Type 3 innate lymphoid cells (ILC3s) are a newly identified group of innate immune cells that participate in the progression of several metabolic diseases by secreting interleukin (IL)-17 and IL-22. These cytokines are associated with hyperuricemia (HUA) severity and development; however, the relationship between ILC3s and HUA remains unclear.To determine the characteristics of circulating ILC3s in patients with HUA.Type 3 innate lymphoid cells and their subsets were detected using flow cytometry in peripheral blood mononuclear cells (PBMCs) of 80 HUA patients and 30 healthy controls (HC). Plasma levels of IL-17A and IL-22 were measured with enzyme-linked immunosorbent assay (ELISA). Clinical data of enrolled subjects were collected from electronic medical records.In patients with HUA, the frequency of circulating ILC3s was elevated and positively correlated with levels of serum uric acid and serum creatinine (Scr). Although there was no significant difference in the plasma concentration of IL-17A between the patients with HUA and healthy controls, positive correlations between plasma IL-17A and the concentration of serum uric acid and frequency of circulating ILC3s were observed in the patients with HUA.In patients with HUA, positive correlations were detected between circulating ILC3 levels, plasma IL-17A and serum uric acid. Therefore, ILC3s and IL-17A may be useful indicators of disease severity, and are potential new therapeutic targets in HUA.

BackgroundThe CD47-SIRPα pathway acts as an important myeloid cell immune checkpoint and targeting the CD47/SIRPα axis represents a promising strategy to promote antitumor immunity. Several CD47-targeting agents show encouraging early activity in clinical trials. However, due to ubiquitous expression of CD47, the antigen sink and hematologic toxicity, such as anemia and thrombocytopenia, are main problems for developing CD47-targeting therapies. Considering the limited expression of SIRPα, targeting SIRPα is an alternative approach to block the CD47-SIRPα pathway, which may result in differential efficacy and safety profiles.MethodsSIRPα-targeting antibody BR105 was generated by hybridoma fusion and following humanization. BR105 was characterized for binding to human SIRPα alleles and blockade of the interaction with CD47. The functional activity was determined in in vitro phagocytosis assays by using human macrophages. The effect of BR105 on human T cell activation was studied using an OKT3-induced T-cell proliferation assay and an allogeneic mixed lymphocyte reaction. Human SIRPα-humanized immunodeficient mice were used in cancer models for evaluating the in vivo antitumor efficacy of BR105. Safety was addressed in a repeat-dose toxicity study in cynomolgus monkeys, and toxicokinetic analysis was further evaluated.ResultsBR105 shows broad binding activity across various SIRPα variants, and potently blocks the interaction of SIRPα and CD47. In vitro functional assays demonstrated that BR105 synergizes with therapeutic antibodies to promote phagocytosis of tumor cells. Moreover, the combination of BR105 and therapeutic antibody significantly inhibits tumor growth in a xenograft tumor model. Although BR105 may slightly bind to SIRPγ, it does not inhibit T cell activation, unlike other non-selective SIRPα-targeting antibody and CD47-targeting agents. Toxicity studies in non-human primates show that BR105 is well tolerated with no treatment-related adverse effects noted.ConclusionsThe novel and differentiated SIRPα-targeting antibody, BR105, was discovered and displays promising antitumor efficacy in vitro and in vivo. BR105 has a favorable safety profile and shows no adverse effects on T cell functionality. These data support further clinical development of BR105, especially as a therapeutic agent to enhance efficacy when used in combination with tumor-targeting antibodies or antibodies that target other immune checkpoints.

We investigate theoretically the carrier transport in a two-dimensional topological insulator of (001) HgTe/CdTe quantum-well heterostructure with inverted band, and find distinct switchable features of the transmission spectra in the topological edge states by designing the double-electric modulation potentials. The transmission spectra exhibit the significant Fabry–Pérot resonances for the double-electric transport system. Furthermore, the transmission properties show rich behaviors when the Fermi energy lies in the different locations in the energy spectrum and the double-electric barrier regions. The opacity and transparency of the double-modulated barrier regions can be controlled by tuning the modulated potentials, Fermi energy and the length of modulated regions. This electrical switching behavior can be realized by tuning the voltages applied on the metal gates. The Fabry–Pérot resonances leads to oscillations in the transmission which can be observed in experimentally. This electric modulated-mechanism provides us a realistic way to switch the transmission in edge states which can be constructed in low-power information processing devices.

BACKGROUND Complications are the most important outcome determinants for acute pancreatitis (AP). We designed this single-center retrospective study to evaluate the clinical findings (complications, disease severity, and outcomes) of 218 patients with AP and to identify variables associated with ascites. MATERIAL AND METHODS We extracted clinical data from consecutive patients with AP and divided them into 2 groups based on presence or absence of ascites. BACKGROUND Renal dysfunction is a leading cause of death in patients with acute pancreatitis (AP) and often occurs later than respiratory complications. Whether respiratory complications can predict renal impairment remains unclear. The aim of this study was to investigate the association between pleural effusion and renal dysfunction in AP. MATERIAL AND METHODS Medical records were reviewed from individuals who were hospitalized with AP from January 1, 2015 to December 31, 2019. The patients were divided into 2 groups, based on the presence or absence of pleural effusion on admission. Disease severity, renal function parameters, and outcomes were compared between the 2 groups. RESULTS A total of 222 patients were enrolled, 25 of whom had pleural effusion on admission and 197 who did not. The main process is to determine the quantitative mathematical calculation method, collect the perceptual evaluation index of the bank experience contact at the design stage, use the analytic hierarchy process to establish the evaluation index system, build the design participation model of the bank experience contact, and use the fuzzy analytic hierarchy process The contact design participation is calculated and the results are quantitatively evaluated. This research can provide reference for decision makers when setting up service contacts, and provide scientific and effective data evaluation scales for bank design transformation, development process, interaction design, service design, etc., for subsequent banking user experience. A series of 6- and 9-substituted cleistopholine derivatives has been designed, synthesized and investigated to inhibit the aggregation of acetylcholinesterase (AChE), butyrylcholinesterase (BChE) and β-myloid (A β). Results showed that these synthetic compounds had excellent AChE inhibitory activity and a significant in vitro inhibitory potency toward the self-induced A β aggregation. When SH-SY5Y cells were treated with these substituted cleistopholine derivatives during they overexpressed the Swedish mutant form of human β -amyloid precursor protein (APPsw), A β 42 secretion levels were significantly reduced. According to a parallel artificial membrane permeation assay for BBB, seven out of these sixteen synthetic compounds probably could cross the blood-brain barrier (BBB) to reach their targets in the central nervous system (CNS).

A series of 4- substituted sampangine derivatives (4-aminoalkylaminosampangine Ar-NH(CH2)nNR1R2) has been designed, synthesized, and tested for their ability to inhibit acetylcholinesterase (AChE), butyrylcholinesterase (BChE) and β-myloid (Aβ) aggregation. The synthetic compounds exhibited high AChE inhibitory activity and a significant in vitro inhibitory potency toward the self-induced Aβ aggregation. While, treatment of SH-SY5Y cells overexpressing the Swedish mutant form of human β-amyloid precursor protein (APPsw) with derivatives was associated with significant reduction of Aβ42 secretion levels. Moreover, most of the synthetic compounds were predicted to be able to cross the blood-brain barrier (BBB) to reach their targets in the central nervous system (CNS) according to a parallel artificial membrane permeation assay for BBB. The result encourages us to study this class of compounds thoroughly and systematically.

Objective To observe the influences of regional citrate or systemic heparin anticoagulation on acid-base balance, coagulation, electrolytes, serum creatinine, alanine aminotransferase (ALT) and cardiac index (CI) during continuous renal replacement therapy (CRRT) rewarming in accidental hypothermia dogs. Methods Nineteen adult beagles were given abdominal trauma coupled with deep hypothermia [(28±0.5)℃] induced by seawater immersion for establishing animal model of trauma. According to rewarming ways, the animal models were randomly divided into three groups, comparison group (warm bath rewarming, n=5), CRRT rewarming-systemic heparin anticoagulation group (heparin anticoagulation group, n=7) and CRRT rewarming-regional citrate anticoagulation group (citrate anticoagulation group, n=7). During the rewarming routine blood examination was performed, and blood chemistry, coagulation function, blood gas and hemodynamic status were assayed, at the same time the mortality was recorded. Results During the rewarming, the mortality was 14.3% (1/7) in heparin anticoagulation group, 40.0% (2/5) in warm bath group and 0 in regional citrate anticoagulation group. Blood temperature in creased to 38℃, the heparin anticoagulation group showed a significant decrease of platelet compared with citrate anticoagulation group and comparison group (P0.05). Conclusions CRRT plus warm bath rewarming have better effect on improving metabolic acidosis than warm bath rewarming alone in accidental hypothermia dogs. In comparison with systemic heparin anticoagulation, regional citrate anticoagulation has smaller influences on coagulation system and platelet when CRRT rewarming performed in accidental hypothermia dogs. Gastric cancer (GC) is a serious health problem worldwide. The potential involvement of long noncoding RNAs in GC progression remains largely unexplored. Here, we identified a novel long noncoding RNA referred to as onclncRNA-626 (oncogenic lncRNA RP11-626H12.3), which was highly upregulated in GC tissues. The high expression levels of onclncRNA-626 in GC patients predicted poor prognoses. Functional assays indicated that onclncRNA-626 could promote the proliferation and metastasis of GC cells in vitro and in vivo. In exploring the molecular mechanisms guiding these functions, we found that onclncRNA-626 specifically interacted with serine- and arginine-rich splicing factor 1 (SRSF1) and increased its stability. SRSF1 was upregulated in GC tissues and correlated with onclncRNA-626 expression and patient survival. Furthermore, RNA-seq data revealed that onclncRNA-626 affected multiple signaling pathways, including the p53 signaling pathway. Rescue experiments showed that onclncRNA-626 probably performed its biological function through SRSF1 mediation of the p53 pathway. Together, our findings demonstrate that onclncRNA-626 promotes GC progression by binding SRSF1; further, this lncRNA is a potential prognostic biomarker for GC patients.

We present theoretical study on developing a one-dimensional (1D) photonic crystal heterojunction (h-PhC) that consists of a monolayer molybdenum disulfide (MoS2) structure. By employing the transfer matrix method, we obtained the analytical solution of the light absorption and emission of two-dimensional materials in 1D h-PhC. Simultaneously enhancing the light absorption and emission of the medium in multiple frequency ranges is easy as h-PhC has more modes of photon localization than the common photonic crystal. Our numerical results demonstrate that the proposed 1D h-PhC can simultaneously enhance the light absorption and emission of MoS2 and enhance the photoluminescence spectrum of MoS2 by 2–3 orders of magnitude.

Biochar, a production of cotton/sludge pyrolysis, was used as an adsorbent for the removal of heavy metals from aqueous solutions. In this paper, it was assessed that the effect of biochar produced parameters including pyrolysis temperature and heating rate, adsorption time, solution pH and biochar modification on removal of Cd from aqueous solution, and the removal effect of heavy metals from mixed aqueous solution was also studied. The results showed that the optimum conditions were pyrolysis temperature of 550°C, heating rate of 5°C/min, adsorption time of 90min, biochar dosage of 10g/L and solution pH=6, respectively. And the effect was a little increased when the biochar were impregnated with chemicals. About 99% Cd, Pb and Zn were removed from aqueous solution using biochar under the optimum conditions.

To clarify the precipitation behavior of beneficial inclusions and mechanism of their effects on microstructure, the effect of aluminum content on inclusion’s characteristics and their influence on the refinement of microstructure in Al-Ti complex deoxidized magnesium-treated steels were systematically investigated based on experiment and calculation. The results showed that due to the dual effects of Ti and Mg deoxidation, a large amount of finely dispersed Al2O3-TiO x -MgO inclusions in low aluminum steel with a complex multilayer or mosaic structure were formed, whereas a relatively smaller amount of Al2O3-MgO inclusions with the simple bundle structure were observed in high aluminum steel. The Al2O3-TiO x -MgO core oxides are more conducive to the precipitation of multiple manganese sulfides with thinner thickness on their local surfaces. Thus, the inclusion deformation, which mainly depends on the surface manganese sulfides layer, is smaller in low aluminum steel than that in high aluminum steel. Complex inclusions in low aluminum steel can pin austenite grain boundaries and induce interlocking acicular ferrite effectively. In addition to the small size and chemical composition of inclusions, the complex structure of oxides and the precipitation of multiple MnS on their surface are important to the nucleation of interlocking AFs on inclusions in Ti-deoxidized Mg-treated steel. The AFs quantity is much more, and the grain size is more uniform in low aluminum steel than that in high aluminum steel.

Soil washing experiments were carried out with citric acid as washing reagent for the remediation of soils highly contaminated with heavy metals, then activated carbon was used in absorption processing for leaching solution. In this study, the effects of the main operating variables for removal of metals from soils were first discussed. The results showed that 36.% Pb, 47.74% Cu and 61.88% Cd were removed from the contaminated soils by optimizing the washing parameters at citric acid concentration 0.2 mol/l, mixing time 2 h, liquid-soil ratio 20 and solution pH=4, respectively. In the adsorption experiments of leachates, the optimum conditions were found as follows: solution pH=7, mixing time of 2 h, standing time of 60 min and activated carbon dosage of 1g/100ml.

In order to reveal the heat conduction direction on roadbed, a series of theoretical analysis and mathematical derivations are used. Through simplifying the frozen soil layer as an equivalent homogeneous frozen body, the physical model of heat conduction was founded. Also, applying the entropy change of the thermodynamics second law, the direction of the actual heat transfer was analyzed. Further more, the three-dimensional and two-dimensional differential equation of unsteady heat transfer in Cartesian model were obtained by using Fourier's Law and importing the micro unit. It has significant senses for deep research of roadbed tympanites.

The physical model and calculating model of temperature field are founded according to the founded controlling equation. By simulating the changing progress of climate features, the temperature field of roadbed in permafrost region in different season was obtained. And some results are got by analyzing the diversity of temperature of key points. The last, the technology assumption of effectively controlling tympanites which combines replacing part frozen soil and resisting highway tympanites structure is brought up.

Two new symmetric double-armed oxadiazole-bridged ligands, 4-methyl-{5-[5-methyl-2-(pyridin-3-ylcarbonyloxy)phenyl]-1,3,4-oxadiazol-2-yl}phenyl pyridine-3-carboxylate (L1) and 4-methyl-{5-[5-methyl-2-(pyridin-4-ylcarbonyloxy)phenyl]-1,3,4-oxadiazol-2-yl}phenyl pyridine-4-carboxylate (L2), were prepared by the reaction of 2,5-bis(2-hydroxy-5-methylphenyl)-1,3,4-oxadiazole with nicotinoyl chloride and isonicotinoyl chloride, respectively. By the implicit C-D pairs, a new approach to auto-B¨acklund transformations of nonlinear evolution equations is presented in this paper. The auto-B¨acklund transformations for combined KdV and MKdV equations are obtained by this approach.

Cement concrete pavement as a common form of the road has been widely used because of its high strength, stiffness, and good durability characteristics. In order to explore failure modes and the strength control factors of the cement concrete pavement in the region, site investigation and the orthogonal test method in laboratory are used. Test combination design for the main control factors of the cement concrete strength was executed based on orthogonal test method. Strength of concrete specimen was determined by using indoor pressure test machine. The results show that the chiefly control parameter of affecting concrete strength is water cement ratio and the following are sand ratio and the coarse aggregate grading.

In eukaryotes, the spindle checkpoint acts as a surveillance mechanism that ensures faithful chromosome segregation. The spindle checkpoint prevents premature separation of sister chromatids and the onset of anaphase until every chromosome is properly attached to the mitotic spindle. Tumorigenesis might result from generation of aneuploidy by dysfunction of the spindle checkpoint. Differences of the checkpoint system in normal cells versus tumor cells might provide a new opportunity in cancer drug development; therefore, efforts to identify the spindle checkpoint inhibitors have been fostered. Based on spindle checkpoint inhibitors being able to induce cells to exit mitotic arrest caused by microtubule drug treatment, we developed a cell-based assay to screen compounds that were potential spindle checkpoint inhibitors. EM algorithm is a common method to solve mixed model parameters in statistical classification of remote sensing image. The EM algorithm based on fuzzification is presented in this paper to use a fuzzy set to represent each training sample. Via the weighted degree of membership, different samples will be of different effect during iteration to decrease the impact of noise on parameter learning and to increase the convergence rate of algorithm. The function and accuracy of classification of image data can be completed preferably.

In order to analyze impact of roller loads applying in subgrade connecting, numerical software had been executed to calculate deformation caused by three types of roller loads to 6m subgrade. In this analysis, surface layer of the old subgrade, slope toe point and connecting point had been chosen as key points to study deformation under different construction processes. The results show that the displacement in the vertical direction of point A and point B increases with the roller load's increasing. Deformation of key points has the non-linear relationship with construction steps. According to the minimum deformation, the first scheme is chosen which is beneficial to the train’s safty and stability of whole subgrade. It will be instructive to control differentia deformation for the connecting subgrade projects in high-speed railway

It is a good alternative that uses reduced IPv6 as network layer protocol of wireless sensor network (WSN). This will generate a new problem: how to choose some nodes as router for homogenous, dense deployment WSN. This paper proposed a centralized router election method, which is based on facility location model, for IP-WSN. This method creates a 0-1 integer programming model by regarding router election as uncapacitated facility location problem firstly. Then genetic algorithm is used for solving the created model. The simulation results show that this method can elect small number of nodes as router with high efficiency.
