12. Expert consensus on the construction of surveillance pathways and systems for vector-borne tropical diseases.

Abstract

With the growth of the global economy, changes in climate and ecological environments, and increased mobility of humans and animals, the transmission risk of vector-borne tropical diseases continues to rise. To address this challenge, strengthening surveillance of vector-borne tropical diseases is urgent. This consensus brought together 29 renowned experts in related professional fields from 26 institutions in China, who, through analyzing the epidemic trend and hazard situation of vector-borne tropical diseases and summarizing the working experiences of experts, have firstly reached following consensus: the burden of vector-borne tropical diseases is heavy with great threats to human health; China has achieved remarkable results in prevention and control of vector-borne tropical diseases, but still needs to strengthen the surveillance and response actively. Secondly, a unanimous consensus has been reached on the aspects of surveillance definition, objectives, contents, and methods of vector-borne tropical diseases. Thirdly, detail requirements have been agreed including: strengthening the concept of early surveillance and forecast, standarding the function, evaluation steps, and construction requirements of surveillance system for vector-borne tropical diseases. Fourthly, key tasks were put forward that need to be investigated and strengthened in the future. 13. Vector Biology and Integrated Management of Malaria Vectors in China.

Abstract

Malaria is an infectious disease caused by Plasmodium parasites, transmitted by Anopheles sinensis, Anopheles lesteri, Anopheles minimus, and Anopheles dirus in China. In 2021, the disease was eliminated in China after more than 70 years of efforts implementing an integrated mosquito management strategy. This strategy comprised indoor residual spray, insecticide-treated bed nets, irrigation management, and rice–fish coculture based on an understanding of taxonomic status and ecological behaviors of vector species, in conjunction with mass drug administration and promotion of public education. However, China still faces postelimination challenges, including the importation of approximately 2,000–4,000 cases of malaria into the country each year, as well as widespread resistance to pyrethroid insecticides in An. sinensis; these challenges require long-term vector surveillance to understand the distribution, population density, and development of resistance in vector mosquitoes to prevent local epidemics caused by imported malaria cases. Abstract

Globally distributed mosquitoes play an important role in emerging and resurging infectious diseases in humans. The critical factor affecting vector competence is susceptibility of the vector to viruses, and the ability of midgut epithelial cells to resist viral infection is the main factor determining the susceptibility of mosquitoes to arbovirus infections and is a key index of vector competence. This study predicted the novel siRNA and piRNA sequences, and then attempted to identify the small RNAs derived from viruses through alignment with the DENV‐2 genome. Our results show that most of the novel piRNAs were down regulated post‐infection, but that the majority of novel siRNAs were upregulated after infection. Interestingly, there were no vsiRNA and vpiRNA sequences that had a high expression level and/or significant differential expression; moreover, all vpiRNAs and vsiRNAs were upregulated after infection. The novel_pirs, novel_sirs, vpiRNAs and vsiRNAs examined in this study play a role in resistance to viral infection. [ABSTRACT FROM AUTHOR]

Abstract

The Japanese encephalitis virus (JEV), a mosquito-borne zoonotic pathogen, is the major cause of viral encephalitis worldwide. An investigation of mosquito species diversity, JEV infection rate, and seasonal population fluctuations of Culex tritaeniorhynchus in Huaihua County, Hunan Province, China, revealed the distribution of vector mosquito populations and genotypes and molecular characteristics of current, common JEV strains in this region. Research on mosquito species diversity in different habitats in Huaihua revealed that local community composition was relatively simple, including five species from four genera (two Culex spp., one Anopheles sp., one Aedes sp., and one Armigeres sp.). Cx. tritaeniorhynchus was clearly the dominant species comprising 94.2–98.6% of all specimens and was always the most common species captured in paddy fields, pigpens, and human dwellings. The seasonal abundance of Cx. tritaeniorhynchus was relatively even, with a single seasonal peak in late August. Two Huaihua JEV strains isolated from the mosquito and pig were highly congruent. The genetic affinities were determined by analyzing capsid/premembrane (C/PrM) and envelope (E) gene variation. The results showed that they were of genotype III and most closely related to the live, attenuated vaccine strains SAl4-14-2 and SA14 and JaGAr01. The Huaihua E protein shares high similarity (mosquito 98.8% and pig 97.6%) at the nucleotide level with the SA14-14-2 vaccine. Although we found that the E gene sequences of the Huaihua JEV mosquito strain and pig strain have 11 and 15 amino acid site substitutions compared with the SAl4-14-2 vaccine, key sites that associated with JEV's antigenic activity and neurovirulence were unchanged.
