7. Angiopoietin-like 4 promotes epidermal stem cell proliferation and migration and contributes to cutaneous wound re-epithelialization

Abstract

Proliferation and migration of epidermal stem cells (EpSCs) are essential for epithelialization during skin wound healing. Angiopoietin-like 4 (ANGPTL4) has been reported to play an important role in wound healing, but the mechanisms involved are not fully understood. Here, we investigate the contribution of ANGPTL4 to full-thickness wound re-epithelialization and the underlying mechanisms using Angptl4-knockout mice. Immunohistochemical staining reveals that ANGPTL4 is significantly upregulated in the basal layer cells of the epidermis around the wound during cutaneous wound healing. ANGPTL4 deficiency impairs wound healing. H&E staining shows that ANGPTL4 deficiency significantly reduces the thickness, length and area of the regenerated epidermis postwounding. Immunohistochemical staining for markers of EpSCs (α6 integrin and β1 integrin) and cell proliferation (PCNA) shows that the number and proliferation of EpSCs in the basal layer of the epidermis are reduced in ANGPTL4-deficient mice. In vitro studies show that ANGPTL4 deficiency impedes EpSC proliferation, causes cell cycle arrest at the G1 phase and reduces the expressions of cyclins D1 and A2, which can be reversed by ANGPTL4 overexpression. ANGPTL4 deletion suppresses EpSC migration, which is also rescued by ANGPTL4 overexpression. Overexpression of ANGPTL4 in EpSCs accelerates cell proliferation and migration. Collectively, our results indicate that ANGPTL4 promotes EpSC proliferation by upregulating cyclins D1 and A2 expressions and accelerating the cell cycle transition from G1 to S phase and that ANGPTL4 promotes skin wound re-epithelialization by stimulating EpSC proliferation and migration. Our study reveals a novel mechanism underlying EpSC activation and re-epithelialization during cutaneous wound healing.

17. Gr\'obner--Shirshov bases for commutative dialgebras

Abstract

We establish Gr\"obner--Shirshov bases theory for commutative dialgebras. We show that for any ideal $I$ of $Di[X]$, $I$ has a unique reduced Gr\"obner--Shirshov basis, where $Di[X]$ is the free commutative dialgebra generated by a set $X$, in particular, $I$ has a finite Gr\"obner--Shirshov basis if $X$ is finite. As applications, we give normal forms of elements of an arbitrary commutative disemigroup, prove that the word problem for finitely presented commutative dialgebras (disemigroups) is solvable, and show that if $X$ is finite, then the problem whether two ideals of $Di[X]$ are identical is solvable. We construct a Gr\"obner--Shirshov basis in associative dialgebra $Di\langle X\rangle$ by lifting a Gr\"obner--Shirshov basis in $Di[X]$.

Abstract

Let $Di\langle X\rangle$ be the free dialgebra over a field generated by a set $X$. Let $S$ be a monic subset of $Di\langle X\rangle$. A Composition-Diamond lemma for dialgebras is firstly established by Bokut, Chen and Liu in 2010 \cite{Di} which claims that if (i) $S$ is a Gr\"{o}bner-Shirshov basis in $Di\langle X\rangle$, then (ii) the set of $S$-irreducible words is a linear basis of the quotient dialgebra $Di\langle X \mid S \rangle$, but not conversely. Such a lemma based on a fixed ordering on normal diwords of $Di\langle X\rangle$ and special definition of composition trivial modulo $S$. In this paper, by introducing an arbitrary monomial-center ordering and the usual definition of composition trivial modulo $S$, we give a new Composition-Diamond lemma for dialgebras which makes the conditions (i) and (ii) equivalent. We show that every ideal of $Di\langle X\rangle$ has a unique reduced Gr\"{o}bner-Shirshov basis. The new lemma is more useful and convenient than the one in \cite{Di}. As applications, we give a method to find normal forms of elements of an arbitrary disemigroup, in particular, A.V. Zhuchok's (2010) and Y.V. Zhuchok's (2015) normal forms of the free commutative disemigroups and the free abelian disemigroups, and normal forms of the free left (right) commutative disemigroups., Comment: 26 pages

Abstract

The Active Particle-induced X-ray Spectrometer (APXS) is one of the payloads on board the Yutu rover of Chang'E-3 mission. In order to assess the instrumental performance of APXS, a ground verification test was done for two unknown samples (basaltic rock, mixed powder sample). In this paper, the details of the experiment configurations and data analysis method are presented. The results show that the elemental abundance of major elements can be well determined by the APXS with relative deviations < 15 wt. % (detection distance = 30 mm, acquisition time = 30 min). The derived detection limit of each major element is inversely proportional to acquisition time and directly proportional to detection distance, suggesting that the appropriate distance should be < 50mm., Comment: 6 pages, 10 figures

Abstract

Tissue‐engineered skin is an effective material for treating large skin defects in a clinical setting. However, its use is limited owing to vascular complications. Human adipose tissue‐derived microvascular fragments (HaMVFs) are vascularized units that form vascular networks by rapid reassembly. In this study, we designed a vascularized bionic skin tissue using a three‐dimensional (3D) bioprinter of HaMVFs and human fibroblasts encapsulated in a hybrid hydrogel composed of GelMA, HAMA, and fibrinogen. Tissues incorporating HaMVFs showed good in vitro vascularization and mechanical properties after UV crosslinking and thrombin exposure. Thus, the tissue could be sutured appropriately to the wound. In vivo, the vascularized 3D bioprinted skin promoted epidermal regeneration, collagen maturation in the dermal tissue, and vascularization of the skin tissue to accelerate wound healing. Overall, vascularized 3D bioprinted skin with HaMVFs is an effective material for treating skin defects and may be clinically applicable to reduce the necrosis rate of skin grafts.
