To explore the relationship between family resilience and preschool children's problem behaviours and the role of maternal parenting stress and parenting self-efficacy in this relationship, a total of 1282 mothers of preschool children were surveyed. The results showed that: (1) family resilience could significantly negatively predict preschool children's problem behaviours. (2) Maternal parenting stress completely mediated the relationship between family resilience and preschool children's problem behaviours. (3) Maternal parenting self-efficacy completely mediated the relationship between family resilience and preschool children's problem behaviours. (4) Maternal parenting stress and parenting self-efficacy played a mediating chain role between family resilience and preschool children's problem behaviours. This study deepened the understanding of the relationship between family resilience and children's social adjustment by discussing and outlining the relationship between family resilience to its influence on children's problem behaviours.

Abstract Background Streptococcus mutans (S. mutans) is an important pathogenic bacterium that causes dental caries, while Streptococcus gordonii (S. gordonii) is a non-cariogenic bacterium that inhibits the growth of S. mutans. The SepM protein can promote the inhibitory ability of S. mutans against S. gordonii by cleaving CSP-21 and activating the ComDE two-component system. This study was designed to explore sepM mutation in S. mutans clinical isolates and related function in the regulation of interactions with S. gordonii. Methods The S. mutans clinical strains that can inhibit the growth of S. gordonii constitute the inhibitory group. 286 C-serotype S. mutans strains were categorized into S. gordonii inhibitory (n = 114) and non-inhibitory bacteria (n = 172). We detected sanger sequencing of sepM gene, the expression levels of related genes and proteins in clinical isolates, obtained prokaryotic expression and purification of mutated proteins, and analyzed the effect of the target mutations on the binding between SepM and CSP-21. Results We found that C482T, G533A, and G661A missense mutations were presented at significantly higher frequency in the inhibitory group relative to the non-inhibitory group. There was no significant difference in the expression of the sepM gene between selected clinical isolates harboring the G533A mutation and the control group. The expression levels of SepM, phosphorylated ComD, and ComE in the mutation group were significantly higher than those in the control group. SepM_control and SepM_D221N (G661A at the gene level) were found to contain two residues close to the active center while SepM_G178D (G533A at the gene level) contained three residues close to the active center. At 25 °C and a pH of 5.5, SepM_D221N (G661A) exhibited higher affinity for CSP-21 (KD = 8.25 µM) than did the SepM control (KD = 33.1 µM), and at 25 °C and a pH of 7.5, SepM_G178D (G533A) exhibited higher affinity (KD = 3.02 µM) than the SepM control (KD = 15.9 µM). It means that it is pH dependent. Conclusions Our data suggest that increased cleavage of CSP-21 by the the mutant SepM may be a reason for the higher inhibitory effect of S. mutans on S. gordonii .

The development of coal seam wind oxidation zone has a huge impact on the production organization and safe mining of coal mines. This paper focuses on the technical difficulties in delineating the coal seam wind oxidation zone in the eastern uplift zone of Junggar Coalfield, and studies the feasibility of using transient electromagnetic method to explore the coal seam wind oxidation zone. Firstly, the electrical resistivity logging curves of normal strata and coal seam wind oxidation areas in the research area were analyzed, and the differences in electrical properties between normal areas and coal seam wind oxidation areas were summarized. A geoelectric model of coal seam wind oxidation areas was constructed based on the electrical resistivity logging curves; then, MAXWELL software was used to numerically simulate the transient electromagnetic response of the theoretical model. It was found that the resistivity cross-section of the transient electromagnetic inversion in the coal seam wind oxidation zone exhibited abnormal characteristics of shallow low resistance layer thickening and reduced low resistance anomaly amplitude, which can be used to delineate the distribution range of the coal seam wind oxidation zone; finally, based on numerical simulation, on-site experiments were conducted in the Chuancao Gedan Coal Mine. Combined with drilling conditions, the experimental data were analyzed from the perspectives of transient electromagnetic inversion resistivity cross-section and plane. The results showed that the transient electromagnetic method is feasible for detecting and dividing wind oxidation zones.

Abstract Triple-negative breast cancer (TNBC) stands as the breast cancer subtype with the highest recurrence and mortality rates, with the lungs being the common site of metastasis. The pulmonary microenvironment plays a pivotal role in the colonization of disseminated tumor cells. Herein, this study highlights the crucial role of exosomal LAP-TGF-β1, the principal form of exosomal TGF-β1, in reshaping the pulmonary vascular niche, thereby facilitating TNBC lung metastasis. Although various strategies have been developed to block TGF-β signaling and have advanced clinically, their significant side effects have limited their therapeutic application. This study demonstrates that in lung metastatic sites, LAP-TGF-β1 within exosomes can remarkably reconfigure the pulmonary vascular niche at lower doses, bolstering the extravasation and colonization of TNBC cells in the lungs. Mechanistically, under the aegis of the acetyltransferase TIP60, a non-canonical KFERQ-like sequence in LAP-TGF-β1 undergoes acetylation at the K304 site, promoting its interaction with HSP90A and subsequent transport into exosomes. Concurrent inhibition of both HSP90A and TIP60 significantly diminishes the exosomal burden of LAP-TGF-β1, presenting a promising therapeutic avenue for TNBC lung metastasis. This study not only offers fresh insights into the molecular underpinnings of TNBC lung metastasis but also lays a foundation for innovative therapeutic strategies.

Abstract Continuous-flow biocatalysis utilizing immobilized enzymes emerged as a sustainable route for chemical synthesis. However, inadequate biocatalytic efficiency from current flow reactors, caused by non-productive enzyme immobilization or enzyme-carrier mismatches in size, hampers its widespread application. Here, we demonstrate a general-applicable and robust approach for the fabrication of a high-performance enzymatic continuous-flow reactor via integrating well-designed scalable isoporous block copolymer (BCP) membranes as carriers with an oriented and productive immobilization employing material binding peptides (MBP). Densely packed uniform enzyme-matched nanochannels of well-designed BCP membranes endow the desired nanoconfined environments towards a productive immobilized phytase. Tuning nanochannel properties can further regulate the complex reaction process and fortify the catalytic performance. The synergistic design of enzyme-matched carriers and efficient enzyme immobilization empowers an excellent catalytic performance with >1 month operational stability, superior productivity, and a high space-time yield (1.05 × 105 g L−1 d−1) via a single-pass continuous-flow process. The obtained performance makes the designed nano- and isoporous block copolymer membrane reactor highly attractive for industrial applications.

The position of makeup air inlets has significant impacts on fire combustion and the smoke spread characteristics. Therefore, influences of makeup air inlets located at different heights on fire combustion and smoke diffusion were discussed through experiments. Experimental results show that when the air inlet was 0.3 m above the ground, the flame width was the largest and the heat release rate (HRR) was the highest. When the height of air inlets was 2.3 m, both the heights of fire flame and smoke layer were the highest. The heights of air inlets were 0.3, 1.3, 2.3 and 3.3 m, the corresponding HRR of the fire was 1.12, 1.06, 0.87 and 0.81 times that of the HRR of the fire natural combustion. There is a logarithmic function relationship between the air inlets height and the HRR, and a polynomial function relationship between the flame height and the air inlets height. When the air inlets was installed at 3.3 m above the ground, the smoke layer height was the lowest, only approximately 1.0 m. Therefore, for rooms with similar dimensions, the makeup air inlets are recommended installed at 1.3–2.3 m above the ground.

T cell development in the thymus is dependent on the thymic microenvironment, in which thymic epithelial cells (TECs) are the major component. However, TECs undergo both a qualitative and quantitative loss during aging, which is believed to be the major factor responsible for age-dependent thymic atrophy. FOXN1 plays a critical role in TEC development and adult TECs maintenance. We have previously reported that intrathymic injection of a recombinant (r) protein containing murine FOXN1 and a protein transduction domain increases the number of TECs in mice, leading to enhanced thymopoiesis. However, intrathymic injection may not be an ideal choice for clinical applications. In this study, we produced a rFOXN1 fusion protein containing the N-terminal of CCR9, human FOXN1 and a protein transduction domain. When injected intravenously into 14-month-old mice, the rFOXN1 fusion protein enters the thymus and TECs, and enhances thymopoiesis, resulting in increased T cell generation in the thymus and increased number of T cells in peripheral lymphoid organ. Our results suggest that the rFOXN1 fusion protein has the potential to be used in preventing and treating T cell immunodeficiency in older adults.

Abstract The interactions between the rumen microbiota and the host are crucial for the digestive and absorptive processes of ruminants, and they are heavily influenced by the climatic conditions of their habitat. Owing to the harsh conditions of the high-altitude habitat, little is known about how ruminants regulate the host transcriptome and the composition of their rumen microbiota. Using the model species of goats, we examined the variations in the rumen microbiota, transcriptome regulation, and climate of the environment between high altitude (Lhasa, Xizang; 3650 m) and low altitude (Chengdu, Sichuan, China; 500 m) goats. The results of 16 S rRNA sequencing revealed variations in the abundance, diversity, and composition of rumen microbiota. Papillibacter, Quinella, and Saccharofermentans were chosen as potential microbes for the adaptation of Xizang goats to the harsh climate of the plateau by the Spearman correlation study of climate and microbiota. Based on rumen transcriptome sequencing analysis, 244 genes were found to be differentially expressed between Xizang goats and low-altitude goats, with 127 genes showing up-regulation and 117 genes showing down-regulation. SLC26A9, GPX3, ARRDC4, and COX1 were identified as potential candidates for plateau adaptation in Xizang goats. Moreover, the metabolism of fatty acids, arachidonic acids, pathway involving cytokines and their receptors could be essential for adaptation to plateau hypoxia and cold endurance. The expression of GPX3, a gene linked to plateau acclimatization in Xizang goats, was linked to the abundance of Anaerovibrio, and the expression of SLC26A9 was linked to the quantity of Selenomonas, according to ruminal microbiota and host Spearman correlation analysis. Our findings imply that in order to adapt harsh plateau conditions, Xizang goats have evolved to maximize digestion and absorption as well as to have a rumen microbiota suitable for the composition of their diet.

ObjectiveLeucine-rich α-2 glycoprotein 1 (LRG1) promotes inflammation and myocardial injury, but its clinical role in ST-elevation myocardial infarction (STEMI) is rarely disclosed. Herein, this prospective study aimed to explore the value of plasma LRG1 at different time points to predict major adverse cardiovascular event (MACE) risk in patients with STEMI.MethodsIn total, 209 patients with STEMI were enrolled for determining plasma LRG1 at admission and on day (D)1/D7/D30 after admission via enzyme-linked immunosorbent assay, as well as for determination of peripheral blood T helper 17 (Th17) cells and regulatory T (Treg) cells by flow cytometry. In addition, plasma LRG1 was obtained from 30 healthy controls at enrollment.ResultsLRG1 was increased in patients with STEMI at admission compared with healthy controls (P 60 μg/ml (P = 0.031) and D7 > 60 μg/ml (P = 0.018) were linked with increased accumulating MACE. Importantly, LRG1 at D7 > 60 μg/ml was independently correlated with increased MACE risk (hazard ratio = 5.216, P = 0.033).ConclusionPlasma LRG1 increases from admission to D1 and gradually declines until D30, which positively links with Th17 cells and MACE risk in patients with STEMI.

Abstract Background Premature loss of primary molars can be treated with a band loop space maintainer (SM). However, fabricating a conventional band loop SM requires multiple clinical and laboratory procedures, which can potentially affect the accuracy of the SM. Moreover, the conventional SM is unable to fully restore masticatory function and maintain the vertical dimension of the edentulous space. In this current study, a fully digital workflow to fabricate a semi-rigid bridge SM made from polyetheretherketone (PEEK) has been described and evaluated for its clinical effectiveness. Methods A total of 15 children (eight males and seven females) between the ages of 4–8 years, who experienced the premature loss of a single primary molar, were included in this study. Digital impressions were taken using the CEREC CAD/CAM chair system and imported into CAD software to design the semi-rigid bridge SM, which was fabricated using PEEK block as the maintainer material. The digital SM was tried-in and bonded to the abutment with resin cement. The edentulous space was measured immediately after bonding (T0) and 1 month (T1), 3 months (T2), and 6 months (T3) after treatment. The periodontal condition and mobility of the SM and abutment were also examined. Results The use of digital impressions resulted in a decreased occurrence of the pharyngeal reflex. The digital semi-rigid bridge SM, fabricated with PEEK, was both convenient and aesthetically pleasing, and successfully restored the anatomy and masticatory function of the missing primary molar. None of the 15 semi-rigid bridge SMs or abutments became loose or fell off during the study, and only one child presented with gingivitis. Furthermore, the difference in the edentulous space at T0, T1, T2, and T3 was not statistically significant (all P > 0.05). Conclusions The digital semi-rigid bridge SM fabricated with PEEK was clinically effective in maintaining the missing space and had advantages over the traditional band/crown loop SM.

Proteases have the advantages of high catalytic efficiency and strong specificity, which are widely used in food, chemical, medicine and other industrial fields. However, natural proteases are susceptible to environmental acidity and inappropriate environmental acidity reduces their catalytic efficiency. Effective strategies to improve the acidity resistance of enzymes are key to promote their industrial application. Rational design strategy is an effective technique to improve the acidic stability of enzymes, such as homologous sequence alignment, surface charge optimization, and intramolecular force optimization. Considering most foods and food raw materials which are subject to contamination by ochratoxin A (OTA) are acidic, this review summarizes the common strategies for improving the acidic stability of enzymes and their prospects for application in the field of OTA degradation. It is expected that this review will provide a reference for research on the acid resistance of industrial enzymes and provide a theoretical basis for the application of mycotoxin-degrading enzymes.

Abstract Objective The pathogenesis of idiopathic pulmonary fibrosis (IPF) remains unclear. We sought to identify IPF-related genes that may participate in the pathogenesis and predict potential targeted traditional Chinese medicines (TCMs). Methods Using IPF gene-expression data, Wilcoxon rank-sum tests were performed to identify differentially expressed genes (DEGs). Protein–protein interaction (PPI) networks, hub genes, and competitive endogenous RNA (ceRNA) networks were constructed or identified by Cytoscape. Quantitative polymerase chain reaction (qPCR) experiments in TGF-β1-induced human fetal lung (HFL) fibroblast cells and a pulmonary fibrosis mouse model verified gene reliability. The SymMap database predicted potential TCMs targeting IPF. The reliability of TCMs was verified in TGF-β1-induced MRC-5 cells. Materials Multiple gene-expression profile data of normal lung and IPF tissues were downloaded from the Gene Expression Omnibus database. In order to improve the inhibition effect of gel on coal spontaneous combustion, a chitosan (CS)/polyacrylamide (PAM)/metal ion (Al3+) composite double-network gel was developed in this study. The optimum formula of the composite double-network gel was determined using orthogonal experimentation. The microstructure, water retention, compressibility, and anti-destruction properties of the composite double-network gel were analyzed. The inhibition effect of the composite double-network gel on coal spontaneous combustion was studied via infrared spectroscopy and a synchronous thermal analyzer from the micro and macro perspectives. The results show that the composite double-network gel has a denser interpenetrating double-network structure and a larger void ratio than the ordinary gel. The water retention rate was 55% after standing at 150 °C for 12 h. The deformation memory ratio of the composite double-network gel was 78%, which was 26.8% higher than that of the ordinary gel, and the compressive strength also increased by 59.96%. In addition, the critical temperature point and the maximum thermal weight-loss rate temperature point decreased by 7.01 °C and 39.62 °C, respectively, and the composite double-network gel effectively reduced active functional groups in the treated coal sample, such as hydroxyl and aliphatic hydrocarbons. In this study, a CS/PAM/Al3+ composite double-network gel was produced, which exhibited good gel performance and inhibition effects, with physical effects such as the covering, wetting, and cementation of coal.

Abstract Refractory diabetic wounds are associated with high incidence, mortality, and recurrence rates and are a devastating and rapidly growing clinical problem. However, treating these wounds is difficult owing to uncontrolled inflammatory microenvironments and defective angiogenesis in the affected areas, with no established effective treatment to the best of our knowledge. Herein, we optimized a dual functional therapeutic agent based on the assembly of LL‐37 peptides and diblock copolymer poly(ethylene glycol)–poly(propylene sulfide) (PEG–PPS). The incorporation of PEG–PPS enabled responsive or controlled LL‐37 peptide release in the presence of reactive oxygen species (ROS). LL‐37@PEG–PPS nanomicelles not only scavenged excessive ROS to improve the microenvironment for angiogenesis but also released LL‐37 peptides and protected them from degradation, thereby robustly increasing angiogenesis. Diabetic wounds treated with LL‐37@PEG–PPS exhibited accelerated and high‐quality wound healing in vivo. This study shows that LL‐37@PEG–PPS can restore beneficial angiogenesis in the wound microenvironment by continuously providing angiogenesis‐promoting signals. Thus, it may be a promising drug for improving chronic refractory wound healing.

Freely engineering the operation frequency of frequency comb sources is crucial for various applications, e.g., high-precision spectroscopy, ranging, communications, and so on. Here, by employing band structure simulations, group velocity dispersion (GVD) analysis, and experimental verifications, we demonstrate that the operation frequency of terahertz (THz) quantum cascade laser frequency combs can be engineered from 4.2 to 4.0 THz. First of all, from the viewpoint of the band structure engineering, we shift the frequency corresponding to the optical transitions in the active region from 4.2 to 4.0 THz by slightly altering the thicknesses of quantum wells. Meanwhile, a GVD analysis is applied to evaluate the potential comb performance. Finally, experimental characterizations, e.g., emission spectra, inter-mode beatnote, dual-comb operation, are performed to validate the exceptional comb operation at 4.0 THz. The advancement in simulations and experimental results present a comprehensive method to customize the desired THz radiative frequency for comb generation, which facilitates the practical development of broadband, high-precision THz comb sources.

Land degradation is a major impediment to achieving sustainable development. However, there is currently no harmonized global map of land degradation status and hotspots. This paper aims to obtain the status and hotspots map of global land degradation by multiple methods and indicators to give essential references for land degradation neutrality. The results show that there are significant differences in the distribution and degree of land degradation between the different methods and indicators. Validation through observation points reveals that most of the methods and indicators can reflect land degradation in arid and semi-arid areas, while there are suitable methods or indicators in tropical and high-latitude areas. The degree of degradation has a large difference after overlay analysis, which shows that there are shortcomings of different methods and indicators for monitoring the degree of land degradation. However, the overlay of land degradation extent displays a high consistency, reflecting the current state of global land degradation to a certain extent. These areas with high overlay value can be recognized as hotspots of land degradation. It is also found there are consistent water-energy change characteristics in the hotspot area, such as increased land surface temperature and air temperature and decreased soil moisture and precipitation. These results conclude that studies on the degree of land degradation need to be considered in an integrated manner about the regional background. The combination of multiple methods and indicators is recommended for land degradation extent studies in large areas. Comparison of different methods and indicators is important guidance for global land degradation research. Accelerating ecological monitoring and restoration of land degradation hotspots is the first step towards land degradation neutrality.

BackgroundInflammation is the common pathogenesis of coronary atherosclerosis disease (CAD) and rheumatoid arthritis (RA). Although it is established that RA increases the risk of CAD, the underlining mechanism remained indefinite. This study seeks to explore the molecular mechanisms of RA linked CAD and identify potential target gene for early prediction of CAD in RA patients.Materials and methodsThe study utilized five raw datasets: GSE55235, GSE55457, GSE12021 for RA patients, and GSE42148 and GSE20680 for CAD patients. Gene Set Enrichment Analysis (GSEA) was used to investigate common signaling pathways associated with RA and CAD. Then, weighted gene co-expression network analysis (WGCNA) was performed on RA and CAD training datasets to identify gene modules related to single-sample GSEA (ssGSEA) scores. Overlapping module genes and differentially expressed genes (DEGs) were considered as co-susceptible genes for both diseases. Three hub genes were screened using a protein-protein interaction (PPI) network analysis via Cytoscape plug-ins. The signaling pathways, immune infiltration, and transcription factors associated with these hub genes were analyzed to explore the underlying mechanism connecting both diseases. Immunohistochemistry and qRT-PCR were conducted to validate the expression of the key candidate gene, PPARG, in macrophages of synovial tissue and arterial walls from RA and CAD patients.ResultsThe study found that Fc-gamma receptor-mediated endocytosis is a common signaling pathway for both RA and CAD. A total of 25 genes were screened by WGCNA and DEGs, which are involved in inflammation-related ligand-receptor interactions, cytoskeleton, and endocytosis signaling pathways. The principal component analysis(PCA) and support vector machine (SVM) and receiver-operator characteristic (ROC) analysis demonstrate that 25 DEGs can effectively distinguish RA and CAD groups from normal groups. Three hub genes TUBB2A, FKBP5, and PPARG were further identified by the Cytoscape software. Both FKBP5 and PPARG were downregulated in synovial tissue of RA and upregulated in the peripheral blood of CAD patients and differential mRNAexpreesion between normal and disease groups in both diseases were validated by qRT-PCR.Association of PPARG with monocyte was demonstrated across both training and validation datasets in CAD. PPARG expression is observed in control synovial epithelial cells and foamy macrophages of arterial walls, but was decreased in synovial epithelium of RA patients. Its expression in foamy macrophages of atherosclerotic vascular walls exhibits a positive correlation (r = 0.6276, p = 0.0002) with CD68.ConclusionOur findings suggest that PPARG may serve as a potentially predictive marker for CAD in RA patients, which provides new insights into the molecular mechanism underling RA linked CAD.

Abstract Background: Cancer health research relies on large-scale cohorts to derive generalizable results for different populations. While traditional epidemiological cohorts often use costly random sampling or self-motivated, preselected groups, a shift toward health system-based cohorts has emerged. However, such cohorts depend on participants remaining within a single system. Recent consumer engagement models using smartphone-based communication, driving projects, and social media have begun to upend these paradigms. Methods: We initiated the Healthy Oregon Project (HOP) to support basic and clinical cancer research. HOP study employs a novel, cost-effective remote recruitment approach to effectively establish a large-scale cohort for population-based studies. The recruitment leverages the unique email account, the HOP website, and social media platforms to direct smartphone users to the study app, which facilitates saliva sample collection and survey administration. Monthly newsletters further facilitate engagement and outreach to broader communities. Results: By the end of 2022, the HOP has enrolled approximately 35,000 participants aged 18–100 years (median = 44.2 years), comprising more than 1% of the Oregon adult population. Among those who have app access, ∼87% provided consent to genetic screening. The HOP monthly email newsletters have an average open rate of 38%. Efforts continue to be made to improve survey response rates. Conclusion: This study underscores the efficacy of remote recruitment approaches in establishing large-scale cohorts for population-based cancer studies. The implementation of the study facilitates the collection of extensive survey and biological data into a repository that can be broadly shared and supports collaborative clinical and translational research.

Abstract Human pluripotent stem cells (hPSCs) can generate insulin-producing beta cells for diabetes treatment, but residual undifferentiated cells may cause tumors. We developed a highly sensitive assay to detect these cells in islet cells derived from human chemically induced pluripotent stem cells (hCiPSCs), which are transgene-free and safer. We used RNA-seq data to find protein-coding and non-coding RNAs that were only expressed in hCiPSCs, not in islet cells. We confirmed these biomarkers by RT-qPCR and ddPCR. We chose long non-coding RNA (lncRNA) markers, which performed better than protein-coding RNA markers. We found that LNCPRESS2, LINC00678 and LOC105370482 could detect 1, 1 and 3 hCiPSCs in 106 islet cells by ddPCR, respectively. We tested our method on several hCiPSC lines, which could quantify 0.0001% undifferentiated cell in 106 islet cells by targeting hCiPSCs-specific lncRNA transcripts, ensuring the safety and quality of hCiPSC-derived islet cells for clinical use.

Abstract Under the shared big data environment, the existing shared data auditing schemes rarely consider the authorization management of group users. Thus, in this paper, we propose a novel remote shared data checking possession scheme that support group authority management. To implement group user rights management, we firstly introduce a trusted entity group manager. To assist the group manager in authorization management, we formalize a new algebraic structure operator named authorization invisible authenticator (AIA). Meanwhile, we provide a basic AIA scheme for general security scenarios and a standard AIA scheme suitable for high-level security scenarios respectively. The standard AIA scheme can fully meet the needs of the group manager to safely perform rights management work. It is composed of the User Information Table (UIT) and the basic AIA scheme, which has higher security and is applicable to a wider range of scenarios. By distributing AIA through the standard AIA scheme, the group manager can easily carry out authority management, including enrolling, revoking, updating. After solving the problem of authorization management, the detailed design of the scheme based on identity-based encryption (IBE) is given. Furthermore, the security analysis and performance evaluation demonstrate that the scheme is safe and feasible.

Environmental changes induced by urbanization may significantly alter plant survival strategies, thereby introducing uncertainties in their ability to withstand extreme heat. This study, centered on Jinhua City, distinguished urban, suburban, and rural areas to represent the various intensities of urbanization. It examined the leaf function properties of evergreen and deciduous trees common in these regions, focusing on leaf and branch characteristics. Employing an analysis of variance (ANOVA), principal component analysis (PCA), and path analysis (PA) of the plant functional traits and the climatic factors of each region, this study assessed the impact of urbanization on plant survival strategies. By tracking changes in plant functional traits from June to August, it explored the capacity of plants to acclimate to urban-warming-related heat stress across different urbanization gradients. The findings revealed that leaf thickness (LT) and stomatal size (SS) initially decreased and then increased, whereas specific leaf area (SLA) and leaf tissue density (LTD) first rose and then declined, from rural to urban regions. From June to August, branch wood density (WD), chlorophyll (Chl) content, LTD, and leaf dry matter content (LDMC) increased, whereas SLA and leaf water content (LWC) diminished, in all regions. PCA suggested that there was no significant change in the resource allocation strategy of plants (p > 0.05), with drought tolerance significantly reduced in the suburbs on the gradient of urbanization (p < 0.05). During the summer, with high temperature, plants were predominantly biased towards slow-return, conservative strategies, particularly among evergreen species. Compared to precipitation, PA revealed a significant urban warming effect. During summer, temperature was the main factor influencing resource investment strategy and drought resistance, with a notably stronger impact on the former. The high temperature in summer promoted a conservative survival strategy in plants, and the urbanization effect increased their tolerance to high temperatures.

Abstract ClC-2 transports chloride ions across plasma membranes and plays critical roles in cellular homeostasis. Its dysfunction is involved in diseases including leukodystrophy and primary aldosteronism. AK-42 was recently reported as a specific inhibitor of ClC-2. However, experimental structures are still missing to decipher its inhibition mechanism. Here, we present cryo-EM structures of apo ClC-2 and its complex with AK-42, both at 3.5 Å resolution. Residues S162, E205 and Y553 are involved in chloride binding and contribute to the ion selectivity. The side-chain of the gating glutamate E205 occupies the putative central chloride-binding site, indicating that our structure represents a closed state. Structural analysis, molecular dynamics and electrophysiological recordings identify key residues to interact with AK-42. Several AK-42 interacting residues are present in ClC-2 but not in other ClCs, providing a possible explanation for AK-42 specificity. Taken together, our results experimentally reveal the potential inhibition mechanism of ClC-2 inhibitor AK-42.

Abstract Biomolecular condensates play key roles in various biological processes. However, specific condensation modulators are currently lacking. PROTAC is a new technology that can use small molecules to degrade target proteins specifically. PROTAC molecules are expected to regulate biomolecular condensates dynamically by degrading/recovering key molecules in biomolecular condensates. In this study, we employed a BRD4-targeting PROTAC molecule to regulate the super-enhancer (SE) condensate and monitored the changes of SE condensate under PROTAC treatment using live-cell imaging and high-throughput sequencing technologies. As a result, we found that BRD4-targeting PROTACs can significantly reduce the BRD4 condensates, and we established a quantitative method for tracking BRD4 condensates by PROTAC and cellular imaging. Surprisingly and encouragingly, BRD4 condensates were observed to preferentially form and play specialized roles in biological process regulation for the first time. Additionally, BRD4 PROTAC makes it possible to observe the dynamics of other condensate components under the continued disruption of BRD4 condensates. Together, these results shed new light on research methods for liquid-liquid phase separation (LLPS), and specifically demonstrate that PROTAC presents a powerful and distinctive tool for the study of biomolecular condensates.

Human milk, which provides personalized, comprehensive, and balanced nutrients, and immune protection, is the best nutritional source for 0~6 month-old infants, and it is the gold standard for infant food. A cohort study of human milk omics is a necessary prerequisite for comprehensively analyzing the composition and characteristics of Chinese human milk, establishing Chinese infant food norms and standards, developing infant nutrition guidelines, and creating infant foods. This article presents the research progress of the National Engineering Research Center of Dairy Health for Maternal and Child pertaining to the China Maternal and Infant Nutrition Health Birth Cohort Study (MINC) in terms of breast milk lipidomics, glycomics, proteomics, and microbiomics detection techniques, composition characteristics, differences from bovine milk, and industrial applications in infant food. This will lay a science and technical support for the omics research of Chinese human milk and provide the necessary data and demonstration for industrial application.

Abstract For the process of photovoltaic conversion in organic solar cells (OSCs) and quantum‐dot solar cells (QDSCs), three of four steps are determined by exciton behavior, namely, exciton generation, exciton diffusion, and exciton dissociation. Therefore, it is of great importance to regulate exciton behavior in OSCs and QDSCs for achieving high power conversion efficiency. Due to the rapid development in materials and device fabrication, great progress has been made to manage the exciton behavior to achieve prolonged exciton diffusion length and improved exciton dissociation in recent years. In this review, we first introduce the parameters that affect exciton behavior, followed by the methods to measure exciton diffusion length. Then, we provide an overview of the recent advances with regard to exciton behavior investigation in OSCs and QDSCs, including exciton lifetime, exciton diffusion coefficient, and exciton dissociation. Finally, we propose future directions in deepening the understanding of exciton behavior and boosting the performance of OSCs and QDSCs.

Abstract The trade-off and synergy between heritage conservation and tourism has become the focus of natural world heritage research. To gain a better understanding of the global researches on natural World Heritage conservation and tourism, we comprehensively reviewed relevant peer-reviewed research literature based on Web of Science (WOS) and China National Knowledge Infrastructure (CNKI). We find that (1) the theoretical research of natural heritage conservation and tourism has gone through a process from emphasizing the protection of heritage value to pursuing the synergy of heritage protection and tourism development; (2) the main research methods include investigation research methods, indirect research methods and experimental research methods; (3) "3S" technology (remote sensing, geographic information system, global positioning system), three-dimensional laser scanning technology, virtual reality (VR) technology, augmented reality (AR) technology, holographic projection technology and other modern technological means are applied to the protection and tourism development of natural properties; (4) the common coordinated development models include ecological science tourism, community participation in tourism, ecological compensation model, world heritage—buffer zone—surrounding areas coordinated protection model and so on. We analyzed the research progresses through (1) the theories proposed in the literature, (2) the main methods applied to address the issues on natural heritage conservation and tourism, (3) the technologies applied in the researches and (4) the coordinated models of heritage conservation and tourism. Furthermore, we put forward the following research prospects: (1) systematically explore the conservation methods and theories based on world heritage criteria; (2) formulate corresponding conservation systems and ecological restoration standards for different types of world heritage; (3) give full play to the complementary advantages of various research methods and reveal the mutual feedback mechanism between tourism and heritage conservation; (4) develop ecological restoration technology based on biodiversity restoration, establish radial ecological corridor, and expand the benign ecological environment of the properties to wider periphery; (5) build ecological compensation development models based on the perspective of heritage tourism and value realization of world heritage.

Background Recent studies about the use of Traditional Chinese Medicine (TCM) to treat cough variant asthma (CVA) , a common respiratory disease, are increasing. TCM has proven to be effective in improving symptoms and the overall treatment efficacy of CVA, but relevant research is still in the exploratory stage, and there are no unified and standardized diagnostic criteria for CVA. Objective To explore the common syndromes and symptom characteristics of CVA. Methods The literature related to CVA syndromes or symptoms treated by TCM was searched in CNKI, Wanfang Data, VIP, and SinoMed databases from inception to October 2, 2021. SPSS 25.0 software was used to support the input of extracted information to establish a database, and the statistical treatment of the frequencies of CVA-related syndromes and symptoms. The symptoms with a frequency over 5% were selected for factor analysis, and the common factors were extracted for further hierarchical cluster analysis, and the potential syndromes were inferred from the results. Using Lantern 5.0 software, based on the LTM-EAST (Latent Tree Model-Extension Adjustment Simplification until Termination algorithm) , a latent structure model was constructed and a comprehensive cluster analysis was performed to infer potential syndromes. Then the common syndromes and symptom characteristics of CVA were obtained by combining the results of frequency statistics, hierarchical clustering, and latent structure analysis. Results A total of 621 studies were included, involving 115 symptoms, including cough, pharyngeal itching, low sputum volume, etc. Thirty-nine syndromes were involved, among which the highly frequent syndromes were pathogenic wind invading the lung, wind and cold invading the lung, and lung yin deficiency. Six potential syndromes were derived from hierarchical cluster analysis, including wind and heat invading the lung, liver fire invading the lung, lung qi deficiency and so on. The latent structure analysis was performed on the symptoms with frequency over 3%, by which 21 latent variables, and 42 latent classes were obtained, and 12 potential syndromes were further inferred by comprehensive clustering, including pathogenic wind invading the lung, wind and cold invading the lung, wind and heat invading the lung, and so on. Based on the above analyses, 11 common syndromes were finally identified. Each syndrome has specific symptom characteristics, for example, the syndrome of pathogenic wind invading the lung manifests itself as cough, sneezing, running nose, and nasal congestion. Conclusion We identified 11 common syndromes of CVA with typical symptom characteristics, which are pathogenic wind invading the lung, wind and cold invading the lung, wind and heat invading the lung, wind and dryness damaging the lung, phlegm and heat congesting in the lung, phlegm and dampness accumulating in the lung, liver fire invading the lung, cold fluid retention in the lung, lung qi deficiency, lung yin deficiency, and lung-spleen qi deficiency. Our analysis will provide a basis for clinical syndrome differentiation and clinical investigation of CVA.

Karst nature reserve (NR) ecosystems are vulnerable, and the development of regulatory and cultural services industries is still in its infancy. Realization of ecological product value (EPV) is a crucial way to promote the integration of eco-protection and industrial development in nature reserves (NRs). This study calculates EPV using a modified equivalent factor, analyzes the spatial pattern of EPV using the fishnet tool in Arc GIS, and constructs a model for realizing EPV. Finally, the driving factors for the formation of eco-industry are identified using the grey correlation analysis (GCA) method. The results show that (1) the regulatory service products in karst NRs play a decisive role in EPV, and forest ecosystems have significant EPV potential; (2) high-value grids are concentrated in the core and buffer zones of karst NRs and the spatial distribution of EPV in the experimental zone is highly heterogeneous, with high and low-value grids in mixed distribution; (3) the realization model of EPV in karst NRs follows the logic of “preservation, transformation, and appreciation”; (4) human resources (0.813), ecology (0.798), industry (0.693), policy (0.783), the market (0.778), and economy (0.715) are essential drivers for the formation of eco-industry in karst NRs. Overall, karst NRs can promote the realization of EPV through planning and regulation of land use, based on the interaction between internal and external factors, to promote the coordinated development of eco-industry in karst NRs. This study provides a reference for the scientific management of eco-resources and the sustainable development of eco-industry in karst NRs.

Ecosystem services (ESs) value realization and ecological industry (eco-industry) are essential components of regional development. Due to the vulnerability and non-renewability of natural landscape resources in Karst areas, once unreasonable resource development takes place, it is easy to pose threats and causes damage to its ecosystem. This article selects the grain data correction equivalent factor coefficients in Guizhou Province, China, and establishes accounting indicators for Karst scenic areas. It is found that the total economic value of ESs is as follows: Shanmuhe Scenic Area (CNY 5096.3 thousand) > DaXiaoQiKong Scenic Area (CNY 2778.6 thousand) > Huangguoshu Scenic Area (CNY 2130.7 thousand). Among them, the value of regulating services plays a dominant role in the composition of ESs, and the value of forests accounts for the largest proportion. Through web crawlers, tourism data can be acquired, and the Product–Experience–Feedback–Improvement (PEFI) model can be applied to reveal that tourists have a predominantly positive perception of Karst scenic areas. This indicates that karst landscape resources are deeply loved by domestic and foreign tourists, especially mountain, water and forest landscapes. Based on the results of the ES value accounting of the scenic area and the external perception data of tourists towards the scenic area, the eco-industry spatial layout development plan is formulated. According to local conditions, the development, construction, and protection of the scenic area are carried out, jointly promoting the realization of the ecological product value (EPV) of the Karst scenic area, to determine the optimal development direction of the eco-industry, and to promote the coordinated development of ecological resources and assets.

Peanuts play a pivotal role as an economic crop on a global scale, serving as a primary source of both edible oil and protein. Peanut rust (Puccinia arachidis Speg.) disease constitutes a significant global biotic stress, representing a substantial economic threat to the peanut industry by inducing noteworthy reductions in seed yields and compromising oil quality. This comprehensive review delves into the distinctive characteristics and detrimental symptoms associated with peanut rust, scrutinizing its epidemiology and the control strategies that are currently implemented. Notably, host resistance emerges as the most favored strategy due to its potential to surmount the limitations inherent in other approaches. The review further considers the recent advancements in peanut rust resistance breeding, integrating the use of molecular marker technology and the identification of rust resistance genes. Our findings indicate that the ongoing refinement of control strategies, especially through the development and application of immune or highly resistant peanut varieties, will have a profound impact on the global peanut industry.

Pneumonia remains a major global health challenge, necessitating the development of effective therapeutic approaches. Recently, necroptosis, a regulated form of cell death, has garnered attention in the fields of pharmacology and immunology for its role in the pathogenesis of pneumonia. Characterized by cell death and inflammatory responses, necroptosis is a key mechanism contributing to tissue damage and immune dysregulation in various diseases, including pneumonia. This review comprehensively analyzes the role of necroptosis in pneumonia and explores potential pharmacological interventions targeting this cell death pathway. Moreover, we highlight the intricate interplay between necroptosis and immune responses in pneumonia, revealing a bidirectional relationship between necrotic cell death and inflammatory signaling. Importantly, we assess current therapeutic strategies modulating necroptosis, encompassing synthetic inhibitors, natural products, and other drugs targeting key components of the programmed necrosis pathway. The article also discusses challenges and future directions in targeting programmed necrosis for pneumonia treatment, proposing novel therapeutic strategies that combine antibiotics with necroptosis inhibitors. This review underscores the importance of understanding necroptosis in pneumonia and highlights the potential of pharmacological interventions to mitigate tissue damage and restore immune homeostasis in this devastating respiratory infection.

The Beijing–Tianjin–Hebei region in China is experiencing a serious ecological water scarcity problem in the context of climate warming and drying. There is an urgent need for practical adaptation measures to cope with the adverse impacts of climate change and provide a scientific basis for urban water supply planning, water resource management, and policy formulation. Urban ecological water can maintain the structure and function of urban ecosystems, both as an environmental element and as a resource. Current research lacks quantitative analysis of the impact of regional meteorological factors on ecological water use at the small and medium scales. Based on the meteorological data and statistical data of water resources in the Beijing–Tianjin–Hebei (BTH) region, this paper analyzed the trend of climate change and established an ecological climatic water model using gray correlation analysis, polynomial simulation, and singular spectrum analysis to predict the ecological water consumption. And, we assessed the climatic sensitivity of ecological water use and estimated the future ecological climatic water use in the BTH region based on four climate scenarios’ data. The results showed that the average multi-year temperature was 13.2 °C with a clear upward trend from 1991 to 2020 in the BTH region. The multi-year average precipitation was 517.1 mm, with a clear shift in the period of abundance and desiccation. Ecological climatic water modeling showed that a 1 °C increase in temperature will increase ecological water use by 0.73 × 108 m3~1.09 × 108 m3 in the BTH region; for a 100 mm increase in precipitation, ecological water use will decrease by 0.49 × 108 m3~0.88 × 108 m3; under the four climate scenarios of SSP1–2.6, SSP2–4.5, SSP3–7.0, and SSP5–8.5, the regional ecological climatic water use will be 5.14 × 108 m3, 6.64 × 108 m3, 7.82 × 108 m3, and 9.06 × 108 m3 in 2035, respectively; and in 2050, the ecological climatic water use will be 8.16 × 108 m3, 9.75 × 108 m3, 10.71 × 108 m3, and 12.41 × 108 m3, respectively. The methodology and results of this study will support the quantification of climate change impacts on ecological water use in the BTH region and serve as a theoretical basis for future research on ecological water use adaptation to climate change. This study can provide a basis for the development of the overall planning of urban ecological water supply, and at the same time, it can lay a foundation for the study of measures to adapt to climate change by ecological water use.

A high glucose diet (HGD) is associated with many metabolic diseases including type 2 diabetes, and cardiovascular diseases. Additionally, a HGD increases the oxidative stress resistance of young animals but shortens their lifespan. To investigate the role of HGD feeding on the aging of aged animals, we tested for oxidative stress resistance and changes in lifespan using C. elegans. We showed that a HGD extends the lifespan of aged worms that are dependent on oxidative stress resistance. Furthermore, we measured the lifespan of oxidative stress responding genes of HGD-fed worms. We found that gpdh-1 and col-92 are highly expressed in HGD and paraquat (PQ) treated worms. Further experiments indicated that intestinal gpdh-1 is essential for the HGD induced lifespan extension of aged worms. Our studies provide new insights into understanding the correlation between glucose metabolism, oxidative stress resistance, and aging.

To improve the quality of mango puree, this study adopted three non-thermal processing methods of ultra-high pressure, irradiation and low-temperature plasma to treat mango puree and analyzed and compared the sterilization effects of different methods. The effect of the optimum method (ultrahigh pressure) on the flavor, color, content of soluble solids and storage stability of mango puree was further studied. The results showed that both ultrahigh pressure (≥200 MPa) and irradiation (≥3 kGy) had good bactericidal effect on mango puree, and the total number of bacteria in mango puree ≤2 lg(CFU/g), reached the commercial sterile standard. However, the effect of low temperature plasma treatment was not ideal. When the treatment time was less than 6 min, the total number of bacteria in fruit puree did not reach the commercial sterile standard. The results showed that the taste of mango puree was similar to that of the control sample. The storage life of mango puree treated with pressure greater than 500 MPa was stable, especially the storage life of samples treated with 600 MPa could reach at least 9 months. The results showed that the high pressure could not only killed the harmful microorganisms in mango puree, but also improved the stability of mango puree, and maintained the original quality of mango puree to the maximum extent. This study provided some theoretical basis and method guidance for the application of ultrahigh pressure technology in food processing.

It is essential to assess the cancer risk for patients with chronic obstructive pulmonary disease (COPD). Comparing gene expression data from patients with lung cancer (a total of 506 samples) and those with cancer-adjacent normal lung tissues (a total of 370 samples), we generated a qualitative transcriptional signature consisting of 2046 gene pairs. The signature was verified in an evaluation dataset comprising 18 subjects with severe disease and 52 subjects with moderate disease (Wilcoxon rank-sum test; p = 7.33 × 10−5). Similar results were obtained in other independent datasets. Among the gene pairs in the signature, 326 COPD stage-related gene pairs were identified based on Spearman’s rank correlation tests and those gene pairs comprised 368 unique genes. Of these 368 genes, 16 genes were significantly dysregulated in COPD rat model data compared with control data. Some of these genes (Dhx16, Upf2, Notch3, Sec61a1, Dyrk2, and Hmmr) were altered when the COPD rat model was treated with traditional Chinese medicines (TCM), including Bufei Yishen formula, Bufei Jianpi formula, and Yiqi Zishen formula. Overall, the signature could predict the cancer incidence-risk of COPD and the identified key genes might provide guidance regarding both the treatment of COPD using TCM and the prevention of cancer in patients with COPD. KEY MESSAGESA cancer risk assessment signature was identified in patients with COPD.The signature is insensitive to batch effects and is well verified.COPD key genes identified in this study might play a crucial role in TCM treatment and cancer prevention.
