Abstract

Fungi are highly diverse organisms, which provide multiple ecosystem services. However, compared with charismatic animals and plants, the distribution patterns and conservation needs of fungi have been little explored. Here, we examined endemicity patterns, global change vulnerability and conservation priority areas for functional groups of soil fungi based on six global surveys using a high-resolution, long-read metabarcoding approach. We found that the endemicity of all fungi and most functional groups peaks in tropical habitats, including Amazonia, Yucatan, West-Central Africa, Sri Lanka, and New Caledonia, with a negligible island effect compared with plants and animals. We also found that fungi are predominantly vulnerable to drought, heat and land-cover change, particularly in dry tropical regions with high human population density. Fungal conservation areas of highest priority include herbaceous wetlands, tropical forests, and woodlands. We stress that more attention should be focused on the conservation of fungi, especially root symbiotic arbuscular mycorrhizal and ectomycorrhizal fungi in tropical regions as well as unicellular early-diverging groups and macrofungi in general. Given the low overlap between the endemicity of fungi and macroorganisms, but high conservation needs in both groups, detailed analyses on distribution and conservation requirements are warranted for other microorganisms and soil organisms.

Abstract

The tests of the Toroidal Field Model Coil (TFMC) are completed in the TOSKA facility of Forschungszentrun Karlsruhe. The TFMC isolated strand is characterized at different applied uniaxial strain, and preliminary results indicate a stronger reduction of carrying capacity compared to the extrapolation from summers scaling used in the analysis.

Abstract

The aims of the TF Model Coil are to test the manufacturing feasibility of the ITER TF magnet concepts, to assess the reliability of the fully integrated system by dedicated testing and to qualify the quality assurance and the testing methods. The 3.8 m long racetrack shaped TFMC will be tested in the TOSKA facility at Karlsruhe in a configuration using the Euratom LCT Coil to provide in-plane and out-of-plane loads, relevant to the ITER coils.

Abstract

A 30 kA/23 kV current lead cooled by forced-flow supercritical helium has been developed at KfK. It allows position independent installation and well controlled operation which would be advantageous for operation of all kind of super-conducting forced-flow cooled magnets. The design of the lead is described in this paper. The main feature is the insertion of [Nb.sub.3]Sn wires inside the conductor of the heat exchanger allowing the operation at minimum mass flow in a wide current range. Measurement results are presented for steady-state operation up to 30 kA, for short time operation up to 50 kA, for pulsed operation up to [+ or -] 1000 A at 10 Hz, and for simulation of loss of mass flow. A high-voltage test up to 28 kV DC has been done. The mass flow rate normalized to current is about 0.055 g/(s-kA) for 15 to 30 kA proving the effectiveness of the [Nb.sub.3]Sn inserts. Comparison to numerical calculations shows good agreement allowing extrapolation for the design of leads for currents up to 80 kA for ITER model coil tests in the TOSKA test facility at KfK.

Abstract

The regulation of metabolic processes by the Indy (I'm Not Dead Yet) (SLC13A5/NaCT) gene was revealed through studies in Drosophila melanogaster and Caenorhabditis elegans. Reducing the expression of Indy in these species extended their life span by a mechanism resembling caloric restriction, without reducing food intake. In D. melanogaster, mutating the Indy gene reduced body fat content, insulin-like proteins and reactive oxygen species production. Subsequent studies indicated that Indy encodes a citrate transporter located on the cell plasma membrane. The transporter is highly expressed in the mammalian liver. We generated a mammalian knock out model deleting the mammalian homolog mIndy (SLC13A5). The knock out animals were protected from HFD induced obesity, fatty liver and insulin resistance. Moreover, we have shown that inducible and liver selective knock down of mIndy protects against the development of fatty liver and insulin resistance and that obese humans with type 2 diabetes and non-alcoholic fatty liver disease have increased levels of mIndy. Therefore, the transporter mINDY (NaCT) has been proposed to be an 'ideal target for the treatment of metabolic disease'. A small molecule inhibitor of the mINDY transporter has been generated, normalizing glucose levels and reducing fatty liver in a model of diet induced obese mice. Taken together, studies from lower organisms, mammals and humans suggest that mINDY (NaCT) is an attractive target for the treatment of metabolic disease.
