It is of great clinical importance to explore more efficacious treatments for OCD. Recently, cognitive-coping therapy (CCT), mainly focusing on recognizing and coping with a fear of negative events, has been reported as an efficacious psychotherapy. However, the underlying neurophysiological mechanism remains unknown. This study of 79 OCD patients collected Yale-Brown Obsessive Compulsive Scale (Y-BOCS) and resting-state functional magnetic resonance imaging (rs-fMRI) scans before and after four weeks of CCT, pharmacotherapy plus CCT (pCCT), or pharmacotherapy. Amygdala seed-based functional connectivity (FC) analysis was performed. Compared post- to pretreatment, pCCT-treated patients showed decreased left amygdala (LA) FC with the right anterior cingulate gyrus (cluster 1) and with the left paracentral lobule/the parietal lobe (cluster 2), while CCT-treated patients showed decreased LA-FC with the left middle occipital gyrus/the left superior parietal/left inferior parietal (cluster 3). The z-values of LA-FC with the three clusters were significantly lower after pCCT or CCT than pretreatment in comparisons of covert vs. overt and of non-remission vs. remission patients, except the z-value of cluster 2 in covert OCD. CCT and pCCT significantly reduced the Y-BOCS score. The reduction in the Y-BOCS score was positively correlated with the z-value of cluster 1. Our findings demonstrate that both pCCT and CCT with large effect sizes lowered LA-FC, indicating that FCs were involved in OCD. Additionally, decreased LA-FC with the anterior cingulate cortex (ACC) or paracentral/parietal cortex may be a marker for pCCT response or a marker for distinguishing OCD subtypes. Decreased LA-FC with the parietal region may be a common pathway of pCCT and CCT. Trial registration: ChiCTR-IPC-15005969.

Laboratory biosafety has become a core focus in biological analysis, owing to the frequent occurrence of laboratory-acquired infections caused by the leakage of pathogenic microorganisms. For this purpose, the authors developed a safe pretreatment device combining a sealing technique with a direct injection technique. In this study, several bacteria and viruses were used to validate the filtration effect of the invention. Data show that the new device can completely filter bacteria and that the filtration rates for hepatitis B virus and hepatitis C virus reached 94% and 96%, respectively. The results show that the new preparation device can effectively block these pathogens and can improve biological safety and provide powerful protection for technicians.

Kiwifruit (Actinidia deliciosa) is a typical climacteric fruit, and its ripening is closely associated with ethylene. In this study, we present evidence that H2S alleviated ethylene-induced ripening and senescence of kiwifruit. Kiwifruit were fumigated with ethylene released from 0.4 g·L−1 ethephon solution or H2S with 1 mm sodium hydrosulfide (NaHS) as the donor or in combination. Fumigation with ethylene was found to accelerate kiwifruit ripening and H2S treatment effectively alleviated ethylene-induced fruit softening in parallel with attenuated activity of polygalacturonase (PG) and amylase. Ethylene + H2S treatment also maintained higher levels of ascorbic acid, titratable acid, starch, soluble protein, and reducing sugar compared with ethylene group, whereas suppressed the increase in chlorophyll and carotenoid. Kiwifruit ripening and senescence under ethylene treatment was accompanied by elevation in reactive oxygen species (ROS) levels, including H2O2 and superoxide anion and malondialdehyde (MDA), but combined treatment of ethylene plus H2S alleviated oxidative stress in fruit. Furthermore, the activities of antioxidative enzymes catalase (CAT) and ascorbate peroxidase (APX) were increased by ethylene + H2S treatment in comparison with ethylene alone, whereas the activities of lipoxygenase (LOX) and polyphenol oxidase (PPO) were attenuated by H2S treatment. Further investigations showed that H2S repressed the expression of ethylene synthesis-related genes AdSAM, AdACS1, AdACS2, AdACO2, and AdACO3 and cysteine protease genes, such as AdCP1 and AdCP3. Taken together, our findings suggest that H2S alleviates kiwifruit ripening and senescence by antagonizing the effect of ethylene through reduction of oxidative stress and inhibition of ethylene synthesis pathway.

Cognitive-coping therapy (CCT), integrating cognitive theory with stress-coping theory, is an efficacious therapy for obsessive-compulsive disorder (OCD). However, the potential brain mediation for the effectiveness remains unclear. We sought to investigate differences of resting-state brain function between OCD and healthy controls and if such differences would be changed by a four-week CCT.Thirty-one OCD patients were recruited and randomized into CCT (n=15) and pharmacotherapy plus CCT (pCCT, n=16) groups, together with 25 age-, gender- and education-matched healthy controls. The Yale-Brown Obsessive Compulsive Scale (Y-BOCS) was scored to evaluate the severity in symptoms. Resting-state functional magnetic resonance imaging was scanned pre- and post-treatment.For patients, Y-BOCS scores were reduced during four-week treatment for CCT and pCCT (P0.001), but no group difference was observed. No differences in amplitude of low-frequency fluctuation (ALFF) values were found between CCT and pCCT either pre- or post-treatment. Compared to controls, ALFF in OCD patients was higher in the left hippocampus, parahippocampus, and temporal lobes, but lower in the right orbitofrontal cortex, rectus, bilateral calcarine, cuneus, lingual, occipital, left parietal, postcentral, precentral, and parietal (corrected P0.05). The ALFF in those regions was not significantly correlated to the severity of OCD symptoms. After a 4-week treatment, the ALFF differences between OCD patients and controls disappeared.The pharmacotherapy group was not included since OCD patients generally do not respond to pharmacotherapy in four weeks.Our data indicated that resting-state brain function was different between OCD and controls; such differences disappeared after OCD symptoms were relieved.

Objective Clostridioides difficile may colonize healthy infants and young children asymptomatically and for the long-term. C. difficile genotypes and the rate and determinants of colonization differ substantially and vary among countries and regions. A 1-year follow-up study was performed to determine the incidence, kinetics and influencing factors of C. difficile intestinal colonization. Methods Twenty-nine healthy infants (14 girls and 15 boys) living at home with their parents in Handan City were followed by survey from birth to 1 year of age, specifically from October 2014 through December 2015. C. difficile isolates were typed by PCR ribotyping and analyzed for the presence of toxin genes. Results During the follow-up study period in the first year of life, 20 of the 29 total enrolled infants acquired C. difficile. A total of 437 fecal samples were obtained, and 111 (25.4%) samples contained C. difficile, including 79 (71.2%) toxigenic strains. The toxigenic isolates comprised six PCR ribotypes, and two PCR ribotypes were identified as nontoxigenic strains. Conclusion Our study showed that C. difficile colonization increase with age during the 12-month period, and the dominant toxigenic types of C. difficile isolates in infants were those involved in long-term colonization. Feeding patterns may affect the dynamic progress of C. difficile colonization.

Clostridium difficile multilocus sequence type 37 (ST37), which mainly corresponds to ribotype 017, has been a dominant genotype circulating in China. In this study, we report the use of matrix-assisted laser desorption ionization–time of flight mass spectrometry (MALDI-TOF MS) to analyze and characterize 204 C. difficile clinical isolates, including 49 ST37 and 155 non-ST37 isolates collected in China and other countries. The distributions of two major protein peaks ( m/z 3,242 and 3,286) were significantly different between ST37 and non-ST37 prototype strains and clinical isolates. This difference was reproducible when analysis was performed on different colonies in different runs. This finding was repeated and confirmed by both bioMérieux Vitek MS and Bruker Microflex LT systems on isolates recovered from a variety of geographic regions worldwide. The combination of the two peaks was present in 47 of 49 ST37 isolates, resulting in a sensitivity of 95.9%. In contrast, the peak combination was absent in 153 of 155 non-ST37 isolates, resulting in a specificity of 98.7%. Our results suggest that MALDI-TOF MS is a rapid and reliable tool to identify C. difficile genotype ST37. Work is in progress to characterize the two molecules having peaks at m/z 3,242 and 3,286, which appear to be specific to C. difficile genotype ST37.

Hydrogen sulfide (H2S) has been identified as an important gaseous signal in plants. Here, we investigated the mechanism of H2S in alleviating postharvest senescence and rotting of Kyoho grape. Exogenous application of H2S released from 1.0 mM NaHS remarkably decreased the rotting and threshing rate of grape berries. H2S application also prevented the weight loss in grape clusters and inhibited the decreases in firmness, soluble solids, and titratable acidity in grape pulp during postharvest storage. The data of chlorophyll and carotenoid content suggested the role of H2S in preventing chlorophyll breakdown and carotenoid accumulation in both grape rachis and pulp. In comparison to water control, exogenous H2S application maintained significantly higher levels of ascorbic acid and flavonoid and total phenolics and reducing sugar and soluble protein in grape pulp. Meanwhile, H2S significantly reduced the accumulation of malondialdehyde (MDA), hydrogen peroxide (H2O2), and superoxide anion (O2∙-) in grape pulp. Further investigations showed that H2S enhanced the activities of antioxidant enzymes ascorbate peroxidase (APX) and catalase (CAT) and decreased those of lipoxygenase (LOX) in both grape peels and pulp. In all, we provided strong evidence that H2S effectively alleviated postharvest senescence and rotting of Kyoho grape by modulating antioxidant enzymes and attenuating lipid peroxidation.

Broccoli (Brassica oleracea var. italica) is an important vegetable crop rich in vitamins and sulforaphane. However, the floral heads of broccoli experience rapid postharvest senescence. Here we found that hydrogen sulfide (H2S) treatment alleviated dark-promoted senescence in broccoli florets. H2S delayed the symptoms of senescence and maintained higher levels of chlorophyll and Rubisco and lower protease activity compared with water control. Gene expression analysis showed that H2S down-regulated the expression of chlorophyll degradation-related genes BoSGR, BoNYC, BoCLH1, BoPPH, and BoRCCR. Expression of lipoxygenase gene BoLOX1 and the genes involved in the ethylene synthesis pathway, BoACS2 and BoACS3, were also down-regulated by H2S. The reduced expression level in cysteine protease gene BoCP3 and aspartic protease gene BoLSC807 suggested the role of H2S in alleviating protein degradation during broccoli senescence. H2S up-regulated the expression of sulfur metabolism genes BoSR and BoOASTL, and the antioxidant gene BoCAT. These results show that H2S plays a vital role in alleviating broccoli senescence through a broad regulation on gene expression of reactive oxygen species (ROS) metabolism genes, ethylene synthesis genes, and protease genes.

Clostridium difficile is the causative pathogen for antibiotic-related nosocomial diarrhea. For epidemiological study and identification of virulent clones, a new binary typing method was developed for C. difficile in this study. The usefulness of this newly developed optimized 10-loci binary typing method was compared with two widely used methods ribotyping and multilocus sequence typing (MLST) in 189 C. difficile samples. The binary typing, ribotyping and MLST typed the samples into 53 binary types (BTs), 26 ribotypes (RTs), and 33 MLST sequence types (STs), respectively. The typing ability of the binary method was better than that of either ribotyping or MLST expressed in Simpson Index (SI) at 0.937, 0.892 and 0.859, respectively. The ease of testing, portability and cost-effectiveness of the new binary typing would make it a useful typing alternative for outbreak investigations within healthcare facilities and epidemiological research.

Mining important persons is significant to network security and computer forensics nowadays in researches on email network centralization. Traditional PageRank algorithm is prone to be affected by interferential nodes because it distributes PR values evenly. This paper proposes a method which decomposes email network into different layers based on the core number, eliminates the interferential nodes in outer layers to decrease impact of interferential nodes and complexity of following procedure. Besides, it proposes an improved PageRank algorithm which partially solves the bias problem on nodes’ weighting, ranks the nodes quantitatively. The experiments indicate that it improves the accuracy and reduces the computational complexity in mining important nodes from email network.

We study a single machine slack due date assignment (usually referred to as SLK) scheduling problem with deteriorating jobs and a rate-modifying activity. The deterioration effect manifest such that the job processing time is a function of its starting time in a sequence. The rate-modifying activity is an activity that changes the processing rate of machine, i.e., the machine performs a rate-modifying activity. Hence the actual processing time of a job is a variable, which depends not only on its starting time in a sequence but also on whether it is scheduled before or after a rate-modifying activity. The goal is to schedule the rate-modifying activity, the optimal common flow allowance and the sequence of jobs to minimize the total earliness, the total tardiness and the common flow allowance cost. We show that the problem remains polynomially solvable under the proposed model.

Filling unbalance is a critical defect for injection mould. When the upper and lower covers of soap plastic box are produced by injection mold at the same time, filling unbalance in injection would appear because of the different dimensions of the two parts. For advancing the quality of the soap plastic box, the runner system is optimized with the filling analysis module and flow runner balance module of moldflow simulation software. The three-dimensional geometrical models of the two covers are constructed using Pro/e software. In moldflow the runner balance optimization of the soap box compounding cavity is analysis. The results indicate the optimized cross section of the runners can reduce the flow unbalance ratio from 3.38% to 0.73%, and the filling time and pressure can satisfy the demands. According to the analysis results moldflow is appropriate for runner balance design of the plastic products.

As inner cooling grinding can break the closure of grinding area and solve practical engineering problems resulting from too high grinding temperature, it can be used to realize the grinding of difficult-to-machine materials, such as Titanium alloy, in a general machine such as M7130. An inner cooling grinding wheel, a grinding carriage and cutting fluid system are the key parts of a inner cooling grinding system, and an inner cooling grinding wheel must be fixed on a plain grinder by a grinding carriage, so this paper mainly studies how to design and manufacture a tooling for an inner cooling grinding wheel, which has simple construction and reliable performance. Grinding experiment shows it can dramatically reduce cutting temperature and effectively avoid grinding burn, thus grinding quality is improved.

For differences of time-domain energy distribution of different gear fault vibration signal, an analytical method based on local mean decomposition (LMD) and least squares support vector machine (LS-SVM) is proposed to apply to gear fault diagnosis. First vibrational signal of gear is decomposed into a series of product functions (PF) by LMD method. Then extracting energy characteristic parameters of PF components which contain main fault information to constitute a fault feature vectors, which is considered as input sample of well-trained LS-SVM, and then identifying working state and fault type of different gear can be identified accurately and effectively by diagnostic method based on LMD and LS-SVM.

Ultrasonic EDM which is a novel machining approach for hard materials is developed quickly. In ultrasonic EDM processing the piezoelectric vibrator has a very important influence on the quality of the machining. The purpose of the paper is to study the performance parameters such as the first longitudinal modal frequency, node plane and vibration amplitude of the vibrator . Through FEM (finite element method ) simulation method the relation between structure parameters and the performance parameters are obtained. The results of the paper are very effective to design a vibrator for ultrasonic EDM.

The dynamic performance of the piezoelectric vibrator used in ultrasonic EDM machine in natural frequencies has a great effect on machining precision. Firstly, Through theoretical analysis the dynamic characteristics of the piezoelectric vibrator is obtained. Then the three-dimensional model of the piezoelectric vibrator is constructed by using PRO/E software, and model analysis is carried by using FEM software. Through theoretical analysis and FEM simulation, the appropriate working frequency and mode of the piezoelectric vibrator was found, and the piezoelectric vibrator was fabricated. Experimented results show that the model analysis of frequency is accord with that of FEM.

A piezoelectric vibrator in ultrasonic EDM processing is designed and fabricated. The structure parameters of the vibrator have a very significant impact on the dynamic characteristics. The effect of different structural parameters on dynamic characteristics of the vibrator is analyzed by using Finite Element Method(FEM), and the modal frequency and vibration amplitude sensitivities of the vibrator to the structural parameters are investigated by modal and harmonic analysis.. According the results of the sensitivities analysis, the structural parameters which have an important impact on the working modal frequency and the vibration amplitude are achieved. The vibrator is designed and made based on the above theoretical analysis. The dynamic characteristics of the piezoelectric vibrator are tested. In the case of exciting voltage of 100V (peak-peak value), the first-order longitudinal vibration frequency (working frequency) of the vibrator is 27.36 kHz, vibration amplitude is about9.8μm, which is in compliance with the theoretically analytical results.

In this paper we consider single-machine group scheduling problems with effects of learning and deterioration at the same time. By effects of learning and deterioration, we mean that the group setup times are general linear functions of their starting times and the jobs in the same group have general position-dependent and time-dependent learning effects. The objective of scheduling problems is to minimize the makespan and the sum of completion times, respectively. We show that the problems remain solvable in polynomial time under the proposed model.

A V-shaped linear ultrasonic motor with a stator and a slider has been developed. The stator consists of two orthogonal Langevin actuators which is joined together in their ends. The Langevin actuators are symmetrical and have same first order longitudinal mode. A linear motion of the slider is generated by friction between the stator and the slider when the two first order longitudinal modes are combined. In this paper, structural dynamic design on the stator is completed with Finite Element Method(FEM). The prototype motor is manufactured based on the design results. The operation modes of the stator and the output performances of the motor are tested. The diameter and length of the Langevin actuator are 20mm and 58mm, respectively. The mass of the stator is 290g. The first order longitudinal frequency of the Langevin actuator is 27.47 kHz (FEM simulating result) and 27.36 kHz (expermental result), respectively. The maximum driving force of the motor approaches 40N. The maximum velocity of the motor is about 100 mm/s. So the stator’s operation modes are consistent with the design results, and the motor output properties have characteristics of slower velocity and stronger force.

Pharmacotherapy and cognitive-behavioral therapy (CBT) present limitations when they are used to treat obsessive-compulsive disorder (OCD), a severe and debilitating psychiatric disorder. To search for more efficacious treatment, we investigated the effects of pharmacotherapy plus cognitive-coping therapy (pCCT) on adult OCD patients with overt or covert compulsions. Two hundred and fifteen OCD patients were randomized into pharmacotherapy plus psychological support (PPS, n=107) and pCCT (n=108). The Yale-Brown Obsessive Compulsive Scale (Y-BOCS) was used to measure severity of symptoms in the OCD patients. The Y-BOCS scores were significantly lower in pCCT than in PPS in both acute term (

Vast amount of information is accumulated in many forms during the usage and maintenance of new equipments under information circumstance. The method of data mining is presented in this paper to take full advantage of information and increase scientific decision level of equipment maintenance management. The method includes maintenance information structure analysis, data collection, data coordination, association rule mining and decision tree classification. A case study is applied to explain the method.

To study the natural history of HIV-1 infection among intravenous drug users (IDUs) detected in late 1989 in the study area and the factors related to survival of these IDUs infected with HIV.196 injecting drug users first detected during August and December, 1989 were observed in Ruili county, Yunnan province. Data gathered from the 20-year follow-up program was collected and analyzed retrospectively.After 20 years' follow-up period, 90.3% of the 196 IDUs with HIV infection died, 5.1% of them were still alive, and 4.6% were lost. The crude pre-AIDS mortality rate was 98.1/1000 person-years, and the AIDS mortality rate was 54.9/1000 person-years. Malaria, septicemia were the main causes of death among the natural diseases whereas overdose and accidental causes were the principal causes related to those non-disease deaths. The median survival time from sero-conversion to death was 8.6 years (95%CI: 7.6 - 9.7). The median survival time from sero-conversion to death due to AIDS was 11.3 years (95%CI: 10.3 - 12.8) with the incubation time as around 10.3 years. People older than 30 years at seroconversion and length of drug usage were associated with shorter survival time, with hazards ratios as 1.9 and 0.7, respectively.A high pre-AIDS mortality was observed among IDUs. Both the median survival time from sero-conversion to death and the HIV incubation period were shorter than that observed in the developed countries. Age of HIV infection seemed to have a strong effect on survival.

The distillery wastewater of Guangdong Jiujiang Distillery, which is characteristic of containing high organic matters and rich total nitrogen, was treated by a combination of methane fermentation and denitrification/nitrification processes. 80% of COD in the raw wastewater was removed by methane fermentation at the COD volumetric loading rate of 20 kg COD/(m3 x d) using the expanded granule sludge bed (EGSB) process. However, almost all the organic nitrogen in the raw wastewater was converted into ammonia by ammonification there. Ammonia and volatile fatty acids (VFA) remaining in the anaerobically treated wastewater were simultaneously removed utilizing VFA as an electron donor by denitrification occurring in the other EGSB reactor and nitrification using PEG-immobilized nitrifying bacteria with recirculation process. An aerobic biological contact oxidization reactor was designed between denitrification/nitrification reactor for further COD removal. With the above treatment system, 18000-28000 mg/L of COD in raw wastewater was reduced to less than 100 mg/L. Also, ammonia in the effluent of the system was not detected and the system had a high removal rate for 900-1200 mg/L of TN in the raw wastewater, only leaving 400 mg/L of nitrate nitrogen.
