Abstract

In recent years, the rate of implant failure has been increasing. Microbial infection was the primary cause, and the main stages included bacterial adhesion, biofilm formation, and severe inhibition of implant osseointegration. Various biomaterials and their preparation methods have emerged to produce specific implants with antimicrobial or bactericidal properties to reduce implant infection caused by bacterial adhesion and effectively promote bone and implant integration. In this study, we reviewed the research progress of bone integration promotion and antibacterial action of superhydrophilic surfaces based on titanium alloys. First, the adverse reactions caused by bacterial adhesion to the implant surface, including infection and bone integration deficiency, are briefly introduced. Several commonly used antibacterial methods of titanium alloys are introduced. Secondly, we discuss the antibacterial properties of superhydrophilic surfaces based on ultraviolet photo-functionalization and plasma treatment, in contrast to the antibacterial principle of superhydrophobic surface morphology. Thirdly, the osteogenic effects of superhydrophilic surfaces are described, according to the processes of osseointegration: osteogenic immunity, angiogenesis, and osteogenic related cells. Finally, we discuss the challenges and prospects for the development of this superhydrophilic surface in clinical applications, as well as the prominent strategies and directions for future research.

36. Association Between Serum 25-hydroxy-vitamin D Level, Liver Function and Liver Fibrosis in Biliary Atresia Infants One Year After Kasai Procedure

Abstract

Objective: To investigate the relationship between serum 25 (OH) D level, liver function, and liver fibrosis in infants with in biliary atresia (BA) infants one year after Kasai procedure. Methods: This retrospective study was conducted using the clinical data of children with BA confirmed by cholangiography and/or surgery admitted to Children Hospital of Shanxi Province between October 2018 and October 2020, as well as the follow-up data at 1, 3, 6, and 12 months after Kasai Portoenterotomy so as to analyze the relationship between serum 25(OH) D level, liver function, and liver fibrosis. Results: Preoperative data of 57 infants with BA were collected. All infants had vitamin D insufficiency or deficiency status before surgery. Three months after surgery, data were collected from 55 infants, 15 of whom did not take vitamin D supplements and 40 of whom received regular vitamin D supplementation. It was found that 15.38%, 47.22%, 75%, and 92.31% of infants living with regular vitamin D supplementation and native liver had sufficient vitamin D levels at 3, 6, and 12 months after Kasai surgery, respectively. The serum 25(OH) D level was significantly lower in patients without vitamin D supplements at 3 months after surgery (t=-2.974，P＜0.05), and the aspartate aminotransferase to platelet ratio index (APRI) was higher in patients with vitamin D supplementation. There was no significant correlation between vitamin D level, liver function, and liver fibrosis preoperatively (P＞0.05). The level of serum 25(OH) D was negatively correlated with liver fibrosis and liver function in infants with the native liver at 12 months after surgery (P < 0.05).Conclusion: Vitamin D deficiency was commonly found in children with BA before operation. Nearly 70% of infants living with native liver reached sufficient vitamin D levels within 6 months after surgery with routine vitamin D supplementation. Serum 25 (OH)D level can reflect the degree of liver fibrosis and liver function in biliary atresia infants living with the native liver at 12 months after the Kasai procedure.

Abstract

Osteoclastogenesis requires activation of RANK signaling as well as costimulatory signals from immunoreceptor tyrosine-based activation motif-containing (ITAM-containing) receptors/adaptors, predominantly tyrosine kinase-binding proteins DAP12 and FcRγ, in osteoclast precursors. It is not well understood how costimulatory signals are regulated and integrated with RANK signaling. Here, we found that osteopetrotic bone phenotypes in mice lacking DAP12 or DAP12 and FcRγ are mediated by the transcription factor RBP-J, as deletion of Rbpj in these mice substantially rescued the defects of bone remodeling. Using a TNF-α-induced model of inflammatory bone resorption, we determined that RBP-J deficiency enables TNF-α to induce osteoclast formation and bone resorption in DAP12--deficient animals. Thus, RBP-J imposes a requirement for ITAM-mediated costimulation of RANKL or TNF-α-induced osteoclastogenesis. Mechanistically, RBP-J suppressed induction of key osteoclastogenic factors NFATc1, BLIMP1, and c-FOS by inhibiting ITAM-mediated expression and function of PLCγ2 and activation of downstream calcium-CaMKK/PYK2 signaling. Moreover, RBP-J suppressed Plcg2 expression and downstream calcium oscillations indirectly by a TGF-β/PLCγ2/calcium axis. Together, our findings indicate that RBP-J suppresses ITAM-mediated costimulation, thereby limiting crosstalk between ITAM and RANK/TNFR signaling and allowing fine tuning of osteoclastogenesis during bone homeostasis and under inflammatory conditions. Abstract

Bone metabolism results from a balance between osteoclast-driven bone resorption and osteoblast-mediated bone formation. Diseases such as periodontitis and rheumatoid arthritis are characterized by increased bone destruction due to enhanced osteoclastogenesis (1,2). Here we report that interferon regulatory factor-8 (IRF-8), a transcription factor expressed in immune cells, is a key regulatory molecule for osteoclastogenesis. IRF-8 expression in osteoclast precursors was downregulated during the initial phase of osteoclast differentiation induced by receptor activator of nuclear factor-[kappa]]B ligand (RANKL), which is encoded by the Tnfsf11 gene. Mice deficient in Irf8 showed severe osteoporosis, owing to increased numbers of osteoclasts, and also showed enhanced bone destruction after lipopolysaccharide (LPS) administration. [Irf8.sup.-/-] osteoclast precursors underwent increased osteoclastogenesis in response to RANKL and tumor necrosis factor-[alpha] (TNF-[alpha]). IRF-8 suppressed osteoclastogenesis by inhibiting the function and expression of nuclear factor of activated T cells c1 (NFATc1). Our results show that IRF-8 inhibits osteoclast formation under physiological and pathological conditions and suggest a model where downregulation of inhibitory factors such as IRF-8 contributes to RANKL-mediated osteoclastogenesis., Osteoclasts are multinucleated giant cells derived from the monocyte and macrophage lineage.
