Patients with familial hypercholesterolemia usually present with high levels of serum low-density lipoprotein, xanthomas and early coronary artery disease. A 13 years old female patient was admitted to Children's Hospital of Chongqing Medical University presenting symptoms of heart failure. Laboratory tests showed that her cholesterol and low-density lipid levels were extremely high. Electrocardiogram test revealed that she had sinus tachycardia, QT lengthening and ST-T change. Multiple cardiac function abnormalities were diagnosed by echocardiogram. Multiple coronary artery stenosis was determined by computed tomography angiography. After the combination of lipid lowering, anti-thrombosis, and cardiac remodeling therapies, the patient's symptoms were significantly improved and the patient was discharged. [ABSTRACT FROM AUTHOR]

Objective To analyze the role of peroxisome proliferator-activated receptors ( PPAR- y ) expression in P. acnes-induced pyroptosis, cell proliferation and apoptosis in human keratinocytes (HaCaT) so as to provide a new theoretical basis for clinical treatment. Methods Expression patterns of PPAR- y in acne and normal skin tissues were evaluated using IHC. By lentiviral transfection, PPAR-y was upregulated in HaCaT cells, and the expression of PPAR-y was verified by Western blot. ELISA was performed to analyze the levels of P. acnes-induced pyroptotic-associated inflammatory factors IL-1p and IL-18. EdU assay and flow cytometry were used to detect the cell proliferation and apoptosis ability. Western blot was used to determine the levels of NLR family pyrin domain containing 3 (NLRP3), cysteinyl aspartate specific proteinase-1 (caspase-1), Gasdermin D (GSDMD) and PPAR-y expression. Results PPAR-y expression was significantly decreased in acne (P < 0.05). Compared with the control, the P. acnes-induced pyroptotic-associated inflammatory factors IL-ip and IL-18 were increased ( P < 0.01 ), the pyroptotic-associated proteins were increased significantly ( P < 0.05 ) . After treatment with P. acnes, the up-regulated PPAR-y reduced the expression of IL-ip and IL-18 and HaCaT cell apoptosis, promoted the cell proliferation, up-regulated the expression of NLRP3, Caspase-1 and GSDMD in human keratinocytes all in a statistically significant way (all P < 0.05). The combined method with high oxygen filling under low temperature and waterless condition is suggested to be a more effective strategy to keep fish alive during turbot transportation. To investigate the biological mechanism of turbot, when turbot was subjected to waterless treatment under different oxygen conditions (water, air and oxygen), the global expression profiling of the gill tissues was performed by transcriptome analysis using Illumina HiSeq 4000 platform. The results showed that a total of 104,886 unigenes were detected, and differential expression analysis revealed 662 differentially expressed genes (322 upregulated (fold >2) and 340 downregulated (fold <0.5)) in the Air-group, and 244 up-regulated and 197 down-regulated genes in the Oxygen-group in comparison with the control (in water at 2 °C), and 354 up-regulated and 212 down-regulated genes in the Oxygen-group compared to the Air-group were identified. In addition, the reliability of the sequencing results was confirmed using qPCR, the fold change of 6 randomly selected genes were consistent with the results of the sequencing. Moreover, gene ontology analysis was conducted, which indicated that the air environment with waterless has a greater impact on the metabolism of fish compared with the water environment. It was supposed that turbot may regulate their metabolism by increasing the expression of genes involved in transcription and promoting the oxidative stress response in order to maintain homoeostasis. Furthermore, GO and Kyoto Encyclopedia of Genes and Genomes pathway enrichment analysis showed that the differentially expressed genes were enriched in lyase activity, immune response, cytokine activity and nitrogen metabolism, revealing that oxygen availability influences above metabolic activities in response to waterless conditions.
