7. In situ structure of actin remodeling during glucose-stimulated insulin secretion using cryo-electron tomography.

Abstract

Actin mediates insulin secretion in pancreatic β-cells through remodeling. Hampered by limited resolution, previous studies have offered an ambiguous depiction as depolymerization and repolymerization. We report the in situ structure of actin remodeling in INS-1E β-cells during glucose-stimulated insulin secretion at nanoscale resolution. After remodeling, the actin filament network at the cell periphery exhibits three marked differences: 12% of actin filaments reorient quasi-orthogonally to the ventral membrane; the filament network mainly remains as cell-stabilizing bundles but partially reconfigures into a less compact arrangement; actin filaments anchored to the ventral membrane reorganize from a netlike to a blooming architecture. Furthermore, the density of actin filaments and microtubules around insulin secretory granules decreases, while actin filaments and microtubules become more densely packed. The actin filament network after remodeling potentially precedes the transport and release of insulin secretory granules. These findings advance our understanding of actin remodeling and its role in glucose-stimulated insulin secretion.

Abstract

Background The effective evaluation of scientific and technological achievements in traditional Chinese medicine (TCM) is of great significance in promoting the innovative development of TCM. Previous evaluations of scientific and technological achievements in TCM often used a generalized evaluation method, leading to problems such as inappropriate indicators and limited dimensions, which is not conducive to reflecting the multiple values of achievements in a scientific and comprehensive way. Consequently, there is an urgent need to establish categorized and multi-dimensional evaluation index system for the evaluation of scientific and technological achievements in TCM in the new period. Objective To establish a scientific, categorized, reliable and comprehensive evaluation index system that reflects the principles of TCM and highlights its unique advantages aiming at the scientific and technological achievements in TCM, in accordance with the contemporary requirements for scientific and technological evaluation in China. Methods This study began in November 2022 and was completed in May 2023. A preliminary index framework was developed through literature review and focus group discussions. Delphi expert consultation was then conducted over two rounds to determine the evaluation indexes and points. The hierarchical analysis method was used to calculate the weight of each index. Results The final evaluation index system of scientific and technological achievements in TCM clinical research comprised 10 threshold indicators, 4 primary indicators, 17 secondary indicators, and 17 evaluation points. After testing, the positive coefficient of experts in both rounds of Delphi method implementation was 100%, the authority coefficient was 0.94 and 0.93, respectively. Abstract

Achieving superconductivity at room temperature could lead to substantial advancements in industry and technology. Recently, a compound known as Cu-doped lead-apatite, Pb10-xCux(PO4)6O (0.9 < x < 1.1), referred to as "LK-99", has been reported to exhibit unusual electrical and magnetic behaviors that appear to resemble a superconducting transition above room temperature. In this work we collected multiphase samples containing the nominal Pb10-xCux(PO4)6O phase (no superconductivity observed in our measured samples), synthesized by three independent groups, and studied their chemical, magnetic, and electrical properties at the microscale to overcome difficulties in bulk measurements. Through the utilization of optical, scanning electron, atomic force, and scanning diamond nitrogen-vacancy microscopy techniques, we are able to establish a link between local magnetic properties and specific microscale chemical phases. Our findings indicate that while the Pb10-xCux(PO4)6O phase seems to have a mixed magnetism contribution, a significant fraction of the diamagnetic response can be attributed to Cu-rich regions (e.g., Cu2S derived from a reagent used in the synthesis). Additionally, our electrical measurements reveal the phenomenon of current path switch and a change in resistance states of Cu2S. 36. Application of continuous nursing based on EMS management mode in preschool children with wheezing diseases

Abstract

Objective·To explore the effect of continuous nursing based on EMS [environment management (E), medicine direction (M) and self monitoring (S)] management mode on the preschool children with asthmatic diseases.Methods·A total of 67 children aged 0 to 6 years with asthmatic diseases admitted to the Department of Respiratory Medicine, Shanghai Children's Hospital, Shanghai Jiao Tong University School of Medicine from December 2019 to November 2020 were selected and divided into observation group (33 cases) and control group (34 cases) according to the random number table method, with 3 cases lost, and finally 32 cases in each group. The observation group received continuous nursing care based on EMS management mode, while the control group received routine care and discharge follow-up through the telephone. The children in the two groups were followed up at 1, 3, and 6 months after discharge to evaluate the results of Test for Respiratory and Asthma Control in Kids (TRACK) and wheezing recurrence; Medication Adherence Report Scale for Asthma (MARS-A) and Nursing Job Satisfaction Questionnaire were used to evaluate medication adherence and nursing job satisfaction 6 months after discharge.Results·There was no significant difference in demographic characteristics and clinical baseline characteristics between the two groups. Repeated measures analysis of variance showed that effects of time, groups and the interaction of groups×time on the total score of TRACK were statistically significant. The total scores of TRACK in the observation group were significantly higher than those in the control group at 1, 3, and 6 months after discharge (P=0.000). The total scores of TRACK in the two groups gradually increased with time (P=0.000). The recurrence rates of wheezing in the observation group were 25.0%, 18.7%, and 9.4% at 1, 3, and 6 months after discharge, which were significantly lower than those in the control group (50.0%, 43.7%, and 31.3%, respectively, P

42. A hierarchical residual network with compact triplet-center loss for sketch recognition

Abstract

With the widespread use of touch-screen devices, it is more and more convenient for people to draw sketches on screen. This results in the demand for automatically understanding the sketches. Thus, the sketch recognition task becomes more significant than before. To accomplish this task, it is necessary to solve the critical issue of improving the distinction of the sketch features. To this end, we have made efforts in three aspects. First, a novel multi-scale residual block is designed. Compared with the conventional basic residual block, it can better perceive multi-scale information and reduce the number of parameters during training. Second, a hierarchical residual structure is built by stacking multi-scale residual blocks in a specific way. In contrast with the single-level residual structure, the learned features from this structure are more sufficient. Last but not least, the compact triplet-center loss is proposed specifically for the sketch recognition task. It can solve the problem that the triplet-center loss does not fully consider too large intra-class space and too small inter-class space in sketch field. By studying the above modules, a hierarchical residual network as a whole is proposed for sketch recognition and evaluated on Tu-Berlin benchmark thoroughly. The experimental results show that the proposed network outperforms most of baseline methods and it is excellent among non-sequential models at present.
