Gut microbes especially the beneficial ones play an important role in maintaining human health. At present, the research methods for intestinal microbes are mainly based on non-culture technologies such as metagenomics. The relationship between intestinal microbes and the body’s health can be found by using metagenomic sequencing technology. Researchers have found that most of the bacteria in the gut are uncultured, and their molecular mechanisms of action are unclear. Culturomics can be used to successfully isolate and culture some intestinal bacteria difficult to culture by improving the composition of culture medium and optimizing the culture conditions. The application of culturomics provides technical support for research on the functions of intestinal bacteria in the host and further screening of beneficial intestinal bacteria. Therefore, using culturomics technology to cultivate more beneficial intestinal bacteria and studying their phenotypes and gene functions are future research priorities. This article reviews the culturomics of and the cultivation methods for intestinal beneficial bacteria for the purpose of providing a reference for the cultivation of beneficial intestinal microorganisms for human health.

The aim of this study was to investigate the effect of Bifidobacterium lactis Probio-M8 on glucolipid metabolism and gut microbiota (GM) composition in type 2 diabetes mellitus (T2DM) mice. The glucolipid metabolic profiles were analyzed. The 16S rRNA gene sequencing was employed to investigate GM. The levels of farnesyl X receptor (FXR) and cytochrome p450 7A1 (CYP7A1) were detected by quantitative polymerase chain reaction and western blot assays. The total bile acids (TBAs), ceramide (CE), glucagon-like peptide-1 (GLP-1), and fibroblast growth factor (FGF)-15 were also detected. The morphological features of liver and pancreas were also analyzed. Compared with the model group, Probio-M8 restored body weight, food intake and water intake, as well as improved hyperglycemia symptoms, serum glucolipid parameters, and the composition of intestinal microbes in T2DM diabetic mice. Moreover, the reduced level of FXR and the increased level of CYP7A1 in T2DM mice were reversed by Probio-M8 treatment. The increased levels of TBA and CE and the reduced levels of GLP-1 and FGF-15 in T2DM mice were altered after Probio-M8 stimulation. Besides, the altered morphology of liver and ileum in T2DM mice was alleviated by Probio-M8 treatment. Taken together, we suggested that the symptoms of T2DM could be ameliorated by Probio-M8 in T2DM mice.

In desert areas, the process of mobile sandy land changing to semi‐fixed sandy land and eventually to fixed sandy land after undergoing vegetation restoration is inevitable. The presence of shrub patches and herb patches is common in this restoration process. No relevant studies have reported the soil bacterial community characteristics of different vegetation‐type patches (shrub patches and herb patches) under different stages of restoration. Therefore, we utilized long‐established experimental plots to collect soil from 0–20 cm soil layer under shrub patches (dominated by Salix psammophila) and herb patches under different stages of restoration (i.e., mobile sand land, semi‐fixed sand land, and fixed sand land), by determining soil physicochemical properties, enzyme activities, and soil bacterial communities. Our results found that soil bacterial α‐diversity under different restoration stages showed higher shrub patches than herb patches. The dominant bacterial communities (phyla) in shrub patches and herb patches at different recovery stages were Actinobacteria, Proteobacteria, and Bacteroidota. When the mobile sandy land returned to fixed sandy land, the relative abundance of Actinobacteria and Bacteroidota gradually decreased under shrub patches and herb patches, while the relative abundance of Proteobacteria increased significantly. In addition, herb patches significantly increased the relative abundance of bacteria (genus) relative to shrub patches at different stages of recovery. Soil nutrients, soil fine particles, and soil enzyme activities were significantly higher under shrub patches than under herb patches when fixed sandy land due to differences in life form and architecture between shrub patches and herb patches. Based on this, soil bacterial community composition and diversity under shrub patches were driven by more soil properties during the restoration of sandy land. This study complements the dynamic recovery processes and driving mechanisms of soil bacterial community structure under different vegetation patches in sandy areas, especially in the context of global climate change. Researchers have applied a series of global models to investigate the link between the built environment and ride-hailing ridership based on ride-hailing data from one specific transportation network company (TNC). However, these research designs inadequately represent real ride-hailing demand within a specific spatial range and cannot reflect spatiotemporal heterogeneity in the link. For the first time, this study collects all demand data of TNCs in Nanjing and analyzes their relationship with the built environment. The effect of taxi demand is considered. We adopt a multiscale geographically weighted regression model to account for the spatial non-stationarity and the multiscale effect of each built environment variable. The findings reveal spatiotemporal heterogeneous relationships of the built environment with ride-hailing ridership. Although the relationship between taxi and ride-hailing ridership varies across spatial locations, ride-hailing always acts as a cooperator for traditional taxis. The findings provide implications for policy making, urban planning, and travel demand management of ride-hailing. [ABSTRACT FROM AUTHOR]
