17. Characterization of Fungal Species Isolated from Cankered Apple Barks Demonstrates the Alternaria alternata Causing Apple Canker Disease.

Abstract

Apple canker disease, also named as apple Valsa canker, is one of the most destructive diseases for apples (Malus domestica Borkh.). Cytospora/Valsa spp. are the dominant causal agent of this disease, but many studies have revealed that fungi from some other genus can also cause typical apple canker symptoms. In this study, we performed fungal pathogen isolation from cankered 'Fuji' apple barks. Six representative morphologically different fungi (Strain 1–6) were further subjected to ITS sequencing and evolutionary analysis. Molecular identification results revealed that Strains 1–6 are Cytospora mali, Fusarium cf. solani, Alternaria alternata, C. mali, Diplodia seriata and F. proliferatum, respectively. All these fungi have been reported to be causal agents of apple diseases. By inoculating fungal plugs onto trunks of 'Fuji' apple trees, the pathogenicity of the six fungi were accessed. Only the inoculations of the two C. mali strains (Strain 1 and Strain 4) and the A. alternata strain (Strain 3) resulted in typical apple canker symptoms in trunks. It is worth noting that Strain 1 caused much more severe canker symptoms and higher pathogenicity incidence than the other two fungi. A. alternata has been identified as a pathogen causing diseases on apple fruits and leaves. By further assessing its pathogenicity on apple fruits and leaves, we verified that it can also cause typical fruit rot and leaf spot symptoms. To the best of our knowledge, this is the first report on apple canker disease caused by A. alternata in China. Abstract

Climate warming can lead to permafrost degradation, potentially resulting in slope failures such as retrogressive thaw slumps (RTSs). The formation of and changes in RTSs could exacerbate the degradation of permafrost and the environment in general. The mechanisms of RTS progression and the potential consequences on the analogous freeze–thaw cycle are not well understood, owing partly to necessitating field work under harsh conditions and with high costs. Here, we used multi-source remote sensing and field surveys to quantify the changes in an RTS on Eboling Mountain in the Qilian Mountain Range in west-central China. Based on optical remote sensing and SBAS-InSAR measurements, we analyzed the RTS evolution and the underlying drivers, combined with meteorological observations. The RTS expanded from 56 m2 in 2015 to 4294 m2 in 2022, growing at a rate of 1300 m2/a to its maximum in 2018 and then decreasing. Changes in temperature and precipitation play a dominant role in the evolution of the RTS, and the extreme weather in 2016 may also be a primary contributor to the accelerated growth, with an average deformation of −8.3 mm during the thawing period, which decreased slope stability. The RTS evolved more actively during the thawing and early freezing process, with earthquakes having potentially contributed further to RTS evolution. 31. Characterization and functional analysis of gerberaplant defensin(PDF) genes reveal the role of GhPDF2.4in defense against the root rot pathogen Phytophthora cryptogea

Abstract

Gerbera (Gerbera hybrida), a major fresh cut flower crop, is very susceptible to root rot disease. Although plant defensins (PDFs), a major group of plant antimicrobial peptides, display broad-spectrum antifungal and antibacterial activities, PDFgenes in gerbera have not been systematically characterized. Here, we identified and cloned nine PDFgenes from gerbera and divided them into two classes based on phylogenetic analysis. Most Class I GhPDFgenes were highly expressed in petioles, whereas all Class II GhPDFgenes were highly expressed in roots. Phytophthora cryptogeainoculation strongly upregulated all Class II GhPDFgenes in roots and upregulated all Class I GhPDFgenes in petioles. Transient overexpression of GhPDF1.5and GhPDF2.4inhibited P. cryptogeainfection in tobacco (Nicotiana benthamiana) leaves. Transient overexpression of GhPDF2.4, but not GhPDF1.5, significantly upregulated ACOand LOXgene expression in tobacco leaves, indicating that overexpressing GhPDF2.4activated the jasmonic acid/ethylene defense pathway and that the two types of GhPDFs have different modes of action. Prokaryotically expressed recombinant GhPDF2.4 inhibited mycelial growth and delayed the hyphal swelling of P. cryptogea, in vitro, indicating that GhPDF2.4 is a morphogenetic defensin. Moreover, the addition of GhPDF2.4 to plant culture medium alleviated the root rot symptoms of in vitro-grown gerbera seedlings and greatly reduced pathogen titer in P. cryptogea-inoculated gerbera roots in the early stages of treatment. Our study provides a basis for the use of GhPDFs, especially GhPDF2.4, for controlling root rot disease in gerbera.

Abstract

Oxidative stress contributes greatly to initiation and progression of liver injury. Activation of nuclear-factor erythroid 2-related factor 2 (Nrf2) has been considered as an attractive strategy for preventing and treating the oxidative damage related to liver injury. This study aimed to find an efficacious agent to activate Nrf2/HO-1 signaling pathway from clinically used therapeutic agents and to characterize the usefulness for preventing and treating CCl4-induced acute liver injury. For this purpose, a series of clinically used therapeutic agents were collected and their activation potentials on Nrf2 were assayed by using 293T-Nrf2-luc cell line. Among all tested therapeutic agents, midazolam was found with good Nrf2 activation effect and this agent could significantly ameliorate CCl4-induced damage to HepG2 cells. In vivo animal tests showed that pretreatment with midazolam reduced the liver pathological tissue damage and the serum levels of ALT and AST in CCl4-induced liver injury mice. Further investigations showed that midazolam could strongly up-regulate the expression of both Nrf2 and HO-1 in the mice liver, accompanied by increasing of the levels of antioxidant enzyme SOD and reducing the production of MDA, as well as reducing the pro-inflammatory cytokines (IL-6, TNF-α) secretion. Collectively, our results clearly demonstrate that midazolam can ameliorate CCl4-induced acute liver injury and oxidative stress via activating the Nrf2 signaling pathway.
