Interleukin-2 (IL-2) exhibits the unique capacity to modulate immune functions, potentially exerting antitumor effects by stimulating immune responses, making it highly promising for immunotherapy. However, the clinical use of recombinant IL-2 protein faces significant limitations due to its short half-life and systemic toxicity. To overcome these challenges and fully exploit IL-2's potential in tumor immunotherapy, this study reports the development of a tumor-activated IL-2 mRNA, delivered via lipid nanoparticles (LNPs). Initially, ionizable lipid U-101 derived nanoparticles (U-101-LNP) were prepared using microfluidic technology. Subsequent in vitro and in vivo delivery tests demonstrated that U-101-LNP achieved more effective transfection than the approved ALC-0315-LNP. Following this, IL-2F mRNAs, encoding fusion proteins comprising IL-2, a linker, and CD25 (IL-2Rα), were designed and synthesized through in vitro transcription. A cleavable linker, consisting of the peptide sequence SGRSEN↓IRTA, was selected for cleavage by matrix metalloproteinase-14 (MMP-14). IL-2F mRNA was then encapsulated in U-101-LNP to create U-101-LNP/IL-2F mRNA complexes. After optimization, assessments of expression efficiency, masking, and release characteristics revealed that IL-2F with linker C4 demonstrated superior performance. Finally, the antitumor activity of IL-2F mRNA was evaluated. The results indicated that U-101-LNP/IL-2F mRNA achieved the strongest antitumor effect, with an inhibition rate of 70.3%. Immunohistochemistry observations revealed significant expressions of IL-2, IFN-γ, and CD8, suggesting an up-regulation of immunomodulation in tumor tissues. This effect could be ascribed to the expression of IL-2F, followed by the cleavage of the linker under the action of MMP-14 in tumor tissue, which sustainably releases IL-2. H&E staining of tissues treated with U-101-LNP/IL-2F mRNA showed no abnormalities. Further evaluations indicated that the U-101-LNP/IL-2F mRNA group maintained proper levels of inflammatory factors without obvious alterations in liver and renal functions. Taken together, the U-101-LNP/IL-2F mRNA formulation demonstrated effective antitumor activity and safety, which suggests potential applicability in clinical immunotherapy. Developing suitable molecular tools for monitoring SO2 and its derivatives in living organisms is attractive due to their importance for human health. Herein, the first near-infrared fluorescent probe Mito-HN with aggregation-induced emission enhancement (AIEE) characteristics for ratiometric sensing of SO2 derivatives in vitro, in cells, and in zebrafish was designed and synthesized. By connecting the electron-donating naphthopyran and electron-withdrawing hemicyanine via the alkenyl group, this probe displays a near-infrared fluorescence emission and notable solid-state luminescence with AIEE features. Based on the specific 1,4-addition reaction of HSO3−/SO32− with the C＝C double bond of Mito-HN, this probe can specifically sense HSO3− in real time with an excellent near-infrared ratiometric fluorescence effect, a low detection limit of 0.17 μM, and a fast response time of less than 30 s. Cell imaging and zebrafish imaging results demonstrate that Mito-HN can ratiometrically not only sense endogenous SO2 derivatives in mitochondria but also monitor SO2 in living organisms. Mycobacterium tuberculosis, the pathogen of tuberculosis (TB), can survive in host macrophages and induce macrophages to M2 phenotype might result in latent MTB infection. During the latent phase, the expression of MTB heat-shock protein 16.3 (Hsp16.3) is markedly increased among most of bacterial proteins, but the role of Hsp16.3 in macrophage M2 polarization is not clear. In this work, we found that macrophages incubated with 100 ng/ml MTB Hsp16.3 increased the production of Arg-1, IL-10, TGF-beta, and CD206. These results showed that MTB Hsp16.3 may induce macrophage M2 phenotype. And the interaction of Hsp16.3 with macrophages was found to depend on chemokine receptors CCRL2 and CX3CR1. Additionally, we used overexpression and silencing techniques to further verify the effect of CCRL2 and CX3CR1 on MTB Hsp16.3-induced M2 polarization macrophages. Furthermore, we explored the downstream signaling molecules of CCRL2 and CX3CR1 and we found MTB Hsp16.3 altered the signal transduction of the AKT/ERK/p38-MAPK. A novel dual-shell magnetic nanoparticle coated with a cationic covalent organic framework, containing ethidium bromide, is easily prepared, characterized and applied as an adsorbent for fast, simple and highly selective capture of nine hydroxylated polycyclic aromatic hydrocarbons in urine samples of non-smokers and smokers who smoked cigarettes with different tar yields. This is the first time that a cationic crystalline framework with high thermal and chemical stability was used for magnetic solid phase extraction. Multiple probes and quantum chemistry theory calculations were conducted to describe the versatile adsorption property directly and quantifiably. A method using high-performance liquid chromatography with a fluorescence detector based on the prepared magnetic adsorbent was established and used to investigate differences in the exposure levels of OH-PAHs in non-smokers and smokers smoking cigarettes with different tar yields. All the OH-PAH analyses present good linearities in the range of 0.1–100 ng mL−1, with R2 > 0.9965. The LOD for the 9 OH-PAHs ranged from 0.0030 to 0.0096 ng mL−1 and the LOQ ranged from 0.096 to 0.030 ng mL−1. The recoveries of the 9 OH-PAHs ranged from 93.3 to 121.3% with the RSD ranging from 0.47 to 3.53%. [Display omitted] Polypeptides are a highly promising carrier for delivering hydrophobic drugs, due to their excellent biocompatibility, non-toxicity, and non-immunogenicity. Herein, a redox and pH dual-responsive poly(ethylene glycol)-SS-b-polypeptide micelles encapsulated with disulfide bridged paclitaxel-pentadecanoic acid prodrug was developed for cancer chemotherapy. First of all, disulfide bridged paclitaxel-pentadecanoic acid prodrug (PTX-SS-COOH) and poly(ethylene glycol)-SS-b-polylysine-b-polyphenylalanine (mPEG-SS-b-PLys-b-PPhe, ESLP) were synthesized and confirmed via NMR, MS, FT-IR or GPC. After that, PTX-SS-COOH (PSH) embedded mPEG-SS-b-PLys-b-PPhe (ESLP/PSH) micelles were prepared by mixing method based on electrostatic interactions and hydrophobic forces. For comparison, mPEG-b-PLys-b-PPhe (ELP) was mixed with PTX-SS-COOH to generate another kind of micelles (ELP/PSH). The characterization of ESLP/PSH micelles through dynamic light scattering (DLS) and transmission electron microscopy (TEM) revealed a spherical structure with a diameter of approximately 170 nm. It is noteworthy that ESLP/PSH micelles displayed a high drug-loading rate of 22.84%, and excellent stability, which can be attributed to the specific interactions between the prodrug and copolymer. Drug release analysis demonstrated that the micelles exhibited a substantial release of PTX in the presence of GSH at pH 5.0, indicating a pH and redox dual responsiveness. In vivo pharmacokinetic study revealed the ESLP/PSH micelles had increased bioavailability and an extended circulation time. Ultimately, antitumor efficacy and systemic toxicity evaluation in 4 T1 tumor-bearing mice confirmed that ESLP/PSH micelles achieved the highest level of tumor growth inhibition (ca. 83%) and the lowest systemic toxicity in comparison with ELP/PSH micelles and commercialized Taxol®. Staphylococcus aureus (S. aureus) contamination commonly occurs in orthopedic internal fixation operations, leading to a delayed healing of the defected bone tissue. However, antibiotic treatments are ineffective in dealing with S. aureus bone infections due to the rise in multiple antimicrobial resistances. Here, we reported the protective effects of a recombinant five-antigen S. aureus vaccine (rFSAV) in an S. aureus infected bone defect model. In this study, we found the number of M2 macrophages markedly increased in the defect site and played a critical role in the healing of defected bone mediated by rFSAV. Mechanistically, rFSAV mediated increased level of IL-13 in bone defect site predominant M2 macrophage polarization. Abstract A new method for the determination of 19 polyaromatic hydrocarbons (PAHs) and 12 hydroxy-polyaromatic hydrocarbons (OHPAHs) in fine particulate matter (PM 2.5) was developed using atmospheric pressure gas chromatography-tandem mass spectrometry (APGC-MS/MS). Better sensitivity was achieved by APGC-MS/MS than conventional liquid chromatography or gas chromatography coupled with mass spectrometry. The method detection limits (< 0.4 pg m−3 for PAHs and < 0.076 pg m−3 for OHPAHs), recoveries (67.7–120%), repeatability (RSD of 0.9–19.1%) and linearity (R2 > 0.99) were obtained for the analysis of PM 2.5 samples. The content profile of 19 PAHs and 12 OHPAHs were successfully obtained in 55 PM 2.5 samples collected from two major Chinese megacities, Taiyuan and Guangzhou, in both summer and winter by this method. Based on the concentrations, possible emission sources for the pollutants were also identified. Abstract Calixarenes and their derivatives have been widely used in chromatographic studies due to unique host-guest recognition properties. However, stationary phases in hydrophilic interaction chromatography associated with them have yet to be studied because of their general hydrophobicity. In this paper, a tetra-proline modified calix[4]arene bonded stationary phase (DTPCSP) was prepared and characterized by FT-IR spectra, elemental analysis, solid state 13C NMR, SEM, EDS and thermogravimetric analysis. The chromatographic performance and retention mechanism of the developed stationary phase were validated in hydrophilic interaction mode and compared with a commercial column using a variety of hydrophilic or hydrophobic compounds including phenols, nucleosides and sulfonamides. The developed stationary phase exhibited better selectivity than conventional calixarene phases, and the retention behaviors of phenols on DTPCSP column were elucidated by the calculation of quantum chemistry using ODS as contrast. All the results indicate that the developed DTPCSP stationary phase can be beneficial for simultaneously separating complex hydrophilic samples with high selectivity. In this contribution, we reported the synthesis of polyhedral oligomeric silsesquioxane (POSS)-capped poly(N-vinyl pyrrolidone) (PVPy) via reversible addition-fragmentation chain transfer/macromolecular design via interchange of xanthate (RAFT/MADIX) polymerization. First, a POSS macromer bearing xanthate moiety was synthesized and was then used as the chain transfer agent to mediate the radical polymerization of N-vinylpyrrolidone (NVP). By controlling the mass ratios of the POSS-CTA to NVP, a series of the POSS-capped PVPy amphiphiles were successfully synthesized with various molecular weights. It was found that in bulks, the POSS-capped PVPy was microphase-separated and the POSS end groups were self-organized into the spherical microdomains with the size of 10~100 nm in diameter. In the solvent selective for PVPy (e.g., water), the POSS-capped PVPy was capable of self-assembling into the spherical micelles with an average diameter of 20~50 nm as evidenced by dynamic laser scattering (DLS) and transmission electron microscopy (TEM). Owing to the amphiphilicity, POSS-capped PVPy also displayed the self-assembly behavior in poly(vinylidene fluoride) (PVDF), in which the POSS cages were aggregated into 10~30 nm microdomains. In the nanocomposites of PVDF with POSS-capped PVPy, the spherical POSS microdomains were readily etched by using hydrofluoric acid, leaving the nanopores in the materials.ᅟ [ABSTRACT FROM AUTHOR]

The performance of Cd, Ni, and Cu release from river sediment at different pH was investigated by a leaching test using deionised water and river water as leachants. Visual MINTEQ geochemical software was used to model the experimental results to predict heavy metal release from sediments. The distribution and speciation of heavy metals in the sediments after leaching test were analyzed by Tessier sequential extraction. Leaching test results showed that the release amounts of Cd, Ni, and Cu are in the range of 10.2–27.3 mg·kg−1, 80.5–140.1 mg·kg−1, and 6.1–30.8 mg·kg−1, respectively, with deionised water as leachant at different pH. As far as the river water was used as the leaching solution in the test, the results show similar metal leaching contents and tendencies to that of the deionised water as leaching solution. The results of Tessier sequential extraction indicate that Cd of residual fraction easily forms obvious precipitate under the acidic condition, especially in the range of pH 0–4 with the residual of Cd over 50% of the total Cd in the sediment. The exchangeable content of Ni decreases with the increase of pH under the range of 0–5. The Fe-Mn oxide fraction of Cu in the sediments changes significantly from pH 0 to pH 9. Based on the effect of pH on the leaching of Cd, Ni, and Cu from the polluted sediment in the tests, more accurate information could be obtained to assess the risk related to metal release from sediments once it is exposed to the changed acid/alkali water conditions. [Display omitted] Lipid nanoparticles (LNPs)-based mRNA vaccines have shown great potential in the fight against the severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) pandemic. However, it remains still a challenge to improve the delivery efficiency of LNPs and the long-term stability of their mediated mRNA vaccines. Herein, a novel ionizable lipid 2-hexyldecyl 6-(ethyl(3-((2-hexyldecyl)oxy)-2-hydroxypropyl)amino)hexanoate (HEAH) derived LNPs were developed for delivering the receptor binding domain (RBD) mRNAs. In vitro cell assays confirmed that the ionizable lipid HEAH with one ether bond and one ester bond derived LNPs possessed higher mRNA delivery efficiency compared with the approved ALC-0315 with two ester bonds used in the BNT162b2 vaccine. Notably, the HEAH-derived LNPs powder lyophilized did not significantly change for 30 days after storage at 37 °C indicating good thermostability. After two RBD mRNAs of Delta and Omicron variants were encapsulated into the HEAH-derived LNPs, a bivalent mRNA vaccine was obtained as a nanoparticle formulation. Importantly, the bivalent mRNA vaccine not only resisted Delta and Omicron and also generated protective antibodies against ancestral SARS-CoV-2. The HEAH-mediated bivalent vaccine induced stronger humoral and cellular immunity than those of the ALC-0315 group. A new atmospheric pressure gas chromatography-tandem mass spectrometry (APGC-MS/MS) was developed to simultaneously separate, identify and quantify 18 nitro-polyaromatic hydrocarbons (NPAHs) in air fine particulate matter (PM 2.5 ). Compared with traditional negative chemical ionization (NCI) or electron impact ionization (EI)-MS/MS methods, APGC-MS/MS equipped with an atmospheric pressure chemical ionization (APCI) source provided better sensitivity and selectivity for NPAHs analysis in PM 2.5 .18 NPAHs were completely separated, and satisfactory linear response (R 2  > 0.99), low instrumental detection limits (0.20–2.18 pg mL −1 ) and method detection limits (0.001–0.015 pg m −3 ) were achieved. Due to the reliable performance of the instrument, only minimal sample pretreatment is needed. It ensured the satisfactory method recovery (70%–120%) and qualified repeatability (RSD: 1.1%–17.2%), which met the requirement of trace analysis of NAPHs in the real environmental PM 2.5 . Using the developed method, the actual PM 2.5 samples collected from Taiyuan, China in both summer and winter were analyzed, and 17 NPAHs but 2-nitrofluorene were detected and quantified. According to the obtained NAPH concentration results, the generation mechanism of NPAHs in PM 2.5 and the effects on NPAHs formation caused by some ambient air pollutants were preliminarily discussed: secondary photochemical reaction might be the dominant source of NPAHs in PM 2.5 collected from Taiyuan in both summer and winter; ambient air pollutants (NO 2 , SO 2 , CO) had more contribution on the NPAHs secondary formation of PM 2.5 in winter. Preparation procedure, endocytosis process and release mechanism of human serum albumin (HSA)/disulfide bond bridged paclitaxel-pentadecanoic acid conjugate (PTX-SS-C10-COOH) nanoparticles (HPTX NPs). [Display omitted] Human serum albumin (HSA) is an important nanocarrier of hydrophobic drugs due to its biocompatibility, bioresorbability, non-immunogenicity and intrinsic targetability. However, HSA/drug nanocomplexes have to experience complicated manufacturing process including multiple high-pressure homogenization and removing organic solvent under reduced pressure condition. Besides, the clinical application of these HSA/drug nanocomplexes is often limited because of their unsatisfactory stability and restricted dose. To overcome these issues, a redox-responsive paclitaxel-pentadecanoic acid prodrug conjugate embedded human serum albumin nanoparticles (NPs) was developed as a model in this report. First, PTX was activated and conjugated with 11-mercaptoundecanoic acid through a disulfide bond. The resultant disulfide bond bridged paclitaxel-pentadecanoic acid conjugate (PTX-SS-C10-COOH) was characterized by NMR and MS. After that, PTX-SS-C10-COOH dissolved in ethanol was mixed with HSA in water followed by lyophilization to generate HSA/PTX-SS-C10-COOH nanoparticles (HPTX NPs). Dynamic light scattering (DLS) and transmission electron microscopy (TEM) characterization indicated that the HPTX NPs have spherical structure with an average diameter of approximately 120 nm. The formation of HSA/PTX-SS-C10-COOH NPs was confirmed by fluorescence quenching technology, ascribed to electrostatic and hydrophobic interactions. The HPTX NPs displayed a high drug loading of 29.78 % and an entrapment efficiency of 94.16 %. Their reduced responsiveness was validated by glutathione (GSH)-triggered fast release of PTX. The pharmacokinetics, antitumor efficacy and systemic toxicity of HPTX NPs were thoroughly evaluated. The results showed that the HPTX NPs had longer retention, more effective tumor growth inhibition and lower toxicity compared with commercialized Taxol®. Importantly, the HPTX NPs could be administered at much high dose to achieve a significant tumor growth inhibition compared with Abraxane®. Crowd behavior analysis has recently attracted extensive attention in research. However, the existing research mainly focuses on investigating motion patterns in crowds, while the emotional aspects of crowd behaviors are left unexplored. Analyzing the emotion of crowd behaviors is indeed extremely important, as it uncovers the social moods that are beneficial for video surveillance. In this paper, we propose a novel crowd representation termed crowd mood. Crowd mood is established based upon the discovery that the social emotional hypothesis of crowd behaviors can be revealed by investigating the spacing interactions and the structural levels of motion patterns in crowds. To this end, we first learn the structured trajectories of crowds by particle advection using low-rank approximation with group sparsity constraint, which implicitly characterizes the coherent motion patterns. Second, rich emotional motion features are explicitly extracted and fused by support vector regression to reflect social characteristics. In particular, we construct weighted features in a boosted manner by considering the features’ significance. Finally, crowd mood is intuitively presented as affective curves to track the emotion states of the crowd dynamics, which is robust to noise, sensitive to semantic shift, and compact for pattern expressions. Extensive evaluations on crowd video data sets demonstrate that our approach effectively models crowd mood and achieves significantly better results with comparisons to several alternative and state-of-the-art approaches for various tasks, i.e., crowd mood classification, global abnormal mood detection, and crowd emotion matching. Duck infectious serositis is an acute and infectious disease caused by Riemerella anatipestifer ( R. anatipestifer ) that leads to perihepatitis, pericarditis, meningitis, and airbag inflammation in ducks, which causes serious economic losses to the global duck industry. The phoP/phoR is a novel 2-component signal transduction system first reported in gram-negative bacteria, of which phoP acts as a global regulator and virulence factor. In this study, the phoP gene from the R. anatipestifer YM strain was knocked out using homologous recombination technology and replaced with the spectinomycin resistance gene (Spec). The virulence of the R. anatipestifer YMΔ phoP strain was reduced by approximately 47,000 times compared to that of the wild-type R. anatipestifer YM strain. Ducks were immunized with live R. anatipestifer YMΔ phoP strain by subcutaneous inoculation at a dose of 106 to 107 CFU (0.2 mL per duck) and challenged with the wild-type R. anatipestifer YM strain 14 days later. The protection rate in the immunized group was 100%. The growth characteristics of ducks in the immunized and negative control groups were normal, and the research demonstrated R. anatipestifer YMΔ phoP strain have suitable immunogenicity and protective effects. Thus, the study findings suggest that the novel R. anatipestifer YMΔ phoP strain may provide a candidate for the development of a gene deletion activated vaccine against duck infectious serositis. Interactions among pedestrians usually play an important role in understanding crowd behavior. However, there are great challenges, such as occlusions, motion, and appearance variance, on accurate analysis of pedestrian interactions. In this paper, we introduce a novel social attribute-aware force model (SAFM) for detection of abnormal crowd events. The proposed model incorporates social characteristics of crowd behaviors to improve the description of interactive behaviors. To this end, we first efficiently estimate the scene scale in an unsupervised manner. Then, we introduce the concepts of social disorder and congestion attributes to characterize the interaction of social behaviors, and construct our crowd interaction model on the basis of social force by an online fusion strategy. These attributes encode social interaction characteristics and offer robustness against motion pattern variance. Abnormal event detection is finally performed based on the proposed SAFM. In addition, the attribute-aware interaction force indicates the possible locations of anomalous interactions. We validate our method on the publicly available data sets for abnormal detection, and the experimental results show promising performance compared with alternative and state-of-the-art methods. The associations of nitric oxide ( NO) with many diseases are still being discovered, and sometimes, NO levels in the body fluids are too low to be detected. Thus, the determination methods for trace nitrites and nitrates in urine and plasma of healthy volunteers and rheumatoid arthritis ( RA) patients are necessary. Using trimethylamine aminated polychloromethyl styrene nano-latex coated capillary column ( ccc-TMAPL) and field-amplified sample stacking ( FASS) injection, the sensitive open-tubular capillary electrochromatography ( OT-CEC) methods for determining trace nitrites and nitrates in urine and plasma of healthy volunteers and RA patients were set up. The limits of detection for both nitrite and nitrate were 1 ng/mL. The intra- and inter-day relative standard deviations ( RSDs) of the methods were less than 1.9%, and recoveries were between 89 and 96%. Plasma nitrate (diluted 10 times) levels were 93-301 ng/mL for healthy volunteers and 222-421 ng/mL for RA patients, respectively. Meanwhile, plasma nitrite levels were 7-18 and 40-82 ng/mL, respectively. The urinary nitrite levels were significantly elevated in RA patients. The proposed sensitive FASS-OT-CEC methods for trace nitrites and nitrates hold the promise of practical applications in a wide array of biological analyses, clinical and pathophysiology studies of the diseases related with NO hyperproduction and metabolites. To evaluate the actual performance of the H2-based polyvinyl chloride hollow fiber membrane biofilm reactor (HF-MBfR), we used HF-MBfR to remove nitrate from the nitrate contaminated groundwater with the dissolved oxygen (~6.2 mg/L) in Zhangqiu city (Jinan, China). The reactor was operated over 135 days with the actual nitrate contaminated groundwater. The result showed that maximum of nitrate denitrification rate achieved was over 133.8 g NO3--N/m3d (1.18 g NO3--N/m2d) and the total nitrogen removal was more than 95.0% at the conditions of influent nitrate 50 mg/L, hydrogen pressure 0.05 MPa, and dissolved oxygen (DO) 6.2 mg/L, with the nitrate in effluent under the value limits of drinking water. The fluxes analysis showed that the electron-equivalent fluxes of nitrate, sulfate, and oxygen account for about 81.2%, 15.2%, and 3.6%, respectively, which indicated that nitrate reduction could consume more electrons than that of sulfate reduction and dissolved oxygen reduction. The nitrate reduction was not significantly influenced by sulfate reduction and the dissolved oxygen reduction. To prevent the transmission of the novel coronavirus disease 2019 (COVID-19), China adopted nationwide lockdown measures on January 25, 2020, leading to an evident diminution in the observed air pollutants. To investigate the influence of the lockdown on atmospheric chemistry, the specific molecular composition, oxidative potential of organic aerosols (OAs) in PM 2.5 were studied using a high-resolution orbitrap mass spectrometry at a typical coal-combustion city, Linfen, in the North China Plain (NCP). The major air pollutants including PM 2.5 , PM 10 , SO 2 , NO 2 , and CO were observed to be diminished by 28.6–45.4%, while O 3 was augmented by 52.5% during the lockdown compared to those before the lockdown. A significant decrease of oxygen-containing (CHO) compounds (24.7%) associated with anthropogenic acids was observed during the lockdown, implying a reduction in fossil fuel combustion. The coal-burning related sulfur-containing organosulfates (CHOS-) and nitrooxy-sulfates (CHONS-) have also shown attenuated in both their relative abundances and anthropogenic/biogenic ratios. Amine/amide-like CHON + components have decreased by 27.6%, while nitro/nitrooxy-containing CHON- compounds have only decreased by 7.1%. Multi-source nitrogen-containing (CHN) compounds have shown a moderate elimination of 24.0%, while the identified high-condensed azaarenes have fallen from 17.7% to 14.7%, implying a potential reduction in the health risk of OAs during quarantine. The measurement of OAs' oxidative potential through dithiothreitol (DTT) assay has confirmed that as it had dropped from 0.88 nmol min−1 m−3 to 0.80 nmol min−1 m−3. High correlations were observed between the abundance of OA subgroups with the concentration of PM 2.5 after the execution of the lockdown, suggesting a potential elevation in the contribution of organic components to the total PM 2.5 level. A 25,27- bis( l-phenylalaninemethylester- N-carbonylmethoxy)-26,28-dihydroxy- para- tert-butylcalix[4]arene-bonded silica gel stationary phase was synthesized, structurally characterized and used for LC. Abstract: A continuous stirred hydrogen-based polyvinyl chloride (PVC) membrane biofilm reactor (MBfR) was investigated to remove nitrate from the drinking water. The reactor was operated over 100days, and the result showed that the average nitrate denitrification rate of 1.2g –N/m2d and the total nitrogen (TN) removal of 95.1% were achieved with the influent nitrate concentration of 50mg –N/L and the hydrogen pressure of 0.05MPa. Under the same conditions, the average rate of hydrogen utilization by biofilm was 0.031mgH2/cm2d, which was sufficient to remove 50mg –N/L from the contaminated water with the effluent nitrate and nitrite concentrations below drinking water limit values. The average hydrogen utilization efficiency was achieved as high as 99.5%. Flux analysis demonstrated that, compared to sulfate reduction, nitrate reduction competed more strongly for hydrogen electron, and obtained more electrons in high influent nitrate loading. Abstract: A hollow fiber membrane biofilm reactor (MBfR) using polyvinyl chloride (PVC) hollow fiber was evaluated in removing nitrate form contaminated drinking water. During a 279-day operation period, the denitrification rate increased gradually with the increase of influent nitrate loading. The denitrification rate reached a maximum value of 414.72gN/m3 d (1.50gN/m2 d) at an influent NO3 −–N concentration of 10mg/L and a hydraulic residence time of 37.5min, and the influent nitrate was completely reduced. At the same time, the effluent quality analysis showed the headspace hydrogen content (3.0%) was lower enough to preclude having an explosive air. Under the condition of the influent nitrate surface loading of 1.04gN/m2d, over 90% removal efficiencies of the total nitrogen and nitrate were achieved at the hydrogen pressure above 0.04MPa. The results of denaturing gel gradient electrophoresis (DGGE), 16S rDNA gene sequence analysis, and hierarchical cluster analysis showed that the microbial community structures in MBfR were of low diversity, simple and stable at mature stages; and the beta-Proteobacteria, including Rhodocyclus, Hydrogenophaga, and beta-Proteobacteria HTCC379, probably play an important role in autohydrogenotrophic denitrification. We reported the discovery and identification of emerging sulfur-containing polycyclic aromatic hydrocarbons, namely polycyclic aromatic sulfur heterocycles (PASHs), in PM 2.5 collected from two typical regions of China, Taiyuan and Guangzhou. Until now, there is no research on contamination status, sources and potential health risks of this unexpected group of organic contaminants in PM 2.5. High atmospheric concentrations (ng m−3) and significant time-dependent variations were determined in PM 2.5 of Taiyuan from 2017 to 2018. Coal combustion/secondary formation and traffic emission/secondary formation were apportioned as possible pollution sources for the PM 2.5 -bound PASHs in Taiyuan and Guangzhou, respectively. Dithiothreitol and cell viability assays were applied for evaluations of PASH-induced reactive oxygen species (ROS) production and cell toxicity based on the determined real exposure levels for adults. The results illustrated that PASHs in PM 2.5 possibly caused oxidative stress and inhibition of human bronchial epithelial cells in seriously polluted regions such as Taiyuan, suggesting that the pollutant-induced health concerns may need more investigations. Rapid increases in urban sprawl affect the observational environment around meteorological stations by changing the land use/land cover (LULC) and the anthropogenic heat flux (AHF). Based on remote sensing images and GIS technology, we investigated the impact of changes in both LULC and AHF induced by urbanization on the meteorological observational environment in the Yangtze River Delta (YRD) during 2000–2018. Our results show that the observational environments around meteorological stations were significantly affected by the rapid expansion of built-up areas and the subsequent increase in the AHF, with a clear spatiotemporal variability. A positive correlation was observed between the proportion of built-up areas and the AHF around meteorological stations. The AHF was in the order urban stations > suburban stations > rural stations, but the increases in the AHF were greater around suburban and rural stations than around urban stations. Some meteorological stations need to be relocated to address the adverse effects induced by urbanization. The proportion of built-up areas and AHF around the new stations decreased significantly after relocation, weakening the urban heat island effect on the meteorological observations and substantially improving the observational environment. As a result, the observed daily mean temperature (relative humidity) decreased (increased) around the new stations after relocation. Our study comprehensively shows the impact of rapid urban sprawl on the observational environment around meteorological stations by assessing changes in both LULC and the AHF induced by urbanization. These findings provide scientific insights for the selection and construction of networks of meteorological stations and are therefore helpful in scientifically evaluating and correcting the impact of rapid urban sprawl on meteorological observations. A dispersive SPE strategy using DMP-Cu as adsorbent combined with atomic fluorescence spectroscopy was developed for the selective capture of trace total mercury in rice sample. The adsorption mechanism showed that the Hg2+ removal process was fitted with pseudo second-order kinetics and the Langmuir adsorption model. The adsorbent was easy to be regenerated and the maximum adsorption capacity for the removal of Hg2+ was 249.5 mg g−1 at the optimal pH of 4. X-ray photoelectron spectroscopy and Raman spectra verified the selective and strong interaction between Hg2+ and thiol/nitrogen-containing functional groups of DMTZ on DMP-Cu. The trace total mercury in rice samples was determined with detection limit of 0.0125 ng mL−1 and relative standard deviation below 6%. This article considers simultaneous localization and mapping (SLAM) problem for robots in situations where accurate estimates for some of the robot poses, termed anchors, are available. These may be acquired through external means, for example, by either stopping the robot at some previously known locations or pausing for a sufficient period of time to measure the robot poses with an external measurement system. The main contribution is an efficient algorithm for selecting a fixed number of anchors from a set of potential poses that minimizes estimated error in the SLAM solution. Based on a graph-topological connection between the D-optimality design metric and the tree-connectivity of the pose-graph, the anchor selection problem can be formulated approximately as a submatrix selection problem for reduced weighted Laplacian matrix, leading to a cardinality-constrained submodular maximization problem. Two greedy methods are presented to solve this submodular optimization problem with a performance guarantee. These methods are complemented by Cholesky decomposition, approximate minimum degree permutation, order reuse, and rank-1 update that exploit the sparseness of the weighted Laplacian matrix. POSS is a class of unique building blocks for preparation of organic-inorganic nanocomposites. In this work, we investigated the influence of POSS microdomains on the properties of homogenous thermosetting blends of epoxy with poly(ethylene oxide). Herein, we first synthesized POSS-terminated poly (ethylene oxide) (PEO-POSS) and then incorporated it into epoxy to afford epoxy/PEO-POSS nanocomposites. Incorporation of PEO-POSS resulted in a microphase separation in the materials due to the self-assembly of PEO-POSS in the epoxy matrix. The influence of POSS microdomains on thermomechanical, surface and shape memory properties of nanocomposites were investigated in detail. Epoxy/PEO-POSS nanocomposites displayed higher glass transition temperature and surface hydrophobicity than epoxy/PEO blends with the same PEO content. Background: MYO1C, an actin-based motor protein, is involved in the late stages of autophagosome maturation and fusion with the lysosome. The molecular mechanism by which MYO1C regulates autophagosome-lysosome fusion remains largely unclear. Methods: Western blotting was used to determine the expression of autophagy-related proteins. Transmission electron microscopy (TEM) was used to observe the ultrastructural changes. An immunoprecipitation assay was utilized to detect protein-protein interactions. Immunofluorescence analysis was used to detect autophagosome-lysosome fusion and colocalization of autophagy-related molecules. An overexpression plasmid or siRNA against MYO1C were sequentially introduced into human breast cancer MDA-MB-231 cells. Results: We show here that cepharanthine (CEP), a novel autophagy inhibitor, inhibited autophagy/mitophagy through blockage of autophagosome-lysosome fusion in human breast cancer cells. Mechanistically, we found for the first time that MYO1C was downregulated by CEP treatment. Furthermore, the interaction/colocalization of MYO1C and F-actin with either LC3 or LAMP1 was inhibited by CEP treatment. Knockdown of MYO1C further decreased the interaction/colocalization of MYO1C and F-actin with either LC3 or LAMP1 inhibited by CEP treatment, leading to blockade of autophagosome-lysosome fusion. In contrast, overexpression of MYO1C significantly restored the interaction/colocalization of MYO1C and F-actin with either LC3 or LAMP1 inhibited by CEP treatment. Conclusion: These findings highlight a key role of MYO1C in the regulation of autophagosome-lysosome fusion through F-actin remodeling. Silver zeolite filters have potential applications in nuclear power plant filtration and emission systems due to their high-efficiency adsorption properties for radioactive methyl iodide. The adsorption performance of the filter is closely related to the flow distribution characteristics. To analyze the flow distribution characteristics of the filter element, we established a reasonable numerical calculation model and verified the model through experiments. Then based on the model, the advantages and disadvantages of different filter unit structures are analyzed. We proposed two new structural forms of filters and evaluated the performance of the filters based on the pressure distribution and flow increment curve, considering the difficulty of processing and space utilization. Comparative analysis and characteristic research have been carried out under different sizes, from the cylindrical filter to the round table filter and, finally, to the square filter. Our results show that round table and square filters have significantly improved flow distribution compared to traditional cylindrical filters. However, the structure of the round table filter is irregular, and the space utilization rate is low. Finally, through optimization design, the square filter with channel spacing L of 250 mm has the best performance. The H9N2 avian influenza virus causes reduced production performance and immunosuppression in chickens. The chicken yolk sac immunoglobulins (IgY) receptor (FcRY) transports from the yolk into the embryo, providing offspring with passive immunity to infection against common poultry pathogens. FcRY is expressed in many tissues/organs of the chicken; however, there are no reports investigating FcRY expression in chicken macrophage cells, and how H9N2-infected HD11 cells (a chicken macrophage-like cell line) regulate FcRY expression remains uninvestigated. This study used the H9N2 virus as a model pathogen to explore the regulation of FcRY expression in avian macrophages. FcRY was highly expressed in HD11 cells, as shown by reverse transcription polymerase chain reactions, and indirect immunofluorescence indicated that FcRY was widely expressed in HD11 cells. HD11 cells infected with live H9N2 virus exhibited downregulated FcRY expression. Transfection of eukaryotic expression plasmids encoding each viral protein of H9N2 into HD11 cells revealed that nonstructural protein (NS1) and matrix protein (M1) downregulated FcRY expression. In addition, the use of a c-jun N-terminal kinase (JNK) activator inhibited the expression of FcRY, while a JNK inhibitor antagonized the downregulation of FcRY expression by live H9N2 virus, NS1 and M1 proteins. Finally, a dual luciferase reporter system showed that both the M1 protein and the transcription factor c-jun inhibited FcRY expression at the transcriptional level. Taken together, the transcription factor c-jun was a negative regulator of FcRY, while the live H9N2 virus, NS1, and M1 proteins downregulated the FcRY expression through activating the JNK signaling pathway. Background: Triple-negative breast cancer (TNBC) is often aggressive and associated with a poor prognosis. Due to the lack of available targeted therapies and to problems of resistance with conventional chemotherapeutic agents, finding new treatments for TNBC remains a challenge and a better therapeutic strategy is urgently required. Methods: TNBC cells and xenograft mice were treated with a combination of chloroquine (CQ) and isorhamnetin (IH). Mitochondrial fission, apoptosis, and related signaling pathways were determined by flow cytometry, immunofluorescence, and related molecular biological techniques. Results: The inhibition of autophagy/mitophagy by CQ selectively enhances IH-induced mitochondrial fission and apoptosis in TNBC cells but not in estrogen-dependent breast cancer cells. These events were accompanied by mitochondrial translocation of Bax and the release of cytochrome c. Mechanistically, these effects were associated with oxidative stress-mediated phosphorylation of CaMKII (Thr286) and Drp1 (S616), and subsequent mitochondrial translocation of CaMKII and Drp1. The interruption of the CaMKII pathway by genetic approaches (e.g. CaMKII mutant or siRNA) attenuated combination-mediated mitochondrial fission and apoptosis. The combination of CQ/IH was a marked inhibitor tumor growth, inducing apoptosis in the TNBC xenograft mouse model in association with the activation of CaMKII and Drp1 (S616). Conclusions: Our study highlights the critical role of ROS-mediating CaMKII/Drp1 signaling in the regulation of mitochondrial fission and apoptosis induced by combination of CQ/IH. These findings also suggest that IH could potentially be further developed as a novel chemotherapeutic agent. The effects of adding organic carbon on the performance of different partial nitrification‐anammox (PNA) process (the activated sludge process and biofilm process) were studied, especially nitrogen removal, functional microbial activity, and microbial community structure. The potential influences of quorum sensing (QS) on the nitrogen metabolism were also analyzed. The results showed that the addition of organic carbon in biofilm systems could reduce total nitrogen (TN) removal percentages, while in activated systems it could increase TN removal percentages. The TN removal percentages in SBBR‐CN (the biofilm system with addition of organic carbon) and SBR‐CN (the activated sludge system with addition of organic carbon) were 15% and 45%, respectively, and those in SBBR‐N (the biofilm system without addition of organic carbon) and SBR‐N (the activated sludge system without addition of organic carbon) were 75% and 21%, respectively. Batch experiments have proved that organic carbon inhibited the activities of nitrite‐oxidizing bacteria (NOB) and anaerobic ammonia oxidation (anammox) bacteria, and organic carbon could promote the activity of denitrifying bacteria in activated sludge systems. Compared with activated sludge systems, biofilm systems could protect the activity of anammox bacteria. The relative abundances of ammonia oxidizing bacteria (AOB) and anammox bacteria were decreased, while the relative abundances of denitrifying bacteria (Thauera) were increased with the addition of organic carbon. The biofilm systems were more conducive to the growth of anammox bacteria. Metagenomics revealed that the same bacteria might be involved in different nitrogen metabolism, and nitrogen metabolism was achieved through the complex cooperation among functional bacteria. Besides, functional bacteria involving in the nitrogen metabolism had genes related to QS, indicating that QS might affect the nitrogen metabolism by regulating the functional bacteria activity. The outbreak of COVID-19 has caused concerns globally. To reduce the rapid transmission of the virus, strict city lockdown measures were conducted in different regions. China is the country that takes the earliest home-based quarantine for people. Although normal industrial and social activities were suspended, the spread of virus was efficiently controlled. Simultaneously, another merit of the city lockdown measure was noticed, which is the improvement of the air quality. Contamination levels of multiple atmospheric pollutants were decreased. However, in this work, 24 and 14 air fine particulate matter (PM 2.5) samples were continuously collected before and during COVID-19 city lockdown in Linfen (a typical heavy industrial city in China), and intriguingly, the unreduced concentration was found for environmentally persistent free radicals (EPFRs) in PM 2.5 after normal life suspension. The primary non-stopped coal combustion source and secondary Cu-related atmospheric reaction may have impacts on this phenomenon. The cigarette-based assessment model also indicated possible exposure risks of PM 2.5 -bound EPFRs during lockdown of Linfen. This study revealed not all the contaminants in the atmosphere had an apparent concentration decrease during city lockdown, suggesting the pollutants with complicated sources and formation mechanisms, like EPFRs in PM 2.5 , still should not be ignored. Pullulan, with its excellent characteristics of film-forming, water solubility, and biodegradability, is attracting more and more attention in agricultural products preservation. However, high pullulan production cost largely restricts its widely application due to its low production. In order to improve pullulan production by Aureobasidium pullulans NCPS2016, the medium was optimized using single factor experiment and response surface methodology. Based on the single factor experiments, the contents of soybean meal hydrolysates (SMHs), (NH4)2SO4, and K2HPO4·3H2O were considered to be main factors influencing the extracellular polysaccharide (EPS) production, and were further optimized by Box–Behnken design. The optimal content of SMHs of 7.71 g/L, (NH4)2SO4 of 0.35 g/L, and K2HPO4·3H2O of 8.83 g/L were defined. Finally, EPS production of 59.8 g/L was obtained, 39% higher in comparison with the production in the basal medium. The purified EPS produced by NCPS2016 was confirmed to be pullulan. This is the first time fructose is reported to be the optimal carbon source for pullulan production by Aureobasidium pullulans, which is of great significance for the further study of the mechanism of the synthesis of pullulan by NCPS2016. A novel method of atmospheric pressure gas chromatography-tandem mass spectrometry (APGC-MS/MS) for the analysis of organochlorine pesticides (OCPs) in human serum was developed. Better sensitivity and selectivity were achieved compared to conventional gas chromatography-mass spectrometry (GC-MS) and GC-MS/MS. The limits of detection (LODs) of OCPs ranged from 0.22 to 0.90 pg/mL for the analysis in serum samples. The recoveries and repeatability of the developed method were evaluated from the analysis of spiked serum at three levels (0.05, 0.5, and 5 ng/mL). The recoveries ranged from 84.5 to 118.2%, and the relative standard deviations (RSDs) were less than 20%. The developed APGC-MS/MS method was applied to measure OCPs in pooled human serum from the interlaboratory comparison study. The obtained results of OCPs were within the acceptable ranges. Abstract A new stationary phase (TTCPSP) has been prepared by covalently bonding 1,3,5-triazine tetrad-aza cyclophanes (TTCP) onto silica gel using 3-aminopropyl triethoxysilane as the coupling reagent. The stationary phase developed in this way was characterized by IR spectroscopy, elemental analysis and thermogravimetric analysis. The chromatographic performance and retention mechanisms of the new stationary phase were validated and compared with octadecylsilyl (ODS) stationary phases using a variety of compounds including monosubstituted benzenes, polycyclic aromatic hydrocarbons (PAHs) and flavonoids. The results indicate that the 1,3,5-triazine tetrad-aza cyclophanes (TTCPSP) stationary phase addresses solutes with multiple interactions, such as hydrophobic, hydrogen bonding, π-π, and inclusion interactions. Abstract Toxic chemicals such as flame retardants are associated with atmospheric fine particles (PM 2.5), which are an important pollutant in the world. In this study, a sensitive method using gas chromatography-tandem triple quadrupole mass spectrometry combined with ultrasonic-assisted extraction and multi-phase column purification was developed for simultaneously determination of 29 flame retardants including halogenated flame retardants (HFRs) and organophosphate flame retardants (OPFRs) in PM 2.5 samples. Extraction time, as well as the type and volume of elution solvent were optimized. The recoveries of all target flame retardants from PM 2.5 samples were in the range of 70%‒130%, with good repeatability and reproducibility (0.39%‒17.4%). The instrumental detection limits (IDLs) and method quantitation limits (MQLs) of HFRs were in the range of 0.004‒5.44 pg and 0.002‒2.52 pg m−3, while those of OPFRs were 0.003‒0.23 pg and 0.07‒5.41 pg m−3, respectively. The developed method has been successfully applied for analysis of target flame retardants in atmospheric PM 2.5 collected from the cities of Guangzhou and Taiyuan, China. HFRs were found at concentrations of 45.7‒1230 pg m−3 and 51.2‒82.3 pg m−3 in Guangzhou and Taiyuan. The concentrations of HFRs were lower than those of OPFRs, which were detected at concentrations of 6004‒49,800 pg m−3 and 16,741‒34,346 pg m−3 in Guangzhou and Taiyuan. Accordingly, the analytical method is robust and useful for further elucidating the contamination profiles of flame retardants in PM 2.5. Abstract Exposure to airborne fine particulate matter (PM 2.5) is associated with various adverse effects. However, the molecular mechanism involved in PM 2.5 -elicited energy metabolic reprogramming and the toxic chemical determinants within PM 2.5 are not well elucidated. In this study, nontargeted and targeted metabolomics research were conducted to investigate the overall metabolic changes and relevant toxicological pathways caused by Taiyuan winter total PM 2.5 and its water soluble and organic soluble fractions in human lung bronchial epithelial cells (BEAS-2B). The results showed that significant metabolome alterations in BEAS-2B cells were observed after the exposure of total PM 2.5 and its organic soluble fraction. Purine metabolism, arginine and proline metabolism, glutathione (GSH) metabolism, tricarboxylic acid (TCA) cycle and glycolysis were mainly affected. Along with a significant increase of reactive oxygen species (ROS), malondialdehyde (MDA), nitric oxide (NO) and pro-inflammatory cytokines (TNF-α, IL-6 and IL-1β), obvious metabolic phenotype remodeling from oxidative phosphorylation to glycolysis was found in BEAS-2B cells treated with total PM 2.5 and its organic soluble fraction. Compared with water soluble fraction, organic soluble fraction was found to play the dominant role in PM 2.5 toxicity. Abstract The Chinese government is encouraging domestic cement producers to move from traditional coal power sources to the co-processing of waste as the primary energy source for the industry. In this study, 32 samples collected from the soil surrounding a cement plant in Beijing were analyzed for the presence of 16 U.S. EPA priority polycyclic aromatic hydrocarbons (PAHs) and 12 heavy metals. Ten samples were selected for polychlorinated dibenzo-p-dioxins and polychlorinated dibenzofurans (PCDD/Fs) analysis. The pollution distribution patterns, sources, and potential risks to human health and the environment were investigated and evaluated. The highest concentrations of PCDD/Fs occurred 1200 m downwind from the cement plant. The levels of ∑16 PAHs ranged from 130.6 to 1134.3 μg kg−1 in the sampled soils. Source identification analysis suggested that the cement plant was the most likely source of PAH contamination. The concentrations of most of the heavy metals detected in the sampled soils were close to background levels, except for the levels of cadmium (Cd) and mercury (Hg), which were, on average, two times and six times higher than background values, respectively. The co-incineration of sludge, coal, and hazardous waste in the cement plant is a major contributing cause for the high levels of Hg in the surrounding soil. Risk assessment models, both the incremental lifetime cancer risks (ILCRs) for PAHs and the potential ecological risk index (RI) for heavy metals, indicate potential risks to the population and the environment surrounding the cement plant. This study reports a novel strategy for the preparation of porous aromatic framework (PAF-6) coated magnetic nanoparticles (PAF-6 MNPs) using cyanuric chloride as a planar trigonal basis upon which to build a linear piperazine linker unit. The PAF-6 MNPs were examined as an efficient solid-phase extraction (SPE) sorbent for enrichment of trace organic pollutants including phenol, 2,4,6-trinitrophenol, naphthalene, naphthol, bisphenol A, 2,4-dichlorophenol and 3-nitrochlorobenzene in water. The high-performance liquid chromatography detection limits of such analytes were in the range of 0.08–5.02 ng/mL and recoveries were found to be 84.0–94.0% in well water, tap water, river water and wastewater. The main toxic components of cigarette smoke, including phenolic compounds and benzo[a]pyrene, are efficiently adsorbed by PAF-6 MNPs, and over 50% of such toxins are removed. Theoretical computations were performed to understand the molecular interaction mechanism between PAF-6 and such analytes. The results demonstrate that the PAF-6 MNPs sorbents show excellent adsorption of phenols, polycyclic aromatic hydrocarbons and nitroaromatics based on multiple π-π stacking and hydrogen-bond interactions. At present, achieving good storability and water damage resistance remains challenging for cold-patching asphalt mixtures (CAMs). To address this issue, this study selects styrene–ethylene–butadiene–styrene copolymer (SEBS) and diesel as a modifier and diluent, respectively, to improve the water stability and storability of CAMs. The diesel oil content is determined through the Brookfield rotational viscosity test, and the modifier content is obtained through the Marshall stability test. With the empirical formula method, paper trail test, and modified Marshall test, mixed designs of CAMs modified with and without SEBS are established to determine the best cold-patching asphalt content. On this basis, the modification effect of SEBS is verified by comparing the test results of the modified and unmodified CAMs, and the water stability and Marshall stability tests are conducted before and after CAM storage, respectively. Results show that the optimum contents of SEBS and diesel oil are 7.5% and 40% of the base asphalt weight, respectively, and the best modified asphalt content is 4.6% of the mineral material weight in CAM. The Marshall residual stability and freeze–thaw splitting strength ratio of the 7.5% SEBS-modified CAM are increased by 20.1% and 15.7%, respectively, relative to the unmodified CAM, and the storage performance requirement of at least two months can be guaranteed. In this report, lipid nanoparticles encapsulated with new shRNA PKN3 (SS-LNP/shPKN3) with redox-responsiveness were developed in order to specifically down-regulate the expression of PKN3 for breast cancer treatment. The SS-LNP/shPKN3 was prepared by microfluidic method using disulfide bonds based ionizable lipid as main component. The as-prepared SS-LNP/shPKN3 lipid nanoparticles were characterized via using dynamic light scattering (DLS) and transmission electron microscopy (TEM). The results indicated that the obtained SS-LNP/shPKN3 exhibited uniform particle size and regular spherical morphology. Moreover, glutathione (GSH) triggered release of shPKN3 confirmed the redox-responsiveness of the SS-LNP/shPKN3. Finally, the anti-tumor effect of SS-LNP/shPKN3 was evaluated against MDA-MB-231 cells and derived xenograft tumor bearing mice. It was found that the SS-LNP/shPKN3-2 had the highest PKN3 protein inhibition rate of 60.8% and tumor inhibition rate of 62.3%. A novel multi-function and mixed-mode stationary phase based on 4-chloro-6-pyrimidinylferrocene modified silica (NFcS) was synthesized and characterized by infrared spectroscopy, elemental analysis and thermogravimetric analysis. Linear solvation energy relationship method was successfully employed to evaluate the new phase with a set of 27 solutes including aromatic and aliphatic compounds. Multiple mechanisms including hydrophobic, π–π, hydrogen-bonding, charge-transfer, acid–base equilibrium and anion-exchange interactions are involved. Based on these interactions, successful separation could be achieved among polycyclic aromatic hydrocarbons, mono-substituted benzenes, aromatic amines, phenols, quinolines, pyridines and nucleosides in reversed-phase (RP) or normal-phase (NP) chromatography. Inorganic anions were also shown to be individually separated in anion-exchange chromatography by using the same column. Moreover, the results here also demonstrated that NFcS based stationary phase could effectively reduce the adverse effect of residual silanol in the separation process. Such stationary phase with characteristics of multi-interaction mechanism and mixed-mode separation is potential for the analysis of complex samples. Matrix deposition plays a critical role in image quality of matrix-assisted laser desorption/ionization mass spectrometry imaging (MALDI-MSI). To improve the ionization efficiency and overcome the limitation of traditional matrix deposition methods in the face of difficult-to-sublimate or difficult-to-dissolve matrix, covalent organic frameworks (COFs) named COF-DhaTab was successfully synthesized and firstly used as matrix film. It was fabricated by imprinting of sieved COF-DhaTab powder on the surface of a double-sided adhesive tape. Outstanding reproducibility and uniformity of COF-DhaTab film were demonstrated by relative standard deviation (RSD) within 8.37% and 7.71% from dot-to-dot and plate-to-plate, respectively. With the introduction of double-sided adhesive tape, water contact angle (WCA) of COF-DhaTab film increased from 55° to 141°, resulting in significant suppression of analyte diffusion. Moreover, the intensity of potassium perfluorooctanic sulfonate (PFOS, C 8 F 17 SO 3 −, m/z 498.93) was 9.3 × 105, more than six hundred times higher than that using DHB matrix. This enhancement was attributed to the rough surface and multiple branches of the synthesized COF-DhaTab. To verify the ability of COF-DhaTab film as substrate, the spatial distribution of PFOS in zebrafish, rat liver and kidney tissues was explored. Superior imaging capability was displayed with high-spatial resolution and reliable location distribution. These results not only demonstrate the outstanding ability of COF-DhaTab as matrix for MALDI-MS and MALDI-MSI, but also provide a facile approach for fabrication of novel matrix films for MALDI-MSI.
