Background: Higher vocational college students face more life stress, which can easily result in depression and hinder their healthy growth. This study aimed to explore the roles of survival situation and personality temperament in the relationship between life stress and depression., Methods: A self-compiled "College Students' Life Stress and Mental Health Questionnaire" was used to survey 4800 students in a Chinese higher vocational college. The questionnaire consisted of five subscales: life stressors scale, stress response scale, depression scale, personality temperament types scale, and survival situations scale. The sample included 4705 students, of whom 3449 (73.30%) were males and 1256 (26.70%) were females, with 990 urban students (21.04%), 3715 rural students (78.96%). The age of the participants ranged from 17 to 33 years. The data were analyzed using SPSS v26, PROCESS v3.3, and AMOS v23., Results: (1) The depression rate of higher vocational students was 18.10% (with a severe depression rate of 1.60%). Life stress could explain 43.80% of depressive episodes (p < 0.01), (2) Among survival situations, the depression degree and rate of students in adversity were the highest (M = 1.56, 24.10%), (3) Among temperament types, the depression degree and rate of melancholic students were the highest (M = 2.13, 36.05%), (4) Survival situation and personality temperament had significant moderating interaction effects on depression caused by life stress (p < 0.01), students in adversity and depressive temperament were more susceptible, (5) Survival situations moderated three paths of the "life stressors-stress response-depression" partial mediation model, and personality temperament types moderated "stress response-depression" path., Conclusion: Prosperity and sanguine temperament are protective factors of depression caused by life stress in higher vocational students. Dilemma, adversity and melancholic temperament are risk factors of depression caused by life stress in higher vocational students., (© 2023. The Author(s).)

The majority of men with defects in spermatogenesis remain undiagnosed. Acephalic spermatozoa is one of the diseases causing primary infertility. However, the causes underlying over half of affected cases remain unclear. Here, we report by whole-exome sequencing the identification of homozygous and compound heterozygous truncating mutations in PMFBP1 of two unrelated individuals with acephalic spermatozoa. PMFBP1 was highly and specifically expressed in human and mouse testis. Furthermore, immunofluorescence staining in sperm from a normal control showed that PMFBP1 localizes to the head-flagella junction region, and the absence of PMFBP1 was confirmed in patients harboring PMFBP1 mutations. In addition, we generated Pmfbp1 knock-out (KO) mice, which we found recapitulate the acephalic sperm phenotype. Label-free quantitative proteomic analysis of testicular sperm from Pmfbp1 KO and control mice showed 124 and 35 proteins, respectively, increased or decreased in sperm from KO mice compared to that found in control mice. Gene ontology analysis indicates that the biological process of Golgi vesicle transport was the most highly enriched in differentially expressed proteins, indicating process defects related to Golgi complex function may disturb formation of the head-neck junction. Collectively, our data indicate that PMFBP1 is necessary for sperm morphology in both humans and mice, and that biallelic truncating mutations in PMFBP1 cause acephalic spermatozoa., (© 2018 John Wiley & Sons A/S. Background: Metabolomics is emerging as a valuable tool in clinical science. However, one major challenge in clinical metabolomics is the limited use of standardized guidelines for sample collection and handling. In this study, we conducted a pilot analysis of serum and plasma to determine the effects of processing time and collection tube on the metabolome. Methods: Blood was collected in 3 tubes: Vacutainer serum separator tube (SST, serum), EDTA (plasma) and P100 (plasma) and stored at 4 degrees for 0, 0.5, 1, 2, 4 and 24 h prior to centrifugation. Compounds were extracted using liquid-liquid extraction to obtain a hydrophilic and a hydrophobic fraction and analyzed using liquid chromatography mass spectrometry. Differences among the blood collection tubes and sample processing time were evaluated (ANOVA, Bonferroni FWER ≤ 0.05 and ANOVA, Benjamini Hochberg FDR ≤ 0.1, respectively). Results: Among the serum and plasma tubes 93.5% of compounds overlapped, 382 compounds were unique to serum and one compound was unique to plasma. There were 46, 50 and 86 compounds affected by processing time in SST, EDTA and P100 tubes, respectively, including many lipids. In contrast, 496 hydrophilic and 242 hydrophobic compounds differed by collection tube. Forty-five different chemical classes including alcohols, sugars, amino acids and prenol lipids were affected by the choice of blood collection tube. Conclusion: Our results suggest that the choice of blood collection tube has a significant effect on detected metabolites and their overall abundances. Perhaps surprisingly, variation in sample processing time has less of an effect compared to collection tube; however, a larger sample size is needed to confirm this., Competing Interests: The authors do not have any conflict of interest to declare.

Aim: To evaluate the efficacies of six derivatives of Compound 2, a novel YycG histidine kinase inhibitor with the thiazolidione core structure in the treatment of medical device-related biofilm infections., Methods: The minimal inhibitory concentration (MIC) of the derivatives was determined using the macrodilution broth method, and the minimal bactericidal concentration (MBC) was obtained via sub-culturing 100 μL from each negative tube from the MIC assay onto drug-free Mueller-Hinton agar plates. Biofilm-killing effect for immature (6 h-old) biofilms was examined using a semiquantitative plate assay, and the effect on mature (24 h-old) biofilms was observed under a confocal laser scanning microscope (CLSM)., Results: The derivatives potently suppressed the growth of Staphylococcus epidermidis. The MIC values of the derivatives H2-10, H2-12, H2-20, H2-29, H2-27, and H2-28 on S epidermidis ATCC 35984 were 24.3, 6.5, 6.2, 3.3, 3.1, and 1.5 μg/mL, respectively. The MBC values of these derivatives were 48.6, 52.2, 12.4, 52.6, 12.4, and 6.2 μg/mL, respectively. The derivatives killed all bacteria in immature (6 h-old) biofilms and eliminated the biofilm proliferation. The derivatives also displayed strong bactericidal activities toward cells in mature (24 h-old) biofilms, whereas they showed low cytotoxicity and hemolytic activity toward Vero cells and human erythrocytes., Conclusion: The bactericidal and biofilm-killing activities of the new anti-YycG compounds were significantly better than the parent Compound 2.

Objective: To investigate the early diagnosis and treatment of congenital adrenal hyperplasia (CAH) complicated by testicular adrenal rest tumors (TART)., Methods: We retrospectively analyzed the clinical data of 1 case of late-onset CAH complicated by TART diagnosed and treated in Xiamen Women and Children Health Care Hospital., Results: The patient was a 15 years old boy, short statured and dark skinned, with skin pigmentation in the gum and external genital, secondary sex characteristics of the adult and irregular tubercles palpable in the bilateral testes. Laboratory examinations showed obviously increased levels of ACTH, 17-KS, DHEA-S and progesterone and evidently decreased levels of FSH, LH and CO. The low-dose dexamethasone suppression test reduced ACTH and DHEA-S to normal. Imaging examinations revealed soft tissue density in the bilateral adrenal glands, especially on the right, and irregularly increased volume of the bilateral testes, particularly on the left, with heterogeneous signals and septas and surrounded by the fluid signals. Histopathological examinations showed the eosinophilic cytoplasm to be polygon- or round-shaped, interstitium-like cells arranged in line, and lipopigment in the endochylema. Immunohistochemical results were negative for testicular interstitial cell tumor. The clinical signs of the patient were improved after 3 months of dexamethasone treatment, the hyperplastic nodules in the left testis decreased obviously and those in the right testis disappeared after 6 months, and the hyperplastic nodules in the adrenal glands vanished after 9 months., Conclusion: Based on the clinical manifestations and the results of auxiliary examinations, this case was diagnosed as late-onset CAH complicated by TART, which was attributed to the continued surge of ACTH induced by corticoadrenal insufficiency. Sufficient dexamethasone treatment could make the TART decrease or disappear and the CAH vanish; it could also improve the clinical symptoms and bring the laboratory results to normal.

Unlabelled: To establish a method for the determination of lead in meconium from 144 samples of newborn through nitric acid digestion by means of graphite furnace atomic absorption spectrometry., Methods: after being baked for at least 12 h at 60 degrees C, 0.3 g of meconium was digested by nitric acid and hydrogen peroxide in turn at 80 degrees C under water-bath condition for 1h and then metered to the whole volume of to 2 mL. After correcting the background with D2 lamp, specimen basal corpuscle matched standard curve was used to detect the lead content., Results: The mean lead content of 93 experimental samples was 1.934 microg x g(-1) with the standard deviation (SD) of 1.551, and that of the 51 control samples was 1.012 microg x g(-1), with the SD of 1.084. There was a significant difference in lead levels of in meconnt between the experimental group and control group (p = 0.000)., Conclusion: The lead content of the experimental was significantly higher than that of the control group detected by this method. This method was stable and efficient.

Objective: To construct the prokaryotic plasmid of FUS1 gene for efficient FUS1 expression in E.coli strain Rosetta(DE3)2plys., Methods: The full-length FUS1 gene was amplified by PCR from the total RNA of umbilical mesenchymal stem cells and cloned into pET-32a(+) vector followed by identification with PCR and sequencing. The recombinant plasmid pET-32a(+)-FUS1 was transformed into the E.coli strain Rosetta(DE3)2plys and the target protein expression was induced by IPTG., Results: The plasmid pET-32a(+)-FUS1 was obtained successfully as verified by PCR and sequence analysis. High expression of the fused FUS1 protein was achieved after induction by low-concentration IPTG (25 micromol/L) for 3 h, and the recombinant FUS1 protein accounted for 40% of the total bacterial protein of Rosetta(DE3)2plys., Conclusion: The recombinant FUS1 plasmid has been successfully cloned, which allows highly efficient FUS1 expression in Rosetta (DE3)2 plys.

Aim: To investigate the role of human papillomavirus (HPV) HPV16/18 E6 oncogene in the carcinogenesis of esophageal cell carcinoma (ESCC)., Materials and Methods: Tissue microarray (TMA) block was constructed from 60 cases of paraffin-embedded ESCC tissues and pair-matched controls (adjacent normal epithelium). Immunohistochemistry (IHC) methods were applied to detect the expression of HPV16/18 E6, p53, p21(WAF1), MDM2, Ki67 and cyclin D1 proteins on TMA slides. In situ hybridization (ISH) targeting HPV gene was also used., Results: In ESCC samples, 18.3% (11/60) were revealed HPV16/18 E6 positive by IHC, while 40.0% (24/60) HPV positive by ISH; HPV16/18 E6 expression was significantly higher than that of control samples. In ESCC samples, the expressions of p53, p21(WAF1), Cyclin D1, MDM2 and Ki67 were recorded in 60.0% (36/60), 40.0% (24/60), 51.7% (31/60), 65.0% (39/60) and 88.3% (53/60) cases respectively, In ESCC samples, p53, MDM2 and Ki67 expression correlated with the HPV16/18 E6 expression (p less, similar 0.01), p21(WAF1) expression - with these of MDM2 and cyclin D1 (p less, similar 0.01) whilst expression of Ki67 - with ESCC grade (p less, similar 0.01)., Conclusion: HPV might be one of etiological factor of esophageal carcinoma in Shantou, China. p53, MDM2 proteins may play important roles in the pathogenesis of HPV-associated ESCC.

Objective: To assess the effect of benzene on sperm DNA., Methods: Twenty-seven workers exposed to benzene for over two years were included in the experiment group and 35 unexposed ones were chosen as controls. Damage by benzene on sperm DNA was detected by modified single cell gel electrophoresis (SCGE)., Results: The benzene concentration in the air of the workplace of the exposed workers was (86.49 +/- 2.83) mg/m3, twice that of the national maximum allowance (40 mg/cm3). Urinary, trans-muconic acid in the exposed group was (1.40 +/- 0.62) mg/L (n = 27), significantly higher than that of the controls [(0.82) +/- 0.16) mg/L, n = 35]. After SCGE, the percentage of head DNA in the exposed group [(70.18% +/- 7.36%, n = 13 ] was significantly lower than that of the controls (90.62% +/- 2.94%, n = 16), P < 0.001., Conclusion: Higher concentration of benzene could cause damage to the sperm DNA of the workers exposed to it.

Objective: To investigate the frequency of numerical aberrations for chromosomes 7 and 8 in the sperms of workers exposed to benzene series., Methods: Numerical aberrations in the sperms of workers were detected by two-color fluorescence in situ hybridization with biotin labeled chromosome 7 probe (D7Z1) and digoxingenin labeled chromosome 8 probe (D8Z1)., Results: The time-weight average air concentration (TWA) of benzene in the workshop was 42.29 mg/m(3), which was significantly higher than that of the national maximum allowable concentration [6 mg/m(3)]. The concentration of urinary trans,trans-muconic acid(ttMA) in the exposed group was also higher than that of the control group. In all, 155721 sperm nuclei from 15 workers in the exposed group and 123771 sperm nuclei from 12 individuals in the control group were examined. The results showed that the frequency of diploidy sperms and the frequencies of disomic sperm for chromosomes 7 and 8 in the exposed group (0.129%, 0.170%, 0.078%) were significantly higher than those of the control group (0.055%, 0.053%, 0.033%), respectively. The frequencies of nullisomic sperm for chromosomes 7 and 8 in the exposed group (0.165%, 0.088%) were also significantly increased, compared with those of the control group (0.056%, 0.029%). A statistically significant difference in the frequency of overall numerical chromosome aberrations was seen between the exposed group (0.745%) and the control group (0.289%)., Conclusion: The results suggested that higher concentration of benzene may induce higher frequencies of numerical aberrations in the sperms of workers exposed to benzene series.

To study the frequencies of numerical and structural aberrations for chromosome in sperm of benzene exposed workers, the multi-color FISH was used. Four DNA probes(one for chromosome 1 centromere and one for 1 p terminal, and two for chromosome 18 centromere) were hybridized with interphase sperms, and the frequencies of numerical aberrations for chromosome 1, 18 and structural aberrations of chromosome 1 were detected simultaneously. The time weighted average concentration (TWA) of benzene in workplace (42.29 mg/m3) was higher than that of our national maximum allowable concentration (6 mg/m3). The geometric concentration of urinary trans, trans-muconic acid(tt-MA) in exposed group was significantly higher than that of control group. A total of 144,282 sperm of 15 benzene-exposed workers and 135,937 sperm in 14 controls were scored. The frequency of hybridization efficiency was 99.85%. The mean frequencies of disomic sperms for chromosome 1 and 18 in exposed group(0.088% +/- 0.041%, and 0.087% +/- 0.049%, respectively) were statistically increased over that of the control group(0.045% +/- 0.024%, and 0.035% +/- 0.028%), and the mean frequencies of nullisomic sperms for chromosome 1 and 18(0.11% +/- 0.059%, 0.075% +/- 0.035%) in exposed group were statistically increased over that of control group too (0.048% +/- 0.018%; 0.045% +/- 0.024%). The frequencies of diploidy sperm were no difference in both exposed and control groups. The mean frequencies of terminal duplication and terminal deletion for chromosome 1 p(0.16% +/- 0.037%; 0.14% +/- 0.053%, respectively) were significantly increased over that of control group(0.082% +/- 0.023%; 0.069% +/- 0.028%, respectively). The mean frequencies of centromeric duplication and centromeric deletion for chromosome 1(0.10% +/- 0.035%; 0.10% +/- 0.041%, respectively) were significantly increased over that of control group(0.075% +/- 0.023%; 0.060% +/- 0.029%). Our experiments showed that exposed to benzene at higher concentration(42.29 mg/m3) may induce increases in frequencies not only of numerical aberrations for chromosome 1 and 18, but also of structural aberrations for chromosome 1 of sperms in exposed workers.

To explore acrylonitrile (ACN)-induced DNA strand breakage and sex chromosome aneuploidy in human spermatozoa, semen parameters were examined among 30 acrylonitrile-exposed workers according to WHO laboratory manual for the examination of human sperm. DNA strand breakage of sperm cells was investigated among 30 ACN-exposed workers using single cell gel electrophoresis (SCGE). The frequency of sex chromosome aneuploidy in sperm cells was analyzed among nine ACN-exposed workers using fluorescence in situ hybridization (FISH). The geometrical mean of sperm density was 75 x 10(6)ml(-1) in exposure group, significantly lower than 140 x 10(6)ml(-1) in the control. The geometrical mean of sperm number per ejaculum was 205 x 10(6) in exposure group, significantly lower than 280 x 10(6) in the control. The rates of comet sperm nuclei were 28.7% in exposure group, significantly higher than 15.0% in the control. Mean tail length was 9.8 microm in exposure group, longer than 4.3 microm in the control. The frequency of sex chromosome disomy was 0.69% in exposure group, significantly higher than 0.35% in the control. XY-bearing sperm was the most common sex chromosome disomy, with an average rate of 0.37% in exposure group, and 0.20% in the control. XX- and YY-bearing sperm accounted for an additional 0.09 and 0.23% in exposure group, and 0.05 and 0.10% in the control. The results indicate that ACN affect semen quality among ACN-exposed workers. ACN or its metabolites could induce reproductive defects as an in vivo multipotent genotoxic agent by inducing DNA strand breakage and sex chromosome non-disjunction in spermatogenesis.

Objectives: To assess the effect of freezing on the sperm DNA., Methods: To assess the sperm DNA preserved at -80 degrees C by using the single cell gel electrophoresis (SCGE)., Results: There was no statistical difference on the time factor by analysis of variance (ANOVA)., Conclusions: Integrity of sperm DNA could not be devastated in frozen states.

To study the frequency of numerical chromosome aberrations in sperm of workers exposed to benzene series, the aneuploidy frequencies of 9 and 18 chromosomes were detected in sperm of workers by two color fluorescence in situ hybridization with digoxingenin labeled 9 chromosome probe (D9z1) and biotin labelled 18 chromosome probe (D18z1). The timeweight average air concentration (TWA) of benzene in the workplace was 86.49 mg/m3, it was two fold higher than the national maximum allowable concentration. The concentration of urinary trans, transmuconic acid (ttMA) in exposed group was significantly higher than that of control group. The sperms counted in 14 exposed workers and 16 control workers were 136,401 and 156,955 respectively. The results showed that the disomy frequencies of sperm for chromosome 9 and 18 (0.168% +/- 0.063% and 0.055% +/- 0.031% respectively) and the frequency of diploidy sperm (0.073% +/- 0.045%) were statistically increased over that of the control group (0.050% +/- 0.030%; 0.033% +/- 0.025% and 0.040% +/- 0.036%, respectively). The nullisomic frequencies of 9 and 18 chromosome in the exposed group (0.206% +/- 0.047%, 0.068% +/- 0.044%) were statistically increased over that of the control group too (0.067% +/- 0.037%, 0.048% +/- 0.034%). The frequency of overall numerical chromosome aberrations in the exposed group was 0.570% +/- 0.144%, when this value was compared with that of control (0.218% +/- 0.071%), a statistically significance was presented. Our experiments showed that exposed to benzene at higher concentration may induce increase in aneuploidy frequency of sperm autosomal chromosome in exposed workers.
