10. Impacts of Apis cerana gut microbes on Nosema ceranae proliferation in Apis mellifera.

Abstract

The gut microbiota is a complex ecosystem including both beneficial and harmful microbes, which is essential for the metabolism, health and immunity of the host. Fecal microbiota transplantation (FMT) is an effective treatment for diseases of altered intestinal microbiota. Nosema ceranae, a parasitic fungus in intestinal epithelial cells, destroys the honey bee's gut integrity. This study provided the first report that the number of spores was significantly lower in the native host (Asian honey bee) compared with the novel host (European honey bee). The treatment effect of FMT was denoted by feeding the gut tissue solution from the original host to the new host. The results showed that FMT did not significantly decrease the spore load but enhanced the expression levels of immune genes in the Toll pathway. Our data confirmed that the native host inhibits N. ceranae proliferation. Our data suggest microbes of the native host could be an alternative approach to treat the parasite in the novel host. Abstract

This study aims to investigate the effect of queen cell size (9.4 mm, 9.6 mm, 9.8 mm and 10.0 mm) and mother queen caged time (0 day, 2 days and 4 days) on rearing young queens without grafting larvae. The birth weight, ovarian tubes, thorax length and width were significantly increased with the increasing diameter of queen cell size. The expression level of Vitellogenin (Vg) in young queen ovaries was also up-regulated with the increased queen cell size diameter. These results indicate that the queen cell size can strongly affect the rearing queen quality and reproductive ability. Moreover, the weight, length and width of laying eggs rose with the mother queen caging time, and young queens reared with the hatched larvae from these eggs were also increased in terms of birth weight, ovarian tubes, thorax length and width. Furthermore, the expression level of Vg in reared queen ovaries was also up-regulated with the caged time. These results reveal that the caged time of queens could significantly influence egg size and their relative queen quality.

Abstract

Triadica cochinchinensis honey (TCH) is collected from the nectar of the medicinal plant T. cochinchinensis and is considered the most important honey variety in southern China. TCH has significant potential medicinal properties and commercial value. However, reliable markers for application in the authentication of TCH have not yet been established. Herein, a comprehensive characterization of the botanical origin and composition of TCH was conducted by determining the palynological characteristics and basic physicochemical parameters. Liquid chromatography tandem-mass spectrometry (LC-MS/MS) was used to investigate the flavonoid profile composition of TCH, T. cochinchinensis nectar (TCN) and 11 other common varieties of Chinese commercial honey. (−)-Gallocatechin gallate (GCG) was identified as a reliable flavonoid marker for TCH, which was uniquely shared with TCN but absent in the other 11 honey types. Furthermore, the authentication method was validated, and an accurate quantification of GCG in TCH and TCN was conducted. Abstract

To assess the feasibility and effectiveness of retrograde intramedullary nail (RIN) revision surgeries for locking compression plate (LCP) failure in distal femoral fractures. This retrospective study included 13 patients who suffered from metalwork failures after they initially underwent open reduction and LCP fixation. In patients who eventually underwent RIN revision from January 2014 to December 2016, range of motion (ROM) and Hospital for Special Surgery (HSS) scores obtained before surgery and at the final follow-up time were analysed. The average operative time was 155 minutes (range, 120–210 minutes), and the average blood loss volume was 650 ml (range, 200–1350 ml). There were two cases of complications (15.38%): one was calf muscle vein thrombosis, and the other was a superficial infection. No deep tissue infection or deep vein thrombosis was observed post-operatively. The average follow-up time was 16 months (range, 12–24 months). All fractures healed in a mean of 6.5 months (range, 4–12 months), and one patient underwent an additional bone graft surgery that did not involve a bone graft during the RIN revision operation (this eventually healed at 12 months post-operatively). Abstract

Identifying the components of Camellia oleiferahoney and pollen and conducting corresponding toxicological tests are essential to revealing the mechanism of Camellia oleiferatoxicity to honey bees. In this research, we investigated the saccharides and alkaloids in honey, nectar, and pollen from Camellia oleifera, which were compared with honey, nectar, and pollen from Brassica napus, a widely planted flowering plant. The result showed that melibiose, manninotriose, raffinose, stachyose, and lower amounts of santonin and caffeine were found in Camellia oleiferanectar, pollen, and honey but not in B. napusnectar, pollen, and honey. Toxicological experiments indicated that manninotriose, raffinose, and stachyose in Camellia oleiferahoney are toxic to bees, while alkaloids in Camellia oleiferapollen are not toxic to honey bees. The toxicity mechanism of oligosaccharides revealed by temporal metabolic profiling is that oligosaccharides cannot be further digested by honey bees and thus get accumulated in honey bees, disturbing the synthesis and metabolism of trehalose, ultimately causing honey bee mortality.

Abstract

The electron beam welding experiments of TC4 titanium alloy and kovar alloy were carried out with Cu or Ag Interlayer. The microstructure and composition of the welded joint were analyzed by optical microscope, scanning electron microscope and x-ray diffraction. The mechanical properties of welded joints were evaluated by microhardness and tensile strength tests. The results show that when Cu is used as the interlayer, there are mainly Cu solid solution and TiFe ,Fe2Ti ,CuTi2 in the joint. The highest hardness of weld is 380HV and the tensile strength is 168 MPa. The joint welded with Ag interlayer exhibit brazing characteristics. The hardness ofjoint is slightly higher than pure silver, and the tensile strength is 243 MPa.

Abstract

The Asian honeybee Apis cerana is one of two bee species that have been commercially kept with immense economic value. Here we present the analysis of genomic sequence and transcriptomic exploration for A. cerana as well as the comparative genomic analysis of the Asian honeybee and the European honeybee A. mellifera. The genome and RNA-seq data yield new insights into the behavioral and physiological resistance to the parasitic mite Varroa the evolution of antimicrobial peptides, and the genetic basis for labor division in A. cerana. Comparison of genes between the two sister species revealed genes specific to A. cerana, 54.5% of which have no homology to any known proteins. The observation that A. cerana displayed significantly more vigilant grooming behaviors to the presence of Varroa than A. mellifera in conjunction with gene expression analysis suggests that parasite-defensive grooming in A. cerana is likely triggered not only by exogenous stimuli through visual and olfactory detection of the parasite, but also by genetically endogenous processes that periodically activates a bout of grooming to remove the ectoparasite. This information provides a valuable platform to facilitate the traits unique to A. cerana as well as those shared with other social bees for health improvement.

Abstract

Reflow soldering is the primary method for Flip-chip bonding without high bonding pressure. Reflow process during flip-chip technology in short wavelength infrared (SWIR) InGaAs/InP Focal Plane array (FPA) with indium solder was studied in this paper. In order to analyze the formation of Indium oxide and its effects on Indium bump reflow process. Indium bumps were investigated by X-ray Photoelectron Spectroscopy (XPS). The profiles of Indium bumps after reflow were observed by scanning electron microscopy (SEM). The interaction between Indium and the metal in under bump metallization (UBM) during reflow process was discussed. The current–voltage (I–V) curves of InGaAs/InP photodiodes were measured before and after the reflow process. The dark current density at 0.1 V reverse bias of InGaAs/InP photodiodes were studied. It was confirmed that the characteristics of InGaAs photodetectors haven’t degenerated after reflow in this paper.
