Abstract Background There is a lack of research regarding the relationship between creatinine to cystatin C to waist circumference ratio (CCR/WC ratios) and the development of type 2 diabetes mellitus (T2DM). We aimed to evaluate the association between CCR/WC ratios and incident T2DM in Chinese adults. Methods This prospective study was from the China Health and Retirement Longitudinal Study (2011, 2013, 2015, and 2018). The participants were divided into three groups by tertiaries of the CCR/WC ratios. Cox proportional‐hazards models were used to identify the relationship between CCR/WC and T2DM. Results Overall, 5938 participants were included for analysis, 766 of whom developed T2DM between 2011 and 2018. Risk of incident T2DM was decreased with tertiaries 2, 3 versus tertiary 1 of the CCR/WC index (adjusted hazard ratio [HR] 0.772 [95% confidence interval 0.647–0.921] and 0.724 [0.596–0.880], p for trend = .001 across tertiaries of the CCR/WC index). The results were consistent excluding participants with T2DM in the first 2 years. Conclusions This study demonstrated that CCR/WC was negatively correlated with the risk of T2DM in Chinese adults. Early detection is necessary to control the development of T2DM in Chinese with low CCR/WC levels.

Abstract Background and Aims Previous studies have shown that sarcopenic obesity (SO) was associated with nonalcoholic fatty liver disease (NAFLD). However, research is limited in the context of the NAFLD renamed as metabolic dysfunction‐associated steatotic liver disease (MASLD) defined by updated diagnostic criteria. The aim of this study was to use the index skeletal muscle mass to visceral fat area ratio (SVR) to describe SO in a large and representative US population (National Health and Nutrition Examination Survey 2017–2018) of adults and investigate their association with MASLD. Methods A total of 2087 individuals were included in the analysis. SVR was calculated according to the measurement of dual‐energy x‐ray absorptiometry and MASLD was diagnosed with controlled attenuation parameter scores and cardiometabolic risk factors. SVR was divided into tertiles. Logistic regression adjusted for confounders was used to evaluate the association between SVR and MASLD. Several sensitivity analyses were performed to test the robustness of our findings. Results In a multivariate logistic regression analysis, a significant association between SVR and MASLD was shown (odds ratio [OR]: 3.11, 95% confidence interval [CI]: 1.31–7.39, p = .010 for middle levels of SVR; OR: 3.82, 95% CI: 1.45–10.08, p = .007 for lowest levels of SVR). The sensitivity analyses confirmed that the association was robust. Conclusion Our findings imply that decreased SVR is linked to MASLD.

Crohn’s disease (CD) is a nonspecific inflammatory disease of digestive tract with limited known biomarkers. We aimed to identify potential molecules involved in CD severity. Weighted gene co-expression network and differentially expressed gene analyses were used to explore hub genes associated with inflammation and involved in pathological processes of CD in the GSE83448 cohort. The expression and relationship of genes with clinical characteristics in normal and CD intestinal mucosa were validated in GSE208303 and our research cohorts. Five genes related to disease and inflammation were identified in the clinically significant modules that were differentially expressed in healthy controls and CD patients. Among them, solute carrier family 39 member 5 (SLC39A5) expression was reduced in CD tissues compared with that in normal tissues; low SLC39A5 expression was associated with CD-simplifying endoscopic score and stenosis. Furthermore, we broadened the bioinformatic threshold with qRT-PCR verification and western blot, and found that the expressions of cytochrome P450 family 2 subfamily S member 1 (CYP2S1), fatty acid 2-hydroxylase (FA2H), fatty acid binding protein 1 (FABP1), and Rho family GTPase 3 (RND3) were reduced in CD tissues. Therefore, SLC39A5, CYP2S1, FA2H, FABP1, and RND3 may be novel biomarkers for CD and associated with severity. Highlights SLC39A5 expression was reduced in CD tissues. Low SLC39A5 expression was associated with CD-simplifying endoscopic score and stenosis. CYP2S1, FA2H, FABP1, and RND3 were reduced in CD tissues.

BackgroundPrevious studies have highlighted changes in the cerebral cortical structure and cognitive function among nonalcoholic fatty liver disease (NAFLD) patients. However, the impact of NAFLD on cerebral cortical structure and specific affected brain regions remains unclear. Therefore, we aimed to explore the potential causal relationship between NAFLD and cerebral cortical structure.MethodsWe conducted a Mendelian randomization (MR) study using genetic predictors of alanine aminotransferase (ALT), NAFLD, and percent liver fat (PLF) and combined them with genome-wide association study (GWAS) summary statistics from the ENIGMA Consortium. Several methods were used to assess the effect of NAFLD on full cortex and specific brain regions, along with sensitivity analyses.ResultsAt the global level, PLF nominally decreased SA of full cortex; at the functional level, ALT presented a nominal association with reduced SA of parahippocampal gyrus, TH of pars opercularis, TH of pars orbitalis, and TH of pericalcarine cortex. Besides, NAFLD presented a nominal association with reduced SA of parahippocampal gyrus, TH of pars opercularis, TH of pars triangularis and TH of pericalcarine cortex, but increased TH of entorhinal cortex, lateral orbitofrontal cortex and temporal pole. Furthermore, PLF presented a nominal association with reduced SA of parahippocampal gyrus, TH of pars opercularis, TH of cuneus and lingual gyrus, but increased TH of entorhinal cortex.ConclusionNAFLD is suggestively associated with atrophy in specific functional regions of the human brain.
