Abstract Physcion is an anthraquinone compound observed dominantly in medicinal herbs. This anthraquinone possesses a variety of pharmaceutically important activities and has been developed to be a widely used antifungal biopesticide. Herein, we report on the effective preparation of 3R-torosachrysone (4), a tetrahydroanthracene precursor of physcion, in Aspergillus oryzae NSAR1 by heterologous expression of related genes mined from the phlegmacins-producing ascomycete Talaromyces sp. F08Z-0631. Conditions for converting 4 into physcion were studied and optimized, leading to the development of a concise approach for extracting high-purity physcion from the alkali-treated fermentation broth of the 4-producing A. oryzae strain. Snakebite envenoming adversely affects human health and life worldwide. Presently, no suitable diagnostic tools for snakebite envenoming are available in China. Therefore, we sought to develop reliable diagnostic tests for snakebite management. We conducted affinity purification experiments to prepare species-specific antivenom antibody (SSAb). In brief, affinity chromatography with an antibody purification column (Protein A) was conducted to purify immunoglobulin G from Bungarus multicinctus (BM) venom hyperimmunized rabbit serum. The cross-reactive antibodies were removed from commercial BM antivenin by immune adsorption on the affinity chromatography columns of the other three venoms, Bungarus Fasciatus (FS), Naja atra (NA), and O. hannah (OH), generating SSAb. The results of western blot analysis and enzyme-linked immunosorbent assay (ELISA) showed the high specificity of the prepared SSAb. The obtained antibodies were then applied to ELISA and lateral flow assay (LFA) to detect BM venom. The resulting ELISA and LFA could specifically and rapidly detect BM venom in various samples with the limits of quantification as 0.1 and 1 ng/ml, respectively. This method could effectively detect snake venom in experimentally envenomed rats (simulating human envenomation), which could distinguish positive and negative samples within 10-15 min. This method also showed promise in serving as a highly useful tool for a rapid clinical distinguishing of BM bites and rational use of antivenom in emergency centers. The study also revealed cross-reactivity between BM and heterogenous venoms, suggesting that they shared common epitopes, which is of great significance for developing detection methods for venoms of the snakes belonging to the same family.

Direct interspecies electron transfer (DIET) is an effective mechanism for microbial species to exchange electrons cooperatively during syntrophic metabolism. It is generally accepted that DIET is mainly mediated by electrically conductive pili and outer surface c-type cytochromes (c-Cyts). However, as an extracellular matrix is ubiquitous and abundant on the surface of microorganisms, the effect and mechanism of exopolysaccharides on DIET are still unclear. This study constructed a co-culture of exopolysaccharides-deficient Geobacter sulfurreducens with Geobacter metallireducens to explore the role of exopolysaccharides in DIET. Results revealed that the deficiency of exopolysaccharides extended the metabolic period of the co-culture by 44.4% and changed the proportions of each species in the co-culture. The exopolysaccharides-deficient co-culture failed to form large, tight spherical aggregates and the expression of c-Cyts and pili was decreased. The addition of magnetite and granular activated carbon (GAC), respectively, might compensate for the functions of c-Cyts and pili in the first generation of co-culture, but the stimulatory effect on the metabolic stable period co-culture was fairly limited. These findings demonstrate that non-conductive exopolysaccharides are an important component of DIET aggregates and an extracellular matrix for DIET-required c-Cyts.

ABSTRACT: In this paper, substantial domestic and foreign research results of microscopic shale reservoir space were systemically reviewed, the research history consisting of simple observation and qualitative classification, quantitative research, the combination of qualitative and quantitative research successively as well as the characteristics of each research stage were summarized. In addition, the current problems existing in the characterization methods of shale reservoir space were also analyzed. Furthermore, based on massive actual detection of typical core samples obtained from more than 50 global shale gas wells and relevant practical experience, a comprehensive characterization method of combining qualitative with the semi-quantitative characterization was put forward. In detail, the indicators of the qualitative characterization include pore combination type and organic-matter microscopic morphology type, while the core elements of the semi-quantitative characterization include the percentage of the organic-matter area and the plane porosity of the pores of different types. Based on the reference of the naming and classification of rocks, the three-end-member diagram method was used to characterize microscopic shale reservoir space. This is achieved by plotting the three end-member diagram of 3 kinds of first-order critical reservoir spaces, i.e., organic-matter pores, matrix pores, and micro-fractures, in order to intuitively present the features of the microscopic pore combination. Meanwhile, statistic histograms of organic-matter microscopic morphology type and the plane porosity of different types of pores were adopted to characterize the development degree of second-order pores quantitatively. By this comprehensive characterization method, the importance of both pore combination and the microscopic morphology of organic matter were emphasized, revealing the control of organic-matter microscopic morphology over the organic-matter pores. What is more, high-resolution FE-SEM was adopted to obtain semi-quantitative statistics results. In this way, the features of pore development and pore combination were quantified, not only reflecting the types and storage capacity of the microscopic shale reservoir space, but also presenting the hydrocarbon-generating potential of organic matter in shale. Therefore, the results of this research are capable of providing in-depth microscopic information for the assessment and exploration and development of shale gas resources.
