Alloying can make conventional metals reach ultra-high strength, but this usually comes at dramatic loss of toughness. In this work, a desirable strength–toughness combination in high-strength low-alloy steel achieved via trace carbon addition. The significance of carbon in tailoring variant pairing and tuning impact toughness was elucidated from the perspective of crystallography and thermodynamics. As the carbon content increases, the packets and blocks are refined, and the − 40 ℃ impact toughness improves. The enhancement of impact toughness results from the higher density of block boundaries, and the fracture mode shifts from brittle fracture to ductile–brittle combined fractures, then to ductile fracture due to the increased carbon. Increasing the carbon content would lower the martensite start temperature (MS) temperature and driving force for martensitic transformation, and increase the strength of austenite matrix, which in turn contributes to producing more V1/V2 variant pairs to accommodate the transformation strain.

We elucidate here the effects of annealing twins on phase transformation products based on electron back-scatter diffraction analysis and corresponding microstructure visualization and quantification methods. Martensite and bainite were obtained by rapid continuous cooling and isothermal processing at different temperatures, respectively, which were designed to study variants formed in austenite with Σ3 boundary. The isothermal transformation near martensite start (Ms) temperature was most conducive in obtaining the highest content of twin-related V1/V2 variant pair. Based on the classical nucleation theory of martensite and bainite, respectively, the role of austenite Σ3 boundary in martensite and bainite transformation is illustrated.

Heart rate is still a controversial and unclear factor of stroke risk in atrial fibrillation. Indices combining parameters are more accurate predictors than single parameters. This article assessed the association of the BNP-to-albumin ratio (BAR), with the risk of stroke, and evaluated the relationship between heart rate and stroke risk. Data were retrospectively collected from the Zhongnan Hospital electronic records. Binary logistic regression assessed the association between BAR and the prediction of acute stroke in atrial fibrillation. Spearman's correlation analysis evaluated the correlation between heart rate and BAR. The specificity and sensitivity of the BAR index were determined by ROC curve analysis. A total of 197 participants were involved, including 119 cases and 78 controls. The mean BAR was significantly higher for cases than for controls P = 0.00 while the difference in mean heart rate did not reach statistical significance P = 0.08. Using binary logistic analysis, BAR was a significant predictor of stroke in AF, OR = 1.67 95%CI [1.09-2.55] P = 0.018. The correlation between BAR and heart rate was significant, the correlation coefficient was r = 0.15 P = 0.03. ROC curve analysis showed that at a cut-off value of 2.01*10-9 g/L 93% of patients with a BAR of less than 2.01 did not have an acute stroke and only the 60% with a BAR greater than 2.01 experienced an acute stroke. It's been suggested that the BNP to albumin ratio and heart rate could be used to estimate the risk of stroke among hospitalized atrial fibrillation patients, thus contributing to the implementation of appropriate measures.

Background Post cardiac injury syndrome (PCIS) is a troublesome but not uncommon complication following catheter ablation of arrhythmias. We aimed to study the clinical features of ablation-associated PCIS. Methods For this purpose, we conducted a computerised literature search that identified 19 published cases, and we additionally included another two new cases from our centres. Twenty-one (21) cases of PCIS following ablation were analysed. Results Among the 21 cases, PCIS most commonly occurred after atrial flutter/fibrillation (AFL/AF) ablation (71.4%), followed by atrioventricular re-entrant tachycardia (AVRT) ablation (9.5%), atrioventricular node (AVN) ablation (9.5%), atrioventricular nodal re-entrant tachycardia (AVNRT) ablation (4.8%) and ventricular tachycardia (VT) ablation (4.8%). Thirty-eight (38) per cent of PCIS was suggested to be secondary to cardiac perforation. Specific symptoms or features include pleuritic chest pain (76.2%), fever (76.2%), elevated markers of inflammation (76.2%), pericardial effusion (90.5%), pleural effusion (71.4%) and pulmonary infiltrates (28.6%). Interestingly, all the six cases with pulmonary infiltrates were following AFL/AF ablation (6/15, 40%). Serious clinical manifestations include cardiac tamponade, massive pleural effusion with hypoalbuminaemia and hyponatraemia, and massive pulmonary infiltrates with hypoxaemia. Notably, empiric antibiotic therapy was used in seven cases including five with pulmonary infiltrates but failed to work. No mortality occurred during a mean follow-up of 4.1 ± 5.3 (1 to 19) months. Conclusions Catheter ablation of AFL/AF was most commonly involved in ablation-associated PCIS. Pulmonary infiltrate is an important feature of PCIS following AFL/AF ablation and may be misdiagnosed as pneumonia. Although PCIS is troublesome and even dangerous, it does carry a benign prognosis.

Implantable cardioverter-defibrillator (ICD) has been increasingly used to prevent sudden death in patients with high risk of ventricular arrhythmias (VAs). Chronic total occlusion (CTO) is a severe condition of coronary artery disease. However, the prognostic impact of CTO in ICD recipients remains controversial. We systematically searched the PubMed, EMBASE, and Cochrane databases for documents published before October 1, 2017 to identify studies that investigated the prognostic impact of CTO on VAs and all-cause mortality (ACM) in ICD recipients. The effect size was expressed by hazard ratio (HR) and 95% confidence interval (CI). Overall, 6 studies with 1,423 patients were included. We found that CTO independently predicted the occurrence of VAs in the global population (HR 1.77, 95% CI 1.14 to 2.76, p = 0.01). Subgroup analysis revealed that CTO only predicted VAs in patients receiving ICD as secondary prevention (HR 1.96, 95% CI 1.55 to 2.48, p0.01). However, CTO was not associated with ACM in the global population (HR 1.51, 95% CI 0.99 to 2.32, p = 0.06). Further subgroup analysis still showed no association between CTO and ACM in patients receiving ICD as primary prevention (HR 1.49, 95% CI 0.43 to 5.21, p = 0.53) or secondary prevention (HR 1.56, 95% CI 0.87 to 2.78, p = 0.13). In conclusion, CTO is an independent predictor of VAs in patients with secondary-prevention ICD, but not for ACM. Our study provided additional evidence for improving the risk stratification and management of VAs in ICD recipients.

The present paper investigated the relationship between low temperature impact toughness and microstructure of bainite in coarse-grained heat affected zone (CGHAZ) and intercritically rehazed CGHAZ (ICCGHAZ) of an offshore engineering steel from both the microstructure morphological and crystallographic aspects. In this work, six groups of samples simulated CGHAZ and ICCGHAZ were designated at three different cooling rates. The Charpy test results showed that the toughness in CGHAZ decreases dramatically with decrease of cooling rate, which was attributed to the microstructural evolution from lath bainite to granular bainite, accompanying with the size increase of Bain zone and the change of M/A morphology from film to block. The increase in hardenability by cooling rate promotes more crystallographic variants from different Bain groups. Meanwhile, the combination with controlled inter-spacing of block boundaries by self-accommodation below the critical Griffith crack length, micro-crack can be arrested by these high angle grain boundaries thereby suppressed brittle fracture initiation and increased fracture properties. However, the variation in toughness of ICCGHAZ is not a concern, since obtaining excellent toughness is scarcely accessible even if the matrix microstructure is analogous to CGHAZ. It was due to the formation of coarse M/A constituents (~2 μm) necklacing at the prior austenite grain boundary. The visualized crystallography suggested that the impact toughness was partially correlated to the configuration manner and the size of Bain zones as well via promoting highly misoriented angle (>45°) boundaries, which in turn effectively deflected or arrested the brittle crack propagation.

Aims We evaluated the feasibility of left atrial appendage (LAA) closure guided by the image fusion of integrating fluoroscopy into 3D computed tomography (CT). Methods and results A total of 117 consecutive patients who underwent LAA closure with or without the image fusion were matched (1:2). Each LAA closure step of the Image fusion group was guided by the preprocedure CT and image fusion, especially in the plan of LAA measurement and transseptal puncture. All patients were successfully implanted with a WATCHMAN closure device. Comparing the two groups, the mean number of recapture times and the number of devices per patient of the Image fusion group were significantly lower (0.4 ± 0.5 vs. 0.7 ± 0.8, P = 0.031 and 1.0 ± 0.2 vs. 1.1 ± 0.3, P = 0.027, respectively). The one-time successful deployment rate by the support of the image fusion was higher than in the control group (66.7% vs. 44.9%, P = 0.026). Each case of the Image fusion group was completely occluded with one transseptal puncture, while five of the Non-image fusion group required redo transseptal punctures. During the 45-day follow-up, both group cases presented occlusion efficiency and no major adverse cardiac events were observed. Conclusion Image fusion technique integrating fluoroscopy into the 3D CT is safe and feasible which can be easily incorporated into the procedural work-flow of percutaneous LAA closure. The fusion image can play an important alternative role in the plan of LAA measurement and transseptal puncture site for improving the LAA closure procedure.

Doxorubicin (DOX) is a widely used antineoplastic agent for a variety of carcinomas. However, it is cardiotoxic and leads to cardiomyopathy. Previous studies have indicated that omega-3 polyunsaturated acids (ω-3 PUFAs) have therapeutic effects on dilated and diabetic cardiomyopathies. However, whether ω-3 PUFAs exert therapeutic effects on DOX-induced cardiomyopathy remains unclear. In this study, we explored the effect and underlying mechanisms of docosahexaenoic acid (DHA), an important type of ω-3 PUFA, on DOX-induced cardiotoxicity and inflammation. H9C2 cardiac cells were exposed to DOX (5 μM) and interfered with by DHA (10 μM) for 4 hours. The effect of DHA on H9C2 cell viability was measured by Cell Counting Kit-8 assay. The levels of mRNA and protein expression of inflammatory cytokines were determined by real-time polymerase chain reaction and enzyme-linked immunosorbent assay, respectively. Reactive oxygen species and nitric oxide (NO) levels were determined by corresponding kits. The protein expression of key molecules in the nuclear factor-kappa B/inducible isoform of nitric oxide synthase/nitric oxide (NF-κB/iNOS/NO) signaling pathway was determined by western blotting. DOX-induced significant cytotoxicity and reactive oxygen species production in H9C2 cardiac cells. It also induced cardiac inflammation as evidenced by significantly increased expressions of inflammatory cytokines, including tumor necrosis factor-α, interleukin-6, interleukin-1β, monocyte chemoattractant protein-1, and inducible isoform of NO synthase. However, DHA effectively attenuated DOX-induced cytotoxicity and inflammation. A further mechanistic study revealed that DHA suppressed DOX-induced activation of the NF-κB/iNOS/NO signaling pathway in H9C2 cells. Our results indicate that DHA may protect against DOX-induced cardiotoxicity by inhibiting NF-κB/iNOS/NO signaling pathway activation.

Two regions with different degrees of centerline segregation in a bainitic steel were studied to explore the influence of segregation on hardenability and mechanical properties. With increased severity of segregation, the microstructure at centerline regions changed from lath bainite to martensite, associated with a deterioration of toughness, the mechanism of which was investigated from the perspective of crystallographic features. The high-level local enrichment of alloying elements led to a large packet and Bain zone size and strong variant selection, which were unfavorable for crack deflection and transformation strain accommodation. As a result, the toughness of severely segregated regions was decreased.

To evaluate the effect of different bone cement injection methods during percutaneous vertebroplasty(PVP) on vertebral morphology and cement diffusion.The clinical data of 52 patients with single-segment osteoporotic vertebral compression fracture treated from January 2016 to December 2017 were retrospectively analyzed. The patients were divided into hydraumatic group (28 cases) and pusher group (24 cases) according to bone cement injection method during PVP. By comparing visual analogue scale(VAS), height of anterior vertebral body, compression ratio, kyphosis angle before and after operation and analyzing filling ratio of bone cement in the first 1/3, median line and back 1/3 of the vertebral body in lateral X-rays and the conditions of bone cement diffusion in AP X-rays were to evaluate the effect of different bone cement injection methods on vertebral morphology and cement diffusion.Postoperative VAS was obviously improved in all patients and hydraumatic group was better than pusher group(Hydraulic delivery vertebroplasty (HDVP) has better clinical efficacy and it can guarantee sufficient distribution of bone cement into the fractured vertebra and preferably restore the morphology of vertebral body, which is worthy of clinical application.

The inhibition of transient outward potassium current (Ito) is the major ionic mechanism for quinidine to treat Brugada syndrome; however, quinidine is inaccessible in many countries. The present study compared the inhibitory effect of the nonselective β-adrenergic blocker carvedilol with quinidine on human Kv4.3 (hKv4.3, encoding for Ito) channel and action potential notch using a whole-cell patch technique in HEK 293 cell line expressing K C N D 3 as well as in ventricular epicardial myocytes of rabbit hearts. It was found that carvedilol and quinidine inhibited hKv4.3 current in a concentration-dependent manner. The IC50 of carvedilol was 1.2 μM for inhibiting hKv4.3 charge area, while the IC50 of quinidine was 2.9 μM (0.2 Hz). Both carvedilol and quinidine showed typical o p e n channel blocking properties (i.e. decreasing the time to peak of activation and increasing the inactivation of hKv4.3), negatively shifted the V1/2 of activation and inactivation, and slowed the recovery from inactivation of the channel. Although carvedilol had weaker in use- and rate-dependent inhibition of hKv4.3 peak current than quinidine, its reduction of the charge area was more than quinidine at all frequencies (0.2–3.3 Hz). Moreover, the inhibitory effect of carvedilol on action potential notch was greater than quinidine. These results provide the novel information that carvedilol, like quinidine, significantly inhibits hKv4.3 and action potential notch, suggesting that carvedilol is likely an alternative drug for preventing malignant ventricular arrhythmias in patients with Brugada syndrome in countries where quinidine is unavailable.

In this study, three different strains of Taxodium hybrid 'Zhongshanshan' varieties [T. hybrid 302 (T. distichum♀×T. mucronatum♂), T. hybrid 407 (T. mucronatum♀×T. distichum♂), T. hybrid 118 (T. hybrid 302 ♀×T. macronatum ♂)] and their parents, T. distichum and T. mucronatum, were applied to investigate the response of photosynthetic characteristics, antioxidant enzyme systems and morphological characteristics to drought stress and recovery. The results indicated that as drought days were prolonged, all plants' net photosynthetic rate (P本文研究了落羽杉和墨杉及其杂交后代中山杉302(落羽杉♀×墨杉♂)、中山杉407(墨杉♀×落羽杉♂)、回交代中山杉118(中山杉302♀×墨杉♂)在自然干旱胁迫和复水过程中,光合特征、抗氧化酶系统和形态特性等的响应.结果表明:随干旱时间的延长,所有植株的净光合速率逐渐降低、脯氨酸开始积累且抗氧化酶系统逐渐清除丙二醛的毒性.胁迫至第8天,落羽杉净光合速率的下降幅度最大,而中山杉118的水分利用效率最高、丙二醛含量最少;墨杉的超氧化物歧化酶活性和脯氨酸含量增长最大.复水2 d后,所有植物的参数均有不同程度的恢复,其中,中山杉118恢复速率最快,其净光合速率和脯氨酸含量分别恢复了74.4%和60.2%.复水9 d后,所有植株的测定指标基本恢复至或接近正常水平,其中,中山杉118的生物量未受影响且根冠比显著增加.植物的耐旱能力依次为墨杉中山杉118中山杉407中山杉302落羽杉.回交品种中山杉118的杂种优势明显,较大程度地遗传了墨杉的耐旱性,该结论可为耐旱中山杉品种的杂交选育和推广应用提供科学依据.

Severe postoperative complications can affect cardiac surgery patients. Levosimendan is a novel calcium sensitizer commonly administered after cardiac surgery. However, the patient benefits are controversial. PubMed, Embase, and the Cochrane library were systematically searched for randomized controlled trials comparing levosimendan with control in adult cardiac surgery patients. Twenty-five studies (3247 patients) were included. To examine microRNA-133a (miR-133a) endogenous expression in cardiomyocytes after ischemia-reperfusion (I/R) injury and study the effects of miR-133a overexpression on I/R injury-induced cardiomyocyte apoptosis. Dual-Luciferase Reporter Assay detected dynamic expression of miR-133a. In an in vitro hypoxia-reoxygenation (HR) injury model and an in vivo rat model of I/R injury, rat cardiomyocytes were transfected with miR-133a mimic to test the effects of miR-133a overexpression on apoptosis. MiR-133a and Death Associated Protein Kinase 2 (DAPK2) mRNA expression was measured using real-time-PCR, and DAPK2 protein expression was detected by western blotting. Annexin V-fluorescein isothiocyanate/propidium iodide (PI) double-staining measured the apoptosis rate in H9C2 cells and transferase dUTP nick end labeling assay quantified the cardiomyocyte apoptosis rate in tissues obtained from in vivo the rat model. DAPK2 is a target of miR-133a. Both in vitro and in vivo results confirmed that after expression of miR-133a mimics, miR-133a levels increased, which was accompanied by decrease in DAPK2 mRNA and protein expression. In H9C2 cells, HR injury caused a sharp decrease in miR-133a expression and a significant upregualtion of DAPK2 mRNA and protein levels. However, exogenous miR-133a expression led to a significant reduction in DAPK2 mRNA and protein levels despite HR injury. Similar results were obtained from in vivo I/R injury model. After HR injury or I/R injury the apoptosis rate of myocardial cells was highly elevated and decreased significantly only after transfection of miR-133a into cardiomyocytes. MiR-133a overexpression may inhibit I/R injury-mediated cardiomyocyte apoptosis by targeting DAPK2, leading to reduced DAPK2 protein, thus miR-133a may potentially have a high therapeutic value in I/R injury.

For decades, digoxin has been widely used to control ventricular rate in atrial fibrillation (AF). However, it remains controversial as to whether digoxin is associated with increased mortality in AF. In this study, we searched relevant studies that were published before December 1, 2014, in PubMed, EMBASE, and the Cochrane central databases. We systematically reviewed the references and performed a meta-analysis of 8 carefully selected studies with 302,738 patients who were included for the final analysis. It was shown that digoxin use was associated with increased risk of all-cause mortality in AF overall [hazard ratio (HR) = 1.375, 95% confidence intervals (CI), 1.201-1.574, P = 0.0001]. Subgroup analysis further revealed that digoxin was associated with increased all-cause mortality in patients with AF, which was complicated by heart failure (HF) (HR = 1.201, CI, 1.074- 1.344, P = 0.001), and in those subjects without HF (HR = 1.172, CI, 1.148-1.198, P = 0.0001). Sensitivity analyses found results to be robust. Our findings indicated that digoxin use was associated with significantly increased all-cause mortality in patients with AF regardless of concomitant HF. We suggest that digoxin should not be preferentially used over other rate control medications in AF.

It is known that adipokines can regulate the hypothalamic-pituitary-adrenal axis (HPAA). In this study, we confirmed that adiponectin regulates the HPAA by affecting pituitary corticotroph cells. Using RT-PCR and immunofluorescence, we determined that adiponectin receptors were expressed in pituitary corticotroph tumour cells (AtT-20 cells and human corticotroph tumours). Adiponectin stimulated calcium influx and increased basal ACTH secretion without affecting corticotrophin-releasing hormone (CRH)-stimulated ACTH secretion, which was most likely due to the expression of adiponectin repressing CRH receptor 1 (CRHR1). Adiponectin also acutely stimulated ACTH release in primary culture pituitary cells. Lastly, adiponectin directly phosphorylated 5' AMP-activated protein kinase (AMPK) in AtT-20 cells. The effects of adiponectin were mimicked by AICAR, which was blocked by compound C. Taken together, our results suggested that adiponectin stimulated ACTH secretion and down-regulated CRHR1, possibly via an AMPK-dependent mechanism in pituitary corticotroph cells.

This research is to explore the effects of traditional Chinese medicine Ginseng-spikenard heart-nourishing capsule on the inactivation of c-type Kv1.4 channels (Kv1.4∆N) in Xenopus laevis oocytes with two-electrode voltageclamp technique. Defolliculated oocytes (stage V-VI) were injected with transcribed cRNAs of ferret Kv1.4δN channels. During recording, oocytes were continuously perfused with ND96 solution (control group) and solution prepared from Ginseng-spikenard heart-nourishing capsule (experimental group). Results found that, at the command potential of +50 mV, the current of experimental group was reduced to 48.33±4.0% of that in control group. The inactivation time constants in control and experimental groups were 2962.56±175.35 ms and 304.13±36.22ms, respectively (P0.05, n=7). The recovery time of fKv1.4∆N channel after inactivation in control group and experimental groups was 987±68.39 ms and 1734.15±98.45 ms, respectively (P0.05, n=5). Ginseng-spikenard heart-nourishing capsule can inhibit the Kv1.4δN channel, which may be one of the mechanisms of underlying antiarrhythmia.

In order to set controller parameters correctly, a graphical stability analysis approach for active queue management (AQM) is proposed. The model of TCP/AQM is converted into a second-order system with time delay. The stability of the closed-loop AQM system is described in terms of characteristic quasi-polynomial. New necessary and sufficient stability criterion is deduced based on the inverse Nyquist curve and the negative frequency characteristic line. The relations between stabilizing boundary of proportional gain in PID controller and network parameters are investigated. Different stabilizing regions are compared to show the less conservatism of our approach, and simulation experiments implemented by both Matlab and Network Simulator validate our analysis. The specialty of the proposed approach lies in the lower complexity of the calculation procedure and intuition in the complex plane.

Objective There is definite therapeutic effect in tuina treating low back pain. The comparative observation was performed on the two types of tuina manipulation for degenerative lumbar spondylolisthesis, in order to explore the best tuina manipulation for lumbar spondylolisthesis.

Based on the multi-objective optimization strategy and linear matrix inequality approach, the problem of reliable guaranteed cost control with multiple criteria constraints is investigated for a class of uncertain discrete-time systems subject to actuator faults. A fault model in actuators, which considers outage or partial degradation in independent actuators, is adopted. The quadratic stability is proved to be independent of the disturbance and the upper bound of a quadratic cost index is improved. The reliable feedback controller is designed to minimize the upper bound of the quadratic cost index, place all the closed-loop poles in a specified disk, constrain the H∞ norm level of the disturbance attenuation into a given range and guarantee the magnitudes of control inputs less than the given bound, as well. Thus, the resulting closed-loop system can provide satisfactory stability, transient behavior, disturbance rejection level and optimized upper bound of the quadratic cost performance despite possible actuator faults. Glial-cell-line-derived neurotrophic factor (GDNF) is a most potent survival factor for dopaminergic neurons. In addition, GDNF was also found to promote neurite outgrowth in dopaminergic neurons. However, despite the potential clinical and physiological importance of GDNF, its mechanism of action is unclear. Therefore, we employed a state-of-the-art proteomic technique, DIGE (Difference in two-dimensional gel electrophoresis), to quantitatively compare profiles of phosphoproteins of PC12-GFRalpha1-RET cells (that stably overexpress GDNF receptor alpha1 and RET) 0.5 and 10 h after GDNF challenge with control. A total of 92 differentially expressed proteins were successfully identified by mass spectrometry. Among them, the relative levels of phosphorylated Hsp27 increased significantly both in 0.5 and 10 h GDNF-treated PC12-GFRalpha1-RET cells. Confocal microscopy and Western blot results showed that the phosphorylation of Hsp27 after GDNF treatment was accompanied by its nuclear translocation. After the mRNA of Hsp27 was interfered, neurite outgrowth of PC12-GFRalpha1-RET cells induced by GDNF was significantly blocked. Furthermore, the percentage of neurite outgrowth induced by GDNF was also reduced by the expression of dominant-negative mutants of Hsp27, in which specific serine phosphorylation residues (Ser15, Ser78 and Ser82) were substituted with alanine. Our data also revealed that p38 MAPK and ERK are the upstream regulators of Hsp27 phosphorylation. Hence, in addition to the numerous novel proteins that are potentially important in GDNF mediated differentiation of dopaminergic cells revealed by our study, our data has indicated that Hsp27 is a novel signaling molecule involved in GDNF-induced neurite outgrowth of dopaminergic neurons.

The suboptimal reliable guaranteed cost control (RGCC) with multi-criterion constraints is investigated for a class of uncertain continuous-time systems with sensor faults. A fault model in sensors, which considers outage or partial degradation of sensors, is adopted. The influence of the disturbance on the quadratic stability of the closed-loop systems is analyzed. The reliable state-feedback controller is developed by a linear matrix inequalities (LMIs) approach, to minimize the upper bound of a quadratic cost function under the conditions that all the closed-loop poles be placed in a specified disk, and that the prescribed level of H∞ disturbance attenuation and the upper bound constraints of control inputs’ magnitudes be guaranteed. Thus, with the above multi-criterion constraints, the resulting closed-loop system can provide satisfactory stability, transient property, a disturbance rejection level and minimized quadratic cost performance despite possible sensor faults.

Hydrolysis reaction of Fe(NO3)3 at a high temperature in the presence of urea as the homogeneous precipitant was studied. With the prepared ceramic filter balls loaded with α-Fe2O3 after high temperature calcination, the loading of α-Fe2O3 on the porous ceramic filter balls from Fe(NO3)3 solutions of different concentrations and mechanical stability of the loaded α-Fe2O3 were studied. The product was characterized using XRD and SEM. Adsorption experiments were conducted to evaluate the performance of the product in adsorbing NH3-N. It turned out that the specific surface area of the ceramic filter balls loaded with α-Fe2O3 had increased to 36.5387 m2/g from original 4.6127 m2/g. When the concentration of Fe(NO3)3 was 0.40 mol/L, the loading of α-Fe2O3 on the ceramic filter balls accounted for 8.4% of the total mass of the adsorbent and α-Fe2O3 was adsorbed on the filter balls very well. The adsorption isotherm of NH3-N on the ceramic filter ball adsorbent loaded with α-Fe2O3 was of Langmuir type. The saturated adsorption capacity was 3.33 mg/L, and the adsorption constant K was 0.1873. NH3-N was adsorbed by α-Fe2O3 more easily, which was a kind of specific adsorption.

Background Purkinje cells (PCs) have a steeper rate dependence of repolarization and are more susceptible to arrhythmic activity than do ventricular myocytes (VMs). Late sodium current (I NaL ) is rate dependent and contributes to rate dependence of repolarization. Objective This study sought to test our hypothesis that PCs have a larger rate dependence of I NaL , contributing to their steeper rate dependence of repolarization and higher susceptibility to arrhythmic activity, than do VMs. Methods I NaL was recorded in isolated rabbit PCs and VMs with the whole-cell patch clamp technique. Action potential was examined using the microelectrode technique. Results Compared with VMs, PCs exhibited a significantly larger rate dependence of I NaL with a larger I NaL to basic cycle length (BCL) slope. Moreover, PCs had a larger rate dependence of I NaL decay and slower recovery kinetics. Interestingly, the larger rate dependence of I NaL matched to a steeper rate dependence of action potential duration (APD) in PCs. The I NaL blocker tetrodotoxin significantly blunted, while the I NaL enhancer anemone toxin (ATX-II) significantly increased, the rate dependence of I NaL and APD in PCs and VMs. In the presence of ATX-II, the rate dependence of I NaL in PCs was markedly larger than that in VMs, causing a much steeper rate dependence of APD in PCs. Accordingly, PCs exhibited greater rate-dependent electrical instability and were more prone to ATX-II–induced early afterdepolarizations, which were completely inhibited by the I NaL inhibitor ranolazine. Conclusion PCs have a significantly larger rate dependence of I NaL than do VMs because of distinctive I NaL decay and recovery kinetics, which contributes to their larger rate adaptation, and simultaneously predisposes them to a higher risk of arrhythmogenesis.

We report a case of severe fulminant myocarditis that closely mimicked acute inferior ST-segment elevation myocardial infarction (STEMI) and presented with refractory cardiogenic shock, multiple life-threatening arrhythmias and rapidly progressive liver failure. This case was successfully differentiated from STEMI by emergency coronary angiography. Recurrent cardiogenic shock was reversed by intra-aortic balloon pumping (IABP). Life-threatening arrhythmias including ventricular tachycardia, ventricular fibrillation, and high-degree atrioventricular block (AVB) were terminated by immediate cardioversion and temporary pacemaker. High-dose hydrocortisone effectively attenuated the inflammatory injury to the myocardium. The patient recovered and was well at the follow-up visit four months after discharge. Resumo: Relatamos um caso real de miocardite fulminante grave simulando enfarte agudo do miocárdio agudo inferior com supra desnivelamento ST (STEMI) com choque cardiogénico refratário, arritmias fatais múltiplas e lesão hepática funcional progressiva. Detetou-se com eficácia que não se tratava de STEMI através de angiografia coronária de urgência. O choque cardiogénico recorrente foi resolvido com eficiência através de balão intra-aórtico. Arritmias fatais incluindo a taquicardia ventricular, a fibrilhação ventricular e o bloqueio auriculoventricular de alto grau foram ultrapassadas por cardioversão imediata e pacemaker temporário. Uma dose elevada de hidrocortisona atenuou eficazmente lesões inflamatórias no miocárdio. O doente ultrapassou esta situação e estava totalmente recuperado na consulta de follow-up quatro meses após a alta. Warfarin is yet the most widely used oral anticoagulant for thromboembolic diseases, despite the recently emerged novel anticoagulants. However, difficulty in maintaining stable dose within the therapeutic range and subsequent serious adverse effects markedly limited its use in clinical practice. Pharmacogenetics-based warfarin dosing algorithm is a recently emerged strategy to predict the initial and maintaining dose of warfarin. However, whether this algorithm is superior over conventional clinically guided dosing algorithm remains controversial. We made a comparison of pharmacogenetics-based versus clinically guided dosing algorithm by an updated meta-analysis. We searched OVID MEDLINE, EMBASE, and the Cochrane Library for relevant citations. The primary outcome was the percentage of time in therapeutic range. The secondary outcomes were time to stable therapeutic dose and the risks of adverse events including all-cause mortality, thromboembolic events, total bleedings, and major bleedings. Eleven randomized controlled trials with 2639 participants were included. Our pooled estimates indicated that pharmacogenetics-based dosing algorithm did not improve percentage of time in therapeutic range [weighted mean difference, 4.26; 95% confidence interval (CI), -0.50 to 9.01; P = 0.08], but it significantly shortened the time to stable therapeutic dose (weighted mean difference, -8.67; 95% CI, -11.86 to -5.49; P < 0.00001). Additionally, pharmacogenetics-based algorithm significantly reduced the risk of major bleedings (odds ratio, 0.48; 95% CI, 0.23 to 0.98; P = 0.04), but it did not reduce the risks of all-cause mortality, total bleedings, or thromboembolic events. Our results suggest that pharmacogenetics-based warfarin dosing algorithm significantly improves the efficiency of International Normalized Ratio correction and reduces the risk of major hemorrhage.

Embryoid bodies (EBs) with large starting numbers of embryonic stem cells (ESCs) have a greater degree of cardiac differentiation than from low numbers of EBs. However, the biological roles of signaling molecules in these effects are not well understood. Here, we show that groups of EBs with different starting numbers of ESCs had differential gene expression patterns for Wnt5a and Wnt11. Wnt11 significantly increased the percentage of beating EBs by up-regulating the expression of the cardiac-specific genes. Wnt5a did not show these effects. Moreover, Wnt11 significantly increased the level of phosphorylated Jun N-terminal kinase. The inhibition of the JNK pathway by SP600125 blocked the effects of Wnt11. Thus, enrichment of cardiac differentiation in groups of EBs with a larger starting number of ESCs is mediated by the Wnt11-JNK pathway.

Neuregulin-1β (NRG-1β)/ErbB signaling plays crucial roles in the cardiac differentiation of mouse embryonic stem cells (ESCs), but its roles and the underlying mechanisms in cardiac differentiation are incompletely understood. This study showed that NRG-1β significantly increased the percentage of beating embryoid bodies (EBs) and up-regulated the gene expressions of Nkx2.5, GATA4, α-actin, MLC-2v, and ANF in a time-dependent manner, with no effect on the gene expressions of HCN4 and Tbx3. Inhibition of ErbB receptors with AG1478 significantly decreased the percentage of beating EBs; down-regulated the gene expressions of Nkx2.5, GATA4, MLC-2v, ANF, and α-actin; and concomitantly up-regulated the gene expressions of HCN4 and Tbx3 in a time-dependent manner. Moreover, the up-regulation of transcripts for Nkx2.5 and GATA4 by NRG-1β was blocked by the extracellular signal-related kinases (ERK) 1/2 inhibitor, U0126. However, U0126 could not inhibit the transcript up-regulations of MLC-2v and ANF by NRG-1β. The protein quantitation results were consistent with those of gene quantitation. Our results suggest that NRG-1β/ErbB signaling plays critical roles in the cardiac differentiation of mouse ESCs and in the subtype specification of cardiomyocytes in a time-dependent manner. The ERK1/2 pathway may be involved in the early cardiogenesis, but not in the subtype specification of cardiomyocytes.

Purpose: To investigate if ellagic acid exerts neuroprotective effects in hypoxic ischemic (HI) brain injury by inhibiting apoptosis and inflammatory responses. Methods: Separate groups of rat pups from post-natal day 4 (D4) were administered with ellagic acid (10, 20 or 40 mg/kg body weight) orally till post- natal day 10 (D10). On D10, the rats were subjected to HI brain injury. Following HI injury, infarct size, weight and volume of the brain were measured. Apoptosis was assessed by Fluoro-Jade C staining. Expression of caspases (caspase-3, 8 and 9), apoptotic pathway proteins (Bax, Bad, Bcl-2 and Bcl-xL), MAPKs, NF κB(p65) and p-IK-Bα were assessed by western blotting. mRNA levels of inflammatory mediators (TNF-α, IL-1α, IL-1β, iNOS, COX-2) were analyzed. Type 2 diabetes (T2D) is associated with perturbed innate immunity. Macrophages, bridging innate immunity and metabolic disturbances, play important roles in controlling immune homeostasis. However, the effect of long-term diabetic milieu (DM) on the functions and phenotypes of macrophages is still not clear. In this study, we used resident peritoneal macrophages (RPMs) from 5-month-old db/db mice to investigate the changes of macrophages. It was found that RPMs in db/db mice significantly reduced phagocytosis and adhesion capacity. After standardization with body weight, the number of F4/80+ RPMs markedly reduced in db/db mice, and, furthermore, the macrophages skewed to M2-polarizated macrophages. The results of morphology found that the RPMs shape of db/db mice was nearly round, but the RPMs shape of control mice was spindle-shaped and irregular. In this study, we found the cell numbers, morphology, and innate immunity functions of RPMs in 5-month-old type 2 diabetic mice (db/db mice) obtained by abdominal cavity lavage were significantly altered. Importantly, we also found the remarkably increased M2-RPMs in diabetic mice for the first time.
