Abstract It has been demonstrated that circ_0001874 and circ_0001971 are potential biomarkers for the diagnosis of oral squamous carcinoma (OSCC). MiR-186 was reported to serve as a tumor suppressor in OSCC, and the down-regulation of miR-186 was reported to lead to higher expression of oncogenic factor SHP2 and the activation of growth promoting signaling. In this study, we aimed to explore the possible molecular role of circ_0001874 and circ_0001971 signaling in the pathogenesis of OSCC. RT-qPCR, Western blot, online bioinformatics tools and luciferase assay were utilized to study the molecular signaling pathways of circ_0001874 and circ_0001971. MTT assay and FCM assay were performed to investigate the synergistic effect of circ_0001971 and circ_0001874 on cell proliferation and apoptosis. By observing the effect of different miRNAs on the levels of circ_0001847 and circ_0001971, it was identified that circ_0001847 and circ_0001971 respectively sponged the expression of miR-296 and miR-186 via binding to these miRNAs. Also, SHP2 mRNA and PLK1 mRNA were respectively targeted by miR-186 and miR-296-5p. We also established two signaling pathways, i.e., circ_0001971/miR-186/SHP2 and circ_0001874/miR-296-5p/PLK1, and validated the synergistic effect of circ_0001971 and circ_0001874 via observing their positive effect on cell proliferation and negative effect on cell apoptosis. The expression of miR-186 and miR-296-5p was generally lower in saliva of OSCC patients compared with that in OLK patients, while the expression of miR-186 and miR-296-5p was specifically up-regulated in saliva of OSCC patients. In conclusion, the finding of this study demonstrated that the relative level of hsa_circ_0001971 and hsa_circ_0001874 were different in the saliva of OSCC patients and could be used as predictive biomarkers for the development of OSCC. Furthermore, oncogenic effects of hsa_circ_0001971 and hsa_circ_0001874 in the development of OSCC might be, at least partially, mediated by its downstream signaling pathways including hsa_circ_0001971/microRNA-186/SHP2 and hsa_circ_0001874/microRNA-297/PLK1.

Polyploid rice and its reverted diploid show rich phenotypic variation and strong heterosis, showing great breeding value. However, the genomic differences among tetraploids, counterpart common diploids, tetraploid-revertant diploids, and hybrid descendants are unclear. In this work, we bred a new excellent two-line hybrid rice variety, Y Liang You Duo Hui 14 (HTRM12), using Haitian tetraploid self-reverted diploid (HTRM2). Furthermore, we comparatively analyzed the important agronomic traits and genome-wide variations of those closest relatives, Haitian diploid (HT2), Haitian tetraploid (HT4), HTRM2, and HTRM12 in detail, based on multiple phenotypic investigations, genome resequencing, and bioinformatics analysis. The results of agronomic traits analysis and genome-wide variation analysis of single nucleotide polymorphism (SNP), insertion–deletion (InDel), and copy number variation (CNV) show that HT4 and HTRM2 had abundant phenotypic and genomic variations compared to HT2. HTRM2 can inherit important traits and variations from HT4. This implies that tetraploid self-reverted diploid has high potential in creating excellent breeding materials and in breeding breakthrough hybrid rice varieties. Our study verifies the feasibility that polyploid rice could be used as a mutation carrier for creating variations and provides genomic information, new breeding materials, and a new way of application for tetraploid rice breeding. Exploring the effects of different additives on the improvement of newly cultivated farmland in mountainous areas can provide rational soil fertilization plans for regions lacking means of production. We conducted a paddy planting experiment in Ankang City, Shaanxi Province. Six treatments were set up, including sole chemical fertilizer (CK); fertilizer + bacteria agent (NB); chemical fertilizer + alginate bio-organic fertilizer (NO); fertilizer + fulvic acid biomass nutrient solution (NF); chemical fertilizer + acid soil conditioner (NC); fertilizer + silicon–calcium–magnesium–potassium fertilizer (NSi). We collected topsoil samples after paddy harvest, analyzed their physical, chemical, and biological properties, and selected indicators to construct a Total Data Set (TDS) and a Minimum Data Set (MDS). The Soil Quality Index (SQI) was used to evaluate the soil improvement effects after different fertilization regimes. The SQI calculated by the TDS and the MDS showed that the SQI after NF treatment was higher than that under other treatments. The SQI based on the TDS (SQITDS) and the SQI based on the MDS (SQIMDS) were significantly positively correlated with yield. The SQI calculated based on both the TDS and the MDS can objectively reflect the soil fertility quality. The paddy yield and total dry matter accumulation were the highest under the NF treatment, and the SQI was the largest. Thus, the effect of chemical fertilizer combined with fulvic acid biomass nutrient solution on soil fertility was the most significant. [ABSTRACT FROM AUTHOR]
