INTRODUCTION: During proliferative plant cell division, the new cell wall, called the cell plate, is first built in the middle of the cell and then expands outward to complete cytokinesis. This dynamic process requires coordinated movement and arrangement of the cytoskeleton and organelles. METHODS: Here we use live-cell markers to track the dynamic reorganization of microtubules, nuclei, endoplasmic reticulum, and endomembrane compartments during division and the formation of the cell plate in maize leaf epidermal cells. RESULTS: The microtubule plus-end localized protein END BINDING1 (EB1) highlighted increasing microtubule dynamicity during mitosis to support rapid changes in microtubule structures. The localization of the cell-plate specific syntaxin KNOLLE, several RAB-GTPases, as well as two plasma membrane localized proteins was assessed after treatment with the cytokinesis-specific callose-deposition inhibitor Endosidin7 (ES7) and the microtubule-disrupting herbicide chlorpropham (CIPC). While ES7 caused cell plate defects in Arabidopsis thaliana, it did not alter callose accumulation, or disrupt cell plate formation in maize. In contrast, CIPC treatment of maize epidermal cells occasionally produced irregular cell plates that split or fragmented, but did not otherwise disrupt the accumulation of cell-plate localized proteins. DISCUSSION: Together, these markers provide a robust suite of tools to examine subcellular trafficking and organellar organization during mitosis and cell plate formation in maize.

This study proposes a novel rolling bearing fault diagnosis technique based on a synchrosqueezing wavelet transform (SWT) and a transfer residual convolutional neural network (TRCNN) designed to address the difficulties of feature extraction caused by the non-stationarity of fault signals, as well as the issue of low fault diagnosis accuracy resulting from small sample quantities. This approach transforms the one-dimensional vibration signal into time–frequency diagrams using an SWT based on complex Morlet wavelet basis functions, which redistributes (squeezes) the values of the wavelet coefficients at different localized points in a time–frequency plane to the estimated instantaneous frequencies. This allows the energy to be more fully concentrated in actual corresponding frequency components. This strategy improves both the time–frequency aggregation and the resolution, which better reflects the eigenvalues of non-stationary signals. In this process, transfer learning and a residual structure are used in the training of a convolutional neural network. The resulting time–frequency diagrams, acquired using the steps discussed above, are then input to the TRCNN for diagnosis. A series of validation experiments confirmed that applying the TRCNN structure made it possible to achieve high diagnostic accuracy, even when training the network with only a small number of fault samples, as all 12 fault types from the test dataset were diagnosed correctly. Further simulation experiments demonstrated that our proposed method improved fault diagnosis accuracy compared to that of conventional techniques (with increases of 1.74% over RCNN, 1.28% over TCNN, 1.62% over STFT, 1.73% over WT, 2.83% over PWVD, and 1.39% over STFA-PD). In addition, diagnostic accuracy reached 100% during the application of three-time transfer learning, validating the effectiveness of the proposed method for rolling bearing fault diagnosis. The segmentation and recognition of power lines are crucial for the UAV-based inspection of overhead power lines. To address the issues of class imbalance, low sample quantity, and long-range dependency in images, a specialized semantic segmentation network for power line segmentation called Axial-UNet++ is proposed. Firstly, to tackle the issue of long-range dependencies in images and low sample quantity, a gated axial attention mechanism is introduced to expand the receptive field and improve the capture of relative positional biases in small datasets, thereby proposing a novel feature extraction module termed axial-channel local normalization module. Secondly, to address the imbalance in training samples, a new loss function is developed by combining traditional binary cross-entropy loss with focal loss, enhancing the precision of image semantic segmentation. Lastly, ablation and comparative experiments on the PLDU and Mendeley datasets demonstrate that the proposed model achieves 54.7% IoU and 80.1% recall on the PLDU dataset, and 79.3% IoU and 93.1% recall on the Mendeley dataset, outperforming other listed models. Additionally, robustness experiments show the adaptability of the Axial-UNet++ model under extreme conditions and the augmented image dataset used in this study has been open sourced. Continuous accurate positioning in global navigation satellite system (GNSS)-denied environments is essential for robot navigation. Significant advances have been made with light detection and ranging (LiDAR)-inertial measurement unit (IMU) techniques, especially in challenging environments with varying lighting and other complexities. However, the LiDAR/IMU method relies on a recursive positioning principle, resulting in the gradual accumulation and dispersion of errors over time. To address these challenges, this study proposes a tightly coupled LiDAR/IMU/UWB fusion approach that integrates an ultra-wideband (UWB) positioning technique. First, a lightweight point cloud segmentation and constraint algorithm is designed to minimize elevation errors and reduce computational demands. Second, a multi-decision non-line-of-sight (NLOS) recognition module using information entropy is employed to mitigate NLOS errors. Finally, a tightly coupled framework via a resilient mechanism is proposed to achieve reliable position estimation for quadruped robots. Experimental results demonstrate that our system provides robust positioning results even in LiDAR-limited and NLOS conditions, maintaining low time costs. [ABSTRACT FROM AUTHOR]
