Abstract Primary central nervous system lymphoma (PCNSL) is a rare and frequently fatal lymphoma subtype. The programmed death-1 (PD-1) pathway has emerged as a potential therapeutic target, but the effectiveness of PD-1 antibody sintilimab in combination with immunochemotherapy as a frontline treatment for PCNSL remains to be determined. In this phase 2 trial (ChiCTR1900027433) with a safety run-in, we included patients aged 18–70 with newly diagnosed PCNSL. Participants underwent six 21-day cycles of a SMTR regimen, which includes sintilimab (200 mg, Day 0), rituximab (375 mg/m2, Day 0), methotrexate (3.0 g/m2, Day 1 or 1.0 g/m2 for patients aged ≥65 years), and temozolomide (150 mg/m2/d, Days 1–5). Among 27 evaluable patients, the overall response rate (ORR) was 96.3% (95% confidence interval: 81–99.9%), with 25 complete responses. At a median follow-up of 24.4 months, the medians for duration of response, progression-free survival (PFS), and overall survival were not reached. The most common grade 3–4 treatment-related toxicities were increased levels of alanine aminotransferase (17.9%) and aspartate aminotransferase (14.3%). Additionally, baseline levels of interferon-α and the IL10/IL6 ratio in cerebrospinal fluid emerged as potential predictors of PFS, achieving areas under the curve of 0.88 and 0.84, respectively, at 2 years. Whole-exome sequencing revealed a higher prevalence of RTK-RAS and PI3K pathway mutations in the durable clinical benefit group, while a greater frequency of Notch and Hippo pathway mutations in the no durable benefit group. These findings suggest the SMTR regimen is highly efficacious and tolerable for newly diagnosed PCNSL, warranting further investigation.

Abstract Deep learning techniques were used in ophthalmology to develop artificial intelligence (AI) models for predicting the short-term effectiveness of anti-VEGF therapy in patients with macular edema secondary to branch retinal vein occlusion (BRVO-ME). 180 BRVO-ME patients underwent pre-treatment FFA scans. After 3 months of ranibizumab injections, CMT measurements were taken at baseline and 1-month intervals. Patients were categorized into good and poor prognosis groups based on macular edema at the 4th month follow-up. FFA-Net, a VGG-based classification network, was trained using FFA images from both groups. Class activation heat maps highlighted important locations. Benchmark models (DesNet-201, MobileNet-V3, ResNet-152, MansNet-75) were compared for training results. Performance metrics included accuracy, sensitivity, specificity, F1 score, and ROC curves. FFA-Net predicted BRVO-ME treatment effect with an accuracy of 88.63% and an F1 score of 0.89, with a sensitivity and specificity of 79.40% and 71.34%, respectively.The AUC of the ROC curve for the FFA-Net model was 0.71. The use of FFA based on deep learning technology has feasibility in predicting the treatment effect of BRVO-ME. The FFA-Net model constructed with the VGG model as the main body has good results in predicting the treatment effect of BRVO-ME. The typing of BRVO in FFA may be an important factor affecting the prognosis.

Abstract Objective To assess whether diffusion and perfusion MRI derived parameters could non-invasively predict PD-L1 and Ki-67 status in primary central nervous system diffuse large B-cell lymphoma (PCNS-DLBCL). Methods We retrospectively analyzed DWI, DSC-PWI, and morphological MRI (mMRI) in 88 patients with PCNS-DLBCL. The mMRI features were compared using chi-square tests or Fisher exact test. Minimum ADC (ADCmin), mean ADC(ADCmean), relative minimum ADC (rADCmin), relative mean ADC (rADCmean), and relative maximum CBV (rCBVmax) values were compared in PCNS-DLBCL with different molecular status by using the Mann-Whitney U test. The diagnostic performances were evaluated by receiver operating characteristic curves. Results PCNS-DLBCL with high PD-L1 expression demonstrated a significantly higher ADCmin value than those with low PD-L1. The ADCmean and rADCmean values were significantly lower in PCNS-DLBCL with high Ki-67 status compared with those in low Ki-67 status. Other ADC, CBV parameters, and mMRI features did not show any association with these molecular statuses The diagnostic efficacy of ADC values in assessing PD-L1 and Ki-67 status was relatively low, with area under the curves (AUCs) values less than 0.7. Conclusions DWI-derived ADC values can provide some relevant information about PD-L1 and Ki-67 status in PCNS-DLBCL, but may not be sufficient to predict their expression due to the rather low diagnostic performance.

Abstract The transformative role of artificial intelligence (AI) in various fields highlights the need for it to be both accurate and fair. Biased medical AI systems pose significant potential risks to achieving fair and equitable healthcare. Here, we show an implicit fairness learning approach to build a fairer ophthalmology AI (called FairerOPTH) that mitigates sex (biological attribute) and age biases in AI diagnosis of eye diseases. Specifically, FairerOPTH incorporates the causal relationship between fundus features and eye diseases, which is relatively independent of sensitive attributes such as race, sex, and age. We demonstrate on a large and diverse collected dataset that FairerOPTH significantly outperforms several state-of-the-art approaches in terms of diagnostic accuracy and fairness for 38 eye diseases in ultra-widefield imaging and 16 eye diseases in narrow-angle imaging. This work demonstrates the significant potential of implicit fairness learning in promoting equitable treatment for patients regardless of their sex or age.

PurposeWe aimed to explore the prognostic significance of collagen content in solitary fibrous tumors (SFTs) of the central nervous system (CNS) and preliminarily investigate its relationship with magnetic resonance imaging (MRI) features of SFTs.MethodsCollagen content was identified using Masson’s trichrome staining, and quantitatively assessed. Radiomic methods were applied to extract quantitative MRI features of SFTs, which were then analyzed in relation to collagen content.ResultsThe collagen content in CNS SFTs was categorized into high- and low-content groups, with a cutoff value of 6%. Survival analysis indicated a positive correlation between collagen content and overall survival (OS). In multivariate Cox regression analysis, incorporating factors such as mitosis, necrosis, Ki67, and collagen content and other indicators, collagen content emerged as an independent prognostic factor. Collagen content demonstrated a negative correlation with tumor histological phenotype, Ki67, WHO grade, mitosis, necrosis, and brain invasion. Additionally, the signal intensity of SFTs on T2-weighted imaging (T2WI) decreased with increasing collagen content. Radiomics analysis identified 1,702 features from each patient’s region of interest, with 12 features showing significant differences between the high and low collagen content groups. Among the quantitative parameters and radiomic models, the combined T1- and T2WI models exhibited the highest diagnostic performance.ConclusionThese findings suggest that collagen content is an independent prognostic risk factor for OS. Furthermore, combined radiomic models based on T1-and T2WI sequences may offer a more comprehensive, objective, and accurate assessment of collagen content in CNS SFTs.

Background: This study aimed to evaluate the efficacy of ranibizumab for the treatment of macular edema secondary to branch retinal vein occlusion (BRVO-ME), changes in retinal volume and central retinal thickness (CRT) before and after therapy, and the connection between visual prognosis and changes in retinal volume. Methods: The 120 patients(121 eyes) of BRVO-ME were recruited from July 2020 to October 2022 at the Affiliated Hospital of Weifang Medical University. The clinical data of patients were retrospectively examined for changes in best-corrected visual acuity (BCVA), retinal volume, and CRT at 1 day, 1 week, 1 month, 3 months, 6 months and 1year after treatment. Findings: Visual acuity improved gradually and became steady approximately 1 months after treatment, whereas retinal volume decreased gradually in both the outer and full layers and stabilized around 6 month after treatment. The decline in retinal volume and CRT was more visible in the deeper layers than in the inner levels. A higher correlation was observed between retinal volume and BCVA than between CRT and BCVA. BCVA after one year of treatment had a high correlation with baseline outer retinal volume. Interpretation: Treatment of BRVO-ME with ranibizumab is highly effective, and the recovery of visual function was depends more on early treatment. The outer retina is the major site of edema. Changes in retinal volume may serve as a better predictor of visual prognosis than changes in CRT. Baseline ourter retinal volume is correlated with long-term visual prognosis.

Accurately distinguishing the origin and variety of rice types is of paramount importance to conducting research on this staple crop. While various methods are currently employed for this purpose, few approaches can verify the identity of single grains rapidly and accurately. In this study, we present a method that integrates machine learning with infrared (IR) microspectroscopy for swift detection of the origin and variety of a single rice grain. To establish the validity of our approach, we assembled a diverse collection of rice samples, comprising 14 distinct types with different origins or varieties. Each rice sample yielded 100 microspectroscopy spectra, resulting in 1400 spectra. We applied two deep learning algorithms, deep neural network (DNN) and convolutional neural network (CNN), for spectral analysis. The 1400 spectra were randomly partitioned into calibration and validation sets at a ratio of 3:1. These datasets were subjected to both DNN and CNN analysis for classification of samples by origin and variety. Following 10,000 iterations, we selected optimal DNN and CNN models. The predication accuracies of the optimal DNN model for calibration and validation sets were 95.4% and 90.0%, respectively. In comparison, the optimal CNN model demonstrated superior accuracy, with 99.8% for the calibration set and 92.0% for the validation set. Based on these results, we selected the CNN model as the final model for field use in rice grain classification.

Background: To assess the feasibility of multiparametric magnetic resonance imaging in predicting tumor recurrence in nonenhancing peritumoral regions in patients with glioblastoma at baseline. Methods: Fifty-eight patients with recurrent glioblastoma underwent multiparametric magnetic resonance imaging, including T2-weighted fluid-attenuated inversion recovery, diffusion-weighted imaging, and dynamic susceptibility contrast perfusion-weighted imaging. Nonenhancing peritumoral regions with glioblastoma recurrence were identified by coregistering preoperative and post-recurrent magnetic resonance images. Regions of interest were placed in nonenhancing peritumoral regions with and without tumor recurrence to calculate the apparent diffusion coefficient value, and relative ratios of T2-weighted fluid-attenuated inversion recovery signal intensity, apparent diffusion coefficient, and cerebral blood volume values. Results: Significant lower relative T2-weighted fluid-attenuated inversion recovery signal intensity, apparent diffusion coefficient, and relative apparent diffusion coefficient but higher relative cerebral blood volume values were found in the nonenhancing peritumoral regions with tumor recurrence than without recurrence (all P

Abstract Surgical removal of the thyroid gland (TG) for treating thyroid disorders leaves the patients on lifelong hormone replacement that partially compensates the physiological needs, but regenerating TG is challenging. Here, an approach is reported to regenerate TG within the spleen for fully restoring the thyroid's functions in mice, by transplanting thyroid tissue blocks to the spleen. Within 48 h, the transplanted tissue efficiently revascularizes, forming thyroid follicles similar to the native gland after 4 weeks. Structurally, the ectopically generated thyroid integrates with the surrounding splenic tissue while maintaining its integrity, separate from the lymphatic tissue. Functionally, it fully restores the native functions of the TG in hormone regulation in response to physiological stimuli, outperforming the established method of oral levothyroxine therapy in maintaining systemic homeostasis. The study demonstrates the full restoration of thyroid functions post‐thyroidectomy by intrasplenic TG regeneration, providing fresh insights for designing novel therapies for thyroid‐related disorders.

PurposeThis bi-institutional study aimed to establish a robust model for predicting clinically significant prostate cancer (csPCa) (pathological grade group ≥ 2) in PI-RADS 3 lesions in the transition zone by comparing the performance of combination models.Materials and methodsThis study included 243 consecutive men who underwent 3-Tesla magnetic resonance imaging (MRI) and ultrasound-guided transrectal biopsy from January 2020 and April 2022 which is divided into a training cohort of 170 patients and a separate testing cohort of 73 patients. T2WI and DWI images were manually segmented for PI-RADS 3 lesions for the mean ADC and radiomic analysis. Predictive clinical factors were identified using both univariate and multivariate logistic models. The least absolute shrinkage and selection operator (LASSO) regression models were deployed for feature selection and for constructing radiomic signatures. We developed nine models utilizing clinical factors, radiological features, and radiomics, leveraging logistic and XGboost methods. The performances of these models was subsequently compared using Receiver Operating Characteristic (ROC) analysis and the Delong test.ResultsOut of the 243 participants with a median age of 70 years, 30 were diagnosed with csPCa, leaving 213 without a csPCa diagnosis. Prostate-specific antigen density (PSAD) stood out as the only significant clinical factor (odds ratio [OR], 1.068; 95% confidence interval [CI], 1.029–1.115), discovered through the univariate and multivariate logistic models. Seven radiomic features correlated with csPCa prediction. Notably, the XGboost model outperformed eight other models (AUC of the training cohort: 0.949, and validation cohort: 0.913). However, it did not surpass the PSAD+MADC model (P > 0.05) in the training and testing cohorts (AUC, 0.949 vs. 0.888 and 0.913 vs. 0.854, respectively).ConclusionThe machine learning XGboost model presented the best performance in predicting csPCa in PI-RADS 3 lesions within the transitional zone. However, the addition of radiomic classifiers did not display any significant enhancement over the compound model of clinical and radiological findings. The most exemplary and generalized option for quantitative prostate evaluation was Mean ADC+PSAD.

The effects of different enclosure years on soil physicochemical properties provide scientific basis for soil fertility evaluation and ecological restoration. However, the details on the effects of long-term fencing measures on soil physicochemical properties in alpine regions are still lacking. The soil physicochemical properties, soil nutrient content, and stoichiometric characteristics of desert grassland were examined in the desert steppe of northern Tibetan Plateau, where grazing exclusion using fences has been implemented. This study included conducting experiments on different soil layers during different duration (10, 11, and 12 years) of grazing exclusion using fences; natural plots with no grazing exclusion were used as a control variable. Results showed that several soil physicochemical indexes, such as soil organic matter, soil water content, soil capillary porosity, soil non-capillary porosity, and soil capillary water capacity, in plots under the fencing measures were higher than those in the natural plots. The soil bulk density of the 12- and 11-year grazing exclusion plots was lower than that of the natural plots. The clay content of the 10- and 12-year grazing exclusion plots was higher than that of the natural plots, and the sand content was lower than that of the natural plots. The soil nutrient content was especially low with less than the average level of soil nutrient content in China. The stoichiometric C/N, C/P, N/P, and C/K ratios were 10.28–16.39, 2.74–16.69, 0.24–1.53, and 0.27–0.82, respectively. This result indicates that the degree of organic carbon mineralization was relatively high, and there existed a certain N limitation in the study area. The implementation of enclosure is conducive to the accumulation of C and N. The results provide support for elucidating the physicochemical characteristics, soil nutrients, and nutrient cycling of desert steppe in the northern Tibetan Plateau and for the study of the allotted time of fencing project in the alpine region.

Abstract Nonhealing diabetic wounds, with persistent inflammation and damaged vasculature, have failed conventional treatments and require comprehensive interference. Here, inspired by tumor‐associated macrophages (TAMs) that produce abundant immunosuppressive and proliferative factors in tumor development, we generate macrophages to recapitulate TAMs' reparative functions, by culturing normal macrophages with TAMs' conditional medium (TAMs‐CM). These TAMs‐educated macrophages (TAMEMs) outperform major macrophage phenotypes (M0, M1, or M2) in suppressing inflammation, stimulating angiogenesis, and activating fibroblasts in vitro. When delivered to skin wounds in diabetic mice, TAMEMs efficiently promote healing. Based on TAMs‐CM's composition, we further reconstitute a nine‐factor cocktail to train human primary monocytes into TAMEMsC‐h, which fully resemble TAMEMs' functions without using tumor components, thereby having increased safety and enabling the preparation of autologous cells. Our study demonstrates that recapitulating TAMs' unique reparative activities in nontumor cells can lead to an effective cell therapeutic approach with high translational potential for regenerative medicine.

Abstract The formation of biomolecular condensates via liquid‒liquid phase separation (LLPS) is an advantageous strategy for cells to organize their subcellular compartments for diverse functions. Recent findings suggest that RNA or RNA-related LLPS techniques have potential for the development of new cellular regulation strategies. However, manipulating RNA LLPS in living cells has great challenges. Herein, we report that cationic polymers (CPs) have strong RNA LLPS-inducing activity. By introducing CPs into living cells or RNA solutions, significant RNA LLPS was verified through confocal imaging, turbidity assays, and fluorescence recovery after photobleaching (FRAP) tests. Among them, turbidity kinetics determinations indicated that the hydrophilic positively charged amino groups on the CPs play essential roles in RNA phase separation. Moreover, the LLPS induced by the cationic polymers dramatically changed the gene expression patterns in the cells. Interestingly, we found that TGFβ1 mRNA was highly encapsulated in the RNA droplets, which lowered the immunosuppressive capability of the tumor cells and triggered marked antitumor reactions in a mouse breast cancer model. Thus, we present here the CP-based modulation of RNA LLPS as a novel transcriptional manipulation method with potential for cancer immunotherapy drug development. Graphical Abstract

Understanding vegetation C, N, and P stoichiometry helps us not only to evaluate biogeochemical cycles and ecosystem functions but also to predict the potential impact of environmental change on ecosystem processes. The foliar C, N, and P stoichiometry in Northern Tibetan grasslands, especially the controlling factors, has been highlighted in recent years. In this study, we have collected 340 plant samples and 162 soil samples from 54 plots in three grassland types, with the purpose of evaluating the foliar C, N, and P stoichiometry and underlying control factors in three grassland types along a 1,500-km east-to-west transect in the Northern Tibetan Plateau. Our results indicated that the averaged foliar C, N, and P concentrations were 425.9 ± 15.8, 403.4 ± 22.2, and 420.7 ± 30.7 g kg−1; 21.7 ± 2.9, 19.0 ± 2.3, and 21.7 ± 5.2 g kg−1; and 1.71 ± 0.29, 1.19 ± 0.16, and 1.59 ± 0.6 g kg−1 in the alpine meadow (AM), alpine steppe (AS), and desert steppe (DS) ecosystems, respectively. The foliar C and N ratios were comparable, with values of 19.8 ± 2.8, 20.6 ± 1.9, and 19.9 ± 5.8 in the AM, AS, and DS ecosystems, respectively. Both the C/P and N/P ratios are the lowest in the AM ecosystem, with values of 252.2 ± 32.6 and 12.8 ± 1.3, respectively, whereas the highest values of 347.3 ± 57.0 and 16.2 ± 3.2 were obtained in the AS ecosystem. In contrast, the soil C, N, C/P, and N/P values decreased from the AM to DS ecosystem. Across the whole transects, leaf C, N, and P stoichiometry showed no obvious trend, but soil C and N concentrations showed an increasing trend, and soil P concentrations showed a decreasing trend with the increasing longitude. Based on the general linear model analysis, the vegetation type was the dominant factor controlling the leaf C, N, and P stoichiometry, accounting for 42.8% for leaf C, 45.1% for leaf N, 35.2% for leaf P, 52.9% for leaf C/N, 39.6% for leaf C/P, and 48.0% for leaf N/P; the soil nutrients and climate have relatively low importance. In conclusion, our results supported that vegetation type, rather than climatic variation and soil nutrients, are the major determinants of north Tibet grassland leaf stoichiometry.

The vertical distribution understanding of undergrowth herbaceous plants in high altitude mountain areas is of great significance to reveal the environmental gradient change law of plant diversity. However, the species composition of herbaceous plants at different altitude gradients and the relationship between diversity and environmental factors is still vague. This study takes the Sejila Mountain in southeast Tibet at an altitude of 2,000–3,300 m as the study area to analyze the vertical distribution differences of the understory herbs and their driving factors on shady and sunny slopes. The results showed that: (1) The number of families, genera and species of herbaceous plants on sunny slope is greater than that on shady slope. Asteraceae on shady and sunny slopes has the largest number of species; (2) The α diversity index of the herbs on shady slope shows a U-shaped trend. The Cody heterogeneity index (βC) is negatively correlated with the β diversity index. The herb richness index (R) on sunny slope is unimodal. The β diversity index displays the same as that of shady slope; (3) Different environmental factors have different effects on herb diversity. The correlation between environmental factors and diversity indicators indicates that there is a certain degree of clustering among samples. The environmental factors of shady slope and sunny slope are separated; and (4) The difference of water and heat conditions caused by altitude and slope aspect is an important driving factor for the diversity of herbage species, followed by soil clay content, silt content and slope gradient. This study will help to understand the species diversity and ecosystem characteristics of the region, and provide a reference for the study of plant community structure and function and rational utilization of understory plant resources in high-altitude mountain areas.

Abstract Background The accurate grading of IDH-mutant astrocytoma is essential to make therapeutic strategies and assess the prognosis of patients. The purpose of this study was to investigate the usefulness of DWI, SWI and DSC-PWI in grading IDH-mutant astrocytoma. Methods One hundred and seven patients with IDH-mutant astrocytoma who underwent DWI, SWI and DSC-PWI were retrospectively reviewed. Minimum apparent diffusion coefficient (ADCmin), intratumoral susceptibility signal intensity(ITSS) and maximum relative cerebral blood volume (rCBVmax) values were assessed. ADCmin, ITSS and rCBVmax values were compared between grade 2 vs. grade 3, grade 3 vs. grade 4 and grade 2 + 3 vs. grade 4 tumors. Logistic regression, tenfold cross-validation,and receiver operating characteristic (ROC) curve analyses were used to assess their diagnostic performances. Abstract Background Pathological alterations of lateral pterygoid muscle (LPM) are implicated in temporomandibular joint anterior disk displacement (ADD). However, quantification of the fatty infiltration of LPM and its correlation with ADD have rarely been reported. The aim of this study was to evaluate the fatty infiltration, morphological features and texture features of LPM in patients with ADD using T1-weighted Dixon sequence. Methods This retrospective study included patients who underwent temporomandibular joint MRI with T1-weighted Dixon sequence between December 2018 and August 2020. The temporomandibular joints of the included patients were divided into three groups according to the position of disk: Normal position disk (NP) group, Anterior disk displacement with reduction (ADDWR) group and Anterior disk displacement without reduction (ADDWOR) group. Fat fraction, morphological features (Length; Width; Thickness), and texture features (Angular second moment; Contrast; Correlation; Inverse different moment; Entropy) extracted from in-phase image of LPM were evaluated. One-way ANOVA, Welch’s ANOVA, Kruskal–Wallis test, Spearman and Pearson correlation analysis were performed. Intra-class correlation coefficient was used to evaluate the reproducibility. Results A total of 53 patients with 106 temporomandibular joints were evaluated. Anterior disk displacement without reduction group showed higher fat fraction than normal position disk group (P = 0.024). Background Chloroplast (cp) genomes are useful and informative molecular markers used for species determination and phylogenetic analysis. Bulbophyllum is one of the most taxonomically complex taxa in Orchidaceae. However, the genome characteristics of Bulbophyllum are poorly understood. Methods Based on comparative morphological and genomic analysis, a new species Bulbophyllum pilopetalum from eastern Himalaya belonging to section Cirrhopetalum is described and illustrated. This study used chloroplast genomic sequences and ribosomal DNA (nrDNA) analysis to distinguish the new Bulbophyllum species and determine its phylogenetic position. An additional phylogenetic analysis was conducted using 74 coding sequences from 15 complete chloroplast genomes from the genus Bulbophyllum, as well as nrDNA sequences and two chloroplast DNA sequences from 33 Bulbophyllun species. Results The new species is morphologically similar to B. pingnanense, B. albociliatum, and B. brevipedunculatum in vegetative and floral morphology, but it can be distinguished by its ovate-triangle dorsal sepal without a marginal ciliate. The chloroplast genome of the new Bulbophyllum species is 151,148 bp in length, and includes a pair of inverted repeats (IRs) of 25,833 bp, a large single-copy region (LSC) of 86,138 bp, and a small single-copy region (SSC) of 13,300 bp. The chloroplast genome includes 108 unique genes encoding 75 proteins, 30 tRNAs, and four rRNAs. Compared with the cp genomes of its two most closely-related species, B. pingnanense and B. albociliatum, this chloroplast genome exhibited great interspecific divergence and contained several Indels that were specific to the new species. The plastid tree showed that B. pilopetalum is most closely-related to B. pingnanense. The phylogenetic tree based on combined nrDNA and chloroplast DNA sequences indicated that section Cirrhopetalum was monophyletic and B. pilopetalum was a member of this section. Discussion The taxonomic status of the new species is strongly supported by cp genome data. Our study highlights the importance of using the complete cp genome to identify species, elucidate the taxonomy, and reconstruct the phylogeny of plant groups with complicated taxonomic problems.

Intestinal autochthonous bacteria play important roles in the maintenance of the physiological homeostasis of animals, especially contributing to the host immune system. In the present study, the variation of autochthonous bacterial community in the intestinal tract of 2-7 months-old tiger pufferfish Takifugu rubripes and bacterial communities in the seawater of recirculating aquaculture system (RAS) and the following offshore sea cage aquaculture system (OSCS) were analyzed during the aquaculture period from May to October 2021. Proteobacteria was found to be the most dominant phyla in both intestinal and seawater bacterial communities, which accounted for 68.82% and 65.65% of the total bacterial abundance, respectively. Arcobacter was the most core bacterial taxon in the intestinal bacterial community, with the most dominant abundance (42.89%) at the genus level and dominant positions in co-occurrence relationships with other bacterial taxa (node-betweenness value of 150). Enterococcaceae was specifically enriched in the intestinal bacterial community of pufferfishes from RAS, while Vibrionaceae was enriched in the intestinal bacterial community from OSCS. The F-values of beta diversity analysis between intestinal and seawater bacterial communities generally increased from May (6.69) to October (32.32), indicating the increasing differences between the intestinal and seawater bacterial communities along with the aquaculture process. Four bacterial taxa of Weissella sp., Akkermansia muciniphila, Dietzia sp. and Psychrobacter pacificensis had significant correlations with immune response parameters, and they were suggested to be the indicators for immune status and pathological process of pufferfish. The knowledge about the specific core bacteria, potentially pathogenic bacteria and the change of bacterial community in the intestinal tract of cultured pufferfish is of great scientific significance and will contribute to the understanding of intestinal bacterial homeostasis and biosecurity practice in pufferfish aquaculture.

Transient receptor potential vanilloid 4 (TRPV4) is one of the major non-selective cation channel proteins, which plays a crucial role in sensing biotic and abiotic stresses, such as pathogen infection, temperature, mechanical pressure and osmotic pressure changes by regulating Ca2+ homeostasis. In the present study, a TRPV4 homologue was identified in Pacific oyster Crassostrea gigas, designated as CgTRPV4. The open reading frame (ORF) of CgTRPV4 was of 2298 bp encoding a putative polypeptide of 765 amino acid residues with three typical ankyrin domains and six conserved transmembrane domains of TRPV4 subfamily proteins, as well as multiple N-glycosylation sites, cAMP- and cGMP-dependent protein kinase phosphorylation sites, protein kinase C phosphorylation sites, casein kinase II phosphorylation sites, and prokaryotic membrane lipoprotein lipid attachment site. The deduced amino acid sequence of CgTRPV4 shared 20.5%-26.2% similarity with TRPV4s from other species. During the early ontogenesis stages of oyster, the mRNA transcripts of CgTRPV4 were detectable in all the stages with the highest expression level in fertilized eggs and the lowest in D-hinged larvae. In adult oyster, the CgTRPV4 mRNA could be detected in all the examined tissues, including gill, hepatopancreas, adductor muscle, labial palp, mantle and haemocyte, with the highest expression level in gill (45.08-fold of that in hepatopancreas, p < 0.05). In immunocytochemical assay, the CgTRPV4 positive signals were distributed in both endoplasmic reticulum and cytoplasmic membrane of oyster haemocytes. The mRNA expression of CgTRPV4 in gill was significantly up-regulated after high temperature stress at 30°C (p < 0.05) and after Vibrio splendidus stimulation (p < 0.05). These results indicated that CgTRPV4 was a classical member of TRPV4 family in oyster, which was induced by either biotic or abiotic stimulations and involved in mediating the stress response of oysters.

Soil quality evaluation is the foundation for understanding the current situation of soil quality and promoting the sustainable utilization of land resources. However, the evaluation of the impacts of the different long-term afforestation timescales on the soil quality in a typical watershed valley area is still insufficient. Here, based on the soil samples collected from different afforestation years of 2, 5, 11, 15, and 31 years and natural sample plots with similar site conditions in the same region, the gray target model and criteria importance through intercriteria correlation (CRITIC) method are used in this study to evaluate the soil quality of the sample plots with different afforestation years and the corresponding natural sample plots in the Tibet Plateau. Results show that: (1) The most positive soil physical and chemical indexes increase with the increase in afforestation years based on the analysis of the characteristics, variation, and fertility grade of 13 soil physical and chemical indexes in different areas. The content of most soil physical and chemical indexes is greater than that in the corresponding natural sample plots in 31 years of afforestation. (2) The bullseye degree of the soil quality in the study area is divided into five grades according to the principle of equal division and integration. The soil quality grade of the corresponding sample plot after 15 years of afforestation is grade 4. The other plots are grade 5. (3) The soil quality index (SQI) generally increases with the increase of in forestation years. The different afforestation years indicate SQI (0.709) of 15 years > SQI (0.662) of 31 years > SQI (0.426) of 11 years > SQI (0.348) of 2 years > SQI (0.309) of 5 years. (4) The full data set method verifies the effectiveness of the gray target model. The two soil quality evaluation methodologies are consistent and can verify each other’s accuracy. The correlation coefficient of the two results reaches 0.957. This study finds that the long-term afforestation timescales have improved the soil quality of watershed valley. Available phosphorus, silt, soil water content are the main factors impacting the soil quality in the afforestation timescales.

A non-motile, Gram-staining-negative, orange-pigmented bacterium called herbae pc1-10T was discovered in Tibet in the soil around Pyrola calliantha H. Andres’ roots. The isolate thrived in the temperature range of 10–30 °C (optimal, 25 °C), pH range of 5.0–9.0 (optimum, pH = 6.0), and the NaCl concentration range of 0–1.8% (optimal, 0%). The DNA G+C content of the novel strain was 37.94 mol%. It showed the function of dissolving organophosphorus, acquiring iron from the environment by siderophore and producing indole acetic acid. Moreover, the genome of strain herbae pc1-10T harbors two antibiotic resistance genes (IND-4 and AdeF) encoding a β-lactamase, and the membrane fusion protein of the multidrug efflux complex AdeFGH; antibiotic-resistance-related proteins were detected using the Shotgun proteomics technology. The OrthoANIu values between strains Chryseobacterium herbae pc1-10T; Chryseobacterium oleae CT348T; Chryseobacterium kwangjuense KJ1R5T; and Chryseobacterium vrystaatense R-23566T were 90.94%, 82.96%, and 85.19%, respectively. The in silico DDH values between strains herbae pc1-10T; C. oleae CT348T; C. kwangjuense KJ1R5T; and C. vrystaatense R-23566T were 41.7%, 26.6%, and 29.7%, respectively. Chryseobacterium oleae, Chryseobacterium vrystaatense, and Chryseobacterium kwangjuense, which had 16S rRNA gene sequence similarity scores of 97.80%, 97.52%, and 96.75%, respectively, were its closest phylogenetic relatives. Chryseobacterium herbae sp. nov. is proposed as the designation for the strain herbae pc1-10T (=GDMCC 1.3255 = JCM 35711), which represented a type species based on genotypic and morphological characteristics. This study provides deep knowledge of a Chryseobacterium herbae characteristic description and urges the need for further genomic studies on microorganisms living in alpine ecosystems, especially around medicinal plants.

High-performance donor-acceptor (D-A) polymers, as an important class of electrochromic (EC) materials, have attracted extensive attention. In this paper, a series of novel poly (aryl amino ketone) (PAAK) and poly (aryl amino sulfone) (PAAS) type high-performance polymers (HPP) with electrochromism were prepared by a simple C-N coupling reaction and were coated on an indium tin oxide (ITO) substrate as EC films. All four polymers were prepared by a nucleophilic substitution reaction using commercially purchased amine monomers with difluoride sulfone/ketone using potassium carbonate as a catalyst. A series of tests were performed to compare and analyze the effects of the different electron-withdrawing abilities of sulfone and carbonyl groups, and the different conjugation lengths of these two TPA structures were connected to the EC properties of the polymer. The different phenyl or biphenyl of the two TPA structures mainly affected the oxidation potential of the polymer, while the sulfone group and the carbonyl group, with a different electron absorption ability, had a greater influence on the energy band and cyclic stability. The optical contrast of PAAS−BT at 850 nm was up to 58% and maintained 450 cycles, indicating that this series of materials had a broad application prospect waiting for further research. In addition to the performance, the raw materials used in this work could be directly and commercially purchased for a low price; the two aniline monomers were priced at about $0.43 /g and $0.15 /g, respectively. This method significantly reduces the cost and provides a new idea for subsequent large-scale production and practical applications.

Abstract Background Tibet Orbivirus (TIBOV) is a recently discovered Orbivirus known to infect cattle, Asian buffalo and goats in south-western China. It was first isolated from mosquitoes and subsequently from biting midges (Culicoides spp.) in Yunnan, China, indicating that it is an arbovirus. Little is known of its potential to cause disease, but the economic importance of related viruses promoted an investigation of potential Culicoides spp. vectors of TIBOV. Methods Biting midges were collected approximately once per week between May and December 2020, at a cattle farm in Wulong village, Shizong County, Yunnan Province, China. Approximately 3000 specimens of nine species were subsequently used in attempts to isolate virus, and a further 2000 specimens of six species were tested for the presence of bluetongue virus (BTV) and TIBOV using a RT-qPCR test. Results Virus isolation attempts resulted in the isolation of three viruses. One isolate from a pool of Culicoides jacobsoni was identified as TIBOV, while the other two viruses from C. orientalis and C. tainanus remain unidentified but are not BTV or TIBOV. RT-qPCR analysis did not detect BTV in any specimens, but a single pool containing five specimens of C. jacobsoni and another containing five specimens of C. tainanus produced PCR quantification cycle (Cq) values of around 28 that may indicate infection with TIBOV. Conclusions The isolation of TIBOV from C. jacobsoni satisfies one criterion required to prove its status as a vector of this virus. This isolation is supported by a low Cq value produced from a different pool of this species in the RT-qPCR test. The low Cq value obtained from a pool of C. tainanus suggests that this species may also be able to satisfy this criterion. Both of these species are widespread throughout Asia, with C. jacobsoni extending into the Pacific region, which raises the possibility that TIBOV may be more widespread than is currently known. Abstract Background Exercise training is well established as the most effective way to enhance muscle performance and muscle building. The composition of skeletal muscle fiber type affects systemic energy expenditures, and perturbations in metabolic homeostasis contribute to the onset of obesity and other metabolic dysfunctions. Long noncoding RNAs (lncRNAs) have been demonstrated to play critical roles in diverse cellular processes and diseases, including human cancers; however, the functional importance of lncRNAs in muscle performance, energy balance, and obesity remains elusive. We previously reported that the lncRNA H19 regulates the poly-ubiquitination and protein stability of dystrophin (DMD) in muscular dystrophy. Methods Here, we identified mouse/human H19-interacting proteins using mouse/human skeletal muscle tissues and liquid chromatography–mass spectrometry (LC-MS). Human induced pluripotent stem-derived skeletal muscle cells (iPSC-SkMC) from a healthy donor and Becker Muscular Dystrophy (BMD) patients were utilized to study DMD post-translational modifications and associated proteins. We identified a gain-of-function (GOF) mutant of H19 and characterized the effects on myoblast differentiation and fusion to myotubes using iPSCs. We then conjugated H19 RNA gain-of-function oligonucleotides (Rgof) with the skeletal muscle enrichment peptide agrin (referred to as AGR-H19-Rgof) and evaluated AGR-H19-Rgof’s effects on skeletal muscle performance using wild-type (WT) C57BL/6 J mice and its anti-obesity effects using high-fat diet (HFD)- and leptin deficiency-induced obese mouse models. Results We demonstrated that both human and mouse H19 associated with DMD and that the H19 GOF exhibited enhanced interaction with DMD compared to WT H19. DMD was found to associate with serine/threonine-protein kinase MRCK alpha (MRCKα) and α-synuclein (SNCA) in iPSC-SkMC derived from BMD patients. Inhibition of MRCKα and SNCA-mediated phosphorylation of DMD antagonized the interaction between H19 and DMD. These signaling events led to improved skeletal muscle cell differentiation and myotube fusion. The administration of AGR-H19-Rgof improved the muscle mass, muscle performance, and base metabolic rate of WT mice. Furthermore, mice treated with AGR-H19-Rgof exhibited resistance to HFD- or leptin deficiency-induced obesity. Conclusions Our study suggested the functional importance of the H19 GOF mutant in enhancing muscle performance and anti-obesity effects.

Abstract Background Culicoides (Diptera: Ceratopogonidae) are vectors for many arboviruses. At least 20 species are considered as vectors or potential vectors of bluetongue virus (BTV) which cause bluetongue disease in ruminants. A BTV prevalence of 30–50% among cattle and goats in tropical southern Yunnan Province, China, prompted an investigation of the potential BTV vectors in this area. Methods Culicoides were collected by light trapping at three sites in the tropical region of Yunnan Province. Species were identified based on morphology and DNA sequences of cytochrome c oxidase subunit 1 (cox1). PCR and quantitative PCR following reverse transcription were used to test for the presence of BTV RNA in these specimens. Phylogenetic analysis was used to analyze the cox1 sequences of Culicoides specimens infected with BTV. Results Approximately 67,000 specimens of Culicoides were collected, of which 748 were tested for the presence of BTV. Five specimens, including two of Culicoides jacobsoni, one of C. tainanus and two of C. imicola, were identified as infected with BTV. No specimens of C. (subgenus Trithecoides) or C. oxystoma tested were positive for BTV infection. Conclusions To our knowledge this is the first report of C. jacobsoni as a potential BTV vector and the fourth report of an association between C. tainanus and BTV, as well as the first direct evidence of an association between BTV and C. imicola in Asia. A fourth potential cryptic species within C. tainanus was identified in this study. Further analysis is required to confirm the importance of C. jacobsoni and C. tainanus in BTV epidemiology in Asia.

Abstract Background Rectal douching (RD) is widely practiced by men who have sex with men (MSM), and is associated with increased risk of HIV infection. However, the mechanism of how RD increases the risk of HIV infection is not well understood, and there is limited data on RD behavior in MSM practicing anal sex in China. We examine the purpose of RD, its timing in relation to anal sex, the types of RD products used, and risky sexual behaviors among MSM reporting anal sex. Methods Between August 2017 and December 2018, a cross-sectional study was conducted among adult MSM in Shenyang, China. Data were collected on demographics, sexual behaviors, and RD for the most recent sexual intercourse by means of interviewer-administered face-to-face questionnaires. Blood samples were collected to test for antibodies to HIV and syphilis. Multivariable logistic regression models were used to assess the risk factors associated with HIV infection. Results A total of 515 eligible MSM participated in this survey (median age: 31 years). During the most recent anal intercourse, 28.3% (146/515) had condomless receptive anal intercourse (CRAI), 21.4% (110/515) practiced serosorting, and more than half (61.6%, 317/515) reported RD before or after anal sex. Of those practicing RD, 96.8% (307/317) conducted RD before sex, while 62.5% (198/317) conducted RD after sex. The douching devices used were primarily shower hoses (85.3%, 262/307), and relatively few MSM used commercial RD products (8.1%, 25/307) before sex. The prevalence of HIV-1 and syphilis was 11.7% and 13.2%, respectively. HIV infection was positively associated with RD, practicing RD before sex, the interaction between RD and CRAI using a shower hose for RD and other risk factors, practicing RD after sex, CRAI, using nitrite inhalants, main sexual role with males as bottom and syphilis infection. Conclusions RD is popular among Chinese MSM. Improper noncommercial RD tools use (such as shower hose), the interaction effect between RD and CRAI associated with HIV infection. Public health workers and the MSM community should publicize scientific knowledge and prevention approaches relating to RD and HIV transmission to MSM. We recommend that further studies should be conducted to understand the detailed mechanism between RD and increased HIV prevalence.

ObjectiveTo investigate the clinicopathological characteristics, molecular genetic characteristics and prognosis of extraventricular neurocytoma located in the sellar/suprasellar region.MethodsSeven archived tumor samples derived from 4 patients with neurocytoma in the sellar/suprasellar region were collected from the First Affiliated Hospital of Fujian Medical University and the Affiliated Hospital of Qingdao University and retrospectively analyzed for clinical manifestations, imaging features, and histopathological features. Neuronal and pituitary biomarkers and molecular features were detected in these tumor tissues by immunohistochemistry and FISH or Sanger sequencing. The related literature was reviewed.ResultsThree patients were female, while 1 was male, with an average age of 35.5 years (range: 27 to 45 years). The initial manifestations were mainly headache and blurred vision in both eyes. The first MRI examination showed marginally enhancing masses in the intrasellar or intra- to suprasellar region. The diagnosis of pituitary adenomas was based on imaging features. The levels of pituitary hormones were normal. Histologically, the tumor cells were arranged in a sheet-like, monotonous architecture and were uniform in size and shape with round to oval, exquisite and hyperchromatic nuclei, which densely packed close to one another and were separated only by a delicate neuropil background. There was no evident mitosis, necrosis or microvascular proliferation. The three cases of recurrent tumors were highly cellular and showed increased mitotic activity. Immunohistochemically, the tumor cells were positive for syn, CR, CgA, and vasopressin and were focally positive for NeuN, TTF-1, NF, CK8, vimentin, and S100 proteins. Other markers, including IDH1, BRAF VE1, Olig-2, and EMA, were negative. Pituitary transcription factors and anterior pituitary hormones were negative. Molecular genetic testing showed that the tumor cells lacked IDH gene mutations, LOH of 1p/19q, MYCN amplification, and EGFR alteration. With a median follow-up of 74.5 months (range 23 to 137 months), 3 patients relapsed at 11, 50, and 118 months after the initial surgery.ConclusionThe morphological features and immunophenotypes of neurocytoma in the sellar/suprasellar region are similar to those of classic central neurocytoma. The prognosis is relatively good. Gross-subtotal resection and atypical subtype may be related to tumor recurrence.

Although fine particulate matter (FPM) in air pollutants and tobacco smoke is recognized as a strong carcinogen and global threat to public health, its biological mechanism for inducing lung cancer remains unclear. Here, by investigating FPM’s bioactivities in lung carcinoma mice models, we discover that these particles promote lung tumor progression by inducing aberrant thickening of tissue matrix and hampering migration of antitumor immunocytes. Upon inhalation into lung tissue, these FPM particles abundantly adsorb peroxidasin (PXDN) – an enzyme mediating type IV collagen (Col IV) crosslinking – onto their surface. The adsorbed PXDN exerts abnormally high activity to crosslink Col IV via increasing the formation of sulfilimine bonds at the NC1 domain, leading to an overly dense matrix in the lung tissue. This disordered structure decreases the mobility of cytotoxic CD8+ T lymphocytes into the lung and consequently impairs the local immune surveillance, enabling the flourishing of nascent tumor cells. Meanwhile, inhibiting the activity of PXDN abolishes the tumor-promoting effect of FPM, indicating the key impact of aberrant PXDN activity on the tumorigenic process. In summary, our finding elucidates a new mechanism for FPM-induced lung tumorigenesis and identifies PXDN as a potential target for treatment or prevention of the FPM-relevant biological risks.

ObjectivesThe performance of multiparametric MRI-based radiomics models for predicting H3 K27M mutant status in diffuse midline glioma (DMG) has not been thoroughly evaluated. The optimal combination of multiparametric MRI and machine learning techniques remains undetermined. We compared the performance of various radiomics models across different MRI sequences and different machine learning techniques.MethodsA total of 102 patients with pathologically confirmed DMG were retrospectively enrolled (27 with H3 K27M-mutant and 75 with H3 K27M wild-type). Radiomics features were extracted from eight sequences, and 18 feature sets were conducted by independent combination. There were three feature matrix normalization algorithms, two dimensionality-reduction methods, four feature selectors, and seven classifiers, consisting of 168 machine learning pipelines. Radiomics models were established across different feature sets and machine learning pipelines. The performance of models was evaluated using receiver operating characteristic curves with area under the curve (AUC) and compared with DeLong’s test.ResultsThe multiparametric MRI-based radiomics models could accurately predict the H3 K27M mutant status in DMG (highest AUC: 0.807–0.969, for different sequences or sequence combinations). However, the results varied significantly between different machine learning techniques. When suitable machine learning techniques were used, the conventional MRI-based radiomics models shared similar performance to advanced MRI-based models (highest AUC: 0.875–0.915 vs. 0.807–0.926; DeLong’s test, p > 0.05). Most models had a better performance when generated with a combination of MRI sequences. The optimal model in the present study used a combination of all sequences (AUC = 0.969).ConclusionsThe multiparametric MRI-based radiomics models could be useful for predicting H3 K27M mutant status in DMG, but the performance varied across different sequences and machine learning techniques.

Abstract Background Diffusion and perfusion MRI can invasively define physical properties and angiogenic features of tumors, and guide the individual treatment. The purpose of this study was to investigate whether the diffusion and perfusion MRI parameters of primary central nervous system lymphomas (PCNSLs) are related to the tumor locations. Methods We retrospectively reviewed the diffusion, perfusion, and conventional MRI of 68 patients with PCNSLs at different locations (group 1: cortical gray matter, group 2: white matter, group 3: deep gray matter). Relative maximum cerebral blood volume (rCBVmax) from perfusion MRI, minimum apparent diffusion coefficients (ADCmin) from DWI of each group were calculated and compared by one-way ANOVA test. In addition, we compared the mean apparent diffusion coefficients (ADCmean) in three different regions of control group. Results The rCBVmax of PCNSLs yielded the lowest value in the white matter group, and the highest value in the cortical gray matter group (P

The echo signal of light detection and ranging (LiDAR) system is easily disturbed by various noises under the influence of intense background light. The signal to noise ratio (SNR) decreases with increasing distance, which seriously affects the retrieval accuracy of the LiDAR system. This paper combines local mean decomposition and an improved thresholding method (LMD-ITM) to process the Lidar signal with a lot of noise, so as to avoid the loss of useful information. In this research, the correlation coefficient and energy entropy are used to distinguish between the relevant and irrelevant components decomposed by the local mean decomposition. Subsequently, the improved thresholding method is applied to process the irrelevant components. It can extract the effective signals which are difficult to be separated from the high-frequency signals. Finally, all components are reconstructed to get the denoising signal. The experimental results demonstrate that this method can effectively avoid the short-range migration, suppress the long-range noise, and adapt to different weather conditions. Compared with the previous denoising method, the proposed method increases the SNR by about 15%.

In recent years, donor-acceptor (D-A)-type conjugated polymers have been widely used in the field of organic solar cells (OSCs) and electrochromism (EC). Considering the poor solubility of D-A conjugated polymers, the solvents used in material processing and related device preparation are mostly toxic halogenated solvents, which have become the biggest obstacle to the future commercial process of the OSC and EC field. Herein, we designed and synthesized three novel D-A conjugated polymers, PBDT1-DTBF, PBDT2-DTBF, and PBDT3-DTBF, by introducing polar oligo (ethylene glycol) (OEG) side chains of different lengths in the donor unit benzodithiophene (BDT) as side chain modification. Studies on solubility, optics, electrochemical, photovoltaic and electrochromic properties are conducted, and the influence of the introduction of OEG side chains on its basic properties is also discussed. Studies on solubility and electrochromic properties show unusual trends that need further research. However, since PBDT-DTBF-class polymers and acceptor IT-4F failed to form proper morphology under the low-boiling point solvent THF solvent processing, the photovoltaic performance of prepared devices is not ideal. However, films with THF as processing solvent showed relatively desirable electrochromic properties and films cast from THF display higher CE than CB as the solvent. Therefore, this class of polymers has application feasibility for green solvent processing in the OSC and EC fields. The research provides an idea for the design of green solvent-processable polymer solar cell materials in the future and a meaningful exploration of the application of green solvents in the field of electrochromism.

(1) Background: The structure, function, and community interactions of soil microbial communities of cultivated Meconopsis integrifolia were characterized by studying this alpine flower and traditional endangered Tibetan medicine. (2) Methods: Soil bacteria and fungi were studied based on high-throughput sequencing technology. Bacteria were isolated using culturomics and functionally identified as IAA-producing, organic phosphorus-dissolving, inorganic phosphorus-dissolving, and iron-producing carriers. (3) Results: The dominant bacterial phyla were found to be Proteobacteria and Acidobacteria, and unclassified_Rhizobiales was the most abundant genus. Ascomycota and Mortierellomycota were the dominant fungal phyla. The bacteria were mainly carbon and nitrogen metabolizers, and the fungi were predominantly Saprotroph—Symbiotroph. The identified network was completely dominated by positive correlations, but the fungi were more complex than the bacteria, and the bacterial keystones were unclassified_Caulobacteraceae and Pedobacter. Most of the keystones of fungi belonged to the phyla Ascomycetes and Basidiomycota. The highest number of different species of culturable bacteria belonged to the genus Streptomyces, with three strains producing IAA, 12 strains solubilizing organic phosphorus, one strain solubilizing inorganic phosphorus, and nine strains producing iron carriers. (4) Conclusions: At the cost of reduced ecological stability, microbial communities increase cooperation toward promoting overall metabolic efficiency and enabling their survival in the extreme environment of the Tibetan Plateau. These pioneering results have value for the protection of endangered Meconopsis integrifolia under global warming and the sustainable utilization of its medicinal value.

A novel double-layer all-solid anti-resonant fiber (DL-AAF) is proposed to develop large mode area (LMA) fiber for high-power fiber laser applications. Single-mode property and mode field area scaling are investigated in simulations. Numerical results show the fiber can achieve strong suppression of higher order modes (HOMs) even with a large core size. With a core diameter of 80 μm, the loss ratio defined as the ratio of the lowest loss of HOMs to that of fundamental mode can exceed 3500, and the mode field area reaches 3030 μm2 at the wavelength of 1.064 μm. By further expanding the core size, the mode filed area can be enlarged to 7481 μm2 while the fiber is ensured to be single-mode. This is a significant enhancement in mode field area scale for the current works on LMA AAF.

A novel hybrid hollow-core polarization-maintaining fiber is proposed by combining the photonic bandgap mechanism and anti-resonant effect. High birefringence can be achieved by introducing a pair of negative curvature anti-resonant layers into the air core of a 7-cell HC-PBGF to obtain two-fold rational symmetry. Within the wavelength range from 1.45 μm to 1.65 μm, the birefringence reaches up to the magnitude order of 10-3 and high-order mode suppression ratio, which is defined as the ratio of the lowest loss of high-order modes to the highest loss of fundamental modes, is greater than 100. Hence, the proposed fiber can realize high birefringence and single mode simultaneously in the 200 nm wide wavelength range. Besides, an extended structure by changing the position of the anti-resonant layers also proves the advantages of hybrid structure in achieving high birefringence and single mode operation. The proposed hybrid structure owns great potential for polarization-sensitive applications and provides a new idea to design hollow-core polarization-maintaining fibers with high birefringence and single mode.

Abstract The blood‐brain barrier (BBB) is the most restrictive and complicated barrier that keeps most biomolecules and drugs from the brain. An efficient brain delivery strategy is urgently needed for the treatment of brain diseases. Based on the studies of brain‐targeting extracellular vesicles (EVs), the potential of using small apoptotic bodies (sABs) from brain metastatic cancer cells for brain‐targeting drug delivery is explored. It is found that anti‐TNF‐α antisense oligonucleotide (ASO) combined with cationic konjac glucomannan (cKGM) can be successfully loaded into sABs via a transfection/apoptosis induction process and that the sABs generated by B16F10 cells have an extraordinarily high brain delivery efficiency. Further studies suggest that ASO‐loaded sABs (sCABs) are transcytosed by b. End3 (brain microvascular endothelial cells, BMECs) to penetrate the BBB, which is mediated by CD44v6, and eventually taken up by microglial cells in the brain. In a Parkinson's disease (PD) mouse model, sCABs dramatically ameliorate PD symptoms via the anti‐inflammatory effect of ASO. This study suggests that sABs from brain metastatic cancer cells are excellent carriers for brain‐targeted delivery, as they have not only an extraordinary delivery efficiency but also a much higher scale‐up production potential than other EVs.

AIM: To explore the effects of sevoflurane combined with dexmedetomidine on emergence agitation(EA)in children under went general anesthesia in ophthalmology department. METHODS: Totally 84 children underment elective ophthalmic surgery in the hospital from May to December 2018 were selected as the research objects. They were randomly divided into study group and control group with 42 cases in each group. All patients in haled sevoflurane to maintain general anesthesia. After anesthesia intubation, children in the study group were treated with intravenous injection of 0.5μg/kg of dexmedetomidine in a constant-speed while children in the control group were treated with same volume of normal saline. The monitoring data of heart rate(HR), mean arterial pressure(MAP)and serum stress response indexes such as norepinephrine(NE)and cortisol(Cor)during recovery period were obtained. The anesthesia recovery, EA and occurrence of complications were recorded. RESULTS: There were obvious fluctuations of HR and MAP in the control group while awake, at 1min, 5min and 10min after extubation. There were significant differences at different time points(PP>0.05). HR and MAP were significantly lower than those in control group at each time point(PPP>0.05). Compared with the control group, the extubation time was significantly shorter and score of extubation quality was significantly lower in the study group(Pvs 31%)(PP>0.05). CONCLUSION: Sevoflurane combined with dexmedetomidine can effectively stabilize hemodynamics of children under went general anesthesia in ophthalmology department during recovery period, and reduce stress response, thus effectively reducing the incidence of EA, without affecting anesthesia recovery quality and with high safety. However, excessive sedation should be payed attention to.

The all-optical vector signal processing technology is of great significance in the evolution of optical network due to its low delay and high efficiency.Therefore,a comprehensive introduction to the technical background and application scheme of the current all-optical signal processing was given.The all-optical signal modulation format was firstly introduced,and the basic characteristics and receiving modes of each modulation format were described.Furthermore,the nonlinear optical effects,which were the basis of all-optical signal processing,were classified and introduced.The occurrence conditions,occurrence process,optical phenomena and common nonlinear media were also illustrated.The advantages of all-optical signal processing technology were briefly introduced and three typical research fields of all-optical signal regeneration,all-optical format conversion and all-optical phase quantization were introduced in detail.Finally,the application prospect of future all-optical vector signal processing technology was prospected.

Abstract Background Cupressus gigantea, a rare and endangered tree species with remarkable medicinal value, is endemic to the Tibetan Plateau. Yet, little is known about the underlying genetics of the unique ecological adaptability of this extremely long-lived conifer with a large genome size. Here, we present its first de novo and multi-tissue transcriptome in-depth characterization. Results We performed Illumina paired-end sequencing and RNA libraries assembly derived from terminal buds, male and female strobili, biennial leaves, and cambium tissues taken from adult C. gigantea. In total, large-scale high-quality reads were assembled into 101,092 unigenes, with an average sequence length of 1029 bp, and 6848 unigenes (6.77%) were mapped against the KEGG databases to identify 292 pathways. A core set of 41,373 genes belonging to 2412 orthologous gene families shared between C. gigantea and nine other plants was revealed. In addition, we identified 2515 small to larger-size gene families containing in total 9223 genes specific to C. gigantea, and enriched for gene ontologies relating to biotic interactions. We identified an important terpene synthases gene family expansion with its 121 putative members. Conclusions This study presents the first comprehensive transcriptome characterization of C. gigantea. Our results will facilitate functional genomic studies to support genetic improvement and conservation programs for this endangered conifer.

In this paper, an all-optical quadrature phase shift keying (QPSK) phase regeneration scheme using vector phase sensitive amplification (PSA) is proposed, analyzed, and simulated. Compared with the conventional scalar PSA scheme, the vector superposition of harmonics makes it easy to obtain high gain extinction ratio (GER) with low pump powers. A subsequent phase-preserving amplitude regenerating device is also introduced to achieve full-quadrature regeneration of signal without phase conjugation and frequency conversion. In addition to the theoretical analysis of the implementation principle, a series of simulation results' analysis, including comparison with traditional scheme and regeneration performance assessment, is also included. The results show that undesired harmonics are greatly reduced by nearly 40 dB, and high GERs are available independent of input power. What is more, various levels of phase and amplitude noise can be effectively compressed for 10-Gbd QPSK signal. The regeneration system also reduces the signal bit error rate and gets 2-dB receiver optical signal noise ratio (OSNR) improvements at 13-dB OSNR input, which has potential applications in enhancing transmission distance in the future optical communication network.

ObjectiveTo explore the value of MR-DWI and T1 mapping in predicting radiation-induced soft tissue fibrosis and its correlation with radiation inflammation.Methods① a total of 30 C57BL/6 mice were randomly divided into a control group (Nor group), irradiation group (IR group) and irradiation plus glycyrrhetinic acid group (GA group). The IR group and GA group were treated with 6MV X-rays to irradiate the right hind limbs of mice for 30 Gy in a single shot. MRI examinations were performed before and on the 7th day after irradiation to measure the apparent diffusion coefficient (ADC) value and the longitudinal relaxation time (T1) value of the hind limb muscles of the mice. On the 90th day after irradiation, the hind limb contracture was measured, and the right hind limb muscle was taken for HE staining, masson staining, immunohistochemical staining and Western blot analysis to detect the expression of a-SMA and Fibronectin. ② The other 30 mice were grouped randomly as above. On the 7th day after irradiation, the right hind limbs of the mice were examined by MRI to measure the ADC value and T1 value of the thigh muscles, and then the right hind thigh muscles were immediately sacrificed to detect IL-1β, IL-6, TNF-a and TGF-β1 expression with ELISA.ResultsOn the 7th day after irradiation, the ADC values ​​of right hind thigh muscles of mice in Nor group, IR group and GA group were (1.35 ± 0.11)*10-3mm2/s, (1.48 ± 0.07) *10-3mm2/s and (1.36 ± 0.13)*10-3mm2/s, respectively, by which the differences between the IR group and Nor group (P=0.008) and that between IR group and GA group (P=0.013) were statistically significant; T1 values ​​were (1369.7 ± 62.7)ms, (1483.7 ± 127.7)ms and (1304.1 ± 82.3)ms, respectively, with which the differences in the T1 value between the IR group and Nor group (P=0.012) and between IR group and GA group (P

Summary: Administration of probiotics to regulate the immune system is a potential anti-tumor strategy. However, oral administration of probiotics is ineffective because of the poor inhabitation of exogenous bacteria in host intestines. Here we report that smectite, a type of mineral clay and established anti-diarrhea drug, promotes expansion of probiotics (especially Lactobacillus) in the murine gut and subsequently elicits anti-tumor immune responses. The ion-exchangeable microstructure of smectite preferentially promotes lactic acid bacteria (LABs) to form biofilms on smectite in vitro and in vivo. In mouse models, smectite laden with LAB biofilms (Lactobacillus and Bifidobacterium) inhibits tumor growth (when used alone) and enhances the efficacy of chemotherapy or immunotherapy (when used in combination with either of them) by activating dendritic cells (DCs) via Toll-like receptor 2 (TLR2) signaling. Our findings suggest oral administration of smectite as a promising strategy to enrich probiotics in vivo for cancer immunotherapy.
