Abstract Background The timing of surgical intervention for Hirschsprung’s disease (HSCR) has been a topic of continued discussion. The objective of this study was to evaluate the significance of age at surgery in the management of HSCR by conducting a comparative analysis of the correlation between surgical age and midterm outcomes. Methods We conducted a retrospective analysis of children with HSCR who underwent one-stage laparoscopic assisted pull-through surgery with modified Swenson technology at our hospital between 2015 and 2019. The study population was stratified into two groups based on surgical age: patients who underwent surgery within a period of less than 3 months and those who underwent surgery between 3 and 12 months. The basic conditions, complications at 3–7 years after surgery, anal function (Rintala scale) and quality of life (PedsQLTM4.0) were compared between the groups. Results A total of 235 children (196 males and 39 females) were included in the study. No statistically significant differences in postoperative bowel function (P = 0.968) or quality of life (P = 0.32) were found between the two groups. However, there was a significant reduction in the incidence of Hirschsprung-associated enterocolitis (HAEC) among individuals under the age of three months prior to undergoing surgical intervention (69.1%) compared to the incidence observed postsurgery (30.9%). Although sows do not directly enter the market, they play an important role in piglet breeding on farms. They consume large amounts of feed, resulting in a significant environmental burden. Pig farms can increase their income and reduce environmental pollution by increasing the litter size (LS) of swine. PCR-RFLP/SSCP and GWAS are common methods to evaluate single-nucleotide polymorphisms (SNPs) in candidate genes. We conducted a systematic meta-analysis of the effect of SNPs on pig LS. We collected and analysed data published over the past 30 years using traditional and network meta-analyses. Trial sequential analysis (TSA) was used to analyse population data. Gene set enrichment analysis and protein–protein interaction network analysis were used to analyse the GWAS dataset. The results showed that the candidate genes were positively correlated with LS, and defects in PCR-RFLP/SSCP affected the reliability of candidate gene results. However, the genotypes with high and low LSs did not have a significant advantage. Current breeding and management practices for sows should consider increasing the LS while reducing lactation length and minimizing the sows’ non-pregnancy period as much as possible.

Cold stress significantly affects gene expression in adipocytes; studying this phenomenon can help reveal the pathogeneses of conditions such as obesity and insulin resistance. Adipocyte triglyceride lipase (ATGL); cell death-inducing deoxyribonucleic acid (DNA) fragmentation factor subunit alpha (DFFA)-like effector (CIDEA); and uncoupling protein genes UCP1, UCP2, and UCP3 are the most studied genes in pig adipose tissues under cold stress. However, contradictory results have been observed in gene expression changes to UCP3 and UCP2 when adipose tissues under cold stress were examined. Therefore, we conducted a meta-analysis of 32 publications in total on the effect of cold stress on the expression of ATGL, CIDEA, UCP2, and UCP3. Our results showed that cold stress affected the expression of swine adipocyte genes; specifically, it was positively correlated with the expression of UCP3 in swine adipocytes. Conversely, expression of ATGL was negatively affected under cold stress conditions. In addition, the loss of functional UCP1 in pigs likely triggered a compensatory increase in UCP3 activity. We also simulated the docking results of UCP2 and UCP3. Our results showed that UCP2 could strongly bind to adenosine triphosphate (ATP), meaning that UCP3 played a more significant role in pig adipocytes.

Cdon and boc are members of the cell adhesion molecule subfamily III Ig/fibronectin. Although they have been reported to be involved in muscle and neural development at late developmental stage, their early roles in embryonic development remain unknown. Here, we discovered that in zebrafish, cdon, but not boc, is expressed in dorsal forerunner cells (DFCs) and the epithelium of Kupffer’s vesicle (KV), suggesting a potential role for cdon in organ left-right (LR) patterning. Further data showed that liver and heart LR patterning were disrupted in cdon morphants and cdon mutants. Mechanistically, we found that loss of cdon function led to defect in DFCs clustering, reduced KV lumen, and defective cilia, resulting in randomized Nodal/spaw signaling and subsequent organ LR patterning defects. Additionally, predominant distribution of a cdon morpholino (MO) in DFCs caused defects in DFC clustering, KV morphogenesis, cilia number/length, Nodal/spaw signaling, and organ LR asymmetry, similar to those observed in cdon morphants and cdon−/− embryos, indicating a cell-autonomous role for cdon in regulating KV formation during LR patterning. In conclusion, our data demonstrate that during gastrulation and early somitogenesis, cdon is essential for proper DFC clustering, KV formation, and normal cilia, thereby playing a critical role in establishing organ LR asymmetry.

Abstract Background Porcine deltacoronavirus (PDCoV) is one of the emerging swine enteric coronaviruses (SECoVs), which has been widely prevalent in the North America and Asia. In addition to causing severe diarrhea in piglets, PDCoV also shows the potential to infect diverse host species, including calves, chickens, turkey poults, and humans. However, the clinical pathogenicity and genetic evolution of PDCoV is still not fully understood. Results Here, we recorded an outbreak of a novel recombinant PDCoV strain (CHN-HeN06-2022) in a large nursery fattening pig farm. Genomic analysis showed that the CHN-HeN06-2022 strain shared 98.3-98.7% sequence identities with the Chinese and American reference strains. To clarify the evolutionary relationships, phylogenetic analysis was performed using the PDCoV genome sequences available in the GenBank database. Based on genetic distance and geographical distribution, the phylogenetic tree clearly showed that all the PDCoV sequences could be divided into lineage 1 and lineage 2, which were further classified into sublineage 1.1 (Chinese strains), 1.2 (the North American strains), 2.1 (the Southeast Asian strains), and 2.2 (Chinese strains). Corresponding to the evolutionary tree, we found that, compared to lineage 1, lineage 2 strains usually contain a continuous 6-nt deletion in Nsp2 and a 9-nt deletion in Nsp3, respectively. Furthermore, recombination analysis suggested that the CHN-HeN06-2022 occurred segments exchange crossed Nsp2 and Nsp3 region between sublineage 1.1 and sublineage 2.1. Combined with previously reported recombinant strains, the highest recombination frequency occurred in Nsp2, Nsp3, and S gene. Additionally, we identified a total of 14 amino acid sites under positive selection in spike protein, most of which are located in the regions related with the viral attachment, receptor binding, and membrane fusion. Conclusions Taken together, our studies provide novel insights into the genetic diversity and adaptive evolution of PDCoV. It would be helpful to the development of vaccine and potential antiviral agent.

IntroductionSoil has the highest carbon sink storage in terrestrial ecosystems but human farming activities affect soil carbon deposition. In this study, land cultivated for 70 years was selected. The premise of the experiment was that the soil could be finely categorized by subtype classification. We consider that farming activities affect the soil bacterial community and soil organic carbon (SOC) deposition differently in the three subtypes of albic black soils.MethodsNinety soil samples were collected and the soil bacterial community structure was analysed by high-throughput sequencing. Relative changes in SOC were explored and SOC content was analysed in association with bacterial concentrations. ResultsThe results showed that the effects of farming activities on SOC deposition and soil bacterial communities differed among the soil subtypes. Carbohydrate organic carbon (COC) concentrations were significantly higher in the gleying subtype than in the typical and meadow subtypes. RB41, Candidatus-Omnitrophus and Ahniella were positively correlated with total organic carbon (TOC) in gleying shallow albic black soil. Corn soybean rotation have a positive effect on the deposition of soil carbon sinks in terrestrial ecosystems.DiscussionThe results of the present study provide a reference for rational land use to maintain sustainable development and also for the carbon cycle of the earth.

IntroductionCardiovascular diseases, including myocardial infarction, remain a leading cause of death globally. Emerging evidence suggests the gut microbiota plays a crucial role in cardiovascular health. This study aims to explore the impact of gut microbiota on myocardial infarction using a mouse model.MethodsThe research utilizes a multi-omics approach, including 16S rDNA sequencing and LC-MS-based metabolomics to analyze fecal and serum samples from mice modeled to mimic myocardial infarction. This methodology allows for a comprehensive analysis of microbial populations and their metabolic output.ResultsThe findings reveal a significant reduction in gut microbiota α-diversity in mice with induced myocardial infarction compared to healthy controls. Notably, there is an increase in populations of Fusobacteria and Clostridia. Metabolomic analysis indicates disruptions in amino acid and energy metabolism, suggesting a metabolic dysregulation linked to myocardial health.DiscussionThe study proposes a novel microbiota-metabolite-myocardium axis, where specific microbial metabolites may directly affect heart health. This connection points to the gut microbiota as a potential player in the pathogenesis of myocardial infarction and may open new therapeutic avenues targeting the gut microbiome to combat cardiovascular diseases.

With the rapid development of power grid, the structure and technology of the secondary system in substations are also constantly innovating. The new generation of intelligent substations has achieved online monitoring functions for secondary equipment, making some state variables of relay protection equipment become observable indicators. Based on this, this paper proposes a novel relay protection equipment status evaluation strategy. Firstly, considering the fuzziness and uncertainty of the boundary division of relay protection evaluation levels, a relay protection risk assessment method based on normal cloud model has been proposed. Then, due to the particularity of historical statistical data, a weight calculation method combining analytical hierarchy process (AHP) and entropy weight method is adopted to eliminate subjective factors in the weight calculation process. Meanwhile, the equipment operation risk level was determined by calculating the certainty of the corresponding evaluation level for each indicator. Finally, in case study, the proposed method is used to evaluate the status of relay protection equipment, and the feasibility and accuracy of this method are verified through the analysis of the results.

IntroductionMelatonin can treat androgenetic alopecia in males. Goats can be used as animal models to study melatonin treatment for human alopecia. In this study, a meta-analysis of melatonin’s effects on goat hair follicles was pursued.MethodsLiterature from the last 20 years was searched in Scopus, Science Direct, Web of Science and PubMed. Melatonin’s effect on goat hair follicles and litter size were performed through a traditional meta-analysis and trial sequential analysis. A network meta-analysis used data from oocyte development to blastocyst. The hair follicle genes regulated by melatonin performed KEGG and PPI. We hypothesized that there are differences in melatonin receptors between different goats, and therefore completed melatonin receptor 1A homology modelling and molecular docking.ResultsThe results showed that melatonin did not affect goat primary follicle or litter size. However, there was a positive correlation with secondary follicle growth. The goat melatonin receptor 1A SNPs influence melatonin’s functioning. The wild type gene defect MR1 is a very valuable animal model.DiscussionFuture studies should focus on the relationship between goat SNPs and the effect of embedded melatonin. This study will provide theoretical guidance for the cashmere industry and will be informative for human alopecia research.

Abstract Background The status of anastomotic blood perfusion is associated with the occurrence of anastomotic leakage after intestinal anastomosis. Fluorescence angiography (FA) with indocyanine green (ICG) can objectively assess intestinal blood perfusion. This study aims to investigate whether anastomotic perfusion assessment with ICG influences surgical decision-making during laparoscopic intestinal resection and primary anastomosis for colonic stricture after necrotizing enterocolitis. Methods Patients who underwent laparoscopic intestinal resection and primary anastomosis between January 2022 and December 2022 were retrospectively analyzed. Before intestinal anastomosis, the ICG fluorescence technology was used to evaluate the blood perfusion of intestinal tubes on both sides of the anastomosis. After the completion of primary anastomosis, the anastomotic blood perfusion was assessed again. Results Of the 13 cases, laparoscopy was used to determine the extent of the diseased bowel to be excised, and the normal bowel was preserved for anastomosis. The anastomosis was established under the guidance of ICG fluorescence technology, and FA was performed after anastomosis to confirm good blood flow in the proximal bowel. The anastomotic intestinal tube was changed in one case because FA showed a difference between the normal range of intestinal blood flow and the macroscopic prediction. There was no evidence of ICG allergy, anastomotic leakage, anastomotic stricture, or other complications. The median follow-up was 6 months, and all patients recovered well. Conclusions The ICG fluorescence technology is helpful in precisely and efficiently determining the anastomotic intestinal blood flow during stricture resection and in avoiding anastomotic leakage caused by poor anastomotic intestinal blood flow to some extent, with satisfactory short-term efficacy.

The mutation types GDF9 S395F and S427R increase the ovulation rate in heterozygotes, while the homozygous form leads to infertility. This study presents a meta-analysis of GDF9 and BMP15 SNPs related to sheep litter size (LS). The results showed that simulation of the molecular recognition of GDF9 R453H, S395F, and S427R was performed to study the effect on the GDF9 dimer. S395F and S427R dimer binding ability of mutant heterozygotes was improved, thereby improving LS. The binding ability of mutant homozygous dimers was reduced, resulting in infertility. The results of this study will provide important references for animal breeding.

Nutrient fertilizer application to agricultural land has led to greenhouse gas emissions and has altered soil nitrogen (N) deposition. In soil, N can be degraded in four ways: entering surface water through water flow, absorption by plants and microorganisms, decomposition into gas, and deposition as minerals. This study proposes the concept of N degradability and aims to clarify how farming activities affect N degradability in soil. Over 260 soil profiles were excavated, and the effective soil depth, coordinates, soil types, and vegetation were recorded at each measurement point. The following characteristics were determined in the soil samples: pH, organic matter, total N, total phosphorus, total potassium, total soluble N, available phosphorus, and available potassium. The sample characteristics were subjected to Pearson correlation analysis, principal component analysis, and one-way analysis of variance. The 260 samples included four soil types: dark brown soil, black soil, albic black soil, and meadow soil. Black soil exhibited more stable N levels compared with the other three soil types, showing a tendency towards N accumulation. Ground vegetation was categorized into seven types: forest, rice, maize, red adzuki bean, grassland, soybean, and others. Forests contributed the most to N deposition. Conversely, planting maize led to a tendency for N loss compared with forests. This study can provide a reference for the sustainable development of agriculture and the balance of ecological protection.

Glutinous rice (GR), an important food crop in Asia, provides prolonged energy for the human body due to its high amylopectin content. The non-volatile metabolites generated by different cooking methods that affect the nutritional value and color of GR are still poorly understood. Herein, a widely targeted metabolomics approach was used to understand the effects of different cooking methods (steaming, baking, and frying) on the metabolite profiles of GR. Compared with other treatments, steamed GR had a brighter color and significantly lower contents of total sugar, starch, amylopectin, and amylose, at 40.74%, 14.13%, 9.78%, and 15.18%, respectively. Additionally, 70, 108, and 115 metabolites were significantly altered in the steaming, baking, and frying groups respectively, and amino acid and carbohydrate metabolism were identified as the representative metabolic pathways based on KEGG annotations. Further evaluation of 14 amino acids and 12 carbohydrates in steamed GR, especially 4-aminobutyric acid, suggested its high nutraceutical value. Additionally, multivariate analysis indicated that total sugar content, amylose content, beta-alanine methyl ester hydrochloride, and 4-aminobutyric acid played a critical role in color formation in raw and cooked GR. Finally, the levels of major amino acids and carbohydrates were quantified by conventional methods to verify the reliability of the metabolome. Consequently, this in-depth understanding of metabolite profiling in normal cooking methods has provided a foundation for the processing of GR products.

In practical applications, the effectiveness of biological control agents such as Bacillus is often unstable due to different soil environments. Herein, we aimed to explore the control effect and intrinsic mechanism of Bacillus in black soil and red soil in combination with tomato Verticillium wilt. Bacillus application effectively controlled the occurrence of Verticillium wilt in red soil, reducing the incidence by 19.83%, but played a limited role in black soil. Bacillus colonized red soil more efficiently. The Verticillium pathogen decreased by 71.13% and 76.09% after the application of Bacillus combinations in the rhizosphere and bulk of the red soil, respectively, while there was no significant difference in the black soil. Additionally, Bacillus application to red soil significantly promoted phosphorus absorption. Furthermore, it significantly altered the bacterial community in red soil and enriched genes related to pathogen antagonism and phosphorus activation, which jointly participated in soil nutrient activation and disease prevention, promoting tomato plant growth in red soil. This study revealed that the shaping of the bacterial community by native soil may be the key factor affecting the colonization and function of exogenous Bacillus.

Thin layers and high-yield dolomite reservoirs were recently discovered in the Permian Guadeloupian Maokou Formation. The genetic mechanism of this reservoir is controversial because of its complex sedimentation and diagenesis in the Maokou Formation. Traditionally, the genesis has focused on sedimentation, karst, and fracture, whereas the influence of the Emeishan mantle plume activity (EMP) has been ignored. In this study, we enumerated petrographic (grouped into micritic bioclastic limestone, limy dolomite, grain dolomite, dolomite cement, calcite cement, and saddle dolomite) and geochemical data (δ13C, δ18O, REE, and 87Sr/86Sr) from a microscopic perspective to support the impact of EMP on reservoirs. We conclude that EMP activity altered the sedimentary environment and induced a complex diagenesis. The paleogeomorphic reconstruction data indicate that the EMP caused an uplift zone in the NE–SW direction, depositing advantageous high-energy beach facies. In terms of diagenesis, the abnormally high 87Sr/86Sr ratios and REE with positive Eu anomalies suggest that dolomitization was influenced by both seawater and hydrothermal fluids. Based on the above evidence, we established a reservoir genetic model for the Maokou Formation related to the intensity of the EMP. This study provides a new perspective on the mantle plume activity for the study of carbonate reservoir genesis.

Plant FLOWERING LOCUS T-Like (FTL) genes often redundantly duplicate on chromosomes and functionally diverge to modulate reproductive traits. Rice harbors thirteen FTL genes, the functions of which are still not clear, except for the Hd3a and RFT genes. Here, we identified the molecular detail of OsFTL12 in rice reproductive stage. OsFTL12 encoding protein contained PEBP domain and localized into the nucleus, which transcripts specifically expressed in the shoot and leaf blade with high abundance. Further GUS-staining results show the OsFTL12 promoter activity highly expressed in the leaf and stem. OsFTL12 knock-out concurrently exhibited early flowering phenotype under the short- and long-day conditions as compared with wild-type and over-expression plants, which independently regulates flowering without an involved Hd1/Hd3a and Ehd1/RFT pathway. Further, an AT-hook protein OsATH1 was identified to act as upstream regulator of OsFTL12, as the knock-out OsATH1 elevated the OsFTL12 expression by modifying Histone H3 acetylation abundance. According to the dissection of OsFTL12 molecular functions, our study expanded the roles intellectual function of OsFTL12 in the mediating of a rice heading date.

Abstract Background Rice is one of the most important cereal crops in the world but is susceptible to cold stress (CS). In this study, we carried out parallel transcriptomic analysis at the reproductive stage on the anthers of two Japonica rice varieties with contrasting CS resistance: cold susceptible Longjing11 (LJ11) and cold resistant Longjing25 (LJ25). Results According to the obtained results, a total of 16,762 differentially expressed genes (DEGs) were identified under CS, including 7,050 and 14,531 DEGs in LJ25 and LJ11, respectively. Examining gene ontology (GO) enrichment identified 35 up- and 39 down-regulated biological process BP GO terms were significantly enriched in the two varieties, with ‘response to heat’ and ‘response to cold’ being the most enriched. Kyoto Encyclopedia of Genes and Genomes (KEGG) analysis identified 33 significantly enriched pathways. Only the carbon metabolism and amino acid biosynthesis pathways with down-regulated DEGs were enriched considerably in LJ11, while the plant hormone signal transduction pathway (containing 153 DEGs) was dramatically improved. Eight kinds of plant hormones were detected in the pathway, while auxin, abscisic acid (ABA), salicylic acid (SA), and ethylene (ETH) signaling pathways were found to be the top four pathways with the most DEGs. Furthermore, the protein-protein interaction (PPI) network analysis identified ten hub genes (co-expressed gene number ≥ 30), including six ABA-related genes. Various DEGs (such as OsDREB1A, OsICE1, OsMYB2, OsABF1, OsbZIP23, OsCATC, and so on) revealed distinct expression patterns among rice types when the DEGs between LJ11 and LJ25 were compared, indicating that they are likely responsible for CS resistance of rice in cold region. Conclusion Collectively, our findings provide comprehensive insights into complex molecular mechanisms of CS response and can aid in CS resistant molecular breeding of rice in cold regions.

BackgroundThe severe acute abdomen associated with Henoch-Schonlein purpura (HSP) is an acute intussusception (AI). There is no reliable specific marker for AI with abdominal-type HSP. The serum total bile acid (TBA) level is a new prognostic marker associated with the severity of intestinal inflammation. The purpose of this study was to identify the prognostic value of serum TBA levels for the diagnosis of AI in children with abdominal-type HSP.MethodsA retrospective study of 708 patients with abdominal-type HSP was conducted, with demographic data, clinical symptoms, hepatic function index, immune function markers, and clinical outcomes assessed. Patients were divided into two groups: HSP (613 patients) and HSP with AI (95 patients). The data were analysed using SPSS 22.0.ResultsOf the 708 patients, the serum TBA levels were higher in the HSP with AI group than in the HSP group (P 3 μmol/L for predicting AI in children with abdominal-type HSP. In this group of HSP patients with AI, a serum TBA level ≥6.98 μmol/L was significantly associated with an increased incidence of operative treatment (51.85% vs. 75.61%, P = 0.0181), intestinal necrosis (9.26% vs. 29.27%, P = 0.0117), and length of hospital stay [15.76 ± 5.31 vs. 10.98 ± 2.83 (days), P

Planting economic crops in soil with salt imbalances can improve land use efficiency. Hemp, which can be planted in low-salinity soil, is a crop with a very high economic value. To reveal the salt tolerance mechanism of hemp, the leaves of Longma #3 and #9 were collected at 0, 2, 4, and 6 days after salt stress in this study, and transcriptome and metabolome joint analysis were performed. The concentration of glutamic acid, succinic acid and gamma-aminobutyric acid (GABA) was 12 times that of the control group. The large increase in succinic acid led to the maintenance of peroxidase and superoxide dismutase levels at normal values and a decrease in H2O2 and catalase. The enriched KEGG pathway citrate cycle (TCA cycle) contains the metabolic regulation of succinic acid. Succinic acid was positively correlated with the MAPK gene. The MAPK pathway is a key pathway of plant tolerance in a high-salt environment. This study provides a reference for research on NaCl tolerance in plants and can also provide an important reference for the cultivation of NaCl-tolerant hemp varieties.

Abstract Purpose It has been shown that abnormalities of coagulation and fibrinolysis system are involved in the pathogenesis of necrotizing enterocolitis (NEC), but not well studied challenge in the context of early detection of disease progression. The present study mainly explores the predictive significance of coagulation parameters at the time of NEC diagnosis in identifying the patients who eventually received surgery and/or NEC-related deaths. Methods The retrospective study of 114 neonates with NEC was conducted with assessments of demographic data, laboratory results at the time of NEC diagnosis, treatment methods and prognosis. According to treatment methods, patients were divided into surgical intervention group and medical treatment group. Predictive factors were put forward and determined by receiver operating characteristic (ROC) curve analysis. An analysis of the surgical intervention and prognosis was performed. Results Of 114 patients, 46 (40.4%) cases received surgical intervention and 14 (12.3%) deaths. prothrombin time (PT), PT international normalized ratio, activated partial thromboplastin time (APTT), fibrinogen and platelet count at the time of NEC diagnosis were independently associated with surgical NEC. The APTT could identify patients at high risk for surgical NEC, with 67.39% sensitivity, 86.76% specificity, better than that of other serological parameters. Abstract The sensitivity of rice to low-temperature stress (LTS), especially at the reproductive stage, is a primary factor of rice yield fluctuation in cold cultivate region. Here, the changes of reactive oxygen species (ROS), osmotic adjustment substances, and antioxidants in different tissues were analyzed during rice growing under low temperatures (LT) at the reproductive stage. Results showed that LTS increases the levels of proline (Pro), soluble protein (SP), glutathione (GSH), superoxidase (SOD), and ascorbate peroxidase (APX) in LJ25 (LTS-resistant) and LJ11 (LTS-sensitive). The activities of catalase (CAT) and peroxidase (POD) were significantly increased in LJ25 but decreased in LJ11 under LTS, while an opposite trend in ROS and malondialdehyde (MDA) was observed in both varieties. Moreover, most physicochemical properties were higher in flag leaves and panicles compared with those in leaf sheaths. The expression patterns of OsCOIN, OsCATC, OsMAP1, OsPOX1, and OsAPX were the same with phenotypic changes in Pro and the enzymes encoded by them, confirming the accuracy of the physicochemical analysis. Therefore, only CAT and POD increased more in LJ25, suggesting they could be the key factors used for LT-tolerant breeding of rice in cold regions.

IntroductionThe mechanisms by which male hormones affect the development of ovaries and follicles has been studied by injecting exogenous androgens into sows. This may provide a reference for human polycystic ovary syndrome (PCOS), and can also provide guidance for improving the litter size of sows.MethodsWe present a meta-analysis of studies published in the past 30 years on the effect of androgens on the ovulation rate of sows. A total of 517 papers were analyzed.ResultsThe results showed that both testosterone (T) and dihydrotestosterone (DHT) injected into sows were positively related to the ovulation rate. T did not have a relevant effect on swine in vivo blastocyst survival rate. DHT had a negative phase with respect to blastocyst survival rate. Pig T-androgen receiver affinity was higher than the analogous affinity for DHT; this is different in humans. This suggests that sows are not suitable as human PCOS experimental animal models.DiscussionTo improve the litter size of sows, future research should focus on the mixed use of T and DHT, and the timing of use should be consistent with the periodic changes in androgen levels in sows. In addition, the welfare of experimental sows should be considered with reference to the clinical symptoms of PCOS.

BackgroundSystemic Immune-Inflammation Index (SII), known as an easy, economical and useful marker, correlates with the severity of inflammatory response. However, the usefulness of SII in necrotizing enterocolitis (NEC) remains unclear. Therefore, we evaluated the correlation of SII at NEC diagnosis and subsequent surgery.MethodsRetrospective review of 131 neonates with NEC in a tertiary-level pediatric referral hospital was conducted with assessments of demographic data, general blood examination results at NEC diagnosis, treatment strategies and clinical outcomes. The receiver operating characteristic (ROC) curve determined the optimal cut-off values of SII, platelet-to-lymphocyte ratio (PLR), and neutrophil-to-lymphocyte ratio. Univariate/multivariate logistic regression analysis and ROC curve were conducted to evaluate the predictive significance of SII in identifying the patients who eventually received surgery. Additionally, NEC-related deaths were assessed.ResultsOverall, 49 (37.4%) cases received surgical intervention and mortality was 12.3% (14/131). The area under ROC curve of SII at NEC diagnosis to predict subsequent surgery was 0.833 (optimal cut-off value: 235.85). Abstract Background Porcine reproductive and respiratory syndrome virus (PRRSV) infection can cause severe reproductive failure in sows and respiratory distress in pigs of all ages, leading to major economic losses. To date, there are still no effective strategies to prevent and control PRRSV. Antibody-dependent enhancement (ADE), a phenomenon in which preexisting non-neutralizing antibodies or sub-neutralizing antibodies facilitate virus entry and replication, may be a significant obstacle in the development of effective vaccines for many viruses, including PRRSV. However, the contribution of ADE to PRRSV infection remains controversial, especially in vivo. Whether attenuated PRRSV vaccines prevent or worsen subsequent disease in pigs infected by novel PRRSV strains requires more research. In the present study, in vivo experiments were conducted to evaluate ADE under different immune statuses, which were produced by waiting different lengths of time after vaccination with a commercially available attenuated highly pathogenic PRRSV (HP-PRRSV) vaccine (JXA1-R) before challenging the pigs with a novel heterologous NADC30-like strain. Results Piglets that were vaccinated before being challenged with PRRSV exhibited lower mortality rates, lower body temperatures, higher bodyweight gain, and lower viremia. These results demonstrate that vaccination with JXA1-R alleviated the clinical signs of PRRSV infection in all vaccinated groups. Conclusions The obtained data indicate that the attenuated vaccine test here provided partial protection against the NADC30-like strain HNhx. No signs of enhanced PRRSV infection were observed under the applied experimental conditions. Our results provide some insight into the molecular mechanisms underlying vaccine-induced protection or enhancement in PRRSV.

Straw return is considered an effective way to improve the soil organic carbon (SOC) content of farmland. Most studies have suggested that a straw application increases the SOC content; however, some suggest that a straw application reduces the SOC content when used in combination with mineral fertilisation. Therefore, a meta-analysis of the effect of a straw application on the SOC change is needed. This study comprises a meta-analysis of 115 observations from 65 research articles worldwide. Straw applications can significantly increase the proportion of the SOC in the soil. Straw applications caused a significant microbial biomass carbon (MBC) increase in tropical and warm climatic zones. The MBC increase was higher than the SOC increase. For agriculture, the most important soil functions are the maintenance of the crop productivity, the nutrient and water transformation, the biological flora and activity, and the maintenance of the microbial abundance and activity. These functions should be prioritised in order to maintain the SOC function and services. Straw applications should not be excessive, especially when combined with mineral fertilisation, in order to avoid the loss of carbon from the straw in the form of greenhouse gases. A large amount of unused fertiliser also leads to a series of environmental problems.

Abstract Background Smooth endoplasmic reticulum aggregation (SERa, SER+) has been reported to increase the risk of birth malformations and other abnormal outcomes, miscarriage, and perinatal complications. Other studies, however, suggest that SER+ embryos may develop into healthy infants. One report indicates that 25% of in vitro fertilization (IVF) centers discard SER+ oocytes. Thus, we investigated the effect of SER+ on birth outcomes in IVF and intracytoplasmic sperm injection. Methods We performed a literature search using PubMed, ScienceDirect, Cochrane, Embase, Ovid, and Scopus. We found a total of 1500 relevant studies between 1978 and 2020 and conducted a meta-analysis to study the effects of SER+ on live births, birth weight, and the number of metaphase II (MII) oocytes retrieved per cycle. Results Eleven eligible studies were included. If the SER+ zygote was evaluated again at the embryo transfer (ET) stage, SER+ did not affect birth or infant body weight. Stimulated ovaries producing too many oocytes per cycle were positively correlated with SER+ (OR = 1.28, 95% CI = 0.41–2.15; p = 0.004). SER+ was positively correlated with oocyte maturation rate, and observed heterogeneity in a previous meta-analysis was likely due to maternal age. Our data also showed that SER+ cycles produced more oocytes but achieved the same number of births from ET. Conclusions The use of SER+ MII oocytes is rare, with the collection of many oocytes in 1 cycle potentially inducing SER+. SER+ may be more common than we originally thought, as some SER+ is found in all oocytes. Although SER+ positively affected oocyte maturation rate, it did not affect births. We hypothesized that this is because the best embryos are chosen at every step of the process, and the oocytes with the poorest characteristics are removed. We therefore suggest a standard method for measuring SER+. Although embryos produced from SER+ cycles can be used, they should only be transferred when no other suitable embryos are available over several cycles.

Early seedling vigor (ESV) is a major breeding target in rice, especially under direct seeding. To identify quantitative trait locus (QTL) affecting ESV, a recombinant inbred line population derived from a cross between 02428 and YZX, two cultivars differing in vigor during early seedling growth, was used for QTL analysis. Nine traits associated with ESV were examined using a high-density map. Of 16 additive loci identified, three were detected in two generations and thus considered stable. Four epistatic interactions were detected, one of which was repeated in two generations. Further analysis of the pyramiding effect of the three stable QTL showed that the phenotypic value could be effectively improved with an increasing number of QTL. These results were combined with results from our previous QTL analysis of the germination index. The lines G58 and G182 combined all the favourable alleles of all three stable QTL for ESV and three QTL for germination speed. These two lines showed rapid germination and strong ESV. A total of 37 candidate differentially expressed genes were obtained from the regions of the three stable QTL by analysis of the dynamic transcriptomic expression profile during the seedling growth period of the two parents. The QTL are targets for ESV breeding and the candidate genes await functional validation. This study provides a theoretical basis and a genetic resource for the breeding of direct-seeded rice.

Understanding whether and how melatonin (MT) may impact sheep oocyte development competence is central to our ability to predict how sheep oocytes will respond to artificially regulated estrus. Implanting MT can make sheep enter estrus during the non-breeding season. One study found that the blastocyst rate increased under MT treatment, while another found that the blastocyst rate decreased. Therefore, we conducted a meta-analysis of MT directly and indirectly influencing sheep oocytes. A total of 433 articles were collected from which 20 articles and 34 treatments were finally selected. A method for estimating the default value was established for the litter size analysis. We found that exogenous MT add into in vitro maturation medium was positively related to the blastocyst rate in the lab. However, subcutaneous implanting MT did not affect the in vivo ovulation rate, fertilization rate, blastocyst rate, or pregnancy rate at farm. MT did not affect the in vitro cleavage rate. However, MT improved the in vivo cleavage rate. We hypothesized that implanted MT could increase the concentration of MT in oviduct fluid in vivo, and also that in vitro MT could increase the early cleavage rate of sheep zygotes without affecting the total cleavage rate. In the analysis of oocyte apoptosis caused by injury, the results suggested that pyroptosis would be more suitable for further research. MT produces responses in all body organs, and thus implanting of MT during non-breeding seasons should consider the effect on animal welfare.

Outbreaks of hydropericardium hepatitis syndrome caused by fowl adenovirus serotype 4 (FAdV-4) with a novel genotype have been reported in China since 2015, with significant economic losses to the poultry industry. Fiber2 is one of the important structural proteins on FAdV-4 virions. In this study, the C-terminal knob domain of the FAdV-4 Fiber2 protein was expressed and purified, and its trimer structure (PDB ID: 7W83) was determined for the first time. A series of affinity peptides targeting the knob domain of the Fiber2 protein were designed and synthesized on the basis of the crystal structure using computer virtual screening technology. A total of eight peptides were screened using an immunoperoxidase monolayer assay and RT-qPCR, and they exhibited strong binding affinities to the knob domain of the FAdV-4 Fiber2 protein in a surface plasmon resonance assay. Treatment with peptide number 15 (P15; WWHEKE) at different concentrations (10, 25, and 50 μM) significantly reduced the expression level of the Fiber2 protein and the viral titer during FAdV-4 infection. P15 was found to be an optimal peptide with antiviral activity against FAdV-4 in vitro with no cytotoxic effect on LMH cells up to 200 μM. This study led to the identification of a class of affinity peptides designed using computer virtual screening technology that targeted the knob domain of the FAdV-4 Fiber2 protein and may be developed as a novel potential and effective antiviral strategy in the prevention and control of FAdV-4.

The broodiness traits of domestic geese are a bottleneck that prevents the rapid development of the goose industry. To reduce the broodiness of the Zhedong goose and thus improve it, this study hybridized it with the Zi goose, which has almost no broody behavior. Genome resequencing was performed for the purebred Zhedong goose, as well as the F2 and F3 hybrids. The results showed that the F1 hybrids displayed significant heterosis in growth traits, and their body weight was significantly greater than those of the other groups. The F2 hybrids showed significant heterosis in egg-laying traits, and the number of eggs laid was significantly greater than those of the other groups. A total of 7,979,421 single-nucleotide polymorphisms (SNPs) were obtained, and three SNPs were screened. Molecular docking results showed that SNP11 located in the gene NUDT9 altered the structure and affinity of the binding pocket. The results suggested that SNP11 is an SNP related to goose broodiness. In the future, we will use the cage breeding method to sample the same half-sib families to accurately identify SNP markers of growth and reproductive traits.

Abstract Background African swine fever (ASF) is the most important disease to the pigs and cause serious economic losses to the countries with large-scale swine production. Vaccines are recognized as the most useful tool to prevent and control ASF virus (ASFV) infection. Currently, the MGF505 and MGF360 gene-deleted ASFVs or combined with CD2v deletion were confirmed to be the most promising vaccine candidates. Thus, it is essential to develop a diagnosis method to discriminate wide-type strain from the vaccines used. Results In this study, we established a duplex TaqMan real-time PCR based on the B646L gene and MGF505-2R gene. The sequence alignment showed that the targeted regions of primers and probes are highly conserved in the genotype II ASFVs. The duplex real-time assay can specifically detect B646L and MGF505-2R gene single or simultaneously without cross-reaction with other porcine viruses tested. The limit of detection was 5.8 copies and 3.0 copies for the standard plasmids containing B646L and MGF505-2R genes, respectively. Clinical samples were tested in parallel by duplex real-time PCR and a commercial ASFV detection kit. The detection results of these two assays against B646L gene were well consistent. Conclusion We successfully developed and evaluated a duplex TaqMan real-time PCR method which can effectively distinguish the wide type and MGF505 gene-deleted ASFVs. It would be a useful tool for the clinical diagnosis and control of ASF.

Based on well test interpretation, production performance analysis, overburden permeability and porosity test, gas-water core flooding test and high-pressure mercury injection, a quantitative correlation has been built of in-situ effective permeability with routine permeability and water saturation, and the ranges of Main Flow Channel Index (MFCI) are determined for different permeability levels in porous sand gas reservoirs. A new method to evaluate the in-situ effective permeability of porous sand reservoir and a correlation chart of reserves producing degree and main flow channel index are established. The results reveal that the main flow channel index of porous sand gas reservoirs has close correlation with routine matrix permeability and water saturation. The lower the routine matrix permeability and the higher the water saturation, the lower the MFCI is. If the routine matrix permeability is greater than 5.0×10–3, the MFCI is generally greater than 0.5. When the routine matrix permeability is from 1.0×10–3 to 5.0×10–3, the MFCI is mainly between 0.2 and 0.5. When the routine matrix permeability is less than 1.0×10–3, the MFCI is less than 0.2. The evaluation method of in-situ effective permeability can be used to evaluate newly discovered or not tested porous sand gas reservoirs quickly and identify whether there is tight sand gas. The correlation chart of reserves producing degree and main flow channel index can provide basis for recoverable reserves evaluation and well infilling, and provide technical support for formulation of reasonable technical policy of gas reservoir.

Natural gas from ultra-deep reservoirs has been a major contributor for reserves boost, deliverability construction, and profits growth in natural gas industry in China. As a significant strategic domain in the future development of upstream business, the high-quality development of ultra-deep gas resources has great significance for economic benefits enhancement and sustained regional supply assurance. In this paper, based on the appraisal of development characteristics and effectiveness in the developed large ultra-deep gas fields, challenges for high quality development were indicated, which include the difficulties in structure confirm, uncertainties in reserves define and production optimization, risks of aquifer water early breakthrough and high investment of deep wells. Through indoor physical simulation experiments, reservoir characterization, performance evaluation, reservoir simulation and knowledge acquisition from analogous fields at home and abroad, the connotation and requirements for high quality development were discussed, and furthermore, strategies and proposals were thus proposed as follows: to strengthen the pre-development reservoir evaluation to define movable gas reserves and quantify rational production rate so as to avoid facility waste; to optimize both well pattern and well flow rate to achieve uniform depletion and high EUR; to continuously enhance drilling & completion technologies to further reduce drilling and completion circle and cost and targeted reservoir stimulation technologies to enhance movable reserves and single-well productivity and increase the depletion of inferior reserves; and to innovate management modes to establish scientific programs and procedures for the construction, production and operation of ultra-deep gas fields and strictly control the upper limit index of production rate so as to emphasize quality benefits. In conclusion, high-quality development of ultra-deep gas fields, a hard and complex system though, will be possibly achieved only by continuous innovation of exploration and development technologies and management modes.

The processes involved in soil domestication have altered the soil microbial ecology. We examined the question of whether animal manure application affects the soil microbial ecology of farmlands. The effects of global animal manure application on soil microorganisms were subjected to a meta-analysis based on randomized controlled treatments. A total of 2303 studies conducted in the last 30 years were incorporated into the analysis, and an additional 45 soil samples were collected and sequenced to obtain 16S rRNA and 18S rRNA data. The results revealed that manure application increased soil microbial biomass. Manure application alone increased bacterial diversity (M-Z: 7.546 and M-I: 8.68) and inhibited and reduced fungal diversity (M-Z: −1.15 and M-I: −1.03). Inorganic fertilizer replaced cattle and swine manure and provided nutrients to soil microorganisms. The soil samples of the experimental base were analyzed, and the relative abundances of bacteria and fungi were altered compared with no manure application. Manure increased bacterial diversity and reduced fungal diversity. Mrakia frigida and Betaproteobacteriales, which inhibit other microorganisms, increased significantly in the domesticated soil. Moreover, farm sewage treatments resulted in a bottleneck in the manure recovery rate that should be the focus of future research. Our results suggest that the potential risks of restructuring the microbial ecology of cultivated land must be considered.

Abstract Background Porcine circovirus type 2 (PCV2) is the pathogen of porcine circovirus associated diseases (PCVAD) and one of the main pathogens in the global pig industry, which has brought huge economic losses to the pig industry. In recent years, there has been limited research on the prevalence of PCV2 in Henan Province. This study investigated the genotype and evolution of PCV2 in this area. Results We collected 117 clinical samples from different regions of Henan Province from 2015 to 2018. Here, we found that the PCV2 infection rate of PCV2 was 62.4%. Thirty-seven positive clinical samples were selected to amplify the complete genome of PCV2 and were sequenced. Based on the phylogenetic analysis of PCV2 ORF2 and complete genome, it was found that the 37 newly detected strains belonged to PCV2a (3 of 37), PCV2b (21 of 37) and PCV2d (13 of 37), indicating the predominant prevalence of PCV2b and PCV2d strains. In addition, we compared the amino acid sequences and found several amino acid mutation sites among different genotypes. Furthermore, the results of selective pressure analysis showed that there were 5 positive selection sites. Conclusions This study indicated the genetic diversity, molecular epidemiology and evolution of PCV2 genotypes in Henan Province during 2015–2018.

Alloteropsis is a widely-distributed genus with C3 and C4 photosynthetic species belonging to the Poaceae family. This study characterized the complete chloroplast genomes of two Alloteropsis species from Chinese mainland, i.e. Alloteropsis semialata with C4 photosynthetic type and Alloteropsis cimicina with C3 photosynthetic type. The chloroplast genomes of A. semialata and A. cimicina are 138,188 bp and 139,335 bp in length, with 38.48% and 38.59% GC contents, respectively. These two chloroplast genomes contain the same number of genes, i.e. 107 unique genes including 27 tRNA, 4 rRNA, and 76 protein-coding genes. Phylogenomic analysis confirmed the species identity of A. semialata and A. cimicina and supported a close relationship of Alloteropsis with species of Setaria and Panicum in grass family.

Developing low carbon city (LCC) has been widely appreciated as the key strategy for achieving emission mitigation goal. Despite the impact factors on cities’ carbon emission reduction have been extensively studied in recent years, little attention has been paid to investigating the key driving force (KDF) on the development of LCC. This study aims to explore the KDF affecting LCC within the context of China through identifying systematically representative forces (RFs) and examining the interactive relationships between RFs. Based on topic modeling of Latent Dirichlet Allocation (LDA), RFs for LCC are identified firstly from the policies issued by the pilot LCCs in China. Then KDFs for LCC are investigated through establishing the interactive relationships between RFs by adopting the Interpretive Structure Modeling (ISM). Results show that: (1) 16 RFs on LCC are identified from the pilot LCC policies in China; (2) emission trading system, carbon emission accounting system, and low carbon planning are identified as the KDFs on the development of LCC; (3) carbon sink is positioned at the topmost level and performs as the most frontier force on the development of LCC. Results of the KDFs identification can provide policy-makers with insight for designing LCC improvement path, and consequently promote emission mitigation in China.

Abstract Background Porcine circovirus 3 (PCV3) is an emerging etiological agent to the swine industry. However, its circulating status and genetic characteristics were still unclear in Henan, central China. Here, 318 porcine oral fluid specimens were collected from asymptomatic pigs in five farms and tested by PCR . Results The results showed that the positive rate of PCV3 was 12.3% (39/318) for the total samples, and 15.06% (25/166) in the stall-based samples, 9.21% (14/152) in the pen-based samples. Of the PCV3-positive samples, 41.0% were also positive for porcine circovirus 2 (PCV2). Nucleotide sequence comparison indicated that the 10 complete genomes and 34 capsid (cap) genes in this study shared 98.7–99.9% and 98–100% pairwise identities to each other, respectively. According to phylogenetic analysis and sequence alignment of cap gene, all the isolated sequences were clustered into 3 clades, including subgroup 1 (21/39, 61.8%), subgroup 2 (5/39, 14.7%) and subgroup 3 (8/39, 23.5%). Similar to previous reports, four amino acids (V24A, K27R, S77 T and I150L) in cap protein were identified as a conserved subgroup specific molecular marker. Conclusion Our research provided new insights into the epidemiology surveillance and genetic characteristics of PCV3 in China.

We successfully achieved low-temperature assembly by reflowing the 13.5Sn-37.5Bi-45In-4Pb quaternary eutectic solder paste and the SAC 305 solder ball together at 140 °C for 5 min. The wetting angle of the mixed solder joint is 17.55°. The overall atomic percent of Pb in the mixed solder joint is less than 1%, which can be further reduced or eliminated. Moreover, after aging at 80 °C for 25 days, we observed no obvious decrease in shear strength of the fully mixed solder joint, which is the most advantage of this assembly technique over Sn58Bi solder assembly. The Bi phase segregation at the interface is slowed down compared with Sn-Bi solder joint. This low-temperature assembly is promising to be applied in advanced packaging technology to replace the eutectic Sn-Bi solder.

Planting rice in saline–alkali land can effectively improve saline–alkali soil and increase grain yield, but traditional identification methods for saline–alkali-tolerant rice varieties require tedious and time-consuming field investigations based on growth indicators by rice breeders. In this study, the Python machine deep learning method was used to analyze the Raman molecular spectroscopy of rice and assist in feature attribution, in order to study a fast and efficient identification method of saline–alkali-tolerant rice varieties. A total of 156 Raman spectra of four rice varieties (two saline–alkali-tolerant rice varieties and two saline–alkali-sensitive rice varieties) were analyzed, and the wave crests were extracted by an improved signal filtering difference method and the feature information of the wave crest was automatically extracted by scipy.signal.find_peaks. Select K Best (SKB), Recursive Feature Elimination (RFE) and Select F Model (SFM) were used to select useful molecular features. Based on these feature selection methods, a Logistic Regression Model (LRM) and Random Forests Model (RFM) were established for discriminant analysis. The experimental results showed that the RFM identification model based on the RFE method reached a higher recognition rate of 89.36%. According to the identification results of RFM and the identification of feature attribution materials, amylum was the most significant substance in the identification of saline–alkali-tolerant rice varieties. Therefore, an intelligent method for the identification of saline–alkali-tolerant rice varieties based on Raman molecular spectroscopy is proposed.

Based on performance data of over 600 wells in 32 large gas fields of different types in China, the correlation is established between per-well average dynamic reserves (G¯) and average initial absolute open flow potential (q¯IAOF) of each field, and its connotation and applicability are further discussed through theoretical deduction. In log-log plot,G¯ vs.q¯IAOF exhibit highly dependent linear trend, which implicates the compatibility betweenG¯ andq¯IAOF attained through development optimization to reach the balance among annual flow capacity, maximum profits and certain production plateau, that is to match productivity with rate maintenance capacity. The correlation can be used as analogue in new gas field development planning to evaluate the minimum dynamic reserves which meet the requirement of stable and profitable production, and facilitate well pattern arrangement. It can also serve as criteria to appraise the effectiveness and infill drilling potential of well patterns for developed gas fields. Soil microorganisms are important components of agricultural ecosystems; they are important in agricultural soil nutrient cycle and are easily affected by soil tillage. The response of soil microbial community to tillage is very complex, and the effect of the no tillage and residue mulching method on soil microbial diversity remains unclear. In 2019, the soil was collected from an experimental field after 10 years of continuous cultivation in the black soil area of the Sanjiang Plain in Northeastern China. In this study, the diversity and composition of the soil bacterial community and their relationship with soil properties were explored via high-throughput sequencing under no tillage with four residue mulching treatments. No tillage with 60% residue mulching (NTR3) significantly increased the alpha diversity of the rhizosphere soil bacteria and changed the composition of the bacterial community-consistent with changes in soil physicochemical properties. Proteobacteria, Acidobacteria, and Actinobacteria were the dominant phyla in the sample soil. Soil physicochemical properties explained 80.6% of the changes in soil diversity and composition, of which soil organic carbon, soil pH, and soil temperature were the principal contributors. Our results suggest that no tillage and residue mulching is conducive to increasing soil organic carbon and soil nutrient content, which is a beneficial conservation tillage measure for black soil protection in Sanjiang Plain of Northeast China. The no tillage with residue mulching, especially 60% residue mulching, alters soil bacterial community and highlights the importance of soil physicochemical properties in shaping the diversity and composition of the soil bacterial community. Our findings contribute to a broad understanding of the effects of no tillage and residue mulching on bacterial community differences and provide a scientific basis for the optimization of no tillage measures and sustainable utilization of the black soil of the Sanjiang Plain in Northeastern China.

Rice is sensitive to low temperatures, specifically at the booting stage. Chilling tolerance of rice is a quantitative trait loci that is governed by multiple genes, and thus, its precise identification through the conventional methods is an arduous task. In this study, we investigated the candidate genes related to chilling tolerance at the booting stage of rice. The F2 population was derived from Longjing25 (chilling-tolerant) and Longjing11 (chilling-sensitive) cross. Two bulked segregant analysis pools were constructed. A 0.82 Mb region containing 98 annotated genes on chromosomes 6 and 9 was recognized as the candidate region associated with chilling tolerance of rice at the booting stage. Transcriptomic analysis of Longjing25 and Longjing11 revealed 50 differentially expressed genes (DEGs) on the candidate intervals. KEGG pathway enrichment analysis of DEGs was performed. Nine pathways were found to be enriched, which contained 10 DEGs. A total of four genes had different expression patterns or levels between Longjing25 and Longjing11. Four out of the 10 DEGs were considered as potential candidate genes for chilling tolerance. This study will assist in the cloning of the candidate genes responsible for chilling tolerance and molecular breeding of rice for the development of chilling-tolerant rice varieties.

As one of the abiotic stresses, low temperature severely threatens rice production during its entire growth period, especially during the booting stage. In the present study, transcriptome analysis was performed comparing Longjing (LJ) 25 (chilling-tolerant) and LJ 11 (chilling-sensitive) rice varieties to identify genes associated with chilling tolerance in rice spikelets. A total of 23 845 expressed genes and 13 205 differentially expressed genes (DEGs) were identified, respectively. Gene ontology (GO) enrichment analyses revealed ‘response to cold’ (containing 180 DEGs) as the only category enriched in both varieties during the entire cold treatment period. Through MapMan analysis, we identified nine and six DEGs related to the Calvin cycle and antioxidant enzymes, respectively, including OsRBCS3, OsRBCS2, OsRBCS4, OsAPX2 and OsCATC, that under chilling stress were markedly downregulated in LJ11 compared with LJ25. Furthermore, we predicted their protein–protein interaction (PPI) network and identified nine hub genes (the threshold of co-expressed gene number ≥ 11) in Cytoscape, including three RuBisCO-related genes with 14 co-expressed genes. Under chilling stress, antioxidant enzyme activities (peroxidase (POD) and catalase (CAT)) were downregulated in LJ11 compared with LJ25. However, the content of malondialdehyde (MDA) was higher in LJ11 compared with LJ25. Collectively, our findings identify low temperature responsive genes that can be effectively used as candidate genes for molecular breeding programmes to increase the chilling tolerance of rice.

Human survival is impossible without oxygen, and as the body load continues to increase, the need for oxygen intake becomes greater. However, oxygen is also a double-edged sword for the human body. A large number of studies have proved that excessive intake of oxygen might lead to oxygen poisoning. Even under normal oxygen uptake, there is still a certain proportion of SOD conversion in oxygen. Superoxide dismutase (SOD) is one of the main causes of oxygen poisoning and chronic diseases. It is of great significance to study the changes of SOD in a large amount of oxygen environment. However, there are a few research studies in this field at home and abroad. Therefore, this paper puts forward the influence of basketball sports on serum superoxide dismutase (SOD) and its relationship with the nanoparticle drug delivery system. The research of this paper is mainly divided into three parts. The first part is the research of theoretical basis and core concepts. Through this part of the study, this paper shows that exercise can make the human body strong, while controlling the transformation of SOD, and only in this way can we achieve the true meaning of sports health. The second part is the establishment method of the test model of the influence of basketball on SOD and the nanoparticle drug delivery system. In this part, the principle and operation steps of the design method are given in detail. In order to ensure the effect of the experiment, the test standard was established, and the whole process data were recorded for the retrospective study. The third part is the comparative experiment, which includes the influence of different exercise intensities on SOD activity and the preservation stability of nanoparticles. Through the analysis of experimental data, it was found that basketball increased the risk of SOD transformation, but at the same time, using nanoparticles intervention can effectively reduce the harm of SOD to the human body.

Senecavirus A (SVA), an emerging swine picornavirus of swine, is one of the causative agents of vesicular disease which is clinically indistinguishable from foot-and-mouth disease in pigs. Here, 3 cases of vesicular disease were reported which was caused by SVA in November 2018 in Henan, China. Three new SVA strains were identified and conducted a genetically evolutionary analysis. The isolates shared 98.1–99.0% genomic pairwise identity to each other and had the highest similarity, of 98.3–98.7%, with the American strain KS15-01, respectively. Phylogenetic analysis indicated that the Chinese prevalent strains could be clearly divided into cluster 1, cluster 2, and cluster 3. Furthermore, one isolate (HeNNY-1/2018) and two previously reported strains (HB-CH-2016 and SVA/CHN/10/2017) were identified as recombinants using several algorithms. It revealed that the recombination among SVA strains has occurred in China since 2016 or earlier. The findings of studies updated the prevalent status of SVA in China. Besides, the genetic evolution and recombinant events of SVA should be attracted more attentions in the future.

The enteric nervous system (ENS) is derived from neural crest cells (NCCs). Defects in ENS NCCs colonizing in the intestines lead to an absence of enteric ganglia in the colon and results in Hirschsprung’s disease (HSCR). Bone morphogenetic proteins (BMPs) play diverse roles in the proliferation, migration and survival of ENS NCCs; however, whether BMPs are involved in HSCR and the underlying mechanism remains largely unknown. In this study, we found that BMP2 expression is significantly decreased in HSCR patients. Further experiments demonstrated that BMP2 is involved in the regulation of NCC proliferation, migration and differentiation. In a detailed analysis of the role of BMP2 in HSCR development in vivo, we demonstrated that BMP2b regulates the proliferation, migration and differentiation of vagal NCCs in zebrafish and that BMP2b is required for intestinal smooth muscle development. In addition, we showed that BMP2b is involved in regulating the expression of glial cell line-derived neurotrophic factor (GDNF) in the intestine, which mediates the regulation of ENS development by BMP2b in zebrafish. These results highlight a central role of the BMP-GDNF cascade in intestinal patterning and ENS development. Our results further demonstrate the key role of BMP2 in the etiology of HSCR in vitro and in vivo.

Palmprint gender classification can revolutionise the performance of authentication systems, reduce searching space and speed up matching rate. However, to the best of their knowledge, there is no literature addressing this issue. The authors design a new convolutional neural network (CNN) structure, fine‐tuning Visual Geometry Group Network, up to 19 layers to achieve a 20‐layer network, for palmprint gender classification. Experimental results show that the proposed structure could achieve good performance for gender classification. They also investigate palmprint images with 15 different kinds of spectra. They empirically find that a palmprint image acquired by the Blue spectrum could achieve 89.2% correct classification and could be considered as a suitable spectrum for gender classification. Obtaining high-quality plant materials for experiments is challenging for many research projects. Therefore, it is of special importance to determine the best method for preserving biological macromolecules like DNA, which degrade over time. Although some research has demonstrated that DNA degradation has little effect on traditional molecular markers, the effects of DNA degradation on ddRAD-seq, a popular reduced-representation sequencing technology, have not been adequately investigated. In this study, we first chose six woody bamboo species (Bambusoideae, Poaceae) to explore appropriate methods for preserving molecular materials with two DNA extraction approaches. Then we sequenced twenty-one bamboos and examined the effects of DNA quality on data generation using the ddRAD-seq technique (MiddRAD-seq). Finally, we reconstructed phylogenies of twenty woody bamboo species. We found that the integrity of dry-powdered DNA was preserved longer than that of TE-dissolved DNA, regardless of whether the DNA was extracted by a modified CTAB protocol or DNAsecure plant kit. The ddRAD-seq data were robust, except when DNA was severely degraded. In addition, we resolved the phylogenetic positions of the sampled Phyllostachys spp. Our results suggest that dry-powdered DNA is the most appropriate preservation method for plant molecular materials. Furthermore, a moderate level of DNA degradation has little effect on reduced representation sequencing techniques represented by ddRAD-seq.
