3. Influence of PABPC1 on translation activity in human monocytic leukemia cell line THP-1

Abstract

Objective To investigate the effect of cytoplasmic poly(A) binding protein-1(PABPC1) on translation of human monocytic leukemia cells line THP-1. Methods Specific short hairpin RNA(shRNA) was transfected into THP-1 cells. Western blot was used to verify the interference efficiency. The polysome profiling was used to examine the translation ability of THP-1 cells. Results The expression of PABPC1 protein was decreased in shPABPC1 transfected THP-1(P＜0.05). Compared with control group, the translation ability of THP-1 cells in shPABPC1 group was reduced. Conclusions Knockdown of PABPC1 may inhibit the translation in THP-1 cells and PABPC1 is a potential target for the treatment of acute myeloid leukemia (AML).

13. Effect of zeta potential on coating morphology of SiO2-coated copper powder and conductivity of copper film

Abstract

In this study, the zeta potential on the surface of copper powder was adjusted by changing the pH value and adding cetyltrimethylammonium bromide (CTAB). Silica sol was coated directly on copper powder through physical adsorption. The as-prepared SiO2-coated copper powders were mixed with organic vehicle and screen printed on low-temperature co-fired ceramic substrate to form copper films. The binder-burn-out process of the films was carried out at 550 °C for 1 h in N2/H2O atmosphere and then sintered at 910 °C for 1 h in N2atmosphere. The results proved that zeta potential had a major effect on the coating morphology of SiO2-coated copper powder. The SiO2-coated copper powder with a weight ratio of 1% showed a uniform and smooth surface. When the CTAB concentration was 1 × 10–4mol/L, the copper powder exhibited the highest zeta potential of 98.2 mV, and the copper film had the best conductivity with a sheet resistance of 1.8 mΩ/□.

Abstract

In the present study, the use of a 3 wt% Fe2O3 catalyst improved the crystallisation of polysilazane PSN2. Crystallisation peaks of higher intensity were observed for Si3N4 and SiC at 1250 °C by using a 3 wt% Fe2O3 catalyst than at 1500 °C without Fe2O3. Besides, the effects of temperature on the morphology of the samples were determined through scanning electron microscopy. Porous structures were observed at 1250 °C and these structures were transformed to flaky and near‐spherical nanoparticles as the temperatures were increased to 1350 and 1450 °C, respectively. Finally, the mechanism of Fe origination from Fe2O3 reduction, the production of a supersaturated liquid alloy, and the release and diffusion of N2 were proposed for Si3N4 formation. 26. The effect of biochanin A on cell growth, apoptosis, and migration in osteosarcoma cells

Abstract

Biochanin A has been reported to be associated to tumour cell proliferation, apoptosis and drug resistance in hepatocellular carcinoma, prostate cancer, and colorectal cancer, etc, while the relation of biochanin A on osteosarcoma is not clear. Hence, in this study, we examined the effects of biochanin A treatment on osteosarcoma cell lines MG63 and U2OS on proliferation, apoptosis, invasion and migration. We then investigated the involved molecular mechanism and found a time- and dose-dependent inhibition of cell viability in MG63 and U2OS cells with biochanin A treatment. Under the same circumstances, an increased ratio of cells in G0/G1 phase but a decreased ratio of cells in G2/M phase was observed. In addition, after biochanin A treatment, apoptotic rates clearly increased and decreased migration and invasion ability were observed in MG63 and O2OS cells. Meanwhile, relevant genes involved in cell proliferation, apoptosis, invation and migration demonstrated altered expressions in MG63 and U2OS cells. The present study supports the assumption that biochanin A has suppressive effects on osteosarcoma through regulating cell proliferation, apoptosis, invasion, and migration.

Abstract

Objective To study the clinical treatment effect of treating the facial contact dermatitis with interferential current and topical medicine delivered through acupuncture points. Methods No.2 detoxication medication was delivered to the affected through acupuncture points,one time a day for 20 min;the control group orally took cetirizine hydrochloride tablets,10 mg/d;a 3%boric acid solution was applied to patients who suffered engorgement,and a topical solution of 3%unguentum boric acid was applied to those who suffered desquamation and itching. Both groups were put under observation for 20 d. Results The effective rate of the treatment group was 83%,and the control group,only 46%,χ2=5.472,P<0.05. Conclusion Treating facial contact dermatitis with interferential current penetrating medicine and topical medicine delivered through acupuncture points can improve the efficacy of the medicaments,and prolongs the pharmacal effect on the treated area.
