OBJECTIVE To investigate the value of CT radiomics in predicting epidermal growth factor mutation status in lung cancer.MATERIALS AND METHODS Data from Hospital 1 (n = 119) and Hospital 2 (n = 49) were used as the internal dataset and data from the Cancer Genome Project (TCGA) database (n = 14) were used as the external validation set. After image segmentation, feature extraction and feature downscaling screening, radiomics models were constructed. Clinical features and conventional imaging markers used to predict EGFR mutation status were screened by univariate and multivariate logistic regression to construct the clinical model. Combining radiomic features and clinical features by multivariate logistic regression to construct a combined model. The predictive performance of each model was assessed using operator operating characteristic (ROC) curves, and the clinical utility of each model was assessed using clinical decision curves.RESULTS Three models based on CT flat-scan were developed: a radiomics model, a clinical model and a combined model. The area under the curve (AUC) for the training set radiomics, clinical and combination models were 0.0.87 (0. 79-0.95), 0.68 (0.57–0.80) and 0.87 (0.80–0.95) respectively; the AUC values for the validation set radiomics, clinical and combination models were 0.73 (0.55–0.90), 0.71 (0.54–0.88) and 0.73 (0.56–0.90) respectively.The clinical decision curves indicate that the combined model has better clinical decision benefits than the radiomics model alone and the clinical model alone.CONCLUSION The combined model combining radiomic features and clinical features further improved the predictive efficacy compared to the radiomic model alone and the clinical model alone.

Objective. To investigate the significance of PAX8-PPARγ expression in thyroid cancer and the application of a PAX8-PPARγ-targeted ultrasound contrast agent in the early diagnosis of thyroid cancer. Methods. In this study, the expression of PAX8-PPARγ in thyroid cancer tissues, paracancer groups, and normal thyroid tissues was detected by western and immunohistochemical techniques; the effects of PAX8-PPARγ expression inhibition on thyroid cancer cell growth, clonogenic ability, and antiapoptosis were examined. The terminal carboxylactic acid/hydroxyacetic acid copolymer (PLGA-COOH) nanoparticles were prepared by the double emulsification solvent volatilization method. The in vitro cytotoxicity of the targeted contrast agent was detected by MTS and other methods; LD50 was used to evaluate its short-term in vivo toxicity after intraperitoneal injection in mice. The number of patients with lung cancer is difficultly diagnosed in the early stage. The purpose of the study was to investigate the effects of CT- and ultrasound-guided percutaneous transthoracic needle biopsy combined with serum CA125 and CEA on the diagnosis of lung cancer. 120 patients with suspected lung cancer admitted to our hospital from January 2019 to January 2020 were selected and divided into an ultrasound group (n = 60) and CT group (n = 60), according to different percutaneous transthoracic needle biopsy modalities. All patients received serum tumor markers detection, so as to compare the CT- and ultrasound-guided percutaneous transthoracic needle biopsy results and pathology results, levels of serum tumor markers among all patients and the patients with different lung cancer types, and diagnostic efficacy of tumor markers, as well as complication rate (CR) in patients. The sensitivity and specificity of ultrasound-guided percutaneous transthoracic needle biopsy were 0.880 and 0.800, respectively, while those of CT-guided percutaneous transthoracic needle biopsy were 0.909 and 0.625, respectively; the CA125 and CEA levels in the lung cancer group were higher than those in the benign group ( P < 0.001 ); the CA125 and CEA levels of the patients with adenocarcinoma were higher than those with squamous carcinoma, and the CEA levels of the patients with small-cell carcinoma were lower than those with adenocarcinoma ( P < 0.05 ); the sensitivity, specificity, and Youden indexes of CA125 were 0.638, 0.833, and 0.471, respectively, while those of CEA were 0.766, 0.778, and 0.544, respectively; there were no significant differences in CR between the two groups ( P > 0.05 ). CT- and ultrasound-guided percutaneous transthoracic needle biopsy is a safe and feasible diagnostic modality for lung cancer, and its combination with serum CA125 and CEA can significantly improve the accuracy of the detection results, which is worthy of promotion and application in clinical practice.

Severe soil erosion occurs on the Loess Plateau in China, which makes the Yellow River the most sediment-laden river in the world. Construction of about 60,000 sediment check dams has remarkably controlled soil erosion on the Loess Plateau and reduced the sediment load of the middle and lower Yellow River. Nonetheless, little is known about the mechanism of erosion control and vegetation development of sediment check dams. The function of a single check dam mainly is trapping sediment, while the function of a train of check dams comprising dozens of or over hundreds of check dams in a gully encompasses controlling bed incision and reducing erosion energy. A formula was proposed to calculate the potential energy of bank failure and slope failure in a gully, which essentially constitutes the erosion energy. The erosion energy increases when gully incision occurs, which is induced by the incision of the Yellow River and its tributaries on the Loess Plateau. Sediment deposition in many gullies due to construction of check dams reduces the erosion energy to almost zero, which in turn greatly reduces soil erosion and sediment yield. Construction of check dams promotes vegetation development. The vegetation-erosion dynamics model was used to study the effect of check dams on vegetation development. Simulation results show that reforestation without check dam construction might result in an increase of vegetation cover in the first ten years and then a drop of vegetation cover to less than 10% in the later years. The check dams provide a foundation for vegetation development.

Disruption of endothelial cell (ECs) and pericytes interactions results in vascular leakage in acute lung injury (ALI). However, molecular signals mediating EC–pericyte crosstalk have not been systemically investigated, and whether targeting such crosstalk could be adopted to combat ALI remains elusive. Using comparative genome-wide EC–pericyte crosstalk analysis of healthy and LPS-challenged lungs, we discovered that crosstalk between endothelial nitric oxide and pericyte soluble guanylate cyclase (NO–sGC) is impaired in ALI. Indeed, stimulating the NO–sGC pathway promotes vascular integrity and reduces lung edema and inflammation-induced lung injury, while pericyte-specific sGC knockout abolishes this protective effect. Mechanistically, sGC activation suppresses cytoskeleton rearrangement in pericytes through inhibiting VASP-dependent F-actin formation and MRTFA/SRF-dependent de novo synthesis of genes associated with cytoskeleton rearrangement, thereby leading to the stabilization of EC–pericyte interactions. Collectively, our data demonstrate that impaired NO–sGC crosstalk in the vascular niche results in elevated vascular permeability, and pharmacological activation of this crosstalk represents a promising translational therapy for ALI.

Photoelectrochemical (PEC) biosensing as a promising and largely developing technique has been widely applied in biological analysis in recent years because of its low background signal and high sensitivity. By utilizing suitable PEC active materials to establish a photoelectric (PE) conversion system, selective and sensitive measurements can be achieved with the help of specific biological recognition elements. PEC biosensors rely on the change of photocurrent that depends on the electron transfer process of nanomaterials. Therefore, the electron manipulation of PEC active nanomaterials is crucial for PEC sensing. In this review, from the perspective of the electron transfer manipulation of PEC active nanomaterials, we summarize the principle of PEC biosensors in three parts

Using miRNA-21 as a model analyte and combining with DSN-assisted amplification, a dual signal amplification strategy for ultrasensitive miRNA biosensing with a low detection limit (0.53 fM) and satisfactory selectivity was designed.

Hyperconcentrated flows often occur in the middle and lower Yellow River (MLYR) and its tributaries, within which the main sediment source originates from the Loess Plateau of China due to serious water erosion. Little is known about the properties of river sediment that is transported by hyperconcentrated flows, particularly with respect to the mineral composition and size distribution. Samples of sediment and loess were collected in the northern, middle, and southern Loess Plateau and the mainstream and tributaries of the MLYR. A total of 18 loess samples and 24 river sediment samples were analyzed to determine their sediment size distribution and mineral composition. The bottom loess samples reflected the original sedimentary features of the Loess Plateau, and the median particle size reduced, and the clay content increased from the north to the south of the study region. The surface loess has been weathered under the action of wind and rainfall, and the clay particle content in the surface loess samples was higher than that in the undisturbed bottom loess. Erosion of the surface soil due to rainfall and surface runoff means that fine particles (mostly clay) have been washed away. The median diameter of surface loess particles was a little larger than that of the bottom loess particles where water erosion dominates. The particle size became coarser with increasing distance from the estuary in the MLYR, which reflects depositional sorting in the river channel. Significant logarithmic relations were found between the median diameter of the sediment particles and the i) non-clay mineral content and ii) clay mineral content. Thus, clay and non-clay mineral compositions can be conveniently estimated from the particle size distribution.

Since the discovery of the enzyme-like activities of nanomaterials, the study of nanozymes has become one of the most popular research frontiers of diverse areas including biosensors. DNA also plays a very important role in the construction of biosensors. Thus, the idea of combined applications of nanozymes with DNA (DNA-nanozyme) is very attractive for the development of nanozyme-based biosensors, which has attracted considerable interest of researchers. To date, many sensors based on DNA-functionalized or templated nanozymes have been reported for the detection of various targets and highly accelerated the development of nanozyme-based sensors. In this review, we summarize the main applications and advances of DNA-nanozyme-based sensors. Additionally, perspectives and challenges are also discussed at the end of the review.

The fluvial geomorphology in tectonically active (particularly rapid uplift) regions often undergoes continuous change. The rapid uplift is coincident with high erosion rates; consequently, incised valleys are formed. Mass flows (for example, avalanches, landslides, and debris flows) in incised valleys can markedly influence fluvial processes and even reshape valley geomorphology. However, these processes and long-term evolution corresponding to mass flows require further clarification. Field campaigns were carried out in the region near the Yigong Tsangpo and Palong Tsangpo Rivers (hereafter the Yigong and Palong Rivers), the two largest tributaries of the lower Yarlung Tsangpo River, to examine the feedback between fluvial processes and mass flows. Remote sensing images from recent decades were used to compare the channel morphology before and after typical mass flows (particularly catastrophic ones). The morphology of the lower Yigong River has evidently been impacted by landslides, while that of the Palong River has mainly been shaped by glacial processes and debris flows. At present, the morphology of the latter consists of alternating sections of gorges and wide valleys, with a staircase-like longitudinal profile. The gorge sections exhibit single and deeply incised channels with a high-gradient channel bed and terraces. In contrast, the wide valley sections consist of lakes, braided or anabranching channels, gentle bed gradients, and thick alluvial deposits. Debris flows occur more frequently in gullies in the reaches of the gorge sections and rarely in gullies along the wide valley sections. The occurrence of mass flow events has resulted in an imbalance of the previous (quasi-)equilibrium in the river morphology; however, this has triggered negative feedback that is driving the transient river morphology to a new state of (quasi-)equilibrium.

In this work, a facile and sensitive colorimetric detection method was firstly reported for RNase A activity detection based on target regulated protection effect of chimeric DNA probe on the salt-induced aggregation of plasmonic gold nanoparticles. Compared with previous works of RNase A activity detection, this colorimetric assay integrated the advantages of sensitive, low cost, facile operation, rapid response and low biological toxicity.

An iron-nickel bimetallic metal−organic framework (FeNi-MOF) with inner peroxidase-like activity as a support for enzyme immobilization was synthesized for the first time. And the GOx-mediated synthesis of mimic multienzyme system (GOx/FeNi-MOF) was obtained by a one-step biomimetic mineralization. FeNi-MOF played a dual role of a peroxidase mimic and a protective coating. Therefore, the obtained multienzyme system showed both peroxidase-like activity of FeNi-MOF and the biological activity of natural enzyme. FeNi-MOF acted as a peroxidase mimic, showing a higher affinity for hydrogen peroxide (H2O2). The enhanced catalytic activity of the FeNi-MOF may result from synergistic effect between iron and nickel. In addition, the GOx/FeNi-MOF was selected as a proof of concept and succeeded in a one-step colorimetric detection of glucose by tandem catalysis. A small amount of the product was used for glucose detection, and the detection range was 0.3–35 mM, which indicated that our sensor can meet the clinical needs for diagnose diabetes.

In this work, a method for quantifying the activity of formamidopyrimidine DNA glucosylase (Fpg) was designed based on phosphate group (P)-modulated multi-enzyme catalysis and fluorescent copper nanoclusters (CuNCs). By eliminating 8-oxoguanine from double-stranded DNA, Fpg generates a nick with P at both 3' and 5' termini. Subsequently, part of the DNA is digested by 5'P-activated lambda exonuclease (λ Exo), and the generated 3'P disables exonuclease I (Exo I), resulting in the generation of single-stranded DNA containing poly(thymine) (poly(T)). Using poly(T) as templates, CuNCs were prepared to emit intense fluorescence as the readout of this method. However, in the absence of Fpg, the originally modified 5'P triggers the digestion of λ Exo. In this case, fluorescence emission is not obtained because CuNCs cannot be formed without DNA templates. Therefore, the catalysis of λ Exo and Exo I can be tuned by 5'P and 3'P, which can be further used to determine the activity of Fpg. The fluorescent Fpg biosensor works in a "signal-on" manner with the feature of "zero" background noise, and thus shows desirable analytical features and good performance. Besides, Fpg in serum samples and cell lysate could be accurately detected with the biosensor, indicating the great value of the proposed system in practical and clinical analysis.

Quantification of circulating tumor DNA (ctDNA) is of great importance in liquid biopsy but difficult due to its low amount in bodily fluids. To meet this high demand, a novel method for ctDNA detection is established by quantifying cyclic DNA polymerization using lanthanide coordination polymers (Ln-CPs). Relying on the coordination between the pyrophosphate ion (PPi) and trivalent cerium ion (Ce3+), organic ligand-free PPi–Ce coordination polymer networks (PPi–Ce CPNs) with enhanced fluorescence are prepared for the first time. By surveying the optical properties of PPi–Ce CPNs, it is found that PPi regulates electric-dipole transition of Ce3+ to the lowest excited state, thus facilitating the emission of fluorescence. Therefore, fluorescence enhancement of PPi–Ce CPNs originates from the ligand field effect rather than the normal antenna effect. Moreover, a new strategy to quantify DNA polymerization is developed based on PPi–Ce CPNs. By introducing multifold cyclic DNA polymerization, a small amount of ctDNA triggers the exponential generation of PPi to form plenty of PPi–Ce CPNs. Accordingly, a biosensor is constructed for sensitive ctDNA detection by measuring the intense fluorescence of PPi–Ce CPNs. The biosensor is capable of sensing ctDNA at the sub-femtomolar level, which is far better than the analytical performances of commercial dyes. Besides, the analytical method is able to detect single nucleotide polymorphism and determine ctDNA in real samples. Considering that DNA polymerization is widely used in bio-recognition, bio-assembly and biomineralization, the work provides a versatile quantitative strategy of making relevant processes precise and controllable., DNA polymerization is quantified by fluorescent pyrophosphate–cerium lanthanide coordination polymer networks, and is used to detect circulating tumor DNA sensitively.

Debris flows in waterways can transport large amounts of sediment downstream, which can cause serious damage and economic losses. The vegetation cover in the valley of the Xiaojiang River in Yunnan Province, China—classified as a dry-hot valley—was significantly reduced by logging in the 1950s. Soil erosion intensified and 107 gullies developed, which led to debris flows along the 86 km length of the river. Jiangjia Gully is a tributary of the Xiaojiang River. Historically, debris flows have occurred frequently, blocking the Xiaojiang River seven times between 1957 and 2000. Since 2000, the construction of check dams and afforestation have decreased the volume of debris flows in the three tributaries of Jiangjia Gully. However, different combinations of check dams and afforestation were adopted in the three tributaries of Jiangjia Gully, which has led to the different trends in debris flows behaviour. A new method was established to evaluate the mitigative effect of check dams and afforestation on debris flows. We found that the debris flow volume was proportional to the gravity energy of soil and rock on the gully bank and inversely proportional to the vegetation coverage in a dry-hot valley setting. The method revealed that under different gravity energy conditions, the implementation order of check dam construction and afforestation is important for debris flow mitigation.

RIPK1, a death domain-containing kinase, has been recognized as an important therapeutic target for inhibiting apoptosis, necroptosis, and inflammation under pathological conditions. RIPK1 kinase inhibitors have been advanced into clinical studies for the treatment of various human diseases. One of the current bottlenecks in developing RIPK1 inhibitors is to discover new approaches to inhibit this kinase as only limited chemotypes have been developed. Here we describe Necrostatin-34 (Nec-34), a small molecule that inhibits RIPK1 kinase with a mechanism distinct from known RIPK1 inhibitors such as Nec-1s. Mechanistic studies suggest that Nec-34 stabilizes RIPK1 kinase in an inactive conformation by occupying a distinct binding pocket in the kinase domain. Furthermore, we show that Nec-34 series of compounds can synergize with Nec-1s to inhibit RIPK1 in vitro and in vivo. Thus, Nec-34 defines a new strategy to target RIPK1 kinase and provides a potential option of combinatorial therapy for RIPK1-mediated diseases.

Polarization of photoactive materials in current photoelectric (PE) systems is difficult to be adjusted, and thus electron-transfer routes of these systems are unchangeable, which limits their performance in photoelectrochemical (PEC) analysis. Herein, we attempted to modulate the polarization of perovskite-based heterostructures by both in situ semiconductor generation and enzyme catalysis. Owing to their band alignments, Cs3Bi2Br9 quantum dots (QDs) and BiOBr are confirmed to construct a Z-scheme structure, leading to a large anodic photocurrent. In the presence of ascorbic acid 2-phosphate (AAP), BiPO4 is generated on the surface of the Cs3Bi2Br9 QDs/BiOBr heterostructure, reassigning energy bands of BiOBr. Accordingly, polarization of the photoactive materials is converted, and a new Z-scheme structure with a reversed electron-transfer route is constructed, which leads to an evident cathodic photocurrent. Furthermore, abundant electron donors can be obtained by catalyzing AAP with alkaline phosphatase (ALP). In this case, photogenerated holes in BiOBr are preferentially annihilated by electron donors, thereby blocking transfer of photogenerated electrons in the Cs3Bi2Br9 QDs/BiOBr/BiPO4 heterostructure. Consequently, a second polarization conversion is triggered by enzyme catalysis, resulting in the recovery of an anodic photocurrent. Benefited from the polarization conversion, a PEC biosensor with a feature of two-wing signal switch is designed, which remarkably enlarges the range of the signal response and subsequently improves the analytical performance. As a result, ALP in small volume of human serum can be quantified with this method. In this work, polarization of perovskite-based photoactive materials is tuned, proposing an alternative perspective on the design of advanced PE systems.

Antibody-drug conjugates (ADCs) are complex pharmaceutical molecules that combine monoclonal antibodies with biologically active drugs through chemical linkers. ADCs are designed to specifically kill disease cells by utilizing the target specificity of antibodies and the cytotoxicity of chemical drugs. However, the traditional ADCs were only applied to a few disease targets because of some limitations such as the huge molecular weight, the uncontrollable coupling reactions, and a single mechanism of action. Here we report a simple, one-pot, successive reaction method to produce dual payload conjugates with the site-specifically engineered cysteine and p-acetyl-phenylalanine using Herceptin (trastuzumab), an anti-HER2 antibody drug widely used for breast cancer treatment, as a tool molecule. This strategy enables antibodies to conjugate with two mechanistically distinct cytotoxic drugs through different functional groups sequentially, therefore, rendering the newly designed ADCs with functional diversity and the potential to overcome drug resistance and enhance the therapeutic efficacy.

Although attractive for their low toxicity, CuInS2/ZnS core/shell quantum dots (CIS/ZnS QDs) still suffer from poor luminescence efficiency and poor water solubility. Herein, two amino acids (AAs), i.e., cysteine (Cys) and threonine (Thr), are used to tune the properties of CIS/ZnS QDs by capping them in both core and shell. It is found that Thr can regulate the density of Cys on the surface of QDs, thus causing a synergistic effect on the enhancement of photoluminescence (PL) intensity. Capping in the shell mainly leads to the enhancement of PL intensity, and capping in the core results in a red-shift of PL wavelength. Accordingly, a new kind of near-infrared region CIS/ZnS QDs with improved optical properties has been prepared. In addition, the Cys- and Thr-capped CIS/ZnS QDs possess outstanding water solubility and biocompatibility. In this work, the QDs are further employed in Cd2+ determination and multicolor imaging, indicating their potential applications. Relying on the enhancement of PL intensity via cation exchange, the Cys- and Thr-capped CIS/ZnS QDs can sense Cd2+ sensitively. Notably, because ZnS shells of the QDs will not be affected by Zn2+, the analytical method can discriminate Cd2+ from Zn2+ depending on the inherent characteristics of QDs. Moreover, intercellular Cd2+ can also be evaluated by the bright PL from the QDs, and the QDs can achieve multicolor imaging. Overall, this work demonstrates that various properties of QDs may be tuned by capping with AAs, and AA-capped QDs are of great value in advanced biosensing and bioimaging.

Currently, reported affinity pairings still lack in diversity, and thus terminal protection relying on steric hindrance is restricted in designing nucleic acid-based analytical systems. In this work, resistance to exonuclease is testified by group modification or backbone replacement, and the 3′-phosphate group (P) reveals the strongest exonuclease I-resistant capability. Due to the substrate specificity of enzymatic catalysis, this 3′-P protection works in a “direct mode”. By introducing DNA templated copper nanoparticles, an alkaline phosphatase assay is performed to confirm the 3′-P protection. To display the application of this novel terminal protection, a multifunctional DNA is designed to quantify the model circulating microRNA (hsa-miR-21-5p) in serums from different cancer patients. According to our data, hsa-miR-21-5p-correlated cancers can be evidently distinguished from non-correlated cancers. Meanwhile, the effect of chemotherapy and radiotherapy on breast cancer is evaluated from the perspective of hsa-miR-21-5p residue in serums. Since greatly reducing the limitations of DNA design, this P-induced terminal protection can be facilely integrated with other DNA manipulations, thereby constructing more advanced biosensors with improved analytical performances for clinical applications.

Throughout the history of China, the Yellow River has been associated with flood disasters and changes in the course of its lower reaches because of sedimentation. From 602 B.C. to 1949 the river experienced 1593 levee bursts, flooding vast areas, and claiming millions of human lives. The river shifted its main course by avulsion 26 times with the apex around Zhengzhou, resulting in devastating calamities and numerous old channels. Training of the Yellow River has a history of more than 3000 yr. Levee construction has been the major strategy for flood control. Two extremely different strategies has been proposed and practiced in the past 2000 yr, i.e. the “wide river and depositing sediment” strategy and the “narrow river and scouring sediment” strategy. This paper analyzes the levee breaches and flood disasters in the past 2000 yr and compares the results of the two extremely different strategies. The “narrow river and scouring sediment” strategy has only short term effects on levee breach control and flood mitigation. The “wide river and depositing sediment” strategy can essentially mitigate flood disasters and reduce levee breaches for a long term period of time. The “wide river and depositing sediment” strategy has been used and no levee breach has occurred in the past 67 yr, which has been the only periods of more than 50 yr with no levee breaches in the history of the Yellow River since 700 A.D. Modern flood and sedimentation management methods have also been introduced, and the strategy of applying the ‘“widen the river and enhance the levees” approach for the upper and lower reaches management is proposed.

The excitonic response between nanomaterials is distance-dependent, and thus, interparticle distance is a key factor in fabricating diverse photoelectrochemical (PEC) systems. Current studies focus on DNA-mediated regulation of interparticle distance. However, limited by high demands of base-pairing and flexibility of DNA, it is hard for DNA to achieve precise regulation, especially in a short distance. To pursue better PEC performances in bioanalyses, alternative biological materials should be explored to replace DNA as new "distance controllers". In this work, a peptide with three functional sequences is designed to control interparticle distance between positive-charged Au nanoparticles ((+) AuNPs) and negative-charged CdTe quantum dots ((-) CdTe QDs). Relying on the function of binding sequence, (+) AuNPs and (-) CdTe QDs may be separated to a certain distance by the multifunctional peptide. In this case, the excitonic response is relatively weak, and an evident PEC response can be observed. Because it contains the substrate sequence of caspase-3, the peptide is cleaved in the presence of caspase-3. As a result, without the support of intact peptide, electrostatic attraction plays a dominant role, leading to the aggregation of oppositely charged AuNPs and CdTe QDs, which strengthens the excitonic response and attenuates the PEC response. On the basis of these principles, a novel PEC approach was fabricated to sensitively quantify caspase-3. Meanwhile, caspase-3 in staurosporine-treated A549 cells are also determined by the approach, and the obtained results agree well with the fluorescent intensity of confocal images, manifesting that the proposed PEC method can monitor apoptosis in a label-free strategy. Overall, the study reveals the capability of peptides in controlling interparticle distance of nanomaterials, which may accelerate the development of peptide-based PEC analytical methods.

Enzyme cascade sensors usually could not discriminate between the target and intermediate product. Herein, based on "AND" logic-controlled activation of the glucose oxidase-copper peroxide sensing system, enzyme cascade detection for glucose with resistance to inherently existing intermediate product H2O2 was reported for the first time, which may provide a novel way for facilitating enzyme cascade sensing.
