1. Characterization of the biological and transcriptomic landscapes of bone marrow-derived mesenchymal stem cells in patients with multiple myeloma

Abstract

Abstract Background Mesenchymal stem/stromal cells (MSCs) have been acknowledged as the most important stromal cells in the bone marrow (BM) microenvironment for physiologic hematopoiesis and the concomitant hematologic malignancies. However, the systematic and detailed dissection of the biological and transcriptomic signatures of BM-MSCs in multiple myeloma (MM) are largely unknown. Methods In this study, we isolated and identified BM-MSCs from 10 primary MM patients and 10 healthy donors (HD). On the one hand, we compared the multifaceted biological characteristics of the indicated two BM-MSCs, including biomarker expression pattern, multilineage differentiation potential, stemness and karyotyping, together with the cellular vitality and immunosuppressive property. On the other hand, we took advantage of RNA-SEQ and bioinformatics analysis to verify the similarities and differences at the transcriptomic level between MM-MSCs and HD-MSCs. Results As to biological phenotypes and biofunctions, MM-MSCs revealed conservation in immunophenotype, stemness and differentiation towards adipocytes and chondrocytes with HD-MSCs, whereas with impaired osteogenic differentiation potential, cellular vitality and immunosuppressive property. As to transcriptomic properties, MM-MSCs revealed multidimensional alterations in gene expression profiling and genetic variations. Conclusions Overall, our date systematic and detailed reflected the multifaceted similarities and variations between MM-MSCs and HD-MSCs both at the cellular and molecular levels, and in particular, the alterations of immunomodulation and cellular viability of MM-MSCs, which wound benefit the further exploration of the pathogenesis and new drug application (NDA) of multiple myeloma from the view of BM-MSCs.

Abstract

Tuning space mapping (TSM) with tuning exponent parameter of ABCD-Matrix is proposed and verified. A section of design interest in the electromagnetic (EM) model is replaced by ‘n’ pieces cascaded ABCD-Matrixes, and each ABCD-Matrix is the EM-simulated ABCD-parameter of pre-assigned cell circuit (PCC). Finally the optimal exponent parameter of ABCD-Matrix is transferred to original optimization variables. The proposed TSM not only has the advantages of circuit tuning element-less (CTEL) TSM in prior art but also clears the number range limitation that the tuning parameter must be positive for tuning in CTEL TSM. The proposed method has the second minimal specification error and lowest simulation time comparing with other TSM methods. Verification examples and comparisons are also implemented.

Abstract

A compact lumped-element low temperature co-fired ceramic (LTCC) quadrature hybrid with harmonics suppression is proposed. The proposed topology enables significant circuit size reduction using spiral inductors with orthogonal input/output ports and vertical interdigital capacitors (VICs) placed in center of the hybrid. The overall size of the LTCC hybrid is only 11 × 11 × 1 mm. Electromagnetic (EM) — simulated S 11 , S 21 , S 31 and S 41 are better than −25, −2.8, −3.9 and −25 dB at 130 MHz, respectively. And the phase balance between the coupled and direct ports is 89.9° at 130 MHz. Moreover, 24 dB suppression for the second harmonic and 36 dB suppression for the third harmonic are obtained.

Abstract

A wide upper stopband and nonorthogonal input/output (I/O) feed dual-mode bandpass filter (BPF) is proposed. The BPF is implemented in a 2-layer low temperature co-fired ceramic (LTCC) substrate and the wide upper stopband is achieved using broadside parallel-coupled lines with stepped impedance feed lines. 40 dB suppression of a 2nd harmonic (2f 0 ) and a prediction of 20 dB suppression up to 2.4f 0 are achieved. Furthermore, yield analysis, performance comparisons and discussions are also implemented.
