In the era of rapid technological development, the integration of artificial intelligence and virtual reality technology has gradually become a research hotspot. Interpersonal interaction, as an important part of virtual reality, also presents opportunities and challenges. The study builds the virtual interpersonal interaction model using the emotion model and hierarchical emotion as the learning algorithm after exploring the key technology of virtual interpersonal interaction based on the UGC model. Finally, the complex interpersonal interaction in virtual reality is simulated to examine the use of virtual community users and the factors affecting interpersonal interaction in virtual communities. According to the results, the percentage of female users of virtual communities is 14.28 percentage points higher than that of male users. The majority of people aged 18-40 who use virtual communities account for 88.01% of the population. In terms of education distribution, the group of bachelor’s degree holders is the group that uses virtual communities the most frequently, accounting for 66.52%. The study concluded that 8 out of the 11 hypotheses it proposed were valid. Among them, hypothesis 3 and hypothesis 11 were supported by 0.01 significance, and hypothesis 9 had a significance of 0.017, which reached 0.05 significance level.

Virtual reality, as a computer simulation system that can create virtual environments, is a fusion of multiple key technologies that are applied in different fields with important practical value and significance. This paper focuses on optimizing the artificial intelligence-based dynamic scene generation and perception mechanism in virtual reality. Firstly, a virtual dynamic scene generation method combining scene management technology and image rendering technology is proposed. In order to optimize the dynamic scene perception mechanism in virtual reality, a perception model based on spatial interaction perception and based on the analysis of user dynamic interaction is proposed. A two-layer perception management strategy is proposed. The paper finally uses the virtual reality technology proposed in this paper to create a Chinese classical garden attraction in a scenic spot. It carries out modeling accuracy assessment and comparison experiments and invites garden professionals and tourists to evaluate the overall effect of the virtual scene. The survey results show that more than 80% of the tourists are satisfied with the generation of the dynamic scene and think that it has spatial continuity and immersive experience.

WWP2 is a HECT-type E3 ubiquitin ligase that regulates various physiological and pathological activities by binding to different substrates, but its role in atherosclerosis (AS) remains largely unknown. The objective of the present study is to investigate the role and underlying molecular mechanisms of WWP2 in endothelial injury. We found that WWP2 expression is significantly decreased in Apolipoprotein E (ApoE)–/– mice. Overexpression of WWP2 attenuates oxidative stress and inflammation in AS mice, while knockdown of WWP2 has opposite effects. WWP2 overexpression alleviates oxidized low-density lipoprotein (ox-LDL)-induced human umbilical vein endothelial cell (HUVEC) injury, evidenced by the decreased oxidative stress levels and the secretion of inflammatory cytokines. Programmed cell death 4 (PDCD4) is identified as a potential substrate of WWP2. Co-immunoprecipitation (Co-IP) further demonstrates that WWP2 interacts with PDCD4, which is enhanced by ox-LDL treatment. Furthermore, the level of PDCD4 ubiquitination is significantly increased by WWP2 overexpression under the condition of MG132 treatment, while WWP2 knockdown shows opposite results. Subsequently, rescue experiments demonstrate that WWP2 knockdown further aggravates oxidative stress and inflammation in ox-LDL-treated HUVECs, while knockdown of PDCD4 alleviates this effect. Moreover, the use of sn-protoporphyrin (SnPP), an inhibitor of HO-1 pathway, confirms that PDCD4 enhances endothelial injury induced by ox-LDL through inhibiting HO-1 pathway. In conclusion, our results suggest that WWP2 protects against atherosclerosis progression via the PDCD4/HO-1 pathway, which may provide a novel treatment strategy for atherosclerosis.

Aimed at the problem of intelligent classification of crack damage in different positions of the plate, a method for plate crack damage detection based on long short-term memory (LSTM) neural network is proposed. The Abaqus secondary development is used to build the plate crack damage model and calculate the acceleration response of the plate under Gaussian white noise excitation. The data set is generated by data augmentation, and the influence of noise on damage detection is considered. An intelligent crack detection model based on LSTM is established, which directly takes the acceleration response of the plate as the input and does not require additional damage feature extraction. With the goal of minimizing prediction error, the hyperparameter of the model is selected and the model configuration is optimized. The comparison of the multi-layer perceptron model and the multi-layer perceptron model based on wavelet packet transform shows that the LSTM model proposed in this paper has a higher damage location accuracy and a better applicability in plate crack detection.

The lubrication model of the slope-platform pad thrust bearing with different structural parameters are built,the effects of the slope angle,oil film thickness and angular speed on bearing temperature field are studied. And the effects of the speed on the load capacity of the bearing is also analyzed under conjugate heat transfer circumstance,which provide the theoretical guidance for design and use of thrust bearing. The results show that the maximum temperature of the bearing pad appear in the circumferential outlet and near the outer diameter side,which decreases with the increase of the slope angle. When the other conditions are constant,the larger the thickness of the oil film is,the lower the temperature rise of bearing,and the film thickness does not affect the temperature field distribution of the bearing. The shear stress and bearing temperature are increased with the increase of speed. There is no longer a linear relationship between the axial bearing capacity and speed when considering the coupling heat transfer.

The different structural helicoidal surface thrust bearing lubrication models is built and calculated with FLUENT to analyze the effects of the thickness of the oil film,pitch of the helicoidal surface and angular speed on load capacity of the thrust bearing,which provide the theoretical basis for design of helicoidal surface thrust bearing. The results show that with the increase of the pitch and oil film thickness,the maximum temperature of the oil film is decreased. The axial load capacity decreases with the increase of the oil film thickness.There is an ratio of pad height to the minimum film thickness which maximizes the load capacity when the minimum thickness of oil film and angular speed are constant,the best ratio of pad height to the minimum film thickness is 1. 4. The maximum temperature and load capacity of the oil film are linearly related to the angular speed. The load capacity of the helicoidal surface is much higher than the flat.

Abstract Background Activation of nuclear factor erythroid 2-related factor (Nrf2), which belongs to the basic leucine zipper transcription factor family, is a strategy for cancer chemopreventive phytochemicals. It is an important regulator of genes induced by oxidative stress, such as glutathione S-transferases, heme oxygenase-1 and peroxiredoxin 1, by activating the antioxidant response element (ARE). We hypothesized that (1) the citrus coumarin auraptene may suppress premalignant mammary lesions via activation of Nrf2/ARE, and (2) that Nrf2 knockout (KO) mice would be more susceptible to mammary carcinogenesis. Methods Premalignant lesions and mammary carcinomas were induced by medroxyprogesterone acetate and 7,12-dimethylbenz[a]anthracene treatment. The 10-week pre-malignant study was performed in which 8 groups of 10 each female wild-type (WT) and KO mice were fed either control diet or diets containing auraptene (500 ppm). A carcinogenesis study was also conducted in KO vs. WT mice (n = 30-34). Comparisons between groups were evaluated using ANOVA and Kaplan-Meier Survival statistics, and the Mann-Whitney U-test. Results All mice treated with carcinogen exhibited premalignant lesions but there were no differences by genotype or diet. In the KO mice, there was a dramatic increase in mammary carcinoma growth rate, size, and weight. Although there was no difference in overall survival, the KO mice had significantly lower mammary tumor-free survival. Also, in the KO mammary carcinomas, the active forms of NF-κB and β-catenin were increased ~2-fold whereas no differences in oxidized proteins were observed. Many other tumors were observed, including lymphomas. Interestingly, the incidences of lung adenomas in the KO mice were significantly higher than in the WT mice. Conclusions We report, for the first time, that there was no apparent difference in the formation of premalignant lesions, but rather, the KO mice exhibited rapid, aggressive mammary carcinoma progression.

Abstract Background Thymoquinone (TQ) is a compound extracted from Black Caraway seeds of Nigella Sativa and is active against various cancers. Cisplatin (CDDP) is the most active chemotherapeutic agent in Lung Cancer. Here we report activity of TQ against non-small cell lung cancer (NSCLC) and small cell lung cancer (SCLC) cell lines alone and in combination with Cisplatin (CDDP). Methods For proliferation MTT assay, cell viability trypan blue assay and for apoptosis Annexin-V FITC assay were used in NCI-H460 and NCI-H146 cell lines. Inhibition of invasion by TQ was assessed using Matrigel assay and its affect on release of various cytokines was determined using RayBio Human Cytokine detection kit. Mouse xenograft model using NCI-H460 was used to determine in vivo activity of TQ and CDDP. Inhibition of LPS induced NF-κB expression by TQ was determined using transgenic mice expressing a luciferase reporter. Results TQ was able to inhibit cell proliferation, reduce cell viability and induce apoptosis. TQ at 100 μM and CDDP at 5 μM inhibited cell proliferation by nearly 90% and the combination showed synergism. TQ was able to induced apoptosis in both NCI-H460 and NCI-H146 cell lines. TQ also appears to affect the extracellular environment inhibiting invasion and reducing the production of two cytokines ENA-78 and Gro-alpha which are involved in neo-angiogenesis. Using a mouse xenograft model we were able to demonstrate that combination of TQ and CDDP was well tolerated and significantly reduced tumor volume and tumor weight without additional toxicity to the mice. In the combination arms (TQ5 mg/kg/Cis 2.5 mg/kg) tumor volume was reduced by 59% and (TQ20 mg/kg/Cis 2.5 mg/kg) by 79% as compared to control which is consistent with in vitro data. TQ down regulated NF-κB expression which may explain its various cellular activities and this activity may prove useful in overcoming CDDP resistance from over expression of NF-κB. Conclusions Thus TQ and CDDP appear to be an active therapeutic combination in lung cancer.
