Background: Giardia duodenalis is a parasitic organism that can cause giardiasis, an intestinal infection, particularly prevalent in young children, with clinical symptoms of diarrhea. We previously reported that extracellular G. duodenalis triggers intracellular nucleotide-binding oligomerization-like receptor 3 (NLRP3) inflammasome activation and regulates the host inflammatory response by secreting extracellular vesicles (EVs). However, the exact pathogen-associated molecular patterns in G. duodenalis EVs (GEVs) involved in this process and the role of the NLRP3 inflammasome in giardiasis remain to be elucidated. Methods: Recombinant eukaryotic expression plasmids of pcDNA3.1(+)-alpha-2 and alpha-7.3 giardins in GEVs were constructed, transfected into primary mouse peritoneal macrophages and screened by measuring the expression levels of the inflammasome target molecule caspase-1 p20. The preliminary identification of G. duodenalis alpha-2 and alpha-7.3 giardins was further verified by measuring the protein expression levels of key molecules of the NLRP3 inflammasome (NLRP3, pro-interleukin-1 beta [IL-1β], pro-caspase-1, and caspase-1 p20), the secretion levels of IL-1β, the level of apoptosis speck-like protein (ASC) oligomerization and the immunofluorescence localization of NLRP3 and ASC. The roles of the NLRP3 inflammasome in G. duodenalis pathogenicity were then evaluated using mice in which NLRP3 activation was blocked (NLRP3-blocked mice), and body weight, parasite burden in the duodenum and histopathological changes in the duodenum were monitored. In addition, we explored whether alpha-2 and alpha-7.3 giardins triggered IL-1β secretion in vivo through the NLRP3 inflammasome and determined the roles of these molecules in G. duodenalis pathogenicity in mice. Results: Alpha-2 and alpha-7.3 giardins triggered NLRP3 inflammasome activation in vitro. This led to caspase-1 p20 activation, upregulation of the protein expression levels of NLRP3, pro-IL-1β and pro-caspase-1, significant enhancement of IL-1β secretion, ASC speck formation in the cytoplasm and also induction of ASC oligomerization. Deletion of the NLRP3 inflammasome aggravated G. duodenalis pathogenicity in mice. Compared to wild-type mice gavaged with cysts, mice gavaged with cysts in NLRP3-blocked mice displayed increased trophozoite loads and severe duodenal villus damage, characterized by necrotic crypts with atrophy and branching. In vivo assays revealed that alpha-2 and alpha-7.3 giardins could induce IL-1β secretion through the NLRP3 inflammasome and that immunization with alpha-2 and alpha-7.3 giardins decreased G. duodenalis pathogenicity in mice. Conclusions: Overall, the results of the present study revealed that alpha-2 and alpha-7.3 giardins trigger host NLRP3 inflammasome activation and decrease G. duodenalis infection ability in mice, which are promising targets for the prevention of giardiasis. Currently, Mikania micrantha (M. micrantha) has invaded Guangdong, Guangxi and other provinces in China, causing serious harm to the forests of southeastern China. Soil microorganisms play an important role in the establishment of M. micrantha invasion, affecting plant productivity, community dynamics, and ecosystem function. However, at present, how M. micrantha invasion affects soil carbon, nitrogen, and phosphorus phase functional genes and the environmental factors that cause gene expression changes remain unclear, especially in subtropical forest ecosystems. This study was conducted in Xiangtoushan National Forest Park in Guangdong Province to compare the changes in soil nutrients and microorganisms after M. micrantha invasion of a forest. The microbial community composition and metabolic function were explored by metagenome sequencing. Our results showed that after M. micrantha invasion, the soil was more suitable for the growth of gram-positive bacteria (Gemmatimonadetes). In addition, the soil microbial community structure and enzyme activity increased significantly after M. micrantha invasion. Correlation analysis and Mantel test results suggested that total phosphorus (TP), nitrate nitrogen (NO3–-N), and soil dissolved organic matter (DOM; DOC and DON), were the strong correlates of soil microbial nitrogen functional genes, while soil organic matter (SOM), soil organic carbon (SOC), total nitrogen (TN), and available phosphorus (Soil-AP) were strongly correlated with the expression of soil microbial phosphorus functional gene. Mikania micrantha invasion alters soil nutrients, microbial community composition and metabolic function in subtropical forests, creates a more favorable growth environment, and may form a positive feedback process conducive to M. micrantha invasion. Climate change is a significant force influencing catchment hydrological processes, such as baseflow, i.e., the contribution of delayed pathways to streamflow in drought periods and is associated with catchment drought propagation. The Weihe River Basin is a typical arid and semi-arid catchment on the Loess Plateau in northwest China. Baseflow plays a fundamental role in the provision of water and environmental functions at the catchment scale. However, the baseflow variability in the projected climate change is not well understood. In this study, forcing meteorological data were derived from two climate scenarios (RCP4.5 and RCP8.5) of three representative general circulation models (CSIRO-Mk3-6-0, MIROC5, and FGOALSg2) in CMIP5 and then were used as inputs in the Soil and Water Assessment Tool (SWAT) hydrological model to simulate future streamflow. Finally, a well-revised baseflow separation method was implemented to estimate the baseflow to investigate long-term (historical (1960–2012) and future (2010–2054) periods) baseflow variability patterns. We found (1) that baseflow showed a decreasing trend in some simulations of future climatic conditions but not in all scenarios (p < 0.05), (2) that the contribution of baseflow to streamflow (i.e., baseflow index) amounted to approximately 45%, with a slightly increasing trend (p ≤ 0.001), and (3) an increased frequency of severe hydrological drought events in the future (2041–2053) due to baseflows much lower than current annual averages. As an attractive electrochemiluminescence (ECL) emitter, graphitic carbon nitride (g-CN) still suffers from weak anodic ECL signals. In this work, a simple molecular engineering strategy is employed to modulate the anodic ECL of g-CN by thiophene doping. Thiophene doped g-CN (CNA) nanosheets show ca. a 6-fold enhanced and red-shift ECL emissions compared to pristine g-CN nanosheets. The optical and ECL property studies reveal that thiophene doping can not only evaluate g-CN electric conductivity by intramolecular electron transfer but also narrow the bandgap, leading to the enhancement and bathochromic shift of ECL emissions. The effects of various factors including the thiophene doping amount, solution pH, coreactant concentration and adopted potential range on the anodic ECL of CNA nanosheets are further investigated for understanding the mechanism of ECL enhancement and obtaining a strong and stable anodic ECL emission. On this basis, the CNA nanosheets modified electrode is used as a signal probe for Cu2+ detection, and the probe shows a linear response to Cu2+ in the concentration range of 5 nM to 50 μM and high selectivity towards other possible interfering metal ions. Our work may provide a promising pathway for the modulation of g-CN ECL by molecular engineering, and the improved anodic ECL may promote its sensing application. [ABSTRACT FROM AUTHOR]

In contrast to unary assemblies comprising π‐conjugated organic species, elaborate modulation of dimensions, polymorphisms, and compositions of multicomponent assembled architectures with two or more constituent materials has not yet been systematically studied. Herein, a combinatorial library of organic microcrystals made of four components via a solution‐phase assembly route is reported. With the involvement of growth kinetics, four organic species with slight structural modifications can assemble into five unary assemblies, which may endow binary combinations with highly and partially structural miscibility. Consequently, a variety of binary alloyed assemblies and microscale heterostructures with tailorable dimensions, polymorphisms, and emission colors are realized by rational compositional and growth control. The effects of structural relations of binary combinations on their miscibility are systematically uncovered, that gives rise to these intricate microscale architectures as well as diverse energy transfer (ET) efficiencies. Highly efficient ET process in binary alloyed assemblies can be beneficial to steady‐state photoluminescence anisotropy amplification. Benefiting from the information of binary combinations, white‐light‐emitting ternary microsheets can be modulated in a predictable manner. The present work uncovers the rational control of multicomponent microcrystals, which further stretches the boundaries of molecular self‐assembly and may be used to achieve integrated optoelectronic properties, such as multicolor lasers and p–n junctions. Background: Although selective serotonin reuptake inhibitors (SSRIs) are usually considered safe to use in patients with Parkinson’s disease (PD), there are mixed data about their effectiveness, and only a few investigations have led to a total improvement of depressive symptoms in patients with PD.We aimed to conduct a comprehensive systematic review and meta-analysis of all studies that investigated the effectiveness of SSRIs in treating depression in the context of PD.From its commencement to June 2024, the databases of MEDLINE via PubMed, Scopus, Embase, and Google Scholar were electronically searched for the relevant papers. All full-text journal articles assessing the effectiveness of SSRIs in treating depression in patients with PD were included. The tool developed by the Cochrane Collaboration was utilized to evaluate the bias risk. Data were analyzed utilizing a pair-wise comparison meta-analysis using the standardized mean difference.A total of 19 articles and 22 separate interventions were included. We found that SSRI treatment attenuated depression in patients with PD (1.242 standardized mean difference, 95% confidence interval 0.956, 1.529, p < 0.001). The general heterogeneity of the studies was medium (ϰ2 = 72.818, T2 = 0.317, df = 21, I2 = 71.15%, p < 0.001). The funnel plot was reasonably symmetrical. However, three studies were trimmed to the left of the mean. Begg’s test (p = 0.080), Egger’s test (p = 0.121), and funnel plot showed no significant risk of publication bias. The meta-regression showed that the treatment effect increased as a function of paroxetine treatment duration (slope p = 0.001) but decreased as a function of sertraline treatment duration (slope p = 0.019).There are few controlled antidepressant trials on the PD population, even though patients with PD frequently experience depression and use antidepressants. The global temperature is continuously rising, and the impacts of nighttime warming (NW) and daytime warming (DW) on ecosystems have drawn widespread attention. However, most studies on the effects of daytime and nighttime warming on ecosystems have focused primarily on aboveground processes, but little is known about how they affect underground processes, particularly soil microorganisms. Therefore, we examined the effects of daytime and nighttime experimental warming (CK: control, DW: daytime warming, NW: nighttime warming, DW + NW: daily warming) on soil bacterial and fungal diversities at the Experimental Station of Zengcheng Forestry Science Research Institute in Guangdong, China. The results showed that, compared with the CK treatment, the NW rather than DW treatment significantly increased the abundance-based coverage estimator (ACE) diversity index of bacteria and fungi. Fungal beta diversity significantly differed between the DW and NW treatments, while there was no significant difference in bacterial beta diversity. Our research indicated that the NW treatment creates a more favourable microenvironment than the DW treatment, accelerating the mineralization of soil carbon, nitrogen, and phosphorus nutrients, which may be the reason for changes in microbial diversity. Summary: Fruit morphology is an important agronomical trait of many crops. Here, we identify Sly‐miR159 as an important regulator of fruit morphology in tomato, a model species of fleshy‐fruit development. We show that Sly‐miR159 functions through its target SlGAMYB2 to control fruit growth. Suppression of Sly‐miR159 and overexpression of SlGAMYB2 result in larger fruits with a reduced length/width ratio, while loss of function of SlGAMYB2 leads to the formation of smaller and more elongated fruits. Gibberellin (GA) is a major phytohormone that regulates fruit development in tomato. We show the Sly‐miR159‐SlGAMYB2 pathway controls fruit morphology by modulating GA biosynthesis. In particular, we demonstrate that Sly‐miR159 promotes GA biosynthesis largely through the direct repression of the GA biosynthetic gene SlGA3ox2 by SlGAMYB2. Together, our findings reveal the action of Sly‐miR159 on GA biosynthesis as a previously unidentified mechanism that controls fruit morphology in tomato. Anshen Dingzhi prescription (ADP), which was first published in the masterpiece of traditional Chinese Medicine in the Qing Dynasty, " Yi Xue Xin Wu " (1732 CE), is documented to interrupt panic-related disorders. However, the mechanism of its action is still not clear. This study aims to investigate the effects of ADP on post-traumatic stress disorder (PTSD)-like behaviors and explore the mechanism from perspective of sirtuin1 (SIRT1)-peroxisome proliferator-activated receptor gamma co-activator 1 alpha (PGC-1α)-dependent mitochondrial function. The changes of SIRT1-PGC-1α signal and mitochondrial function were evaluated in the hippocampus of mice receiving single prolonged stress (SPS). Later, the roles of this signaling pathway played in fear memory generalization and anxiety-like behavior in SPS mice was investigated using two agonists of this signaling pathway. On this basis, the effects of ADP (36.8 mg/kg) with definite therapeutic effects, on mitochondrial function were investigated and further confirmed by a SIRT1 inhibitor. Finally, the possible components of ADP targeting PGC-1α were monitored through bioinformatics. Compared with control mice, SIRT1-PGC-1α signal in the hippocampus was impaired in SPS mice, accompanied with dysfunction of mitochondria and abnormal expression of synaptic proteins. The agonists of SIRT1-PGC-1α signal, ZLN005, as well as resveratrol improved the behavioral changes of mice caused by SPS, reversed the decline of proteins in SIRT1-PGC-1α signal, mitochondrial dysfunction, and the abnormal expression of synaptic proteins. The fingerprint was established for the quality control of ADP. At a dose of 36.8 mg/kg, ADP could prevent fear memory generalization and anxiety-like behavior in SPS mice. Mechanically, ADP promoted SIRT1-PGC-1α signal and repaired mitochondrial function. Importantly, SIRT1 inhibitor, selisistat eliminated the ameliorative effects of ADP on behavioral and mitochondrial function. Through molecular docking simulation, the brain-entering components of ADP, including malkangunin, Rg5, fumarine, frutinone A, celabenzine, and inermin had high binding energy with PGC-1α. Dysfunction of SIRT1-PGC-1α-dependent mitochondrial function is attributed to SPS-triggered fear generalization and anxiety-like behavior, and ADP could improve PTSD-like behaviors likely through activating this signaling pathway. We present high‐resolution 3D images of the crustal S‐wave velocity structure beneath northeastern Tibet, via ambient noise imaging techniques. The data come from continuous recordings taken in one year by a seismic array consisting of 366 broadband stations in the study region. The results show a substantial and pervasive middle crust low‐velocity structure (<3.4 km/s) beneath the plateau region, which partially protrudes northeastward across the West Qinling fault, reflecting channel flow from inside the plateau and material extrusion in the periphery. The growth of the northeastern Tibetan Plateau in the study region is manifested in its northeastward expansion and thickening driven by crustal channel flow, accompanied with upper crust shortening. Plain Language Summary: The growth mechanisms of the Tibetan Plateau (TP) have been considered controversial for decades. Expansion of the northeastern TP and the interaction with its peripheral area have become of major interest to researchers. Background: Giardia duodenalis is an extracellular protozoan parasite that causes giardiasis in mammals. The presentation of giardiasis ranges from asymptomatic to severe diarrhea, and the World Health Organization lists it in the Neglected Diseases Initiative. Extracellular vesicles (EVs) are a key mediator of intracellular communication. Although previous studies have shown that G. intestinalis can regulate a host's innate immune response, the role of G. intestinalis EVs (GEVs) in triggering a G. intestinalis-induced innate immune response remains to be further explored. Methods: In this study, GEVs, G. intestinalis and GEVs + G. intestinalis were inoculated into macrophages, respectively. The transcription and secretion levels of proinflammatory cytokines, including interleukin (IL)-1β, IL-6 and tumor necrosis factor alpha (TNF-α), were measured using real-time quantitative PCR (qPCR) and enzyme-linked immunosorbent assays (ELISAs). The phosphorylation levels of the MAPK, AKT and NF-κB signaling pathways in GEV-stimulated mouse macrophages were examined using western blotting and immunofluorescence methods. The roles of activated pathways in the GEV-triggered inflammatory response were determined using inhibition assays, western blotting and ELISAs. Results: The results showed that pretreatment with GEVs enhanced with G. intestinalis (GEVs + G. intestinalis) induced IL-1β, IL-6 and TNF-α transcription and secretion from mouse macrophages compared to stimulation with either GEVs or G. intestinalis alone. Inoculation of mouse macrophages with GEVs upregulated the phosphorylation levels of the p38 MAPK, p44/42 MAPK (Erk1/2), AKT and NF-κB signaling pathways and led to the nuclear translocation of NF-κB p65. Blocking the activated p38, Erk and NF-κB signaling pathways significantly downregulated the secretion of proinflammatory cytokines, and blocking the activated AKT signaling pathway demonstrated reverse effects. Conclusions: The results of this study reveal that GEVs can enhance G. intestinalis-induced inflammatory response levels in mouse macrophages through activation of the p38, ERK and NF-κB signaling pathways. We present high‐resolution 3D images of the crustal S‐wave velocity structure beneath northeastern Tibet, via ambient noise imaging techniques. The data come from continuous recordings taken in one year by a seismic array consisting of 366 broadband stations in the study region. The results show a substantial and pervasive middle crust low‐velocity structure (<3.4 km/s) beneath the plateau region, which partially protrudes northeastward across the West Qinling fault, reflecting channel flow from inside the plateau and material extrusion in the periphery. The growth of the northeastern Tibetan Plateau in the study region is manifested in its northeastward expansion and thickening driven by crustal channel flow, accompanied with upper crust shortening. Giardia duodenalis, also known as G. intestinalis or G. lamblia, is the major cause of giardiasis leading to diarrheal disease with 280 million people infections annually worldwide. Extracellular vesicles (EVs) have emerged as a ubiquitous mechanism participating in cells communications. The aim of this study is to explore the roles of G. duodenalis EVs (GEVs) in host-pathogen interactions using primary mouse peritoneal macrophages as a model. Multiple methods of electron microscopy, nanoparticle tracking analysis, proteomic assays, flow cytometry, immunofluorescence, qPCR, western blot, ELISA, inhibition assays, were used to characterize GEVs, and explore its effects on the host cell innate immunity as well as the underlying mechanism using primary mouse peritoneal macrophages. Results showed that GEVs displayed typical cup-shaped structure with 150 nm in diameter. GEVs could be captured by macrophages and triggered immune response by increasing the production of inflammatory cytokines Il1β, Il6, Il10, Il12, Il17, Ifng, Tnf, Il18, Ccl20 and Cxcl2. Furthermore, activation of TLR2 and NLRP3 inflammasome signaling pathways involved in this process. In addition, CA-074 methyl ester (an inhibitor of cathepsin B) or zVAD-fmk (an inhibitor of pan-caspase) pretreatment entirely diminished these effects triggered by GEVs exposure. Cellulose nanocrystals (CNC) have attracted great attention due to their excellent physical properties, abundant functional groups, and extensive use in many fields. However, how to facilely and efficiently prepare CNC and utilize the cellulosic raw materials comprehensively are still major challenges. Herein, a comprehensive utilization strategy of cellulose is implemented by facile, high-yield, controllable CNC preparation and effective enzymatic hydrolysis of the cellulosic residue. CNC with a mixed crystal form of cellulose I and cellulose II was successfully prepared through the partial dissolution of cellulose in an aqueous tetrabutylphosphonium hydroxide (TBPH) solution under mild conditions followed by regeneration, short-term ultrasonic dispersion, resulting in a high CNC yield of up to 72.2%, high crystallinity of up to 72.1%, and tunable dimensions and crystal structure of CNC. The characterization results confirmed that the present method produced a nonderivative form of cellulose and changed its crystalline structure to varying degrees, and CNC exhibited thermal stability comparable to that of the raw materials. A partial dissolution/regeneration/ultrasonic dispersion mechanism was proposed for this CNC preparation process. Cellulose residue obtained from the CNC production process led to a high enzymatic hydrolysis rate of up to 89.1%, which was twice as high as that of the raw material. The overall process finally resulted in a comprehensive cellulose utilization rate of more than 95%. The present strategy provides a candidate approach for the comprehensive utilization of cellulose through the synchronous production of advanced high-performance materials and fine chemicals. Objectives: As a traditional Chinese medicine, lotus leaf was reported to have significant hepatoprotective effect. To explore the hepatoprotective mechanism of lotus leaf, a rapid and reliable UPLC‐MS/MS method was conducted to simultaneously determine six specific endogenous substances including 5‐oxoproline, phenylalanine, tryptophan, C18‐phytosphingosine, lysophosphatidylcholine (16 : 0) and lysophosphatidylcholine (18 : 1). Methods: With the help of HPLC–FT‐ICR‐MS, the chemical constituents of louts leaf extract were elucidated. By observing histopathological changes and determining hepatotoxicity‐related biochemical indicators, rat model of liver injury was developed and the hepatoprotective effect of lotus leaf was verified. With the developed UPLC‐MS/MS method, six endogenous metabolites related to hepatotoxicity were monitored to investigate the hepatoprotective mechanism of lotus leaf. Key findings: In the qualitative analysis, a total of twenty compounds including ten flavonoids, nine alkaloids and one proanthocyanidin were identified. Based on the results of determining six endogenous metabolites related to hepatotoxicity, it was predicted that the hepatoprotective mechanism of lotus leaf might be related to glutathione metabolism, phenylalanine metabolism, tryptophan metabolism, sphingolipid metabolism and phospholipid metabolism. Conclusions: This study could be a meaningful investigation to provide mechanistic insights into the hepatoprotective effect of lotus leaf and further lay a theoretical basis for the clinical application of lotus leaf. Vibrio parahaemolyticus is an important pathogenic bacterium in both food safety management and mariculture. Rapid and accurate detection technologies are critical for effective control of its outbreak and spreading. Conventional technologies and polymerase chain reaction (PCR)‐based approaches have limited usage because of the requirement of laboratory instruments and trained personnel. Using the isothermal recombinase polymerase amplification (RPA) technology, several detection assays have been developed with added convenience. Combining the lateral flow strip (LFS) test with RPA can further simplify the detection. In this study, an improved RPA assay using LFS for visual detection of V. parahaemolyticus was developed. Primers were designed targeting the virulence genes and screened for amplification efficiency, nonspecific amplification, and primer–dimer formation. Probes were designed for the best primer pairs, and the weakness of LFS tests, being easily affected by primer‐dependent artifacts, was overcome by sequence modifications on primers and probe. The RPA–LFS assay took 25 min at 35 to 45 °C, and showed excellent specificity. It detected as low as one colony forming unit (CFU) of V. parahaemolyticus per reaction without DNA purification, or 10 CFU/10 g spiked food samples with 2 hr of enrichment. The detection limit was better than the currently available RPA‐based detection methods. Application of the RPA–LFS assay for simulated samples or real clinical samples showed accurate and consistent detection results compared to bioassay and quantitative PCR. The RPA–LFS assay provided a rapid, accurate, and convenient V. parahaemolyticus detection method suitable for on‐site detection in resource‐limited conditions. Practical Application: This research developed a rapid and visual detection technology for Vibrio parahaemolyticus that is not dependent on complicated equipment. The detection process takes 25 min and the result is read with the naked eye. Background: Vibrio alginolyticus is an important pathogen that has to be closely monitored and controlled in the mariculture industry because of its strong pathogenicity, quick onset after infection and high mortality rate in aquatic animals. Fast, simple and specific methods are needed for on-site detection to effectively control outbreaks and prevent economic losses. The detection specificity towards the pathogenic strains has to be emphasized to facilitate pointed treatment and prevention. Polymerase chain reaction (PCR)-based molecular approaches have been developed, but their application is limited due to the requirement of complicated thermal cycling machines and trained personnel. Results: A fast, simple and highly specific detection method for V. alginolyticus pathogenic strains was established based on isothermal recombinase polymerase amplification (RPA) and lateral flow dipsticks (LFD). The method targeted the virulence gene toxR, which is reported to have good coverage for V. alginolyticus pathogenic strains. To ensure the specificity of the method, the primer-probe set of the RPA system was carefully designed to recognize regions in the toxR gene that diverge in different Vibrio species but are conserved in V. alginolyticus pathogenic strains. The primer-probe set was determined after a systematic screening of amplification performance, primer-dimer formation and false positive signals. The RPA-LFD method was confirmed to have high specificity for V. alginolyticus pathogenic strains without any cross reaction with other Vibrio species or other pathogenic bacteria and was able to detect as little as 1 colony forming unit (CFU) per reaction without DNA purification, or 170 fg of genomic DNA, or 6.25 × 103 CFU/25 g in spiked shrimp without any enrichment. The method finishes detection within 30 min at temperatures between 35 °C and 45 °C, and the visual signal on the dipstick can be directly read by the naked eye. In an application simulation, randomly spiked shrimp homogenate samples were 100% accurately detected. Conclusions: The RPA-LFD method developed in this study is fast, simple, highly specific and does not require complicated equipment. Plants generally adopt different reproductive strategies to adapt to their environments and increase their fitness. Here, we studied the effects of enclosure cultivation on the reproductive allocation of a wheatgrass species Agropyron mongolicum in the Desert steppes of Northern China. The results showed that: (a) after enclosure cultivation, the height, clump width, coverage, and clump biomass of A. mongolicum significantly increased by 78.96% (p =.040), 63.50% (p =.013), 50.89% (p =.032), and 205.38% (p =.022), respectively, whereas density did not show a significant change (p =.330). (b) Enclosure cultivation significantly affected the biomass of A. mongolicum. Compared with cultivation outside the fence, root, leaf, and spike biomass of A. mongolicum inside the fence significantly increased by 183.52% (p =.020), 334.09% (p =.011), and 381.25% (p =.005), respectively. In addition, root biomass was the highest among the components (38.53 and 13.59 g inside and outside the fence, respectively) and spike biomass was the lowest (6.16 and 1.28 g inside and outside the fence, respectively). (c) Enclosure cultivation affected elemental nutrient allocation and the caloric values of various components of A. mongolicum, and the caloric values are positively correlated with carbon, nitrogen, and phosphorus contents. Enclosure cultivation significantly reduced carbon, nitrogen, and phosphorus in the roots, as well as nitrogen and phosphorus in the spikes, but significantly increased nitrogen in the spikes by 9.78%. The caloric values of A. mongolicum inside and outside the fence in decreasing order were as follows: spikes > leaves > stems > roots. Comparison of cultivation effects between inside and outside the fence showed that the caloric values of the spikes and roots significantly increased by 0.92% and 1.60%, respectively, whereas those of the leaves significantly decreased by 0.70%. The likelihood changes of the numbers of occurrences, duration and intensity of hydrological droughts in the Weihe River Basin in China were investigated by using the bias-corrected future climate projections from three selected Global Climate Models (GCMs) with two emission scenarios (RCP 4.5, RCP 8.5), and Soil and Water Assessment Tool (SWAT) hydrological model. Results showed that: 1) The frequency of hydrological droughts, drought durations and intensities would have obvious increasing trends in the future under dry GCM condition. However, an opposite decreasing trend was shown under wet GCM condition; 2) Future precipitation changing direction and magnitudes were dominating factor for the likelihood changes of drought characteristics. Forty GCMs project a future precipitation change from −3.42% to +14.24%, which is the largest source of uncertainties. If the observed trends of precipitation and temperature during the last 50 years continue, then the Weihe River Basin would likely be in the dry condition of GCMs; 3) The impacts of temperature on hydrological droughts cannot be neglected and it has direct (evaporation and runoff generation) and larger magnitude indirect effects (precipitation patterns). In this study, aluminum hypophosphite (AP) and melamine cyanurate (MCA) were used as flame retardant in polypropylene (PP)/wood flour (WF) composite. The flammability of the PP/WF composites was examined by the limiting oxygen index (LOI), vertical burning test (UL-94) and cone calorimeter test. When the PP/WF composite was loaded with 20% AP/MCA (the mass ratio of AP and MCA was 5: 1), the UL-94 achieved V-0 rating and the LOI was increased to 29.5%. In addition, the flexural strength was increased by about 11.0%. The results of cone calorimeter test revealed that the heat release rate and heat release rate peak of the PP/WF composite with AP/MCA were significantly reduced. The thermal degradation mechanism of PP/WF composites was investigated by thermo-gravimetric analysis, Fourier transform infrared spectrometry, energy-dispersive X-ray spectroscopy and scanning electron microscopy. The results indicate that AP/MCA had the effect of gas-phase and condensed-phase flame retardancy during the combustion and degradation of PP/WF composite. In this examine, melamine pyrophosphate (MPP) and aluminum hypophosphite (AHP) were selected as flame retardants. The synergistic effect between the MPP and AHP on flame retardancy and thermal stability of the polypropylene (PP)/wood flour (WF) composite were investigated. The thermal stability and char forming ability were enhanced considerably for the PP/WF composite with 20 wt% MPP/AHP (3:1), which were indicated by the thermal gravimetric analysis (TG), along with reductions in the heat release rate (HRR) and the smoke production release rate (SPR). The outcomes of TG, scanning electron microscopy and energy-dispersive X-ray spectroscopy revealed that there were inert gases as well as more phosphates and phosphoric acid formed. Which effectively diluted flammable gases and enabled more intumescent char residue to form. In summary, the system of MPP/AHP was equipped with perfect effect of gas phase and condensed phase flame retardancy, endowing PP/WF composite with outstanding capacity of inflaming retarding. The objective of this study was to identify the key changed subtype of T helper cells (Th cells) and their cytokines in pristane-induced arthritis (PIA) in rats. The severity of arthritis was evaluated by body weight, clinical score, the perimeter of ankle and mid-paw and histological assessment of ankle joints. Cytokines of Th1, Th2 and Th17 were determined in the spleen and inguinal lymph nodes at 28 days after pristane injection by real-time qPCR. The mRNA levels of IL-22 receptors, IL-22R1 and IL-22BP, in the spleen were quantified by real-time qPCR. Additionally, IL-22 expression in synovial membrane was detected by Western blotting, and serum IL-22 concentration was determined by ELISA. Correlation between IL-22 concentration and clinical score was analyzed. By screening the cytokines of Th1, Th2 and Th17 expression profile, we found that the mRNA levels of Th17 cytokines were significantly increased in PIA rats. Particularly, a significant increase in the protein expression of IL-22 was determined in synovial membrane and serum from PIA rats, and correlated with clinical score. We conclude that IL-22 expression level was increased and correlated with disease severity, which indicated that IL-22 may play an important role in development of PIA, and was helpful to explorer the pathogenesis of rheumatoid arthritis. NiCrAlY coatings with various Cr and Al contents were prepared on Ni-based superalloy K438G from a single NiCrAlY target by adjusting the substrate bias during high vacuum arc ion plating. The oxidation behavior of the coatings follows the oxide map laws proposed by Wallwork and Hed rather than the guide line based on Cr/Al = 4. As the coatings degrade due to Al depletion, formation of Cr 2 O 3 scales beneath the external α-Al 2 O 3 scales was observed. The mechanism for the oxidation of Ni-Cr-Al alloys is discussed. The Longmen Shan (LMS) fault zone is located on the eastern margin of the Tibetan Plateau and is a representative area for studying the eastward growth of the plateau. In this study, we present a 3-D crustal S-wave velocity structure beneath the LMS fault zone and its adjacent regions with a lateral resolution of ∼ 10 km down to a depth of 25 km from data recorded at 57 stations over one year using seismic ambient noise imaging techniques. The high and steep topography of the LMS is considered when building the high-resolution image of the upper and middle crust. The imaging provides further evidence for the high-angle listric characteristics of the LMS faults south of Beichuan, and the feature disappears toward the north, indicating differences in deformation from south to north. The structural characteristics are well correlated with the aftershock distribution and the rupture of the Wenchuan earthquake, which indicate a change in the mechanism from mostly thrust in the south to mostly strike slip in the north. The velocity structure controls the downward stretch of the faults. The results support that crustal channel flow is responsible for the growth of eastern Tibet. The eastward movement of the weak crustal material was blocked by the hard crust surrounding the Minshan uplift, resulting in great differences in the velocity structures and fault properties south and north of Beichuan. Arsenic, a naturally occurring heavy metal pollutant, is one of the functioning risk factors for neurological toxicity in humans. However, little is known about the effects of arsenic on the nervous system of Gallus Gallus . To investigate whether arsenic induce neurotoxicity and influence the oxidative stress and heat shock proteins (Hsps) response in chickens, seventy-two 1-day-old male Hy-line chickens were treated with different doses of arsenic trioxide (As 2 O 3 ). The histological changes, antioxidant enzyme activity, and the expressions of Hsps were detected. Results showed slightly histology changes were obvious in the brain tissues exposure to arsenic. The activities of Glutathione peroxidase (GSH-Px) and catalase (CAT) were decreased compared to the control, whereas the malondialdehyde (MDA) content was increased gradually along with increase in diet-arsenic. The mRNA levels of Hsps and protein expressions of Hsp60 and Hsp70 were up-regulated. These results suggested that sub-chronic exposure to arsenic induced neurotoxicity in chickens. Arsenic exposure disturbed the balance of oxidants and antioxidants. Increased heat shock response tried to protect chicken brain tissues from tissues damage caused by oxidative stress. In remote sensing–based forest aboveground biomass (AGB) estimation research, data saturation in Landsat and radar data is well known, but how to reduce this problem for improving AGB estimation has not been fully examined. Different vegetation types have their own species composition and stand structure, thus they have different data saturation values in Landsat or radar data. Optical and radar data also have different characteristics in representing forest stand structures, thus effective use of their features may improve AGB estimation. This research examines the effects of Landsat Thematic Mapper (TM) and ALOS PALSAR L-band data and their integrations in forest AGB estimation of Zhejiang Province, China, and the roles of textural images from both datasets. The linear regression models of AGB were conducted by using (1) Landsat TM alone, (2) ALOS PALSAR data alone, (3) their combination as extra bands, and (4) their data fusion, based on non-stratification and stratification of vegetation types, respectively. The results show that (1) overall, Landsat TM data perform better than PALSAR data, but the latter can produce more accurate estimates for bamboo and shrub, and for forests with AGB values less than 60 Mg/ha; (2) the combination of TM and PALSAR data as extra bands can greatly improve AGB estimation performance, but their fusion using the modified high-pass filter resolution-merging technique cannot; (3) textures are indeed valuable in AGB estimation, especially for forests with complex stand structures such as mixed forests and pine forests with understories of broadleaf species; (4) stratification of vegetation types can improve AGB estimation performance; and (5) the results from the linear regression models are characterized by overestimation and underestimation for the smaller and larger AGB values, respectively, and thus, selecting non-linear models or non-parametric algorithms may be needed in future research. Raspberry-shaped microparticles with adjustable protrusions and functional groups were synthesized and applied as carriers for the immobilization of lipase. The structural advantages of the raspberry-shaped microcarriers with immobilized lipase (RM-IL) are not only that the protrusions are constructed to play the role of a spacer arm, but the block structure enhances the bioactivity of lipase at the oil-water interface. The activation method used for the carrier has a significant effect on the performance of immobilized lipase. RM-IL prepared by carboxylic acid activation has a 3.5-fold increase of activity compared with that from amine activation and generates a fourfold increase of protein bound to 164 mg g−1 of carrier. Finally, RM-IL is used as an environmentally friendly and high-efficiency biocatalyst to catalyze the esterification of oleic acid for biodiesel under mild conditions, and the conversion can reach up to 82.1 %. Water shortage problems have received wide attention since the past decades as the global environment and climate continue to change. To tackle with this escalating challenge, generations of study has been conducted in the research for water utilization efficiency. Reclaimed water has been widely used in many countries for industry, agriculture and landscape. Despite the economic value and environmental benefits the reclaimed water brings,. contents such as heavy metal ions, organic pollutants and pathogenic microorganisms may enter the soil-plant-atmosphere system, thus posing potential environmental health risks. Therefore, many laws, guidelines and standards have been published to maintain the security of irrigation with reclaimed water. This paper compares the standards of irrigation with reclaimed water among different organizations and countries by analyzing the similarities and differences from different types of classification and water quality parameters requirements, in which their advantages and limitations are demonstrated. Polymer microcavities with adjustable openings and surface roughness are fabricated on a large scale via single-hole poly(glycidyl methacrylate) (PGMA) swelling seed particles. The size of openings of these microcavities can be adjusted by changing the amount of hydrophilic monomer, and the degree of surface roughness is easily regulated relying on the adjustment of the polarity of monomer. Furthermore, the morphology of PGMA/poly(styrene-methacrylic acid) (PGMA/P(S-MAA)) microparticles from microcavity to erythrocyte shape is controlled by the polarity of seed surface. From transmission electron microscopy images of PGMA/P(S-MAA) microparticles, a fresh polymer particle appears in the cavity. To confirm this phenomenon, thermal annealing process in dioxane/water solution is carried out. Considering the flexibility of polymers, the openings and closing of the prepared microparticles are regulated following the increase in volume ratio of dioxane/water. The substrate bias effects on high vacuum arc ion plating (HV-AIP) of NiCrAlY coatings on superalloy were investigated at the vacuum level of 10 − 3 –10 − 4 Pa, which are 2–3 orders in magnitude better than that of conventional low vacuum arc ion plating (LV-AIP). The negative bias applied to the substrate was in the range of − 50 V to − 450 V. Microstructures, deposition rates, and chemical compositions of the coatings are significantly influenced by the negative bias. Except for commonly observed bias induced densification in microstructure and decrease in deposition rate, strong preferential sputtering effect at high bias was observed. The sputtering yield for Cr was estimated to be ranged from ~ 0.1 to ~ 0.2 at bias from − 50 V to − 450 V, while those for Ni and Al were from ~ 0.1 to 1.5. As a result, the major phase constitution of the coatings changed from γ-Ni/γ′-Ni 3 Al phases at bias of − 50 V to α-Cr phase at bias of − 450 V. Such stronger preferential sputtering effect in HV-AIP fosters a strategy for preparing compositionally gradient NiCrAlY coatings just by control of the substrate bias without the need of further treatments or multi-targets. In this work, combining poly (methylene green)/glucose dehydrogenase (PMG/GDH) bioanode with Prussian blue/glucose oxidase (PB/GOD) biocathode, we developed a hybrid bioelectrochemical device based on glucose/O 2 enzymatic biofuel cell (EBFC) to achieve energy conversion and storage. Depending on the reversible redox behaviors, PMG and PB play roles of dual-functional nanomaterials, not only as mediators to accelerate electrons transport between enzymes and electrodes surface, but also as pseudocapacitive materials to achieve energy storage. As a result, this hybrid device exhibited superior performance towards energy storage with a specific capacitance of 158.5 mF cm−2 at the current density of 1 mA cm−2. Meanwhile, a maximum output power density of 783.5 μW cm−2 in the pulse mode could be obtained, which had a prominent increment than that of the prototypical EBFC (87.5 μW cm−2) in the steady state. Furthermore, this hybrid device also exhibited better operational stability in long-term charge/discharge cycles test, at the current pulse of 1 mA cm−2. Through the combination of bioelectrocatalysis and capacitive energy storage, this developed hybrid device achieves effective energy conversion and storage, and provides a high output power to address the insufficient power supply of traditional EBFCs, which may enlarge the practical application of EBFCs in self-powered implantable and wearable devices. High vacuum (HV) arc ion plating (AIP) was developed for depositing NiCrAlY coatings at vacuum levels of 10 −3 –10 −4 Pa, which are 2–3 orders in magnitude better than that of conventional low vacuum (LV) AIP. Microstructure and oxidation behavior of the NiCrAlY coatings prepared by HV- and LV-AIP were comparatively investigated. As compared with the LV coatings, the HV coatings showed dramatically lower porosity and better oxidation resistance by forming purer alumina scales during oxidation. H3PW12O40 (PW12) was hybridized with 3-aminopropyltriethoxysilane modified graphene oxide (GO–NH2) sheets using a facile impregnation method, for the removal of organic pollutants in water. The obtained hybrid (PW12/GO–NH2) was characterized by elemental analysis, X-ray diffraction (XRD), Fourier transform infrared spectroscopy (FT-IR), Raman spectroscopy, X-ray photoelectron spectroscopy (XPS), UV-vis diffuse reflectance spectroscopy (DRS), photoluminescence spectroscopy (PL), transmission electron microscopy (TEM), and photo-electrochemical measurements. Its removal efficiency for organic pollutants was evaluated by the removal of methyl orange (MO). The results show that about 61.8% of MO is adsorbed onto the PW12/GO–NH2 hybrid upon 2 h of mixing, and after 2 h of irradiation under a high-pressure mercury lamp, MO is further decreased to 17.7%. The adsorption capacity of PW12/GO–NH2 hybrids is enhanced as compared with GO–NH2, probably due to enhanced electrostatic attraction and hydrogen-bonding interactions between the hybrid and MO. And the hybrid exhibits a better photocatalytic activity than the homogenous photocatalyst PW12 (1.36 times), which is probably attributed to the effective separation of photo-generated electron–hole pairs. Furthermore, the trapping experiment of active species during the photocatalytic degradation was carried out over PW12/GO–NH2, revealing that the oxidation of MO by O2Ṗ− and ṖOH plays a major role in the photocatalytic degradation process. The R2R3MYB proteins comprise one of the largest families of transcription factors and play regulatory roles in developmental processes and defense responses in plants. However, there has been relatively little effort to systematically carry out comprehensive genomic and functional analyses of these genes in tomato ( Solanum lycopersicum L.), a reference species for Solanaceae plants, and the model plant for fruit development. In this study, a total of 121 R2R3MYB genes were identified in the tomato genome released recently and further classified into 29 subgroups based on the phylogenetic analysis of the complete protein sequences. Phylogenetic comparison of the members of this superfamily among tomato, Arabidopsis, grape, rice, poplar, soybean, cucumber and apple revealed that the putative functions of some tomato R2R3MYB proteins were clustered into the Arabidopsis functional clades. The chromosome distribution pattern revealed that tomato R2R3MYB genes were enriched on several chromosomes and 52 % of the family members were tandemly duplicated genes. Tissue specificity or different expression levels of SlR2R3MYBs in different tissues suggested differential regulation of tissue development as well as metabolic regulation. The transcript abundance level analysis during abiotic conditions identified a group of R2R3MYB genes that responded to one or more treatments suggesting that the SlR2R3MYBs played major roles in the plant response to abiotic conditions and involved in signal transduction pathways. Immunoglobulin Y (abbreviated as IgY) is a type of immunoglobulin that is the major antibody in bird, reptile, and lungfish blood. IgY consists of two light (λ) and two heavy (υ) chains. In the present study, polyclonal antibody against IgYFc was generated and evaluated. rIgYCυ3/Cυ4 was expressed in Escherichia coli, purified and utilized to raise polyclonal antibody in rabbit. High affinity antisera were obtained, which successfully detected the antigen at a dilution of 1:204,800 for ELISA assay. The antibody can specifically recognize both rIgYCυ3/Cυ4 and native IgY by Western bolt analysis. Furthermore, the serum of Grus japonensis or immunoglobulin of chicken, duck, turkey, and silkie samples and dynamic changes of serum GoIgY after immunogenicity with GPV-VP3-virus-like particles (GPV-VP3-VLPs) can be detected with the anti-GoIgYFc polyclonal antibody. In the present study, ultrasound (400 W, U), microwave heating (75 ℃ for 15 min, M) and ultrasound synergized with microwave heating (UM) pretreatments of whey protein isolate (WPI) were applied to investigate and compare their influence on structure, physicochemical and functional characteristic of transglutaminase (TGase)-induced WPI. From the results of size exclusion chromatography, it could be seen that all three physical pretreatments could promote the formation of polymers in TGase cross-linked WPI, whose polymer amounts were increased by the order of U, UM and M pretreatment. Among three physical methods, M pretreatment had the strongest effect on structure and functional characteristics of TGase-induced WPI. Furthermore, compared with TGase-induced WPI, α-helix and β-turn of M−treated TGase-induced WPI (M−WPI−TGase) were reduced by 7.86% and 2.93%, whereas its β-sheet and irregular curl were increased by 15.37% and 7.23%. Zeta potential, emulsion stability and foaming stability of M−WPI−TGase were increased by 7.8%, 59.27% and 28.95%, respectively. Abstract: Interferon-α (IFN-α) genes have been cloned from a variety of animals, but information regarding crane IFN-α has not been reported to date. In this study, we cloned a full-length Red-crowned Crane interferon-α (crIFN-α) gene sequence consisting of a 486bp partial 5′ UTR, 741bp complete ORF and 559bp partial 3′ UTR. This gene encodes a protein of 246 amino acids and shares 60 to 80% identity with avian IFN-α and less than 45% identity with mammalian IFN-α. The expression of crIFN-α with an N-terminal His-tag was investigated in Escherichia coli, and the protein was purified on a nickel column. To obtain activated proteins, crIFN-α inclusion bodies were renatured by dialysis. In vitro cytopathic inhibition assays indicated that the recombinant crIFN-α could inhibit the replication of vesicular stomatitis virus in chicken fibroblasts. These antiviral activities were abrogated by rabbit anti-crIFN-α antibodies in vitro. In addition, an immunofluorescence assay indicated that crIFN-α could be expressed in chicken fibroblasts and was primarily located in the cytoplasm. The WD-repeat ( WDR) proteins comprise an astonishingly diverse superfamily of regulatory proteins. To date, genome-wide characterization of this family has only been conducted in Arabidopsis and little is known about WDR genes in cucumber ( Cucumis sativus L.). This study identified 191 cucumber WDR genes in the latest cucumber genome and the CsWDR family contained a smaller number of identified genes compared to Arabidopsis. The results of this study were also supported by genome distribution and gene duplication analysis. Phylogenetic analysis showed that the WDR proteins could be classified into 21 subgroups. Moreover, an additional 12 AtWDR proteins were also identified and a complete overview of this gene family in Arabidopsis is presented, including the phylogeny, chromosome locations and duplication events. In addition, a comparative analysis between these genes in cucumber and Arabidopsis was performed and it suggested that there was strong gene conservation and that there was an expansion of particular functional genes during the evolution of the two species. The transcript abundance level analysis during abiotic stress (NaCl, ABA and low temperature treatments) identified six CsWDR genes that responded to one or more treatments. Tissue-specific expression profiles of these six genes were also analyzed. This study has produced a comparative genomics analysis of the WDR gene family in cucumber and Arabidopsis and provides the first steps towards the selection of CsWDR genes for cloning and functional dissection that can be used in further studies into their roles in cucumber stress resistance. A herpesvirus was isolated during a diagnostic investigation of severe cases of conjunctivitis in feral pigeons ( Columba livia f. domestica ). Isolates of the virus were recovered from throat swabs of the pigeons followed by inoculation of the swab samples in chicken embryo fibroblasts. Pigeons inoculated with the isolated virus had similar clinical signs to those observed in naturally infected birds. Transmission electron microscopy revealed viral structures with typical herpesvirus morphology. Polymerase chain reaction amplification, using herpesvirus-identifying primers resulted in an amplicon of the expected size for herpesvirus. Sequencing of these amplicons and database comparisons identified the herpesvirus UL30 homologue. Phylogenetic reconstructions suggested that the isolated herpesvirus belongs to the Mardivirus genus of Alphaherpesvirinae . Floating solar still is considered as a land-saving, easy-operating and environmental-friendly approach for freshwater production. However, the low yield and work instability in actual weather conditions limits its application. To overcome these disadvantages, a bionic floating solar still inspired by water intake of plant roots was presented in this paper. The channels for water supply were distributed in array and wrapped in a whole thermal insulation sheet. With the design of simulating roots, the present desalination system achieves low heat losses, water-absorbing uniformity and structural stability. Cellulose fabric and glass were separately selected as evaporation and condensation materials of the experimental prototype after preliminary test. The outdoor experiment was conducted in Xingtai, China, and the accumulated radiation in the test day was 21.96 MJ/m2. The maximum evaporation temperature and daily freshwater yield reached 59.2 °C and 1.5 kg/m2/d, respectively, which were the highest in comparison with existing floating solar stills. Next, the effect of rolling motion on humid air flow inside the still was investigated by CFD simulation. Vibrio vulnificus is an important foodborne pathogenic bacterium that mainly contaminates seafood. Rapid and accurate technologies that suitable for on-site detection are critical for effective control of its spreading. Conventional detection methods and polymerase chain reaction (PCR)-based and qPCR-based approaches have application limitations in on-site scenarios. Application of loop-mediated isothermal amplification (LAMP) technology was a good step towards the on-site detection. In this study, a recombinase polymerase amplification (RPA)-based detection method for V. vulnificus was developed combining with lateral flow strip (LFS) for visualized signal. The method targeted the conservative empV gene encoding the extracellular metalloproteinase, and finished detection in 35 min at a conveniently low temperature of 37 °C. It showed good specificity and an excellent sensitivity of 2 copies of the genome or 10−1 colony forming unit (CFU) per reaction, or 1 CFU/10 g in spiked food samples with enrichment. The method tolerated unpurified templates directly from sample boiling, which added the convenience of the overall procedure. Application of the RPA-LFS method for clinical samples showed accurate and consistent detection results compared to bioassay and quantitative PCR. Efficient allocation of the essential nutrient potassium (K+) is a central determinant of plant ion homeostasis and involves AKT2 K+ channels. Here, we characterize four AKT2 K+ channels from cotton and report that xylem and phloem expressed GhAKT2bD facilitates K+ allocation and that AKT2‐silencing impairs plant growth and development. We uncover kinase activity‐dependent activation of GhAKT2bD‐mediated K+ uptake by AtCBL4–GhCIPK1 calcium signalling complexes in HEK293T cells. Moreover, AtCBL4–AtCIPK6 complexes known to convey activation of AtAKT2 in Arabidopsis also activate cotton GhAKT2bD in HEK293T cells. Collectively, these findings reveal an essential role for AKT2 in the source‐sink allocation of K+ in cotton and identify GhAKT2bD as subject to complex regulation by CBL–CIPK Ca2+ sensor–kinase complexes. The middle and lower reaches of the Yangtze River (MLYR) plain represent the second-largest wheat producing area in China; the winter wheat-rice system is one of the main planting systems in this region. The use of the agricultural production system simulator (APSIM)-wheat model to simulate wheat production potential and evaluate the impact of future climate change on wheat production in this region is of great importance. In this study, the adaptability of the APSIM-wheat model in the MLYR was evaluated based on observational data collected in field experiments and daily meteorological data from experimental stations in Wuhan, Jingmen, and Xiangyang in Hubei province. The results showed significant positive relationships between model-predicted wheat growth duration from sowing to anthesis and maturity and the observed values, with coefficients of determination (R2) in ranges of 0.90–0.97 and 0.93–0.96, respectively. The normalized root-mean-square error (NRMSE) of the simulated growth durations and measured values were lower than 1.6%, and the refined index of agreement (dr-values) was in the range of 0.74–0.87. The percent mean absolute relative error (PMARE) was cited here as a new index, with a value below 1.4%, indicating that the model's rating was excellent. The model's performance in terms of grain yield and above-ground biomass simulation was also acceptable, although it was not as good as the growth periods simulation. The R2 value was higher than 0.75 and 0.72 for the simulation of grain yield and biomass, respectively. The indices NRMSE and PMARE were lower than 19.8% and 19.9%, and the dr-value was higher than 0.71. According to our results, APSIM-wheat was an effective and accurate model for simulating the phenology and yield production processes of wheat in the MLYR, and the results also provided a theoretical basis and technical support for further research on the yield potential of wheat-rice rotation planting systems with clarification of the key factors limiting the yield gap in this region. Background: Age-related cognitive decline has a significant impact on the health and longevity of older adults. Circulating very long-chain saturated fatty acids (VLSFAs) may actively contribute to the improvement of cognitive function. The objective of this study was to investigate the associations between arachidic acid (20:0), docosanoic acid (22:0), tricosanoic acid (23:0), and lignoceric acid (24:0) with cognitive function in older adults. Methods: This study used a dataset derived from the 2011–2014 National Health and Nutrition Examination Survey (NHANES). A total of 806 adults (≥ 60 years) were included who underwent comprehensive cognitive testing and plasma fatty acid measurements. Multivariable linear regression, restricted cubic spline (RCS), and interaction analyses were used to assess associations between VLSFAs and cognitive function. Partial Spearman' s correlation analysis was used to examine the correlations between VLSFAs and palmitic acid (16:0), high-density lipoprotein cholesterol, low-density lipoprotein cholesterol, total cholesterol, triglycerides, systemic inflammatory markers, and dietary nutrients. Results: Multivariable linear regression analysis, adjusting for sociodemographic, clinical conditions, and lifestyle factors, showed that 22:0 and 24:0 levels were positively associated with better global cognitive function (β = 0.37, 95% confidence interval [CI] = 0.01, 0.73; β = 0.73, 95% CI = 0.29, 1.2, respectively) as well as better CEARD-DR Z-score (β = 0.82, 95% CI = 0.36, 1.3 and β = 1.2, 95% CI = 0.63, 1.8, respectively). RCS analysis showed linear associations between higher 22:0 and 24:0 levels and better cognitive performance in both global cognitive function and CERAD-DR tests. Conclusions: The study suggests that higher levels of 22:0 and 24:0 are associated with better global cognitive function in older adults. 22:0 and 24:0 may be important biomarkers for recognizing cognitive impairment, and supplementation with specific VLSFAs (22:0 and 24:0) may be an important intervention to improve cognitive function. As a potential diagnostic index, it has attracted the interest of many physicians. However, the correlation between the PNI and post-stroke cognitive impairment (PSCI) remains unclear.A total of 285 patients with acute ischemic stroke were included. PNI was assessed as serum albumin (g/L) + 5× lymphocyte count (109/L) and was dichotomized according to the prespecified cut-off points 48.43 for the high and low groups. PSCI was defined as Mini-Mental State Examination (MMSE) < 27 at the 6–10 months follow-up. Multiple logistic regression and linear regression analyses were performed to examine the association between PNI and cognitive outcomes.A low PNI was independently associated with PSCI after adjusting for age, sex, education, National Institutes of Health Stroke Scale (NIHSS), deep white matter hyperintensity (DWMH), and stroke history (odds ratio [OR]: 2.158; 95% confidence interval [CI]: 1.205–3.863). The PNI scores were significantly associated with MMSE and attention domain (β = 0.113, p = 0.006; β = 0.109, p = 0.041, respectively). The PNI improved the model’s discrimination when added to the model with other clinical risk factors.A low PNI was independently associated with the occurrence of PSCI and the PNI scores were specifically associated with the scores of global cognition and attention domain. Polyporaceae is an important fungal family that has been a source of natural products with a range of pharmaceutical activities in China. In our previous study, two polysaccharides, PCWPW and PCWPS, with significant antioxidant and antidepressant activity were obtained from Poria cocos. In this study, we evaluated their potential molecular mechanisms in the immunomodulation of macrophages. PCWPW and PCWPS were characterized by GC–MS analysis to contain 1,3-linked Glcp. ELISA assays results demonstrated that the secretion of TNF-α was significantly enhanced by PCWPW/PCWPS. RNA-seq data demonstrated that PCWPS treatment modulated the expression of immune-related genes in macrophages, which was further confirmed by RT-qPCR assays. The activation of TNF-α secretion was found to be mannose receptor (MR) dependent and suppressed by MR inhibitor pretreatment. Moreover, the amount of TNF-α cytokine secretion in PCWPW/PCWPS-induced RAW264.7 cells was decreased when pretreated with NF-κB or MAPK signaling pathway inhibitors. Collectively, our results suggested that PCWPW and PCWPS possessed immunomodulatory activity that regulates TNF-α expression through the NF-κB/MAPK signaling pathway by binding to mannose receptors. Background: Abdominal obesity has become an important public health issue in China. Socioeconomic disparities are thought to be closely related to the prevalence of abdominal obesity. Exploring socioeconomic disparities in abdominal obesity over the life course in China could inform the design of new interventions to prevent and control abdominal obesity. Methods: The China Health and Nutrition Survey (CHNS) was a prospective household-based study involving seven rounds of surveys between 1993 and 2011. Twenty three thousand, two hundred and forty-three individuals were followed up over an 18-year period. The mixed effects models with random intercepts were used to assess the effects on abdominal obesity. Six key socioeconomic indicators, with age and age-squared added to the models, were used to identify socioeconomic disparities in abdominal obesity over the adult life course. Results: Prevalence of abdominal obesity increased non-linearly with age over the adult life course. Abdominal obesity was more prevalent in younger than older birth cohorts. Positive period effects on the prevalence of abdominal obesity were substantial from 1993 to 2011, and were stronger among males than females. Prevalence of abdominal obesity was higher among ethnic Han Chinese and among the married [coefficient (95% confidence intervals): 0.03(0.003, 0.057) and 0.035(0.022, 0.047), respectively], and was lower among males [coefficient (95% confidence intervals): − 0.065(− 0.075,-0.055)]. A higher-level of urbanization and higher household income increased the probability of abdominal obesity [coefficient (95% confidence intervals): 0.160(0.130, 0.191), 3.47E− 4 (2.23E− 4, 4.70E− 4), respectively], while individuals with more education were less likely to experience abdominal obesity [coefficient (95% confidence intervals): − 0.222 (− 0.289, − 0.155)] across adulthood. Conclusions: In China, abdominal obesity increased substantially in more recent cohorts. Simple Summary: Tritrichomonas muris (T. muris) mainly parasitizes the ceca of different rodent species worldwide. To date, there are no ideal methods for the detection of T. muris infections in laboratory mice. This study developed a nested PCR reaction using a novel specific primer based on the conserved regions of the SSU rRNA gene of T. muris for a molecular epidemiological survey (investigation) of T. muris infections in laboratory mice. The results showed that the nested PCR system has higher sensitivity, reliability, and specificity. The nested PCR system showed an infection rate of T. muris of 18.96% (58/306), which was higher than the infection rate of 14.05% (43/306) that was detected via smear microscopy in fecal samples from five mouse strains. The sensitivity and specificity of nested PCR in detecting T. muris was found to be 100%, and it demonstrated a 26% increase in diagnostic sensitivity compared to the smear microscopy method. The present study provides a new method for the molecular epidemiological investigation of T. muris infections in laboratory mice. A variety of rodent ceca are parasitized by Tritrichomonas muris (T. muris), a flagellated protozoan. The nested PCR system for T. muris showed good specificity and high sensitivity for at least 100 T. muris trophozoites/mL and 0.1 ng/μL of fecal genomic DNA, which means that 176 trophozoites per gram of mouse feces could be detected. When using this nested PCR system, the detection rate was 18.96% (58/306), which was higher than the detection rate of 14.05% (43/306) detected via smear microscopy in fecal samples from five mouse strains. The sensitivity and specificity of nested PCR in detecting T. muris was found to be 100%, and it demonstrated a 26% increase in diagnostic sensitivity compared to the smear microscopy method in the present study. In conclusion, the nested PCR developed with novel primers based on the SSU rRNA gene of T. muris has good accuracy, specificity, and sensitivity for the detection of T. muris infections in laboratory mice.
