Ecological security patterns (ESPs) provide a solution for balancing the conflicts between ecological conservation and socio-economic development and maintaining sustainable regional development. Identifying sustainable ESPs should be a matter given attention to in order to meet future urban expansion needs. Based on multi-source eco-environmental datasets and remote sensing (RS) containing the history of land-use patterns, our study incorporated future land-use patterns into the ESP identification framework to reduce the potential impact of future urban expansion on the identified ESPs (i.e., ecological corridors and nodes) and to reserve space for future urban development from an urban development perspective. Considering the Changsha–Zhuzhou–Xiangtan (CZX) urban agglomeration, the results show that 57 corridors are extracted, within which 36 key corridors are mainly distributed in the Lukou and Wangcheng districts and Xiangtan and Changsha counties; an additional 21 potential corridors are mainly distributed in the urban built-up area and these corridors connect to Xiangjiang River or its tributary. In addition, 18 pinch points were identified, mainly located adjacent to the Xiangjiang River, Liuyang River, and Laodao River; 9 barrier points are distributed at the junctions of the main roads. A comparison of ESP identification with and without simulated land-use patterns showed the incorporation of future land-use patterns favoured sustainable ESP construction. Furthermore, we discuss the rationality and application of ESP identification from an urban development perspective and emphasize the trade-off between future land-use patterns and ecological conservation, better reflecting the forward-looking thinking of spatial planning.

This paper focuses on the distributed model predictive control for multi-agent systems subject to packet loss and actuator saturation. The multi-agent systems, which can be decoupled into several subsystems, have a coupled global cost function. By decoupling the global cost function, the distributed model predictive control for multi-agent systems is approximately cast into centralized model predictive control for each agent with a cost function including the states of the neighboring agents. The exchange of states between adjacent agents realized by communication network may subject to packet loss which is assumed to obey a Bernoulli distribution with probability being known. Actuator saturation is also considered and dealt with by the Nth-step set invariance approach. To ensure the multi-agent systems is asymptotically stable, a compatibility condition is provided. The problem of controller design for agent is converted into a Linear matrix inequality optimization problem involving compatibility constraint. Furthermore, a control algorithm is obtained to ensure the asymptotic stability of the global closed-loop system as well as the recursive feasibility of the optimization problem. Finally according to a simulation example, it is concluded that the presented method is effective.

CSA inhibits Wnt/β-catenin signaling activity and GICs self-renewal

Clinicopathological characteristics

Purpose: Glioma-initiating cells (GIC) are glioma stem–like cells that contribute to glioblastoma (GBM) development, recurrence, and resistance to chemotherapy and radiotherapy. They have recently become the focus of novel treatment strategies. Cyclophilin A (CypA) is a cytosolic protein that belongs to the peptidyl–prolyl isomerase (PPIase) family and the major intracellular target of the immunosuppressive drug cyclosporin A (CsA). In this study, we investigate the functions of CypA and its mechanism of action in GICs' development.Experimental Design: We analyzed differences in CypA expression between primary tumors and neurospheres from the GDS database, both before and after GIC differentiation. A series of experiments was conducted to investigate the role of CypA in GIC stemness, self-renewal, proliferation, radiotherapy resistance, and mechanism. We then designed glutathione S-transferase (GST) pulldown and coimmunoprecipitation assays to detect signaling activity.Results: In this study, we demonstrated that CypA promotes GIC stemness, self-renewal, proliferation, and radiotherapy resistance. Mechanistically, we found that CypA binds β-catenin and is recruited to Wnt target gene promoters. By increasing the interaction between β-catenin and TCF4, CypA enhances transcriptional activity.Conclusions: Our results demonstrate that CypA enhances GIC stemness, self-renewal, and radioresistance through Wnt/β-catenin signaling. Due to its promotive effects on GICs, CypA is a potential target for future glioma therapy. Clin Cancer Res; 23(21); 6640–9. ©2017 AACR.

Hippocampal sclerosis (HS) is one of the most prevalent pathological types of temporal lobe epilepsy (TLE), and it significantly affects patient prognoses. The methylation of DNA plays an important role in the development of epilepsy. However, few studies have focused on HS subtypes to determine DNA methylation profiles in TLE. This study aimed to determine the pathogenesis of TLE from an epigenetic perspective in patients with TLE-HS type I (TLE-HSTI) and TLE without HS (TLE-nHS) using whole-genome bisulfite sequencing (WGBS). We defined 1171 hypermethylated and 2537 hypomethylated regions and found 632 differentially methylated genes (DMG) in the promoter region that were primarily involved in the regulation of various aspects of epilepsy development. Twelve DMG overlapped with differentially expressed genes (DEG) in the promoter region, and RT-qPCR findings revealed significant overexpression of the SBNO2, CBX3, RASAL3, and TMBIM4 genes in TLE-HSTI. We present the first systematic analysis of methylation profiles of TLE-HSTI and TLE-nHS from an epigenetic perspective using WGBS. Overall, our preliminary data highlight the underlying mechanism of TLE-HSTI, providing a new perspective for guiding treatment of TLE.

Background: Although many biomarkers have been reported for detecting glioma, the prognosis for the disease remains poor, and therefore, new biomarkers need to be identified. GNG5, which is part of the G-protein family, has been associated with different malignant tumors, though the role of GNG5 in glioma has not been studied. Therefore, we aimed to identify the relationship between GNG5 and glioma prognosis and identify a new biomarker for the diagnosis and treatment of gliomas.Methods: We used data on more than a thousand gliomas from multiple databases and clinical data to determine the expression of GNG5 in glioma. Based on clinical data and CGGA database, we identified the correlation between GNG5 and multiple molecular and clinical features and prognosis using various analytical methods. Co-expression analysis and GSEA were performed to detect GNG5-related genes in glioma and possible signaling pathways involved. ESTIMATE, ssGSEA, and TIMER were used to detect the relationship between GNG5 and the immune microenvironment. Functional experiments were performed to explore the function of GNG5 in glioma cells.Results: GNG5 is highly expressed in gliomas, and its expression level is positively correlated with pathological grade, histological type, age, and tumor recurrence and negatively correlated with isocitrate dehydrogenase mutation, 1p/19 co-deletion, and chemotherapy. Moreover, GNG5 as an independent risk factor was negatively correlated with the overall survival time. GSEA revealed the potential signaling pathways involved in GNG5 function in gliomas, including cell adhesion molecules signaling pathway. The ssGSEA, ESTIMATE, and TIMER based analysis indicated a correlation between GNG5 expression and various immune cells in glioma. In vivo and in vitro experiments showed that GNG5 could participate in glioma cell proliferation and migration.Conclusions: Based on the large data platform and the use of different databases to corroborate results obtained using various datasets, as well as in vitro and in vivo experiments, our study reveals for the first time that GNG5, as an oncogene, is overexpressed in gliomas and can inhibit the proliferation and migration of glioma cells and lead to poor prognosis of patients. Thus, GNG5 is a potential novel biomarker for the clinical diagnosis and treatment of gliomas.

Background XRCC2, a homologous recombination-related gene, has been reported to be associated with a variety of cancers. However, its role in glioma has not been reported. This study aimed to find out the role of XRCC2 in glioma and reveal in which glioma-specific biological processes is XRCC2 involved based on thousands of glioma samples, thereby, providing a new perspective in the treatment and prognostic evaluation of glioma. Methods The expression characteristics of XRCC2 in thousands of glioma samples from CGGA and TCGA databases were comprehensively analyzed. Wilcox or Kruskal test was used to analyze the expression pattern of XRCC2 in gliomas with different clinical and molecular features. The effect of XRCC2 on the prognosis of glioma patients was explored by Kaplan–Meier and Cox regression. Gene set enrichment analysis (GSEA) revealed the possible cellular mechanisms involved in XRCC2 in glioma. Connectivity map (CMap) was used to screen small molecule drugs targeting XRCC2 and the expression levels of XRCC2 were verified in glioma cells and tissues by RT-qPCR and immunohistochemical staining. Results We found the overexpression of XRCC2 in glioma. Moreover, the overexpressed XRCC2 was associated with a variety of clinical features related to prognosis. Cox and meta-analyses showed that XRCC2 is an independent risk factor for the poor prognosis of glioma. Furthermore, the results of GSEA indicated that overexpressed XRCC2 could promote malignant progression through involved signaling pathways, such as in the cell cycle. Finally, doxazosin, quinostatin, canavanine, and chrysin were identified to exert anti-glioma effects by targeting XRCC2. Conclusions This study analyzed the expression pattern of XRCC2 in gliomas and its relationship with prognosis using multiple datasets. This is the first study to show that XRCC2, a novel oncogene, is significantly overexpressed in glioma and can lead to poor prognosis in glioma patients. XRCC2 could serve as a new biomarker for glioma diagnosis, treatment, and prognosis evaluation, thus bringing new insight into the management of glioma.

We introduce a novel dimension to the fleet renewal decisions of a shipping company, taking into account the effects of ship scrapping subsidies. The fleet renewal behavior of a company is analyzed with focus on early scrapping decisions prompted by the subsidy. We present a fleet renewal decision model which maximizes firm profit given market demand and the features of a scrapping subsidy. To assess the effects of a potential subsidy on corporate decisions, we compare the optimal fleet renewal schemes with and without the subsidy. We calculate the outcome of such a policy measure for China COSCO Shipping.

Temporal lobe epilepsy (TLE) is associated with neurodegeneration, often leading to hippocampal sclerosis (HS). Type 1 HS, which is characterized by severe neuronal loss and gliosis predominantly in regions CA1 and CA4, is the most common subtype and is associated with the best prognosis according to the ILAE classification system. MiRNAs participate in the biological processes underlying many nervous system diseases, including epilepsy. However, the miRNA expression profile of HS ILAE type 1 is not completely understood. A total of 14 patients were identified as having the ILAE subtype, as determined by NeuN immunohistochemistry (ILAE type 1 = 7; no-HS = 7). Next-generation sequencing and reverse transcription polymerase chain reaction technology were used to validate the dysregulated miRNAs. Bioinformatics analysis of the predicted target genes was conducted using Gene Ontology and Kyoto Encyclopedia of Genes and Genomes analyses. In total, 1643 mature miRNAs were detected in this study, along with 5 miRNAs that were upregulated and 2 miRNAs that were downregulated in the type 1 group. Bioinformatics analysis showed that 1545 target genes were predicted using the miRDB and Targetscan databases and that these predicted genes showed enrichment in pathways associated with nucleic acid binding, intracellular and cellular macromolecule metabolic processes, and the PI3K-Akt signaling pathway. This study is the first to report the miRNA expression profile of HS ILAE type 1 compared with those of no-HS. These results provide new insights into the neuronal loss pathology of type 1 HS.

Background: Stem cell transplantation maybe an advantaged method for curing spinal cord injury (SCI). However, the adverse environment of the injured area makes the treatment effects on transplantation unsatisfactory. The aim of this study was to investigate whether AC-YVAD-CMK could change enviroment by inhibiting caspase-1-mediated imflammatory response to improve Bone marrow stromal cells (BMSCs) transplantation on neuroprotection after SCI.Methods: Firstly, BMSCs were prepared and cultured in vitro and the SCI rat model was constructed. Then, AC-YVAD-CMK and BMSCs were injected into the SCI model at the same time. Western blot and ELISA were used to detect the production of inflammatory factors and neurotrophic factors in the injured area, meanwhile immunohistochemistry was employed to clarify the effect of AC-YVAD-CMK on BMSCs transplantation. Finally, we evaluated the recovery of limb motor function by BBB score in SCI rats.Result: In the AC-YVAD-CMK + BMSCs group, the active caspase-1, IL-18 and IL-1β expressed less than those in the BMSCs group or AC-YVAD-CMK group at 1 week after SCI (P < 0.01). Meanwhile, increasing brain derived neurotrophic factor (BDNF) and neurotrophin-3 (NT-3) levels, the PKH26 labeled BMSCs with some being neurofilament protein-200 (NF-200)-positive were still observed in the contusion site. Furthermore, the restoration of motor connections across the injury sites was positively correlated with the recovery of spinal cord function in cotreatment group (P < 0.01).Conclusions: These results show that caspase-1-mediated inflammatory response is inhibited by AC-YVAD-CMK to enhance BMSCs transplantation to improve neuroprotection after SCI.

The resource scheduling ability of hidden fault channel cloud test platform based on cache data replacement method is not good. A hidden fault channel cloud test platform resource access algorithm based on deep learning is proposed. The resource access and data mining structure model of hidden fault channel cloud test platform is constructed, the task code is distributed to the multi-path monitoring node by distributed code execution and KD tree data index, the two-dimensional KD tree is constructed to realize the path data monitoring, the cloud computing core technology is used to test the hidden fault channel cloud test platform, and the hidden fault channel cloud test platform is designed with the resource integration method. Improve the parallel processing ability of hidden fault channel cloud test platform. The simulation results show that the output stability and reliability of the hidden fault channel cloud test platform designed by this method are good, and the ability of hidden fault channel cloud testing is improved.

Objective Studies have shown that microRNAs play a role in the development of epilepsy by regulating downstream target messenger (m)RNA. The present study aims to determine the changes associated with microRNA-21-5p (miR-21-5p) during epileptogenesis in a kainic acid rat model, and to assess whether the PTEN-mTOR pathway is a target of miR-21-5p. Method Reverse transcription polymerase chain reaction (RT-PCR) was used to examine the quantitative expressions of miR-21-5p and PTEN, and Western blotting was used to test the activity of mTOR in the acute, latent, and chronic stages of epileptogenesis. The antagomir of miR-21-5p was injected into the intracerebroventricular space using a microsyringe. Neuronal death and epilepsy discharge were assessed by Nissl staining and electroencephalography (EEG), respectively. The Morris water maze (MWM) was used to assess the cognitive impairment in rats after status epilepticus (SE). Results Both miR-21-5p and mTOR were upregulated and PTEN was downregulated in rats during acute, latent, and chronic stages of epileptogenesis when compared with those of the control. After using antagomir miR-21-5p in vivo, miR-21-5p and mTOR decreased and the expression of PTEN increased compared with that in the SE model. The silencing of miR-21-5p diminished the number of abnormal spikes on EEG and decreased the number of neuron deletions on Nissl staining. The cognitive and memory impairment caused by epilepsy could also be improved after miR-21-5p knockdown in vivo. Conclusion The results of the present study demonstrate that PTEN-mTOR is the target of miR-21-5p in a kainic acid model of epilepsy. The knockout of miR-21-5p decreases the neuronal damage in stages of epileptogenesis. The miR-21-5p/PTEN/mTOR axis may be a potential target for preventing and treating seizures and epileptic damage.

In order to improve teaching effect of the “Java Design Patterns” course, the teaching process uses cases as the main line through the pattern concept, pattern structure diagram, pattern realization, application scenarios and the advantages and disadvantages of the pattern. This teaching method combines the orientation of application-oriented undergraduate, the characteristics of students’ knowledge structure and course’s abstract nature. In addition, this method combines traditional classrooms and MOOC platforms, and has achieved good results in actual teaching.

Shot blasting is an important means of metal surface treatment, and the fracture of rotating blade is the main reason for the abnormal operation of shot blasting machine. In order to prolong the service life of the equipment, the dynamic fatigue characteristics of the blade are studied in this paper. A three-dimensional model of the blade is established and its constraint modal are analyzed by both simulation and experiment. The results show that the deformation of blade is proportional with the increase of natural frequency, and the stress concentration is easy to occur at the root of blade. Meanwhile, the dynamic fatigue test of the blade is carried out, and it can be found that the root of the blade breaks at the rotational speed of 750r/min, which verifies the correctness of simulation results. Therefore, it is necessary to effectively adjust the control speed to ensure the normal rotation of the blades and reliable operation of the machine.

In this paper, ABAQUS finite element simulation software was used to establish a more reliable three-dimensional milling titanium alloy Ti6Al4V finite element simulation model. This model sets up important links such as the material constitutive model, the tool-chip contact friction model, and the chip separation criterion. And the accuracy of the model is verified by experiments. Based on this model, orthogonal experiment was used to explore the effects of axial cutting depth, tool speed and feed speed on the milling force. And the morphology of the chip and the surface morphology of the machined workpiece were investigated. The results show that the influence of cutting parameters on milling force from high to low is as follows: axial cutting depth > tool speed > feed rate. The change of cutting parameters has different influence on milling force in all directions, and has the largest influence on milling force in X direction.

Soybean (Glycine max [L.] Merrill) seeds are a major source of tocopherols (Toc), which could significantly improve immune system health of human and prevent or treat many serious diseases. Selection for higher Toc contents of seeds could increase nutritional value of soybean-derived food, laying on an important breeding goal for many soybean breeders. The present objectives of the work were to evaluate various genetic effects of QTL associated with individual and total Toc content based on a RIL population (“Beifeng 9” × “Freeborn”) in six environments to improve the efficiency of molecular marker-assisted selection (MAS) for high-Toc breeding. The results described that eighteen, thirteen, eleven and thirteen QTL were associated with α-Toc, γ-Toc, δ-Toc and total Toc content, respectively, and have additive main effects (a) and/or additive × environment interaction effects (ae) in certain environments. Among them, four QTL for α-Toc, two QTL for γ-Toc, one QTL for δ-Toc and four QTL for total Toc could increase α-Toc, γ-Toc, δ-Toc and total Toc content via significant a effect, respectively, which have stronger stability in different years and locations. It implied a value for MAS. Additionally, twenty-five, fifteen, eleven and twenty epistatic pairwise QTL associated with α-Toc, γ-Toc, δ-Toc and total Toc contents, respectively, were detected. The genetic information of the QTL effects obtained here would be beneficial for breeding soybean variety with high-Toc content by MAS.

Glioblastoma multiforme (GBM), one of the most aggressive human malignant brain tumors, is induced by multiple complex pathological mechanisms. The main cause of mortality in patients with GBM is the invasion-metastasis cascade of tumor cells. The dysfunction of Parkinson protein 2 E3 ubiquitin protein ligase (PARK2) is closely linked with the development of certain human cancers. However, whether PARK2 is associated with metastasis in GBM remains unknown. The present study demonstrated that the metastasis and invasion of U87 cells were significantly repressed by PARK2 overexpression. Conversely, knockdown of PARK2 facilitated the metastasis and invasion of A172 cells. Furthermore, PARK2 downregulated zinc finger E-box-binding homeobox 1 (ZEB1) expression and mitigated epithelial-mesenchymal transition (EMT). Promoter effects of PARK2 knockdown on cell metastasis and EMT were antagonized by silencing ZEB1 expression. These results indicated that PARK2 participated in regulating the invasion-metastasis cascade of cancer cells by depressing ZEB1 expression and acting as a metastasis suppressor in GBM progression, providing a potential therapeutic approach for GBM treatment.

Vitamin E (VE) is an important antioxidant supplement for human health. Soybean seed extracts are the main source of VE supplements. Therefore, increasing the VE content of soybean seeds is important issue in breeding programmes. To detect quantitative trait loci (QTL) associated with VE in soybean seeds, 238 F6:7 recombinant inbred lines (RILs) were created by crossing a high VE cultivar, ‘Beifeng 9’, with a low VE cultivar, ‘Freeborn’. A genetic map was constructed using 218 polymorphic simple sequence repeat (SSR) markers. Composite interval mapping analysis detected 66 QTLs for contents of individual and total VE, 21 for α-tocopherol, seventeen for γ-tocopherol, thirteen for δ-tocopherol and fifteen for total VE. The QTLs were located on chromosomes 9, 10, 15, 18 and 19. Phenotypic variance underlain by each QTL ranged from 2.4% to 32.6%. Two major QTLs (BARCSOYSSR_10_1140–BARCSOYSSR_10_1188 and BARCSOYSSR_15_0855 to BARCSOYSSR_15_0887) associated with α-Toc, γ-Toc, δ-Toc and total VE contents were mapped on chromosomes 10 and 15. They explained 12.0% and 32.6% of phenotypic variance for α-Toc; 5.5% and 13.0% for γ-Toc; 6.6% and 23.6% for δ-Toc; and 19.6% and 21.8% for total VE. QTL intervals BARCSOYSSR_15_0790–BARCSOYSSR_15_0855 (Qα15_1, Qγ15_1), BARCSOYSSR_15_1113–BARCSOYSSR_15_1159 (Qα15_3, Qδ15_2, QTVE15_4) and BARCSOYSSR_15_1159–BARCSOYSSR_15_1190 (Qα15_4, Qγ15_5, QTVE15_5) were associated with α-Toc and explained 22.2%, 23.8% and 24.4% of the phenotypic variation in multiple environments. BARCSOYSSR_09_1098–BARCSOYSSR_09_1128 (QTVE9_1) and BARCSOYSSR_15_0887–BARCSOYSSR_15_0935 (QTVE15_2, Qγ15_3) associated with total VE content explained 21.8% and 16.4% of the phenotypic variation in two environments. These QTLs allow for marker-assisted selection for cultivars with high VE contents.

Objectives-Elevated protein O-GlcNAcylation could benefit cell survival and promote organ functional recovery. Thiamet-G (O-GlcNAcase inhibitor) could upregulate protein O-GlcNAcylation level to improve dyskinesia in models of neurodegenerative diseases without any obvious detrimental side-effects. Therefore, we conducted this study to investigate the effects of protein O-GlcNAcylation upregulation by Thiamet-G on the spinal cord injury (SCI) in rats. Methods-We randomly assigned 74 rats to three groups: sham-operated group (Sham) with no lesion (n = 22), injured control group (SCI+SS) with saline solution (n = 26), and Thiamet-G treated group (SCI+Thiamet-G) (n = 26). We assessed Locomotor behavior using the Basso, Beattice, and Bresnahan (BBB) scale and evaluated histopathological alterations by morphometry and histochemistry. We also assessed potential inflammatory effects by microglia/macrophages immunohistochemistry, and potential apoptosis effects by caspase-3 western blot. Results-Thiamet-G treatment improved hindlimb motor functional recovery by inducing elevated protein O-GlcNAcylation, and mitigated the severity, reduced the lesion size and promoted the structural recovery of the injured spinal cord. Thiamet-G treatment also inhibited microglia/macrophages infiltration at the injury sites and the caspase-3 mediated apoptosis pathway. Discussion-We conclude that Thiamet-G induced elevated protein O-GlcNAcylation to ameliorate acute SCI, which could provide a potential novel therapeutic approach for SCI treatment.

Most of quantitative trait loci (QTL) underlying soybean seed isoflavone contents were derived from the harvest stage of plant development, which uncover the genetic effects that were expressed in earlier seed developmental stages. The aim of this study was to detect conditional QTL associated with isoflavone accumulation during the entire seed development. A total of 112 recombinant inbred lines developed from the cross between ‘Zhongdou 27’ (higher seed isoflavone content) and ‘Jiunong 20’ (lower seed isoflavone content) were used for the conditional QTL analysis of daidzein (DZ), genistein (GT), glycitein (GC) and total isoflavone (TI) accumulations through composite interval mapping with mixed genetic model. The results indicated that the number and type of QTL and their additive effects for individual and total isoflavone accumulations were different among R3 to R8 developmental stages. Three unconditional QTL and six conditional QTL for DZ, four unconditional QTL and five conditional QTL for GT, six unconditional QTL and five conditional QTL for GC, six unconditional QTL and seven conditional QTL for TI were identified at different developmental stages, respectively. Unconditional and conditional QTL that affect individual and total isoflavone accumulations exhibited multiple expression patterns, implying that some QTL are active for long period and others are transient. Two genomic regions, Satt144-Satt569 in linkage group F (LG F; chromosome 13, chr 13) for DZ, GC, GT and TI accumulations and Satt540-Sat_240 in LG M (chr 07) for TI and GC accumulations, were found to significantly affect individual and total isoflavone accumulations in multiple developmental stages, suggesting that the accumulation of soybean seed isoflavones is governed by time-dependent gene expression.

A solid knowledge associated with lumbar drainage (LD)-related infections in spontaneous subarachnoid hemorrhage (SAH) patients is necessary and that would be useful in taking effective measures to cope with this complication. We aimed to describe incidence rates and risk factors associated with LD-related infections in SAH patients. A retrospective review was performed on SAH patients who underwent LD between July 2010 and August 2015. Patient charts were reviewed to retrieve demographic, clinical, and laboratory data. LD-related infections were defined based on culture results of cerebrospinal fluid in combination with clinical symptoms. Infection rates were calculated, and a logistic regression model was developed to identify risk factors. A total of 629 SAH patients (25–82 years age range, 42.8 % male) were treated with LD in the period. LD-related infections were identified in 36 patients (5.7 %). Introduction: Glioma is one of the most common and most aggressive brain tumors in humans. The molecular and cellular mechanisms responsible for the onset and the progression of glioma are elusive and controversial. Centrosomal protein of 55 (CEP55) was initially described as a highly coiled-coil protein that plays critical roles in cell division, but was recently identified as being overexpressed in many human cancers. The function of CEP55 has not previously been characterized in glioma. We aim to discover the effect and mechanism of CEP55 in glioma development. Method: qRT-PCR and immunohistochemistry were used to analyze CEP55 expression. Glucose uptake, western blot, MTS, CCK-8, Caspase-3 activity and TUNEL staining assays were performed to investigate the role and mechanism of CEP55 on glioma cell process. Results: We found that the levels of CEP55 expression were upregulated in glioma. In addition, CEP55 appeared to regulate glucose metabolism of glioma cells. Furthermore, knockdown of CEP55 inhibited cell proliferation and induced cell apoptosis in glioma. Finally, we provided preliminary evidence that knockdown of CEP55 inhibited glioma development via suppressing the activity of Akt/mTOR signaling. Conclusions: Our results demonstrated that CEP55 regulates glucose metabolism, proliferation and apoptosis of glioma cells via the Akt/mTOR signaling pathway, and its promotive effect on glioma tumorigenesis can be a potential target for glioma therapy in the future.

In eukaryotes, proteins encoded by the 14-3-3 genes are ubiquitously involved in the plant growth and development. The 14-3-3 gene family has been identified in several plants. In the present study, we identified 22 GmGF14 genes in the soybean genomic data. On the basis of the evolutionary analysis, they were clustered into ε and non-ε groups. The GmGF14s of two groups were highly conserved in motifs and gene structures. RNA-seq analysis suggested that GmGF14 genes were the major regulator of soybean morphogenesis. Moreover, the expression level of most GmGF14s changed obviously in multiple stress responses (drought, salt and cold), suggesting that they have the abilities of responding to multiple stresses. Taken together, this study shows that soybean 14-3-3s participate in plant growth and can response to various environmental stresses. These results provide important information for further understanding of the functions of 14-3-3 genes in soybean.

A Synchronous Optimization for Multiship Shuttle Tanker Fleet Design and Scheduling is solved in the context of development of floating production storage and offloading device (FPSO). In this paper, the shuttle tanker fleet scheduling problem is considered as a vehicle routing problem with hard time window constraints. A mixed integer programming model aiming at minimizing total transportation cost is proposed to model this problem. To solve this model, we propose an exact algorithm based on the column generation and perform numerical experiments. The experiment results show that the proposed model and algorithm can effectively solve the problem.

Glioblastomas multiforme (GBM) are the most frequently occurring malignant brain cancers. Treatment for GBM consists of surgical resection and subsequent adjuvant radiation therapy and chemotherapy. Despite this, GBM patient survival is limited to 12-15 months, and researchers are continually trying to develop improved therapy options. Insulin-like growth factor 2 mRNA-binding protein 2 (Imp2) is known to be upregulated in many cancers and is known to regulate the signaling activity of insulin-like growth factor 2 (IGF2). However, relatively little is known about its role in malignant development of GBM. In this study, we first found Imp2 is upregulated in GBM tissues by using clinical samples and public database search. Studies with loss and gain of Imp2 expression in in vitro GBM cell culture system demonstrated the role of Imp2 in promoting GBM cell proliferation, migration, invasion and epithelial-to-mesenchymal transition (EMT). Additionally, our results show that Imp2 regulates the activity of IGF2, which further activates PI3K/Akt signaling, thereby to promote GBM malignancy. Inhibition of Imp2 was also found to sensitize GBM to temozolomide treatment. These observations add to the current knowledge of GBM biology, and may prove useful in development of more effective GBM therapy.

Soybean pod borer (SPB) (Leguminivora glycinivorella (Mats.) Obraztsov) causes severe loss of soybean (Glycine max L. Merr.) seed yield and quality in some regions of the world, especially in north-eastern China, Japan and Russia. Isoflavones in soybean seed play a crucial role in plant resistance to diseases and pests. The aim of this study was to find whether SPB resistance QTL are associated with soybean seed isoflavone content. A cross was made between ‘Zhongdou 27’ (higher isoflavone content) and ‘Jiunong 20’ (lower isoflavone content). One hundred and twelve F5:10 recombinant inbred lines were derived through single-seed descent. A plastic-net cabinet was used to cover the plants in early August, and thirty SPB moths per square metre were put in to infest the soybean green pods. The results indicated that the percentage of seeds damaged by SPB was positively correlated with glycitein content (GC), whereas it was negatively correlated with genistein (GT), daidzein (DZ) and total isoflavone content (TI). Four QTL underlying SPB damage to seeds were identified and the phenotypic variation for SPB resistance explained by the four QTL ranged from 2% to 14% on chromosomes Gm7, 10, 13 and 17. Moreover, eleven QTL underlying isoflavone content were identified, and ten of them were encompassed within the same four marker intervals as the SPB QTL (BARC-Satt208-Sat292, Satt144-Sat074, Satt540-Sat244 and Satt345-Satt592). These QTL could be useful in marker-assisted selection for breeding soybean cultivars with both SPB resistance and high seed isoflavone content.

The food enterprise risk warning problem is an important issue both in the theoretical and practical aspects. For this reason, a novel risk warning technology is proposed to food enterprises based on data mining and information fusion. Experimental results suggest that this technology is feasible, correct and valid. Experimental results and trend analysis has many practical significance for assisting government management and decision of import and export food agricultural product safety.
