Abstract Background Fruit photosynthetic apparatus development comprises a series of biological processes which is essential in determining fruit development and quality formation. However, the understanding of the regulation of fruit photosynthetic apparatus development remains poor. Results In this study, we identified a transcriptional factor SlBES2, the closest homolog of BES1 and BZR1 in tomato BES1 family, is highly expressed in fruit at mature green (MG) stage and exhibited transcriptional inhibition activity. Down-regulation of SlBES2 resulted in fruits showing paler fruit than wild type at MG stage, in contrast, SlBES2-overexpressing tomato lines bore deeper green fruits. Notably, chlorophyll content and number of thylakoids per chloroplast in fruit was substantially increased in SlBES2-overexpressing lines, while markedly decreased in SlBES2-suppressing lines. Comparative transcriptome analysis revealed that multiple genes of the photosystem, chloroplast development and chlorophyll metabolism pathways were regulated by SlBES2. Further verification revealed that SlBES2 can significantly repress the transcriptional activity of SlNYC1 and Green-Flesh, and physically interact with protein SlHY5. Conclusions Collectively, this study demonstrated that SlBES2 plays an important role in regulating fruit photosynthetic apparatus development through either transcriptional repression of genes involved in chlorophyll breakdown, or posttranscriptional regulation of proteins associated with plant photomorphogenesis and chloroplast development. Our findings add a new actor to the complex mechanisms underlying photosynthetic apparatus during fruit development.

To address the optimization problem of electric field intensity in multi-target areas under transmission lines, this work suggests combining the enhanced improved particle swarm optimization (IPSO) with the analytical hierarchy process (AHP). First, using finite element simulation software, a transmission line shielding model was created. Next, the number, erection height, and phase spacing of the shielding lines were studied in relation to electric field strength. Drawing on these results, field strength shielding in many target locations was optimized using the AHP-IPSO algorithm. Research has demonstrated that the electric field intensity beneath a transmission line can be greatly decreased by installing shielded cables, and that the peak field intensity can be decreased by 28.4% by installing three insulated wires. The AHP-IPSO algorithm effectively solved the multi-factor electric field shielding optimization problem, thus offering a novel method for addressing such problems. Compared to other algorithms, the AHP-IPSO algorithm exhibits better optimization performance and offers better shielding efficiency, while also accounting for the economic aspects of shielded wire erection.

Abstract N-hydroxy pipecolic acid (NHP) plays an important role in plant immunity. In contrast to its biosynthesis, our current knowledge with respect to the transcriptional regulation of the NHP pathway is limited. This study commences with the engineering of Arabidopsis plants that constitutively produce high NHP levels and display enhanced immunity. Label-free proteomics reveals a NAC-type transcription factor (NAC90) that is strongly induced in these plants. We find that NAC90 is a target gene of SAR DEFICIENT 1 (SARD1) and induced by pathogen, salicylic acid (SA), and NHP. NAC90 knockout mutants exhibit constitutive immune activation, earlier senescence, higher levels of NHP and SA, as well as increased expression of NHP and SA biosynthetic genes. In contrast, NAC90 overexpression lines are compromised in disease resistance and accumulated reduced levels of NHP and SA. NAC90 could interact with NAC61 and NAC36 which are also induced by pathogen, SA, and NHP. We next discover that this protein triad directly represses expression of the NHP and SA biosynthetic genes AGD2-LIKE DEFENSE RESPONSE PROTEIN 1 (ALD1), FLAVIN MONOOXYGENASE 1 (FMO1), and ISOCHORISMATE SYNTHASE 1 (ICS1). Constitutive immune response in nac90 is abolished once blocking NHP biosynthesis in the fmo1 background, signifying that NAC90 negative regulation of immunity is mediated via NHP biosynthesis. Our findings expand the currently documented NHP regulatory network suggesting a model that together with NHP glycosylation, NAC repressors take part in a ‘gas-and-brake’ transcriptional mechanism to control NHP production and the plant growth and defense trade-off.

Abstract Studying the penetration characteristics of blast‐induced cracks during double water jet slotted boreholes blasting is theoretically and practically significant since it acts both by the “superposition effect” of explosion‐induced stress waves and the “guiding effect” of water jet slots. A combination of experimental testing and numerical simulation was adopted to investigate the transmission law of explosion‐induced stress waves and the propagation law of blast‐induced cracks first. Then the penetration characteristics of blast‐induced cracks was analyzed according to rock blasting theory and rock fracture mechanics theory. The results indicate that the rock between the two boreholes is mainly subjected to a tensile failure mode due to the combined effect of the “superposition effect” of explosion‐induced stress wave and the “guiding effect” of water jet slot during double water jet slotted boreholes blasting. The hole spacing significantly affects the penetration mode of blast‐induced cracks during double water jet slotted boreholes blasting, and unless it reaches a certain value, a single flat through‐crack will form between two slotted boreholes. In addition, when the blast‐induced cracks successfully penetrate each other, the hole spacing of double water jet slotted boreholes increases by 33.3% compared with that of double circular boreholes under the same conditions. The research results can provide guidance for the application of directional controlled blasting technology with water jet assistance.

Cadmium (Cd) contamination of soil threatens human health, food security, and ecosystem sustainability. The in situ stabilization of Cd has been recognized as a potentially economical technology for the remediation of Cd-contaminated soil. Recently, biochar (BC) and activated carbon (AC) have received widespread attention as eco-friendly soil amendments that are more beneficial for plant growth, soil health, and remediation of contaminated soil. An experiment was performed in a paddy field to investigate the effects of two different types of BC (maize straw biochar and bamboo biochar) and AC (coconut shell activated carbon) in combination with rape organic fertilizer (R), calcium magnesium phosphate fertilizer (P), and fulvic acid (F), respectively, on soil Cd immobilization, Cd accumulation in rice, and yield. The results indicated that the BC/AC-based amendments reduced soil bioavailable Cd (DTPA-Cd) and brown rice Cd by 9.58%–27.06% and 19.30%–71.77%, respectively. The transformation of exchangeable Cd (Ex-Cd) to carbonate-bound Cd (Ca-Cd), Fe-Mn oxide bond (Ox-Cd), and residual (Re-Cd) in soil accounted for the mitigation of Cd uptake and enrichment by rice. Additionally, BC-/AC-based amendments altered soil physicochemical properties, which significantly increased the soil pH and cation exchange capacity (CEC), total nitrogen (TN), total phosphorus (TP), soil organic carbon (SOC), and dissolved organic carbon (DOC), directly promoting soil health. All BC-/AC-based amendments significantly increased FeIMP and MnIMP concentrations by 47.31%–160.34% and 25.72%–73.09% in the Fe/Mn plaque (IMP), respectively. Maize straw and bamboo biochar-based amendments significantly increased rice yield by 10.46%–20.41% and 9.94%–16.17%, respectively, while coconut shell-activated carbon severely reduced rice yield by 65.06%–77.14%. The correlation analysis revealed that leaf Cd and IMP primarily controlled Cd uptake by rice, and soil pH, Eh, CEC, SOC, IMP, and TP influenced DTPA-Cd in soil. This field study demonstrated that maize straw and bamboo biochar-based amendments not only reduced soil DTPA-Cd in paddy fields but also decreased the accumulation of Cd in brown rice, as well as improved rice yield, which has potential application in Cd-contaminated agriculture fields. Coconut shell-activated carbon severely decreased rice yields, which is not appropriate for rice production.

Abstract Steroidal glycoalkaloids (SGAs) are major plant defense metabolites against pests, while they are considered poisonous in food. The genetic basis that guides negative selection of SGAs production during tomato domestication remains poorly understood. Here, we identify a distal enhancer, GAME Enhancer 1 (GE1), as the key regulator of SGAs metabolism in tomato. GE1 recruits MYC2-GAME9 transcriptional complex to regulate the expression of GAME cluster genes via the formation of chromatin loops located in the neighboring DNA region. A naturally occurring GE1 76 allelic variant is found to be more active in stimulating GAME expression. We show that the weaker GE1 allele has been the main driver for selecting reduced SGAs levels during tomato domestication. Unravelling the “TFs-Enhancer-Promoter” regulatory mechanism operating in SGAs metabolism opens unprecedented prospects for SGAs manipulation in Solanaceae via precision breeding strategies.

With the continuous innovation of genomics, proteomics and molecular biological detection technology, the treatment of non-small cell lung cancer (NSCLC) has changed from traditional chemotherapy to immunotherapy and targeted therapy. Among them, molecular tumor markers targeting tyrosine kinase pathways play more important roles in clinical practice. For advanced NSCLC patients with positive epidermal growth factor receptor (EGFR) mutations, there are many first-line drugs on the market and they could bring significant efficacy, thus completely subverting the treatment pattern of advanced NSCLC. Common mutations of EGFR in Chinese patients are located on exons 19, 20 and 21, of which exons 19 and 21 mutations are the more common types. Besides, there is also a subtype of EGFR mutations, known as EGFR 20 exon insertion (EGFR 20ins) mutation. The authors summarized the treatment of a lung adenocarcinoma patient with EGFR 20ins mutation accepting Furmonertinib mesylate, in order to provide effective references for clinical diagnosis and treatment.

As an important spice species in Rutaceae, the Sichuan pepper (Zanthoxylum armatum) can provide pungent and numbing taste, as well as aroma in its mature fruit. Here we assembled a chromosome-level genome of green prickly ash which was widely cultivated in a major production area including Chongqing and Sichuan province, China. We generated 712 Gb (∼112 × ) PacBio long reads and 511 Gb (∼82 × ) Hi-C data, and yielded an assembly of 99 pseudochromosomes with total size of 5.32 Gb and contig N50 of 796 kb. The genomic analyses and cytogenetic experiments both indicated that the cultivar ‘Zhuye Huajiao’ was a triploid. We identified a Zanthoxylum-specific whole genome duplication event emerging about 24.8 million years ago (Mya). We also detected a transposition burst event (0.3–0.4 Mya) responsible for the large genome size of Z. armatum. Metabolomic analysis of the Zanthoxylum fruits during development stages revealed profiles of 39 volatile aroma compounds and 528 secondary metabolites, from which six types of sanshools were identified. Based on metabolomic and transcriptomic network analysis, we screened candidate genes encoding long chain acyl-CoA synthetase, fatty acid desaturase, branched-chain amino acid aminotransferase involved in sanshool biosynthesis and three genes encoding terpene synthase during fruit development. The multi-omics data provide insights into the evolution of Zanthoxylum and molecular basis of numbing and aroma flavor of Sichuan pepper.

Abstract The SHI RELATED SEQUENCE (SRS) family plays a vital role in the development of multiple plant organs such as floral meristem determinacy, organ morphogenesis, and signal transduction. Nevertheless, there is little understanding of the biological significance of tomato SRS family at this point. Our research identified eight SlSRS family members and classified them into three subfamilies based on phylogenetics, conserved motifs, and characteristic domain analysis. The intraspecies and interspecies collinearity analysis revealed clues of SRS family evolution. Many cis-elements related to hormones, stresses, and plant development can be found in the promoter region of SlSRS genes. All of eight SlSRS proteins were located in the nucleus and possessed transcriptional activity, half of which were transcriptional activators, and the other half were transcriptional repressors. Except for SlSRS1, which showed high transcript accumulation in vegetative organs, most SlSRS genes expressed ubiquitously in all flower organs. In addition, all SlSRS genes could significantly respond to at least four different plant hormones. Further, expression of SlSRS genes were regulated by various abiotic stress conditions. In summary, we systematically analyzed and characterized the SlSRS family, reviewed the expression patterns and preliminarily investigated the protein function, and provided essential information for further functional research of the tomato SRS genes in the determination of reproductive floral organs and the development of plants, and possibly other plants.

Flower and fruit abscission is a highly programmed physiological process, which is closely related to the yield of horticultural plants. The coordination of many regulatory factors associated with metabolic and signaling pathways plays a key role in the flower and fruit shedding. Hormones, peptides, carbohydrates, polyamines or cell wall modifying proteins regulate flower and fruit shedding. This article reviewed the recent studies of flower and fruit abscission, including the molecular regulation mechanism of abscission zone formation, typical structure and location of abscission zones, and other factors affecting flower and fruit abscission, such as stresses, hormones, peptides, carbohydrates, polyamines and cell wall modifying proteins. Overall, the review summarizes the developmental mechanism and the diversity of abscission zones, and the key factors affecting flower and fruit abscission of horticultural plants, aiming to provide guidance for studying the molecular regulatory mechanism of flower and fruit abscission.

Fruit development can be viewed as the succession of three main steps consisting of the fruit initiation, growth and ripening. These processes are orchestrated by different factors, notably the successful fertilization of flowers, the environmental conditions and the hormones whose action is coordinated by a large variety of transcription factors. Among the different transcription factor families, TEOSINTE BRANCHED 1, CYCLOIDEA, PROLIFERATING CELL FACTOR (TCP) family has received little attention in the frame of fruit biology despite its large effects on several developmental processes and its action as modulator of different hormonal pathways. In this respect, the comprehension of TCP functions in fruit development remains an incomplete puzzle that needs to be assembled. Building on the abundance of genomic and transcriptomic data, this review aims at collecting available TCP expression data to allow their integration in the light of the different functional genetic studies reported so far. This reveals that several Class I TCP genes, already known for their involvement in the cell proliferation and growth, display significant expression levels in developing fruit, although clear evidence supporting their functional significance in this process remains scarce. The extensive expression data compiled in our study provide convincing elements that shed light on the specific involvement of Class I TCP genes in fruit ripening, once these reproductive organs acquire their mature size. They also emphasize their putative role in the control of specific biological processes such as fruit metabolism and hormonal dialogue.

BackgroundBased on pharmacoeconomics, drug availability and actual treatment, optimal treatment regimens for Chinese non-small-cell lung carcinoma (NSCLC) patients over 70 years old are needed.MethodsThis multicenter, single-arm pilot trial enrolled patients with advanced non-squamous NSCLC who refused systemic chemotherapy. Eligible patients received anlotinib (12 mg/day, d1-14, Q3W) until disease progression, intolerant toxicities, or withdrawal from the study. The primary endpoint was progression-free survival (PFS).ResultsForty-nine patients were screened between January 2019 and September 2021, of whom 40 patients were eligible. The median age was 76 years. With a median follow-up period of 16.20 (95% CI: 8.77, 25.10) months, the median PFS was 5.45 months (95% CI: 3.52-9.23) and the median overall survival was 10.32 months (95% CI: 6.44-12.78). Three patients achieved a partial response and 34 had stable disease, with an objective response rate of 7.5% and a disease control rate of 92.5%. Thirty-three (82.5%; 33/40) patients reported treatment-related adverse events (TRAEs) of any grade, and the incidence rate of grade ≥3 TRAEs was 35% (14/40). The most common grade ≥3 TRAEs were hypertension (4/40; 10.0%), hand-foot syndrome (3/40; 7.5%), and proteinuria (2/40; 5.0%).ConclusionAnlotinib treatment was feasible and safe in Chinese elderly patients with advanced non-squamous NSCLC who did not receive any systemic chemotherapy.

Genetic transformation with mature material as the explants could shorten the transgenic period and avoid seed dependence compared with genetic transformation using the epicotyl seedling stem segments as the receptor. Here, we constructed an Agrobacterium tumefaciens-mediated transformation for generation of marker-free transgenic plants from navel orange (Citrus sinensis Osbeck) mature stems using a Cre-loxP recombination system. To efficiently recover the regenerated buds from mature tissues, five recovery methods were compared: in vitro micrografting of 0.1–0.5 (1–2 weeks), > 0.5 cm (3–4 weeks) and > 1 cm long lignified bud and in vitro micrografting of explants with a bud and rooting regenerated bud. The data showed that in vitro micrografting of > 1 cm long regenerated bud with expanded leaves after one month of continuous culture for lignification was the optimal solution for plant recovery from mature tissues. Transgenic plants without selectable marker genes were created from navel orange (Citrus sinensis Osbeck) tissue using a transformation vector PLI-35SPR1aCB containing a Cre/loxP system recombination together with genes encoding the selectable marker isopentenyl transferase (IPT) and an anti-bacterial peptide (PR1aCB). Using IPT positive selection, the transformation efficiency determined by PCR was 0.9%, and in total, 20 transgenic plants were obtained. Southern blotting confirmed further their transgenicity. PCR and sequencing analysis demonstrated that both the Cre and IPT genes had been successfully removed from the transgenic plants (deletion efficiency 100%). Over all, using Cre/loxP system recombination together with the IPT positive selection, marker-free transgenic plants can be recovered efficiently from mature tissues of navel orange (Citrus sinensis Osbeck), which provides a potential method for production of transgenic plants from citrus mature tissue.

Introduction: During the arms race between plants and pathogens, pathogenesis-related proteins (PR) in host plants play a crucial role in disease resistance, especially PR1. PR1 constitute a secretory peptide family, and their role in plant defense has been widely demonstrated in both hosts and in vitro. However, the mechanisms by which they control host-pathogen interactions and the nature of their targets within the pathogen remain poorly understood. Objectives: The present study was aimed to investigate the anti-oomycete activity of secretory PR1 proteins and elaborate their underlying mechanisms. Methods: This study was conducted in the potato-Phytophthora infestans pathosystem. After being induced by the pathogen infection, the cross-kingdom translocation of secretory PR1 was demonstrated by histochemical assays and western blot, and their targets in P. infestans were identified by yeast-two-hybrid assays, bimolecular fluorescence complementation assays, and co-immunoprecipitation assay. Results: The results showed that the expression of secretory PR1-encoding genes was induced during pathogen infection, and the host could deliver PR1 into P. infestans to inhibit its vegetative growth and pathogenicity. The translocated secretory PR1 targeted the subunits of the AMPK kinase complex in P. infestans, thus affecting the AMPK-driven phosphorylation of downstream target proteins, preventing ROS homeostasis, and down-regulating the expression of RxLR effectors. Conclusion: The results provide novel insights into the molecular function of PR1 in protecting plants against pathogen infection, and uncover a potential target for preventing pre- and post-harvest late blight.

Colonic endoscopy is the gold standard for detecting rectal polyps and rectal cancer. In which polyps are a major predisposing factor for colorectal cancer, the precise diagnosis of polyps within colorectal endoscopy is highly dependent on a physician of professional level. With the development of deep learning, some semantic segmentation methods have recently been applied to polyp detection, but there are problems with insufficient accuracy and segmentation speed. To this end, we propose a precision adaptive global context network (AGCNet) based on real-time colon endoscopy. Firstly, in order to adapt to the problem of large-scale variation of polyps, we designed a multi-scale semantic fusion module (MSFM), which enhances the representation capability by varieties of filters to collect contextual information at different scales, thus adapting to the problem of large variation of polyp size, especially smaller polyps. In addition, modelling long-range dependence by simply using complex spatial pixels tends to introduce more background noise and increase the computational effort. To this end, a context-aware pyramid aggregation module (CPAM) was designed, which internally includes a novel dual attention mechanism whereby the CPAM aggregates feature information across different regions to boost the network’s ability to utilize global context and model long-range dependency through dual attention to further reinforce the features information of important regions and efficiently suppress features in non-important regions. Additionally, the CPAM performs multi-level pooling on the input features to extract multi-scale context information from the image and uses an attention mechanism to selectively highlight informative regions of the image that are most relevant to the segmentation task. The module fuses the multi-level pooled features with the attention map to produce enhanced feature representations that capture both global and local information. Thereby achieving precise polyp segmentation and taking real-time into account. Our proposed AGCNet performed extensive experimental studies on datasets Kvasir-SEG, CVC-ClinicDB, CVC-ColonDB and ETIS-LaribPolypDB. Specifically, AGCNet achieved an IoU of 87.40% and a Dice score of 92.63% on the Kvasir dataset, achieving accurate segmentation results faster than many current state-of-the-art models.

Transcranial magnetic stimulation (TMS) is a noninvasive neuromodulation technique for treating various neurological disorders. The geometry of the TMS coil determines the focality, stimulation field strength, and stimulation depth of the induced electric field. In this paper, we introduce a novel coil, named the eccentric folding coil (EFC). We used Sim4Life, a three-dimensional human tissue medical electromagnetic simulation software, and the brain models of the subjects from the population head model repository to conduct finite element simulations of the eccentric folding coil, circular coil, and figure-eight coil, implying that the largest EFC has a focal area of 19.5 cm2 less than the smallest circular coil, and the EFC of any size has a smaller focal area than the figure-8 coil of the same size. Furthermore, we stimulated the model from three directions with the coil and measured the electric field responses of the model, confirming that stimulation varied in different directions. Additionally, we applied EFC to four representative human models and found that the electric field in different subjects’ brains differed by 3.03 mm in stimulation depth and 12.9 cm2 in focal area, suggesting that personalized human head modeling and electromagnetic analysis are essential for TMS treatment. In the future, we will explore using different types of TMS devices for patients with different conditions to achieve more precise and customized treatment.

Giant landslides and dammed lakes occurred frequently during the prehistoric period; however, their mechanisms often remain an open issue because they are complex. This study used field investigations to observe four old landslides and three landslide dams in Daning County, Shanxi Province, China. Remote sensing images captured during different periods were used to compare the landforms of landslides, landslide dams, and their surrounding environments in detail. Subsequently, their shapes, scales, spatial distributions, and positional relationships were determined. The ages of 14C obtained from the paleolandslide dam lake (PDL) deposits were 3742 ± 95, 4115 ± 121, 6544 ± 91, 7814 ± 109 cal a BP. The 14C ages of the deposits under the sliding surfaces of the old landslides were 2748 ± 27, 4470 ± 54, and 8245 ± 79 cal a BP. Based on the actual physical geographical conditions and geomorphological characteristics of the region, combined with the above age data, that the following conclusions were made: (i) During the Holocene, from 2700 to 8200 years ago, heavy rainfall was the main reason for the occurrence of landslides and landslide dams in the region. (ii) Under the condition of heavy rainfall, the slopes with angles >60° in the Loess Plateau are prone to form landslide masses, and the size of the landslide is related to rainfall intensity. A landslide dam was easily formed when the sliding direction was perpendicular to the direction of the valley. It is difficult to form landslide dams when the sliding direction is almost consistent with the downward direction of the valley. (iii) The formation mechanism of old landslides shows that collapse or sliding may occur suddenly in loess geological masses under an external rainfall scenario. Moreover, the process of landslide dams is related to the geomorphic forms of the original valley.

The blasting quality of open-pit mining can be enhanced and the production cost of stope reduced by establishing a mathematical model for step drilling and blasting costs based on stope consumption. By enhancing the Gray Wolf algorithm, the parameters for step drilling and blasting are optimized, resulting in improved effectiveness for step blasting mining, as demonstrated through modeling and calculation. The enhanced Gray Wolf algorithm effectively enhances the blasting performance, reduces production costs, and increases production efficiency. Taking a limestone mine as an example, the optimized drilling and blasting parameters are as follows: hole spacing of 4.62 m, row spacing of 4 m, and explosive consumption rate of 0.23 kg/t; based on these parameters, the stope’s production cost is reduced to CNY 7.7.

The objective of this study was to investigate the effects and molecular mechanisms of total flavonoids from Cortex Juglandis Mandshuricae (TFC) on preventing alcohol-induced chronic liver injury and regulating gut microbiota in mice. The results showed that oral administration of TFC significantly attenuated alcoholic liver injury in mice. TFC improved lipid accumulation in mice with chronic alcoholic liver injury through activation of the AMPK/PPARα pathway. In addition, TFC maintained the integrity of the intestinal barrier in alcoholic mice, reducing endotoxin leakage from the intestine and further inhibiting the TLR4/NF-κB inflammatory pathway. More importantly, TFC regulated the intestinal microbiota composition and certain bacteria, including Akkermansia muciniphila, Lactobacillus and others. At the same time, reduced levels of short-chain fatty acids due to alcohol consumption were restored. In summary, TFC upregulated AMPK/PPARα signaling pathway to improve hepatic fat accumulation and oxidative stress; TFC positively regulated intestinal flora composition to reduce intestinal disorders caused by alcohol consumption, and further inhibited alcohol-induced inflammatory responses through the intestinal-liver axis. The above findings may be the mechanism of TFC’s pharmacological effects against alcoholic liver injury.

Agriculture is pivotal in national economies, with pest classification significantly influencing food quality and quantity. In recent years, pest classification methods based on deep learning have made progress. However, there are two problems with these methods. One is that there are few multi-scale pest detection algorithms, and they often lack effective global information integration and discriminative feature representation. The other is the lack of high-quality agricultural pest datasets, leading to insufficient training samples. To overcome these two limitations, we propose two methods called RS Transformer (a two-stage region proposal using Swin Transformer) and the Randomly Generated Stable Diffusion Dataset (RGSDD). Firstly, we found that the diffusion model can generate high-resolution images, so we developed a training strategy called the RGSDD, which was used to generate agricultural pest images and was mixed with real datasets for training. Secondly, RS Transformer uses Swin Transformer as the backbone to enhance the ability to extract global features, while reducing the computational burden of the previous Transformer. Finally, we added a region proposal network and ROI Align to form a two-stage training mode. The experimental results on the datasets show that RS Transformer has a better performance than the other models do. The RGSDD helps to improve the training accuracy of the model. Compared with methods of the same type, RS Transformer achieves up to 4.62% of improvement.

The all-flesh type of tomato fruits is caused by mutation of the MBP3 gene, however, knocking down MBP3 in certain genotypes also affect plant and fruit development. Here, the authors show that a natural mutation of AGL11, a close homolog of MBP3, is responsible for the phenotypic divergence.

Long noncoding RNAs (lncRNAs), defined as untranslated and tightly-regulated transcripts with a length exceeding 200 nt, are common outputs of the eukaryotic genome. It is becoming increasingly apparent that many lncRNAs likely serve as important regulators in a variety of biological processes. In particular, some of them accumulate in the nucleus and function in diverse nuclear events, including chromatin remodeling, transcriptional regulation, RNA processing, DNA damage repair, etc. Here, we unite recent progresses on the functions of nuclear lncRNAs and provide insights into the future research directions of this field.

Abstract Background As the key regulators in BR signaling, BES1 family genes regulate thousands of target genes involved in various development processes. So far, the functions of BES1 family are poorly understood in tomato, and a comprehensive genomic and expressional analysis is worth to conduct for this family. Results Here, nine SlBES1 family members were identified in tomato and classified into five groups based on the conserved motif, gene structure and phylogenetic analysis. Synteny among tomato, Arabidopsis, pepper and rice were further analyzed to obtain insights into evolutionary characteristics. Several cis-elements related to hormone, stress and plant development were exhibited in the promoter regions of SlBES1 family genes. Subcellular localization showed seven members localized both in the nucleus and cytoplasm, implying the presence of dephosphorylated and phosphorylated form of these seven proteins, furthermore, five of them possessed transcription activation activity whereas the left two functioned as transcriptional repressors. Another two members, however, neither localized in the nucleus nor had transactivation activity. Besides, SlBES1.8 showed flower-specific expression while other members expressed ubiquitously in all organs. Moreover, SlBES1 genes exhibited variational expression in response to nine principal plant hormones. Notably, the expression levels of SlBES1 genes presented a dominant downregulated trend in response to stresses. Conclusions In this study, we systematically analyzed the genomic characterization of SlBES1 family, together with the analyses of protein functional features and expression patterns, our results lay a foundation for the functional research of SlBES1 family.

A rare actinomycetes strain of Saccharothrix texasensis, strain 6-C, has been isolated from the potato rhizosphere and it was shown to act as a biological control agent to potato late blight. It is also the first report on Saccharothrix spp. inhibiting Phytophthora infestans. Here, we present the complete genome data of S. texasensis strain 6-C, assembled by sequencing reads obtained by both PacBio and Illumina technologies with annotation. The final assembled genome length is 9,045,220 bp, without gaps and plasmid, and its GC content is 72.39%. Nine nonribosomal peptides synthetase, five type I polyketide synthase, four terpene, and three lanthipeptide gene clusters were identified in the genome, which would be likely to encode lots of antimicrobial active substances to help host plants against disease. This genome sequence could contribute to investigations of the molecular basis underlying the biocontrol activity of this Saccharothrix strain.

Nanoparticles have been found to possess unique advantages in many fields, especially in the field of cancer treatment. Herein, based on the unique physical and chemical properties of natural polysaccharides, the polysaccharide from the edible and medicinal fruits of Chaenomeles speciosa was prepared, and the complex nanoparticles constructed by combining C. speciosa polysaccharide with selenium have been successfully developed by a chemical method. Monodisperse spherical nanoparticles with the particle size of 80.5 nm were characterized by various methods, which exhibited ideal size distribution and prominent stability under physiological conditions and alkaline conditions. Cellular studies demonstrated the nanoparticles significantly inhibited the growth of MCF-7 cells with an IC50 value of 8.37 ± 0.97 μg/mL through inducing the apoptosis and arresting the cell circle at S phase. Moreover, the zebrafish assays confirmed the antitumor effects of the nanoparticles, which suppressed the proliferation and migration of tumor and blocked the angiogenesis of transgenic zebrafish. Collectively, the results suggested that the nanoparticles may be considered as a candidate agent to treat breast cancer.

Colorectal cancer, also known as rectal cancer, is one of the most common forms of cancer, and it can be completely cured with early diagnosis. The most effective and objective method of screening and diagnosis is colonoscopy. Polyp segmentation plays a crucial role in the diagnosis and treatment of diseases related to the digestive system, providing doctors with detailed auxiliary boundary information during clinical analysis. To this end, we propose a novel light-weight feature refining and context-guided network (FRCNet) for real-time polyp segmentation. In this method, we first employed the enhanced context-calibrated module to extract the most discriminative features by developing long-range spatial dependence through a context-calibrated operation. This operation is helpful to alleviate the interference of background noise and effectively distinguish the target polyps from the background. Furthermore, we designed the progressive context-aware fusion module to dynamically capture multi-scale polyps by collecting multi-range context information. Finally, the multi-scale pyramid aggregation module was used to learn more representative features, and these features were fused to refine the segmented results. Extensive experiments on the Kvasir, ClinicDB, ColonDB, ETIS, and Endoscene datasets demonstrated the effectiveness of the proposed model. Specifically, FRCNet achieves an mIoU of 84.9% and mDice score of 91.5% on the Kvasir dataset with a model size of only 0.78 M parameters, outperforming state-of-the-art methods. Background Circular RNAs (circRNAs) have been closely implicated in competing endogenous RNA (ceRNA) network among human cancers including non‐small cell lung cancer (NSCLC). However, the role of most circRNAs in NSCLC remains to be determined. Here, we aimed to investigate the role of hsa_circ_0007385 (circ_0007385) in NSCLC cells. Methods Expression of hsa_circ_0007385 (circ_0007385), miRNA (miR)‐519d‐5p and high‐mobility group box 1 (HMGB1) was measured by real‐time quantitative PCR and western blotting. Functional experiments were evaluated by cell counting kit (CCK)‐8, flow cytometry, fluorescein active caspase‐3 staining kit, transwell assays, western blotting, and xenograft experiment. The relationship among circ_0007385,miR‐519d‐5p and HMGB1 was testified by dual‐luciferase reporter assay. Kaplan‐Meiersurvival curve identified overall survival in NSCLC patients. Results circ_0007385 expression was higher in NSCLC tissues and cell lines, and was associated with poor overall survival. Silencing circ_0007385 could suppress cell proliferation, migration and invasion in A549 and H1975 cells, as well as cisplatin (DDP) resistance. Moreover, circ_0007385 silence retarded tumor growth of A549 cells in vivo. Molecularly, there was a direct interaction between miR‐519d‐3p and either circ_0007385 or HMGB1; expression of miR‐519d‐3p was downregulated in NSCLC tumors in a circ_0007385‐correlated manner, and circ_0007385 could indirectly regulate HMGB1 via miR‐519d‐3p. Functionally, both inhibiting miR‐519d‐3p and restoring HMGB1 could overturn the suppressive effect of circ_0007385 knockdown on cell proliferation, migration, invasion, and DDP resistance. Conclusions Collectively, circ_0007385 deletion could function anti‐tumor role in NSCLC by suppressing malignant behaviors and DDP resistance in vitro and in vivo via circ_0007385/miR‐519d‐3p/HMGB1 axis. These outcomes might enhance our understanding of the molecular mechanisms underlying the malignant progression of NSCLC. Key points Significant findings of the study circ_0007385 was upregulated in NSCLC tissues and cells, and was associated with poor overall survival. Silenced circ_0007385 suppressed NSCLC cell proliferation, migration, invasion, and DDP resistance in vitro, and tumor growth in vivo. circ_0007385 was upregulated in NSCLC tissues and cells, and was associated with poor overall survival. What this study adds miR‐519d‐3p could directly interact with circ_0007385 and HMGB1 in NSCLC cells. A promising circ_0007385/miR‐519d‐3p/HMGB1 regulatory pathway was determined in NSCLC cells.

Various risk management measures have been applied to reduce risks associated with the debris flow; however, only a few studies have adopted the economic benefit to evaluate measure effectiveness. The present study sought to explore debris flow risks at a catchment scale and establish the appropriate risk-reducing measures. The Chengbei Gully debris flow in Shanxi province (China) was selected for the case study. High-resolution topographic data of the drainage basin were obtained using the airborne LiDAR technology. FLO-2D software was used to simulate the debris flow process to perform hazard zonation. Vulnerability was estimated based on the location of elements at risk within the hazard zones and the field survey. Several structural and non-structural measures for controlling risks were proposed based on the risk assessment results, and the benefit–cost ratio was used to analyze their effectiveness. The findings indicated that the rainfall event triggering the Chengbei Gully debris flow had an 80-year return period. The total risk under this rainfall condition was 2.3 × 105 $, which was an unacceptable level according to the criteria of tolerance risk. The findings showed that the engineering measure was the best mitigation approach for the Chengbei Gully debris flow with a benefit of 1.35 million $ and a benefit–cost ratio of 6.43.

For the purpose of uncovering the influence law on the explosion stress wave transmission and blast-induced crack propagation by inclined natural fracture and water jet slot, the influential mechanism for the transmission law of explosion stress wave due to inclined natural fracture was analyzed based on stress wave theory in this article. Next, the evolution law of explosion stress wave and blast-induced crack propagation affected by inclined natural fracture and water jet slot were simulated by ANSYS/LS-DYNA, and an experiment was conducted to understand the crack propagation law under explosion shock wave loading. The results indicate that the existence of inclined natural fracture will affect the “guiding effect” of water jet slot on blast-induced crack propagation by changing the propagation path and mechanical properties of the explosion stress wave. When the distance between inclined natural fracture and blast hole is too small, the water jet slot cannot play a guiding role in propagation process of blast-induced main crack. With the increase of the distance, the propagation direction of blast-induced crack is affected by both inclined natural fracture and water jet slot. When the distance increases further and exceeds a certain value, inclined natural fracture cannot affect the guiding effect of water jet slot in propagation process of blast-induced main crack.

Botrytis cinerea is one of the most destructive fungal pathogens causing tremendous losses in fresh fruit or vegetables. 3-Methylthio-1-propanol (3-MP) is a naturally occurring food-borne sulfide, which is mainly used to increase the flavor in food. However, the potential application of 3-MP in the postharvest phase to manage fruit fungal diseases has not been explored. In this study, the antifungal activity of 3-MP against B. cinerea was evaluated, and the possible mechanism involved was explored. In vitro 3-MP treatment could effectively inhibit the mycelial growth, spore germination, and germ tube elongation of B. cinerea. 3-MP also impaired the spore viability and membrane integrity of B. cinerea as well as increased the leakage of nucleic acids, proteins, and malondialdehyde (MDA) in B. cinerea. In vivo 3-MP fumigation treatment inhibited the infection of B. cinerea on tomato fruits. Also, the fruits with 3-MP fumigation treatment exhibited higher antioxidant enzyme activity, lower MDA content, and a significant delay of induction of the expression of most of the stress-related genes when compared to the control group. Moreover, a cytotoxicity evaluation revealed that 3-MP had no toxicity to normal cells in a certain concentration range. Collectively, our research results will provide evidence for the development of food-borne sulfide 3-MP as a fungicide in food and agriculture and will provide an important reference for the formulation of B. cinerea biocontrol strategies.

Existing multi-scale exposure fusion (MEF) algorithms which focus on defining weight maps to extract reliable information. However, they are difficult to preserve brightness order among over-exposed regions of a bright image and under-exposed regions of a dark image in a fused image when input images are two large-exposure-ratio images. In this paper, a new concept of exposure interpolation, which generates an appropriate bracketed sequence of virtual images from two input images, is introduced to address the problem. The possibly minimal number of virtual images is determined by using an optimization method. Then, an effective method by employing the intensity mapping function (IMF) is proposed to generate virtual images with proportional exposure times. The final image is produced by fusing both the two input images and the virtual images using multi-scale fusion. Experimental results show that the brightness order is preserved much better and the MEF-SSIM is significantly improved by the exposure interpolation.

It is challenging to align multi-exposed images due to large illumination variations, especially in presence of saturated regions. In this paper, a novel image alignment algorithm is proposed to cope with the multi-exposed images with saturated regions. Specifically, the multi-exposed images are first normalized by using intensity mapping functions (IMFs) in consideration of saturated pixels. Then, the normalized images are coded by using the local binary pattern (LBP). Finally, the coded images are aligned by formulating an optimization problem by using a differentiable Hamming distance. Experimental results show that the proposed algorithm outperforms state-of-the-art alignment methods for multi-exposed images in terms of alignment accuracy and robustness to exposure values.

Abstract Auxin response factors (ARFs) are involved in auxin-mediated transcriptional regulation in plants. In this study, we performed functional characterization of SlARF6A in tomato. SlARF6A is located in the nucleus and exhibits transcriptional activator activity. Overexpression of SlARF6A increased chlorophyll contents in the fruits and leaves of tomato plants, whereas downregulation of SlARF6A decreased chlorophyll contents compared with those of wild-type (WT) plants. Analysis of chloroplasts using transmission electron microscopy indicated increased sizes of chloroplasts in SlARF6A-overexpressing plants and decreased numbers of chloroplasts in SlARF6A-downregulated plants. Overexpression of SlARF6A increased the photosynthesis rate and accumulation of starch and soluble sugars, whereas knockdown of SlARF6A resulted in opposite phenotypes in tomato leaves and fruits. RNA-sequence analysis showed that regulation of SlARF6A expression altered the expression of genes involved in chlorophyll metabolism, photosynthesis and sugar metabolism. SlARF6A directly bound to the promoters of SlGLK1, CAB, and RbcS genes and positively regulated the expression of these genes. Overexpression of SlARF6A also inhibited fruit ripening and ethylene production, whereas downregulation of SlARF6A increased fruit ripening and ethylene production. SlARF6A directly bound to the SAMS1 promoter and negatively regulated SAMS1 expression. Taken together, these results expand our understanding of ARFs with regard to photosynthesis, sugar accumulation and fruit development and provide a potential target for genetic engineering to improve fruit nutrition in horticulture crops.

Infrared (IR) band sensors can capture digital images under challenging conditions, such as haze, smoke, and fog, while visible (VIS) band sensors seize abundant texture information. It is desired to fuse IR and VIS images to generate a more informative image. In this paper, a novel multi-scale IR and VIS images fusion algorithm is proposed to integrate information from both the images into the fused image and preserve the color of the VIS image. A content-adaptive gamma correction is first introduced to stretch the IR images by using one of the simplest edge-preserving filters, which alleviates excessive luminance shifts and color distortions in the fused images. New contrast and exposedness measures are then introduced for the stretched IR and VIS images to achieve weight matrices that are more in line with their characteristics. The IR and luminance components of the VIS image in grayscale or RGB space are fused by using the Gaussian and Laplacian pyramids. The RGB components of the VIS image are finally expanded to generate the fused image if necessary. Comparisons experimentally demonstrate the effectiveness of the proposed algorithm to 10 different state-of-the-art fusion algorithms in terms of computational cost and quality of the fused images.

Ascorbic acid, also known as vitamin C, is a vital antioxidant widely found in plants. Plant fruits are rich in ascorbic acid and are the primary source of human intake of ascorbic acid. Ascorbic acid affects fruit ripening and stress resistance and plays an essential regulatory role in fruit development and postharvest storage. The ascorbic acid metabolic pathway in plants has been extensively studied. Ascorbic acid accumulation in fruits can be effectively regulated by genetic engineering technology. The accumulation of ascorbic acid in fruits is regulated by transcription factors, protein interactions, phytohormones, and environmental factors, but the research on the regulatory mechanism is still relatively weak. This paper systematically reviews the regulation mechanism of ascorbic acid metabolism in fruits in recent decades. It provides a rich theoretical basis for an in-depth study of the critical role of ascorbic acid in fruits and the cultivation of fruits rich in ascorbic acid.

Genetics: one small tweak, one giant leap for tomatoes A single gene has been identified in tomatoes that regulates multiple aspects of fruit quality, including levels of health-promoting anthocyanins, opening the door to engineering more nutritious and better-tasting tomatoes. Fruit breeders have long-manipulated genes to enhance traits like yield or disease resistance, but it is rare to find a single gene that can improve multiple aspects of fruit quality. In this study, Zhengguo Li at Chongquing University in China and colleagues show that over-expressing a transcription factor called SIMYB75 results in striking deep purple tomatoes enriched in anthrocyanins–antioxidants thought to be protective against various diseases - which tomatoes are usually devoid of. They also had higher levels of other health-promoting phytochemicals and enhanced production of aroma volatiles, which can influence the taste and flavor of fruit.

Genetic engineering: Quick and easy CRISPR in tomato Researchers in China have created a toolkit to easily mutate genes in tomato using the CRISPR gene modification technology. Zhengguo Li of Chongqing University developed the system in response to the lack of an open, standardized, reliable gene editing tool in tomato. The toolkit is designed according to an open-source, modular standard known as Biobrick. It consists of several molecular building blocks which can be used to target up to eight genes for editing or deletion. The kit also includes software to help researchers determine the DNA sequences necessary to copy genes of interest and prepare them for the CRISPR reactions. Li’s team tested the toolkit by using it to successfully simultaneously mutate multiple genes involved in tomato ripening. The new toolkit will enable researchers to rapidly and easily manipulate genes in this important crop.

Cadmium (Cd), a toxic heavy metal, affects the yield and quality of crops. Wheat (Triticum aestivum L.) can accumulate high Cd content in the grain, which poses a major worldwide hazard to human health. Advances in our understanding of Cd toxicity for plants and humans, different parameters influencing Cd uptake and accumulation, as well as phytoremediation technologies to relieve Cd pollution in wheat have been made very recently. In particular, the molecular mechanisms of wheat under Cd stress have been increasingly recognized. In this review, we focus on the recently described omics and functional genes uncovering Cd stress, as well as different mitigation strategies to reduce Cd toxicity in wheat.

Abstract Background Lycopene is an important carotenoid pigment in red fruits and vegetables, especially in tomato. Although lycopene biosynthesis and catabolism have been found to be regulated by multiple factors including phytohormones, little is known about their regulatory mechanism. Cytokinins are crucial to various aspects of plant growth. Isopentenyltransferases (IPTs) catalyze the initial rate-limiting step of cytokinins biosynthesis, however, their roles in fruit ripening remain unclear. Results Here, the functions of SlIPT4, encoding an isopentenyltransferase, were characterized via RNAi-mediated gene silencing in tomato. As we expected, silencing of SlIPT4 expression resulted in accelerated leaf senescence. However, down-expression of SlIPT4 generated never-red orange fruits, corresponding with a dramatic reduction of lycopene. Among lycopene biosynthesis-related genes, the fact of remarkable decrease of ZISO transcript and upregulation of other genes, revealed that SlIPT4 regulates positively lycopene biosynthesis via directly affecting ZISO expression, and also supported the existence of regulatory loops in lycopene biosynthesis pathway. Meanwhile, the accumulation of abscisic acid (ABA) was reduced and the transcripts PSY1 were increased in SlIPT4-RNAi fruits, supporting the feedback regulation between ABA and lycopene biosynthesis. Conclusion The study revealed the crucial roles of SlIPT4 in leaf senescence and the regulatory network of lycopene biosynthesis in tomato, providing a new light on the lycopene biosynthesis and fruit ripening.

Environmental stresses, such as temperature, heavy metals, drought, cold, and microbial infections adversely damage various aspects of plant growth and development. Salinity and drought are among major hazardous factors, which adversity affects plant growth and productivity. Transcription factors, such as basic helix-loop-helix play critical roles in regulating plant physiological processes under abiotic stresses. In this study, we presented the characterization of a tomato SlbHLH22 gene under abiotic stresses such as drought and salinity. Plants overexpressing SlbHLH22 showed short height with small leaves and enhanced flavonoid accumulation. In wild type (WT) plant, the elevated levels of SlbHLH22 were detected under salt and D-mannitol stresses. Subcellular localization analysis revealed that SlbHLH22 protein was targeted to the nucleus in onion epidermal cells. Transactivation assay in yeast demonstrated that SlbHLH22 had transcriptional activation ability. The transgenic plants overexpressing SlbHLH22 displayed enhanced vigor and more tolerant to drought and salinity than WT. Overexpression of SlbHLH22 significantly peaked the activities of catalase (CAT), superoxide dismutase (SOD), and peroxidase (POD) to minimize the impacts of reactive oxygen species such as H2O2, which was reduced significantly in transgenic plants along with Malondialdehyde (MDA). Moreover, the expression levels of ROS defense genes (SlPOD, SlCAT, SlSOD), ABA biosynthesis genes, proline biosynthesis, and flavonoids synthesis genes were also activated under salinity and drought. Taken together, our study implies that the overexpression of SlbHLH22 improved tomato plant stress resistance by improving ROS scavenging system, increasing osmotic potential and enhanced accumulation of secondary metabolites in tomato plants.

The structural index is an important quantitative parameter for revealing the structural properties of loess. However, there is no a widely accepted measurement method for structural index at present. This study aims at presenting a novel method for obtaining the loess structural index (LSI), based on the application of computed tomography (CT) scanning techniques and laboratory physico-mechanical tests. The mountainous area of Lvliang in northwest China was taken as the study area, and Late Pleistocene loess samples were taken from various sites in the region. Several physical parameters were first measured using laboratory tests, including dry density, pore ratio, and liquidity index. CT scanning was used to observe sample microstructures, and a mathematical relationship was established between CT image parameters and the physical property indices, through three dimensions (3D) reconstruction and slice porosity analysis. The results revealed that LSI can be expressed as a non-linear function related to CT image parameters, dry density, and the liquidity index of the loess. Compared with traditional calculation methods, this novel technique calculates the LSI by using an empirical formula, which is less labor-intensive. Such results indicate that the method warrants wide application in the future.

The harmonic reducer, with its advantages of high precision, low noise, light weight, and high speed ratio, has been widely used in aerospace solar wing deployment mechanisms, antenna pointing mechanisms, robot joints, and other precision transmission fields. Accurately predicting the performance of the harmonic reducer under various application conditions is of great significance to the high reliability and long life of the harmonic reducer. In this paper, a set of automatic harmonic reducer performance test systems is designed. By using the CANOpen bus interface to control the servo motor as the drive motor, through accurately controlling the motor speed and rotation angle, collecting the angle, torque, and current in real time, the life cycle test of space harmonic reducer was carried out in high vacuum and low temperature environment on the ground. Then, the collected data were automatically analyzed and calculated. The test data of the transmission accuracy, backlash, and transmission efficiency of the space harmonic reducer were obtained. It is proven by experiments that the performance data of the harmonic reducer in space work can be more accurately obtained by using the test system mentioned in this paper, which is convenient for further research on related lubricating materials.

Repetitive operation is extensively present in the inverter compressor refrigeration field. Research illustrates that an approximate periodic load disturbance deteriorates its self-sensing and efficiency in a low frequency range. According to an internal model theorem, a repetitive controller can realize periodic signal tracking or suppression; however, ideal repetitive control (IRC) can not be directly employed in practical application owing to its excessive sensitivity to period-time uncertainties or aperiodic component, etc. In addition, the ratio between period delay to sampling time can not always maintain an integer which causes the resonant frequencies to deviate from the interested. Even if the ratio is a fixed integer, the auxiliary function for stabilization in a conventional repetitive controller can result in the steady state tracking error. In this paper, a revised repetitive controller with a novel Savitzky–Golay (S–G) filter is proposed, the S–G filter cascaded with a traditional delay element can approximate the delay element with any ratio. Aiming at a different operating condition, a second order generalized integrator (SOGI) based observer is employed to extract disturbance fundamental component, and Steiglitz–McBride based adaptive notch filter (SM-ANF) is adopted for frequency estimation. The proposed scheme varies the S–G filter according to variable fundamental frequency and maintains its resonant frequencies corresponding with the fundamental and harmonic disturbance. In the end, simulations and experiments are implemented and results illustrate that speed fluctuation can be effectively suppressed by the proposed method.

Repetitive operations have been extensively used in the inverter compressor refrigeration industry. The approximately periodic disturbance caused by repetitive operations must be compensated to realize stable and high-efficiency operation. In this paper, a periodic disturbance observer (PDOB) is proposed to tackle the speed fluctuation of an inverter compressor in the low-frequency range. Periodic disturbance, consisting of a fundamental wave and corresponding harmonics, can thus be estimated and compensated; in addition, sensitivity and complementary sensitivity can reach a compromise through the use of a certain parameter. Aiming at a different operation environment, an adaptive notch filter based on the Steiglitz–McBride method is employed to estimate the fundamental frequency of periodic disturbance. Finally, the feasibility of our approach is verified by MATLAB simulation, and experiments are implemented to illustrate that speed fluctuation can be more effectively attenuated by the proposed method in comparison with general DOB.

Capturing high-quality images via mobile devices in low-light or backlighting conditions is very challenging. In this paper, a new, single image brightening algorithm is proposed to enhance an image captured in low-light conditions. Two virtual images with larger exposure times are generated to increase brightness and enhance fine details of the underexposed regions. In order to reduce the brightness change, the virtual images are generated via intensity mapping functions (IMFs) which are computed using available camera response functions (CRFs). To avoid possible color distortion in the virtual image due to one-to-many mapping, a least square minimization problem is formulated to determine brightening factors for all pixels in the underexposed regions. In addition, an edge-preserving smoothing technique is adopted to avoid noise in the underexposed regions from being amplified in the virtual images. The final brightened image is obtained by fusing the original image and two virtual images via a gradient domain guided image filtering (GGIF) based multiscale exposure fusion (MEF) with properly defined weights for all the images. Experimental results show that the relative brightness and color are preserved better by the proposed algorithm. The details in bright regions are also preserved well in the final image. The proposed algorithm is expected to be useful for computational photography on smart phones.

Abiotic stresses remain the key environmental issues that reduce plant development and therefore affect crop production. Transcription factors, such as the GRAS family, are involved in various functions of abiotic stresses and plant growth. The GRAS family of tomato (Solanum lycopersicum), SlGRAS7, is described in this study. We produced overexpressing SlGARS7 plants to learn more about the GRAS transcription factors. Plants overexpressing SlGARS7 (SlGRAS7-OE) showed multiple phenotypes related to many behaviors, including plant height, root and shoot length, and flowering time. We observed that many genes in the SlGRAS7-OE seedlings that are associated with auxin and gibberellin (GA) are downregulated and have altered sensitivity to GA3/IAA. SlGRAS7 was upregulated during abiotic stresses following treatment with sodium chloride (NaCl) and D-mannitol in the wild-type (WT) tomato. Tomato plants overexpressing SlGRAS7 showed more resistance to drought and salt stress comparison with WT. Our study of SlGRAS7 in tomato demonstrates how GRAS showed an integrative role, improving resistance to abiotic stresses and enhancing gibberellin/auxin signaling through reproductive as well as vegetative processes.

As the major postharvest disease of citrus fruit, postharvest green mold is caused by the necrotrophic fungus Penicillium digitatum (Pd), which leads to huge economic losses worldwide. Fungicides are still the main method currently used to control postharvest green mold in citrus fruit storage. Investigating molecular mechanisms of plant−pathogen interactions, including pathogenicity and plant resistance, is crucial for developing novel and safer strategies for effectively controlling plant diseases. Despite fruit−pathogen interactions remaining relatively unexplored compared with well-studied leaf−pathogen interactions, progress has occurred in the citrus fruit−Pd interaction in recent years, mainly due to their genome sequencing and establishment or optimization of their genetic transformation systems. Recent advances in Pd pathogenicity on citrus fruit and fruit resistance against Pd infection are summarized in this review.

Phytophthora infestans (P. infestans) is the causal agent of potato late blight, which caused the devastating Irish Potato Famine during 1845-1852. Until now, potato late blight is still the most serious threat to potato growth and has caused significant economic losses worldwide. Melatonin can induce plant innate immunity against pathogen infection, but the direct effects of melatonin on plant pathogens are poorly understood. In this study, we investigated the direct effects of melatonin on P. infestans. Exogenous melatonin significantly attenuated the potato late blight by inhibiting mycelial growth, changing cell ultrastructure, and reducing stress tolerance of P. infestans. Notably, synergistic anti-fungal effects of melatonin with fungicides on P. infestans suggest that melatonin could reduce the dose levels and enhance the efficacy of fungicide against potato late blight. A transcriptome analysis was carried out to mine downstream genes whose expression levels were affected by melatonin. The analysis of the transcriptome suggests that 66 differentially expressed genes involved in amino acid metabolic processes were significantly affected by melatonin. Moreover, the differentially expressed genes associated with stress tolerance, fungicide resistance, and virulence were also affected. These findings contribute to a new understanding of the direct functions of the melatonin on P. infestans and provide a potential ecofriendly biocontrol approach using a melatonin-based paradigm and application to prevent potato late blight.
