4. Witten-type topological field theory of self-organized criticality for stochastic neural networks

Abstract

We study the Witten-type topological field theory(W-TFT) of self-organized criticality(SOC) for stochastic neural networks. The Parisi-Sourlas-Wu quantization of general stochastic differential equations (SDEs) for neural networks, the Becchi-Rouet-Stora-Tyutin(BRST)-symmetry of the diffusion system and the relation between spontaneous breaking and instantons connecting steady states of the SDEs, as well as the sufficient and necessary condition on pseudo-supersymmetric stochastic neural networks are obtained. Suppose neuronal avalanche is a mechanism of cortical information processing and storage \cite{Beggs}\cite{Plenz1}\cite{Plenz2} and the model of stochastic neural networks\cite{Dayan} is correct, as well as the SOC system can be looked upon as a W-TFT with spontaneously broken BRST symmetry. Then we should recover the neuronal avalanches and spontaneously broken BRST symmetry from the model of stochastic neural networks. We find that, provided the divergence of drift coefficients is small and non-constant, the model of stochastic neural networks is BRST symmetric. That is, if the SOC of brain neural networks system can be looked upon as a W-TFT with spontaneously broken BRST symmetry, then the general model of stochastic neural networks which be extensively used in neuroscience \cite{Dayan} is not enough to describe the SOC. On the other hand, using the Fokker-Planck equation, we show the sufficient condition on diffusion so that there exists a steady state probability distribution for the stochastic neural networks. Abstract

ABSTRACTBackgroundConsidering the rise of new SARS-CoV-2 variants that have reduced the efficacy of COVID-19 vaccines, the development of new antiviral medications for the disease has become increasingly necessary. In this study, ASC10, a novel antiviral prodrug, was studied in a phase 1 trial in healthy Chinese participants.Research design and methodsPart 1 involved 60 participants, receiving 50–800 mg ASC10 or placebo twice daily for 5.5 days. Part 2, with 12 participants, explored ASC10 dosing in the fed/fasting states.ResultsASC10-A, the main pharmacologically active metabolite, rapidly appeared in plasma (Tmax: 1.00–2.00 h) and decreased (t1/2: 1.10–3.04 h) without accumulation. The Cmaxand area under the plasma concentration – time curve (AUC) of ASC10-A increased dose-dependently (50–800 mg BID) over 5.5 days, with no accumulation. The Tmaxwas slightly delayed in the fed state; however, the Cmaxand AUC were similar between the fed and fasting states. Adverse events (AEs) were comparable (ASC10/placebo, 66.7%) and mostly mild (95%).ConclusionASC10 was demonstrated to be safe and well tolerated and exhibited dose-proportional exposure and minimal food effects.Clinical trial registrationwww.clinicaltrials.govidentifier is NCT05523141.

Abstract

Background Poliovirus receptor interacts with 3 receptors: T-cell immunoglobulin immunoreceptor tyrosine-based inhibitory motif, CD96, and DNAX accessory molecule 1, which are predominantly expressed on T cells and natural killer (NK) cells. Many solid tumors, including IDH wild-type glioblastoma, have been reported to overexpress poliovirus receptor, and this overexpression is associated with poor prognosis. However, there are no preclinical or clinical trials investigating the use of cell-based immunotherapies targeting poliovirus receptor in IDH wild-type glioblastoma. Methods We analyzed poliovirus receptor expression in transcriptome sequencing databases and specimens from IDH wild-type glioblastoma patients. We developed poliovirus receptor targeting chimeric antigen receptor T cells using lentivirus. The antitumor activity of chimeric antigen receptor T cells was demonstrated in patient-derived glioma stem cells, intracranial and subcutaneous mouse xenograft models. Results We verified poliovirus receptor expression in primary glioma stem cells, surgical specimens from IDH wild-type glioblastoma patients, and organoids. Accordingly, we developed poliovirus receptor–based second-generation chimeric antigen receptor T cells. The antitumor activity of chimeric antigen receptor T cells was demonstrated in glioma stem cells and xenograft models. Tumor recurrence occurred in intracranial xenograft models because of antigen loss. The combinational therapy of tyrosine-based inhibitory motif extracellular domain–based chimeric antigen receptor T cells and NK-92 cells markedly suppressed tumor recurrence and prolonged survival. Conclusions Poliovirus receptor–based chimeric antigen receptor T cells were capable of killing glioma stem cells and suppressing tumor recurrence when combined with NK-92 cells. Abstract

Quantum process may conduce to brain function. Particularly, it was proposed that the Posner molecules, Cag(PO4)6, can serve as neural qubits, which can maintain quantum entanglement between phosphorus nuclei for quite a long time. We study the process from entangled Posner molecules to the synchronized activities of brain. A diffusion model of Posner molecules with appropriate boundary conditions and the mean first arriving time of Posner molecules from one area of brain to another are obtained. We establish linkages between the quantum entangled connectivity of brain and the remaining time of entangled Posner molecules before decoherence. Based on these results, we find direct correlations between entangled Posner molecules and the functional connectivity of human brain.
