Abstract Ring rot, caused by Botryosphaeria dothidea, is an important fungal disease of pear fruit during postharvest storage. Melatonin, as a plant growth regulator, plays an important role in enhancing the stress resistance of pear fruits. It enhances the resistance of pear fruits to ring rot by enhancing their antioxidant capacity. However, the underlying mechanism remains unclear. In this study, we examined the effect of melatonin on the growth of B. dothidea. Results showed that melatonin did not limit the growth of B. dothidea during in vitro culture. However, metabolomics and transcriptomics analyses of ‘Whangkeumbae’ pear (Pyrus pyrifolia) revealed that melatonin increased the activity of antioxidant enzymes, including peroxidase (POD), superoxide dismutase (SOD), and polyphenol oxidase (PPO), in the fruit and activated the phenylpropanoid metabolic pathway to improve fruit resistance. Furthermore, melatonin treatment significantly increased the contents of jasmonic acid and phlorizin in pear fruit, both of which could improve disease resistance. Jasmonic acid regulates melatonin synthesis and can also promote phlorizin synthesis, ultimately improving the resistance of pear fruit to ring rot. In summary, the interaction between melatonin and jasmonic acid and phlorizin enhances the antioxidant defense response and phenylpropanoid metabolism pathway of pear fruit, thereby enhancing the resistance of pear fruit to ring rot disease. Our results provide new insights into the application of melatonin in the resistance to pear fruit ring rot.

The lack of a suitable rootstock to control scion growth has limited the development of high-density plantations in pear production, which is partly attributed to poor understanding of the dwarfing mechanism. In the present study, the rootstock of the dwarf-type pear (Pyrus betulaefolia) ‘PY-9’ was identified and used as the material for anatomical analysis. ‘PY-9’ grew to half the tree height of the normal cultivar ‘Zhengdu’, along with fewer internodes and shorter length. Significant differences in growth rate between ‘PY-9’ and ‘Zhengdu’ were detected at approximately 30 days after full bloom, which corresponded with the time of the greatest difference in water potential between the dwarf and normal cultivar. ‘PY-9’ showed a higher photosynthetic rate than ‘Zhengdu’. Anatomical analysis showed that ‘PY-9’ had higher area ratios of both phloem and xylem and more developed vascular tissues than ‘Zhengdu’. The three-dimensional reconstructed skeleton of the xylem from X-ray computed tomography scanning revealed greater intervessel connectivity in ‘Zhengdu’ than in ‘PY-9’, which could contribute to the more vigorous growth of ‘Zhengdu’. This study thus provides the first comparison of the microstructural properties of xylem elements between a dwarfing-type and vigorous-type pear rootstock, providing new insights into the dwarfing mechanism in pear and facilitating breeding of dwarf pear rootstocks to increase crop productivity.

Diabetic wounds are difficult to heal due to hampered vascularization and tissue regeneration. Wound dressings need frequent replacement and show limited effects on guiding tissue repair. Here, bio-inks containing sodium alginate (SA), oxidized sodium alginate (OSA), gelatin (Gel) and CaCO3 microspheres were prepared. To increase the printability, all bio-inks were pre-crosslinked with calcium ions (Ca2+). Porous hydrogel scaffolds with 4.43 ± 0.14 μm2 pore area and 184 ± 25 μm line diameter were fabricated via 3D printing. After lyophilization and swelling in PBS (pH 6.4), SA/OSA/Gel scaffold showed more excellent structural stability than SA and SA/Gel, which was attributed to the Schiff base reaction between OSA and Gel. NIH-3T3 cells on SA/OSA/Gel proliferated faster and showed better spreading morphology than those on SA and SA/Gel. After placed onto full-thickness wounds on SD rat back, SA/OSA/Gel scaffold guided tissue growth, integrated well with the regenerated tissue and accelerated wound healing, which promoted angiogenesis and showed 93.0 ± 2.5% of collagen deposition between degraded fragments of hydrogel scaffold. Taken together, porous hydrogel scaffolds fabricated via 3D printing bio-inks composed of SA, OSA, Gel and CaCO3 provide a potential strategy to improve diabetic wound healing.

Grapevine is one of the most economically important fruit crops cultivated worldwide. However, grapevine is highly susceptible to virus infections and exposed to the most diverse forms of viral diseases compared to other fruit crops, and virus-induced incompatibility affects plant growth to different degrees ranging from decline to death. The influence of virus-induced incompatibility could be mitigated to an acceptable level by using appropriate rootstocks. However, the viral tolerance of various grapevine rootstocks with diverse genetic backgrounds remains unclear, along with the identification of the specific viral tolerance factors. In this study, the viral tolerance of 21 grapevine rootstocks was evaluated in a green grafting system. Cabernet Franc varieties infected with a single virus [grapevine leafroll associated virus-1 (GLRaV-1)], a co-infection of two viruses (GLRaV-1 plus grapevine virus A—GVA), and no infection were used as the scions, respectively. The vegetative growth and photosynthetic function of the grafts were analyzed 4 months after grafting. The results indicated that some rootstocks could alleviate the influence of the virus infection, with vegetative growth and photosynthetic function sustained at a normal level, whereas other rootstocks were susceptible to the virus infection, resulting in a decline in the growth and photosynthetic function of the grafts. Our research provides evidence for the existence and diversity of viral tolerance among grapevine rootstocks, offering important information for appropriate rootstock selection in the establishment of new vineyards and in the breeding of grapevine rootstocks with enhanced viral tolerance.

Cork spot disorder has affected the fruit of Asian pear since the 1990s and has become serious in recent years with increasingly affected cultivars and areas. The commodity value of affected fruit is greatly decreased, resulting in severe economic losses. Cork spot disorder of pear fruit is a physiological disorder, and the factors responsible are relatively complex. Research on the cause of cork spot disorder is still at an early stage and, thus, further investigations are needed to elucidate the underlying mechanism of the disorder. In this review, current knowledge of the factors associated with the incidence of cork spot disorder in Asian pear fruit is summarized, including fruit growth and development, fruit nutrient status, and environmental factors. Potential preventive measures and priorities for future research are outlined.

The quality of seedlings is an important factor for development of the pear industry. A strong seedling with few branches and suitable internodes is ideal material as a rootstock for grafting and breeding. Several branching mutants of pear rootstocks were identified previously. In the present study, ‘QAU-D03’ (Pyrus communis L.) and it’s mutants were used to explore the mechanism that affects branch formation by conducting phenotypic trait assessment, hormone content analysis, and transcriptome analysis. The mutant plant (MP) showed fewer branches, shorter 1-year-old shoots, and longer petiole length, compared to original plants (OP), i.e., wild type. Endogenous hormone analysis revealed that auxin, cytokinin, and jasmonic acid contents in the stem tips of MP were significantly higher than those of the original plants. In particular, the jasmonic acid content of the MP was 1.8 times higher than that of the original plants. Transcriptome analysis revealed that PcCOI1, which is a transcriptional regulatory gene downstream of the jasmonic acid signaling pathway, was expressed more highly in the MP than in the original plants, whereas the expression levels of PcJAZ and PcMYC were reduced in the MP compared with that of the original plants. In response to treatment with exogenous methyl jasmonate, the original plants phenotype was consistent with that of the MP in developing less branches. These results indicate that jasmonic acid negatively regulates branch growth of pear trees and that jasmonic acid downstream regulatory genes play a crucial role in regulating branching.

Cork spot is a common physiological disorder in pear fruits, which has been found in some pear cultivars. Mineral nutrition imbalance in fruit is regarded as the principal influence factor for disorder incidence, with some ongoing confusion and controversy. In our research, we explored the cork spot characteristics in Japanese pear ‘Akizuki’ (Pyrus pyrifolia Nakai), adopted metabolome and mineral content analysis for healthy and disordered fruits, and made a correlation analysis of mineral and metabolites. Cork spots are mainly distributed on the outer flesh beneath the fruit peel. In cork spotted tissues, superoxide (SOD) and peroxidase (POD) activities, as well as malondialdehyde (MDA) content, increased. A total of 1024 known metabolites were identified from all the samples and more changes in metabolism were detected between normal and cork spotted flesh tissues. Correlation analysis displayed that Ca, especially the Mg/Ca in fruits, could be used to predict whether an orchard will develop cork spot disorder; Mg and B were associated with the appearance of symptoms, and the contents of Zn, Fe, and Mg, as well as Mg/B and Zn/B, might be strongly tied to the formation of cork spots in pears. This research provides insights into the occurrence of pear cork spot disorder and clarifies the role of minerals.

Geological structures of Sima coal mine in Shanxi Province were analyzed to understand the control effect of the geological structures on the occurrence of coalbed methane (CBM) in coal seam #3 of Sima coal mine. The CBM contents in the districts #2 and #3 of Sima coal mine were tested, and the effects of buried depth, fault and collapse column on the distribution of coalbed methane content are studied. The research results showed that: 1) The average content of CBM has a linear relationship with buried depth and overburden thickness, but in the smallscale range of buried depth, the dispersion between CBM and buried depth is very large. 2) Faults and collapse columns significantly affect the content of local CBM nearby, but from the largescale range such as the whole mining area, the average value of CBM content at a certain buried depth will not be affected by faults and collapse columns. 3) In the hanging wall of F29 normal fault, it is roughly estimated that the average escape rate of CBM near the fault is 13.9%, while in the footwall of F29 normal fault, this value is 0.7%–1.1%. The results show that there is a significant difference in the influence of the fault on the CBM content in the hanging wall and footwall. 4) The control effect of collapse column on CBM occurrence is related to the development height of collapse column, the cementation degree of collapse column, groundwater runoff conditions and other factors. It can be divided into three categories: aggregation action, escape action (such as collapse column X8) and no obvious effect (such as collapse column DX7).

The lotus (Nelumbo Adans.) is one of the most economically relevant ornamental aquatic plants. Plant architecture (PA) is an important trait for lotus classification, cultivation, breeding, and applications. However, the underlying genetic and molecular basis controlling PA remains poorly understood. In this study, an association study for PA-related traits was performed with 93 genome-wide microsatellite markers (simple sequence repeat, SSR) and 51 insertion–deletion (InDel) markers derived from the candidate regions using a panel of 293 lotus accessions. Phenotypic data analysis of the five PA-related traits revealed a wide normal distribution and high heritability from 2013 to 2016, which indicated that lotus PA-related traits are highly polygenic traits. The population structure (Q-matrix) and the relative kinships (K-matrix) of the association panels were analyzed using 93 SSR markers. The mixed linear model (MLM) taking Q-matrix and K-matrix into account was used to estimate the association between markers and the traits. A total of 26 markers and 65 marker–trait associations were identified by considering associations with p < 0.001 and Q < 0.05. Based on the significant markers, two QTLs on Chromosome 1 were identified, and two candidate genes were preliminarily determined. The results of our study provided useful information for the lotus breeding aiming at different PA phenotypes using a molecular-assisted selection (MAS) method and also laid the foundation for the illustration of the molecular mechanism underlying the major QTL and key markers associated with lotus PA.

KNOTTED1-like homeobox (KNOX) transcription factors (TFs) belonging to the homeobox TF family play important roles in plant growth, development, and responses to abiotic and biotic stress. However, little information is available on KNOX TF in pear (Pyrus). In this study, 19 PbKNOXs TFs were re-identified in pear (Pyrus bretschneideri Rehd.). Phylogenetic analysis revealed that the TFs were clustered into three groups with 10 conserved motifs, some of which were group- or subgroup-specific, implying that they are important for the functions of the KNOX in these clades. PbKNM1 and PbKNM2 are KNM (encodes a MEINOX domain but not a homeodomain) genes identified in pear for the first time. KNOX genes in Pyrus and Malus were closely related, and a collinear relationship among PbKNOX genes in Pyrus and Malus was observed. Analysis of the expression patterns of PbKNOX genes in different tissues, at various growth stages, and in response to abiotic and biotic stress revealed that PbKNOXs are involved in plant growth and development. Our comparative transcriptional analysis of dwarf mutant varieties revealed that genes belonging to class I are highly expressed compared with genes in other classes. Analysis of the expression of PbKNOX genes in the hybrid offspring of vigorous and dwarf varieties revealed that PbKNOX genes were highly expressed in the vigorous offspring and weakly expressed in the dwarf offspring. These findings provide new insight into the function of KNOX TFs in pear and will aid future studies of dwarf fruit trees.

Fruit pedicel is the bridge linking the parent tree and the fruit, which is an important channel for water and nutrients transport to the fruit. The genetic specificity determines the characteristics of the pedicel and the fruit, but the relationship between the pedicel structure and the fruit characteristics is unexplored. Combining the investigation of fruit characteristics, the statistical analysis of the pedicel structural properties, and the 2D and 3D anatomical observation of the pedicel, this study found distinctive contributions of the pedicel elements to the fruit characteristics in four pear species. The European pear (Conference) showed distinct fruit shape index and pedicel structural properties compared with the oriental pears (Akizuki, Yali, and Nanguoli). The fruit size positively correlated with pedicel length, fiber area, pedicel diameter, the area percentage of the cortex, and the area percentage of phloem; however, fruit firmness and soluble solids concentration are showed a stronger positive correlation with xylem area, pith area, the area percentage of xylem, the area percentage of sieve tube, and the area percentage of pith. Pedicel elements, including pith, fiber, and cortex, likely play a certain role in the fruit growth due to the variations of their characteristics demonstrated in the four pear species. The porosity, the ratio of the surface area to the volume, and the spatial arrangement of the vessels showed significant variations across the pear species, indicating the distinction of the hydraulic conductance of the pedicels. Our findings provided direct evidence that pedicel structural elements contributed distinctively to the fruit characteristics among pear species.

Mixed cation strategy greatly benefits the enhancement of device performance and chemical stability. However, adverse impact also accompanies the mixed cation system simultaneously. It brings the compositional instability, wherein the homogeneous film is likely to segregate into multi-phases during the fabrication and ageing process, thus resulting in the efficiency reduction of perovskite solar cells (PSCs) devices. This review focuses on the cation induced phase segregation, and elucidates the segregation mechanisms from the perspectives of film formation and ageing process, respectively. Furthermore, the influence of cation segregation on device performance and operational stability are discussed. And based on these understandings, viable strategies are proposed for the design of phase-stable mixed composition halide perovskites and for suppressing segregation to benefit its development towards commercial applications.

‘Xinqihong’ is a recently selected and well-colored red pear (Pyrus bretschneideri Rehd.) cultivar that is popular in the marketplace owing to the bright red color and high quality of the fruit. The red pigmentation is strongly associated with the light signal. However, its responses to bagging treatment and to light exposure after shading are unknown. In this study, the fruit were treated with three types of fruit bags. ’Xinqihong’ fruit colored rapidly in response to light stimulation. A white fruit bag was optimal for bagging of ‘Xinqihong’ fruit. To ensure satisfactory red pigmentation, the fruit required exposure to 30 days of light after bag removal. A transcriptome analysis was conducted to screen light-signal-related genes and identify their possible functions. PbCRY1 activated the promoter of PbHY5.2 and enhanced its expression. PbHY5.2 activated the promoter activity of PbUFGT and induced anthocyanin synthesis, and also showed self-activation characteristics. Both PbCRY2 and PbPHY1 induced anthocyanin accumulation. Thus, blue-light receptors played an important role in anthocyanin synthesis. This study provides a theoretical basis for the bagging cultivation of new varieties of ‘Xinqihong’, and lays a foundation for the study of the mechanisms of red pear fruit coloring in response to light signals.

‘Akituki’ (Pyrus pyrifolia Nakai.) is a very popular and profitable pear cultivar in China. However, its high susceptibility to cork spot disorder has limited its expansion of cultivated area. The mechanisms of cork spot disorder have been discussed extensively, focusing on Ca2+ deficiency, yet no consensus has been made. In this study, we applied nitrendipine (NI) as a Ca2+ uptake inhibitor to explore the role of calcium in cork spot disorder occurrence. Results showed that NI treatment on the fruit remarkably increased the incidence of cork spot disorder; alteration of mineral contents happened at the early developmental stage of the fruit, especially on the outer flesh and the peel of the fruit; and this gap was filled gradually along with the expansion of the fruit. Significant differences in the expression levels of Ca2+ transport-related genes were found in the inner flesh, outer flesh and peel during the fruit growth period. The observation of free Ca2+ localization indicated the intracellular imbalance of Ca2+ in the NI-treated fruit. In conclusion, NI treatment reduced the calcium content in the fruit at an early developmental stage, altered the related expression of genes and influenced the cellular Ca2+ balance in the fruit, which prompted the occurrence of cork spot disorder. Measures for the prevention and control of cork spot disorder should be taken at the early stage of the fruit development in the field.
