As the urgency of PM2.5 prediction becomes increasingly ingrained in public awareness, deep-learning methods have been widely used in forecasting concentration trends of PM2.5 and other atmospheric pollutants. Traditional time-series forecasting models, like long short-term memory (LSTM) and temporal convolutional network (TCN), were found to be efficient in atmospheric pollutant estimation, but either the model accuracy was not high enough or the models encountered certain challenges due to their own structure or some specific application scenarios. This study proposed a high-accuracy, hourly PM2.5 forecasting model, poly-dimensional local-LSTM Transformer, namely PD-LL-Transformer, by deep-learning methods, based on air pollutant data and meteorological data, and aerosol optical depth (AOD) data retrieved from the Himawari-8 satellite. This research was based on the Yangtze River Delta Urban Agglomeration (YRDUA), China for 2020–2022. The PD-LL-Transformer had three parts: a poly-dimensional embedding layer, which integrated the advantages of allocating and embedding multi-variate features in a more refined manner and combined the superiority of different temporal processing methods; a local-LSTM block, which combined the advantages of LSTM and TCN; and a Transformer encoder block. Over the test set (the whole year of 2022), the model's R2 was 0.8929, mean absolute error (MAE) was 4.4523 µg/m3, and root mean squared error (RMSE) was 7.2683 µg/m3, showing great accuracy for PM2.5 prediction. The model surpassed other existing models upon the same tasks and similar datasets, with the help of which a PM2.5 forecasting tool with better performance and applicability could be established. Androgen from the testis and weak androgens from the adrenal cortex may interact with each other and affect their synthesis and secretion due to their similar functions. The purpose of this study was to investigate the compensatory effect of adrenal in rats after immunocastration and surgical castration, and the interaction between the hypothalamic–pituitary–testis (HPT) axis and the hypothalamic–pituitary–adrenal (HPA) axis. 24 male SD rats aged 8 weeks were randomly divided into three groups and accepted treatments: surgical castration group, immunocastration group and control group. In both surgical castration and immunocastration groups, the secretion of adrenocorticotropic hormone (ACTH) and dehydroepiandrosterone (DHEA) hormones was significantly increased compared with the control group (p < 0.05). In the HPT axis of the immunocastration group, the KISS1 expression was up-regulated, whereas GPR54, LH and LHR expression were down-regulated (p < 0.05). The expression levels of CRH, POMC and MC2R genes were also significantly up-regulated (p < 0.05). In addition, in the immunocastration group, the expression of adrenal LHR mRNA expression was decreased (p < 0.05). The expression of HPT axis genes and adrenal LHR were up-regulated in the surgical castration group (p < 0.05). These results show that in both immunocastration and surgical castration, adrenal androgen is increased, suggesting that the adrenal gland plays a compensatory role. Subways are widely used in major cities around the world, and subway fine particulate matter (PM 2.5 ) is the main source of daily PM 2.5 exposure for urban residents. Exposure to subway PM 2.5 leads to acute inflammatory damage in humans, which has been confirmed in mouse in vivo studies. However, the concrete mechanism by which subway PM 2.5 causes airway damage remains obscure. In this study, we found that subway PM 2.5 triggered release of pro-inflammatory cytokines such as interleukin 17E, tumor necrosis factor α, transforming growth factor β, and thymic stromal lymphopoietin from human bronchial epithelial cells (BEAS-2B) in a dose-effect relationship. Subsequently, supernatant recovered from the subway PM 2.5 group significantly increased expression of the aforementioned cytokines in BEAS-2B cells compared with the subway PM 2.5 group. Additionally, tight junctions (TJs) of BEAS-2B cells including zonula occludens-1, E-cadherin, and occludin were decreased by subway PM 2.5 in a dose-dependent manner. Moreover, supernatant recovered from the subway PM 2.5 group markedly decreased the expression of these TJs compared with the control group. Furthermore, inhibitors of toll-like receptors (TLRs) and nuclear factor-kappa B (NF-κB), as well as chelate resins (e.g., chelex) and deferoxamine, remarkably ameliorated the observed changes of cytokines and TJs caused by subway PM 2.5 in BEAS-2B cells. Therefore, these results suggest that subway PM 2.5 induced a decline of TJs after an initial ascent of cytokine expression, and subway PM 2.5 altered expression of both cytokines and TJs by activating TLRs/NF-κB-dependent pathway in BEAS-2B cells.
