes, as well as serve as a foundation for the protection of the ecological environment.

Polar mesospheric clouds (PMCs) are ice crystal clouds formed in the mesosphere of high-latitude regions in both the northern (NH) and southern hemispheres (SH). Peak height is an important physical characteristic of PMCs. Satellite observation data from solar occultation for ice experiments (SOFIE) during seven PMC seasons from 2007 to 2014 show that the difference between the height of the mesopause and the peak height of the PMCs (Zmes-Zmax) were inversely correlated with the atmospheric mesopause temperature. The Zmes-Zmax averages for all seasons for the NH and SH were 3.54 km and 2.66 km, respectively. They were smaller at the starting and ending stages of each PMC season and larger in the middle stages. Analysis of the individual cases and statistical results simulated by the PMCs 0-D model also revealed the inverse correlations between the Zmes-Zmax and mesopause temperature, with correlation coefficients of −0.71 and −0.62 for the NH and SH, respectively. The corresponding rates of change of Zmes-Zmax with respect to mesopause temperature were found to be −0.21 km/K and −0.14 km/K, respectively. The formation mechanism of PMCs suggests that a lower temperature around the mesopause can lead to a greater distance and longer time for ice crystals to condense and grow in clouds. Thus, ice crystals sediment to a lower height, making the peak height of the PMCs further away from the mesopause. In addition, disturbances in small-scale dynamic processes tend to weaken the impact of temperature on the peak height of PMCs.

Spacecraft recovery technology is crucial in the field of aerospace, in which the parachute plays a key role in slowing down the descent speed of the spacecraft and realizing a smooth landing. In order to construct a dynamically adjustable parachute deployment strategy, it is necessary to measure the parachute dynamic load accurately in real-time. However, the existing sensor measurement scheme makes it difficult to meet the measurement requirements due to its large structure and complex wiring. In order to meet the current demand for real-time measurement of parachute cords dynamic load, a miniature measuring instrument is designed. According to the function and technical requirements of the miniature measuring instrument, the hardware modules of the acquisition system are selected and designed, and the integration debugging and performance optimization of the microcontroller module, A/D sampling module, signal acquisition circuit, and power supply module are carried out. The software of the parachute cords tension acquisition system based on the miniature measuring instrument is developed. The Load Cell is modeled by using SolidWorks 2022 and statically analyzed by using Ansys 2022 R1 Workbench finite element analysis software. Then the final structure of the Load Cell and the pasting position of the strain gauge are determined through the results analysis as well as experimental verification. The hardware module of the signal acquisition system for the miniature measuring instrument is then encapsulated. The force value of the miniature measuring instrument is calibrated and tested many times by using the microcomputer-controlled electronic universal testing machine. The experimental results show that the designed miniature measuring instrument has accurate data, strong stability, and good real-time performance, which meets the demand for real-time accurate measurement of miniature measuring instruments, and can provide reliable data for parachute cords parameter validation and stepless unfolding design.

Background: Recent studies have established associations between the gut microbiota (GM) and thyroid diseases (TDs). However, their causal relationships remain elusive. Methods: To investigate this causality, we conducted a two-sample Mendelian randomization (MR) analysis using genome-wide association study (GWAS) data from MiBioGen and FinnGen, with GM as the exposure and six TDs as outcomes. Results: We identified 32 microbial taxa linked to the risk of six TDs. The Clostridium innocuum group, Ruminiclostridium5, and Lachnoclostridium exhibited protective effects against nontoxic diffuse goiter (NDG). Conversely, an increased risk of NDG was associated with Ruminococcaceae UCG002, Alistipes, Methanobrevibacter, Marvinbryantia, and Ruminococcaceae UCG014. Bifidobacterium and Sutterella were protective against nontoxic multinodular goiter (NMG), while the Ruminococcus gauvreauii group and Rikenellaceae RC9 gut group heightened NMG risk. Protective effects against nontoxic single thyroid nodule (NSTN) were observed with Defluviitaleaceae UCG011, Ruminococcus1, and Ruminococcaceae UCG010, whereas increased risk was linked to Alistipes, the Ruminococcus gauvreauii group, and Lachnospiraceae UCG010. Ruminiclostridium9, Victivallis, and Butyricimonas offered protection against thyrotoxicosis with Graves’ Disease (GD), while the Eubacterium rectale group, Desulfovibrio, Bifidobacterium, Collinsella, Oscillospira, and Catenibacterium were risk factors. For thyrotoxicosis with Plummer Disease (PD), protective taxa included Butyricimonas and Lachnospira, whereas Dorea, Eggerthella, Odoribacter, Lactobacillus, Intestinimonas, and Phascolarctobacterium increased risk. Lastly, Parasutterella was protective against thyrotoxicosis with toxic single thyroid nodule (TSTN), while increased risk was associated with Sutterella, Oscillibacter, and Clostridium sensu stricto1. Conclusions: Our findings support a causal relationship between specific GM and TDs at the genetic level, laying the foundation for future research into potential mechanisms and the identification of novel therapeutic targets.

Computer systems and applications generate large amounts of logs to measure and record information, which is vital to protect the systems from malicious attacks and useful for repairing faults, especially with the rapid development of distributed computing. Among various logs, the anomaly log is beneficial for operations and maintenance (O&M) personnel to locate faults and improve efficiency. In this paper, we utilize a large language model, ChatGPT, for the log parser task. We choose the BERT model, a self-supervised framework for log anomaly detection. BERT, an embedded transformer encoder, with a self-attention mechanism can better handle context-dependent tasks such as anomaly log detection. Meanwhile, it is based on the masked language model task and next sentence prediction task in the pretraining period to capture the normal log sequence pattern. The experimental results on two log datasets show that the BERT model combined with an LLM performed better than other classical models such as Deelog and Loganomaly.

With the rapid advances in next-generation sequencing technology, numerous non-protein-coding transcripts have been identified, including long noncoding RNAs (lncRNAs), which are functional RNAs comprising more than 200 nucleotides. Although lncRNA-mediated regulatory processes have been extensively investigated in animals, there has been considerably less research on plant lncRNAs. Nevertheless, multiple studies on major crops showed lncRNAs are involved in crucial processes, including growth and development, reproduction, and stress responses. This review summarizes the progress in the research on lncRNA roles in several major crops, presents key strategies for exploring lncRNAs in crops, and discusses current challenges and future prospects. The insights provided in this review will enhance our comprehension of lncRNA functions in crops, with potential implications for improving crop genetics and breeding.

Introduction There is still a lack of therapeutic options for long COVID. Several studies have shown the benefit of hyperbaric oxygen therapy (HBOT) on long COVID. However, the efficacy and safety of HBOT for long COVID remain unclear. Therefore, we will conduct this systematic review to assess the feasibility of HBOT as a primary or complemental therapy for long COVID.Methods and analysis Databases such as Web of Science, PubMed, Embase, Cochrane Database of Systematic Reviews, ClinicalTrials.gov, International Clinical Trials Registry Platform, Wanfang Database, China National Knowledge Infrastructure, SINOMED, VIP Database and the Chinese Clinical Trial Registry will be searched systematically from the establishment to 9 December 2023. All articles will be reviewed by two independent reviewers. Cochrane risk of bias tool will be used to assess the risk of bias in the study. We will evaluate heterogeneity using a visual inspection of the funnel plot. If an available number of studies are identified, we will perform a meta-analysis.Ethics and dissemination No ethical approval is required since this study is based on published articles. Extreme high and low temperatures both exert impacts on terrestrial ecosystems. However, current research still lacks a precise assessment of the risk of vegetation loss under simultaneous consideration of different temperature stresses and lag effects. To this end, we propose a methodology for assessing the risk of vegetation loss under temperature stress that incorporates lag effects, based on weekly normalized difference vegetation index and temperature data. Quantified risk probabilities of different terrestrial ecosystems to warming and cooling stresses in Heilongjiang Province, China. The results of the study revealed a strong association between vegetation and temperature change during the growing season, reaching the most sensitive state around 9 weeks and 23 weeks lag, respectively, with high spatial consistency. The study identifies the eastern and western edges of the study area as high-risk zones for vegetation loss, while the risk is comparatively lower in the northwestern and central regions. The probability of risk increased by about 0.5% for every 1°C of warming in average temperatures and by about 0.7% for every 1°C of cooling. This indicates that cooling has a greater impact on vegetation than warming. Farmland ecosystems had a higher change in risk to temperature stress and forest ecosystems had the least. This study provides new perspectives for understanding the specific impacts of temperature extremes on different ecosystems and provides a scientific basis for developing adaptive management measures.

This study investigated the effect of nitrogen application on the rhizosphere soil microenvironment of sunflower and clarified the relationship between ammonium assimilation and the microenvironment. In a field experiment high (HN, 190 kg/hm2), medium (MN, 120 kg/hm2) and low nitrogen (CK, 50 kg/hm2) treatments were made to replicate plots of sunflowers using drip irrigation. Metagenomic sequencing was used to analyze the community structure and functional genes involved in the ammonium assimilation pathway in rhizosphere soil. The findings indicated that glnA and gltB played a crucial role in the ammonium assimilation pathway in sunflower rhizosphere soil, with Actinobacteria and Proteobacteria being the primary contributors. Compared with CK treatment, the relative abundance of Actinobacteria increased by 15.57% under MN treatment, while the relative abundance decreased at flowering and maturation stages. Conversely, the relative abundance of Proteobacteria was 28.57 and 61.26% higher in the MN treatment during anthesis and maturation period, respectively, compared with the CK. Furthermore, during the bud stage and anthesis, the abundance of Actinobacteria, Proteobacteria, and their dominant species were influenced mainly by rhizosphere soil EC, ammonium nitrogen (NH4+-N), and nitrate nitrogen (NO3--N), whereas, at maturity, soil pH and NO3--N played a more significant role in shaping the community of ammonium-assimilating microorganisms. The MN treatment increased the root length density, surface area density, and root volume density of sunflower at the bud, flowering, and maturity stages compared to the CK. Moreover, root exudates such as oxalate and malate were positively correlated with the dominant species of Actinobacteria and Proteobacteria during anthesis and the maturation period. Under drip irrigation, applying 120 kg/hm2 of nitrogen to sunflowers effectively promoted the community structure of ammonium-assimilating microorganisms in rhizosphere soil and had a positive influence on the rhizosphere soil microenvironment and sunflower root growth.

ObjectivesThe altered neuromelanin in substantia nigra pars compacta (SNpc) is a valuable biomarker in the detection of early-stage Parkinson’s disease (EPD). Diagnosis via visual inspection or single radiomics based method is challenging. Thus, we proposed a novel hybrid model that integrates radiomics and deep learning methodologies to automatically detect EPD based on neuromelanin-sensitive MRI, namely short-echo-time Magnitude (setMag) reconstructed from quantitative susceptibility mapping (QSM).MethodsIn our study, we collected QSM images including 73 EPD patients and 65 healthy controls, which were stratified into training-validation and independent test sets with an 8:2 ratio. Twenty-four participants from another center were included as the external validation set. Our framework began with the detection of the brainstem utilizing YOLO-v5. Subsequently, a modified LeNet was applied to obtain deep learning features. Meanwhile, 1781 radiomics features were extracted, and 10 features were retained after filtering. Finally, the classified models based on radiomics features, deep learning features, and the hybrid of both were established through machine learning algorithms, respectively. The performance was mainly evaluated using accuracy, net reclassification improvement (NRI), and integrated discrimination improvement (IDI). The saliency map was used to visualize the model.ResultsThe hybrid feature-based support vector machine (SVM) model showed the best performance, achieving ACC of 96.3 and 95.8% in the independent test set and external validation set, respectively. The model established by hybrid features outperformed the one radiomics feature-based (NRI: 0.245, IDI: 0.112). Furthermore, the saliency map showed that the bilateral “swallow tail” sign region was significant for classification.ConclusionThe integration of deep learning and radiomic features presents a potent strategy for the computer-aided diagnosis of EPD. This study not only validates the accuracy of our proposed model but also underscores its interpretability, evidenced by differential significance across various anatomical sites.

ObjectivesThe aims of this study were to report the effectiveness and safety of teriflunomide in Chinese patients with relapsing–remitting multiple sclerosis (RRMS) and to explore the association of paramagnetic rim lesion (PRL) burden with patient outcome in the context of teriflunomide treatment and the impact of teriflunomide on PRL burden.MethodsThis is a prospective observational study. A total of 100 RRMS patients treated with teriflunomide ≥3 months were included in analyzing drug persistence and safety. Among them, 96 patients treated ≥6 months were included in assessing drug effectiveness in aspects of no evidence of disease activity (NEDA) 3. The number and total volume of PRL were calculated in 76 patients with baseline susceptibility-weighted imaging (SWI), and their association with NEDA3 failure during teriflunomide treatment was investigated.ResultsOver a treatment period of 19.7 (3.1–51.7) months, teriflunomide reduced annualized relapse rate (ARR) from 1.1 ± 0.8 to 0.3 ± 0.5, and Expanded Disability Status Scale (EDSS) scores remained stable. At month 24, the NEDA3% and drug persistence rate were 43.8% and 65.1%, respectively. In patients with a baseline SWI, 81.6% had at least 1 PRL, and 42.1% had ≥4 PRLs. The total volume of PRL per patient was 0.3 (0.0–11.5) mL, accounting for 2.3% (0.0%–49.0%) of the total T2 lesion volume. Baseline PRL number ≥ 4 (OR = 4.24, p = 0.009), younger onset age (OR = 0.94, p = 0.039), and frequent relapses in initial 2 years of disease (OR = 13.40, p = 0.026) were associated with NEDA3 failure. The PRL number and volume were not reduced (p = 0.343 and 0.051) after teriflunomide treatment for more than 24 months. No new safety concerns were identified in this study.ConclusionTeriflunomide is effective in reducing ARR in Chinese patients with RRMS. Patients with less PRL burden, less frequent relapses, and relatively older age are likely to benefit more from teriflunomide, indicating that PRL might be a valuable measurement to inform clinical treatment decision.

Background The redundant extracellular matrix (ECM) within tumor microenvironment (TME) such as hyaluronic acid (HA) often impairs intratumoral dissemination of antitumor drugs. Oncolytic viruses (OVs) are being studied extensively for cancer therapy either alone or in conjunction with chemotherapy and immunotherapy. Here, we designed a novel recombinant vaccinia virus encoding a soluble version of hyaluronidase Hyal1 (OVV-Hyal1) to degrade the HA and investigated its antitumor effects in combination with chemo drugs, polypeptide, immune cells, and antibodies.Methods We constructed a recombinant oncolytic vaccinia virus encoding the hyaluronidase, and investigated its function in remodeling the ECM of the TME, the antitumor efficacy both in vitro and in several murine solid tumors either alone, or in combination with chemo drugs including doxorubicin and gemcitabine, with polypeptide liraglutide, with immune therapeutics such as PD-L1/PD-1 blockade, CD47 antibody, and with CAR-T cells.Results Compared with control OVV, intratumoral injection of OVV-Hyal1 showed superior antitumor efficacies in a series of mouse subcutaneous tumor models. Moreover, HA degradation by OVV-Hyal1 resulted in increased intratumoral dissemination of chemo drugs, infiltration of T cells, NK cells, macrophages, and activation of CD8+ T cells. When OVV-Hyal1 was combined with some antitumor therapeutics, for example, doxorubicin, gemcitabine, liraglutide, anti-PD-1, anti-CD47 blockade, or CAR-T cells, more profound therapeutic outcomes were obtained.Conclusions OVV-Hyal1 effectively degrades HA to reshape the TME, therefore overcoming some major hurdles in current cancer therapy, such as limited OVs spread, unfavored dissemination of chemo drugs, polypeptides, antibodies, and insufficient infiltration of effector immune cells. OVV-Hyal1 holds the promise to improve the antitumor outcomes of current cancer therapeutics.

Exposure to cadmium (Cd), a toxic heavy metal classified as an environmental endocrine disruptor, can exert significant toxicity in both animals and humans. However, the potential effects of Cd exposure on socioemotional behaviors are still poorly understood, as are the underlying mechanisms. In the present study, employing a series of behavioral tests as well as 16 S rRNA sequencing analysis, we investigated the long-term effects of Cd exposure on socioemotional behaviors and their associated mechanisms in mice based on the brain–gut interaction theory. The results showed that postweaning exposure to Cd reduced the ability to resist depression, decreased social interaction, subtly altered sexual preference, and changed the composition of the gut microbiota in male mice during adolescence. These findings provided direct evidence for the deleterious effects of exposure to Cd in the postweaning period on socioemotional behaviors later in adolescence, and suggested that these effects of Cd exposure may be linked to changes in the gut microbiota.

Abstract Ubiquitination is a post-translational modification initiated by the E1 enzyme UBA1, which transfers ubiquitin to ~35 E2 ubiquitin-conjugating enzymes. While UBA1 loss is cell lethal, it remains unknown how partial reduction in UBA1 activity is endured. Here, we utilize deep-coverage mass spectrometry to define the E1-E2 interactome and to determine the proteins that are modulated by knockdown of UBA1 and of each E2 in human cells. These analyses define the UBA1/E2-sensitive proteome and the E2 specificity in protein modulation. Interestingly, profound adaptations in peroxisomes and other organelles are triggered by decreased ubiquitination. While the cargo receptor PEX5 depends on its mono-ubiquitination for binding to peroxisomal proteins and importing them into peroxisomes, we find that UBA1/E2 knockdown induces the compensatory upregulation of other PEX proteins necessary for PEX5 docking to the peroxisomal membrane. Altogether, this study defines a homeostatic mechanism that sustains peroxisomal protein import in cells with decreased ubiquitination capacity.

【Objective】Sesuvium portulacastrum, a typical coastal and mangrove associate plant, is capable of retaining sand and protecting banks. It can grow normally in seawater and has a strong salt tolerance. Na+/H+ exchange (NHX) plays a key role in salt tolerance and plant growth with development. In order to understand the role of NHX in salt tolerance of S. portulacastrum, the NHX genes were identified and analyzed.【Method】In this study, the S. portulacastrum NHX genes were identified by using informatic method through its full length transcriptome data. Their physicochemical properties, conserved motifs and evolutionary relationships were analyzed, and the expression patterns of NHX genes under s alt stress were studied by quantative real-time PCR (qRT-PCR).【Result】The results showed that 12 SpNHX genes were identified from S. portulacastrum and named as SpNHX1 to SpNHX12. The number of amino acids ranged from 276 to 554, with the molecular weight ranged from 31.22 to 61.21 kD, isoelectric point from 5.55 to 8.64, and instability ind ex from 32.14 to 50.54. All the proteins were hydrophobic. Phylogenetic analysis showed that SpNHX proteins can be divided into two subgroups. Similar conserved motifs composition was found in each subgroup. Multiple sequence alignment results showed that all SpNHX proteins had Na+/H+ exchange conserved domains. RNA-seq data and qRT-PCR results showed that SpNHX genes were induced to different degrees under salt stress, and SpNHX1, SpNHX9, SpNHX10, SpNHX11 and SpNHX12 were significantly up-regulated under salt stress.【Conclusion】This study clarified the expression patterns of NHXs in S. portulacastrum under high salt stress, indicating that SpNHX1, SpNHX9, SpNHX10, SpNHX11 and SpNHX12 may be involved in the response of S. portulacastrum to salt stress, which provided a theoretical basis for further research on the role of NHX in the process of salt tolerance of S. portulacastrum.

Salt stress is one of the important abiotic stresses affecting plant growth and development, and it seriously restricts agricultural production and economic development. The utilization of salinized farmland has been a worldwide problem. It is of great theoretical significance and application value to study the mechanism of plant salt tolerance and cultivate new varieties of salt-tolerant plants to make full use of salinized farmland. At present, a growing number of genes involved in salt stress response have been discovered and revealed. When plants exposed to high-salt environment conditions, many proteins in plant cells are involved in salt stress response. The receptor-like kinases and components of the cell wall respond to salt stress. The glycosyl inositol phosphorylceramide on the cell membrane acts as a Na+ receptor and binds to Na+, then it causes changes in cell surface to generate calcium signals to activate downstream regulatory pathways. The plasma membrane potassium channel proteins and Na+/H+ antiporter mediate Na+ influx and exclusion, and the tonoplast Na+/H+ antiporter compartments the excess Na+ in the cytoplasm into the vacuole. In addition, transcription factors also participate in the transcriptional regulation of plant adaptation to salt stress, and play important roles in the regulation of plant salt tolerance. In this study, the molecular mechanisms of salt-tolerant genes reported in recent years were reviewed based on their localization in plant cells, their functions in improving plant salt tolerance were summarized, and application potential of salt-tolerant genes were prospected, with an aim to provide references for plant salt-tolerance molecular breeding and scientific basis for salinized farmland improvement.

Abstract Alzheimer’s disease (AD) is the most common form of dementia worldwide, with a projection of 151 million cases by 2050. Previous genetic studies have identified three main genes associated with early-onset familial Alzheimer’s disease, however this subtype accounts for less than 5% of total cases. Next-generation sequencing has been well established and holds great promise to assist in the development of novel therapeutics as well as biomarkers to prevent or slow the progression of this devastating disease. Here we present a public resource of functional genomic data from the parahippocampal gyrus of 201 postmortem control, mild cognitively impaired (MCI) and AD individuals from the Mount Sinai brain bank, of which whole-genome sequencing (WGS), and bulk RNA sequencing (RNA-seq) were previously published. The genomic data include bulk proteomics and DNA methylation, as well as cell-type-specific RNA-seq and assay for transposase-accessible chromatin with high-throughput sequencing (ATAC-seq) data. We have performed extensive preprocessing and quality control, allowing the research community to access and utilize this public resource available on the Synapse platform at https://doi.org/10.7303/syn51180043.2 .

Soil mercury (Hg) contamination is a serious threat to local ecology and public health. Exogenous selenium (Se) supplementation can effectively reduce the toxicity of Hg. However, the mechanisms affecting the changes in soil Hg speciation, soil properties and the microbial Hg-resistant system during the Se–Hg interaction after exogenous Se supplementation are not clear. Therefore, in this study, soil culture experiments were conducted to analyze the effects of different Se additions on the transformation of Hg speciation, soil properties and Hg-resistant microorganisms and resistant genes (mer operon). The results indicated that Se supplementation facilitated the transformation of soil Hg from bioavailable (exchangeable and carbonate-bound) to stable forms (organic material-bound and residual), significantly reducing Hg bioavailability. Se supplementation notably decreased the electrical conductivity of Hg-contaminated soil, but had no significant effect on the soil pH, organic matter content, cation exchange capacity or alkaline phosphatase and catalase activities. The maximum activity levels of soil sucrase and urease were observed when 1 mg kg−1 Se was added. Se significantly inhibited soil peroxidase and ascorbate oxidase activities, thereby alleviating the oxidative stress in the soil system caused by Hg. Additionally, Se significantly activated the Hg-resistant system in soil microorganisms by either decreasing or increasing the regulatory genes merD and merR, and it significantly upregulated the cytoplasmic protein gene merP and the membrane protein genes merC, merF and merT. This further increased the abundance of the organomercury lyase gene merB and the mercuric reductase gene merA, promoting the conversion of Hg species to Hg⁰. Furthermore, the abundance of mer operon-containing microorganisms, such as Thiobacillus ferrooxidants, Pseudomonas, Streptomyces and Cryptococcus, significantly increased with Se addition, explaining the role of soil microorganisms in mitigating soil Hg stress via Se supplementation.
