16. Evolutionary history and current population relationships of the Chiru (Pantholops hodgsonii) inferred from mtDNA variation

Abstract

Total population size of the chiru (Pantholops hodgsonii), a species endemic to the Tibetan plateau, has undergone a severe decline in the past several decades. This study investigates the genetic variation, evolutionary history, and current population relationships among the 3 main populations of the chiru. Samples from Xinjiang, Xizang (Tibet), and Qinghai provinces in western China were collected and mitochondrial DNA control region sequences were analyzed. Overall nucleotide diversity ([pi]) was 2.18% and thus indicated a relatively high genetic variation. Neutrality tests suggested a significant historical population expansion (Fu's Fs = -28.152), consistent with a population decline and observed pattern of mismatch distribution. Analysis of molecular variance suggested high degrees of gene flow among all the sampled populations, of which the Xinjiang and Xizang populations were revealed to be the most genetically related. It was inferred that calving ground might play a significant role in the course of gene exchange. These results shed light on the population history and current population status of the chiru, and have strong implications for the continued conservation of the species. 17. Ethylene‐induced NbMYB4L is involved in resistance against tobacco mosaic virus in Nicotiana benthamiana.

Abstract

Several MYB transcription factors are known to play important roles in plant resistance to environmental stressors. However, the mechanism governing the involvement of MYBs in regulating tobacco mosaic virus (TMV) resistance in plants is still unclear. In this study, we found that not only is Nicotiana benthamiana MYB4‐like involved in defence against TMV, but also that the ethylene pathway participates in MYB4L‐mediated resistance. Transcription of NbMYB4L was up‐regulated in N. benthamiana infected with TMV. Silencing of NbMYB4L led to intensified TMV replication, whereas overexpression of NbMYB4L induced significant resistance to TMV. Transcription of NbMYB4L was greater in 1‐aminocyclopropanecarboxylic acid (ACC, ethylene precursor)‐pretreated plants but lower when the ethylene signalling pathway was blocked during TMV infection. Gene expression analysis showed that the transcription of NbMYB4L was largely suppressed in ETHYLENE INSENSITIVE 3‐like 1(EIL1)‐silenced plants. The results of electrophoretic mobility shift assay and chromatin immunoprecipitation‐quantitative PCR (ChIP‐qPCR) experiments indicated that NbEIL1 could directly bind to two specific regions of the NbMYB4L promoter. Furthermore, a luciferase assay revealed that NbEIL1 significantly induced the reporter activity of the MYB4L promoter in N. benthamiana. These results point to NbEIL1 functioning as a positive regulator of NbMYB4L transcription in N. benthamiana against TMV. Collectively, our work reveals that EIL1 and MYB4L constitute a coherent feed‐forward loop involved in the robust regulation of resistance to TMV in N. benthamiana. [ABSTRACT FROM AUTHOR]

Abstract

A high precision inspection method based on image mosaic was proposed for the final optics inspection in an Inertial Confinement Fusion(ICF).The image mosaic method contains image registration stage and image fusion stage.In the image registration stage,a registration method based on feature matching of local damaged area was used to overcome the exposure difference between the subimages and the lack of image information,and the moving information in the inspection system was used in image matching to improve the inspection precision and inspection efficiency.In the image fusion stage,the Poisson image fusion was adopted to eliminate the "stitching line" caused by an exposure difference and to implement the seamless mosaic.The images of final optics inspected by Final Optics Online Inspection(FOOI)was compared with the proposed method and Scale Invariant FeatureTransform(SIFT)method,and the results show that the proposed method is superior to SIFT method.The 4×4mosaic image shows that the inspection resolution of the FOOI device in the ICF has been improved significantly by proposed method.

Abstract

The dynamic process of light-induced agglomeration of carbon nanotubes (CNTs), C60 and Escherichia coli (E.coli) in aqueous solutions is demonstrated using an optical tweezers system. Based on the results, the diameter of the agglomerated region and the agglomeration rate increase with the increasing laser power. After the saturation-stable period, CNTs diffuse completely, C60 clusters only diffuse partially, and E. coli never diffuses in the agglomeration region. Theoretical analyses show that the molecular polarization and thermal diffusion of particles play crucial roles in the diffusion process. The results indicate the possibility of using light to aggregate and sort nanoparticles.
