Visual SLAM technology has been widely used in all aspects of life, but has been applied relatively little in the field of weakly textured environments in agricultural fields. To make visual SLAM technology can be better applied in farmland environment, aiming at the problem of low accuracy of direct image recognition and matching in weak texture environment of farmland, A GMS-PROSAC feature point matching algorithm based on ORB feature point extraction is applied. This algorithm integrates the screening principle of the PROSAC algorithm with the smoothness constraint and multi-region generalization construction principle of the GMS algorithm, embedding a polar geometric constraint model that incorporates projection error fusion. It effectively overcomes the impact of light sensitivity and noise interference on matching accuracy. Due to its excessive plant repeatability, a judgment method is proposed to eliminate the wrong matching pairs under correct matching, so that the intelligent farm machine can correctly recognize the complex road conditions in different farmlands for reasonable harvesting. Through experimental verification, the highest matching accuracy can reach 86.0% in the weak texture environment of the farmland, which can be more effective for feature point extraction and matching, that provide new directions for subsequent unmanned agricultural machines that can harvest intelligently in different kinds of fields.

Cutaneous squamous cell carcinoma (cSCC) is the second most common non-melanoma skin cancer, among which 82% arise from actinic keratosis (AK) characterized by lesions of epidermal keratinocyte dysplasia. It is of great significance to uncover the progression mechanisms from AK to cSCC, which will facilitate the early therapeutic intervention of AK before malignant transformation. Thus, more and more studies are trying to ascertain the potential transformation mechanisms through multi-omics, including genetics, transcriptomics, and epigenetics. In this review, we gave an overview of the specific biomarkers and signaling pathways that may be involved in the pathogenesis from AK to cSCC, pointing out future possible molecular therapies for the early intervention of AK and cSCC. We also discussed current interventions on AK and cSCC, together with future perspectives.

Chitosan oligosaccharide (COS), a water-soluble derivative of chitin, has been recognized for its diverse biological properties. Deoxynivalenol (DON) is a prevalent mycotoxin, causing extreme liver damage. However, the mechanism whereby COS alleviates DON-induced liver injury remains unclear. In the present study, C57BL/6 mice were randomly divided into four groups: control (CON), DON (1.0 mg/d/kg BW DON), COS (200 mg/d/kg BW COS), and COS+DON (200 mg/d/kg BW COS + 1.0 mg/d/kg BW DON), with a period of 28 days. The results indicated that COS effectively reversed DON-induced weight loss, elevated liver index, and liver hemorrhage and swelling in mice. Moreover, COS significantly reduced liver reactive oxygen species (ROS) levels, malondialdehyde (MDA) content, and lactate dehydrogenase (LDH) release in DON-exposed mice, while restoring the activity of superoxide dismutase (SOD), catalase (CAT), glutathione peroxidase (GSH-Px) and total antioxidant capacity (T-AOC). Further investigations revealed that COS modulated the expressions of pro-inflammatory cytokines and anti-apoptotic proteins through stimulation of the Nrf2/HO-1 signaling pathway and suppression of the NF-κB signaling pathway. Additionally, COS inhibited ferroptosis by modulating the SLC7A11/GSH/GPX4 pathway and the expression of FTH1 and FLC proteins, thereby reducing lipid peroxidation accumulation and iron overload. In summary, this research showed that COS mitigated DON-induced liver injury in mice by alleviating DON-induced oxidative stress, inflammation, apoptosis, and ferroptosis via modulating the Nrf2/HO-1/NF-κB and GPX4 signaling pathways. These results offer a theoretical basis for the development and application of COS as a novel liver protectant and propose innovative therapeutic strategies for combating DON-induced liver damage.

PURPOSE: This study aimed to explore the relationship between the time from percutaneous vertebral augmentation (PVA) until subsequent fracture and the risk of new symptomatic fractures (NSFs) in untreated vertebrae at different distances from “augmented vertebrae”. METHODS: Patients who underwent PVA for the treatment of osteoporotic vertebral compression fractures at the West China Hospital of Sichuan University from May 2014 to April 2019 were retrospectively recruited. Vertebrae not treated during PVA were stratified based on their distance from the nearest augmented vertebra and the time elapsed since PVA. Survival curves were plotted to compare the risk of NSFs in untreated vertebrae at different distances from augmented vertebrae. The Cox proportional hazards model was used to identify risk factors of NSFs in untreated vertebrae. RESULTS: total, 162 patients with 228 NSFs (2.760 vertebrae) were analyzed. More than half of the NSFs (56.6%) occurred within the first year after PVA. Rates and hazard ratios (HRs) of NSFs were higher in vertebrae located one segment away from the augmented vertebrae (21.0%, HR: 3.99, P < 0.001), two segments away (10.6%, HR: 1.97, P = 0.003), or three segments away (10.5%, HR: 2.26, P < 0.001) than in vertebrae located five or more segments away (3.81%, HR: 1.00). Similar results were observed regardless of whether the untreated vertebrae were located in the thoracolumbar junction. In addition to distance, other risk factors of NSFs were the thoracolumbar location of untreated vertebrae, the number of augmented vertebrae, and percutaneous vertebroplasty. CONCLUSION: The risk of NSFs is greater for untreated vertebrae located closer to augmented vertebrae than for untreated vertebrae further away. This distance dependence occurs mainly within the three segments closest to the augmented vertebra. The risk of NSFs decreases with time after augmentation, and it is also related to the number of augmented vertebrae, the type of augmentation, and whether the untreated vertebrae are thoracolumbar or not.

Hidroacanthoma simplex (HS) is a rare skin appendage tumor that typically appears on the trunk and lower limbs in the elderly. Although HS is a predominantly benign condition, the presence of cellular atypia and dermal infiltration on histological examination indicates malignant HS (MHS). 5-aminolaevulinic acid photodynamic therapy (ALA-PDT) uses a photosensitizer and corresponding light source to cause irreversible damage or death of target cells through a photochemical reaction. Here, we reported the successful treatment of a MHS patient with ALA-PDT using plum-blossom needle pretreatment combined with CO2 laser. After five courses of ALA-PDT, the lesions were completely resolved, and the autonomic activity and smooth surface of the left ring finger were restored. This suggests that ALA-PDT is an effective, minimally invasive and safe treatment modality for MHS.

Abstract Background Syndrome coronavirus-2 (SARS-CoV-2) has developed various strategies to evade the antiviral impact of type I IFN. Non-structural proteins and auxiliary proteins have been extensively researched on their role in immune escape. Nevertheless, the detailed mechanisms of structural protein-induced immune evasion have not been well elucidated. Methods Human alveolar basal epithelial carcinoma cell line (A549) was stimulated with polyinosinic-polycytidylic acid (PIC) and independently transfected with four structural proteins expression plasmids, including nucleocapsid (N), spike (S), membrane (M) and envelope (E) proteins. By RT-qPCR and ELISA, the structural protein with the most pronounced inhibitory effects on IFN-β induction was screened. RNA-sequencing (RNA-Seq) and two differential analysis strategies were used to obtain differentially expressed genes associated with N protein inhibition of IFN-β induction. Based on DIANA-LncBase and StarBase databases, the interactive competitive endogenous RNA (ceRNA) network for N protein-associated genes was constructed. By combining single-cell sequencing data (GSE158055), lncRNA-miRNA-mRNA axis was further determined. Finally, RT-qPCR was utilized to illustrate the regulatory functions among components of the ceRNA axis. Results SARS-CoV-2 N protein inhibited IFN-β induction in human alveolar epithelial cells most significantly compared with other structural proteins. RNA-Seq data analysis revealed genes related to N protein inhibiting IFNs induction. The obtained 858 differentially expressed genes formed the reliable ceRNA network. The function of LINC01002-miR-4324-FRMD8 axis in the IFN-dominated immune evasion was further demonstrated through integrating single-cell sequencing data. Moreover, we validated that N protein could reverse the effect of PIC on LINC01002, FRMD8 and miR-4324 expression, and subsequently on IFN-β expression level. And LINC01002 could regulate the production of FRMD8 by inhibiting miR-4324. Conclusion SARS-CoV-2 N protein suppressed the induction of IFN-β by regulating LINC01002 which was as a ceRNA, sponging miR-4324 and participating in the regulation of FRMD8 mRNA. Our discovery provides new insights into early intervention therapy and drug development on SARS-CoV-2 infection.

Abstract Kidney diseases result from various causes, which can generally be divided into neoplastic and non-neoplastic diseases. Deep learning based on medical imaging is an established methodology for further data mining and an evolving field of expertise, which provides the possibility for precise management of kidney diseases. Recently, imaging-based deep learning has been widely applied to many clinical scenarios of kidney diseases including organ segmentation, lesion detection, differential diagnosis, surgical planning, and prognosis prediction, which can provide support for disease diagnosis and management. In this review, we will introduce the basic methodology of imaging-based deep learning and its recent clinical applications in neoplastic and non-neoplastic kidney diseases. Additionally, we further discuss its current challenges and future prospects and conclude that achieving data balance, addressing heterogeneity, and managing data size remain challenges for imaging-based deep learning. Meanwhile, the interpretability of algorithms, ethical risks, and barriers of bias assessment are also issues that require consideration in future development. We hope to provide urologists, nephrologists, and radiologists with clear ideas about imaging-based deep learning and reveal its great potential in clinical practice. Critical relevance statement The wide clinical applications of imaging-based deep learning in kidney diseases can help doctors to diagnose, treat, and manage patients with neoplastic or non-neoplastic renal diseases. Abstract Background Beyond the observed alterations in cellular structure and mitochondria, the mechanisms linking rare genetic mutations to the development of heart failure in patients affected by desmin mutations remain unclear due in part, to the lack of relevant human cardiomyocyte models. Methods To shed light on the role of mitochondria in these mechanisms, we investigated cardiomyocytes derived from human induced pluripotent stem cells carrying the heterozygous DES E439K mutation that were either isolated from a patient or generated by gene editing. To increase physiological relevance, cardiomyocytes were either cultured on an anisotropic micropatterned surface to obtain elongated and aligned cardiomyocytes, or as a cardiac spheroid to create a micro-tissue. Moreover, when applicable, results from cardiomyocytes were confirmed with heart biopsies of suddenly died patient of the same family harboring DES E439K mutation, and post-mortem heart samples from five control healthy donors. Results The heterozygous DES E439K mutation leads to dramatic changes in the overall cytoarchitecture of cardiomyocytes, including cell size and morphology. Most importantly, mutant cardiomyocytes display altered mitochondrial architecture, mitochondrial respiratory capacity and metabolic activity reminiscent of defects observed in patient’s heart tissue. Finally, to challenge the pathological mechanism, we transferred normal mitochondria inside the mutant cardiomyocytes and demonstrated that this treatment was able to restore mitochondrial and contractile functions of cardiomyocytes. Conclusions This work highlights the deleterious effects of DES E439K mutation, demonstrates the crucial role of mitochondrial abnormalities in the pathophysiology of desmin-related cardiomyopathy, and opens up new potential therapeutic perspectives for this disease.

Abstract Investigation of the choroid plexus in schizophrenia has seen growing interest due to its role in the interaction between neuroinflammation and brain dysfunction. Most previous studies included treated and long-term ill patients, while antipsychotics and illness course might both affect the choroid plexus. Here, we recruited first-episode antipsychotic-naïve schizophrenia patients, performed high-resolution structural brain scan and manually extracted choroid plexus volume. Choroid plexus volume was compared between patients and healthy controls after controlling for age, sex and intracranial volume. Age and sex effects were examined on choroid plexus volume in patient and healthy control groups respectively. In patients, we also examined the correlation of choroid plexus volume with volume measures of cortical and subcortical gray matter, white matter, lateral ventricular as well as symptom severity and cognitive function. Schizophrenia patients showed significantly enlarged choroid plexus volume compared with healthy controls. Choroid plexus volume was positively correlated with age in only patient group and we found significantly larger choroid plexus volumes in males than females in both patient and healthy control groups, while the sex effects did not differ between groups. Choroid plexus volume was only found correlated with lateral ventricular volume among the brain volume measures. No significant correlation between choroid plexus volume and clinical ratings or cognitive performance was observed. Without potential confounding effects of pharmacotherapy or illness course, our findings indicated the enlargement of choroid plexus in schizophrenia might be an enduring trait for schizophrenia.

Abstract Background As a histone methyltransferase, suppressor of variegation 3–9 homolog 1 (SUV39H1) plays an important role in the occurrence and development of cancer. To explore the mechanism and biological function of SUV39H1 in hepatitis B virus-associated hepatocellular carcinoma (HBV-HCC) can gain an insight into the pathogenesis of HBV-HCC. Methods The effect of HBV infection on SUV39H1 in hepatoma cells was detected. CCK-8, colony growth assay and wound healing assay were used to assess the proliferation and migration of HBV-positive hepatoma cells. RNA sequencing (RNA-seq) was applied to find differential genes and enriched pathways. The serum SUV39H1 level in HBV-HCC patients was detected and its correlation with clinical indicators was analyzed. Results SUV39H1 was increased by HBV infection and promoted the proliferation and migration of hepatoma cells. SUV39H1 could upregulate the expression of mitochondrial oxidative phosphorylation (OXPHOS) pathway-related genes. OXPHOS pathway inhibitors could reduce the capacity of proliferation and migration of hepatoma cells after overexpressing SUV39H1. Serum SUV39H1 levels were higher in chronic hepatitis B (CHB) patients than in healthy controls and higher in HBV-HCC patients than in CHB patients. In the diagnosis of HCC, the predictive value of SUV39H1 combined with alpha-fetoprotein (AFP) was better than that of AFP alone. Conclusion SUV39H1 is regulated by HBV infection and promotes the proliferation and migration of hepatoma cells by targeting OXPHOS pathway. It indicates that SUV39H1 may be a new biomarker of the diagnosis of HCC.

ObjectiveThis study aimed to investigate the association between the risk of suicidal behaviors and student-supervisor relationships and subjective family socioeconomic status (SFSS) in medical graduate students, and to propose preventive strategies to reduce the suicidal risk among medical graduate students.Materials and methodsA total of 1,310 validated questionnaires were collected from medical graduate students, which included demographic information, study programs, the Suicidal Behaviors Questionnaire-Revised (SBQ-R) questionnaire, the Leader-Member Exchange 7 (LMX-7) questionnaire, and SFSS by MacArthur Scale. Multiple regression analysis was employed to examine the associations between variables and adjust for confounders. A moderation analysis, containing simple slope analysis and Johnson-Neyman interval plots were used to analyze the moderating effect of the SFSS in the association of SBQ-R and LMX-7 scores.ResultsA total of 88 participants (6.7%) were at risk of suicidal behaviors. In the high-quality student-supervisor relationship group (LMX-7 score ≥ 25), SFSS was significantly higher than in the low- and moderate-quality relationship group (p=0.002). Myofibrillar myopathy (MFM) is a rare genetic disorder characterized by muscular dystrophy that is often associated with cardiac disease. This disease is caused by mutations in several genes, among them DES (encoding desmin) is the most frequently affected. Peripheral blood mononuclear cells from 5 different MFM patients with different DES mutations were reprogrammed into induced pluripotent stem cells (IPSC) using non-integrative vectors. For each patient, one IPSC clone was selected and demonstrated pluripotency hallmarks without genomic abnormalities. SNP profiles were identical to the cells of origin and all the clones have the capacity to differentiate into all three germ layers.

Abstract Aim Hydrogels with excellent biocompatibility and biodegradability can be used as the desirable dressings for the therapy of diabetic foot ulcer (DFU). This review aimed to summarize the biological functions of hydrogels, combining with the pathogenesis of DFU. Methods The studies in the last 10 years were searched and summarized from the online database PubMed using a combination of keywords such as hydrogel and diabetes. The biological functions of hydrogels and their healing mechanism on DFU were elaborated. Results In this review, hydrogels were classified by their active substances such as drugs, cytokines, photosensitizers, and biomimetic peptide. Based on this, the biological functions of hydrogels were summarized by associating the pathogenesis of DFU, including oxidative stress, chronic inflammation, cell phenotype change, vasculopathy, and infection. This review also pointed out some of the shortcomings of hydrogels in present researches. Conclusions Hydrogels were classified into carrier hydrogels and self‐functioning hydrogels in this review. Besides, the functions and components of existing hydrogels were clarified to provide assistance for future researches and clinical applications.

Abstract Background Large-scale medical equipment, which is extensively implemented in medical services, is of vital importance for diagnosis but vulnerable to various anomalies and failures. Most hospitals that conduct regular maintenance have been suffering from medical equipment-related incidents for years. Currently, the Internet of Medical Things (IoMT) has emerged as a crucial tool in monitoring the real-time status of the medical equipment. In this paper, we develop an IoMT system of Computed Tomography (CT) equipment in the West China Hospital, Sichuan University and collected the system status time-series data. Novel multivariate time-series classification models and frameworks are proposed to predict the anomalies of CT equipment. The important features that are closely related to the equipment anomalies are identified with the model. Methods We extracted the real-time CT equipment status time-series data of 11 equipment between May 19, 2020 and May 19, 2021 from the IoMT, which includes the equipment oil temperature, anode voltage, etc. The arcs are identified as labels of anomalies due to their relationship with decreased imaging quality and CT equipment failures. To improve prediction accuracy, the statistics and transformations of the raw historical time-series data segment in the sliding time window are used to construct new features. Due to the particularity of time-series data, two frameworks are proposed for splitting the training and test sets. Then the Decision Tree, Support Vector Machine, Logistic Regression, Naive Bayesian, and K-Nearest Neighbor classification models are used to classify the system status. We also compare our model to state-of-the-art models. Results The results show that the anomaly prediction accuracy and recall of our method are 79% and 77%, respectively. The oil temperature and anode voltage are identified as the decisive features that may lead to anomalies. The proposed model outperforms the others when predicting the anomalies of the CT equipment based on our dataset. Conclusions The proposed method could predict the state of CT equipment and be used as a reference for practical maintenance, where unexpected anomalies of medical equipment could be reduced. It also brings new insights into how to handle non-uniform and imbalanced time series data in practical cases.

Abstract Objective This study aimed to assess the feasibility of software-aided selection of monoenergetic level for acute necrotising pancreatitis (ANP) depiction compared to other automatic image series generated using dual-energy computed tomography (CT). Methods The contrast-enhanced dual-source dual-energy CT images in the portal venous phase of 48 patients with ANP were retrospectively analysed. Contrast-to-noise ratio (CNR) of pancreatic parenchyma-to-necrosis, signal-to-noise ratio (SNR) of the pancreas, image noise, and score of subjective diagnosis were measured, calculated, and compared among the CT images of 100 kV, Sn140 kV, weighted-average 120 kV, and optimal single-energy level for CNR. Results CNR of pancreatic parenchyma-to-necrosis in the images of 100 kV, Sn140 kV, weighted-average 120 kV, and the optimal single-energy level for CNR was 5.18 ± 2.39, 3.13 ± 1.35, 5.69 ± 2.35, and 9.99 ± 5.86, respectively; SNR of the pancreas in each group was 6.31 ± 2.77, 4.27 ± 1.56, 7.21 ± 2.69, and 11.83 ± 6.30, respectively; image noise in each group was 18.78 ± 5.20, 17.79 ± 4.63, 13.28 ± 3.13, and 9.31 ± 2.96, respectively; and score of subjective diagnosis in each group was 3.56 ± 0.50, 3.00 ± 0.55, 3.48 ± 0.55, and 3.88 ± 0.33, respectively. Currently, graph convolutional network (GCN) is widely used in relation extraction (RE) tasks. Within RE tasks in the form of directed graphs, the placement of entities in the sentence context generates a large number of remote entity nodes in the directed graph. However, the GCN has an acceptance domain limitation and can only aggregate consecutive neighbor nodes, making it ineffective for remote node information extraction. To tackle the above issues, a new model GCN based on weighted diffusion is proposed, including two improvements. First, the pretrained BERT model is introduced to extract the semantic features from the original sentences, and then the diffusion convolution is modified by the weighted matrix using the Gaussian kernel function to calculate the distance of nodes on the graph, enabling the diffusion convolution to selectively aggregate information of remote nodes. The effect of remote node information aggregation can be improved by shortening the distance of related nodes and increasing the distance of unrelated nodes. Second, an identity mapping is added to ensure that the weight matrix can match each layer adaptively. The efficacy of our proposed model has been demonstrated through experimental results on two publicly available datasets, TACRED and SemEval, which have shown that it outperforms other baseline models.

A number of genetic variants in the SYNM gene encoding for the intermediate filament synemin have been reported in patients with cardiomyopathies, skeletal myopathies, cancer and certain neurodegenerative disorders. To better understand its role, we generated a human induced pluripotent stem cell line with a homozygous deletion in the SYNM gene by CRISPR/Cas9 genome editing. The synemin-knockout human induced pluripotent stem cells exhibit typical morphology of pluripotent cells, expression of pluripotency markers, normal karyotype and differentiation capacity in the three germ layers. This line will allow us to investigate the role of synemin in cardiomyopathy upon differentiation into beating cardiomyocytes.

As the pioneering review in the field of ultrasonic defect detection for oil and gas pipelines based on bibliometric methods, this study employs visual analysis to identify the most influential countries, academic institutions, and journals in this domain. Through cluster analysis, it determines the primary trends, research hotspots, and future directions in this critical field. Starting from the current global industrial ultrasonic in-line inspection (ILI) detection level, this paper provides a flowchart for selecting detection methods and a table for defect comparison, detailing the comparative performance limits of different detection devices. It offers a comprehensive perspective on the latest ultrasonic pipeline detection technology from laboratory experiments to industrial practice.

To address the problem of system instability during vehicle low-speed driving, we propose improving the visual odometer using ORB (Oriented FAST and Rotated BRIEF) features. The homogeneity of ORB features leads to poor corner point properties of some feature points. When the environmental texture lacks richness, it leads to poor matching performance and low matching accuracy of the feature points. We solve the problem of the corner point properties of feature points using weight calculation for regions with different textures. When the vehicle speed is too low, the continuous frames captured by the camera will overlap significantly, causing large fluctuations in the system error. We use motion model estimation to solve this problem. Meanwhile, experimental validation using the KITTI dataset achieves good results.

Osteosarcoma (OS) is a highly metastatic bone cancer that usually affects children. Rhizoma Paridis saponins (RPS) have been identified to show a broad-spectrum anti-tumor activity. Our previous study has identified vasculogenic mimicry (VM) as an indicator of poor prognosis for OS. Rhizoma Paridis ethanol extract exhibits potent anti-OS property. However, the anti-metastatic effect of RPS on OS and the detailed mechanisms remain unknown. RPS was characterized by liquid chromatography/quadrupole time-of-flight mass spectrometry (LC/Q-TOF/MS) analysis. The anti-OS, anti-metastasis and anti-VM activities of RPS were investigated using in vitro biological assays and a xenograft mouse model. Western blot, qRT-PCR, ELISA, Phalloidin staining and immunohistochemistry assays were conducted to investigate the molecular mechanism of RPS. A total of 34 phytochemicals from RPS were identified by LC/Q-TOF/MS. RPS dose-dependently suppressed the OS cell proliferation, metastasis and VM formation in vitro and in vivo. Mechanically, we found that RPS downregulated migration-inducing gene 7 (MIG-7) expression, resulting in inhibition of the PI3K/MMPs/Ln-5γ2 pathway and cell protrusion formation. Additionally, we confirmed that RPS downregulated MIG-7 by upregulating miR-520d-3p expression. Our results suggests that RPS inhibits the VM formation and metastasis of OS by modulating the miR-520d-3p/MIG-7 signaling axis.

Cell migration–inducing protein (CEMIP), also known as KIAA1199 and hyaluronan-binding protein involved in hyaluronan depolymerization, is a new member of the hyaluronidase family that degrades hyaluronic acid (HA) and remodels the extracellular matrix. In recent years, some studies have reported that CEMIP can promote the proliferation, invasion, and adhesion of various tumor cells and can play an important role in bacterial infection and arthritis. This review focuses on the pathological mechanism of CEMIP in a variety of diseases and expounds the function of CEMIP from the aspects of inhibiting cell apoptosis, promoting HA degradation, inducing inflammatory responses and related phosphorylation, adjusting cellular microenvironment, and regulating tissue fibrosis. The diagnosis and treatment strategies targeting CEMIP are also summarized. The various functions of CEMIP show its great potential application value.

Shale pore structure characterization is key to shale reservoir evaluation, sweet spot selection, and economic exploitation. It remains a challenge to accurately characterize shale micro-nano pores. Common experimental characterization methods for shale pore systems are listed, and advantages and weaknesses of each method are analyzed. An improved pore structure characterization method for shale is proposed by combining Helium and NMR. The new method does not affect shale samples and has a higher accuracy. The affecting factors for shale pore evolution for shale are also discussed, showing that organic matter content and maturity are key factors in total porosity development. Furthermore, a shale porosity–TOC maturity relationship chart is developed based on the experimental data of shale samples selected from six shale reservoirs. The application of this chart in Well X in the Gulong field of Songliao Basin proves its utility in evaluating shale reservoirs.

Objective: To explore the application of noise index combined with automatic tube current regulation technology in the one-stop scanning of chest and lumbar QCT physical examination. Method: 100 patients who unferwent thoracic CT and lumbar QCT scan in our medical examination were prospectively collected and randomly divided into two groups( conventional dose group and ow dose group). There were 50 patients in the conventianl group (noise index set to NI=8.5) and 50 patients in the low dose group (noise index set to NI=14). Their height, weight and BMI were recorded, as well as the mean BMD of L1, L2 in both radiation and low dose groups, and L1, L2 in the conventional dose group. The differences of radiation dose and mean bone mineral density between the two groups were compared. Results: The average scanning dose was 387.65 mGy in the conventional dose group and 73.18 mGy in the low dose group. There was statistical differences in radiation dose between the two groups. The mean bone mineral density (BMD) of L1 and L2 was 119.71 in the conventional dose group, and that of L1 and L2 was 123.65 in the low dose group. There was no statistical difference between the two groups. Conclusion: Low dose CT scan of thoracic and lumbar spine can not only meet the needs of lung screening and bone mineral density assessment but also greatly reduce radiation dose. It can be widely used in physical examination.

Abstract Background High-resolution computed tomography (HRCT) is recommended diagnosing and monitoring connective tissue disease-associated interstitial lung disease (CTD-ILD). Quantitative computed tomography has the potential to precisely assess the radiological severity of CTD-ILD, but has still been under study. Objective To investigate whether dual-energy computed tomography (DECT), a novel quantitative technique, can be used for quantitative severity assessment in CTD-ILD. Methods This cross sectional study recruited adult CTD-ILD patients who underwent DECT scans from the ICE study between October 2019 and November 2021. DECT parameters, including effective atomic number (Zeff), lung (lobe) volume, and monochromatic CT number (MCTN) of each lung lobe, were evaluated. CTD-ILD was classified into extensive CTD-ILD and limited CTD-ILD by staging algorithm using combined forced vital capacity (FVC)%predicted and total extent of ILD (TEI) on CT. Dyspnea, cough, and life quality were scored by Borg dyspnea score, Leicester cough questionnaire (LCQ), and short-form 36 health survey questionnaire (SF-36), respectively. Wallerian degeneration, the progressive disintegration of distal axons and myelin that occurs after peripheral nerve injury, is essential for creating a permissive microenvironment for nerve regeneration, and involves cytoskeletal reconstruction. However, it is unclear whether microtubule dynamics play a role in this process. To address this, we treated cultured sciatic nerve explants, an in vitro model of Wallerian degeneration, with the microtubule-targeting agents paclitaxel and nocodazole. We found that paclitaxel-induced microtubule stabilization promoted axon and myelin degeneration and Schwann cell dedifferentiation, whereas nocodazole-induced microtubule destabilization inhibited these processes. Evaluation of an in vivo model of peripheral nerve injury showed that treatment with paclitaxel or nocodazole accelerated or attenuated axonal regeneration, as well as functional recovery of nerve conduction and target muscle and motor behavior, respectively. These results suggest that microtubule dynamics participate in peripheral nerve regeneration after injury by affecting Wallerian degeneration. Innovative approaches are under research to study the resistance of chloride ion penetration in concrete containing chloride ions, to minimize the impact of chloride ion penetration test errors in coastal reinforced concrete (RC), which is helpful to the design of coastal RC structures. In this study, the diffusion depth, free ion concentration and diffusion coefficient of chloride, iodide ions with different curing ages and GGBFS content were measured by the Rapid Chloride Migration Test (RCM) and Rapid Iodide Migration tests (RIM). The SEM-EDS and MIP were used to analyze the microstructures, pore size distribution and the hydrated products. The results show that the performance of GGBFS concrete against the diffusion of corrosive ions is affected by the curing age and the content of GGBFS. With the increase of GGBFS content, especially concrete with 60% GGBFS, the influence of chloride, iodide ion penetrating into concrete gradually becomes smaller. The long-age curing system is more conducive to the concrete resistance to the migration and diffusion of chloride, iodine ions. Compared with the ordinary concrete, the total porosity of concrete mixed with GGBFS is lower, the internal microstructures have fewer cracks and defects, the density is better, and the diffusion coefficient of chloride and iodide ions is also lower. In addition, using the concept of corrosive ion adjustment coefficient (conversion coefficient of diffusion between chloride ion and iodide ion) and applying the data regression analysis (DRA), it is found that there is a good quadratic parabolic function relationship between the GGBFS content and the ions adjustment coefficient.

Abstract Background Silent information regulator 6 (SIRT6) is a mammalian homolog of the nicotinamide adenine dinucleotide (NAD)-dependent deacetylase sirtuin family. Prior evidences suggested that the anti-inflammatory function of SIRT6 after spinal cord and brain injury, and it plays a crucial role in macrophages polarization of adipose tissue and skin. However, the role of SIRT6 in macrophages involved peripheral nerve injury is still unknown. Given the prominent role of macrophages in peripheral nerve recovery, we aim to investigate the role of SIRT6 in the regulation of phenotypes shift and functions in macrophages after peripheral nerve injury. Results In the present study, we first identified a significant increase of SIRT6 expression during nerve degeneration and macrophages phagocytosis. Next, we found nerve recovery was delayed after SIRT6 silencing by injected shRNA lentivirus into the crushed sciatic nerve, which exhibited a reduced expression of myelin-related proteins (e.g., MAG and MBP), severer myoatrophy of target muscles, and inferior nerve conduction compared to the shRNA control injected mice. In vitro, we found that SIRT6 inhibition by being treated with a selective inhibitor OSS_128167 or lentivirus transfection impairs migration and phagocytosis capacity of bone marrow-derived macrophages (BMDM). In addition, SIRT6 expression was discovered to be reduced after M1 polarization, but SIRT6 was enhanced after M2 polarization in the monocyte-macrophage cell line RAW264.7 and BMDM. Moreover, SIRT6 inhibition increased M1 macrophage polarization with a concomitant decrease in M2 polarization both in RAW264.7 and BMDM via activating NF-κB and TNF-α expression, and SIRT6 activation by UBCS039 treatment could shift the macrophages from M1 to M2 phenotype. Conclusion Our findings indicate that SIRT6 inhibition impairs peripheral nerve repair through suppressing the migration, phagocytosis, and M2 polarization of macrophages. Therefore, SIRT6 may become a favorable therapeutic target for peripheral nerve injury.

Engineered cement-based composites exhibit excellent deformability, mechanical behavior, fresh performance and durability. However, the traditional cement-based composites incorporating high volume ordinary Portland cement would lead to high carbon footprint. In this study, a new and eco-efficient engineered cement-based composites was designed by incorporating Polypropylene fibers (PPF) and eco-friendly cement with limestone calcined clay (LC3-ECCs). The LC3-ECCs were analyzed and discussed in terms of mechanical properties, microscopic morphology, hydration products and porosity. The study found that the 28-days compressive behavior was above 44.2 MPa, and the flexural strength remained above 4.8 MPa. Because of the formation and gathering of highly polymerized compound products (C-S-H gel, C-A-S-H gel) in the matrix and plenty of ettringite, the bonding effect between PPF and LC3 cementitious matrix is better. Additionally, the LC3-ECCs with 1.5 % volume content of PP fiber showed less porosity beneficial to the mechanical behavior. This study suggests that low-carbon LC3 has the potential to be successfully utilized as the alternative to OPC and is suitable to design sustainable ECCs, and this low-carbon construction product can be also generally applied into some area with the abundant clay sources.

China’s Guizhou is a typical karst landscape province with high production of chili pepper, but it is mostly planted in mountainous areas, while manual harvesting of chili pepper has the deficiencies of high labor intensity, low efficiency, and high labor cost; in addition, there is no harvesting machinery applicable to the dense planting pattern of the chili pepper in mountainous areas in China. The fully hydraulic mountain track-based self-propelled pepper harvester 4JZ-1.0A is designed to solve the above problems. The pepper harvester spiral comb picking head is an important part of the whole machine design, the design of the hydraulic system of the working circuit of the picking head is the key to realizing the hydraulic control part of the whole system. In this paper, the working principle diagram of the improved load-sensitive hydraulic system is designed and analyzed for the study of whether the working circuit of the pepper picking head of the pepper machine can meet the requirements of mountain operation, taking the working circuit of the mountain pepper harvester as the research object. In addition, the load-sensitive pump model and the simulation model of the whole working circuit are established by the AMESim platform 2019.2 (Siemens simcenter amesim). The operating performance of the system under variable flow conditions, variable load conditions, and an improved sensitive system is analyzed. The simulation results show that the improved load-sensitive system can effectively reduce the oscillation and cavitation during cylinder operation and improve the system efficiency and the performance and service life of the components. The performance of the hydraulic system of the working circuit of the mountain pepper harvester was verified in the test, meeting the requirements of working use. This provides a theoretical basis for the improvement and optimal design of a mountain pepper harvester hydraulic system.

Limestone calcined clay cement (LC3) is a new type of low-carbon cement that can reduce energy consumption and carbon dioxide emissions while meeting the performance requirements of ordinary cement. In this study, polypropylene (PP) fibers were mixed into limestone calcined clay cement-based materials to make new low-carbon ECCs. In this study, a total of 24 sets of specimens were designed for 4 groups of curing ages and 6 types of mix ratios. The compressive load–displacement data were measured the compressive curve characteristics were analyzed then, a compressive constitutive model of the composites was deduced and obtained. Through XRD, SEM-EDS and MIP experiments, the reasons and laws of the compressive strength ranges of adding PP fibers and LC3 to engineered cementitious composites (LC3-PP-ECCs) are further explained from the perspective of the pore size, microstructures and hydration products. The results show that, after 28 days, the compressive strength values of LC3-PP-ECCs generally decreases with increasing PP fiber content and the combined effect of PP fibers and hydration products causes the compressive strength of LC3-ECCs with 0.5% PP fibers to drop sharply. In addition, the specimens showed better properties in terms of toughness, ductility and energy absorption. However, in the microstructures, the addition of PP fibers will cause more internal defects and flaws. This results of this study can provide some theoretical experience and technical support for the engineering application of LC3-ECCs.

BackgroundChronic hepatitis B (CHB) remains a significant global health problem, leading to recurrent inflammation and liver-damaging diseases such as fibrosis, cirrhosis, and hepatocellular carcinoma (HCC). Currently, although diagnostic markers for CHB are well established, the indicators for predicting liver injury caused by hepatitis B virus (HBV) infection still need to be further explored. Thus, the identification of credible infectious indicators is urgently needed to facilitate timely clinical intervention and avoid the progression of disease malignancy.MethodsThe Gene Expression Omnibus (GEO) database GSE83148 data set was used to explore the hub genes for HBV infection. The quantitative real-time polymerase chain reaction (qPCR) was used to identify the impact of HBV infection on the expression of hub gene at the cell level. At the same time, serum samples and clinical information were collected from healthy, HBV-free and CHB patients. The enzyme-linked immunosorbent assay (ELISA) was used to verify the results of cell experiments and Pearson correlation analysis was used to clarify hub genes correlation with HBV infection indicators and liver injury-related indicators. Finally, the Gene Expression Profiling Interactive Analysis (GEPIA) database was used to analyze the differences in the expression of hub gene in liver injury diseases.ResultsChemokine (C-X-C motif) ligand (CXCL)8, CXCL9, CXCL10, and CXCL11 were identified as hub genes in HBV infection. After HBV infection, the expression of the four chemokines was significantly increased and the concentrations secreted into serum were also increased. Moreover, the four chemokines were significantly correlated with HBV infection-related indicators and liver injury-related indicators, which were positively correlated with alanine aminotransferase (ALT), aspartate aminotransferase (AST) and hepatitis B e antigen (HBeAg), and negatively correlated with AST/ALT ratio and hepatitis B core antibody (HBcAb). In addition, the expression of CXCL9, CXCL10, and CXCL11 in HCC tissues was significantly higher than in normal tissues.ConclusionUsing a combination of bioinformatics, cell experiments, and clinical correlation analysis, this study showed that CXCL8, CXCL9, CXCL10, and CXCL11 can be used as serum biomarkers to forecast liver injury caused by HBV infection.

In order to explore the measurement law of ultrasonic flowmeters under different rectifiers, a calibration pipeline model with rectifiers was established, the flow field of pipelines with different rectifiers was numerically simulated by Fluent software, and the indication errors of the ultrasonic flowmeters were obtained. Meanwhile, the accuracy of the numerical simulation method was verified based on the filed application data of flowmeters. The three types of rectifiers and ultrasonic flowmeters arranged on A, B and C channels were selected with the numerical simulation method, and the reasons for the measurement variations of ultrasonic flowmeters caused by the rectifiers were analyzed through the flow field distribution in the pipeline and the velocity distribution in the cross section at the flowmeter. The results show that different indication errors of different types of ultrasonic flowmeter are resulted from the three types of rectifiers. Specifically, the maximum and minimum differences of indication error for type A ultrasonic flowmeter are about 0.5% and 0.2%, that for type B ultrasonic flowmeter are about 0.1% and 0.05%, and that for type C ultrasonic flowmeter are about 0.4% and 0.2%, respectively. Base on that, suggestions are put forward for the installation and verification of ultrasonic flowmeters, which have guiding significance for improving the calibration quality of flowmeters.

Abstract Skeletal muscle has a remarkable capacity of regeneration after injury, but the regulatory network underlying this repair process remains elusive. RNA-binding proteins play key roles in the post-transcriptional regulation of gene expression and the maintenance of tissue homeostasis and plasticity. Rbm24 regulates myogenic differentiation during early development, but its implication in adult muscle is poorly understood. Here we show that it exerts multiple functions in muscle regeneration. Consistent with its dynamic subcellular localization during embryonic muscle development, Rbm24 also displays cytoplasm to nucleus translocation during C2C12 myoblast differentiation. In adult mice, Rbm24 mRNA is enriched in slow-twitch muscles along with myogenin mRNA. The protein displays nuclear localization in both slow and fast myofibers. Upon injury, Rbm24 is rapidly upregulated in regenerating myofibers and accumulates in the myonucleus of nascent myofibers. Through satellite cell transplantation, we demonstrate that Rbm24 functions sequentially to regulate myogenic differentiation and muscle regeneration. It is required for myogenin expression at early stages of muscle injury and for muscle-specific pre-mRNA alternative splicing at late stages of regeneration. These results identify Rbm24 as a multifaceted regulator of myoblast differentiation. They provide insights into the molecular pathway orchestrating the expression of myogenic factors and muscle functional proteins during regeneration.

During the coronavirus disease 2019 (COVID-19) pandemic period, container computed tomography (CT) scanners were developed and used for the first time in China to perform CT examinations for patients with clinically mild to moderate COVID-19 who did not need to be hospitalized for comprehensive treatment, but needed to be isolated in Fangcang shelter hospitals (also known as makeshift hospitals) to receive some supportive treatment. The container CT is a multidetector CT scanner installed within a radiation-protected stand-alone container (a detachable lead shielding room) that is deployed outside the makeshift hospital buildings. The container CT approach provided various medical institutions with the solution not only for rapid CT installation and high adaptability to site environments, but also for significantly minimizing the risk of cross-infection between radiological personnel and patients during CT examination in the pandemic. In this article, we described the typical setup of a container CT and how it worked for chest CT examinations in Wuhan city, the epicenter of COVID-19 outbreak.

Abstract Background Secondary hyperparathyroidism is a common complication of end-stage renal disease (ESRD), which may be associated with cardiovascular diseases. Thus, this study aimed to explore myocardial damage using non-contrast cardiovascular magnetic resonance (CMR) in ESRD patients undergoing hemodialysis and further investigate its relationship with parathyroid hormone (PTH) toxicity. Methods Seventy-two adult ESRD patients receiving regular hemodialysis and 30 healthy subjects underwent CMR examination. Continuous CMR cine sections from the mitral valve level to the left ventricular (LV) apex in the short-axis plane, cine series of vertical two-chamber long-axis plane, and horizontal four-chamber plane were acquired. Native T1 mapping was obtained using modified Look-Locker inversion recovery (MOLLI) sequences. Native T1 values and myocardial strain were analyzed. Immunoreactive parathyroid hormone (iPTH) was obtained from all enrolled patients. Results Forty (55.6%) hemodialysis ESRD patients were found to have increased iPTH levels. LV ejection fraction (LVEF) of both ESRD patients with targeted and increased iPTH levels was decreased compared with healthy subjects (55.9 ± 12.0% vs. 65.0 ± 4.5%; 51.7 ± 12.8 vs. 65.0 ± 4.5%, both P

Wallerian degeneration occurs after peripheral nerve injury and provides a beneficial microenvironment for nerve regeneration. Our previous study demonstrated that ascorbic acid promotes peripheral nerve regeneration, possibly through promoting Schwann cell proliferation and phagocytosis and enhancing macrophage proliferation, migration, and phagocytosis. Because Schwann cells and macrophages are the main cells involved in Wallerian degeneration, we speculated that ascorbic acid may accelerate this degenerative process. To test this hypothesis, 400 mg/kg ascorbic acid was administered intragastrically immediately after sciatic nerve transection, and 200 mg/kg ascorbic acid was then administered intragastrically every day. In addition, rat sciatic nerve explants were treated with 200 μM ascorbic acid. Ascorbic acid significantly accelerated the degradation of myelin basic protein-positive myelin and neurofilament 200-positive axons in both the transected nerves and nerve explants. Furthermore, ascorbic acid inhibited myelin-associated glycoprotein expression, increased c-Jun expression in Schwann cells, and increased both the number of macrophages and the amount of myelin fragments in the macrophages. These findings suggest that ascorbic acid accelerates Wallerian degeneration by accelerating the degeneration of axons and myelin in the injured nerve, promoting the dedifferentiation of Schwann cells, and enhancing macrophage recruitment and phagocytosis. In order to study the physical properties of RP-3 aviation kerosene and its impurities, three tanks from different oil inlet and outlet units were sampled during the receiving and dispatching operation of an airport oil depot in North China, and the samples of aviation kerosene and its solid impurities were obtained by filtration. Further, the density, viscosity and water content of aviation kerosene sample were measured with a densimeter, a capillary viscometer, a distillation flask and other experimental instruments, and for the solid impurities, electron microscope scanning and energy dispersive spectrum analysis were carried out. The results show that the kinematic viscosity, density and water content of aviation kerosene in the three tanks are 1.651 2-1.868 8 mm2/s, 0.794 9-0.795 6 g/cm3 and 0.007%-0.009% respectively. The solid impurity particles are spherical, rod-shaped, long block-shaped or tetrahedral. The particles are mainly composed of nonmetallic elements such as C, H, O and Si, and metallic elements such as Ca, Al, Mg, Fe, Mn, Co, Ti, K and W. The particle size is ranged of 8-1.5×104 μm, and the impurity particles sized less than 150 μm, 50-100 μm, 30-50 μm and other size range account for about 83%, 15%, 2.3% and 0.1% respectively. The impurity particles more than 150 μm are easy to be separated, while those less than 150 μm are hard to be separated with the conventional methods. Taking the impurity particles sized 20-150 μm for example, the density was measured by pycnometer method, and the density change function model was fitted on this basis, which could provide effective guidance for separation of aviation kerosene solid impurities.

In order to clarify the top corrosion mechanism of X65 wet gas pipeline in Yanchang Gasfield and to improve thecorrosion protection effect of the pipeline, the coupon comparison test was carried out using a self-made tube top corrosionsimulation test device with high temperature and high pressure, and the variation of corrosion rate of bottom and topcoupons with the time and the morphology characteristics of the surface and cross-section of corrosion coupons were studiedby means of scanning electron microscope. The results indicate that the uniform corrosion rate at the bottom of pipelinedecreases with time, but that on the top has no obvious variation. The uniform corrosion rate at the bottom and on the topvaries greatly, close to 100%. The bottom corrosion is pitted and the corrosion pits tend to connect as a whole. However, obvious local corrosion can be observed on the top, and the top corrosion spots are greater than the bottom corrosion pits.The corrosion product film at the bottom is thick and distributed uniformly, generally having good protective effect to thecoupons. But the corrosion product film on the top is thin and distributed unevenly. No accumulation of corrosion productappears around the top corrosion, and the local corrosion expands rapidly. Most of the top corrosion is localized, with agreater threat to the safe operation of wet gas pipeline.

To study the pressure pulsation characteristics of the cryogenic expander, the numerical simulation was performed on the expander hydraulic model by the numerical simulation method. Firstly, external characteristics of expander were obtained by steady numerical simulation. Simulation results were compared with test results, verifying the accuracy of simulation results. The unsteady numerical simulation was performed based on the results of steady numerical simulation.The time-domain plot at different monitoring points of the guide vane and the runner was drawn, and the frequency-domain plot at monitoring points was obtained through fast Fourier transform. The results show that the effect of dominant frequency at the monitoring point of the runner on fluid is as consistent as that of the number of the guide vanes and the effect of dominant frequency at the monitoring point of the guide vanes on fluid is as consistent as that of the number of the runner. The amplitude of the dominant frequency at the monitoring point is related to the point location. When the monitoring point is close to the vaneless area between the guide vane and the runner, the amplitude is larger. The further the distance from the vaneless area is, the smaller the amplitude is, and the smaller the influence of dynamic and static interference between the guide vane and the runner is. The research could provide reference for the design of cryogenic expander.

As the largest valley glacier in the Qilian Mountains, the Laohugou glacier No. 12 (LHG12) has shrunk significantly since 1957. In this study, two topographic maps and a WorldView-2 satellite stereopair image data were used to assess the volume and cumulative mass balance of LHG12 located at the western Qilian Mountains during 1957–2015. During the study period, the LHG12 exhibited changes in two processes: slightly ablation and stability in a brief period during 1957–1989 and strong melting and accelerated ablation during 1989–2015. During 1957–2015, the volume of LHG12 decreased by 0.38 km3, the average thickness decreased by 17.23 m, the cumulative mass balance (MB) was −14.69 ± 3.00 m w. e., and ablation was found glacier-wide. By comparing the previous MB simulation and digital elevation model (DEM) differencing results, it was found that the MB simulation results underestimated the strong melting trend of LHG12 since the 1990s. Temperature rose, especially in autumn and winter, and could cause the ice temperature of LHG12 to increase, and LHG12 may become more sensitive to climate change.

(1) Background: the miR-301a is well known involving the proliferation and migration of tumor cells. However, the role of miR-301a in the migration and phagocytosis of macrophages is still unclear. (2) Methods: sciatic nerve injury, liver injury models, as well as primary macrophage cultures were prepared from the miR-301a knockout (KO) and wild type (WT) mice to assess the macrophage’s migration and phagocytosis capabilities. Targetscan database analysis, Western blotting, siRNA transfection, and CXCR4 inhibition or activation were performed to reveal miR301a’s potential mechanism. (3) Results: the macrophage’s migration and phagocytosis were significantly attenuated by the miR-301a KO both in vivo and in vitro. MiR-301a can target Yin-Yang 1 (YY1), and miR-301a KO resulted in YY1 up-regulation and CXCR4 (YY1′s down-stream molecule) down-regulation. siYY1 increased the expression of CXCR4 and enhanced migration and phagocytosis in KO macrophages. Meanwhile, a CXCR4 inhibitor or agonist could attenuate or accelerate, respectively, the macrophage migration and phagocytosis. (4) Conclusions: current findings indicated that miR-301a plays important roles in a macrophage’s capabilities of migration and phagocytosis through the YY1/CXCR4 pathway. Hence, miR-301a might be a promising therapeutic candidate for inflammatory diseases by adjusting macrophage bio-functions.

Background: Desmin is a muscle-specific protein belonging to the intermediate filament family. Desmin mutations are linked to skeletal muscle defects, including inherited myopathies with severe clinical manifestations. The aim of this study was to examine the role of desmin in skeletal muscle remodeling and performance gain induced by muscle mechanical overloading which mimics resistance training.Methods: Plantaris muscles were overloaded by surgical ablation of gastrocnemius and soleus muscles. The functional response of plantaris muscle to mechanical overloading in desmin-deficient mice (DesKO, n = 32) was compared to that of control mice (n = 36) after 7-days or 1-month overloading. To elucidate the molecular mechanisms implicated in the observed partial adaptive response of DesKO muscle, we examined the expression levels of genes involved in muscle growth, myogenesis, inflammation and oxidative energetic metabolism. Moreover, ultrastructure and the proteolysis pathway were explored.Results: Contrary to control, absolute maximal force did not increase in DesKO muscle following 1-month mechanical overloading. Fatigue resistance was also less increased in DesKO as compared to control muscle. Despite impaired functional adaptive response of DesKO mice to mechanical overloading, muscle weight and the number of oxidative MHC2a-positive fibers per cross-section similarly increased in both genotypes after 1-month overloading. However, mechanical overloading-elicited remodeling failed to activate a normal myogenic program after 7-days overloading, resulting in proportionally reduced activation and differentiation of muscle stem cells. Ultrastructural analysis of the plantaris muscle after 1-month overloading revealed muscle fiber damage in DesKO, as indicated by the loss of sarcomere integrity and mitochondrial abnormalities. Moreover, the observed accumulation of autophagosomes and lysosomes in DesKO muscle fibers could indicate a blockage of autophagy. To address this issue, two main proteolysis pathways, the ubiquitin-proteasome system and autophagy, were explored in DesKO and control muscle. Our results suggested an alteration of proteolysis pathways in DesKO muscle in response to mechanical overloading.Conclusion: Taken together, our results show that mechanical overloading increases the negative impact of the lack of desmin on myofibril organization and mitochondria. Furthermore, our results suggest that under these conditions, the repairing activity of autophagy is disturbed. Consequently, force generation is not improved despite muscle growth, suggesting that desmin is required for a complete response to resistance training in skeletal muscle.

Existing research on credit risk contagion of supply chain finance pays more attention to the influence of network internal structure on the process of risk contagion. The spread of COVID-19 has had a huge impact on the supply chain, with a large number of enterprises experiencing difficulties in operation, resulting in increased credit risks in supply chain finance. Under the impact of the epidemic, this paper explores the transmission speed and steady state of credit risk when the supply chain finance network is affected by external impact so that we can have a more complete understanding of the ability of supply chain finance to resist risks. The simulation results show that external shocks of different degrees will increase the number of initial infected enterprises and lead to the increase in credit risk contagion speed but have no significant impact on network steady state; the speed of credit risk contagion is positively correlated with network complexity but not significantly affected by network size; core enterprises infected will increase the rate of credit risk contagion. The intensity of policy intervention has obvious curative effect on the risk caused by external shock. When the supply chain financial network is affected by external shocks, the intensity, time, and pertinence of policy response can effectively prevent the credit risk contagion.

Ctip2/Bcl11b is a zinc finger transcription factor with dual action (repression/activation) that couples epigenetic regulation to gene transcription during the development of various tissues. It is involved in a variety of physiological responses under healthy and pathological conditions. Its role and mechanisms of action are best characterized in the immune and nervous systems. Furthermore, its implication in the development and homeostasis of other various tissues has also been reported. In the present review, we describe its role in skin development, adipogenesis, tooth formation and cranial suture ossification. Experimental data from several studies demonstrate the involvement of Bcl11b in the control of the balance between cell proliferation and differentiation during organ formation and repair, and more specifically in the context of stem cell self-renewal and fate determination. The impact of mutations in the coding sequences of Bcl11b on the development of diseases such as craniosynostosis is also presented. Finally, we discuss genome-wide association studies that suggest a potential influence of single nucleotide polymorphisms found in the 3’ regulatory region of Bcl11b on the homeostasis of the cardiovascular system.

Background Increasing studies demonstrated that the cardiac involvements are related to coronavirus disease 2019 (COVID‐19). Thus, we investigated the clinical characteristics of patients with COVID‐19 and further determined the risk factors for cardiac involvement in them. Methods and Results We analyzed data from 102 consecutive laboratory‐confirmed and hospitalized patients with COVID‐19 (52 women aged 19–87 years). Epidemiologic and demographic characteristics, clinical features, routine laboratory tests (including cardiac injury biomarkers), echocardiography, electrocardiography, chest imaging findings, management methods, and clinical outcomes were collected. Patients were divided into acute cardiac injury, with and without cardiac marker abnormities groups according to different level of cardiac markers. In this research, cardiac involvement was found in 72 of the 102 (70.6%) patients: tachycardia (n=20), electrocardiography abnormalities (n=23), echocardiography abnormalities (n=59), elevated myocardial enzymes (n=55), and acute cardiac injury (n=9). Eight patients with acute cardiac injury were aged >60 years; seven of them had ≥2 underlying comorbidities (hypertension, diabetes mellitus, cardiovascular diseases, chronic obstructive pulmonary disease, and chronic kidney disease). We and others propose vimentin as a possible cellular target for the treatment of COVID-19. This innovative idea is so recent that it requires further attention and debate. The significant role played by vimentin in virus-induced infection however is well established: (1) vimentin has been reported as a co-receptor and/or attachment site for SARS-CoV; (2) vimentin is involved in viral replication in cells; (3) vimentin plays a fundamental role in both the viral infection and the consequent explosive immune-inflammatory response and (4) a lower vimentin expression is associated with the inhibition of epithelial to mesenchymal transition and fibrosis. Moreover, the absence of vimentin in mice makes them resistant to lung injury. Since vimentin has a twofold role in the disease, not only being involved in the viral infection but also in the associated life-threatening lung inflammation, the use of vimentin-targeted drugs may offer a synergistic advantage as compared with other treatments not targeting vimentin. Consequently, we speculate here that drugs which decrease the expression of vimentin can be used for the treatment of patients with COVID-19 and advise that several Food and Drug Administration-approved drugs be immediately tested in clinical trials against SARS-CoV-2, thus broadening therapeutic options for this type of viral infection.

It has been demonstrated that arterial stiffness is linked to arterial fibrosis manifested by increased collagen and other extracellular matrix synthesis in smooth muscle cells (SMC). Transmembrane receptors integrins mediating cell-cell and cell-matrix signalling pathways are involved in tissue fibrosis. We study the role of one integrin subunit av in angiotensin II (AngII)-induced SMC proliferation and arterial fibrosis and stiffness using SMC specific knock-out av mouse model (avSMKO) induced in adult mice by injection of tamoxifen. There is no difference in vascular fibrosis in basal conditions between control and mutant mice. However, decreased arterial fibrosis is observed in avSMKO mutant mice after 28-day perfusion of AngII. Analysis of RNA from aorta of control and mutant mice by Affymetrix microarrays indicated an alteration of the TGF-β pathway in AngII-treated mutant mice. In order to examine the mechanism associated to the decreased fibrosis in vascular SMC from avSMKO mice, isolated VSMC from the aorta of control and avSMKO mice were treated with TGF-β1 or AngII. The results indicated that these two treatments increased the expression of collagen, fibronectin and integrin αν at the protein and RNA levels as well as the phosphorylation of ERK and smad2/3 in the control cells. Inactivation of integrin αν partly inhibited all above effects induced by TGF-β1 and AngII. Our study indicates a role of av and TGF-β1 in the arterial fibrogenesis. Therefore, av signaling could be a therapeutic target against arterial fibrosis and stiffness in pathological conditions.

Physical exercise has important effects as secondary prevention or intervention against several diseases. Endurance exercise induces local and global effects, resulting in skeletal muscle adaptations to aerobic activity and contributes to an amelioration of muscle performance. Furthermore, it prevents muscle loss. Serum response factor (Srf) is a transcription factor of pivotal importance for muscle tissues and animal models of Srf genetic deletion/over-expression are widely used to study Srf role in muscle homeostasis, physiology and pathology. A global characterisation of exercise adaptation in the absence of Srf has not been reported. We measured body composition, muscle force, running speed, energy expenditure and metabolism in WT and inducible skeletal muscle-specific Srf KO mice, following three weeks of voluntary exercise by wheel running. We found a major improvement in the aerobic capacity and muscle function in WT mice following exercise, as expected, and no major differences were observed in Srf KO mice as compared to WT mice, following exercise. Taken together, these observations suggest that Srf is not required for an early (within 3 weeks) adaptation to spontaneous exercise and that Srf KO mice behave similarly to the WT in terms of spontaneous physical activity and the resulting adaptive responses. Therefore, Srf KO mice can be used in functional muscle studies, without the results being affected by the lack of Srf. Since lack of Srf induces premature sarcopenia, our observations suggest that the modifications due to the absence of Srf take time to occur and that young, Srf KO mice behave similarly to WT in aerobic physical activities.
