The hybrid between the female channel catfish (Ictalurus punctatus) and the male blue catfish (I. furcatus) is the best genetic type currently available for commercial catfish farming due to their superior traits. However, further genetic improvements can be achieved by selecting parents with increased combining abilities. Twenty female channel catfish and twelve male blue catfish were crossed in a partial factorial mating design, resulting in forty hybrid families. These families were evaluated for early growth in three different rearing systems, including ponds and aquaria. The early growth performance of hybrid catfish was significantly (p < 0.05) affected by the additive gene action of the female parent and the male parent. There were genotype–environment or genotype–age interactions affecting the combining abilities, both the amount and the type of genetic variation. Dam GCA was significant in all environments/ages; however, sire GCA was variable, and SCA was not significant. These findings suggest that reciprocal recurrent selection for growth could potentially improve the performance of F1 hybrid catfish.

Antimicrobial peptides (AMPs) are promising next-generation antibiotics, but are difficult to produce due to the toxicity to bacterial hosts. Here, the authors report the utilization of transplastomic tobacco plants for AMPs production without cytotoxic effects via inducible expression systems and fusions to cleavable carrier protein.

Creeping bentgrass (Agrostis stolonifera) experiences quality decline during summer in the United States transition zone and warmer regions. Various bioproducts have been used to improve creeping bentgrass performance and to mitigate effects of summer stress in the United States transition zone. This 2-year study was carried out to examine if foliar application of seaweed extract (SWE; Ascophyllum nodosum)-based biostimulant Utilize® could enhance creeping bentgrass nitrate reductase (NaR) activity, and root viability under heat and drought stress conditions. The Utilize® was sprayed biweekly on creeping bentgrass foliage at 0, 29, 58, 87, and 116 µL⋅m−2, with application volume of 815 L⋅ha−2. Two weeks after first application, plants were exposed to heat (35/25 °C, day/night) and drought stress (40% to 50% evapotranspiration replacement) conditions for 42 days in an environment-controlled growth chamber. In general, the abiotic stress caused turf quality reduction. Foliar application of Utilize® at 58, 87, and 116 µL⋅m−2 increased turf quality, leaf color ratings, leaf chlorophyll, carotenoid content, and net photosynthetic rate (Pn). Utilize® at 58, 87, and 116 µL⋅m−2 increased NaR activity by 26.5%, 16.3%, and 16.3%, respectively, when compared with the control. Utilize® at 58, 87, and 116 µL⋅m−2 increased root biomass, root length, surface area (SA), and root volume when compared with the control. Utilize® at 58 and 87 µL⋅m−2 improved root viability by 16.3% and 30.9%, respectively, when compared with the control. Our data indicate that the SWE-based biostimulant Utilize® improves nitrogen (N) metabolism and root viability. Utilize® treatment at 58 µL⋅m−2 biweekly can be considered an effective approach for improving creeping bentgrass performance during summer stress.

The Powell River in Tennessee and Virginia, U.S.A. is a biodiversity hotspot for freshwater mussels, a highly imperiled faunal group in North America. Populations have been declining for several decades. Identification of the cause(s) of mussel decline is critical toward improving water quality and restoring populations in the river. We hypothesized that mussel populations may be suppressed because of exposure to toxic levels of trace elements in sediment pore water, a consequence of coal mining in the watershed. We measured concentrations of trace elements in water-column (surface) and substrate interstitial (pore) water seven times over one year at nine locations spanning 157 river kilometers. Concentrations of trace elements in filtered (0.45 μm) surface and pore water were low relative to water-quality criteria. In contrast, trace elements were higher in pore water containing interstitial fine sediment particles (< 300 μm), with all toxic elements except arsenic having potential to exceed their chronic criteria if they were in dissolved form. Abstract The Cumberlandian Combshell (Epioblasma brevidens) is an endangered freshwater mussel endemic to the Tennessee and Cumberland River drainages, major tributaries of the Ohio River of the eastern United States. We conducted mask and snorkel surveys in May and June of 2021 and 2022 to locate, observe, photograph, and video female E. brevidens to document their unique mantle lures at sites in the Clinch River in Tennessee and Virginia. The mantle lure is morphologically specialized mantle tissue that mimics prey items of the host fish. The mantle lure of E. brevidens appears to mimic four distinct characteristics of the reproductive anatomy of the underside (ventral) of a gravid female crayfish, to include: (1) the external apertures of the oviducts located on the base of the third pair of walking legs, (2) crayfish larvae still encased in the egg membrane, (3) pleopods or claws, and (4) postembryonic eggs. Surprisingly, we observed males of E. brevidens displaying mantle lures that were anatomically complex and closely resembled the female mantle lure. The male lure similarly mimics oviducts, eggs, and pleopods but is diminutive (2–3 mm smaller in length or diameter) to those same structures in females. We describe for the first time the mantle lure morphology and mimicry of E. brevidens, showing its close resemblance to the reproductive anatomy of a gravid female crayfish, and a novel form of mimicry in males. To our knowledge, mantle lure displays in males have not been previously documented in freshwater mussels.

During the COVID-19 pandemic, there was perhaps no group of students better prepared to make the switch to online learning than Gen Z. Research supports the fact that Gen Z adapts quickly to and enjoys technology integration in educational spaces (Barber, 2020; Mellman, 2020; Pavao, 2020; Strawser, 2020), yet no research has explored how at-risk students experience online learning and how these students uniquely adopt persistence strategies to succeed. As a result, this content analysis investigates the online discussion board posts from a credit-bearing student success course for at-risk Gen Z students on warning or probationary financial aid status (GPA < 2.00 or below a 67% completed over attempted credit hours ratio) to learn how these students reflected on their online learning experiences and their precarious status at their institution. Findings echoed prior studies where students felt their biggest challenges were distractions and their own procrastination. Moreover, students reported feeling like a burden, students could not learn and socialize effectively in online settings, and students did not have knowledge of institutional support that could have helped them persist during the pandemic. However, students unearthed unique persistence strategies, such as blending tactile learning strategies into their online learning environment and strategically segmenting their technology use during class sessions. Implications for research, policy, and practice are addressed.

Bermudagrass is a warm-season turfgrass species widely used for sports fields, home lawns, and golf courses. Ultradwarf bermudagrass has been used for golf course greens, but its quality declines with abiotic stresses. This 2-year study was designed to investigate if foliar applications of seaweed extract-based biostimulant Utilize® could improve ultradwarf bermudagrass photosynthetic function, nitrate reductase activity, root growth, and root function while under heat stress and drought stress conditions. Utilize® was applied to ultradwarf bermudagrass canopy at 0, 88, 117, 175, and 234 μL⋅m−2. Two weeks after the initial application of Utilize®, bermudagrass was subjected to heat (40/36 °C, day/night) and drought stress (40–50% evapotranspiration replacement) for up to 42 days. Heat stress and drought stress caused decline of the turf quality. Foliar application of Utilize® at 117, 175, and 234 μL⋅m−2 biweekly consistently improved turf quality and leaf color ratings and increased leaf chlorophyll and carotenoid concentrations, net photosynthetic rate, nitrate reductase activity, and root growth and viability. On average, Utilize® at 117, 175, and 234 μL⋅m−2 increased turf quality ratings by 9.1%, 12.1%, and 10.6%, respectively, net photosynthetic rates by 32.4%, 45.0%, and 35.0%, respectively, and nitrate reductase activity by 16.7%, 18.8%, and 14.6%, respectively, compared with the control. Utilize® at 117, 175, and 234 μL⋅m−2 increased the root biomass, root length, surface area, and root volume compared with the control. Utilize® at 88, 117, 175, and 234 μL⋅m−2 increased root viability by 46.2%, 73.1%, 88.5%, and 74.4%, respectively, relative to the control. The results of this study suggest that seaweed extract-based biostimulant Utilize® improves nitrogen metabolism, photosynthetic function, root growth, and root viability. Foliar application of Utilize® at rates between 117 and 175 μL⋅m−2 biweekly can be considered an effective approach to improving ultradwarf bermudagrass performance under heat stress and drought stress environments.

Introduction Increasingly, older adults are turning to emergency departments (EDs) to address healthcare needs. To achieve these research demands, infrastructure is needed to both generate evidence of intervention impact and advance the development of implementation science, pragmatic trials evaluation and dissemination of findings from studies addressing the emergency care needs of older adults. The Geriatric Emergency Care Applied Research Network (https://gearnetwork.org) has been created in response to these scientific needs—to build a transdisciplinary infrastructure to support the research that will optimise emergency care for older adults and persons living with dementia.Methods and analysis In this paper, we describe our approach to developing the GEAR Network infrastructure, the scoping reviews to identify research and clinical gaps and its use of consensus-driven research priorities with a transdisciplinary taskforce of stakeholders that includes patients and care partners. We describe how priority topic areas are ascertained, the process of conducting scoping reviews with integrated academic librarians performing standardised searches and providing quality control on reviews, input and support from the taskforce and conducting a large-scale consensus workshop to prioritise future research topics. The GEAR Network approach provides a framework and systematic approach to develop a research agenda and support research in geriatric emergency care.Ethics and dissemination This is a systematic review of previously conducted research; accordingly, it does not constitute human subjects research needing ethics review. BackgroundPodcasting has become a popular medium for medical education content. Educators and trainees of all levels are turning to podcasts for high-quality, asynchronous content. Although numerous medical education podcasts have emerged in recent years, few student-run podcasts exist. Student-run podcasts are a novel approach to supporting medical students. Near-peer mentoring has been shown to promote medical students’ personal and professional identity formation. Student-run podcasts offer a new medium for delivering near-peer advice to medical students in an enduring and accessible manner. ObjectiveThis paper describes the creation of the UnsCripted Medicine Podcast—a student-run medical education podcast produced at the University of Cincinnati College of Medicine. MethodsThe planning and preparatory phases spanned 6 months. Defining a target audience and establishing a podcast mission were key first steps. Efforts were directed toward securing funding; obtaining necessary equipment; and navigating the technical considerations of recording, editing, and publishing a podcast. In order to ensure that high professionalism standards were met, key partnerships were created with faculty from the College of Medicine. ResultsThe UnsCripted Medicine Podcast published 53 episodes in its first 2 years. The number of episodes released per month ranges from 0 to 5, with a mean of 2.0 episodes. The podcast has a Twitter account with 217 followers. The number of listeners who subscribed to the podcast via Apple Podcasts grew to 86 in the first year and then to 218 in the second year. The show has an average rating of 4.8 (out of 5) on Apple Podcasts, which is based on 24 ratings. The podcast has hosted 70 unique guests, including medical students, resident physicians, attending physicians, nurses, physicians’ family members, graduate medical education leadership, and educators. ConclusionsMedical student–run podcasts are a novel approach to supporting medical students and fostering professional identity formation. Podcasts are widely available and convenient for listeners. Additionally, podcast creators can publish content with lower barriers of entry compared to those of other forms of published content. Medical schools should consider supporting student podcast initiatives to allow for near-peer mentoring, augment the community, facilitate professional identity formation, and prepare the rising physician workforce for the technological frontier of medical education and practice.

Peak power is a critical factor for sub-nanosecond-pulsed transmitters utilizing laser diodes (LD) and applied to long distance LIDARs (light detection and ranging) for drones and automotive applications. Receiver speed is not anymore a limiting factor thanks to replacing linear (typically avalanche) detectors and a broad-band amplifier with a single photon avalanche detector (SPAD). Consequently the transmitters become the bottle neck in the resolution and ranging. The simplest and lowest-possible-cost transmitter consists of a switch, an LD, a storage capacitor C, and unavoidable parasitic loop inductance L. In the resulting resonant circuit, the principal problem consists of suppressing relaxation oscillations. Traditional way of oscillation damping reduce peak current and increase the pulse width. Here we show that specific transient properties of a Si avalanche switch solves the problem automatically provided the inductance is sufficiently low. This finding advances the state-of-the-art by reaching 90 W/1ns/200 kHz pulses from a miniature low-cost transmitter based on Si avalanching bipolar junction transistor (ABJT). Besides, the same self-damping effect may be realized in other switches maintaining significant residual voltage despite of fast current reduction.

Transcription activator-like effector nuclease (TALEN) plasmids targeting the channel catfish gonadotropin-releasing hormone (cfGnRH) gene were delivered into fertilized eggs with double electroporation to sterilize channel catfish (Ictalurus punctatus). Targeted cfGnRH fish were sequenced and base deletion, substitution, and insertion were detected. The gene mutagenesis was achieved in 52.9% of P1 fish. P1 mutants (individuals with human-induced sequence changes at the cfGnRH locus) had lower spawning rates (20.0–50.0%) when there was no hormone therapy compared to the control pairs (66.7%) as well as having lower average egg hatch rates (2.0% versus 32.3–74.3%) except for one cfGnRH mutated female that had a 66.0% hatch rate. After low fertility was observed in 2016, application of luteinizing hormone-releasing hormone analog (LHRHa) hormone therapy resulted in good spawning and hatch rates for mutants in 2017, which were not significantly different from the controls (p > 0.05). No exogenous DNA fragments were detected in the genome of mutant P1 fish, indicating no integration of the plasmids. No obvious effects on other economically important traits were observed after the knockout of the reproductive gene in the P1 fish. Growth rates, survival, and appearance between mutant and control individuals were not different. While complete knock-out of reproductive output was not achieved, as these were mosaic P1 brood stock, gene editing of channel catfish for the reproductive confinement of gene-engineered, domestic, and invasive fish to prevent gene flow into the natural environment appears promising.

Improving the longitudinal modes coupling in layered spherical structure contributes significantly to corneal terahertz sensing, which plays a crucial role in the early diagnosis of cornea dystrophies. Using a steel sphere to calibrate reflection from the cornea sample assists in enhancing the resolution of longitudinal modes. The requirement and challenges toward applying the calibration sphere are introduced and addressed. Six corneas with different properties are spotted to study the effect of perturbations in the calibration sphere in a frequency range from 100 GHz to 600 GHz. A particle-swarm optimization algorithm is employed to quantify corneal characteristics considering cases of accurately calibrated and perturbed calibrated scenarios. For the first case, the study is carried out with signal-to-noise values of 40 dB, 50 dB and 60 dB at waveguide bands WR-5.1, WR-3.4, and WR-2.2. As expected, better estimation is achieved in high-SNR cases. Furthermore, the lower waveguide band is revealed as the most proper band for the assessment of corneal features. For perturbed cases, the analysis is continued for the noise level of 60 dB in the three waveguide bands. Consequently, the error in the estimation of corneal properties rises significantly (around 30%).

Current point-of-care (POC) screening of Coronavirus disease 2019 (COVID-19) requires further improvements to achieve highly sensitive, rapid, and inexpensive detection. Here we describe an immunoresistive sensor on a polyethylene terephthalate (PET) film for simple, inexpensive, and highly sensitive COVID-19 screening. The sensor is composed of single-walled carbon nanotubes (SWCNTs) functionalized with monoclonal antibodies that bind to the spike protein of SARS-CoV-2. Silver electrodes are silkscreen-printed on SWCNTs to reduce contact resistance. We determine the SARS-CoV-2 status via the resistance ratio of control- and SARS-CoV-2 sensor electrodes. A combined measurement of two adjacent sensors enhances the sensitivity and specificity of the detection protocol. The lower limit of detection (LLD) of the SWCNT assay is 350 genome equivalents/mL. The developed SWCNT sensor shows 100% sensitivity and 90% specificity in clinical sample testing. Further, our device adds benefits of a small form factor, simple operation, low power requirement, and low assay cost. This highly sensitive film sensor will facilitate rapid COVID-19 screening and expedite the development of POC screening platforms.

Abstract Background Findings suggest approximately one in six people with intellectual disability engage in ‘challenging behaviours’, which include aggression towards others/property and self-injurious actions. In residential settings, actions of staff members can make challenging behaviours more likely to occur, or make these behaviours worse. In particular, negative attitudes from members of staff and lack of understanding about the reasons for challenging behaviour are contributory factors. ‘Who’s Challenging Who?’ (WCW) training is designed to emphasise the role of staff in residential settings as a challenge also to people with intellectual disability. The course is delivered jointly by a trainer with intellectual disability who has been labelled as having challenging behaviour, along with a trainer without intellectual disability. Methods This is a cluster randomised two-arm trial of WCW training versus a waiting list control. Overall, 118 residential settings will be recruited and randomised on a 1:1 ratio. Within each setting, two members of staff will be invited to take part in the trial. Participants will complete assessments at baseline and at 6 and 20 weeks. WCW is a half day initial training course with some follow-on coaching to ensure implementation. The primary outcome is changes in staff empathy towards people with challenging behaviour. Secondary outcomes at the staff level include confidence, attitudes and work-related well-being. Secondary outcomes at the residential setting level include recorded incidents of aggressive challenging behaviour, and use of any restrictive practices. Discussion If the results of the cluster randomised trial are positive, we will disseminate the findings widely and make all training manuals and materials freely available for anyone in intellectual disability services (and beyond) to use. Our training approach may have wider implications in other areas of social care. It may also provide a generally applicable model for how to train people with intellectual disability to act as co-trainers in intellectual disability social care settings. People with intellectual disability and challenging behaviour have already been involved centrally with the design, development and pilot evaluation of WCW and will also be fully involved throughout this trial. Introduction: Raoultella planticola is a gram-negative rod associated with soil and aquatic environments that has rarely been associated with human infections. Presentation of case: We present the case of a 73 year old female with multiple myeloma and recurrent Clostridium difficile infection who was found to have a urinary tract infection with Raoultella planticola, which has only been reported to cause 29 cases of human infections and 2 cases of urinary tract infections. Discussion and conclusion: Our case and literature review suggest that immunocompromised patients are predisposed to developing Raoultella planticola infection, and that this is a potential emerging pathogen.

Targeted photothrombosis is a method to occlude individual arterioles and venules that lie on the surface of the cerebral cortex. It has been used to study collateral flow patterns within the pial vascular network following occlusion of single surface vessels (Schaffer et al., 2006; Blinder et al., 2010; Nguyen et al., 2011), as well as to generate localized ischemic strokes following occlusion of single penetrating vessels (Nishimura et al., 2007; Drew et al., 2010; Shih et al., 2013). The intravascular clot is formed by irradiation of a target vessel with a focused green laser after injection of a circulating photosensitizing agent, Rose Bengal (Watson et al., 1985). We briefly describe modifications of custom-designed and commercial two-photon imaging systems required to introduce a green laser for photothrombosis. We further provide instructions on how to occlude a single penetrating arteriole within the somatosensory cortex of an anesthetized mouse.
