Abstract

We investigated SARS-CoV-2 transmission dynamics in Italy, one of the countries hit hardest by the pandemic, using phylodynamic analysis of viral genetic and epidemiological data. We observed the co-circulation of multiple SARS-CoV-2 lineages over time, which were linked to multiple importations and characterized by large transmission clusters concomitant with a high number of infections. Subsequent implementation of a three-phase nationwide lockdown strategy greatly reduced infection numbers and hospitalizations. Yet we present evidence of sustained viral spread among sporadic clusters acting as “hidden reservoirs” during summer 2020. Mathematical modelling shows that increased mobility among residents eventually catalyzed the coalescence of such clusters, thus driving up the number of infections and initiating a new epidemic wave. Our results suggest that the efficacy of public health interventions is, ultimately, limited by the size and structure of epidemic reservoirs, which may warrant prioritization during vaccine deployment.

Abstract

In 2018 the activity under the RF R&D task 5.8 of the 2018 Work Programme was addressed to the characterization of the voltage hold off capability of different RF driver mockups on the High Voltage Radio Frequency Test Facility (HVRFTF). Particular effort was addressed to review the facility layout and to improve its high voltage RF circuit in terms of impedance matching, frequency tuning capability, RF power available and protection against fault in circuit components. Furthermore the set of diagnostic tools was improved with a plasma light sensor and a camera. Their respective signals and images, along with the gas pressure signal (already available as diagnostic since 2016) are sent to the data acquisition system.

Abstract

The High Voltage RadioFrequency Test Facility (HVRFTF) has been developed with the aim to be a simple, accessible and flexible test facility to characterize the dielectric strength in vacuum of the RF drivers of SPIDER and MITICA ion source and effectively address the issues related to the ir voltage hold off when subjected to their operating radio frequency E - fields and pressure . This report presents the results of the test campaign s carried out in 2017 on stainless steel planar circular electrodes with Argon injection. Currently the HVRFTF is able to supply RF voltage of more than 10 kV, producing E - fields up to 55 MV/m for a gap of 0.2 mm. The experimental activity, besides the collection of data relevant for the main HVRFTF scope, allowed gaining experience and addressing specific issues related to the setu p and operation of the facility.

12. Installation and site testing of the SPIDER Ion Source and Extraction Power Supplies

Abstract

SPIDER experiment, currently under construction at the Neutral Beam Test Facility (NBTF) in Padua, Italy, is a full-size prototype of the ion source for the ITER Neutral Beam (NB) injectors part of the ITER project. The Ion Source and Extraction Power Supplies (ISEPS) for SPIDER are supplied by OCEM Energy Technology s.r.l. (OCEM) under a procurement contract with Fusion for Energy (F4E) covering also the units required for MITICA and ITER injectors. The installation of SPIDER ISEPS started in June 2015 and was completed in September 2015. Functional checks started thereafter and power testing in January 2016. The formal Site Acceptance Tests (SAT), witnessed by F4E, Consorzio RFX and the ITER Organization were successfully completed in April 2016. ISEPS, with an overall power rating of 5 MVA, form a heterogeneous set of items, ranging from power transformers, medium voltage power distribution equipment at 6.6 kV to solid state power converters and including four 1 MHz radiofrequency generators of 200 kW output power. Both high voltage, down to -12 kV and high current, up to 5kA, power supplies are present. The paper presents the main features of the SPIDER ISEPS installation inside the SPIDER HV Deck, focussing in particular on non-standard technical solutions like the cable terminations on the HV Deck incoming power lines. The arrangements are described for fully checking the integration of the ISEPS local control system with the SPIDER control system are described. Details are given on non-standard site tests verifying immunity to electromagnetic interference (EMI) and the behaviour of the control electronics during fast discharges of electrostatic energy, specific to this application. Performance of ISEPS during the SAT is described and commented against the specifications, with emphasis on demonstration of the 1 hour full load capability of the radiofrequency generator.

Abstract

Interferon (IFN)-alpha affects the growth, differentiation and function of various cell types by transducing regulatory signals through the Janus tyrosine kinase/signal transducers of activation and transcription (Jak/STAT) pathway. The signalling pathways employing the mitogen-activated ERK-activating kinase (MEK) and the extracellular-regulated kinase (ERK) are critical in growth factors signalling. Engagement of the receptors, and subsequent stimulation of Ras and Raf, initiates a phosphorylative cascade leading to activation of several proteins among which MEK and ERK play a central role in routing signals critical in controlling cell development, activation and proliferation. We demonstrate here that 24-48 h following treatment of transformed T- and monocytoid cell lines with recombinant human IFN-alpha2b both the phosphorylation and activity of MEK1 and its substrates ERK1/2 were reduced. In contrast, the activities of the upstream molecules Ras and Raf-1 were not affected. No effect on MEK/ERK activity was observed upon short-term exposure (1-30 min) to IFN. The anti-proliferative effect of IFN-alpha was increased by the addition in the culture medium of a specific inhibitor of MEK, namely PD98059. In conclusion, our results indicate that IFN-alpha regulates the activity of the MEK/ERK pathway and consequently modulates cellular proliferation through a Ras/Raf-independent mechanism.
