12. Terpenoids from the Coral-Derived Fungus Trichoderma harzianum (XS-20090075) Induced by Chemical Epigenetic Manipulation

Abstract

The soft coral-derived fungus Trichoderma harzianum (XS-20090075) was found to be a potential strain to produce substantial new compounds in our previous study. In order to explore its potential to produce more metabolites, chemical epigenetic manipulation was used on this fungus to wake its sleeping genes, leading to the significant changes of its secondary metabolites by using a histone deacetylase (HDAC) inhibitor. The most obvious difference was the original main products harziane diterpenoids were changed into cyclonerane sesquiterpenoids. Three new terpenoids were isolated from the fungal culture treated with 10 mM sodium butyrate, including cleistanthane diterpenoid, harzianolic acid A (1), harziane diterpenoid, harzianone E (2), and cyclonerane sesquiterpenoid, 3,7,11-trihydroxy-cycloneran (3), together with 11 known sesquiterpenoids (4-14). The absolute configurations of 1-3 were determined by single-crystal X-ray diffraction, ECD and OR calculations, and biogenetic considerations. This was the first time to obtain cleistanthane diterpenoid and africane sesquiterpenoid from genus Trichoderma, and this was the first chlorinated cleistanthane diterpenoid. These results demonstrated that the chemical epigenetic manipulation should be an efficient technique for the discovery of new secondary metabolites from marine-derived fungi.

Abstract

NaZn13-type La(Fe0.94Co0.06)11.8Si1.2 alloys were manufactured to investigate the influence of their microstructural change on magnetic refrigeration performance during magnetic field cycling. The magnetic refrigeration performance measurements indicate that both the large magnetic entropy change value (ΔS=14.1 J kg-1 K-1) and maximum adiabatic temperature change (Δ=2.2K) are favorable for the alloys to be superior candidate of magnetic refrigerants. However, the alloys exhibit nearly 10% decrease of ΔS and ΔT when they performed cycling ten-thousand times. More than thousand times of cycles induce local stress and grain cleavages presented by the accumulation of irreversible microstructure changes such as micro-cracks and sub-boundaries. According to the domain observation for the alloys with different field cycles, these microstructure characteristics accompany with the reconfiguration of the local magnetic domains and increase of domain wall energy, which are considered to be the reason of the decrease of ΔS. Abstract

Field-induced unidirectional anisotropy of Co-based amorphous ribbons was discussed by magnetization measurement. The shifted hysteresis loops of Co58Fe5Ni10Si11B16 amorphous ribbons were obtained by annealing the samples in longitudinal magnetic field. Here, the feasibility of employing the longitudinal pulse field to tailor the anisotropy characteristic is demonstrated. It is found that the shifted loops can be technically controlled by enhancing the pulse field to modulate the magnetic anisotropy from unidirectional to uniaxial, and even back to unidirectional. The surface domains scan gives strong evidence that the pulse field can be one of the skillful methods to navigate the unidirectional anisotropy in the amorphous ribbons for potential applications. 29. Effect of inhibition to Yes-related proteinsmediated Wnt/β-catenin signaling pathway through miR-195-5p on apoptosis of gastric cancer cells.

Abstract

OBJECTIVE: This clinical study aimed to explore the expression of miR-195-5p and Yes-associated protein (YAP) in gastric cancer (GC) and their relevant apoptosis mechanism. PATIENTS AND METHODS: Thirty GC patients treated in our hospital were enrolled as a patient group and 30 normal people who underwent physical examination during the same period were enrolled as a normal group. The purchased GC cells and human gastric mucous membrane cells were used to transfect miR-195-5p-mimics, miR-NC, si-YAP, and si-RNA into MKN45 and SGC7901 cells. qRT-PCR was used to detect the expression of miR-195-5p and YAP in samples, and WB was used to detect the expression of YAP, Caspase-3, Caspase-9, β-catenin, c-myc, and cyclin Dl. CCK-8, the transwell, and the flow cytometry were used to detect cell proliferation, invasion, and apoptosis. The dual fluorescent enzyme reporter was used to determine the relationship between miR-195-5p and YAP. RESULTS: MiR-195-5p was lowly expressed in the tissues and serum of patients, but YAP was highly expressed, and the area under the miR-195-5p and YAP curves was more than 0.9. MiR-195-5p and YAP were associated with tumor diameter, TNM stage, lymph node metastasis, and differentiation in GC patients. The overexpression of miR-195-5p and the inhibition of YAP expression can inhibit cell proliferation and invasion and promote apoptosis. WB assay showed that the overexpression of miR-195-5p and the inhibition of YAP could inhibit the expression of β-catenin, c-myc, and cyclin D1 protein and promote the expression of Caspase-3, Caspase-9 protein. The dual fluorescent enzyme reporter revealed that there was a targeting relationship between miR-195-5p and YAP. CONCLUSIONS: The overexpression of miR-195-5p can inhibit YAP-mediated Wnt/β-catenin signaling pathway and promote cell apoptosis, so it may be a potential therapeutic target for GC. 30. Verification of expressions of lncRNA FOXCUT in gastric adenocarcinoma patients and its effects on cell biological function based on TCGA database.

Abstract

OBJECTIVE: This study aimed to investigate expressions of lncRNA FOXCUT in gastric adenocarcinoma patients and its effects on the cell biological function. PATIENTS AND METHODS: Expressions and survival of lncRNA FOXCUT in gastric adenocarcinoma patients (GA) in the Cancer Genome Atlas (TCGA) database were collected. Fifty patients with GA treated in our hospital (patient group) and another 50 contemporaneous normal people (normal group) were collected. Expressions of lncRNA FOXCUT in GES1, SNU-5, HGC-27, SGC-7901, and AGS cells were detected. Also, si-lncRNA FOXCUT and si-NC sequences were transfected to SGC-7901. Si-RNA and si-NC groups were constructed in AGS cells. QRT-PCR was used to detect expressions of lncRNA FOXCUT in samples. MTT, transwell, and flow cytometry were used to detect the proliferation, invasion, and apoptosis of transfected cells. Patients were followed up for 5 years to observe their survival. RESULTS: Expressions of lncRNA FOXCUT in cancer tissues of GA patients in TCGA database were significantly increased (p<0.001). The survival rate of patients with low expressions of lncRNA FOXCUT was significantly increased (p=0.017, p=0.047). LncRNA FOXCUT is closely related to patients' tumor diameter, lymph node metastasis, TNM staging, and differentiation degree (p<0.05). LncRNA FOXCUT has high clinical value in disease diagnosis. Multivariate Cox regression analysis found that tumor diameter, lymph node metastasis, and lncRNA FOXCUT were independent prognostic factors. Compared with GES1, expressions of lncRNA FOXCUT in GA cells increased significantly (p<0.05), the proliferation and invasion ability of si-RNA group decreased significantly (p<0.05) compared with si-NC group, and the apoptosis rate of si-RNA group was significantly lower than that of si-NC group (p<0.05). CONCLUSIONS: We showed that the inhibition of the expressions of lncRNA FOXCUT can reduce the proliferation and invasion of GA cells and increase apoptosis, which can be used as a potential therapeutic target for GA. Exchange bias phenomena of nanocrystalline FeCuNbSiB ribbons have been investigated at room temperature. It was found that the rougher surface of ribbons annealed in longitudinal field played a main role in bringing the bias behavior in our ribbons. The topography of ribbon surfaces and the configuration of magnetic domains were specially observed in order to make clear the dependence of bias behavior on the induced magnetic anisotropy. Abstract

Objectives: We aimed to evaluate the relationship between baseline renal function and changes in telomere length in Han Chinese.Methods: The telomere restriction fragment (TRF) length of leukocytes in the peripheral blood was measured in healthy volunteers recruited in 2014. The estimated glomerular filtration rate (eGFR) was calculated based on serum creatinine (Scr) and serum cystatin C (CysC)-eGFRcys and eGFRScr-cys through the Cockcroft-Gault formula (eGFRC-G) or the Chronic Kidney Disease Epidemiology Collaboration (CKD-EPI / eGFRCKD-EPI) equation. The correlation between telomere length changes over time and renal function was analyzed.Results: Leukocyte TRF lengths were negatively correlated to age (r = -0.393, p < 0.001) and serum CysC (r = -0.180, p < 0.01), while positively associated with eGFRCKD-EPI, eGFRC-G, eGFRcys, and eGFRScr-cys (r = 0.182, 0.122, 0.290, and 0.254 respectively, p < 0.01). The 3-year change of telomere length was 46 bp/years. When adjusted for age, the associations between telomere length changes and baseline, subsequent TRF lengths, and serum CysC were no longer present. No association was observed between TRF length changes and renal function.Conclusion: The rate of telomere length changes was affected by age and baseline telomere length. Abstract

Si3N4 ceramics with different sintering aids are prepared by gas pressure sintering at 1800°C under 8 MPa N2 atmosphere. The mechanical and microwave dielectric properties of the Si3N4 ceramics with and without sintering aids are investigated. The results show that the sintering aids have a significant impact on the densification and mechanical properties of Si3N4 ceramics. Of all, the Si3N4 ceramics using the dry gel of yttrium aluminium garnet as sintering aids exhibit the maximum bending strength and fracture toughness, reaching 395 MPa and 4·25 MPa m1/2 respectively. However, the dielectric properties of Si3N4 ceramics are similar to that using the other two sintering aids. The improved mechanical properties of the Si3N4 ceramics are attributed to the formation of rod-like βSi3N4-particles. [ABSTRACT FROM AUTHOR]

Abstract

Callusing was induced from three kinds of explants of Chinese medical plantScutellaria baicalensiswith thidiazuron (TDZ) as an exogenous growth regulator. Multiple shoots were formed from these calli on hormone-free media. Calli induced by 0.3 mg/l TDZ produced shoots directly on the induction medium after culturing for 30-40 days without any interval, while at a higher concentrations of TDZ (1.0, 2.0, and 5.0 mg/l), regeneration occurred only after transfer to hormone-free medium. Endogenous hormones were detected by reverse-phase HPLC-MS and ELISA. The higher TDZ concentrations decreased endogenous benzyladenine (BA) and IAA levels, while iPA and ZR levels and their ratios to IAA were not correlated with TDZ concentrations, growth of calli, or plant regeneration. BA was the dominant cytokinin during calli growth and bud formation, 10 000-fold more abundant than iPA and ZR. Moreover, low IAA/BA ratio, rather than low IAA or high cytokinin levels alone, is the most important factor for shoot formation by calli. TDZ might act indirectly by changing endogenous hormonal levels, most likely the IAA/BA ratio which controls the calli growth and bud formation. [ABSTRACT FROM AUTHOR]

47. Developing Aerobic Rice Cultivars for Water‐Short Irrigated and Drought‐Prone Rainfed Areas in the Tropics

Abstract

Aerobic rice is a production system in which rice is direct seeded and grown in nonpuddled and nonflooded aerobic soils. Aerobic rice varieties need to be developed to cope with the increasing water scarcity in tropical Asia. Advanced breeding lines were evaluated in 24 yield trials under both nonstressed and stressed aerobic soil conditions in 4 yr (2005–2008) to assess the efficacy of the two‐stress‐level screening protocol and selection strategies for developing aerobic rice and to identify genotypes that are high yielding and drought tolerant. Twenty‐six genotypes outyielded the reference varieties Apo and UPLRi‐7 by more than 10% under both nonstress and stress conditions. High harvest index (HI) and vegetative vigor accounted for the increased yield of the newly developed genotypes. On average, the drought stress reduced grain yield by 60%, HI by 50%, and plant height by 12 cm and delayed flowering by 3 d. There was a low genetic correlation for yield across the two stress levels, and indirect selection under one stress level was not predicted to be effective in improving grain yield under the other, suggesting that screening genotypes under both nonstress and stress conditions for yield is needed to develop aerobic‐adapted varieties combining high yield potential with drought tolerance. The elite germplasm identified in this study is likely to be useful to farmers in water‐short irrigated and rainfed areas in the tropics.
