9. EFFECT OF INDOXYL SULFATE ON THE MORPHOLOGY AND FUNCTION OF THE THYROID GLAND - EX VIVO STUDIES IN RABBITS.

Abstract

Indoxyl sulfates are uremic indolic toxins known to participate in the pathogenesis of cardiovascular diseases during chronic kidney disease in humans and some animal species. However, nothing is known about the indoxyl sulfate effect on the thyroid gland which is especially responsible for the general organism metabolism. This study determines the morpho-functional status of the thyroid gland after exposure to indoxyl sulfate (10, 25, and 50 mM) with the use of an ex vivo system and rabbit (n=10) as an experimental model thyroid gland histology, immunoexpression of thyrotropin receptor (TSHR), and concentrations of thyroxine (T4) and triiodothyronine (T3) were evaluated. Statistical analyses were performed using one-way analysis of the variance (ANOVA) followed by Tukey's post hoc comparison test. Minor alterations in thyroid tissue structure e.g. very rare exfoliated epithelial cells, condensed colloid fluid, or slight loosening of the epithelium were found. In addition, modulated dose dependent-expression of TSHR (p<0.01, p<0.001) together with a decreased level of T4 and T3 (p<0.001, p<0.01) exception of an increased level of T4 after the middle dose of indoxyl sulfate were revealed. We report here, for the first time, that indoxyl sulfate affects the thyroid gland mainly at the molecular level. The rabbit thyroid gland ex vivo system seems to be suitable for further studies on the thyroid gland in health and disease. However, the effect of TSH-TSHR signaling at ultrastructural, and epigenetic levels needs supplementary appraisal. [ABSTRACT FROM AUTHOR]

Abstract

Inverted internal limiting membrane (ILM) flap may constitute a scaffold for Muller cells whose migration and proliferation on its surface begin the process of macular hole closure. The goal of the study was to establish an in vitro model of the interaction between ILM and the Muller cells. Vitrectomy with inverted ILM flap was performed in 23 patients due to a full-thickness macular hole (FTMH). After dissection of the inverted flap, the area of ILM peeling was extended and material was collected for cell culture experiments. Muller cells cultured on adherent cell plates showed significantly better growth than on suspension plates. Our results reveal that the presence of the ILM can overcome the growth inhibitory effect of the non-adhesive surface. Moreover, the ILM appears to be the optimal growth surface under normoxia conditions mimicking the microenvironment after vitrectomy and hypoxia which is natural state for Muller cells. The closure rate of FTMH was 100%. Our study revealed that in non-adhesive culture conditions patient derived ILM constitutes an optimal growth surface for Muller cells. We have demonstrated that the ILM effectively stimulates attachment, proliferation, and survival of Müller cells in conditions of normoxia which is the case after vitrectomy. Abstract

This research aims to contribute to Colombia’s historical memory regarding the knowledges, experiences, and embodied subjectivities of displaced women, as active members of a society determined by more than fifty years of lasting conflict. Drawing on what has been called the ‘affective turn’, this work explores emotion as a pivotal bridge between the individual and the social, opening new possibilities for investigating and conceptualizing the subject of feminism as embodied, located and relational. Working with oral and visual narratives of displaced women, this research explores emotional-embodied-experiences of war, violence, and forced displacement, relevant in the constitution of oppressed/subversive subjectivities. Analyzing different spaces women have inhabited, objects they have interacted with, and social relationships they have established, during their forced mobility, this research aims to trace different actors within emotion’s sociality, recognizing that emotions of gendered violence are not processes inside victim’s minds and bodies, but effects and affects of social dynamics. This analysis aims to contribute to current debates regarding Colombian society’s accountability for maintaining and perpetuating social injustices in displaced women lives. By locating participant’s narratives at the core of knowledge theorization, this research elucidates new ways of entangle theoretical approaches in everyday life dynamics, widening the influence of the affective turn over cultural and social sciences approaches.

Abstract

Love is one of the most important experiences in women’s lives. It has been deconstructed by feminism in its romantic and universal facets. Love still engenders damages as well as new kinds of fulfilment and support for women. This thesis claims the importance of love as a political tool, analysing its manifestations in the stories of a group of migrant women squatters in Florence, Italy. This ethnography identifies both old patterns of subjugation disguised as love that women suffer from their partners, as well as new patterns men’s power in love takes within the migratory experience. However, this research shows also positive manifestations of love and emotions between empowered women who find the strength to free themselves from their partners’ subjugation, and establish a women-only squat. This investigation therefore manages to demonstrate why love is important in connection to migration, a current fundamental geopolitical topic, and how the practice of squatting enables the women I interviewed to fight different layers of stereotypes targeting both migrant women and Muslim women. “Sisterhood”, communal maternal practices and new forms of female multicultural cohabitation, despite the precarious and illegal aspects of living in a squat, emerge as the rich findings of my investigation.

Abstract

This thesis focuses on the political use of violence and specifically on notions, narratives and discourses that construct and defining “terrorism” in contemporary neoliberal capitalist societies. The author looks at the new dynamics of control and repression that are at stake in the current historical and socio-political context, characterized by the interaction of different forms of power – disciplinary, necropolitical and biopolitical – which act to construct new definitions, representations and countermeasures about concepts such as “threat”, “enemy” and “terrorism”. This concerns securitarian and repressive state technologies in the Italian political framework, situated in the broader European context, in relation to social and political movements. The thesis will explore how securitarian narratives and emergencial rhetoric, connected to an increased militarization of territories, act as devices for the criminalization of social movements and political struggles and how these narratives always build up larger spaces for the legitimation of the exercise of state control and oppression within a growing invisible structural systemic violence – “terror” – acted upon bodies along the lines of class, gender, race and ethnicity. Throughout the case study the author analyses queer and transfeminist movements (Atlantide) and focuses specifically on the ways they have been affected by a hegemonic narrative of securitization and on how queer subjectivities become stigmatized and marginalized.

Abstract

The relentless increase of reproducible images often gives rise to complaints that we are being ‘bombarded’ with them. This quasi-military vocabulary echoes the alleged aggressiveness of the visual culture that is enforcing itself upon us (Sturken and Cartwright 2001). Since the 1970s, feminist art critics and scholars in particular have problematised the omnipresence of sexualised, and yet mythologised, female bodies both in the media and in artistic practice, claiming that the tradition of meaning assigned to gender, race and ethnicity is not given, but constructed, and that these constructions take specific visual forms (Nochlin 1970; Parker and Pollock 1981). The underlying conviction of feminist theory has always been that to be able to change the lives and material circumstances of women, we need an alternative imagery with which to identify, as well as alternative modes of making, seeing and interpreting visual culture and its institutions (Pollock 1999; Reckitt 2001). Today, the training in gender sensitive readings of images in general and of images representing women in particular constitutes a broad field of feminist practices. While feminist cultural theorists aimed at deconstructing the idealist concept of representation as an unmediated, transparent practice, postcolonial scholars argued forcefully against the Eurocentrism that pervades many discussions of the visual (Shohat and Stam 1994). Together they challenge traditional ways of seeing as well as the hegemonic character of dominant patriarchal and colonial canons.

This study focuses on the recurrence of visual tropes in press photographs awarded in the annual contest World Press Photo (WPP) in the years 2009–11. By tropes, we mean conventions (e.g. a mourning woman, a civilian facing soldiers, a distressed witness to an atrocity) that remain unchanged despite their travels across the visual sphere, gaining professional and public recognition and having a strong affective impact. We contend that photography contests such as the WPP influence and organize a process of generic understanding of war, disaster and atrocity that is based on a number of persistent tropes, such as the mourner, the protester or the survivor amidst chaos and ruins. We further show that these tropes are gendered along traditional conceptions of femininity and masculinity, appealing strongly to both judges and wider audiences. The evidence for our claim comes from an analysis of the photographs that won awards, observation of the judging sessions, semi-structured interviews with three jury chairmen, and public commentary on the juries’ choices (blogs, newspapers and websites).

De ruimtelijke ontwikkeling van Amsterdam is vastgelopen. De ongezonde hoeveelheid aan leegstand in de stad is een actuele en relevante uitdaging. De relevantie om de stad, stadsplanning en haar beleid te onderzoeken is groot in het licht van groeiende steden anno 2012 en de invloed van technologie op deze groei. Deze scriptie combineert vergelijkend literatuuronderzoek vanuit New Media en Urban Studies en websiteanalyses van platformen van de gemeente Amsterdam. Het onderzoek laat zien op welke manier technologie de mogelijkheid biedt stedelingen te betrekken in een dialoog over de stad, het cultiveren van vertrouwen in de relatie met architecten en ontwikkelaars, en het legitimeren van de rol die is weggelegd voor de stedeling om complexe stedelijke dilemma’s op te lossen. Het begint met een verkenning van de omstandigheden en drijfveren die ten grondslag liggen aan het hedendaagse urbanisme, welke de relevantie toont van tijdelijkheid als permanente conditie. Vervolgens wordt er uitgebreid op de theorieën crowdsourcing en collectieve intelligente ingegaan, in het kader van stadsplanning. Toepassingen als crowdsourcing en collectieve intelligentie onderstrepen de relevantie van het nemen van ‘ownership’ door zowel stedelingen als overheid, in dit geval de gemeente Amsterdam. In de analyses van de websites wordt duidelijk dat digitale technologieën en nieuwe media per uitstek de affordances dragen ter ondersteuning van kortstondig invulling. Technologie moet echter niet als magie worden ervaren, enkel het aanbieden van een platform is geen garantie voor succes. Het betrekken van de probleemeigenaar bij het probleem en het proces om dit probleem op te lossen is een aanbeveling: het resulteert in grotere betrokkenheid van stedelingen.

Abstract

To get a deeper insight into the function of estrogen-related receptors (ERRs) and dissect underlying mechanism in Leydig cells, ERRs (type α, β and γ) were blocked or activated in testes of adult bank voles (Myodes glareolus) which show seasonal changes in the intratesticular sex hormones level. Both actively reproducing animals (long day conditions; LD) and those with regression of the reproductive system (short day conditions; SD) received intraperitoneal injections of selective ERRα antagonist 3-[4-(2,4-Bis-trifluoromethylbenzyloxy)-3-methoxyphenyl]-2-cyano-N-(5-trifluoromethyl-1,3,4-thiadiazol-2-yl)acrylamide (XCT 790) or selective ERRβ/ERRγ agonist N-(4-(Diethylaminobenzylidenyl)-N'-(4-hydroxybenzoyl)-hydrazine (DY131) (50 μ/kg bw; six doses every other day). Markedly more, XCT 790 (P0.05) but also DY131 affected interstitial tissue histology whose volume increased in both LD and SD males while seminiferous epithelium structure was untouched. Ultrastructure analysis revealed alterations in mitochondria number as well as endoplasmic reticulum and Golgi complexes volume and structure especially after ERRα blockage. Diverse and complex ERRs regulation at mRNA level and protein expression (P0.05; P0.01 and P0.001) of steroidogenic (lutropin receptor (LHR), translocator protein (TSPO), steroidogenic acute regulatory protein (StAR)) and secretory (insulin-like protein 3 (INSL3) and relaxin (RLN)) molecules were revealed in relations to endogenous estrogen level in treated males. Notably, immunolocalization of ERRs and above proteins, exclusively in Leydig cells, indicated their involvement in Leydig cell function control based on interactions with endogenous estrogen level and/or estrogen signaling via ERRs. Treatment with XCT 790 or DY131 significantly decreased (P0.05; P0.01 and P0.001) intratesticular estrogens concentration, with exception in SD DY131 males. Abstract

Background/Objectives: Fetal alcohol spectrum disorders comprise a range of neurodevelopmental disorders caused by prenatal alcohol exposure. Recent investigations have revealed that among patients with neurodevelopmental disorders, serum cobalamin (vitamin B12) levels are substantially higher than those of healthy controls. Patients with fetal alcohol spectrum disorders similarly present with higher levels of cobalamin, yet the significance of cobalamin in the pathogenesis of fetal alcohol spectrum disorders remains to be established. This study aimed to examine cobalamin and other cobalamin status markers in patients with fetal alcohol spectrum disorders in comparison with healthy controls. Methods: In total, 80 patients were enrolled in the study-41 diagnosed with fetal alcohol spectrum disorders and 39 healthy controls. The diet history method was used to assess vitamin B12 intake for three days preceding blood sampling. Total vitamin B12 (cobalamin), holotranscobalamin, methylmalonic acid and soluble transcobalamin receptor (CD320) were measured in serum samples. Results: The daily intake of vitamin B12 was higher in patients with fetal alcohol spectrum disorders compared to controls, both in the simple analysis and after adjusting for age (OR for patients with FASDs: 1.82; 95% CI: 1.16-2.87). An elevated serum cobalamin level was noted in some patients from the group with fetal alcohol spectrum disorders. No statistically significant differences were found between the groups in serum levels of cobalamin, holotranscobalamin, CD320 or methylmalonic acid. However, the correlations between cobalamin and its metabolites differed in fetal alcohol spectrum disorders as compared to those in the control group. Conclusions: Our study did not find any deficits of vitamin B12 and its metabolites in patients with fetal alcohol spectrum disorders. Further studies to investigate the role of vitamin B12 in the pathogenesis of fetal alcohol spectrum disorders should be established given the fact that both high and low levels of vitamin B12 may have negative health impacts.

Abstract

Melanoma treatments are necessary when surgically curable treatments are limited. The major challenge of targeted therapy for treating malignant melanoma is acquired drug resistance. Translation and autophagy pathways are interconnected and involved in developing cancer drug resistance. We hypothesized that coordinate inhibition of autophagy and translation would lead to a better anticancer effect. In the present study, we used chloroquine combined with two translation inhibitors (NVP-BEZ235 and CGP57380) acting at different signaling pathway levels, activating the translation. Our study was conducted for human melanoma cell lines with similar genomic alteration (BRAFV600E and PTEN loss). The combination of the drugs suppresses cell invasiveness and growth by inducing apoptosis. We showed multiple direct and indirect interactions, indicating the overlap and interaction between the translation machinery and autophagy. These data suggest that coordinated inhibition of translation and autophagy promotes apoptosis and may be a new therapeutic model for melanoma treatment.

Abstract

The balance between apoptosis and autophagy plays a key role in cancer biology and treatment strategies. The aim of this study was to assess the effect of the mTOR kinase inhibitor everolimus and chloroquine on the regulation of proliferation, caspase-3 activation, and apoptosis in melanoma cells. We studied the activity of caspase-3 and the levels of caspase-3 and -9 using the Western blot technique. Cellular apoptosis was examined using a DNA fragmentation assay, and changes in the cell nucleus and cytoskeleton were examined using fluorescence microscopy DAPI, OA/IP. We also studied the rearrangement of lipid structures using fluorescent dyes: Nile Red and Nile Blue. A low nanomolar concentration of the mTOR kinase inhibitor everolimus in combination with chloroquine activated the apoptosis process and decreased cell proliferation. These changes were accompanied by an obvious change in cell morphology and rearrangement of lipid structures. Alterations in lipid redistribution accompanying the process of apoptosis and autophagy are among the first to occur in the cell and can be easily monitored in in vitro studies. The combination of mTOR inhibitors and chloroquine represents a promising area of research in cancer therapy. It has the potential to enhance treatment efficacy through complementary mechanisms.

Recently we reported expressional alterations in 219 genes and their transcripts in Leydig cell tumors but nowadays there is still a lack of full basic biochemical characteristics of these tumors. The discovery of potential biochemical markers for tumor management from early detection, treatments, and control of therapy results may markedly supplement genetic data. Leydig cell micronodules were obtained from patients with azoospermia who were qualified for testicular biopsy. The biochemistry of Leydig cell tumors was analyzed using histological staining and spectrophotometric measurements of total proteins, carbohydrates, lipids, and nucleic acids. In addition, the levels of calcium (Ca2 +), copper (Cu2 +), zinc (Zn2 +), and selenium (Se2 +) ions were measured. When compared to healthy testis we revealed, for the first time, that in the interstitial tissue with Leydig cell tumors, great amounts of proteins, carbohydrates, lipids, and acids were dislocated from the seminiferous tubules. Measurements of organic compounds showed a decrease (P < 0.05) only in the Cu2 + content in Leydig cell tumors which may be related to their altered biochemical structure. Abstract

The importance and regulation of adrenal androgen production and signaling are not completely understood and are scarcely studied. In addition, there is still a search for appropriate animal models and experimental systems for the investigation of adrenal physiology and disease. Therefore, the main objective of the study was to evaluate the effect of luteinizing hormone (LH) signaling and selenium (Se2+) exposure on androgen adrenal signaling via canonical androgen receptor (AR), and membrane androgen receptor acting as zinc transporter (zinc- and iron-like protein 9; ZIP9). For herein evaluations, adrenals isolated from transgenic mice with elevated LH receptor signaling (KiLHRD 582G ) and adrenals obtained from rabbits used for ex vivo adenal cortex culture and exposure to Se2+ were utilized. Tissues were assessed for morphological, morphometric, and Western blot analyses and testosterone and zinc level measurements.Comparison of adrenal cortex histology and morphometric analysis in KiLHRD 582G mice and Se2+-treated rabbits revealed cell hypertrophy. No changes in the expression of proliferating cell nuclear antigen (PCNA) were found. In addition, AR expression was decreased (p < 0.001) in both KiLHRD 582G mouse and Se2+-treated rabbit adrenal cortex while expression of ZIP9 showed diverse changes. Its expression was increased (P < 0.001) in KiLHRD 582G mice and decreased (P < 0.001) in Se2+-treated rabbits but only at the dose 10 ug/100 mg/ tissue. Moreover, increased testosterone levels (P < 0.05) and zinc levels were detected in the adrenal cortex of KiLHRD 582G mice whereas in rabbit adrenal cortex treated with Se2+, the effect was the opposite (P < 0.001)., (© 2023. Abstract

Bladder cancer is one of the most prevalent cancers worldwide. Moreover, if not optimally treated, bladder cancer is a significant burden on healthcare systems due to multiple recurrences which often require more aggressive therapies. Therefore, targeted anti-cancer therapies, developed based on an in-depth understanding of specific proteins and molecular mechanisms, are promising in cancer treatment. Here, for the first time, we presented the new approaches indicating that intracellular adhesion molecule-1 (ICAM-1) may play a potential role in enhancing therapeutic effectiveness for bladder cancer. In the present study, we presented that ICAM-1 expression as well as its regulation in bladder cancer is strongly correlated with the high expression of N-cadherin. Importantly, the presence of N-cadherin and its regulator-TWIST-1 was abolished when ICAM-1 was silenced. We identified also that ICAM-1 is capable of regulating cellular migration, proliferation, and EMT progression in bladder cancer cells via the N-cadherin/SRC/AKT/GSK-3β/β-catenin signaling axis. Therefore, we propose ICAM-1 as a novel metastatic marker for EMT progression, which may also be used as a therapeutic target in bladder cancer., (© 2024. Abstract

S-allyl-L-cysteine (SAC) is a sulfur compound present in fresh garlic. The reference literature describes its anticancer, antioxidant and neuroprotective effects. Breast cancer is infamously known as one of the most commonly diagnosed malignancies among women worldwide. Its morbidity and mortality make it reasonable to complete and expand knowledge on this cancer's characteristics. Hydrogen sulfide (H 2 S) and its naturally occurring donors are well-known investigation subjects for diverse therapeutic purposes. This study was conducted to investigate the SAC antiproliferative potential and effect on three enzymes involved in H 2 S metabolism: 3-mercaptopyruvate sulfurtransferase (MPST), cystathionine γ-lyase (CTH), and cystathionine β-synthase (CBS). We chose the in vitro cellular model of human breast adenocarcinomas: MCF-7 and MDA-MB-231. The expression of enzymes after 2, 4, 6, 8, and 24 h incubation with 2.24 mM, 3.37 mM, and 4.50 mM SAC concentrations was examined. The number of living cells was determined by the MTS assay. Changes in cellular plasma membrane integrity were measured by the LDH test. Expression changes at the protein level were analyzed using Western blot. A significant decrease in viable cells was registered for MCF-7 cells after all incubation times upon 4.50 mM SAC exposure, and after 6 and 24 h only in MDA-MB-231 upon 4.50 mM SAC. In both cell lines, the MPST gene expression significantly increased after the 24 h incubation with 4.50 mM SAC. S-allyl-L-cysteine had opposite effects on changes in CTH and CBS expression in both cell lines. In our research model, we confirmed the antiproliferative potential of SAC and concluded that our studies provided current information about the increase in MPST gene expression mediated by S-allyl-L-cysteine in the adenocarcinoma in vitro cellular model for the MCF-7 and MDA-MB-231 cell lines. Further investigation of this in vitro model can bring useful information regarding sulfur enzyme metabolism of breast adenocarcinoma and regulating its activity and expression (gene silencing) in anticancer therapy.

Abstract

Despite the development of modern drugs, drug resistance in oncology remains the main factor limiting the curability of patients. This paper shows the use of a group of hydrophobic statins to inhibit drug resistance (Pgp protein). In a chemoresistance melanoma cell model, viability, necroptosis with DNA damage, the absorption of the applied pharmaceuticals, and the functional activity of the ABCB1 drug transporter after administration of docetaxel or docetaxel with a selected hydrophobic statin were studied. Taxol-resistant human melanoma cells from three stages of development were used as a model: both A375P and WM239A metastatic lines and radial growth phase WM35 cells. An animal model ( Mus musculus SCID) was developed for the A375P cell line. The results show that hydrophobic statins administered with docetaxel increase the accumulation of the drug in the tumor cell a.o. by blocking the ABCB1 channel. They reduce taxol-induced drug resistance. The tumor size reduction was observed after the drug combination was administrated. It was shown that the structural similarity of statins is of secondary importance, e.g., pravastatin and simvastatin. Using cytostatics in the presence of hydrophobic statins increases their effectiveness while reducing their overall toxicity.

50. Three generations of mTOR kinase inhibitors in the activation of the apoptosis process in melanoma cells.

Abstract

Many signaling pathways are involved in the mammalian target of rapamycin (mTOR), and this serine/threonine kinase regulates the most important cellular processes such as cell proliferation, autophagy, and apoptosis. The subject of this research was the effect of protein kinase inhibitors involved in the AKT, MEK, and mTOR kinase signaling pathways on the expression of pro-survival proteins, activity of caspase-3, proliferation, and induction of apoptosis in melanoma cells. The following inhibitors were used: protein kinase inhibitors such as AKT-MK-2206, MEK-AS-703026, mTOR-everolimus and Torkinib, as well as dual PI3K and mTOR inhibitor-BEZ-235 and Omipalisib, and mTOR1/2-OSI-027 inhibitor in single-mode and their combinations with MEK1/2 kinase inhibitor AS-703026. The obtained results confirm the synergistic effect of nanomolar concentrations of mTOR inhibitors, especially the dual PI3K and mTOR inhibitors (Omipalisib, BEZ-235) in combination with the MAP kinase inhibitor (AS-703026) in the activation of caspase 3, induction of apoptosis, and inhibition of proliferation in melanoma cell lines. Our previous and current studies confirm the importance of the mTOR signal transduction pathway in the neoplastic transformation process. Melanoma is a case of a very heterogeneous neoplasm, which causes great difficulties in treating this neoplasm in an advanced stage, and the standard approach to this topic does not bring the expected results. There is a need for research on the search for new therapeutic strategies aimed at particular groups of patients.
