Autophagy plays a critical role in the physiology and pathophysiology of hepatocytes. High level of homocysteine (Hcy) promotes autophagy in hepatocytes, but the underlying mechanism is still unknown. Here, we investigate the relationship between Hcy-induced autophagy level and the expression of nuclear transcription factor EB (TFEB). The results show that Hcy-induced autophagy level is mediated by upregulation of TFEB. Silencing of TFEB decreases the level of autophagy-related protein LC3BII/I and increases p62 expression level in hepatocytes after exposure to Hcy. Moreover, the effect of Hcy on the expression of TFEB is regulated by hypomethylation of the TFEB promoter catalyzed by DNA methyltransferase 3b (DNMT3b). In summary, this study shows that Hcy can activate autophagy by inhibiting DNMT3b-mediated DNA methylation and upregulating TFEB expression. These findings provide another new mechanism for Hcy-induced autophagy in hepatocytes.

In order to clarify the stress situation of buried elbow in a certain sulfur oil and gas field, by analyzing the stress factors of a buried elbow in this oil field, the stress parameters affecting the elbow are simulated and calculated.Results showed that the stress at the elbow increased with the increase of the operating temperature, and it presented approximately linear relationship.Under operating conditions with higher operating temperature, the stress at the elbow decreased as the operating pressure increased.Under operating conditions with lower operating temperature, the stress at the elbow increased with increasing operating pressure.Moreover, the stress at the elbow increased first and then decreased as the angle of the elbow increased.According to the change rule of the influence of various factors on the stress of the elbow, the influence factors of the comprehensive parameters of stress evaluation at the elbow were determined, and the stress risk of the elbow in the field engineering was quickly investigated.

Hypoxic–ischemic brain damage (HIBD) is a major cause of brain injury in neonates. Bone marrow mesenchymal stem cells (BMSCs) show therapeutic potential for HIBD, and genetic modification may enhance their neuroprotective effects. The goal of this study was to investigate the neuroprotective effects of hepatocyte growth factor (HGF)-overexpressing BMSCs (BMSCs-HGF) against HIBD and their underlying mechanisms.

Objective To explore the psychological experience and needs of female caregivers of stroke patients in rehabilitation period. Methods The phenomenological research method was used to conduct semi-structured interviews with 12 female caregivers of stroke patients. Results Based on the real experience of female caregivers during stroke rehabilitation period, four themes were extracted, including imbalance in physical and mental health, coping with multiple roles, insufficient of nursing information acquisition and lack of social support. Conclusion The feelings of female caregivers of patients in stroke rehabilitation stage are multi-dimensional, suggesting that we should pay attention to the mental health of female caregivers and guide them to seek positive coping styles, so as to provide better care for the rehabilitation of stroke patients. (目的 探讨脑卒中康复期患者的女性照顾者的照顾心理体验及需求。方法 采用现象学研究方法, 对12例脑卒中康复期患者的女性照顾者进行半结构式访谈, 采用质性研究的类属法进行整理和分析。结果 根据脑卒中康复期女性照顾者的真实体验提炼出4个主题: 身心健康失衡、多重角色应对失调、护理信息获取不足、社会支持缺乏。结论 脑卒中康复期患者的女性照顾者的感受呈现多维度, 提示应关注女性照顾者的心理健康, 引导其寻求积极的应对方式, 进而为脑卒中患者的康复提供更好的照护。)

In this study, the potato whole flour was added to wheat flour with wheat gluten was auxiliary added to make potato noodle. Three groups of the potato powder noodles with different total flour content were studied from three levels: texture characteristics, microstructure and physicochemical properties of noodles. It was found that the potato powder had a certain effect on the mechanical properties of dough, the formation and structure of gluten network, and the cooking parameters and appearance of noodles. The results showed that the overall index was the best when the total amount of the potato powder was 20% (g/g)and the amount of gluten was 0.03% (g/g). It was also found that the effect of the potato powder on dough was bidirectional. The effect of the potato starch on the dough was negative, but the polysaccharides and protein in the potato powder may have a positive effect on the dough and gluten network. At the same time,the addition of wheat gluten can improve the stability of dough and the quality of noodles.

[Purpose/Significance] Basic academic libraries play an important role in popularizing and promoting digital humanities (DH) in the whole ecosystem. The analysis of their development space is helpful to the construction and development of the digital humanistic ecosystem. [Method/Process] A semi-structured interview was conducted with 13 humanities scholars and 4 library managers to find out their awareness, understanding and demand of DH and their views on the position of basic academic libraries in the digital humanistic ecosystem. On this basis, this paper probes into the current situation and development strategies of DH in basic academic libraries. [Results/Conclusions] The development of DH in basic academic libraries has received less attention and does not match the huge demand for DH due to the shortage of funds, resources and technologies, the access of digital human resources is basically dependent on external open channels, and the digital and human foundations of digital human development are weak. In these cases, this paper puts forward three development strategies: strengthening the cooperation among basic academic libraries, strengthening the construction of quality cultivation systems of DH, and winning the support of the state and relevant institutions for DH services.

The giant panda (GP) was the most endangered species in China, and gut microbiota plays a vital role in host health. To determine the differences of the gut microbiota among the male, female and pregnant GPs, a comparative analysis of gut microbiota in GPs was carried out by 16S rRNA and ITS high-throughput sequencing. In 16S rRNA sequencing, 435 OTUs, 17 phyla and 182 genera were totally detected. Firmicutes (53.6%) was the predominant phylum followed by Proteobacteria (37.8%) and Fusobacteria (7.1%). Escherichia/Shigella (35.9%) was the most prevalent genus followed by Streptococcus (25.9%) and Clostridium (11.1%). In ITS sequencing, 920 OTUs, 6 phyla and 322 genera were also detected. Ascomycota (71.3%) was the predominant phylum followed by Basidiomycota (28.4%) and Zygomycota (0.15%). Purpureocillium (4.4%) was the most prevalent genus followed by Cladosporium (2.5%) and Pezicula (2.4%). Comparative analysis indicated that the male GPs harbor a higher abundance of phylum Firmicutes than female GPs with the contribution from genus Streptococcus. Meanwhile, the female GPs harbor a higher abundance of phylum Proteobacteria than male GPs with the contribution from genus Escherichia/ Shigella. In addition, the shift in bacteria from female to pregnant GPs indicated that phylum Firmicutes increased significantly with the contribution from Clostridium in the gut, which may provide an opportunity to study possible associations with low reproduction of the GPs.

As a fuel cladding materials of the nuclear reactor, water side corrosion and hydrogen absorption generated in zirconium alloys which due to the embrittlement of matrix and hydrogen induced delayed cracking. The hydride phases, hydride orientation and the effect of hydride orientation on mechanical properties of zirconium alloys were reviewed. The crystallographic orientation relationship between α-Zr matrix and hydrides(γ and δ), the effect of texture, Q value, stress and annealing temperature on hydride orientation, the effect of hydride orientation on mechanical properties of zirconium alloy were primarily discussed. Meanwhile, the current problems were analyzed, for example, the mechanism of hydride orientation was not clear, the mechanism of hydride stress reorientation was controversial, etc. It was pointed out that the hydride orientation, crystallographic orientation relationship between hydrides and Zr matrix, hydride stress reorientation in Zr alloys require further investigation. Furthermore, the research on the hydrides in domestic new type high performance Zr alloy needs to be carried out.

Tau pathogenesis is a hallmark of many neurodegenerative diseases, including Alzheimers disease (AD). Although the events leading to initial tau misfolding and subsequent tau spreading in patient brains are largely unknown, traumatic brain injury (TBI) may be a risk factor for tau-mediated neurodegeneration. Using a repetitive TBI (rTBI) paradigm, we report that rTBI induced somatic accumulation of phosphorylated and misfolded tau, as well as neurodegeneration across multiple brain areas in 7-month-old tau transgenic PS19 mice but not wild-type (WT) mice. rTBI accelerated somatic tau pathology in younger PS19 mice and WT mice only after inoculation with tau preformed fibrils and AD brain-derived pathological tau (AD-tau), respectively, suggesting that tau seeds are needed for rTBI-induced somatic tau pathology. rTBI further disrupted axonal microtubules and induced punctate tau and TAR DNA binding protein 43 (TDP-43) pathology in the optic tracts of WT mice. These changes in the optic tract were associated with a decline of visual function. Treatment with a brain-penetrant microtubule-stabilizing molecule reduced rTBI-induced tau, TDP-43 pathogenesis, and neurodegeneration in the optic tract as well as visual dysfunction. Treatment with the microtubule stabilizer also alleviated rTBI-induced tau pathology in the cortices of AD-tau-inoculated WT mice. These results indicate that rTBI facilitates abnormal microtubule organization, pathological tau formation, and neurodegeneration and suggest microtubule stabilization as a potential therapeutic avenue for TBI-induced neurodegeneration.

This qualitative study explored perspectives of college students with learning disabilities (LD) regarding their self-determination and parental influences on student self-determination during students' transition from high school to postsecondary education. Data were collected from in-depth interviews with 12 college students with LD. Qualitative analyses were conducted to examine students' participation in secondary transition, self-determination behaviors, and parental influences on students' self-determination. The findings identified five important self-determination behaviors (i.e., postsecondary education goal setting and attainment, self-advocacy, being autonomous, self-awareness, and decision-making) students demonstrated in secondary transition. Four themes pertain to parental influences on student self-determination, including encouraging students to be advocates, fostering students' academic competency, showing moral support and high expectations, and providing opportunities for students to make decisions. Recommendations for engaging parents to promote student self-determination are provided.

BackgroundViral tracers are important tools for mapping brain connectomes. The feature of predominant anterograde transneuronal transmission offers herpes simplex virus-1 (HSV-1) strain H129 (HSV1-H129) as a promising candidate to be developed as anterograde viral tracers. In our earlier studies, we developed H129-derived anterograde polysynaptic tracers and TK deficient (H129-dTK) monosynaptic tracers. However, their broad application is limited by some intrinsic drawbacks of the H129-dTK tracers, such as low labeling intensity due to TK deficiency and potential retrograde labeling caused by axon terminal invasion. The glycoprotein K (gK) of HSV-1 plays important roles in virus entry, egress, and virus-induced cell fusion. Its deficiency severely disables virus egress and spread, while only slightly limits viral genome replication and expression of viral proteins. Therefore, we created a novel H129-derived anterograde monosynaptic tracer (H129-dgK) by targeting gK, which overcomes the limitations of H129-dTK.MethodsUsing our established platform and pipeline for developing viral tracers, we generated a novel tracer by deleting the gK gene from the H129-G4. The gK-deleted virus (H129-dgK-G4) was reconstituted and propagated in the Vero cell expressing wildtype H129 gK (gKwt) or the mutant gK (gKmut, A40V, C82S, M223I, L224V, V309M), respectively. Then the obtained viral tracers of gKmut pseudotyped and gKwt coated H129-dgK-G4 were tested in vitro and in vivo to characterize their tracing properties.ResultsH129-dgK-G4 expresses high levels of fluorescent proteins, eliminating the requirement of immunostaining for imaging detection. Compared to the TK deficient monosynaptic tracer H129-dTK-G4, H129-dgK-G4 labeled neurons with 1.76-fold stronger fluorescence intensity, and visualized 2.00-fold more postsynaptic neurons in the downstream brain regions. gKmut pseudotyping leads to a 77% decrease in retrograde labeling by reducing axon terminal invasion, and thus dramatically improves the anterograde-specific tracing of H129-dgK-G4. In addition, assisted by the AAV helper trans-complementarily expressing gKwt, H129-dgK-G4 allows for mapping monosynaptic connections and quantifying the circuit connectivity difference in the Alzheimer's disease and control mouse brains.ConclusionsgKmut pseudotyped H129-dgK-G4, a novel anterograde monosynaptic tracer, overcomes the limitations of H129-dTK tracers, and demonstrates desirable features of strong labeling intensity, high tracing efficiency, and improved anterograde specificity.
