In the current study, Methylation and expression microarray data related to Schizophrenia (SCZ) were analyzed to identify genes whose aberrant promoter methylation could play a crucial role in the dysregulation of gene expression. After data analysis, two genes including KLF9 and MYH11 were identified. Considering CpG sites in the promoter region of the mentioned genes using Methprimer database, Bisulfite specific primer (BSP) was designed, and Methylation specific-high resolution melting (MS-HRM) analysis was performed to assess the promoter methylation status of the genes, and also real-time PCR was used to evaluate mRNA expression level of KLF9 and MYH11 in 50 SCZ and healthy control specimens. The results show 62% hypermethylation and 38% hypomethylation of KLF9 in SCZ samples and 34% hypermethylation and 66% hypomethylation in healthy control samples. MYH11 promoter shows 54% hypermethylation and 46% hypomethylation in SCZ samples while 28% hypermethylation and 72% hypomethylation in healthy controls. The result of real-time PCR shows downregulation of KLF9 and MYH11 in SCZ samples compared with healthy samples. The aberrant methylation has a direct effect on the expression of mentioned genes.

However, advanced technologies have been developed in the treatment of various cancers, but the mortality rate from cancer is still very high. Drug resistance is a major problem for patients with cancer, which causes the treatment process to fail. In addition to inhibiting drug resistance, targeted therapy is also very important in treatment.Nowadays, miRNAs have gained increasing interest as they play a major role in both drug resistance and targeted therapy. MicroRNA (miRNA) is an important part of non-coding RNA that regulates gene expression at a post-transcriptional level. The prevailing studies about miRNA expression have been expanded into a variety of neoplasms. MiR-424 and miR-631 targets genes involved in various cellular processes and can participate in proliferation, differentiation, apoptosis, invasion, angiogenesis, and drug resistance and sensitivity.In this study, we focus on the role of miR-424 and miR-631 in many cancer types by displaying the potential target genes associated with each cancer, as well as briefly describing the clinical uses of miR-424 and miR-631 as a diagnostic and predictive tool in malignancies.

The high mortality rate in Breast Cancer (BC) is largely due to the difficulty of early detection and inadequate accuracy of the current diagnostic approaches. In this study, we investigated serum and tumor miRNA profiles of 1800 breast cancer miRNA expression profiles of GEO (Gene Expression Omnibus) and also more than 2500 patients in TCGA (The Cancer Genome Atlas) database. After data analysis by some packages like Limma, and WGCNA, we selected 6 hub miRNAs. Then by collecting serum samples of 100 breast cancer patients, real-time PCR was performed and the diagnosis performance of hub miRNAs was evaluated by ROC curve. Due to more validation, Kaplan-Meier Plot of hub miRNAs was evaluated by 1262 BC samples using METABRIC database. The target genes of 6 hub miRNAs were investigated using Mirbase, Mirtarbase, Targetscan, and TransmiR and after the identification of target genes, their expression was evaluated in 1600 BC samples using GEO database. Finally, six miRNAs including Hsa-miR-151a-5p, Hsa-miR-34a-5p, Hsa-miR-1307-3p, Hsa-miR-450b-5p, Hsa-miR-501-3p, and Hsa-miR-532-5p were introduced as effective biomarkers of BC.

Abstract: MicroRNAs are non-coding ribonucleic acids that are evolutionarily protected. MiRNAs control the expression of genes after transcription by mRNA decomposition or inhibition of their translation. These molecular structures control physiological and pathological processes; therefore, many of them can play vital roles as oncogenes or tumor inhibitors. Besides, the occurrence of various mutations in miRNAs can lead to cancer. In this review article, we want to peruse the role of miR-491-5p in various cancers. In recent years, many experiments and studies have been performed on the involvement of miR-491-5p in cancer, invasion, and cell metastasis. Metastasis is an event that makes cancer more advanced and harder to treat. When cancer is invasive, the cancer cells invade nearby tissues or other organs and develop cancer. Tumor studies have shown that miR-491-5p can inhibit cell growth, invasion, and metastasis. Thus, expression enhancement of miR-491-5p disrupts cell migration and improves cancer.

Therefore by considering internationally and locally data, we succeeded to identify the more prevalent and effective breast cancer biomarkers including Hsa-miR-151a-5p, Hsa-miR-34a-5p, Hsa-miR-1307-3p, Hsa-miR-450b-5p, Hsa-miR-501-3p, and Hsa-miR-532-5p.

Drug resistance is the drug-effectiveness reduction in treatment and is a serious problem in oncology and infections. In oncology, drug resistance is a complicated process resulting from enhancing the function of a pump that transports drugs out of tumor cells, or acquiring mutations in drug target. Surprisingly, most drugs are very effective in the early stages, but the response to the drug wears off over time and resistance eventually develops. Drug resistance is caused by genetic and epigenetic changes that affect cancer cells and the tumor environment. The study of inherited changes in the phenotype without changes in the DNA sequence is called epigenetics. Because of reversible changes in epigenetics, they are an attractive target for therapy. Some of these epigenetic drugs are effective in treating cancers like acute myeloid leukemia (AML), which is characterized by the accumulation and proliferation of immature hematopoietic cells in the blood and bone marrow. In this article, we outlined the various contributing factors involved in resistance or sensitivity to epigenetic drugs in the treatment of AML.

More powerful prognostic and diagnostic tools are urgently needed for identifying and treating ovarian cancer (OC), which is the most fatal malignancy in women in developed countries. Circular RNAs (circRNAs) are conservative and stable looped molecules that can regulate gene expression by competing with other endogenous microRNA sponges. This discovery provided new insight into novel methods for regulating genes that are involved in many disorders and cancers. This review focuses on the dysregulated expression of circRNAs as well as their diagnostic and prognostic values in OC. We found that studies have identified twenty-one downregulated circRNAs and fifty-seven upregulated ones. The results of these studies confirm that circRNAs might be potent biomarkers with diagnostic, prognostic and therapeutic target value for OC. We also consider the connection between circRNAs and OC cell proliferation, apoptosis, metastasis, and chemotherapy resistance and sensitivity.

Cancer is still considered a main challenge regarding morbidity and mortality, although an earlier diagnosis through screening programs and more effective cure modalities have led to decreased mortality rates so the development of new genetic editing techniques such as zinc finger and TALENs or CRISPRCas9 has enabled the production of strong animal genetic models that sums up the cooperating oncogenic lesions influencing genes with a confirmed role in the proliferation and formation. This review presents the mechanisms of separate genome-editing approaches and explains each of the common nucleasebased platforms.

Purpose To evaluate the association of CC-cytokine ligand 2 CCL2–2518 (rs1024611) single nucleotide polymorphism, complement factor H (CFH Y402H) and their possible interaction in developing advanced age-related macular degeneration (AMD). Methods In this case–control study, DNA samples from 266 patients with advanced AMD and 229 healthy controls were genotyped for CCL2 polymorphism and also 254 patients and 164 healthy controls were genotyped for CFH polymorphism. The possible associations of these polymorphisms with susceptibility to AMD independently and in different joint combinations were evaluated. We propose a method to k-coverage the field. Our target problem is NP-hard. So, we propose a genetic algorithm (GA) to find solution that is a near optimal solution. In our method, the maximum monitored area fully covered by a minimum active sensors.
