1. A synonymous mutation in IGF-1 impacts the transcription and translation process of gene expression

Abstract: Insulin-like growth factor 1 (IGF-1) is considered to be a crucial gene in the animal development of bone and body size. In this study, a unique synonymous mutation (c.258 A > G) of the IGF-1 gene was modified with an adenine base editor to observe the growth and developmental situation of mutant mice. Significant expression differences and molecular mechanisms among vectors with different alanine synonymous codons were explored. Although modification of a single synonymous codon rarely interferes with animal phenotypes, we observed that the expression and secretion of IGF-1 were different between 8-week-old homozygous (Ho) and wild-type (WT) mice. In addition, the IGF-1 with optimal codon combinations showed a higher expression content than other codon combination modes at both transcription and translation levels and performed proliferation promotion. The gene stability and translation initiation efficiency also changed significantly. Our findings illustrated that the synonymous mutation altered the IGF-1 gene expression in individual mice and suggested that the synonymous mutation affected the IGF-1 expression and biological function through the transcription and translation processes.

2. Effects of corn particle size on growth performance, gastrointestinal development, carcass indices and intestinal microbiota of broilers

Abstract: ABSTRACT: This experiment was conducted to investigate the effects of different corn particle sizes on growth performance, gastrointestinal development, carcass processing yields and intestinal microbiota of caged broilers. One-day-old Ross 308 broilers were randomly divided into 8 treatments with 10 replicates per treatment and 30 birds per replicate pen. The experiment lasted 37 d. Feed and water were provided ad libitum. The results showed as follows: birds fed diets with the FG corn between d 1 and 13 and CG corn between d14 to 37 had increased body weight, daily gain, and feed intake (P < 0.05). Birds fed diets with CG corn between d 24 to 37 had a heavier relative weight of gizzard at d 38 (P < 0.05). Birds fed diets with FG corn from d 1 to 13 and the CG corn from d 14 to 37 had a higher carcass yield and a relative thigh weight at d 38 (P < 0.05). The intestinal microbiota was significantly affected by different corn particle sizes. The relative abundance of Lactobacillaceae was significantly decreased, whereas that of Peptostreptococcaceae was increased (P < 0.05) in birds fed with the CG corn between d1 to 37. The relative abundance of Acinetobacter was significantly increased in birds fed the FG corn between d1 to 37 (P < 0.05). In conclusion, the use of FG corn in the starter phase and CG corn in the grower and finisher phases was beneficial to growth performance, gastrointestinal development and intestinal microbial structure of broilers reared in cages.

Abstract: Using 16 energy points of e+e− annihilation data collected in the vicinity of the J/ψ resonance with the BESIII detector and with a total integrated luminosity of around 100pb−1, we study the relative phase between the strong and electromagnetic amplitudes of J/ψ decays. The relative phase between J/ψ electromagnetic decay and the continuum process (e+e− annihilation without the J/ψ resonance) is confirmed to be zero by studying the cross section lineshape of μ+μ− production. The relative phase between J/ψ strong and electromagnetic decays is then measured to be (84.9±3.6)∘ or (−84.7±3.1)∘ for the 2(π+π−)π0 final state by investigating the interference pattern between the J/ψ decay and the continuum process. This is the first measurement of the relative phase between J/ψ strong and electromagnetic decays into a multihadron final state using the lineshape of the production cross section. We also study the production lineshape of the multihadron final state ηπ+π− with η→π+π−π0, which provides additional information about the phase between the J/ψ electromagnetic decay amplitude and the continuum process. Additionally, the branching fraction of J/ψ→2(π+π−)π0 is measured to be (4.73±0.44)% or (4.85±0.45)%, and the branching fraction of J/ψ→ηπ+π− is measured to be (3.78±0.68)×10−4. Both of them are consistent with the world average values. The quoted uncertainties include both statistical and systematic uncertainties, which are mainly caused by the low statistics. Abstract: This study was conducted to evaluate the effects of 3 rearing systems (FL: flooring litter rearing, MC: multilayer cage rearing, PN: plastic net rearing) with or without supplemental narasin on growth performance, gastrointestine development and health of broilers. A total of 2,400 one-day-old Ross 308 mixed-sex broilers (1:1 ratio of males and females) were used in a completely randomized design utilizing a 3 × 2 factorial arrangement of treatments, with 12 replicates per treatment. Each replicate for FL, MC, and PN consisted of 34 birds per floor pen, 30 birds per cage, and 36 birds per net pen, respectively, ensuring the same stocking density (12 birds/m2) across the 3 systems. Abstract: We report a study of the e+e−→D+D−π+π− process using e+e− collision data samples with an integrated luminosity of 2.5fb−1 at center-of-mass energies from 4.36 to 4.60GeV, collected with the BESIII detector at the BEPCII storage ring. The D1(2420)+ is observed in the D+π+π− mass spectrum. The mass and width of the D1(2420)+ are measured to be (2427.2±1.0stat.±1.2syst.)MeV/c2 and (23.2±2.3stat.±2.3syst.)MeV, respectively. In addition, the Born cross sections of the e+e−→D1(2420)+D−+c.c.→D+D−π+π− and e+e−→ψ(3770)π+π−→D+D−π+π− processes are measured as a function of the center-of-mass energy.

Abstract: Using a data sample of e+e− collision data corresponding to an integrated luminosity of 2.93 fb−1 collected with the BESIII detector at a center-of-mass energy of s=3.773GeV, we search for the singly Cabibbo-suppressed decays D0→π0π0π0, π0π0η, π0ηη and ηηη using the double tag method. The absolute branching fractions are measured to be B(D0→π0π0π0)=(2.0±0.4±0.3)×10−4, B(D0→π0π0η)=(3.8±1.1±0.7)×10−4 and B(D0→π0ηη)=(7.3±1.6±1.5)×10−4 with the statistical significances of 4.8σ, 3.8σ and 5.5σ, respectively, where the first uncertainties are statistical and the second ones systematic. No significant signal of D0→ηηη is found, and the upper limit on its decay branching fraction is set to be B(D0→ηηη)

Abstract: Using a data set of 2.93 fb−1 taken at a center-of-mass energy s=3.773 GeV with the BESIII detector at the BEPCII collider, we perform a search for an extra U(1) gauge boson, also denoted as a dark photon. We examine the initial state radiation reactions e+e−→e+e−γISR and e+e−→μ+μ−γISR for this search, where the dark photon would appear as an enhancement in the invariant mass distribution of the leptonic pairs. We observe no obvious enhancement in the mass range between 1.5 and 3.4 GeV/c2 and set a 90% confidence level upper limit on the mixing strength of the dark photon and the Standard Model photon. We obtain a competitive limit in the tested mass range.

Abstract: Using data samples collected with the BESIII detector at the BEPCII collider at six center-of-mass energies between 4.008 and 4.600 GeV, we observe the processes e+e−→ϕϕω and e+e−→ϕϕϕ. The Born cross sections are measured and the ratio of the cross sections σ(e+e−→ϕϕω)/σ(e+e−→ϕϕϕ) is estimated to be 1.75±0.22±0.19 averaged over six energy points, where the first uncertainty is statistical and the second is systematic. The results represent first measurements of these interactions.

Abstract: We report the first observation of the decay Λc+→Σ−π+π+π0, based on data obtained in e+e− annihilations with an integrated luminosity of 567 pb−1 at s=4.6 GeV. The data were collected with the BESIII detector at the BEPCII storage rings. The absolute branching fraction B(Λc+→Σ−π+π+π0) is determined to be (2.11±0.33(stat.)±0.14(syst.))%. In addition, an improved measurement of B(Λ+c→Σ−π+π+) is determined as (1.81±0.17(stat.)±0.09(syst.))%.

Abstract: Based on e+e− annihilation data samples collected with the BESIII detector at the BEPCII collider at 13 center-of-mass energies from 4.008 to 4.600 GeV, measurements of the Born cross section of e+e−→pp¯π0 are performed. No significant resonant structure is observed in the measured energy dependence of the cross section. The upper limit on the Born cross section of e+e−→Y(4260)→pp¯π0 at the 90% C.L. is determined to be 0.01 pb. The upper limit on the ratio of the branching fractions B(Y(4260)→pp¯π0)B(Y(4260)→π+π−J/ψ) at the 90% C.L. is determined to be 0.02%. Keywords: Hadrons, Cross section measurements, Y(4260)

Abstract: We study the decays of J/ψ and ψ(3686) to the final states Σ(1385)0Σ¯(1385)0 and Ξ0Ξ¯0 based on a single baryon tag method using data samples of (1310.6±7.0)×106 J/ψ and (447.9±2.9)×106 ψ(3686) events collected with the BESIII detector at the BEPCII collider. The decays to Σ(1385)0Σ¯(1385)0 are observed for the first time. The measured branching fractions of J/ψ and ψ(3686) to Ξ0Ξ¯0 are in good agreement with, and much more precise than, the previously published results. The angular parameters for these decays are also measured for the first time. The measured angular decay parameter for J/ψ→Σ(1385)0Σ¯(1385)0, α=−0.64±0.03±0.10, is found to be negative, different to the other decay processes in this measurement. In addition, the “12% rule” and isospin symmetry in the decays of J/ψ and ψ(3686) to ΞΞ¯ and Σ(1385)Σ¯(1385) are tested.

Abstract: We report the first measurement of the absolute branching fraction for Λc+→Λμ+νμ. This measurement is based on a sample of e+e− annihilation data produced at a center-of-mass energy s=4.6 GeV, collected with the BESIII detector at the BEPCII storage rings. The sample corresponds to an integrated luminosity of 567 pb−1. The branching fraction is determined to be B(Λc+→Λμ+νμ)=(3.49±0.46(stat)±0.27(syst))%. In addition, we calculate the ratio B(Λc+→Λμ+νμ)/B(Λc+→Λe+νe) to be 0.96±0.16(stat)±0.04(syst).

Abstract: By analyzing 2.93fb−1 of data taken at the ψ(3770) resonance peak with the BESIII detector, we measure the branching fractions for the hadronic decays D+→KS0KS0K+, D+→KS0KS0π+, D0→KS0KS0 and D0→KS0KS0KS0. They are determined to be B(D+→KS0KS0K+)=(2.54±0.05stat.±0.12sys.)×10−3, B(D+→KS0KS0π+)=(2.70±0.05stat.±0.12sys.)×10−3, B(D0→KS0KS0)=(1.67±0.11stat.±0.11sys.)×10−4 and B(D0→KS0KS0KS0)=(7.21±0.33stat.±0.44sys.)×10−4, where the second one is measured for the first time and the others are measured with significantly improved precision over the previous measurements.

Abstract: The four decay modes D0→ϕπ0, D0→ϕη, D+→ϕπ+, and D+→ϕK+ are studied by using a data sample taken at the centre-of-mass energy s=3.773 GeV with the BESIII detector, corresponding to an integrated luminosity of 2.93 fb−1. The branching fractions of the first three decay modes are measured to be B(D0→ϕπ0)=(1.168±0.028±0.028)×10−3, B(D0→ϕη)=(1.81±0.46±0.06)×10−4, and B(D+→ϕπ+)=(5.70±0.05±0.13)×10−3, respectively, where the first uncertainties are statistical and the second are systematic. In addition, the upper limit of the branching fraction for D+→ϕK+ is given to be 2.1×10−5 at the 90% confidence level. The ratio of B(D0→ϕπ0) to B(D+→ϕπ+) is calculated to be (20.49±0.50±0.45)%, which is consistent with the theoretical prediction based on isospin symmetry between these two decay modes. Abstract: Using a data set of 2.93 fb−1 taken at a center-of-mass energy of s=3.773 GeV with the BESIII detector at the BEPCII collider, we measure the process e+e−→J/ψγ→μ+μ−γ and determine the product of the branching fraction and the electronic width Bμμ⋅Γee=(333.4±2.5stat±4.4sys) eV. Using the earlier-published BESIII result for Bμμ=(5.973±0.007stat±0.037sys)%, we derive the J/ψ electronic width Γee=(5.58±0.05stat±0.08sys) keV.

Abstract: We extract the e+e−→π+π− cross section in the energy range between 600 and 900 MeV, exploiting the method of initial state radiation. A data set with an integrated luminosity of 2.93 fb−1 taken at a center-of-mass energy of 3.773 GeV with the BESIII detector at the BEPCII collider is used. The cross section is measured with a systematic uncertainty of 0.9%. We extract the pion form factor |Fπ|2 as well as the contribution of the measured cross section to the leading-order hadronic vacuum polarization contribution to (g−2)μ. We find this value to be aμππ,LO(600–900MeV)=(368.2±2.5stat±3.3sys)⋅10−10, which is between the corresponding values using the BaBar or KLOE data.

Abstract: By analyzing a data set of 2.92 fb−1 of e+e− collision data taken at s=3.773 GeV and 106.41×106 ψ(3686) decays taken at s=3.686 GeV with the BESIII detector at the BEPCII collider, we measure the branching fraction and the partial decay width for ψ(3770)→γχc0 to be B(ψ(3770)→γχc0)=(6.88±0.28±0.67)×10−3 and Γ[ψ(3770)→γχc0]=(187±8±19) keV, respectively. These are the most precise measurements to date.

Abstract: We study Ds+ decays to final states involving the η′ with a 482 pb−1 data sample collected at s=4.009 GeV with the BESIII detector at the BEPCII collider. We measure the branching fractions B(Ds+→η′X)=(8.8±1.8±0.5)% and B(Ds+→η′ρ+)=(5.8±1.4±0.4)% where the first uncertainty is statistical and the second is systematic. In addition, we estimate an upper limit on the non-resonant branching ratio B(Ds+→η′π+π0)

Abstract: Using the data sets taken at center-of-mass energies above 4 GeV by the BESIII detector at the BEPCII storage ring, we search for the reaction e+e−→γISRX(3872)→γISRπ+π−J/ψ via the Initial State Radiation technique. The production of a resonance with quantum numbers JPC=1++ such as the X(3872) via single photon e+e− annihilation is forbidden, but is allowed by a next-to-leading order box diagram. We do not observe a significant signal of X(3872), and therefore give an upper limit for the electronic width times the branching fraction ΓeeX(3872)B(X(3872)→π+π−J/ψ)

Abstract: We report a measurement of the parameter yCP in D0–D¯0 oscillations performed by taking advantage of quantum coherence between pairs of D0D¯0 mesons produced in e+e− annihilations near threshold. In this work, doubly-tagged D0D¯0 events, where one D decays to a CP eigenstate and the other D decays in a semileptonic mode, are reconstructed using a data sample of 2.92 fb−1 collected with the BESIII detector at the center-of-mass energy of s=3.773 GeV. We obtain yCP=(−2.0±1.3±0.7)%, where the first uncertainty is statistical and the second is systematic. This result is compatible with the current world average. 21. Effect of supplementation of green tea polyphenols on the developmental competence of bovine oocytes in vitro

Abstract: The objective of the present study was to examine the effect of green tea polyphenols (GTPs) supplementation during in vitro maturation, in vitro fertilization, and in vitro culture on the developmental competence of bovine oocytes. Cumulus-oocyte complexes aspirated from the ovaries were matured in vitro (38.5ºC for 24 h) and fertilized (38.5ºC for 15-18 h) and embryos were cultured (38.5ºC for 192 h) in a defined conditioned medium with or without GTPs supplementation. The GTPs used in the present study contained 99% catechin derivatives, with the major components being 50% (-)-epigallocatechin gallate, 22% (-)-epicatechin gallate, 18% (-)-epigallocatechin, and 10% (-)-epicatechin. Four replicate trials were done for each type of experiment. GTPs supplementation (15 µM) of the maturation medium led to a significant increase in the rate of blastocyst formation (34.0 vs 21.4%, P < 0.05). However, the rate of blastocyst formation was not improved when higher GTPs concentrations (20 or 25 µM) were added to the in vitro maturation medium. During in vitro fertilization, supplementation with higher GTPs concentrations (20 or 25 µM) significantly reduced the rate of blastocyst formation (P < 0.05). Supplementation of the culture medium with 15 µM GTPs improved the rate of blastocyst formation, while higher GTPs concentrations (25 µM) significantly reduced embryo development (P < 0.05). In conclusion, these results demonstrate that supplementation with GTPs at low concentration (15 µM) during in vitro maturation and in vitro culture improved the developmental competence of bovine oocytes.

22. Effects of oocyte collection techniques and maturation media on in vitro maturation and subsequent embryo development in Boer goat

Abstract: The oocytes (experiment 1) were harvested by one of the four collection techniques (slicing, puncture, aspiration I and aspiration II) and the total number and the number of each grade of oocytes were counted, respectively. The good-quality oocytes (good and fair grade) were cultured for maturation. In experiment 2, the oocytes were matured in TCM-199 supplemented with 10 ng/ml of epidermal growth factor (EGF) or 10% FCS, either alone or with 1 IU/ml FSH, or the oocytes were matured in TCM-199 supplemented with 1 IU/ml FSH. After maturation, the oocytes in the two experiments were fertilized, respectively. Slicing (6.3) and puncture (5.8) of the ovaries yielded a higher (P < 0.05) number of oocytes per ovary compared to aspiration I (2.9) and aspiration II (3.1). Oocytes matured in the TCM-199 medium supplemented with EGF or FCS with FSH had a significantly higher proportion of blastocysts than the other treatments (P < 0.05). In conclusion, slicing and puncture are alternative techniques of oocyte recovery in Boer goat. The TCM-199 medium supplemented with EGF or FCS in the presence of FSH is suitable for in vitro maturation of oocytes.

23. Creep Behavior Analysis and Creep Rupture Prediction of Carbon-glass Fiber Reinforced Polymer Tendon

Abstract: Carbon-glass fiber reinforced polymer (C-GFRP) tendons is a kind of two components fiber-reinforced material manufactured by using pulltrusion process, consisting of primarily longitudinal glass fibers bound by carbon fibers at a certain angle. The paper attempts to figure out the relationship of C-GFRP tendon’s maximum bearing capacity, loading capacity and creep behavior through tensile testing and creep testing of a C-GFRP tendon with the diameter of 8 mm. The results show that the ultimate tensile strength of the tendon is 1452.22 MPa, its tensile rate is 1.4% and its elastic modulus is 103.48 GPa, and that its failure mode is different from the silky failure mode of one component FRP tendon that is characterized with interface shear failure. When the level of stress C-GFRP tendons bear is 60% more than its ultimate tensile strength in value, there is an obvious impact on creep behavior and creep rate. By analyzing the relationship of stress level, creep rupture and loading time, it can be figure out that the safe work duration of C-GFRP tendons is 500,000 hours when its stress level is 42% of its ultimate tensile strength in value.

Abstract: The phases, microstructures, and their formation mechanisms of cast zirconium-containing 304 stainless steel (304SS-Zr) alloys with excellent mechanical properties were investigated. Optical microscopy, scanning electron microscopy, transmission electron microscopy, and statistical analysis were employed to further understand the structure–property relationships. The in situ three-phase composite microstructure in 304SS-Zr alloys consisting of micron-sized (MS) austenite, MS intermetallic, and nanocrystalline (NC) ferrite was obtained by simple vacuum melting route. As zirconium (Zr) content increased, the grain sizes of austenite and ferrite became significantly refined. The multiple heterogeneous structures in 304SS-Zr alloys produced the super-high strength which also had higher strain hardening than conventional 304SS. For expanding the nanostructure design ideas, in this work, all the phases were identified and the mechanisms related to phases and microstructure evolution were analyzed from both the kinetic and thermodynamic aspects. For the development of high-performance 304SS-Zr alloys, the mechanical behaviors and strain-hardening mechanisms were also discussed.

Abstract: Chromobox 4 (CBX4) is a core component of polycomb-repressive complex 1 with important roles in cancer biology and tissue homeostasis. Aberrant expression of CBX4 has been implicated in several human malignancies. However, its role and underlying mechanisms in the tumorigenesis of lung adenocarcinoma (LUAD) have not been defined in vivo. Here, we found that expression of CBX4 was frequently up-regulated in human LUAD samples and correlated with poor patient survival. Importantly, genetic ablation of CBX4 greatly dampened lung tumor formation and improved survival in the KrasG12D/P53L/L (KP) autochthonous mouse model of LUAD. In addition, CBX4 depletion significantly inhibited proliferation and anchorage-independent growth of KP mouse embryonic fibroblasts. Moreover, ectopic CBX4 expression clearly promoted proliferation and anchorage-independent growth in both human and mouse LUAD cells, whereas silencing of CBX4 exerted opposite effects. Mechanistically, CBX4 promoted growth of LUAD cells through activation of the Wnt/β-catenin pathway. Abstract: Background and aims The conserved Hippo pathway regulates organ size, tissue homeostasis, and tumorigenesis. Interferon regulatory factor 2 binding protein 2 (IRF2BP2) was originally identified as a transcriptional corepressor. However, the association between IRF2BP2 and the Hippo pathway remains largely unknown. In addition, the biological function and regulation mechanism of IRF2BP2 in liver cancer are poorly understood. Approach and results In this study, we uncovered the clinical significance of IRF2BP2 in suppressing hepatocellular carcinogenesis. We showed that IRF2BP2, a direct target repressed by the Yes-associated protein (YAP)/TEA domain transcription factor 4 (TEAD4) transcriptional complex, inhibited YAP activity through a feedback loop. IRF2BP2 stabilized vestigial-like family member 4 (VGLL4) and further enhanced VGLL4's inhibitory function on YAP. Moreover, liver-specific IRF2BP2 overexpression suppressed tumor formation induced by Hippo pathway inactivation. Conclusions These results revealed the important role of IRF2BP2 in repressing liver cancer progression and highlighted a feedback loop underlying the Hippo pathway in organ-size control and tumorigenesis.

Abstract: As a system with both profound physics and promising application potentials, the strain-induced self-assembled semiconductor nanostructures have been investigated for decades of years. The optical and electrical properties of this system are mainly determined by the nanostructures, but can be greatly affected by the generally existent two dimensional structures from which the nanostructures evolve, the so called wetting layer (WL). The WL configurations can be varied with different growth conditions, and further influence the nanostructure morphology and properties. This is a reviewing article introducing some recent progresses in investigating the evolution of WLs in InAs/GaAs system with reflectance difference spectroscopy. Different kinds of WL evolution processes in Stranski-Krastanov growth mode are introduced. The segregation and desorption of indium atoms and the effect of growth interruption are mentioned. The existence and the evolution process of wetting layers in droplet epitaxy method are also discussed.

Abstract: BackgroundEnsete glaucum (2n = 2x = 18) is a giant herbaceous monocotyledonous plant in the small Musaceae family along with banana (Musa). A high-quality reference genome sequence of E. glaucum offers a vital genomic resource for functional and evolutionary studies of Ensete, the Musaceae, and more widely in the Zingiberales.FindingsUsing a combination of Illumina and Oxford Nanopore Technologies (ONT) sequencing, genome-wide chromosome conformation capture (Hi-C), and RNA survey sequence, we report a high-quality assembly of the 481.5Mb genome with 9 pseudochromosomes and 36,836 genes (BUSCO 94.7%). A total of 55% of the genome is composed of repetitive sequences with LTR-retroelements (37%) and DNA transposons (7%) predominant. The 5S and 45S rDNA were each present at one locus, and the 5S rDNA had an exceptionally long monomer length of c.1,056 bp, contrasting with the c. 450 bp monomer at multiple loci in Musa. A tandemly repeated c. 134 bp satellite, 1.1% of the genome (with no similar sequence in Musa), was present around all nine centromeres, with a LINE retroelement also found at Musa centromeres. The assembly, including centromeric positions, enabled us to characterize in detail the chromosomal rearrangements occurring between the x = 9 species and x = 11 species of Musa. Only one chromosome has the same gene content as M. acuminata (ma). Three ma chromosomes represent part of only one E. glaucum (eg) chromosome, while the remaining seven ma chromosomes are fusions of parts of two, three, or four eg chromosomes, demonstrating complex and multiple evolutionary rearrangements in the change between x = 9 and x = 11.ConclusionsThe advance towards a Musaceae pangenome including E. glaucum, tolerant of extreme environments, makes a complete set of gene alleles available for crop breeding and understanding environmental responses. The chromosome-scale genome assembly show how chromosome number evolves, and features of the rapid evolution of repetitive sequences.

Abstract: Ovarian surface epithelium (OSE) undergoes recurring ovulatory rupture and OSE stem cells rapidly generate new cells for the repair. How the stem cell senses the rupture and promptly turns on proliferation is unclear. Our previous study has identified that Protein C Receptor (Procr) marks OSE progenitors. In this study, we observed decreased adherent junction and selective activation of YAP signaling in Procr progenitors at OSE rupture site. OSE repair is impeded upon deletion of Yap in these progenitors. Interestingly, Procr+ progenitors show lower expression of Vgll4, an antagonist of YAP signaling. Overexpression of Vgll4 in Procr+ cells hampers OSE repair and progenitor proliferation, indicating that selective low Vgll4 expression in Procr+ progenitors is critical for OSE repair. In addition, YAP activation promotes transcription of the OSE stemness gene Procr. The combination of increased cell division and Procr expression leads to expansion of Procr+ progenitors surrounding the rupture site. These results illustrate a YAP- dependent mechanism by which the stem/progenitor cells recognize the ovulatory rupture, and rapidly multiply their numbers, highlighting a YAP- induced stem cell expansion strategy.

Abstract: Genetic findings suggested that ethnolinguistically diverse populations in China harbor differentiated genetic structure and complex evolutionary admixture history, which provide the genetic basis and theoretical foundation for forensic biogeographical ancestry inference (BGAI). Forensic assays for BGAI among intracontinental eastern Eurasians were previously conducted mainly based on the SNPs or InDels. Microhaplotypes, as a set of closely linked SNPs within 200 base pairs, possess the advantages of both STR and SNP and have great potential in forensic ancestry inference. However, the developed forensic assay based on the ancestry informative microhaplotypes in the BGAI remained to be comprehensively explored, especially in China with enriching genetic diversity. Here, we described a new BGAI panel based on 21 novel identified ancestry informative microhaplotypes that focused on dissected finer-scale ancestry composition of Chinese populations. We initially screened all possible microhaplotypes with high Fst values among five East Asian populations and finally employed 21 candidate microhaplotypes in two multiplex SNaPshot assays. Forensic amplification efficiency and statistically/physically phased haplotypes of the 21 microhaplotypes were validated using both SNaPshot and massively parallel sequencing (MPS) platforms. Followingly, we validated the efficiency of these microhaplotypes for BGAI in 764 individuals from ten Chinese populations. Fine-scale ancestry source and ancestry proportion estimated by the principal component analysis (PCA), multidimensional scaling (MDS), phylogenetic tree and model-based STRUCTURE among worldwide populations and East Asians showed that our customized panel could provide a higher discrimination resolution in both continental population stratification and East Asian regional substructure. East Asian populations could be further classified into linguistically/geographically different intracontinental subpopulations (Tibeto-Burman, Tai-Kadai and others). Finally, we obtained a higher estimated accuracy using training and tested datasets in the microhaplotype-based panel than traditional SNP-based panels. Generally, the above results demonstrated that this microhaplotype panel was robust and suitable for forensic BGAI in Chinese populations, which not only provided a high discriminatory power for continental populations but also discriminated East Asians into linguistically restricted subpopulations.

Abstract: In order to investigate the double-reverse-jet film cooling (DRJFC), the multi-field coupling calculating method is used to study the effect of geometric parameters on the resultant vortex structure and conjugate thermal-elastic property. The traditional streamwise film cooling is also investigated for comparison. The results indicate that the formation of effective anti-kidney vortices is the key to enhance the dimensionless temperature of DRJFC holes. At low blowing ratios, the streamwise or lateral distance between two DRJFC holes should be increased to widen the transverse shift of the jets, thus, to increase the cooling performance. At high blowing ratios, the lateral distance should be decreased to prevent two jets from separating apart so that the malfunction of the anti-kidney vortices could be avoided. The stress concentration resulting from the nonuniform temperature distribution is considered.

Abstract: In this study, the Ag and In were alloyed to the Sn-58Bi solder, and their individual and combined influences on the SnBi solder and the SnBi/Cu solder joints were investigated. The results indicate that the In can effectively decrease the melting temperature and improve the wettability of the solder, while the Ag shows slight impacts on the melting behavior and the wetting properties. During the isothermal aging at 100 °C, growth rate of the intermetallic compounds (IMCs) layer at the Ag-bearing solders/Cu interfaces are lower, while that at the In-bearing solders/Cu interfaces are much higher, because the substitution of Sn in the Cu6Sn5 IMC by In alters the interfacial diffusion behavior and assist in faster growth of IMCs layer. The Bi segregation at the long-term aged SnBi/Cu interface is eliminated in the aged SnBiAg/Cu, SnBiIn/Cu and SnBiAgIn/Cu solder joints. Tensile strength of all the solders joints decreases with increasing aging time, while the sharp decrease in the strength of the long-term aged SnBi/Cu solder joints is inhibited once the Bi segregation is eliminated, and fracture occurs mainly around the IMCs/solder interface. For the In-bearing solder joints, the Cu6(Sn,In)5 decrease the adhesion force between the solder and the interfacial IMCs, while the SnBiAg/Cu joints shows the highest strength. Based on the results, a more practicable method to inhibit the Bi embrittlement at the aged SnBi/Cu solder joints is proposed.

Abstract: The Hippo pathway plays a critical role in cell growth and tumorigenesis. The activity of TEA domain transcription factor 4 (TEAD4) determines the output of Hippo signaling; however, the regulation and function of TEAD4 has not been explored extensively. Here, we identified glucocorticoids (GC) as novel activators of TEAD4. GC treatment facilitated glucocorticoid receptor (GR)-dependent nuclear accumulation and transcriptional activation of TEAD4. TEAD4 positively correlated with GR expression in human breast cancer, and high expression of TEAD4 predicted poor survival of patients with breast cancer. Mechanistically, GC activation promoted GR interaction with TEAD4, forming a complex that was recruited to the TEAD4 promoter to boost its own expression. Functionally, the activation of TEAD4 by GC promoted breast cancer stem cells maintenance, cell survival, metastasis, and chemoresistance both in vitro and in vivo. Pharmacologic inhibition of TEAD4 inhibited GC-induced breast cancer chemoresistance. In conclusion, our study reveals a novel regulation and functional role of TEAD4 in breast cancer and proposes a potential new strategy for breast cancer therapy. Significance: This study provides new insight into the role of glucocorticoid signaling in breast cancer, with potential for clinical translation.

Abstract: Bulk nanostructured (NS) materials processed by severe plastic deformation (SPD) have received considerable attention for several decades. The physical origin of this processing philosophy is to enable substantial grain refinement from a micrometer to a nanoscale level mainly through the activation of fundamental deformation mechanisms: dislocation glide, deformation twinning, and their sophisticated interactions. The formation of nanostructures in NS metallic materials is significantly governed by the quintessential dominance of these two plasticity carriers during SPD, and their mechanical properties are thereby correspondingly affected. According to conventional crystal plasticity, the stacking fault energy (SFE) of materials is one of the most crucial factors primarily controlling which deformation mechanism plays an overwhelming role in accommodating the plasticity. Therefore, a profound understanding of the vital significance of SFE in NS materials can extend and enrich our comprehension of their structure-property relationship, lead to the design of NS metallic materials with superior properties, and pave the path for their perspective applications. Choosing Cu and its binary alloys as model systems, this review extensively surveys the principal influences of SFE on the preferred choice of deformation mechanisms during SPD, microstructural evolution, grain refinement, deformation behavior, and mechanical properties of NS material including tensile properties and cyclic deformation responses.

Abstract: In triple negative breast cancers (TNBCs), the autocrine activation of Wnt ligands, the epigenetic silencing of Wnt suppressor genes, and the cross talks between signaling pathways stabilize β-catenin in the dephosphorylated state, increase the level of nuclear β-catenin, and aberrantly activate the Wnt/β-catenin signaling pathway. The design of inhibitors for the upstream effectors of the Wnt/β-catenin signaling pathway cannot confer the inhibitory activities to cancer cells that harbor downstream APC, Axin, or β-catenin activation mutations and can perturb the function of β-catenin in cell–cell adhesion. The formation of the β-catenin/T-cell factor (Tcf) complex in the cell nucleus is the penultimate step of the Wnt/β-catenin signaling pathway. The aberrant formation of this protein–protein interaction (PPI) complex has been recognized as a key driving force for many cancers including TNBCs. Since β-catenin was reported in 1992, significant interest has been paid to screen compound libraries to discover small-molecule inhibitors that can bind β-catenin and disrupt the β-catenin/Tcf interaction. Many high-throughput screening campaigns were conducted, but little to no success was obtained. None of the reported compounds were able to deliver any drug candidates to preclinical and clinical trials. We decided to take a rational drug design approach to design new inhibitors based on the electronic properties of the key structural features for β-catenin/Tcf recognition. Our crystallographic and biochemical analyses revealed that the Tcf4 G13ANDE17 binding site of β-catenin could be targeted to design potent small-molecule inhibitors selective for the β-catenin/Tcf interaction. In combination of peptidomimetic strategy, structure-based drug design, and chemical synthesis, and biochemical and cell-based characterizations, we successfully designed and synthesized potent and selective small-molecule inhibitors for the β-catenin/Tcf interaction. The most potent inhibitor exhibited submicromolar inhibitory potency for disruption of the β-catenin/Tcf interaction. This potent inhibitor also exhibited dozens to hundreds folds of selectivities for the β-catenin/Tcf over the β-catenin/E-cadherin and β-catenin/APC interactions. The binding mode of new inhibitors was characterized by the site-directed mutagenesis and structure-activity relationship studies. The cell-based studies demonstrated that new inhibitors passed the cell membrane, significantly attenuated Wnt/β-catenin signaling in TNBC cells, and suppressed growth of Wnt/β-catenin-dependent TNBC cells. These inhibitors also exhibited cell-based selectivities for the β-catenin/Tcf over β-catenin/cadherin and β-catenin/adenomatous polyposis coli (APC) interactions. We have attempted to improve the quality and reduce the surface defect density of thick epilayers. The surface defect density is obviously reduced from 3.46 cm−2 to 0.85 cm−2, owing to the addition of the etching process. The effect of the etching process on the surface defect is discussed based on the measured results. The results demonstrate that the fast epitaxial growth system can grow ultra thick 4H-SiC epilayers with high quality that satisfies the requirement of ultra-high-voltage devices.

45. Study on the Optimized Energy Resolution of Scintillator Detectors Based on SiPMs and LYSO:Ce

Abstract: The cerium-doped lutetium yttrium silicate (LYSO:Ce) crystal has many advantages such as high light output and fast decay time, showing great potential to improve the performance of scintillation detectors. Silicon photomultipliers (SiPMs) are high-performance semiconductor photodetectors, which generally have a higher photon detection efficiency than photomultiplier tubes (PMTs) and are beneficial to realize a better energy resolution. The energy resolution and detected light output of SiPM-coupled LYSO:Ce crystals were studied by optimizing the photon detection efficiency (PDE), the operating voltage, the wrapper, the surface finish and the coupling method between the SiPM and crystal. Considering the output saturation of SiPMs in high light intensity, a preliminary correction method was applied to the saturation response of SiPMs and energy resolution of 662 keV gamma-rays from 137Cs was obtained after correction. The results show that the optimized energy resolution at 662 keV measured by the SiPM of 50 μm microcells can reach 7.6%. The light output of the polished and rough crystal detected by a SiPM of 10 μm microcells were also measured, which can reach 27000 ± 3200 photons/MeV and 36000 ± 3700 photons/MeV, respectively.

46. Piled-up neutron-gamma discrimination system for CLLB using convolutional neural network

Abstract: A piled-up neutron-gamma discrimination system is designed to discriminate single and piled-up events under high counting rate. The data acquired by a Cs2LiLaBr6:Ce (CLLB) detector and an Am-Be neutron source are used to train and test the model in the n-γ discrimination system. The charge comparison method is applied to discriminate the non-piled-up events in the experimental data and label the dataset of single events. As a result of the method, the figure-of-merit (FOM) value is 1.10, which indicates that the wrong labeling ratio is about 0.248%. A dataset of piled-up events is created by adding up waveforms and labels of the events in the single-pulse dataset. The discrimination system consists of three convolutional models, called Model_PulseNum, Model_OnePulse and Model_TwoPulses. All the models are trained and tested by the created dataset. Model_PulseNum is created and trained to define the number of pulses in the waveform of the event, with an accuracy of 99.94%. The other two models (Model_OnePulse and Model_TwoPulses) are created and trained to discriminate the particle types for non-piled-up and two-fold piled-up events with the accuracy of 99.5% and 98.6%, respectively. For the whole discrimination system, the accurcy for the particle identification is over 97% for each class (γ, n, γ + γ, γ + n, n + γ and n + n). These results indicate that CNN model can improve the performance of particle detection systems by effectively discriminate neutron and gamma for both piled-up and non-piled-up events under high counting rates.

47. Retron Editing for Precise Genome Editing without Exogenous Donor DNA in Human Cells

Abstract: CRISPR-Cas9 mediated seamless genome editing can be achieved by incorporating donor DNA into the CRISPR-Cas9 target loci via homology-directed repair (HDR), albeit with relative low efficiency due to the inefficient delivery of exogenous DNA. Retrons are bacterial genetic element composed of a non-coding RNA (ncRNA) and reverse transcriptase (RT). Retrons coupled with CRISPR-Cas9 have been shown to enhance precise genome editing via HDR in yeast through fusing guide RNA (gRNA) to the 3’ end of retron ncRNA, producing multicopy single-stranded DNA (msDNA) covalently tethered to gRNA. Here, we further engineered retrons by fusing Cas9 with E.coli RT from different clades and joining gRNA at the 5’ end of retron ncRNA, and found that retron editing can achieve precise genome editing efficiently in human cells. By co-expression of Cas9-RT fusions and retron-ncRNA gRNA (rgRNA) in HEK293T cells, we demonstrated the rates of retron editing at endogenous genomic loci was up to 10 %. We expect our retron editing system could aid in advancing the ex vivo and in vivo therapeutic applications of retron.

48. Endothelial SIRPα signaling controls thymic progenitor homing for T cell regeneration and antitumor immunity

Abstract: Thymic homing of hematopoietic progenitor cells (HPCs) is an essential step for the subsequent T cell development. Previously we have identified a subset of specialized thymic portal endothelial cells (TPECs), which is important for thymic HPC homing. However, the underlying molecular mechanism remains still unknown. Here we found that signal regulatory protein alpha (SIRPα) is preferentially expressed on TPECs. Disruption of CD47-SIRPα signaling in mice resulted in reduced number of thymic early T cell progenitors (ETPs) and impaired thymic HPC homing. Mechanistically, SIRPα-deficient ECs and CD47-deficient lymphocytes demonstrated impaired transendothelial migration (TEM). Specifically, SIRPα intracellular ITIM motif-initiated downstream signaling in ECs was found to be required for TEM in a SHP2- and Src-dependent manner. Furthermore, CD47-signaling from migrating cells and SIRPα intracellular signaling were found to be required for VE-cadherin endocytosis in ECs. Functionally, SIRPα signaling is required for T cell regeneration upon sub-lethal total body irradiation (SL-TBI); CD47-SIRPα signaling blockade post SL-TBI diminishes antitumor immunity. Thus, our study reveals a novel role of endothelial SIRPα signaling for thymic HPC homing for T cell regeneration and antitumor immunity.Graphic abstractThymic homing of hematopoietic progenitor cells is fundamental to the T cell-based adaptive immunity, yet the molecular basis of this process is less clear.
