Grass carp was an important economic fish for freshwater aquaculture. The exploring and utilization of high-quality grass carp germplasm resources were important for ensuring a domestic supply of high-quality aquatic protein. However, natural grass carp populations in China were facing severe threats because of the effects of frequent human activities, water environment damage, overfishing, etc. Against this background, high-quality grass carp germplasm resources were the foundation for the healthy and sustainable development of the grass carp aquaculture industry, so systematic collection, preservation, identification, evaluation, protection, and utilization of them carry great strategic importance. This paper summarizes major research results of grass carp in morphology, cytogenetics, molecular population genetics, etc. It not only introduced genetic improvement techniques such as gynogenesis, hybrid breeding, and polyploid breeding, but also discussed the existing research on the protection measures of grass carp germplasm resources. On this basis, the paper proposed new methods to improve the protection of grass carp germplasm and create improved varieties, which could provide high-quality resources for the sustainable development of the grass carp industry.

Abstract Anisotropic optical crystals can exhibit a hyperbolic response within the Reststrahlen band (RB) and support directional polaritonic propagations when interacting with light. Most of the reported low-symmetry optical crystals showcase the evolution from hyperbolic to elliptic dispersion topologies, largely owing to their adjacent RBs being either overlapped or separated. Here, we report an exceptional Reststrahlen point (ERP) in rare-earth oxyorthosilicate Y2SiO5, at which two neighboring RBs almost kiss each other. Consequently, we observe the direct hyperbolic-to-hyperbolic topological transition: the hyperbolic branches close and reopen along with the rotating transverse axis (TA). At such ERP, the TA merges to the direction orthogonal to its proximate phonon mode, mainly due to the interplay between these two non-orthogonal phonon modes. We also find that even with the existence of only one single RB, the TA can rotate in-plane. Our findings are prevalent in isostructural rare-earth oxyorthosilicates, such as Lu2SiO5. The universally underlying physics of ERP and its corresponding special class of rare-earth oxyorthosilicates may offer playgrounds for continuously tuning phonon polariton propagation direction, and broadband controlling light dispersion of polaritonic nanodevices.

Abstract Background This study aimed to explore the mechanism of Ge-Gen-Qin-Lian decoction (GGQLD) in the alleviation of symptoms of type 2 diabetes mellitus (T2DM) with inflammatory bowel disease (IBD) by network pharmacology and experimental validation. Methods The active components and targets of GGQLD were identified from the TCMSP database. The potential therapeutic targets of T2DM and IBD were identified from the GEO database and 4 online disease target databases. The PPI network and KEGG/GO analyses were performed with the common targets among GGQLD, T2DM and IBD. Molecular docking was carried out between the core compounds and hub targets. To verify the above results, UHPLC-MS technology was used to identify the chemical compounds in GGQLD, and a T2DM with IBD rat model was used to explore the mechanism by which GGQLD treats T2DM with IBD. Results Totally, 70 potential therapeutic targets were identified among GGQLD, T2DM and IBD. Ten hub genes were selected from the PPI network. KEGG analysis revealed that GGQLD is tightly involved in the AGE-RAGE signaling pathway. Berberine, baicalein, wogonin, and quercitrin are the main active compounds of GGQLD. Animal experiments showed that GGQLD could decrease blood glucose and alleviate intestinal inflammation. Notably, the concentrations of AGEs, the expression of RAGE, c-JUN and NF-κB and the expression of inflammatory cytokines were decreased by GGQLD. Conclusions Our study initially demonstrated that GGQLD has favorable anti-hyperglycemic and anti-intestinal inflammation effects in a T2DM with IBD rat model, and the AGE-RAGE pathway plays a vital role in this process.

The laser modification pretreatment method is an effective way to address the machining difficulties of silicon carbide (SiC), a typical hard-to-machine material. Therefore, the ablation behavior and modification mechanism of SiC under different laser energy were explored in this paper. A new multi-scale model of laser irradiation SiC that couples heat transfer and fluid motion is first established. Then, a series of experiments is carried out to evaluate the model's accuracy, and the interaction between laser and SiC is discussed in detail. The results show that the dominant modification mechanism changes from coulomb explosion to multiphoton absorption, incubation effect, and heat accumulation with the laser energy increase. This leads the surface topography of SiC to transition from nanoparticle formation to disorder to a melting state. In ablative state, micro/nano porous and humps are formed at the edge of ablation groove due to surface tension, generation and rupture of bubbles, respectively. Furthermore, the surface roughness is not proportional to the laser energy due to the plasma shielding effect, and the surface roughness can be reduced by enhancing the flow of the molten material. Amorphous Si–O–C, Si and spherical SiO2 exist in deposition area, leading to SiC elastic modulus decreases from 347 GPa to 103.82 GPa, and the shear strength decreases from 20.9 GPa to 17.25 GPa. The results of this study can provide references for parameters selection and theoretical support for improving the machinability of SiC.

Abstract Background Due to the high mortality and disability rate of intracranial hemorrhage, headache is not the main focus of research on cerebral arteriovenous malformation (AVM), so research on headaches in AVM is still scarce, and the clinical understanding is shallow. This study aims to delineate the risk factors associated with headaches in AVM and to compare the effectiveness of various intervention treatments versus conservative treatment in alleviating headache symptoms. Methods This study conducted a retrospective analysis of AVMs who were treated in our institution from August 2011 to December 2021. Multivariable logistic regression analysis was employed to assess the risk factors for headaches in AVMs with unruptured, non-epileptic. Additionally, the effectiveness of different intervention treatments compared to conservative management in alleviating headaches was evaluated through propensity score matching (PSM). Results A total of 946 patients were included in the analysis of risk factors for headaches. Abstract Background Biological-derived hydroxyapatite is widely used as a bone substitute for addressing bone defects, but its limited osteoconductive properties necessitate further improvement. The osteo-immunomodulatory properties hold crucial promise in maintaining bone homeostasis, and precise modulation of macrophage polarization is essential in this process. Metabolism serves as a guiding force for immunity, and fluoride modification represents a promising strategy for modulating the osteoimmunological environment by regulating immunometabolism. In this context, we synthesized fluorinated porcine hydroxyapatite (FPHA), and has demonstrated its enhanced biological properties and osteogenic capacity. However, it remains unknown whether and how FPHA affects the immune microenvironment of the bone defects. Methods FPHA was synthesized and its composition and structural properties were confirmed. Macrophages were cultured with FPHA extract to investigate the effects of FPHA on their polarization and the related osteo-immune microenvironment. Furthermore, total RNA of these macrophages was extracted, and RNA-seq analysis was performed to explore the underlying mechanisms associated with the observed changes in macrophages. The metabolic states were evaluated with a Seahorse analyzer. Additionally, immunohistochemical staining was performed to evaluate the macrophages response after implantation of the novel bone substitutes in critical size calvarial defects in SD rats. Results The incorporation of fluoride ions in FPHA was validated. FPHA promoted macrophage proliferation and enhanced the expression of M2 markers while suppressing the expression of M1 markers. Additionally, FPHA inhibited the expression of inflammatory factors and upregulated the expression of osteogenic factors, thereby enhancing the osteogenic differentiation capacity of the rBMSCs. RNA-seq analysis suggested that the polarization-regulating function of FPHA may be related to changes in cellular metabolism. Further experiments confirmed that FPHA enhanced mitochondrial function and promoted the metabolic shift of macrophages from glycolysis to oxidative phosphorylation. Moreover, in vivo experiments validated the above results in the calvarial defect model in SD rats. Conclusion In summary, our study reveals that FPHA induces a metabolic shift in macrophages from glycolysis to oxidative phosphorylation. This shift leads to an increased tendency toward M2 polarization in macrophages, consequently creating a favorable osteo-immune microenvironment. These findings provide valuable insights into the impact of incorporating an appropriate concentration of fluoride on immunometabolism and macrophage mitochondrial function, which have important implications for the development of fluoride-modified immunometabolism-based bone regenerative biomaterials and the clinical application of FPHA or other fluoride-containing materials. The immunomodulatory effects of FPHA were investigated through in vitro and in vivo studies, revealing its ability to induce a metabolic shift in macrophages from glycolysis to mitochondrial oxidative phosphorylation (OxPhos). This metabolic remodeling resulted in a notable suppression of M1 macrophage polarization and promotion of M2 macrophage polarization. Furthermore, FPHA was found to enhance osteogenic differentiation and facilitate bone repair. Abstract Lipopolysaccharide (LPS) is known to elicit a robust immune response. This study aimed to investigate the impact of LPS on the transcriptome of human nasal epithelial cells (HNEpC). HNEpC were cultured and stimulated with LPS (1 μg/mL) or an equivalent amount of normal culture medium. Subsequently, total RNA was extracted, purified, and sequenced using next-generation RNA sequencing technology. Differentially expressed genes (DEGs) were identified and subjected to functional enrichment analysis. A protein–protein interaction (PPI) network of DEGs was constructed, followed by Ingenuity Pathway Analysis (IPA) to identify molecular pathways influenced by LPS exposure on HNEpC. Validation of key genes was performed using quantitative real-time PCR (qRT-PCR). A total of 97 DEGs, comprising 48 up-regulated genes and 49 down-regulated genes, were identified. Results from functional enrichment analysis, PPI, and IPA indicated that DEGs were predominantly enriched in chemokine-related signaling pathways. Subsequent qRT-PCR validation demonstrated significant upregulation of key genes in these pathways in LPS-treated HNEpC compared to control cells. In conclusion, LPS intervention profoundly altered the transcriptome of HNEpC, potentially exacerbating inflammatory responses through the activation of chemokine-related signaling pathways.

Understanding the spatial pattern of vegetation in the Gobi area is of paramount importance for preserving its vegetation and the overall stability of the ecosystem. Here, we investigated the populations of the predominant species, Ephedra przewalskii, at various locations within the alluvial fan situated in the southern foothills of the Tianshan Mountains in the Gobi area. The location of the E. przewalskii distribution in the sample plot was extracted from unmanned aerial vehicle (UAV) images. This article analyzes the distribution patterns of the predominant species based on different null models, tests whether the distribution patterns are affected by habitat heterogeneity, and examines whether other ecological processes affect these patterns. Additionally, using the variance decomposition method, we analyze the explanatory power of habitat factors on the distribution patterns of the predominant species. The results show that: (1) Using UAVs to locate the distribution of vegetation information for sparse vegetation pattern analysis in the Gobi region has good applicability and prospects; (2) the frequency, density, and coverage of E. przewalskii decrease sharply with decreasing altitude. At a small scale (0∼10 m), the E. przewalskii population is mainly aggregated, whereas on a large scale (>10 m), it primarily exhibits uniform or random distributions; (3) the E. przewalskii population distribution pattern is significantly affected by habitat heterogeneity, mainly on a large scale. The species distribution in the study area strongly correlates with the altitude, soil moisture, and topographic wetness index. Habitat factors can explain 64.2 % of the reasons for species distribution; and (4) The clustering of E. przewalskii population at the apex of the alluvial fan is primarily driven by facilitation, whereas at the middle and bottom sections, it is predominantly influenced by habitat heterogeneity and restricted diffusion. The results enhance our comprehension of the spatial patterns and environmental adaptation mechanisms exhibited by the predominant species in the study area. Furthermore, they provide a scientific foundation for ecological conservation and biodiversity protection in the environmentally vulnerable Gobi region.

Abstract The Hemiptera insects are the largest incomplete metamorphosis insect group in Insecta and play a vital role in ecosystems and biodiversity. Previous studies on the spatial distribution of Hemiptera insects mainly focused on a specific region and insect, this study explored the spatial distribution characteristics of Hemiptera insects in China (national scale), and further clarified the dominant factors affecting their spatial distribution. We used spatial autocorrelation analysis, hot spot analysis, and standard ellipse to investigate the spatial distribution characteristics of Hemiptera insects in China. Furthermore, we used geographic detectors to identify the main factors affecting their spatial distribution under China's six agricultural natural divisions and explore the influencing mechanism of dominant factors. The results show that: (i) The spatial differentiation characteristics of Hemiptera insects in China are significant, and their distribution has obvious spatial agglomeration. The Hu Huanyong Line is an important dividing line for the spatial distribution of Hemiptera insects in China. From the city scale, the HH type (high‐high cluster) is mainly distributed on both sides of the Hu Huanyong Line. (ii) The hot spots of Hemiptera insects are mainly distributed in southwest China, along the Qinling Mountains, the western side of the Wuyi Mountains, the Yinshan Mountains, the Liupanshan Mountains, the Xuefeng Mountains, the Nanling Mountains, and other mountainous areas. (iii) Under agricultural natural divisions, the influence of natural environmental factors on the spatial distribution of Hemiptera insects is obviously different. Temperature and precipitation are the dominant factors. Natural factors and socio‐economic factors have formed a positive reinforcement interaction mode on the spatial distribution of Hemiptera insects. These can provide the decision‐making basis for biodiversity conservation and efficient pest control.

The improvement and verification of fluid dynamics simulation on temperature uniformity during the heat treatment of ring pieces are investigated in this study. The accuracy of the temperature field model is validated by comparing the simulation results with the measured temperatures. The findings reveal that the vortex generated near the furnace wall during heat treatment significantly affects the uniformity of the temperature field. To improve this, adjustments are made to the placement of ring pieces based on an experimentally validated fluid dynamics simulation model, and subsequent calculations are performed on this adjusted model. It is observed that these adjustments greatly enhance temperature uniformity in the heating process, with a 39.06 % improvement in medium-temperature zone (732.32–743.69 k) within the furnace compared to the original model. Additionally, surface temperatures of ring pieces in another medium-temperature zone (668.89–691.11 k) show a 34.54 % improvement in comparison to those predicted by the original model.

Abstract Currently, the typical combination therapy of programmed death ligand‐1 (PD‐L1) antibodies with radiotherapy (RT) still exhibits impaired immunogenic antitumor response in clinical due to lessened DNA damage and acquired immune tolerance via the upregulation of some other immune checkpoint inhibitors. Apart from this, such combination therapy may raise the occurrence rate of radiation‐induced lung fibrosis (RIPF) due to enhanced systemic inflammation, leading to the ultimate death of cancer patients (average survival time of about 3 years). Therefore, it is newly revealed that mitochondria energy metabolism regulation can be used as a novel effective PD‐L1 and transforming growth factor‐β (TGF‐β) dual‐downregulation method. Following this, IR‐TAM is prepared by conjugating mitochondria‐targeted heptamethine cyanine dye IR‐68 with oxidative phosphorylation (OXPHOS) inhibitor Tamoxifen (TAM), which then self‐assembled with albumin (Alb) to form IR‐TAM@Alb nanoparticles. By doing this, tumor‐targeting IR‐TAM@Alb nanoparticle effectively reversed tumor hypoxia and depressed PD‐L1 and TGF‐β expression to sensitize RT. Meanwhile, due to the capacity of heptamethine cyanine dye in targeting RIPF and the function of TAM in depressing TGF‐β, IR‐TAM@Alb also ameliorated fibrosis development induced by RT.

The temperature of a motor significantly affects its control and lifespan. However, due to the influence of motor structure and operating environment, precise temperature measurement of the motor is challenging with temperature sensors. Therefore, machine learning algorithms are often employed to predict the temperature more accurately. To enhance motor control, integrating machine learning algorithm models with the actual motor control terminal is highly beneficial. This paper proposes a Short and Long Term Memory (LSTM) algorithm model based on Harris's hawk optimization to predict the temperature of the motor stator, which is applied in actual motor control. Furthermore, it evaluates the tracking performance of motor control current. Firstly, an experimental platform for temperature measurement is established to acquire the temperature at different positions of the motor as raw data. Subsequently, the raw data is inputted into three algorithms: LSTM, PSO-LSTM, and HHO-LSTM, for comparison. By comparing evaluation metrics, it is demonstrated that HHO-LSTM exhibits excellent predictive performance. Furthermore, utilizing diverse segments of the motor as model input sets enhances the generalization capability and predictive accuracy of the model. Finally, applied in actual motor control, a comparative analysis of q-axis and d-axis currents reveals reduced fluctuation errors, showcasing improved motor control effectiveness.

Abstract Intragrain impurities can impart detrimental effects on the efficiency and stability of perovskite solar cells, but they are indiscernible to conventional characterizations and thus remain unexplored. Using in situ scanning transmission electron microscopy, we reveal that intragrain impurity nano-clusters inherited from either the solution synthesis or post-synthesis storage can revert to perovskites upon irradiation stimuli, leading to the counterintuitive amendment of crystalline grains. In conjunction with computational modelling, we atomically resolve crystallographic transformation modes for the annihilation of intragrain impurity nano-clusters and probe their impacts on optoelectronic properties. Such critical fundamental findings are translated for the device advancement. Adopting a scanning laser stimulus proven to heal intragrain impurity nano-clusters, we simultaneously boost the efficiency and stability of formamidinium-cesium perovskite solar cells, by virtual of improved optoelectronic properties and relaxed intra-crystal strain, respectively. This device engineering, inspired and guided by atomic-scale in situ microscopic imaging, presents a new prototype for solar cell advancement.

Abstract Wireless and battery-free radio-frequency (RF) sensors can be used to create physical spaces that ambiently sense and respond to human activities. Making such sensors ultra-flexible and transparent is important to preserve the aesthetics of living environments, accommodate daily activities, and functionally integrate with objects. However, existing RF sensors are unable to simultaneously achieve high transparency, flexibility, and the electrical conductivity required for remote room-scale operation. Here, we report 4.5 µm RF tag sensors achieving transparency exceeding 90% that provide capabilities in room-scale ambient wireless sensing. We develop a laser-assisted water-based adhesion-reversion process to digitally realize computer-aided RF design at scale. By individually tagging multiple objects and regions of the human body, we demonstrate multiplexed wireless tracking of human-environment interactions and physiological signals at a range of up to 8 m. These radio-frequency identification sensors open opportunities for non-intrusive wireless sensing of daily living spaces for applications in health monitoring and elderly care.

To obtain high-frequency SAW devices, the interdigital transducer electrodes are prepared narrower and the electrode spacing is smaller, which leads to higher cost and lower reliability of high-frequency SAW devices. In this paper, two other interdigital electrode parallel layout structures are designed based on the traditional IDT flat layer layout structure, and the influence of the three different IDT electrode layout structures on the SAW device of LiNbO3/Diamond/Si multilayer structure is studied by COMSOL Multiphysics. The results show that the designed multi-layer structure SAW device can successfully excite SAW with superior performance, and the parallel layout structure of the interdigital electrode can reduce the lateral size of SAW device, which provides a new idea and direction for the miniaturization of the SAW devices.

Abstract The industrial applications of the κ-carrageenases have been restricted by their poor thermostability. In this study, based on the folding free energy change (ΔΔG) and the flexibility analysis using molecular dynamics (MD) simulation for the alkaline κ-carrageenase KCgCD from Pseudoalteromonas porphyrae (WT), the mutant S190R was identified with improved thermostability. After incubation at 50 °C for 30 min, the residual activity of S190R was 63.7%, 25.7% higher than that of WT. The T m values determined by differential scanning calorimetry were 66.2 °C and 64.4 °C for S190R and WT, respectively. The optimal temperature of S190R was 10 °C higher than that of WT. The κ-carrageenan hydrolysates produced by S190R showed higher xanthine oxidase inhibitory activity compared with the untreated κ-carrageenan. MD simulation analysis of S190R showed that the residues (V186–M194 and P196–G197) in F5 and the key residue R150 in F3 displayed the decreased flexibility, and residues of T169–N173 near the catalytic center displayed the increased flexibility. These changed flexibilities might be the reasons for the improved thermostability of mutant S190R. This study provides a useful rational design strategy of combination of ΔΔG calculation and MD simulation to improve the κ-carrageenase’s thermostability for its better industrial applications.

The Arctic fox (Vulpes lagopus) is a species indigenous to the Arctic and has developed unique lipid metabolism, but the mechanisms remain unclear. Here, the significantly increased body weight of Arctic foxes was consistent with the significantly increased serum very-low-density lipoprotein (VLDL), and the 40% crude fat diet further increased the Arctic fox body weight. The enhanced body weight gain stems primarily from increased subcutaneous adipose tissue accumulation. The adipose triacylglycerol and phosphatidylethanolamine were significantly greater in Arctic foxes. The adipose fatty-acid synthase content was significantly lower in Arctic foxes, highlighting the main role of exogenous fatty-acids in fat accumulation. Considering the same diet, liver-derived fat dominates adipose expansion in Arctic foxes. Liver transcriptome analysis revealed greater fat and VLDL synthesis in Arctic foxes, consistent with the greater VLDL. Glucose homeostasis wasn’t impacted in Arctic foxes. And the free fatty-acids in adipose, which promote insulin resistance, also did not differ between groups. However, the hepatic glycogen was greater in Arctic foxes and transcriptome analysis revealed upregulated glycogen synthesis, improving glucose homeostasis. These results suggest that the superior fat accumulation capacity and distinct characteristics of hepatic and adipose lipid and glucose metabolism facilitate glucose homeostasis and massive fat accumulation in Arctic foxes.

Currently, there is a lack of clarity regarding the differences in the composition and function of rumen microorganisms among buffaloes at different growth stages.MethodsIn this study, metagenomics sequencing technology was applied to examine the compositional and functional differences of rumen microorganisms in adult and breastfed buffaloes.ResultsThe results revealed that the rumen of adult buffaloes had significantly higher levels of the following dominant genera: Prevotella, UBA1711, RF16, Saccharofermentans, F23-D06, UBA1777, RUG472, and Methanobrevibacter_A. Background Irritable bowel syndrome (IBS) is a functional gastrointestinal disorder (FGID) with heterogeneous clinical presentations. There are no clear testing parameters for its diagnosis, and the complex pathophysiology of IBS and the limited time that doctors have to spend with patients makes it difficult to adequately educate patients in the outpatient setting. An increased awareness of IBS means that patients are more likely to self-diagnose and self-manage IBS based on their own symptoms. These factors may make patients more likely to turn to Internet resources. Wikipedia is the most popular online encyclopedia among English-speaking users, with numerous validations. However, in Mandarin-speaking regions, the Baidu Encyclopedia is most commonly used. There have been no studies on the reliability, readability, and objectivity of IBS information on the two sites. This is an urgent issue as these platforms are accessed by approximately 1.45 billion people. Objective We compared the IBS content on Wikipedia (in English) and Baidu Baike (in Chinese), two online encyclopedias, in terms of reliability, readability, and objectivity. Methods The Baidu Encyclopedia (in Chinese) and Wikipedia (in English) were evaluated based on the Rome IV IBS definitions and diagnoses. All possible synonyms and derivatives for IBS and IBS-related FGIDs were screened and identified. Two gastroenterology experts evaluated the scores of articles for both sites using the DISCERN instrument, the Journal of the American Medical Association scoring system (JAMA), and the Global Quality Score (GQS). ABSTRACT In this study, a mixture of estradiol benzoate, progesterone, and testosterone propionate was injected into Diannan small-ear sows to induce estrus. The 16S rRNA technology was used to comparatively analyze the differences in fecal microbial composition and diversity between induced and spontaneous estrus in Diannan small-ear sows. The most abundant phylum in the sows in estrus were Firmicutes, Bacteroidetes, Actinobacteria, and Proteobacteria. There was a significant negative correlation between Firmicutes and Bacteroide and a significant positive correlation between Bacteroides and Bifidobacterium. The relative abundance of Stenotrophomonas, Neisseria, Anaerofustis, and Terrisporobacter in the sows during induced estrus was significantly higher than that during spontaneous estrus. Taken together, induced estrus affects the relative abundance of specific microbes in the feces of Diannan small-ear sows, but it does not affect the overall composition and diversity. These results provide fundamental knowledge about the gut microbiota of sows with induced estrus.

However, chimerism rate is dynamic changes, and the sensitivity of different chimerism requires further research.MethodsTo investigate the predictive value of distinct chimerism for relapse, we measured bone marrow (BM), peripheral blood (PB), and T-cell (isolated from BM) chimerism in 178 patients after allo-HSCT.ResultsReceiver operating characteristic (ROC) curve showed that T-cell chimerism was more suitable to predict relapse after allo-HSCT compared with PB and BM chimerism. The cutoff value of T-cell chimerism for predicting relapse was 99.45%. Leukemia and myelodysplastic syndrome (MDS) relapse patients’ T-cell chimerism was a gradual decline from 2 months to 9 months after allo-HSCT. Higher risk of relapse and death within 1 year after allo-HSCT. The T-cell chimerism rates in remission and relapse patients were 99.43% and 94.28% at 3 months after allo-HSCT (P = 0.009), 99.31% and 95.27% at 6 months after allo-HSCT (P = 0.013), and 99.26% and 91.32% at 9 months after allo-HSCT (P = 0.024), respectively. There was a significant difference (P = 0.036) for T-cell chimerism between early relapse (relapse within 9 months after allo-HSCT) and late relapse (relapse after 9 months after allo-HSCT) at 2 months after allo-HSCT. The urgent need to address climate change has prompted a focus on carbon-intensive industries, such as cement production. This study explores a novel approach by utilizing dredged sludge (DS), a hazardous waste, treated with mechanical-thermal activation to enhance its pozzolanic activity. The resulting sintered sludge ash (SSA) serves as an supplementary cementitious material for the preparation of sintered sludge cement (SSC), potentially reducing the carbon footprint of traditional cement. Compressive strength tests reveal that up to a 30% dosage of SSA maintains minimal change in 28 d strength while improving chloride erosion resistance. However, a 50% SSA dosage leads to reduced strength due to inadequate cement. Freeze-thaw cycle tests indicate a negative impact on frost resistance with increasing SSA dosage. Micro tests, including heat of hydration, mercury intrusion porosimetry (MIP), and scanning electron microscope (SEM), provide insights into the evolving properties of SSC. A lower SSA dosage initially increases porosity, but hydration progress results in a denser microstructure.

Abstract Singularities ubiquitously exist in different fields and play a pivotal role in probing the fundamental laws of physics and developing highly sensitive sensors. Nevertheless, achieving higher-order (≥3) singularities, which exhibit superior performance, typically necessitates meticulous tuning of multiple (≥3) coupled degrees of freedom or additional introduction of nonlinear potential energies. Here we propose theoretically and confirm using mechanics experiments, the existence of an unexplored cusp singularity in the phase-tracked (PhT) steady states of a pair of coherently coupled mechanical modes without the need for multiple (≥3) coupled modes or nonlinear potential energies. By manipulating the PhT singularities in an electrostatically tunable micromechanical system, we demonstrate an enhanced cubic-root response to frequency perturbations. This study introduces a new phase-tracking method for studying interacting systems and sheds new light on building and engineering advanced singular devices with simple and well-controllable elements, with potential applications in precision metrology, portable nonreciprocal devices, and on-chip mechanical computing.

Abstract Salmonellosis is a globally extensive food-borne disease, which threatens public health and results in huge economic losses in the world annually. The rising prevalence of antibiotic resistance in Salmonella poses a significant global concern, emphasizing an imperative to identify novel therapeutic agents or methodologies to effectively combat this predicament. In this study, self-assembly hydrogen sulfide (H2S)-responsive nanoprodrugs were fabricated with poly(α-lipoic acid)-polyethylene glycol grafted rhein and geraniol (PPRG), self-assembled into core–shell nanoparticles via electrostatic, hydrophilic and hydrophobic interactions, with hydrophilic exterior and hydrophobic interior. The rhein and geraniol are released from self-assembly nanoprodrugs PPRG in response to Salmonella infection, which is known to produce hydrogen sulfide (H2S). PPRG demonstrated stronger antibacterial activity against Salmonella compared with rhein or geraniol alone in vitro and in vivo. Additionally, PPRG was also able to suppress the inflammation and modulate gut microbiota homeostasis. In conclusion, the as-prepared self-assembly nanoprodrug sheds new light on the design of natural product active ingredients and provides new ideas for exploring targeted therapies for specific Enteropathogens. Abstract Multi principal element alloys (MPEAs) comprise an atypical class of metal alloys. MPEAs have been demonstrated to possess several exceptional properties, including, as most relevant to the present study a high corrosion resistance. In the context of MPEA design, the vast number of potential alloying elements and the staggering number of elemental combinations favours a computational alloy design approach. In order to computationally assess the prospective corrosion performance of MPEA, an approach was developed in this study. A density functional theory (DFT) – based Monte Carlo method was used for the development of MPEA ‘structure’; with the AlCrTiV alloy used as a model. High-throughput DFT calculations were performed to create training datasets for surface activity/selectivity towards different adsorbate species: O2-, Cl- and H+. Machine-learning (ML) with combined representation was then utilised to predict the adsorption and vacancy energies as descriptors for surface activity/selectivity. The capability of the combined computational methods of MC, DFT and ML, as a virtual electrochemical performance simulator for MPEAs was established and may be useful in exploring other MPEAs.

Abstract Cuproptosis caused by copper overload is mediated by a novel regulatory mechanism that differs from previously documented mechanisms regulating cell death. Cells dependent on mitochondrial respiration showed increased sensitivity to a copper ionophore elesclomol that induced cuproptosis. Maternal embryonic leucine zipper kinase(MELK) promotes tumorigenesis and tumor progression through the PI3K/mTOR pathway, which exerts its effects partly by targeting the pyruvate dehydrogenase complex(PDHc) and reprogramming the morphology and function of mitochondria. However, the role of MELK in cuproptosis remains unclear. Here, we validated that elevated MELK expression enhanced the activity of PI3K/mTOR signaling and subsequently promoted Dihydrolipoamide S-Acetyltransferase (DLAT) expression and stabilized mitochondrial function. This regulatory effect helped to improve mitochondrial respiration, eliminate excessive intracellular reactive oxygen species (ROS), reduce intracellular oxidative stress/damage and the possibility of mitochondria-induced cell fate alternations, and ultimately promote the progression of HCC. Meanwhile, elesclomol reduced translocase of outer mitochondrial membrane 20(TOM 20) expression and increased DLAT oligomers. Moreover, the above changes of MELK to HCC were abolished by elesclomol. In conclusion, MELK enhanced the levels of the cuproptosis-related signature(CRS) gene DLAT (especially the proportion of DLAT monomer) by activating the PI3K/mTOR pathway, thereby promoting elesclomol drug resistance, altering mitochondrial function, and ultimately promoting HCC progression.

The ecology of the aquatic environment in Poyang Lake, the largest fresh lake in China, is notably impacted by the backflow from the Yangtze River, which conveys a high flux of sediments. This study employs a widely recognized numerical model to replicate the backflow in 2007 (the strongest backflow after the operation initiation of the Three Gorges Dam) to investigate the contributions of wind and backflow to the sediment transport process. The results show that the influences of wind and backflow on flow patterns and sediment transport processes have significant spatial heterogeneity. In the narrow waterway leading to the central lake area, hydrodynamics is mainly driven by backflow. Conversely, the hydrodynamics of the open expanse of the lake is primarily influenced by wind forces. Dominant wind leads to the formation of gyres, which significantly alter flow paths and push sediment into the upstream areas. As a result, the suspended sediment area expands at an average rate of 20.1–21.3 km2 daily, marking a 75–85% surge compared to the no wind condition (11.5 km2). The study facilitates a deeper understanding of sediment transport processes in large lakes. Abstract Background Mycobacterium houstonense is a category of rapidly growing mycobacteria that is gram-positive, acid-fast, polycrystalline, and non-spore-forming. There have been few reports of human infection caused by Mycobacterium houstonense worldwide. Case presentation We present a case of chronic periprosthetic joint infection caused by Mycobacterium houstonense in an elderly female patient. The patient developed signs of infection after undergoing total hip arthroplasty. Despite receiving antibiotic treatment and revision surgery, the signs of infection recurred repeatedly. Multiple bacterial cultures during the treatment period were negative. Later, we identified the pathogenic bacteria Mycobacterium houstonense through mNGS testing, isolated the bacteria from the ultrasonically centrifuged fluid of the prosthesis and obtained drug sensitivity results. Finally, we performed a revision surgery and treated the patient with moxifloxacin and clindamycin. After treatment, the patient did not show signs of infection recurrence during 24 months of follow-up. Conclusion Through a relevant literature search, we believe that Mycobacterium houstonense may show higher sensitivity to amikacin and quinolone antibiotics. Additionally, clarifying occult infection sources through methods such as gene testing will improve the diagnosis and treatment of periprosthetic joint infection.

Abstract Background Goat is an important livestock worldwide, which plays an indispensable role in human life by providing meat, milk, fiber, and pelts. Despite recent significant advances in microbiome studies, a comprehensive survey on the goat microbiomes covering gastrointestinal tract (GIT) sites, developmental stages, feeding styles, and geographical factors is still unavailable. Here, we surveyed its multi-kingdom microbial communities using 497 samples from ten sites along the goat GIT. Results We reconstructed a goat multi-kingdom microbiome catalog (GMMC) including 4004 bacterial, 71 archaeal, and 7204 viral genomes and annotated over 4,817,256 non-redundant protein-coding genes. We revealed patterns of feeding-driven microbial community dynamics along the goat GIT sites which were likely associated with gastrointestinal food digestion and absorption capabilities and disease risks, and identified an abundance of large intestine-enriched genera involved in plant fiber digestion. We quantified the effects of various factors affecting the distribution and abundance of methane-producing microbes including the GIT site, age, feeding style, and geography, and identified 68 virulent viruses targeting the methane producers via a comprehensive virus-bacterium/archaea interaction network. Conclusions Together, our GMMC catalog provides functional insights of the goat GIT microbiota through microbiome-host interactions and paves the way to microbial interventions for better goat and eco-environmental qualities. Milking methods have significant impacts on the microbiological composition, which could affect the quality of raw buffalo milk. Hence, the current study was conducted on the impact of milking methods on microorganisms in buffalo tank raw milk from 15 farms in Guangxi, China. The farms were divided into two groups based on the milking method: mechanical milking (MM, n = 6) and hand milking (HM, n = 9). Somatic cell counts, bacterial cell counts and nutrients of the raw buffalo milk samples were analyzed. The comparison of raw buffalo milk samples was analyzed using metagenomic sequencing to detect any differences between the two groups. There was no significant difference in the basic nutritional compositions and somatic cell count of raw buffalo milk between the two milking methods. However, the HM samples had significantly higher bacterial counts and diversity compared to the MM samples. The results showed that Staphylococcus spp., Klebsiella spp., Streptococcus spp., and Pseudomonas spp. were the major microbes present in canned raw buffalo milk. However, the differences between the two milking methods were the relative abundance of core microorganisms and their potential mastitis-causing genera, including the content of antibiotic-resistance genes and virulence genes. Our study revealed that Staphylococcus spp. and Streptococcus spp. were significantly more abundant in the MM group, while Klebsiella spp. was more abundant in the HM group. Regardless of the milking method used, Pseudomonas spp. was identified as the primary genus contributing to antibiotic resistance and virulence genes in canned raw buffalo milk. These findings affirm that there are differences in the microbial and genomic levels in canned raw milk. To prove the functional roles of the discovered genes and how these genes affect milk quality, further research and experimental validation are necessary.

To understand the role of an additional coordination site in the linker in chirality sensing, we designed and synthesized an S-2-methylbutanamido-substituted m-phthalic diamide-linked zinc bisporphyrinate, [Zn2(S-MAABis)] and investigated its ability to sense the chirality of amino acid esters. The 1H NMR spectra and the crystal structure showed that the amido oxygen adjacent to the chiral carbon was coordinated with zinc. NMR and UV–vis titration showed that the binding of [Zn2(S-MAABis)] to amino acid esters occurred via two equilibria, forming 1:1 and 1:2 host–guest complexes. The CD spectra suggested that [Zn2(S-MAABis)] can effectively recognize the absolute configuration of amino acid esters. The sign of the CD spectra remained unchanged during the titration, indicating that the corresponding 1:1 and 1:2 host–guest complexes had the same chirality. This is different from previously studied amino-substituted m-phthalic diamide-linked zinc bisporphyrinate [Zn2(AmBis)], which showed chirality inversion during titration. Theoretical calculations indicated that the additional coordination sites (amido or amino) in the 1:1 host–guest complexes played different roles, leading to differences in chirality. Our studies suggest that the introduction of a coordination site can influence the chirality transfer process, but the results of chirality transfers are dependent on the specific binding modes.

The dissociative ionization of CHBr2Cl molecules in femtosecond laser fields at 800 nm and 400 nm is investigated to enhance the comprehension of ultrafast dynamics phenomena. The kinetic energy distribution of the resulting ions following photo-dissociation is analyzed using time-of-flight mass spectrometry in combination with DC-sliced ion velocity map imaging. The findings from the experimental study indicate that the presence of low kinetic energy components is attributed to the dissociative ionization processes of CHBr2Cl molecules. The complexity of individual dissociation pathways remains unaffected by the laser fields but is determined by factors such as bond energy, ionization energy of neutral groups, and charge distribution. In the case of 400 nm laser fields, distinct elimination channels enable CHBr2Cl+ ions to circumvent the transition state, leading to the formation of BrCl+ and Br2+ fragments.

IntroductionHigh-throughput sequencing (HTS) provides an efficient and cost-effective way to generate large amounts of sequence data, providing a very powerful tool to analyze biodiversity of soil organisms. However, marker-based methods and the resulting datasets come with a range of challenges and disputes, including incomplete reference databases, controversial sequence similarity thresholds for delimitating taxa, and downstream compositional data analysis. MethodsHere, we use HTS data from a soil nematode biodiversity experiment to explore standardized HTS data processing procedures. We compared the taxonomic assignment performance of two main rDNA reference databases (SILVA and PR2). We tested whether the same ecological patterns are detected with Amplicon Sequence Variants (ASV; 100% similarity) versus classical Operational Taxonomic Units (OTU; 97% similarity). Further, we tested how different HTS data normalization methods affect the recovery of beta diversity patterns and the identification of differentially abundant taxa.ResultsAt this time, the SILVA 138 eukaryotic database performed better than the PR2 4.12 database, assigning more reads to family level and providing higher phylogenetic resolution. ASV- and OTU-based alpha and beta diversity of nematodes correlated closely, indicating that OTU-based studies represent useful reference points. For downstream data analyses, our results indicate that loss of data during subsampling under rarefaction-based methods might reduce the sensitivity of the method, e.g. underestimate the differences between nematode communities under different treatments, while the clr-transformation-based methods may overestimate effects. The Analysis of Compositions of Microbiome with Bias Correction approach (ANCOM-BC) retains all data and accounts for uneven sampling fractions for each sample, suggesting that this is currently the optimal method to analyze compositional data.DiscussionOverall, our study highlights the importance of comparing and selecting taxonomic reference databases before data analyses, and provides solid evidence for the similarity and comparability between OTU- and ASV-based nematode studies. Further, the results highlight the potential weakness of rarefaction-based and clr-transformation-based methods. We recommend future studies use ASV and that both the taxonomic reference databases and normalization strategies are carefully tested and selected before analyzing the data.

Corrosion fatigue behavior of a new candidate material for geothermal turbine blade, modified 17-4PH, was investigated in detail. The environmental acceleration of crack growth rate was discussed in terms of ∆K. Fracture surface and side surface were observed by SEM. The crack path on side surface was also analyzed by EBSD. Then, the corrosion mechanism was discussed. Striation width on the fracture surface was compared to the crack growth per cycle. Since pitting occurred during the corrosion fatigue test, anodic polarization test was exercised to measure the pitting potential, and the cause for the pitting was also discussed. The hardness was also characterized by micro-hardness test and auger electron mapping.

Abstract Chronic prostatic inflammation promotes cell survival and fibrosis, leading to benign prostatic hyperplasia (BPH) with aggravated urinary symptoms. It is investigated whether yes‐associated protein 1 (YAP1), an organ size controller and mechanical transductor, is implicated in inflammation‐induced BPH. The correlation between YAP1 expression and fibrosis in human and rat BPH specimens is analyzed. Furthermore, the effects of YAP1 activation on prostatic cell survival and fibrosis, as well as the underlying mechanism, are also studied. As a result, total and nuclear YAP1 expression, along with downstream genes are significantly upregulated in inflammation‐associated human and rat specimens. There is a significant positive correlation between YAP1 expression and the severity of fibrosis or clinical performance. YAP1 silencing suppresses cell survival by decreasing cell proliferation and increasing apoptosis, and alleviates fibrosis by reversing epithelial‐mesenchymal transition (EMT) and extracellular matrix (ECM) deposition in prostatic BPH‐1 and WPMY‐1 cells. Mechanistically, inflammatory stimulus and rigid matrix stiffness synergistically activate the RhoA/ROCK1 pathway to provoke cytoskeleton remodeling, thereby promoting YAP1 activation to exacerbate BPH development. Overall, inflammation‐triggered mechanical stiffness reinforcement activates the RhoA/ROCK1/F‐actin/YAP1 axis, thereby promoting prostatic cell survival and fibrosis to accelerate BPH progression.

A numerical study was conducted on an axial compressor, the NASA Stage 35, with three nonuniform tip clearances to understand the effect of a different shape tip clearances rotor on the compressor's performance. The results demonstrated that by modifying the traditional parallel tip clearance to sine-type tip clearance (STC), hump-type tip clearance (HTC), and concave-type tip clearance (CTC), the compressor’s peak efficiency showed remarkable improvement and the SMI was significantly improved. In comparison to the design rotor, the SMI of STC, HTC, and CTC increased by 3.102 %, 2.672 %, and 0.645 %, respectively. The leakage distribution at the tip-clearance region from LE to TE exhibited an inverse pattern to that of the tip curve. The leakage’s magnitude could not reflect the TLF’s intensity, and the leakage in the middle of the blade tip had a role in the size of the TLV. Leakage at the tip’s TE influenced the corner separation’s scale in the casing. The STC and HTC schemes’ total pressure ratio improved, the low-velocity zone’s area, high-entropy area, and high absolute vorticity area at the LE of rotor’s PS decreased, the detached shock moved backward, the leading-edge spillage flow decreased, the shock action position of the rotor suction surface moved forward, the BLS increased, the TPL in the stator channel decreased, and the CTC exhibited the opposite trend. A nonuniform tip clearance was achieved by reducing the leakage and TLV intensity, thereby reducing the size of the TLV, induced vortex, or CSV in rotor passage and increasing the compressor’s SMI.

Stochastic resonance (SR) typically manifests in nonlinear systems, wherein the detection of a weak signal is bolstered by the addition of noise. Since its first discovery in a study of ice ages on Earth, various types of SRs have been observed in biological and physical systems and have been implemented in sensors to benefit from noise. However, a universally designed sensor architecture capable of accommodating different types of SRs has not been proposed, and the widespread applications of SRs in daily environments have not yet been demonstrated. Here, we propose a sensor architecture to simultaneously realize multi-type SRs and demonstrate their wide applications in mechanical, optical, and acoustic sensing domains. In particular, we find the coexistence of excitable SR and bistable SR in a sensor architecture composed of wirelessly coupled inductor–capacitor resonators connected to a nonlinearly saturable amplifier. In both types of SRs, adding noise to the system leads to a characteristic noise-enhanced signal-to-noise ratio (SNR). We further validate our findings through mechanical, optical, and acoustic sensing experiments and obtain noise-enhanced SNR by 9 dB, 3 dB, and 7 dB, respectively, compared to the standard methods devoid of SR integration. Our findings provide a general strategy to design various types of SRs and pave the way for the development of a distinctive class of sensors leveraging environmental noise, with potential applications ranging from biomedical devices to ambient sensing.

Abstract Cattle and the draught force provided by its skeletal muscle have been integral to agro‐ecosystems of agricultural civilization for millennia. However, relatively little is known about the cattle muscle functional genomics (including protein coding genes, non‐coding RNA, etc.). Circular RNAs (circRNAs), as a new class of non‐coding RNAs, can be effectively translated into detectable peptides, which enlightened us on the importance of circRNAs in cattle muscle physiology function. Here, RNA‐seq, Ribosome profiling (Ribo‐seq), and peptidome data are integrated from cattle skeletal muscle, and detected five encoded peptides from circRNAs. It is further identified and functionally characterize a 907‐amino acids muscle‐specific peptide that is named circNEB‐peptide because derived by the splicing of Nebulin (NEB) gene. This peptide localizes to the nucleus and cytoplasm and directly interacts with SKP1 and TPM1, key factors regulating physiological activities of myoblasts, via ubiquitination and myoblast fusion, respectively. The circNEB‐peptide is found to promote myoblasts proliferation and differentiation in vitro, and induce muscle regeneration in vivo. These findings suggest circNEB‐peptide is an important regulator of skeletal muscle regeneration and underscore the possibility that more encoding polypeptides derived by RNAs currently annotated as non‐coding exist.

Abstract Background The aim of this study was to evaluate the precision and feasibility of patient-specific instruments (PSI) in total hip arthroplasty (THA) as compared to the traditional free-hand (FRH) approach. Methods During the period of January 1, 2021 to December 31, 2022, a randomized allocation was used for patients receiving unilateral primary THA to either the PSI or conventional operation group. The placement and size of the PSI were specifically chosen to guide femoral neck resection and prosthesis implantation. The study analyzed component positions and evaluated radiographic and clinical outcomes in 30 patients who received PSI-assisted THAs and 30 patients who received FRH THAs. This study was registered at China Clinical Trial Registry (number: ChiCTR2300072325) on June 9th, 2023. Results The use of PSI in THA resulted in significantly higher precision in achieving the desired component position as compared to the FRH approach. Abstract Exceptional point (EP) is a special degeneracy of non-Hermitian systems. One-dimensional transmission systems operating at EPs are widely studied and applied to chiral conversion and sensing. Lately, two-dimensional systems at EPs have been exploited for their exotic scattering features, yet so far been limited to only the non-visible waveband. Here, we report a universal paradigm for achieving a high-efficiency EP in the visible by leveraging interlayer loss to accurately control the interplay between the lossy structure and scattering lightwaves. A bilayer framework is demonstrated to reflect back the incident light from the left side ( | r −1 | >0.999) and absorb the incident light from the right side ( | r +1 | < 10–4). As a proof of concept, a bilayer metasurface is demonstrated to reflect and absorb the incident light with experimental efficiencies of 88% and 85%, respectively, at 532 nm. Our results open the way for a new class of nanoscale devices and power up new opportunities for EP physics.

Abstract Implanted bioelectronic devices can form distributed networks capable of sensing health conditions and delivering therapy throughout the body. Current clinically-used approaches for wireless communication, however, do not support direct networking between implants because of signal losses from absorption and reflection by the body. As a result, existing examples of such networks rely on an external relay device that needs to be periodically recharged and constitutes a single point of failure. Here, we demonstrate direct implant-to-implant wireless networking at the scale of the human body using metamaterial textiles. The textiles facilitate non-radiative propagation of radio-frequency signals along the surface of the body, passively amplifying the received signal strength by more than three orders of magnitude (>30 dB) compared to without the textile. Using a porcine model, we demonstrate closed-loop control of the heart rate by wirelessly networking a loop recorder and a vagus nerve stimulator at more than 40 cm distance. Our work establishes a wireless technology to directly network body-integrated devices for precise and adaptive bioelectronic therapies.

Abstract Background The use of degradable magnesium screws to fix the bone flap implanted in the treatment of femoral head necrosis has achieved preliminary good therapeutic results. However, there is no conclusive evidence in the study to demonstrate whether biodegradable magnesium screws promote angiogenesis and no comparison has been made between degradable magnesium screws and traditional screws. Objective To investigate the clinical efficacy and safety of biodegradable magnesium screws in pedicled vascularized iliac bone graft transfer (PVIBGT) for osteonecrosis of the femoral head (ONFH). Materials and methods A total of thirty-six patients (37 hips) with ONFH were recruited from March 2020 to July 2022. The study used a single-blind method, and patients who underwent PVIBGT were randomized into three groups: 12 patients (12 hips) were fixed with biodegradable magnesium screws (Group A), 12 patients (13 hips) were fixed with titanium screws (Group B), 12 patients (12 hips) were directly embedded (Group C). The operating time and the length of stay were recorded. Harris scores, radiological examinations (X-ray, CT, DCE-MRI), blood and serum tests were conducted before and after surgery. The gas yield and degradation rates of the magnesium screws were measured at the 3-months and 6-months post-operative follow-ups in Group A. Results There was no statistically significant difference among these three groups in terms of types, gender, age, course of disease, surgical side, operation time, the length of stay (P > 0.05). Peroxisome proliferator-activated receptor γ (PPARG) has various splicing variants and plays essential roles in the regulation of adipocyte differentiation and lipogenesis. However, little is known about the expression pattern and effect of the PPARG on milk fat synthesis in the buffalo mammary gland. In this study, we found that only PPARG-X17 and PPARG-X21 of the splicing variant were expressed in the buffalo mammary gland. Amino acid sequence characterization showed that the proteins encoded by PPARG-X17 and PPARG-X21 are endonuclear non-secreted hydrophilic proteins. Protein domain prediction found that only the PPARG-X21-encoded protein had PPAR ligand-binding domains (NR_LBD_PPAR), which may lead to functional differences between the two splices. RNA interference (RNAi) and the overexpression of PPARG-X17 and PPARG-X21 in buffalo mammary epithelial cells (BMECs) were performed. Results showed that the expression of fatty acid synthesis-related genes (ACACA, CD36, ACSL1, GPAT, AGPAT6, DGAT1) was significantly modified (p < 0.05) by the RNAi and overexpression of PPARG-X17 and PPARG-X21. All kinds of FAs detected in this study were significantly decreased (p < 0.05) after RNAi of PPARG-X17 or PPARG-X21. Overexpression of PPARG-X17 or PPARG-X21 significantly decreased (p < 0.05) the SFA content, while significantly increased (p < 0.05) the UFA, especially the MUFA in the BMECs. In conclusion, there are two PPARG splicing variants expressed in the BMECs that can regulate FA synthesis by altering the expression of diverse fatty acid synthesis-related genes. This study revealed the expression characteristics and functions of the PPARG gene in buffalo mammary glands and provided a reference for further understanding of fat synthesis in buffalo milk.

Amino acid permeases (AAPs) transporters are crucial for the long-distance transport of amino acids in plants, from source to sink. While Arabidopsis and rice have been extensively studied, research on foxtail millet is limited. This study identified two transcripts of SiAAP9, both of which were induced by NO3− and showed similar expression patterns. The overexpression of SiAAP9L and SiAAP9S in Arabidopsis inhibited plant growth and seed size, although SiAAP9 was found to transport more amino acids into seeds. Furthermore, SiAAP9-OX transgenic Arabidopsis showed increased tolerance to high concentrations of glutamate (Glu) and histidine (His). The high overexpression level of SiAAP9 suggested its protein was not only located on the plasma membrane but potentially on other organelles, as well. Interestingly, sequence deletion reduced SiAAP9’s sensitivity to Brefeldin A (BFA), and SiAAP9 had ectopic localization on the endoplasmic reticulum (ER). Protoplast amino acid uptake experiments indicated that SiAAP9 enhanced Glu transport into foxtail millet cells. Overall, the two transcripts of SiAAP9 have similar functions, but SiAAP9L shows a higher colocalization with BFA compartments compared to SiAAP9S. Our research identifies a potential candidate gene for enhancing the nutritional quality of foxtail millet through breeding.

Sample pretreatment is a key step for qualitative and quantitative analysis of trace substances in complex samples. Cis-dihydroxyl (cis-diol) group-containing substances exist widely in biological samples and can be selectively bound by boronate affinity adsorbents. Based on this, in this article, we proposed a simple method for the preparation of novel spherical three-dimensionally ordered macropore (3DOM) materials based on a combination of the boronate affinity technique and colloidal crystal template method. The prepared 3DOM materials were characterized using Fourier transform–infrared spectroscopy, scanning electron microscopy, X-ray photoelectron spectroscopy, and thermo-gravimetric analysis, and results showed that they possessed the characteristics of a high specific surface area, high porosity, and more boronic acid recognition sites. The adsorption performance evaluation results showed that the maximum adsorption capacity of the boron affinity 3DOMs on ovalbumin (OVA) could reach to 438.79 mg/g. Kinetic and isothermal adsorption experiments indicated that the boronate affinity 3DOM material exhibited a high affinity and selectivity towards OVA and adenosine. Sodium dodecyl sulfate–polyacrylamide gel electrophoresis analysis of the proteins in egg whites was conducted and proved that the glycoprotein in the egg whites could be separated and enriched with a good performance. Therefore, a novel boronate affinity 3DOM material a with highly ordered and interconnected pore structure was prepared and could be applied in the separation and enrichment of molecules with cis-diol groups from complex samples with a good selectivity, efficiency, and high throughput.

Monitoring drought stress is crucial for estimating productivity and assessing the health status of forest ecosystems under global climate change. Solar-induced chlorophyll fluorescence (SIF) is mechanistically coupled to photosynthesis and has advantages over greenness-based vegetation indices in detecting drought. In recent years, SIF has commonly been used in monitoring drought stress in crop ecosystems. However, the response of tower-based SIF to drought stress in forest ecosystems remains unclear. In this study, we investigated the potential of tower-based SIF to monitor drought, which was quantified using the plant water stress index (PWSI) in a Chinese cork oak plantation. The results show the negative effect of drought on SIF, and afternoon depression of SIF emission under drought stress was observed. Canopy SIF (F) exhibited a nonlinear relationship with PWSI, while the quantum yield of SIF (ΦF) exhibited a significant linear relationship with PWSI at 687 nm and 760 nm (ΦF687: R2 = 0.90; ΦF760: R2 = 0.85). Incident radiation (PAR) and canopy structure affected the response of SIF to drought stress, with PAR as the main factor causing the nonlinear relationship between F and PWSI. Afternoon depression was described as the afternoon/morning ratio (AMR). AMRF and AMRΦF exhibited a negative linear response to PWSI. AMRF was less affected than F by PAR and canopy structures, and AMRΦF was more physiologically representative than ΦF. Moreover, AMRΦF was sensitive to VPD and REW, and it might be a good indicator of drought. Red SIF was more sensitive to drought than far-red SIF, as the R2 of PWSI with AMRΦF687 (R2 = 0.89) was higher than that with AMRΦF687 (R2 = 0.84). These results highlight the potential of tower-based SIF, especially red SIF, for drought monitoring in a plantation, and consideration of the physiological diurnal variation in SIF under drought stress is crucial for improving the accuracy of drought stress monitoring.

Background: Gastric adenocarcinoma (GAC) is a malignant tumor with the highest incidence in the digestive system. Macrophages have been proven to play important roles in tumor microenvironment. Methods: Herein, single-cell RNA sequencing (scRNA-seq) profiles from the Gene Expression Omnibus (GEO) and bulk RNA-seq data from the Cancer Genome Atlas (TCGA) database were utilized to construct a macrophage marker gene signature (MMGS) to predict the prognosis of GAC patients. Subsequently, a risk score model based on the MMGS was built to predict the prognosis of GAC patients; further, this was validated in the GEO cohort. The risk score categorized patients into the high- and low-risk groups. A nomogram model based on the risk score and clinic-pathological characteristics was developed. Results: Seven genes, ABCA1, CTHRC1, GADD45B, NPC2, PLTP, PRSS23, and RNASE1, were included in the risk score model. Patients with a low-risk score showed a better prognosis. The MMGS had good sensitivity and specificity for predicting the prognosis inGAC patients. The risk score was an independent prognostic factor. The constructed nomogram exhibited favorable predictability and reliability for predicting GAC prognosis. Conclusion: In conclusion, the risk score model based on the seven MMGSs performed well in the predicting prognosis of GAC patients. Our study may provide new insights into clinical decision-making for the personalized treatment of patients with gastric cancer (GC).

To address the inaccuracy of the Constant Acceleration/Constant Velocity (CA/CV) model as the state equation in describing the relative motion state in UAV relative navigation, an adaptive UAV relative navigation method is proposed, which is based on the UAV attitude information provided by Attitude and Heading Reference System (AHRS). The proposed method utilizes the AHRS output attitude parameters as the benchmark for dead reckoning and derives a relative navigation state equation with attitude error as process noise. By integrating the extended Kalman filter output for relative state estimation and employing an adaptive decision rule designed using the innovation of the filter update phase, the proposed method recalculates motion states deviating from the actual motion using the Tasmanian Devil Optimization (TDO) algorithm. The simulation results show that, compared with the CA/CV model, the proposed method reduces the relative position errors by 12%, 23%, and 32% in the X, Y, and Z directions, respectively, and that it reduces the relative velocity errors by 350%, 330%, and 300%, respectively. There is a significant improvement in the relative navigation accuracy.
