Vitis amurensis Ruprecht contains a large number of polyphenolic compounds which are biologically active components. For the most efficient and safe extraction supercritical carbon dioxide was used. In this work, for the first time, a comparative metabolomic study of biologically active substances of wild grapes collected from five different places of the Primorsky and Khabarovsk territories is carried out. To identify target analytes in ethanol extracts of grape berries, high performance liquid chromatography (HPLC) was used in combination with an amaZon SL ion trap (manufactured by BRUKER DALTONIKS, Germany) equipped with an ESI electrospray ionization source in negative and positive ion modes. The mass spectrometer was used in the scan range m / z 100 - 1.700 for MS and MS / MS. Used fragmentation of the 4th order. Primary mass spectrometric results showed the presence of 89 biologically active compounds corresponding to the species V. amurensis, moreover, salvianolic acids F, D and G, oleanoic, ursolic, myristoleic acids, berbericinin, mearnsetin, esculin, nevadensin, stigmasterol, fucosterol, phlorizin, L-tryptophan identified for the first time in V. amurensis.

The purpose of this work was a comparative metabolomic study of extracts of Blueberried honeysuckle Lonicera caerulea L.: №1043-11 (St. Petersburg); №1043-08 (St. Petersburg) №863; (Japan); №860 (Wild Lonicera from Amur river) from the collection of N.I. Vavilov All-Russian Institute of Plant Genetic Resources. To identify target analytes in extracts HPLC was used in combination with a BRUKER DALTONIKS ion trap. The results showed the presence of 82 target analytes corresponding to family Caprifoliaceae. In addition to the reported metabolites, a number of metabolites were newly annotated in Lonicera caerulea L. There were flavonols: Dihydrokaempferol, Rhamnetin I, Rhamnetin II, Taxifolin-3O-glucoside, Mearnsetin-hexoside, Horridin; flavones: Chrysoeriol, Apigenin-O-pentoside, Chrysoeriol-7-Oglucoside; flavanone Naringenin; flavan-3-ols: Catechin, Epicatechin, Biochanin A-7-O-glucoside; essential amino acids: L-Pyroglutamic acid, Tyrosine; polypeptide 5-Oxo-L-propyl-L-isoleucine; sterols: Ergosterol, Fucosterol, Beta-Sitosterin; triterpenoids: Betunolic acid, Oleanoic acid; anabolic steroid Vebonol, indole sesquiterpene alkaloid Sespendole; iridoids: Monotropein, p-Coumaroyl monotropein, p-Coumaroyl monotropein hexoside; Myristoleic acid, etc.

Actinidia deliciosa (A. Chev.) C.F. Liang & A.R& Ferguson, 1984 contains a large number of target analytes, which are biologically active compounds. HPLC-ion trap-MS/MS (tandem mass spectrometry) was used to identify target analytes in extracts of A. deliciosa (varieties Abbot, Alison, Bruno, Hayward, Monti), originating from the Adler's Branch of N.I. Vavilov All-Russian Institute of Plant Genetic Resources. Separately, the pulp of the kiwi fruit and its peel were analyzed for the composition of biologically active substances. The richest in the composition of active substances was indicators of the cultivar Monti. The results of initial studies revealed the presence of 56 compounds in the extracts of the peel of A. deliciosa and the presence of 64 compounds in the extracts of berry pulp of A. deliciosa. Forty-three compounds were identified for the first time in family Actinidiaceae. New identified metabolites belonged to 17 classes including 13 flavones, 2 flavonols, 2 flavan-3-ols, 5 stilbenes, 4 lignans, 2 carotenoids, 2 sterols, 3 terpenoids, etc. [ABSTRACT FROM AUTHOR]

Triticosecale “Bilinda” (cultivar was bred in the laboratories of the Leningrad Research Agriculture Institute, Branch of Russian Potato Research Centre, Leningrad Province) contains a large number of polyphenolic compounds which are biologically active components. The purpose of this work was to conduct a comparative metabolomic study of extracts of Triticosecale cultivar “Bilinda” seeds harvested at different stages of maturation. To identify target analytes in extracts HPLC was used in combination with an ion trap. The results showed the presence of 93 target compounds corresponding to Triticosecale. In addition to the reported polyphenol constituents, 28 polyphenols were newly annotated in grains of Triticosecale. There were 9 flavones, 7 flavonols, 2 flavan-3-ols, 1 flavanone, 5 hydroxybenzoic acids, 1 hydroxycinnamic acid, 1 anthocyanidin, 1 condensed tannin, 1 hydroxycoumarin, 2 triterpenoids; 1 diterpenoid, 6 sterols, 2 isoquinoline alkaloids, 4 carotenoids, 1 hydroquinone derivative and others bioactive compounds. Metabolic analysis showed a significant difference in the classes and amounts of polyphenolic substances identified in the seeds, depending on the time of seed collection. [ABSTRACT FROM AUTHOR]

Three types of Zostera marina L. collection were extracted using the supercritical CO2-extraction method. For the purposes of supercritical CO2-extraction, old seagrass ejection on the surf edge, fresh seagrass ejection on the surf edge and seagrass collected in water were used. Several experimental conditions were investigated in the pressure range 50–350 bar, with the used volume of co-solvent ethanol in the amount of 1% in the liquid phase at a temperature in the range of 31–70 °C. The most effective extraction conditions are: pressure 250 Bar and temperature 60 °C for Z. marina collected in sea water. Z. marina contain various phenolic compounds and sulfated polyphenols with valuable biological activity. Tandem mass-spectrometry (HPLC-ESI–ion trap) was applied to detect target analytes. 77 different biologically active components have been identified in Z. marina supercritical CO2-extracts. 38 polyphenols were identified for the first time in Z. marina. Several experimental conditions were investigated in the pressure range of 200–500 bar, with the used volume of cosolvent ethanol in the amount of 1% in the liquid phase at a temperature in the range of 31–70°C. The most effective extraction conditions are pressure 350 bar and temperature 60°C. The extracts were analyzed by HPLC with MS/MS identification. 78 target analytes were isolated from Rhodiola rosea (Russia) using a series of column chromatography and mass spectrometry experiments. The results of the analysis showed a spectrum of the main active ingredients Rh. rosea: salidroside, rhodiolosides (B and C), rhodiosin, luteolin, catechin, quercetin, quercitrin, herbacetin, sacranoside A, vimalin, and others. In addition to the reported metabolites, 29 metabolites were newly annotated in Rh. rosea. There were flavonols: dihydroquercetin, acacetin, mearnsetin, and taxifolin-O-pentoside; flavones: apigenin-O-hexoside derivative, tricetin trimethyl ether 7-O-hexosyl-hexoside, tricin 7-O-glucoronyl-O-hexoside, tricin O-pentoside, and tricin-O-dihexoside; flavanones: eriodictyol-7-O-glucoside; flavan-3-ols: gallocatechin, hydroxycinnamic acid caffeoylmalic acid, and di-O-caffeoylquinic acid; coumarins: esculetin; esculin: fraxin; and lignans: hinokinin, pinoresinol, L-ascorbic acid, glucaric acid, palmitic acid, and linolenic acid. The results of supercritical CO2-extraction from roots and rhizomes of Rh. rosea, in particular, indicate that the extract contained all biologically active components of the plant, as well as inert mixtures of extracted compositions. The current review focuses on the connection of telomerase and telomeres with aging. In this review, we describe the changes in telomerase and telomere length (TEL) during development, their role in carcinogenesis processes, and the consequences of reduced telomerase activity. More specifically, the connection of TEL in peripheral blood cells with the development of aging-associated diseases is discussed. The review provides systematic data on the role of telomerase in mitochondria, the biology of telomeres in stem cells, as well as the consequences of the forced expression of telomerase (telomerization) in human cells. Additionally, it presents the effects of chronic stress exposure on telomerase activity, the effect of TEL on fertility, and the effect of nutraceutical supplements on TEL. Finally, a comparative review of the chronographic theory of aging, presented by Olovnikov is provided based on currently available scientific research on telomere, telomerase activity, and the nature of aging by multicellular organisms. Intracellular fucoidanase was isolated from the marine bacterium, Formosa algae strain KMM 3553. The first appearance of fucoidan enzymatic hydrolysis products in a cell-free extract was detected after 4 h of bacterial growth, and maximal fucoidanase activity was observed after 12 h of growth. The fucoidanase displayed maximal activity in a wide range of pH values, from 6.5 to 9.1. The presence of Mg2+, Ca2+ and Ba2+ cations strongly activated the enzyme; however, Cu2+ and Zn2+ cations had inhibitory effects on the enzymatic activity. The enzymatic activity of fucoidanase was considerably reduced after prolonged (about 60 min) incubation of the enzyme solution at 45 °C. The fucoidanase catalyzed the hydrolysis of fucoidans from Fucus evanescens and Fucus vesiculosus, but not from Saccharina cichorioides. The fucoidanase also did not hydrolyze carrageenan. Desulfated fucoidan from F. evanescens was hydrolysed very weakly in contrast to deacetylated fucoidan, which was hydrolysed more actively compared to the native fucoidan from F. evanescens. Analysis of the structure of the enzymatic products showed that the marine bacteria, F. algae, synthesized an α-L-fucanase with an endo-type action that is specific for 1→4-bonds in a polysaccharide molecule built up of alternating three- and four-linked α-L-fucopyranose residues sulfated mainly at position 2. Rhizochalins C and D ( 1, 2), new representatives of two-headed glycosphingolipids, were isolated from the sponge Rhizochalina incrustata. Rhizochalin D is an unexpected C 29 homologue of the canonical C 28 dimeric sphingolipid structures. Their structures including absolute configurations were established using spectroscopic data, micromolar-scale Baeyer-Villiger oxidation, and LCMS interpretation of the products. Application of the latter method leads to a revision of the structure of oceanapiside and placement of the keto group at C-18 rather than C-11.
