Objective: The emergence of multidrug-resistant (MDR) Escherichia coli strains has significantly constrained antibiotic treatment options, while the spread of antimicrobial resistance genes (ARGs) and mobile genetic elements exacerbates the situation. This study delves into an MDR E. coli strain, QMM-01, which uniquely co-expresses β-lactamases from all four recognized classes, aiming to uncover the underlying mechanisms of its resistance and assess its potential for global spread. Methods: E. coli QMM-01, isolated from a burn patient, underwent antibiotic susceptibility testing through standard automated procedures commonly employed in clinical settings, with further test by immunochromatographic tests for carbapenemases. For genomic insights, whole-genome sequencing (WGS) was performed using both PacBio Sequel and Illumina NovaSeq platforms, supplemented by bioinformatics analyses to predict antimicrobial resistance genes, determining serotypes, performing multilocus sequence typing, and conducting comparative genomic analysis. Results: QMM-01 exhibited resistance to a broad spectrum of β-lactam antibiotics, including carbapenems and ceftazidime-avibactam, with aztreonam being the sole exception. The resistance profile of the strain might primarily be due to the production of class B metallo-β-lactamases. WGS revealed the presence of a chimeric plasmid, pQMM-2-NDM-5, carrying the blaNDM-5 gene and exhibiting similarities with plasmids from diverse geographical regions. This plasmid contains 161 predicted coding sequences and harbors resistance genes for 13 different antibiotics, forming a resistance island with a complex genetic environment. Conclusion: This study underscores the global challenge posed by antibiotic resistance and emphasizes the need for international collaboration in antibiotic stewardship to mitigate the spread of resistance genes.

Carbides play an important role in influencing the phase transformation and mechanical properties of steels. In this work, the microstructure of H13 steel is tailored by controlling the pre-precipitation of VC carbides at 950 °C to obtain excellent mechanical properties. It is revealed that the pre-precipitation of VC carbides decreases the carbon content of austenite, thus resulting in a high temperature of Ms. With the increasing amount of pre-precipitated VC carbide, martensite transformation is firstly accelerated by the enhanced transformation driving force, and then decelerated due to the hindering effect of VC particles on the growth of martensite laths. The improvement in strength, ductility and toughness of quenched H13 steel can be attributed to the pre-precipitated VC carbides and the refined martensite blocks. The tempered H13 steel with pre-precipitated VC carbides also exhibits excellent ductility and toughness. It can be concluded that controlling the pre-precipitation of VC carbides from austenite is an effective method to tailor the mechanical properties of H13 steel.

Abstract Background Tracheobronchial mucormycosis is a fatal opportunistic infection that mainly causes airway stenosis and is difficult to manage clinically. Case presentation We report a case of severe tracheal stenosis caused by tracheobronchial mucormycosis in a 37-year-old female with a history of hyperthyroidism. She developed agranulopenia after oral methimazole administration and subsequently experienced asthma with dyspnea. Bronchoscopy, sputum culture, colony mass spectrometry, and microscopic cotton orchid staining confirmed tracheobronchial mucormycosis. The patient received venous-venous extracorporeal membrane oxygenation (VV-ECMO) and prolonged intratracheal instillation of amphotericin B (AmBD), combined with amphotericin B liposome (L-AmB) and isavuconazole intravenous infusion, ultimately resulting in successful treatment. Conclusion VV-ECMO combined with prolonged intratracheal instillation of AmBD is an effective method for the treatment of tracheobronchial mucormycosis.

Empirical analysis of the relative effectiveness of the Giant Panda National Park (GPNP) system can promote the optimization and improvement of its management level. Normalized Difference Vegetation Index (NDVI) is a key indicator to measure the health of ecosystems, which can effectively quantitatively reveal the spatial and temporal changes of ecological protection effects. This study evaluated the relative effectiveness of Normalized Difference Vegetation Index (NDVI) protection in the Sichuan area of the GPNP from 2000 to 2020 using the propensity score matching model (PSM). It also explored the influencing factors and interactions of each period by combining the Optimal Parameter-based Geographical Detector Model (OPGD). The results showed that: 1) The study area’s Relative Effectiveness Index (REI) was positive, suggesting effective ecological protection. The REI fell from 0.044 in 2000 to 0.031 in 2015 and although it then increased to 0.034 in 2020 to a small extent, the REI showed an overall decreasing trend, and the conservation effect has weakened. 2)The REI change patterns varied in different functional zones of the area, with a general fluctuation and decline, in which the Minshan and Baishuijiang Core Protection Area (MBJ-CPA) as a whole first rise and then fall, and it is the area with the best relative effectiveness of protection. 3) Natural factors such as temperature and elevation are the main factors affecting NDVI, while the influence of policy and economic factors such as the level of protected areas and distance to towns are increasing. The Qionglaishan and Adjacent Areas General Control Area (QLA-GCA) is dominated by the interaction of landscape pattern index with its remaining factors, and the rest of the functional areas are dominated by the interaction of natural factors such as temperature, evapotranspiration with its remaining factors. Therefore, in future development, the Qionglaishan Areas need to pay more attention to the optimization of landscape patterns, while the other areas need to pay more attention to the impact of climate change on the ecosystem. This study can provide a reference for the improvement and management of ecological protection of the GPNP system in the future.

Abstract Double-layer island working face main roadway coal pillars are affected by complex mining stress superposition, when different coal pillar width combinations, the surrounding rock stress field will produce different degrees of regional loading increase effect; the study of the surrounding rock stress field regional superposition loading increase law is meaningful to explaining the failure mode of the roadway and determining the critical control area. This study combines numerical simulation with on-site monitoring and other methods and draws the following conclusions: The superimposed loading increase law (“decreasing” → “increasing”) of the abutment pressure and deviatoric stress in the lower coal seam of the double-layer island working face during the mining; the type of the principal stress deflection in the advance working face region; and by obtaining the three types of development morphology of the deviatoric stress peak zone of the roadway and its corresponding nine evolution modes (one type of circular tube → four types of inverse hyperbolic body → four types of hyperbolic body) in the double-layered island working face mining. Indicated the critical reinforcement area corresponding to the main roadway when at different combinations of coal pillar widths; determined the main track roadway protective coal pillars width for 40 m and the shape of the roadway peak deviatoric stress zone is the inverse class hyperbolic body mode; according to the evolution mode of the peak deviatoric stress zone, determined the synergistic failure control program for the asymmetric critical zone of the roadway surrounding rock which is a targeted scientific support method; after the feedback of on-site monitoring and, the support program is reasonable and effective.

Abstract Modeling user intention with limited evidence in short-term historical sequences is a major challenge in session recommendation. In this domain, research exploration extends from traditional methods to deep learning. However, most of them solely concentrate on the sequential dependence or pairwise relations within the session, disregarding the inherent consistency among items. Additionally, there is a lack of research on context adaptation in session intention learning. To this end, we propose a novel session-based model named C-HAN, which consists of two parallel modules: the context-embedded hypergraph attention network and self-attention. These modules are designed to capture the inherent consistency and sequential dependencies between items. In the hypergraph attention network module, the different types of interaction contexts are introduced to enhance the model’s contextual awareness. Finally, the soft-attention mechanism efficiently integrates the two types of information, collaboratively constructing the representation of the session. Experimental validation on three real-world datasets demonstrates the superior performance of C-HAN compared to state-of-the-art methods. The results show that C-HAN achieves an average improvement of 6.55%, 5.91%, and 6.17% over the runner-up baseline method on Precision@K, Recall@K, and MRR evaluation metrics, respectively.

Abstract Topological insulators are characterized by exhibiting an internal insulating state but a surface conductor state, which makes them advantageous for applications in novel devices. However, for most given acoustic topological metamaterials, the operating frequency is relatively fixed and the effect of temperature on their topological properties is rarely considered. Therefore, a temperature-tunable acoustic topological insulator is constructed in this paper. The quadruple degenerate Dirac cone formed at $$\Gamma$$ Γ point can be opened by adjusting the temperature, causing topological band inversion between the doubly degenerate dipolar and quadrupole states, and achieving topological phase transition. The evolution of its topological state with temperature is numerically investigated and a novel topological acoustic waveguide is constructed. The switching effect of temperature on the waveguide device is verified by numerical simulation and experiment. Non-contact active modulation of edge states in the structure is achieved by temperature-controlled topological phases, exhibiting acoustic switching effects. This study can provide corresponding references for the intelligent control of acoustic topology in noise, vibration, and other aspects.

Cell lines are indispensable tools in biological studies. Decades of attempts have failed to establish a cell line in marine mollusks, which contribute to 70 % of the mariculture production in China. In the present study, FBS, scallop serum, scallop ovarian extracts, and FeCl3 were added to the L-15 basic medium to develop an optimized culture medium (L15-CFIIIFe) for culturing trochophore larval cells of Chlamys farreri, an important mariculture scallop in China. C. farreri trochophore larval (CfT) cells exhibited proliferative activity, and a CfT cell line was successfully established, which was passaged for 155 generations in vitro. The origin of the CfT cell line was verified as C. farreri using internal transcribed spacer (ITS) and 18 s rRNA gene amplification, genomic in situ hybridization (GISH), and karyotype analysis. The CfT cell line was a mixture of cells of three sizes (5–7, 9–12, and 18–22 μm), with a population doubling time of 75.6 h. Immunofluorescence detection of KLF4, c-MYC, MYOSIN, and 5-HT indicated that CfT cells may possess a certain degree of pluripotency. Four gene transfer methods were tested in the CfT cell line; however, the transfer efficiency (0.5 %-1 %) requires further optimization. We successfully established the first marine mollusk cell line, providing a valuable cellular tool for mollusk studies.

Copper-based materials play a vital role in the electrochemical transformation of CO2 into C2/C2+ compounds. In this study, cross-sectional octahedral Cu2O microcrystals were prepared in situ on carbon paper electrodes via electrochemical deposition. The morphology and integrity of the exposed crystal surface (111) were meticulously controlled by adjusting the deposition potential, time, and temperature. These cross-sectional octahedral Cu2O microcrystals exhibited high electrocatalytic activity for ethylene (C2H4) production through CO2 reduction. In a 0.1 M KHCO3 electrolyte, the Faradaic efficiency for C2H4 reached 42.0% at a potential of −1.376 V vs. RHE. During continuous electrolysis over 10 h, the FE (C2H4) remained stable around 40%. During electrolysis, the fully exposed (111) crystal faces of Cu2O microcrystals are reduced to Cu0, which enhances C-C coupling and could serve as the main active sites for catalyzing the conversion of CO2 to C2H4.

Zero-refractive index metamaterials have a wide range of applications in directional transmission and wave-front shaping due to their unusual acoustic properties. However, for most given acoustic topological metamaterials, the operating frequency is relatively fixed and the effect of temperature on their topological properties is rarely considered. Therefore, temperature-controlled tunable topological zero-refraction acoustic metamaterials are proposed in this paper. Firstly, a metamaterial with quadruple degenerate Dirac-like points at the center of the Brillouin zone is constructed, and the influence of temperature on the Dirac-like points is analyzed. The results show that the topological bandgap frequency range is more sensitive to temperature. The existence of pseudospin-polarized edge state is demonstrated by analysing the band structure of supercells with different topological phase phonon crystal. The topological zero-refraction property of the edge states outcoupled into free space is numerically demonstrated, and the non-contact active control of their operating frequencies can be realized by temperature. This study can provide a corresponding reference for the intelligent control of near-zero refractive index acoustic topological materials in elastic wave collimation and acoustic communication.

IntroductionIntestinal dysfunction poses a severe problem by preventing the digestion and absorption of nutrients. The gut, being the most vital organ for these processes, plays a crucial role in ensuring our body receives the nutrients it needs. We explored the mitigating effect of Morchella esculenta polysaccharides (MEP) on intestinal injury induced by lipopolysaccharides (LPS) through the modulation of intestinal flora.MethodsFor this purpose, Kunming mice (KM) were divided into three groups, namely, PC, PM, and PY. Group PY was treated with MEP, while groups PM and PY were induced with LPS.ResultsThe results showed that weight loss in the PM group was significantly greater than that in the PY group (P < 0.05), and the organ indexes of the lung and spleen in the PM group were significantly higher than those in the PC (P < 0.01) and PY (P < 0.05) groups. LPS caused severe injuries in KM mice in the PM group, characterized by broken villi. However, MEP treatment could alleviate this damage in the PY group, resulting in relatively intact villi. The serum analysis showed that tumor necrosis factor alpha (TNF-ɑ) (P < 0.01), interleukin 6 (IL-6) (P < 0.01), and 3,4-methylenedioxyamphetamine (MDA) (P < 0.05) levels were significantly higher in the PM group, while IL-10 (P < 0.001), superoxide dismutase (SOD) (P < 0.01) and glutathione peroxidase (GSH-Px) (P < 0.01) were significantly lower in that group. Interestingly, supplementation with MEP could lower the levels of TNF-ɑ, IL-10, IL-6, MDA while increasing the levels of superoxide dismutase (SOD) (P < 0.01) and GSH-Px. The gut microbiota analysis yielded 630,323 raw reads and 554,062 clean reads, identifying 3,390 amplicon sequencing variants (ASVs). One phylum and five genera were notably different among animal groups, including Escherichia_Shigella, Limosilactobacillus, unclassified_Geminicoccaceae, unclassified_Rhodobacteraceae, and Parabacteroides (P. distasonis).DiscussionIn conclusion, we found that MEP could mitigate the intestinal damage caused by LPS by modulating the inflammatory response, oxidative resistance, and intestinal flora of KM mice. Our results may provide insights into novel treatment options for intestine-related diseases.

Abstract Background The lint percentage of seed cotton is one of the most important parameters for evaluating seed cotton quality and affects its price. The traditional measuring method of lint percentage is labor-intensive and time-consuming; thus, an efficient and accurate measurement method is needed. In recent years, classification-based deep learning and computer vision have shown promise in solving various classification tasks. Results In this study, we propose a new approach for detecting the lint percentage using MobileNetV2 and transfer learning. The model is deployed on a lint percentage detection instrument, which can rapidly and accurately determine the lint percentage of seed cotton. We evaluated the performance of the proposed approach using a dataset comprising 66 924 seed cotton images from different regions of China. The results of the experiments showed that the model with transfer learning achieved an average classification accuracy of 98.43%, with an average precision of 94.97%, an average recall of 95.26%, and an average F1-score of 95.20%. Furthermore, the proposed classification model achieved an average accuracy of 97.22% in calculating the lint percentage, showing no significant difference from the performance of experts (independent-sample t-test, t = 0.019, P = 0.860). Conclusion This study demonstrated the effectiveness of the MobileNetV2 model and transfer learning in calculating the lint percentage of seed cotton. The proposed approach is a promising alternative to traditional methods, providing a rapid and accurate solution for the industry.

Abstract Background Larval settlement and metamorphosis represent critical events in the life history of marine benthic animals. Myoinhibitory peptide (MIP) plays a pivotal role in larval settlement of marine invertebrates. However, the molecular mechanisms of MIP involved in this process are not well understood. Results In this study, we evaluated the effects of thirteen MIP mature peptides on triggering the larval settlement of Urechis unicinctus (Xenopneusta, Urechidae), and determined that MIP2 was the principal neuropeptide. Transcriptomic analysis was employed to identify differentially expressed genes (DEGs) between the MIP2-treated larvae and normal early-segmentation larvae. Both cAMP and calcium signaling pathways were enriched in the DEGs of the MIP2-treated larvae, and two neuropeptide receptor genes (Spr, Fmrfar) were up-regulated in the MIP2-treated larvae. The activation of the SPR-cAMP pathway by MIP2 was experimentally validated in HEK293T cells. Furthermore, fourteen cilia-related genes, including Tctex1d2, Cfap45, Ift43, Ift74, Ift22, Cav1 and Mns1, etc. exhibited down-regulated expression in the MIP2-treated larvae. Whole-mount in situ hybridization identified two selected ciliary genes, Tctex1d2 and Cfap45, were specially expressed in circumoral ciliary cells of the early-segmentation larvae. Knocking down Tctex1d2 mRNA levels by in vivo RNA interference significantly increased the larval settlement rate. Conclusion Our findings suggest that MIP2 inhibits the function of the cilia-related genes, such as Tctex1d2, through the SPR-cAMP-PKA pathway, thereby inducing larval settlement in U. unicinctus. The study contributes important data to the understanding of neuropeptide regulation in larval settlement.

Background/Objectives: Bupleuri Radix is a plant in the Apiaceae family Bupleurum Chinense DC. or Bupleurum scorzonerifolium Willd. root. The dissimilarities in the metabolite profiles of plants directly correlate with the disparities in their clinical efficacy. Methods: Therefore, the wild Bupleurum Chinense DC. (YBC) and wild Bupleurum scorzonerifolium Willd. (YNC) were used as research materials. They were analyzed using the UPLC-MS/MS and the similarities and differences were uncovered based on differential metabolites. Results: Our results proved that the differences in clinical efficacy between YBC and YNC may be attributed to their distinct metabolite profiles, as follows: (1) a total of 12 classes of 2059 metabolites were identified in the roots, with phenolic acids, terpenoids, and flavonoids being the most abundant metabolic products, with 2026 shared components between the two, 2045 in YBC, and 2040 in YNC; (2) a total of 718 differential metabolites were identified, accounting for 35.44% of the shared metabolites. Among them, YBC had 452 metabolites with higher content relative to YNC, representing 62.95%, and 266 components with lower content, representing 37.05%; (3) the KEEG enrichment analysis results show that the differential metabolic pathways are flavone and flavonol biosynthesis, linoleic acid metabolism, arachidonic acid metabolism, sesquiterpenoid and triterpenoid biosynthesis, and linolenic acid metabolism. Conclusions: These new findings will serve as a foundation for further study of the BR biosynthetic pathway and offer insights into the practical use of traditional Chinese medicine in clinical settings.

Airwave interference presents a major source of noise in seismic exploration, posing significant challenges to the quality control of raw seismic data. With the increasing data volume in 3D seismic exploration, manual identification methods fall short of meeting the demands of high-density 3D seismic surveys. This study employs the YOLOv5 model, a widely used tool in object detection, to achieve rapid identification of airwave noise in seismic profiles. Initially, the model was pre-trained on the COCO dataset—a large-scale dataset designed for object detection—and subsequently fine-tuned using a training set specifically labeled for airwave noise data. The fine-tuned model achieved an accuracy and recall rate of approximately 85% on the test dataset, successfully identifying not only the presence of noise but also its location, confidence levels, and range. To evaluate the model’s effectiveness, we applied the YOLOv5 model trained on 2D data to seismic records from two regions: 2D seismic data from Ningqiang, Shanxi, and 3D seismic data from Xiushui, Sichuan. The overall prediction accuracy in both regions exceeded 90%, with the accuracy and recall rates for airwave noise surpassing 83% and 90%, respectively. The evaluation time for single-shot 3D seismic data (over 8000 traces) was less than 2 s, highlighting the model’s exceptional transferability, generalization ability, and efficiency. These results demonstrate that the YOLOv5 model is highly effective for detecting airwave noise in raw seismic data across different regions, marking the first successful attempt at computer recognition of airwaves in seismic exploration.

A purified polysaccharide with a galactose backbone (SPR-1, Mw 3,622 Da) was isolated from processed Polygonati Rhizoma with black beans (PRWB) and characterized its chemical properties. The backbone of SPR-1 consisted of [(4)-β-D-Galp-(1]9 → 4,6)-β-D-Galp-(1 → 4)-α-D-GalpA-(1 → 4)-α-D-GalpA-(1 → 4)-α-D-Glcp-(1 → 4,6)-α-D-Glcp-(1 → 4)-α/β-D-Glcp, with a branch chain of R1: β-D-Galp-(1 → 3)-β-D-Galp-(1→ connected to the →4,6)-β-D-Galp-(1→ via O-6, and a branch chain of R2: α-D-Glcp-(1 → 6)-α-D-Glcp-(1→ connected to the →4,6)-α-D-Glcp-(1→ via O-6. Immunomodulatory assays showed that the SPR-1 significantly activated macrophages, and increased secretion of NO and cytokines (i.e., IL-1β and TNF-α), as well as promoted the phagocytic activities of cells. Furthermore, isothermal titration calorimetry (ITC) analysis and molecular docking results indicated high-affinity binding between SPR-1 and MD2 with the equilibrium dissociation constant (KD) of 18.8 μM. It was suggested that SPR-1 activated the immune response through Toll-like receptor 4 (TLR4) signaling and downstream responses. Our research demonstrated that the SPR-1 has a promising candidate from PRWB for the TLR4 agonist to induce immune response, and also provided an easily accessible way that can be used for PR deep processing.

[Abstract]: Objective: This study aims to evaluate the efficacy and safety of using a strip-shaped cymba conchae orthosis for the nonsurgical correction of complex auricular deformities. Methods: Clinical data were collected from 2020 to 2021 for 6 patients who underwent correction using a strip-shaped cymba conchae orthosis. The indications, corrective effects, and complications associated with use of the orthosis were analyzed. Results: There were four indications for treatment: cryptotia with helix adhesion; cryptotia with grade I microtia; cryptotia with excessive helix thickness; and auricular deformity beyond the treatment time window (≥6 months). Excellent corrective effects were observed in all 6 patients. Complications occurred in one patient, who recovered after symptomatic treatment. Conclusion: The use of a strip-shaped cymba conchae orthosis alone or combined with a U-shaped helix orthosis presents a feasible approach for correcting complex auricular deformities or deformities beyond the treatment time window in pediatric patients.

The paper proposes and verifies a small-angle measurement method based on the defect spot mode of the position-sensitive detector (PSD). With the output characteristics of the PSD in the defect spot mode and the size transformation properties of a focused beam, the measurement sensitivity can be significantly improved. Calibration experiments with the piezoelectric transducer (PZT) indicate that compared with the current PSD-based autocollimation method, the proposed method can improve the sensitivity of small-angle measurement by 57 times, and the measurement sensitivity of the proposed method can be further improved by optimizing the system parameters, while the proposed method has the advantages of a simple system and high real-time performance. Therefore, the proposed method is expected to be used in high-precision motion error detection, as well as in shape and position measurement.

Polygonatum cyrtonema Hua (PCH), as an important economic crop, is used as raw industrial materials and traditional Chinese medicine. There are significant variations in the quality of PCH from different geographical origins. It can be due to the change of the endophytic fungi and soil microbial communities of PCH. Therefore, the aim of this study is to investigate the composition and functional prediction of the main microbial communities in the rhizomes and soil of PCH and explore their impact on medicinal quality. High-throughput sequencing techniques targeting ITS and 16S rDNA were employed to compare the structure and biodiversity differences of endophytic fungi in the rhizomes and soil microbial communities of PCH from 12 different locations in Sichuan and Guangxi province. Heatmap analysis was used for comprehensive statistics and visualization of the richness of rhizome and soil microbial communities from all locations. Venn analysis was conducted to determine the total number of shared fungi between rhizomes and soil, and GraphPad Prism analysis was employed to predict and compare the microbial communities related to phenotypes at the genus level in Sichuan and Guangxi. Tax4Fun and Fungild were used for metabolic function prediction of microbial communities in the rhizomes and soil of PCH. The results revealed the identification of 19,387 bacterial amplicon sequence variants (ASVs) in the rhizomes and 37,990 bacterial ASVs in the soil, with 6,889 shared bacterial ASVs. In addition, 2,948 fungal ASVs were identified in the rhizomes and 8,868 in the soil, with 1,893 shared fungal ASVs. Microbial sequencing results indicated that the fungal communities between soil and rhizomes were mainly composed of Ascomycota and Basidiomycota, while bacterial communities included Proteobacteria, Acidobacteria, Bacteroidota, Gammatimonadota, and Firmicutes. Dominant bacterial groups such as Nitrospira, Acidibacter, and fungal groups including Mortierella, Ceratobasidium, and Fusarium were identified as potential contributors to the observed traits. In the top 15 microbial genera, both Sichuan and Guangxi contain 15 bacterial genera, but there are differences in their abundance. Guangxi has three unique fungal genera, including the genera Scleroderma, Russula, and Gliocladiopsis. On the other hand, Sichuan has the unique fungal genus Chamaeota. The correlation analysis between the microbiota and the chemical content from 12 different collecting spots was performed by GraphPad Prism. Burkholderia-Caballeronia-Paraburkholderia, Acidibacter, and Amycolatopsis show an inverse proportionality to total polysaccharides and saponins, while Enterobacter shows a direct proportionality to total polysaccharides and inverse proportionality to saponins. The metabolism pathways show a significant positive correlation with PCH polysaccharides and saponins. This study provide new insights into the mechanisms underlying the quality differences between the two major indigenous areas.

Abstract Glaucoma, the leading cause of irreversible blindness worldwide, is closely linked to aqueous overaccumulation and elevated intraocular pressure (IOP). For refractory glaucoma, aqueous shunts with valves are commonly implanted for effective aqueous drainage control and IOP stabilization. However, existing valved glaucoma implants have the disadvantages of inconsistent valve opening/closing pressures, poor long‐term repeatability due to their reliance on moving parts, and complex architectures and fabrication processes. Here, we propose a novel valving concept, the droplet Laplace valve (DLV), a three‐dimensional printable moving‐parts‐free microvalve with customizable and consistent threshold valving pressures. The DLV uses a flow discretization unit governed by capillarity, comprising a droplet‐forming nozzle, and a separated reservoir to digitize continuous flow into quantifiable droplets. Unlike the classic one‐time‐use Laplace valves, the DLV's unique design allows for its reusability. The opening pressure is adjustable by varying the nozzle size, like the classic Laplace valves (following the Young–Laplace equation), while the closing pressure can be modified by tuning the separation distance and the reservoir size. Various DLVs with customizable opening pressures from 5 to 11 mmHg have been demonstrated, with opening/closing pressure differences suppressed down to 0.99). Preliminary ex vivo validation in an enucleated porcine eye confirms the DLV's efficiency in aqueous shunting and prompt IOP stabilization. The DLV technology holds great promise in glaucoma implants for IOP management and various microsystems for flow control.

Abstract Background and Aim Both intestinal symptoms and comorbidities exist in irritable bowel syndrome (IBS) patients and influence their quality of life (QOL). More research is needed to determine how these variables impact the QOL of IBS patients. This study aimed to determine which specific factors had a higher influence on QOL and to further compare the effects of intestinal symptoms and comorbidities on QOL. Methods IBS patients were recruited from six tertiary hospitals in different regions of China. QOL, gastrointestinal symptoms, and comorbidities were assessed by different scales. Correlation analysis, multiple linear regression, and mediation model were used for statistics. Results Four hundred fifty‐three IBS patients (39.7% women, mean age 45 years) were included and no significant differences in QOL were found across demographic characteristics. Abnormal defecation (r = −0.398), fatigue (r = −0.266), and weakness (r = −0.286) were found to show higher correlation with QOL. More than 40% of IBS patients were found to suffer from varying degrees of anxiety or depression, and anxiety (r = −0.564) and depression (r = −0.411) were significantly negatively correlated with QOL (P

Objective To investigate the resource allocation status of blood testing laboratories in 14 blood stations in Gansu Province, explore the impact of differences in basic conditions on the comprehensive testing ability of laboratories, so as to promote the homogenization and standardization of blood screening capacity in blood stations in Gansu and improve blood safety and effectivenes. Methods An evaluation index system of laboratory resource allocation was constructed and a question-naire was designed. The data of human resources, infrastructure and key equipment of 14 blood stations were collected. The entropy weight -TOPSIS method was used to evaluate and rank the resource allocation of 14 blood stations. Results In the comprehensive evaluation of blood testing laboratory resource allocation in 14 blood stations in Gansu, the top three were laboratories A, B and I, and the last three were laboratories G, M and J. On the whole, the main issue was unreasonable structure of human resources: most laboratories had unreasonable age structure; except for Laboratory A, there was no personnel with bachelor's degree or above in laboratories; most laboratories had not established a team with intermediate professional titles. In terms of infrastructure, the size of seven laboratories could not meet the needs of modern laboratory testing, and all eight blood stations had no spare nucleic acid laboratories nor a mutual spare laboratory with other blood stations As for the key equipment, 5 laboratories had no automatic blood grouping diagnostic instrument, 5 laboratories only had one set of enzyme immunoassay detection system, 3 laboratories had no spare equipment for the key equipment, which means if the equipment failure could not be repaired in time, the release of results would be affected. Conclusion There were significant differences in human resources, infrastructure and key equipment of blood testing laboratories in 14 blood stations in Gansu, which had a great impact on laboratory testing capacity and subsequent development. It is suggested that governments at all levels and health administrative departments optimize the input of laboratory resource allocation according to the blood collection volume of blood stations to gradually narrow the differences in resource distribution between different regions, improve the degree of laboratory automation and optimize the personnel structure, so as to build high-quality and efficient blood testing laboratories and ensure the safety of clinical blood use.

Abstract Ischemic insult stimulates proliferation of neural stem cells (NSCs) in the subventricular zone (SVZ) after stroke. However, only a fraction of NSC‐derived neuroblasts from SVZ migrate toward poststroke brain region. We have previously reported that direct‐current stimulation guides NSC migration toward the cathode in vitro. Accordingly, we set up a new method of transcranial direct‐current stimulation (tDCS), in which the cathodal electrode is placed on the ischemic hemisphere and anodal electrode on the contralateral hemisphere of rats subjected to ischemia–reperfusion injury. We show that the application of this bilateral tDCS (BtDCS) promotes the migration of NSC‐derived neuroblasts from SVZ toward the cathode direction into poststroke striatum. Reversing the position of the electrodes blocks the effect of BtDCS on the migration of neuroblasts from SVZ. BtDCS protects against neuronal death and improves the functional recovery of stroke animals. Thus, the migration of NSC‐derived neuroblasts from SVZ toward poststroke brain region contributes to the effect of BtDCS against ischemia‐induced neuronal death, supporting a potential development of noninvasive BtDCS as an endogenous neurogenesis‐based stroke therapy.

Abstract Graphene, with its two linearly dispersing Dirac points with opposite windings, is the minimal topological nodal configuration in the hexagonal Brillouin zone. Topological semimetals with higher-order nodes beyond the Dirac points have recently attracted considerable interest due to their rich chiral physics and their potential for the design of next-generation integrated devices. Here we report the experimental realization of the topological semimetal with quadratic nodes in a photonic microring lattice. Our structure hosts a robust second-order node at the center of the Brillouin zone and two Dirac points at the Brillouin zone boundary—the second minimal configuration, next to graphene, that satisfies the Nielsen–Ninomiya theorem. The symmetry-protected quadratic nodal point, together with the Dirac points, leads to the coexistence of massive and massless components in a hybrid chiral particle. This gives rise to unique transport properties, which we demonstrate by directly imaging simultaneous Klein and anti-Klein tunnelling in the microring lattice.

ABSTRACT: Objectives: Linezolid-resistant Staphylococcus capitis (LRSC) has become a new challenge for clinical anti-infective therapy. The present study aimed to investigate the trends of LRSC prevalence in a tertiary hospital of China 2017–2020. The resistance mechanisms, virulence genes, biofilm formation, and mass spectrometric characteristics of LRSC isolates were also analysed. Methods: This study retrospectively analysed the antibiotic resistance trends of coagulase negative staphylococci (CoNS) isolated from clinical samples collected between 2017–2020. Antimicrobial resistance profiles were tested by micro-broth dilution and the E-test method. Antimicrobial resistance genes and virulence genes were detected by polymerase chain reaction, and dru-typing sequences were obtained by Sanger sequencing. Crystal violet staining in 96-well plates was used to detect biofilm formation ability. Mass spectrometric characterization of LRSC was analysed by matrix-assisted laser desorption ionization-time of flight mass spectrometry (MALDI-TOF MS) coupled with ClinProTools. Results: The linezolid resistance rate in 3575 CoNS clinical strains was 1.6%, wherein the great majority of was LRSC (91.1%, n = 51/56), with a resistant rate of 15.5% (n = 51/328) in all S. capitis isolates. In this study, 48 out of the 51 LRSC strains and 54 of 277 linezolid-susceptible S. capitis (LSSC) strains were enrolled. G2576T, C2104T, T2130A, C2163T, and T2319C mutations in the 23S rRNA V region and acquisition of cfr were the main linezolid resistant mechanisms in LRSC. The biofilm-forming ability of LRSC was more potent than LSSC, with a higher detection rate of bap (P < 0.05). Eleven mass spectrometric peaks of interest were identified by using MALDI-TOF MS and ClinProTools, which were differently distributed between LRSC and LSSC strains, with the area under the receiver operating characteristic curve of more than 0.8, especially for 5465.37 m/z. Conclusions: Linezolid resistance was mediated by mutations in the 23S rRNA V region and presence of the cfr gene in LRSC strains. LRSC strains have stronger biofilm-forming ability than LSSC strains, which maybe associated with the adhesion-related gene of bap. Further, linezolid-resistant and linezolid-susceptible S. capitis could be rapidly identified with mass spectrometric characterization. To the best of our knowledge, this study is the first to document the biofilm formation ability of LRSC and the potential usefulness of MALDI-TOF MS for the discrimination of LRSC and LSSC.

Millettia speciosa Champ. (MSCP), which is distributed and used in Southern China, is often sulfur-fumigated during postharvest handling to prevent insects and molds. In the present study, a comprehensive strategy was proposed by using ultrahigh-performance liquid chromatography-quadrupole/time-of-flight mass spectrometry (UHPLC–QTOF–MS/MS) with multivariate analysis to rapidly identify potential chemical sulfur-fumigated MSCP (SF-MSCP) samples, discover chemical transformation mechanisms and control the quality of SF-MSCP. Eighty-six compounds were tentatively assigned, and ten compounds were screened out as potential markers. Moreover, the chemical transformation mechanism was deduced by hydrolysis, esterification and dehydration reactions that occurred during the sulfur-fumigated process. The developed method was successfully applied to predict 16 commercial MSCP samples, and 9 samples were found to contain sulfur compounds, ranging in sulfur residues from 100 to 310 mg/kg. These research outcomes can not only be used to distinguish the nonfumigated MSCP from SF-MSCP by chemical markers but also provide a chemical basis for further comprehensive investigating in-depth safety and functional evaluations of SF-MSCP.

Abstract Background Real-time quantitative PCR (RT-qPCR) is a crucial and widely used method for gene expression analysis. Selecting suitable reference genes is extremely important for the accuracy of RT-qPCR results. Commonly used reference genes are not always stable in various organisms or under different environmental conditions. With the increasing application of high-throughput sequencing, transcriptome analysis has become an effective method for identifying novel stable reference genes. Results In this study, we identified candidate reference genes based on transcriptome data covering embryos and larvae of early development, normal adult tissues, and the hindgut under sulfide stress using the coefficient of variation (CV) method in the echiuran Urechis unicinctus, resulting in 6834 (15.82%), 7110 (16.85%) and 13880 (35.87%) candidate reference genes, respectively. Gene Ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) enrichment analyses revealed that the candidate reference genes were significantly enriched in cellular metabolic process, protein metabolic process and ribosome in early development and normal adult tissues as well as in cellular localization and endocytosis in the hindgut under sulfide stress. Subsequently, ten genes including five new candidate reference genes and five commonly used reference genes, were validated by RT-qPCR. The expression stability of the ten genes was analyzed using four methods (geNorm, NormFinder, BestKeeper, and ∆Ct). The comprehensive results indicated that the new candidate reference genes were more stable than most commonly used reference genes. The commonly used ACTB was the most unstable gene. The candidate reference genes STX12, EHMT1, and LYAG were the most stable genes in early development, normal adult tissues, and hindgut under sulfide stress, respectively. The log2(TPM) of the transcriptome data was significantly negatively correlated with the Ct values of RT-qPCR (Ct = − 0.5405 log2(TPM) + 34.51), which made it possible to estimate the Ct value before RT-qPCR using transcriptome data. Conclusion Our study is the first to select reference genes for RT-qPCR from transcriptome data in Echiura and provides important information for future gene expression studies in U. unicinctus.

Strengthening the real-time and accurate identification of civil structures is of great significance for ensuring the safety and service life of civil engineering projects. Therefore, in order to reduce the incidence of civil structural accidents in buildings and improve the safety, availability, and integrity of building structures, the study plans to adopt deep learning methods. In this study, the parallel Convolutional neural network covering one-dimensional and two-dimensional features is combined with the Benchmark numerical model to identify structural damage. This network structure can effectively utilize two parallel branches to extract response features at different scales and time domains, ensuring the coverage of damage feature recognition content to a certain extent. And the Benchmark numerical model can effectively improve the visualization of identification in the simulation of civil structures. By testing the fusion algorithm model, the results show that the network structure can effectively extract damage signal features, and its minimum classification loss value can approach 0.01; The maximum damage indicators on connecting beams, frame beams, and shear walls reached 0.472, 0.117, and 0.055, far higher than other comparative algorithms. The fusion algorithm has a recognition accuracy of over 85% for structural joint damage, showing good damage recognition performance. This fusion algorithm can effectively provide reference value and significance for the development of structural inspection and related risk prevention plans in civil engineering projects, and also provide new ideas and possibilities for relevant researchers to study the field of civil engineering.

To rapidly evaluate the quality of complex herbal preparations, a new strategy was proposed based on multi-color scale and efficacy-oriented high-performance thin-layer chromatography (HPTLC) characteristic fingerprint combined with chemometric method. Firstly, effective components were screened through high-performance liquid chromatography with ultraviolet detection and evaporative light-scattering (HPLC-UV-ELSD), using multi-wavelength fusion combined with network pharmacology and molecular docking techniques. Subsequently, guided by the effective components, the targeted HPTLC characteristic fingerprint was established by multi-color scale scanning. Finally, combined with the chemometric method, the consistency of the preparation quality was evaluated, the marker components leading to quality differences were screened, and the quality control limit was established. Sanwujiao Pills (SWJPs) is a herbal preparation composed of six herbs for treating rheumatoid arthritis (RA). Through this strategy, four HPTLC characteristic fingerprints were established, they were derived from five herbs and guided by eight effective components in SWJPs. Through similarity, clustering heatmap, principal component analysis (PCA), and orthogonal partial least squares discriminant analysis (OPLS-DA), the quality distinctions among the 12 batches of SWJPs were determined. These batches were categorized into two groups based on their production time, and eight components affecting the quality of the preparation were identified. Meanwhile, the quality control threshold for SWJPs was determined based on Hotelling's T2 and DModX methods. This strategy aims to rapidly evaluate the quality of complex herbal preparations by HPTLC and extends the application of HPTLC fingerprint chromatography for identifying herbal medicine species and activity-related quality detection. The proposed strategy is also helpful for the quality control of other complex herbal preparations.

Summary: Background: Mediastinal neoplasms are typical thoracic diseases with increasing incidence in the general global population and can lead to poor prognosis. In clinical practice, the mediastinum's complex anatomic structures and intertype confusion among different mediastinal neoplasm pathologies severely hinder accurate diagnosis. To solve these difficulties, we organised a multicentre national collaboration on the basis of privacy-secured federated learning and developed CAIMEN, an efficient chest CT-based artificial intelligence (AI) mediastinal neoplasm diagnosis system. Methods: In this multicentre cohort study, 7825 mediastinal neoplasm cases and 796 normal controls were collected from 24 centres in China to develop CAIMEN. We further enhanced CAIMEN with several novel algorithms in a multiview, knowledge-transferred, multilevel decision-making pattern. CAIMEN was tested by internal (929 cases at 15 centres), external (1216 cases at five centres and a real-world cohort of 11 162 cases), and human–AI (60 positive cases from four centres and radiologists from 15 institutions) test sets to evaluate its detection, segmentation, and classification performance. Findings: In the external test experiments, the area under the receiver operating characteristic curve for detecting mediastinal neoplasms of CAIMEN was 0·973 (95% CI 0·969–0·977). In the real-world cohort, CAIMEN detected 13 false-negative cases confirmed by radiologists. The dice score for segmenting mediastinal neoplasms of CAIMEN was 0·765 (0·738–0·792). The mediastinal neoplasm classification top-1 and top-3 accuracy of CAIMEN were 0·523 (0·497–0·554) and 0·799 (0·778–0·822), respectively. In the human–AI test experiments, CAIMEN outperformed clinicians with top-1 and top-3 accuracy of 0·500 (0·383–0·633) and 0·800 (0·700–0·900), respectively. Meanwhile, with assistance from the computer aided diagnosis software based on CAIMEN, the 46 clinicians improved their average top-1 accuracy by 19·1% (0·345–0·411) and top-3 accuracy by 13·0% (0·545–0·616). Interpretation: For mediastinal neoplasms, CAIMEN can produce high diagnostic accuracy and assist the diagnosis of human experts, showing its potential for clinical practice. Objective To investigate the hip joint forces, Von Mises stress, contact pressure and micro‐motion of hip prosthesis for developmental dysplasia of the hip (DDH) patients under different hip joint centers using musculoskeletal (MSK) multi‐body dynamics and finite element analysis. Methods Both MSK multi‐body dynamics model and finite element (FE) model were based on CT data of a young female DDH patient with total hip replacement and were developed to study the biomechanics of the S‐ROM hip prosthesis. The same offset of hip joint center along all six orientations compared with the standard position was set to predict its effects on both MSK multi‐body dynamics and contact mechanics during one gait cycle. Results The hip joint forces in the entire walking gait cycle showed two peak values and clear differences between them under different hip joint centers. The hip joint force increased when the hip joint center moved posteriorly (2101 N) and laterally (1969 N) to the anatomical center (1848 N) at the first peak by 13.7% and 6.6%, respectively. The hip joint force increased sharply when the hip center deviated laterally (2115 N) and anteriorly (2407 N), respectively, from the standard position (1742 N) at the second peak. For the sleeve of the S‐ROM prosthesis, the maximum Von Mises stress and contact pressure of the sleeve increased if the hip joint center deviated from the anatomical center posteriorly at the first peak. However, the Von Mises stresses and contact pressure increased at anterior and lateral orientations, compared to that of the standard position at the second peak. Small changes were observed for the maximum relative sliding distance along most of the orientations at both peaks except in the lateral and medial orientations, in which an increase of 8.6% and a decrease of 13.6% were observed, respectively. Conclusion The hip joint center obviously influenced the hip joint forces, stress, contact pressure and micro‐motion of the hip implant for this female patient.

In order to solve the problem of difficult coordination of traditional overcurrent relay protection caused by short supply radius and little difference of fault current along urban distribution network, a coordinated relay protection strategy based on economical fault current limiter (FCL) is proposed. The FCL device is configured at the export of the main transformer, by using quick fault identification method, setting different protection current value and relay protection delay time, once the fault occurs, the FCL can quickly limit the fault current first then cooperate with the circuit break and switches on the feeder line to achieved the coordinated relay protection. Combining with faults occurring at different locations along the feeder line, the composition and basic working principle of the FCL are discussed, the theory of fast fault identification method and the setting method of the coordinated relay protection parameters are emphatically discussed. The simulation results show that the proposed current limiting coordinated relay protection strategy can effectively reduce the short-circuit current level and bus voltage drop during the fault handle processing, thus reduce the damage to the distribution system and influence range of the fault, greatly improve the selectivity of the relay protection finally.

This paper proposes a novel control approach for fault-tolerant control of dual three-phase permanent magnet synchronous motor (PMSM) under one-phase open-circuit fault. A modified six-phase static coordinate transformation matrix and an extended rotating coordinate transformation matrix are investigated considering the influence of the fifth harmonic space on fault-tolerant control. These mathematical models are further analyzed in the fundamental space and the fifth harmonic space after the fault and to eliminate the coupling between the d-q axis voltage equation in the fundamental wave space and the d-q axis voltage equation in the fifth harmonic space, a secondary rotation coordinate transformation matrix is proposed. To achieve the purpose of reducing torque ripple, the fault-tolerant control method proposed in this paper not only takes the minimum copper loss as the constraint condition, but also injects the fifth harmonic current. The experimental result of current and torque is used to verify the accuracy of fault-tolerant control.

In this study, a fingerprint-activity relationship between ultrahigh performance liquid chromatography (UHPLC) fingerprints and antioxidant activity was established to evaluate the quality of Aster yunnanensis Franch.(AYF) from different collecting spots. First, the fingerprint of AYF was established by UHPLC, and the similarity analysis was analyzed based on twenty-one common peaks. Then the chemical constituents from AYF were analyzed and identified by UHPLC-quadrupole time-of-flight tandem mass spectrometry (QTOF-MS/MS). Next, the antioxidant activity of twelve batches of AYF was assessed in vitro. Subsequently, eleven chemical markers were screened out by fingerprints and antioxidant activity utilizing grey relational analysis (GRA) and partial least squares (PLS). Finally, the contents of eleven chemical markers in twelve batches of AYF were detected by UHPLC, and the antioxidant quality of AYF was evaluated using chemometric analysis, such as principal components analysis (PCA) and technique for order preference by similarity to ideal solution (TOPSIS). The results showed that the antioxidant efficacy was associated with the total content of eleven compounds of AYF. Moreover, this method to discover quality markers was reasonable by fingerprint-activity relationship combination with chemometric analysis. The present study will certify quality markers associated with therapeutic effects, and provide a powerful strategy for evaluating the resource quality of AYF.

ABSTRACT: Objectives: Identification and classification of microorganisms is one of the most important but difficult and challenging issues in microbiology. Whole genome sequencing (WGS), which can give a thorough understanding for the genome of bacteria strain, has been universally used for studying bacterial classification, evolution, and drug-related resistant genes. We in this study aimed to identify a Gram-positive, microaerophilic, catalase-negative cocci strain named AV208, which has shown resistance to vancomycin, by whole genome's average nucleotide identity (ANI) and high-throughput sequencing technology. Methods: The AV208 strain was identified by following commercially available identification systems, including API 20 Strep system and Vitek 2 Compact Gram-positive identification system for biochemical phenotypic test. Matrix-assisted laser desorption ionization-time of flight mass spectrometry (MALDI-TOF-MS) and 16S rRNA gene sequencing were used for confirmation identification. The whole genome of AV208 was sequenced by using high throughput sequencing technology and ANI between AV208, and its phylogenetic neighbours were analysed by the Orthologous Average Nucleotide Identity Tool (OAT) software. Polymerase chain reaction (PCR) and DNA sequencing were used to investigate the potential molecular mechanism for vancomycin resistance. Results: The AV208 strain was isolated from an ascites sample from a patient with chronic kidney disease who showed extensive resistance to the drugs detected, such as vancomycin with MIC >256 μg/mL. With combination of biochemical phenotypic test, MALDI-TOF-MS and 16S rRNA gene sequencing, the AV208 strain was tentatively identified as an Aercoccus viridans. By using complete genome sequence, we found a 96.24% ANI between strain AV208 and Aerococcus urinaeequi CCUG 28094T, which was higher than that with A. viridans CCUG4311T (94.9%). The consistency of 16S rRNA sequence of strain AV208 was 100% with A. urinaeequi CCUG 28094T and 99.9% with A. viridans CCUG4311T, with only one base difference between them. PCR and sequencing for van genes revealed that AV208 was positive for the vanA gene. A Tn1546 transposon-like structure with vanA gene was found in the genome, which was predicted locating in plasmid, causing vancomycin resistance phenotypes. Conclusion: Average nucleotide identity analysis based on whole genome sequence is an accurate and effective method for identification of bacteria, especially for strains that are not discernible by existing methods such as Aerococcus.

Objective To analyze the comprehensive factors causing adverse reactions to apheresis platelet donation(ARAPD), so as to provide references for effective prevention of ARAPD. Methods The 272 cases of ARAPD from 2012 to 2019 in Lanzhou were statistically analyzed, and factors that induced ARAPD were studied. Statistical analysis were performed according to the gender, nationality, occupation, age, weight, donation units, and number of donations. Results As to the factors inducing ARAPD, anticoagulant reactions accounted for the first(32.4%, 88/272). Women and students were prone to develop ARAPD. Millettia speciosa Champ. (MSCP) enjoys widespread recognition for its culinary and medicinal attributes. Despite the extensive history of MSCP cultivation, the disparities in quality and bioactivity between wild and cultivated varieties have remained unexplored. In this study, 20 wild and cultivated MSCP samples were collected from different regions in China. We embarked on a comprehensive investigation of the chemical constituents found in both wild and cultivated MSCP utilizing UHPLC-Q-Exactive Orbitrap-MS technology and multivariate analysis such as principal component analysis (PCA) and orthogonal partial least squares discriminant analysis (OPLS-DA). In total, 62 chemical components were unequivocally identified or tentatively characterized. Via the multivariate statistical analysis, we successfully pinpointed nine compounds with the potential to serve as chemical markers, enabling the differentiation between wild and cultivated MSCP varieties. Moreover, both genotypes exhibited substantial antioxidant and anti-fatigue properties. The bioactivities of wild MSCP were marginally higher when compared to their cultivated counterparts. This study illuminates the impressive antioxidant and anti-fatigue potential present in both wild and cultivated MSCP genotypes, further augmenting the allure of this species and opening new avenues for the economic valorization of MSCP. Hence, this study provides a valuable method for the identification and quality control of MSCP and a method in chemistry and pharmacology to assess an alternative possibility for cultivated MSCP.

Rapid non-destructive testing technologies are effectively used to analyze and evaluate the linoleic acid content while processing fresh meat products. In current study, hyperspectral imaging (HSI) technology was combined with deep learning optimization algorithm to model and analyze the linoleic acid content in 252 mixed red meat samples. A comparative study was conducted by experimenting mixed sample data preprocessing methods and feature wavelength extraction methods depending on the distribution of linoleic acid content. Initially, convolutional neural network Bi-directional long short-term memory (CNN-Bi-LSTM) model was constructed to reduce the loss of the fully connected layer extracted feature information and optimize the prediction effect. In addition, the prediction process of overfitting phenomenon in the CNN-Bi-LSTM model was also targeted. The Bayesian-CNN-Bi-LSTM (Bayes-CNN-Bi-LSTM) model was proposed to improve the linoleic acid prediction in red meat through iterative optimization of Gaussian process acceleration function. Results showed that best preprocessing effect was achieved by using the detrending algorithm, while 11 feature wavelengths extracted by variable combination population analysis (VCPA) method effectively contained characteristic group information of linoleic acid. The Bi-directional LSTM (Bi-LSTM) model combined with the feature extraction data set of VCPA method predicted 0.860 Rp2 value of linoleic acid content in red meat. The CNN-Bi-LSTM model achieved an Rp2 of 0.889, and the optimized Bayes-CNN-Bi-LSTM model was constructed to achieve the best prediction with an Rp2 of 0.909. This study provided a reference for the rapid synchronous detection of mixed sample indicators, and a theoretical basis for the development of hyperspectral on-line detection equipment.

Abstract Background In the geriatric patient population, frailty significantly affects a patient’s prognostic outcome. This study aimed to compare the consistency of our constructed geriatric trauma frailty index with previously published indexes. Methods The geriatric trauma frailty index (GTFI) was compared with four previously published frailty indexes, i.e., the hospital frailty risk score (HFRS), Fried index, trauma-specific frailty index (TSFI), and 11-item modified frailty index (mFI) using the Bland-Altman method, intraclass correlation coefficient (ICC), and kappa consistency test. The indexes were calculated based on data collected from 101 questionnaires and medical records from 101 geriatric trauma patients at a tertiary hospital in Shanghai. Results Among the 101 geriatric trauma patients, 64 (63.4%) were women, with a mean age of 71.18 (SD = 9.89) years and mean length of stay (LOS) of 7.51 (SD = 3.89) days. The mean scores of GTFI score(≥ 1.3045 as frail), Fried index score(≥3 items as frail), TSFI score(≥ 4 as frail), and mFI (≥ 3 as frail),were 0.86 (SD = 1.51), 0.76 (SD = 1.07), 1.76 (SD = 1.96), and 1.29 (SD = 1.17). respectively. The GTFI score had good consistency with the HFRS (ICC: 0.716, 95% confidence interval [CI]: 0.596, 0.799, kappa: 0.608, 95% CI: 0.449, 0.766), fair consistency with the TSFI (ICC: 0.407, 95% CI: 0.227, 0.562, kappa: 0.460, 95% CI: 0.239, 0.672), and poor consistency with the mFI (ICC: 0.286, 95% CI: 0.097, 0.455, kappa: 0.305, 95% CI: 0.069, 0.525) and Fried index score (ICC: 0.256, 95% CI: 0.063, 0.426, kappa: 0.188, 95% CI: − 0.028, 0.408). Conclusions Different frailty indexes are based on different concepts of frailty and cannot be assumed to be interchangeable. There is still no gold standard for the current assessment methods of frailty, but it can be compared based on the understanding in terms of the concepts and measures used in each.

Objective To study the current situation of apheresis platelets collection in various regions of Gansu province by comparing and analyzing relevant data from blood stations in 14 prefecture-level cities of Gansu province. Methods The units of collected platelets and rate of double-dose collection in 13 regional blood stations and 1 provincial blood center from 2016 to 2020, as well as the clinical supply and demand was statistically analyzed. Results From 2016 to 2020, the total units of platelets collected by 13 blood stations and 1 blood center in Gansu increased from 11 255 U to 15 270 U, with the increase rate at 35.7% in 5 years, and mainly were collected by the provincial blood center (74.57%, 50 253/ 67 392). Although the rate of double-dose collection in the province showed a steady upward trend, only 3 blood stations realized annual double-dose collection more than 20%. There was still a gap of about 10% between supply and clinical needs. Conclusion Although the number of platelet collections and units in each blood station in Gansu is on the rise in general, the units collected varies in each blood station. Therefore, further measures need to be taken from the aspects of publicity, recruitment, optimizing the collection process, improving the rate of double-dose collection, retention of regular blood donors and regional coordination to increase the collection units, narrow down the regional gap and ensure the balance between supply and demand.

BackgroundDespite years of public cardiopulmonary resuscitation (CPR) training efforts, the training rate and survival following out-of-hospital cardiac arrest (OHCA) have increased modestly in China. Access is imperative to increase the public CPR training rate, which is determined by both demand- (e.g., the lay public) and supply-side (e.g., CPR trainers) factors. We aimed to explore the demand and supply determinants of access to CPR training for the lay public in China.MethodsQualitative semi-structured interviews were conducted with 77 laypeople (demand side) and eight key stakeholders from CPR training institutions (supply side) in Shanghai, China. The interview guide was informed by Levesque et al. healthcare access framework. Data were transcribed, quantified, described, and analyzed through thematic content analysis.ResultsOn the demand side, the laypeople's ability to perceive their need and willingness for CPR training was strong. However, they failed to access CPR training mainly due to the lack of information on where to get trained. Overestimation of skills, optimism bias, and misconceptions impeded laypeople from attending training. On the supply side, trainers were able to meet the needs of the trainees with existing resources, but they relied on participants who actively sought out and registered for training and lacked an understanding of the needs of the public for marketing and encouraging participation in the training.ConclusionInsufficient information and lack of initiative on the demand side, lack of motivation, and understanding of public needs on the supply side all contributed to the persistently low CPR training rate in China. Suppliers should integrate resources, take the initiative to increase the CPR training rate, innovate training modes, expand correct publicity, and establish whole-process management of training programs.

Environment friendly tin perovskite solar cells (TPSCs) have become a promising candidate due to their low toxicity and similar electronic configuration to lead counterparts. However, the easily generated defects due to the Sn2+oxidation and poor morphology originated from rapid crystallization are both barriers for tin-based devices in the enhancement of photovoltaic performance. Here, phenethylammonium chloride (PEACl) is introduced for post-treating FASnI3 films, which plays dual roles in adjusting film topography and passivating defects in bulk and interface of films. First, the film morphology can be improved by eliminating SnF2 impurities. In addition, a small amount of two-dimension perovskite can be formed in the films, inducing preferred orientation in secondary crystallization, well-matched energy level, and effective defect passivation. Finally, the FASnI3-based devices with PEACl treatment obtained a champion conversion efficiency of 11.63% with an average value of 10.76%, along with the obvious enhanced open circuit voltage and fill factor. In contrast, the pristine FASnI3 devices range from 6.92% to 9.40% with an average value of 7.96%. This work not only deepens our understanding of the suppression of defects in tin-based perovskites but also paves a facile way to ameliorate film morphology and interfaces to fabricate promising tin perovskites and devices.

Total flavonoids from Smilax glabra (TFSG) exhibit several biological activities; however, their poor stability limits their application. In this work, zein–lecithin–TFSG complex nanoparticles (Z–L–TFSG NPs) were prepared using the anti-solvent coprecipitation technique. The prepared Z–L–TFSG NPs were spherical with an encapsulation efficiency of 98.0%. Differential scanning calorimetry, Fourier transform infrared spectroscopy, and morphology tests revealed that the TFSG were successfully encapsulated by Z–L NPs. Z–L–TFSG NPs showed superior stability and better controlled release characteristics in simulated gastrointestinal digestion. The encapsulation of TFSG by Z–L NPs could improve their antioxidant capacity in vitro. Moreover, Z–L–TFSG NPs could enhance the protective effects of TFSG against H2O2-induced oxidative damage to HepG2 cells. The results indicated that the Z–L self-assembled NPs could serve as a promising drug delivery system through the integrated encapsulation of multiple flavonoids.

In this study, the chloroplast genomes of Acanthocalyx alba (Hand.-Mazz., 1925) and Acanthocalyx nepalensis subsp. delavayi (Franchet, 1885) were sequenced, and their total lengths were 148,720 bp and 149,253 bp, respectively. The A. alba genome contained two inverted repeat regions (IRs) of 21,849 bp, a large single-copy region (LSC) of 89,084 bp, and a small single-copy region (SSC) of 15,938 bp, whereas A. nepalensis subsp. delavayi contained two IRs of 21,736 bp, one LSC of 89,034 bp, and one SSC of 16,747 bp. The chloroplast genomes of both A. alba and A. nepalensis subsp. delavayi contained 109 genes, including 72 mRNA, 33 tRNA, and four rRNA genes. Phylogenetic analysis suggested that A. alba is in a clade with A. nepalensis subsp. delavayi. This study provides useful data for further phylogenetic studies of A. alba and A. nepalensis subsp. delavayi.

Loess subsurface-erosion refers to the erosion, transport and subsidence of loess caused by surface water infiltration along the dominant seepage channels such as loess macropore joints and fissures. Due to the characteristics of water sensitivity, structural surface development, dominant seepage, etc., loess subsurface-erosion has developed extensively in the loess area, and loess subsurface-erosion has a significant disaster effect, making the mechanism and prevention of loess disasters induced by erosion more complicated. This article first combed the basic concepts of loess subsurface-erosion, based on summarizing the main controlling factors of loess subsurface-erosion, expounded the formation mechanism of loess subsurface-erosion, and expanded the basic connotation of loess subsurface-erosion disaster effect. Finally, according to the current research status of loess submergence, a brief prospect is made for the future research of loess subsurface-erosion, and it is pointed out that more attention should be paid to the research on the key linking role of loess subsurface erosion in the loess disaster chain. This article focuses on the preliminary thinking of several problems in the study of loess subsurface-erosion, and aims to provide scientific suggestions for the subsequent study of loess subsurface-erosion hazard effects.

Mollusca is the second largest animal phylum and represents one of the most evolutionarily successful animal groups. Mulinia lateralis, a small bivalve, is a promising model organism to facilitate studies of mollusc development. However, because of the lack of published genomic and transcriptomic resources, integrated research on the formation of larval shells in this species, which is a representative developmental process of molluscs and of great importance for larva survival, is hindered. In this study, the blastula, gastrula, trochophore larva, and D-shaped larva of M. lateralis were utilized for generating a comprehensive full-length transcriptome through Pacific BioSciences (PacBio) isoform sequencing (Iso-seq) and Illumina RNA-Seq. A total of 238,919 full-length transcripts with an average length of 3,267 bp and 121,424 annotated genes were obtained. Illumina RNA-Seq data analysis showed that 4,512, 10,637, and 17,829 differentially expressed genes (DEGs) were obtained between the two adjacent developmental stages. Functional annotation and enrichment analysis revealed the specific function of genes in shell biomineralization during different developmental stages. Twelve genes that may be involved in the formation of the larval shell of M. lateralis were identified, including insoluble shell matrix protein-encoding gene 1 (ISMP1), ISMP2, ISMP5, chitin synthase, tyrosinase, chitin-binding protein, collagen and pu14 involved in shell matrix deposition, and carbonic anhydrase, solute carrier family 4 member 8 (slc4a8), EF-hand, and a calmodulin coding gene C-2442 participated in ion transportation. In addition, calcium ion binding function, calcium signaling pathway, and endocrine and other factor-regulated calcium reabsorption pathways were significantly enriched. Weighted gene correlation network analysis (WGCNA) identified two modules related to biomineralization and larval shell formation, and slc4a8 and ring finger protein 41 (rnf41) were key hub genes that may be involved in this process. Moreover, it could be implied that the process of ion transport occurs earlier than the deposition of the shell matrix. This work provided a clear view of the transcriptome for M. lateralis and will be valuable in elucidating the mechanisms of larval shell formation as well as other developmental processes in molluscs.

As for the tight sandstones, pore-scale fluid flow ability is one of the most fundamental parameters that determine potential development capacity when compared to conventional reservoirs due to relatively tiny spaces. The determination of pore-throat structures and their impact on fluid flow behavior could improve the forecast ability for development. Therefore, the main purpose of this paper is to study the effects of differences in microscopic pore structures on moveable fluid saturation in tight sandstone reservoirs. Casting thin sections (CTS), scanning electron microscope (SEM), constant-rate mercury injection (CRMI), nuclear magnetic resonance (NMR), etc. are used as experimental methods to qualitatively describe the differences in accumulation space types and to quantitatively compare and analyze the differences in microscopic pore structures between reservoirs for Chang 6 formation in the Jiyuan and Heshui areas. The different characteristic parameters are chosen to analyze their correlations with moveable fluid saturation. Thus, as a result, due to good correlations with moveable fluid saturation, parameters such as throat radius, pore-throat radius ratio, and pore-throat amount allocation ratio are chosen to characterize the differences in microscopic pore structures between reservoirs in those areas. Differences in throat radius, pore-throat radius ratio, and a pore-throat amount allocation ratio of microscopic pore structure of Chang 6 members in these two areas have led to differences in moveable fluid saturation to some extent. This research provided the evidence about the coupling law between pore-scale fluid flow capacity and pore-throat configuration in tight sandstone reservoirs, and the results can provide theoretical guidance for field development of oil and gas fields.
