To explore the different extraction methods and traditional refining process of squid visceral fish oil and its quality changes, the deep-sea squid viscera from the Equator, Argentina, and the North Pacific were used as raw materials in this study. Firstly, different organic reagent extraction methods and compound methods were used to extract fish oil from Argentine squid viscera. The method with the highest oil yield and best quality was selected for refining three types of crude squid visceral oil. The sensory morphology, physicochemical indicators, fatty acid composition and volatile flavor compounds of three types of crude and refined squid visceral oils were analyzed. The results showed that the oil extraction method of the No.6 solvent assisted freeze-thaw was the best, with a yield of 12.90%, and the physical and chemical indexes of squid visceral oil after refining were significantly improved. To investigate the impact response and damage mechanism of CFRP composite materials for high-speed trains under high-speed impact from foreign objects of different shapes and sizes, high-speed impact experiments and simulations were conducted on CFRP composite laminate at a velocity of 163 m/s. A finite element simulation model was established in Abaqus/Explicit. This model adopts the 3D-Hashin failure criterion considering strain rate effects to simulate the failure of fibers and matrix. Bonded elements with zero thickness were inserted along the fiber direction in the laminate to replicate the damage phenomena of fiber bundle debonding and pull-out. The effectiveness of the finite element model was validated by high-speed photography of the impact process and ultrasonic C-scan damage images recorded during the experiments. Based on this simulation model, the influence of projectiles of different shapes and sizes on the impact response of CFRP composite materials was studied. Projectiles of different shapes exhibit their own characteristics in terms of the time of peak force occurrence, manifestation, and characteristics of laminar damage. The peak force of the cylindrical projectile is 245.01% greater than that of the spherical one, while the conical projectile is 52.26% smaller than the spherical one. When the projectile size increases from 10 mm to 20 mm, the peak force of the spherical projectile, conical projectile, and cylindrical projectile increases by 79.67%, 120.12%, and 283.13% respectively. The total area of delamination in laminated plates is positively correlated with the size of the impacting body.

Abstract Background Intra-oral halitosis (IOH) is bad breath produced locally by the mouth in addition to systemic diseases and is one of the main causes of interpersonal communication and psychological disorders in modern society. However, current treatment modalities still only alleviate IOH and do not eradicate it. Therefore, based on the differential performance of oral microecology in IOH patients, we propose a microbiota transplantation treatment aimed at restoring oral microecological balance and analyze its feasibility by oral flora colonization test in Wistar rats. Objective Saliva flora samples were collected from IOH patients and healthy subjects to analyze the feasibility of oral microbiota transplantation (OMT) for the treatment of IOH by the Wistar rat oral flora colonization test. Methods Seven patients with IOH who visited the First Affiliated Hospital of Xinjiang Medical University from June 2017 to June 2022 with the main complaint of halitosis and three healthy subjects were randomly selected. A Halimeter portable breath detector was used to record breath values and collect saliva flora samples. Sixteen SPF-grade male Wistar rats were housed in the Animal Experiment Center of Xinjiang Medical University and randomly divided into an experimental group (Group E) and a control group (Group C) for the oral flora colonization test. Species composition and associated metabolic analysis of oral flora during the Wistar rat test using 16SrRNA sequencing technology and PICRUSt metabolic analysis. The intricate interplay between the gut microbiome and colorectal cancer (CRC) presents novel avenues for early diagnosis and prognosis, crucial for improving patient outcomes. This comprehensive review synthesizes current findings on the gut microbiome’s contribution to CRC pathogenesis, highlighting its potential as a biomarker for non-invasive CRC screening strategies. We explore the mechanisms through which the microbiome influences CRC, including its roles in inflammation, metabolism, and immune response modulation. Furthermore, we assess the viability of microbial signatures as predictive tools for CRC prognosis, offering insights into personalized treatment approaches. Our analysis underscores the necessity for advanced metagenomic studies to elucidate the complex microbiome-CRC nexus, aiming to refine diagnostic accuracy and prognostic assessment in clinical settings. This review propels forward the understanding of the microbiome’s diagnostic and prognostic capabilities, paving the way for microbiome-based interventions in CRC management.

Abstract In the last decade, prophages that possess the ability of lysogenic transformation have become increasingly significant. Their transfer and subsequent activity in the host have a significant impact on the evolution of bacteria. Here, we investigate the role of prophage phi456 with high spontaneous induction in the bacterial genome of Avian pathogenic Escherichia coli (APEC) DE456. The phage particles, phi456, that were released from DE456 were isolated, purified, and sequenced. Additionally, phage particles were no longer observed either during normal growth or induced by nalidixic acid in DE456Δphi456. This indicated that the released phage particles from DE456 were only phi456. We demonstrated that phi456 contributed to biofilm formation through spontaneous induction of the accompanying increase in the eDNA content. The survival ability of DE456Δphi456 was decreased in avian macrophage HD11 under oxidative stress and acidic conditions. This is likely due to a decrease in the transcription levels of three crucial genes—rpoS, katE, and oxyR—which are needed to help the bacteria adapt to and survive in adverse environments. It has been observed through animal experiments that the presence of phi456 in the DE456 genome enhances colonization ability in vivo. Additionally, the number of type I fimbriae in DE456Δphi456 was observed to be reduced under transmission electron microscopy when compared to the wild-type strain. The qRT-PCR results indicated that the expression levels of the subunit of I fimbriae (fimA) and its apical adhesin (fimH) were significantly lower in DE456Δphi456. Therefore, it can be concluded that phi456 plays a crucial role in helping bacterial hosts survive in unfavorable conditions and enhancing the colonization ability in DE456.

Cancer cachexia is a multifactorial syndrome characterized by progressive weight loss and a disease process that nutritional support cannot reverse. Although progress has been made in preclinical research, there is still a long way to go in translating research findings into clinical practice. One of the main reasons for this is that existing preclinical models do not fully replicate the conditions seen in clinical patients. Therefore, it is important to understand the characteristics of existing preclinical models of cancer cachexia and pay close attention to the latest developments in preclinical models. The main models of cancer cachexia used in current research are allogeneic and xenograft models, genetically engineered mouse models, chemotherapy drug-induced models, Chinese medicine spleen deficiency models, zebrafish and Drosophila models, and cellular models. This review aims to revisit and summarize the commonly used animal models of cancer cachexia by evaluating existing preclinical models, to provide tools and support for translational medicine research.

Abstract Macrophages regulate essential aspects of innate immunity against pathogens. In response to microbial components, macrophages activate primary and secondary inflammatory gene programs crucial for host defense. The liver X receptors (LXRα, LXRβ) are ligand‐dependent nuclear receptors that direct gene expression important for cholesterol metabolism and inflammation, but little is known about the individual roles of LXRα and LXRβ in antimicrobial responses. Here, the results demonstrate that induction of LXRα transcription by prolonged exposure to lipopolysaccharide (LPS) supports inflammatory gene expression in macrophages. LXRα transcription is induced by NF‐κB and type‐I interferon downstream of TLR4 activation. Moreover, LPS triggers a reprogramming of the LXRα cistrome that promotes cytokine and chemokine gene expression through direct LXRα binding to DNA consensus sequences within cis‐regulatory regions including enhancers. LXRα‐deficient macrophages present fewer binding of p65 NF‐κB and reduced histone H3K27 acetylation at enhancers of secondary inflammatory response genes. Mice lacking LXRα in the hematopoietic compartment show impaired responses to bacterial endotoxin in peritonitis models, exhibiting reduced neutrophil infiltration and decreased expansion and inflammatory activation of recruited F4/80lo‐MHC‐IIhi peritoneal macrophages. Together, these results uncover a previously unrecognized function for LXRα‐dependent transcriptional cis‐activation of secondary inflammatory gene expression in macrophages and the host response to microbial ligands.

This study aimed to assess the safety and toxicity of peony protein and provide a scientific basis for its application in food industry. The safety evaluation included acute oral toxicity tests in mice, as well as 28 d and 90 d oral toxicity tests in rats. The results demonstrated that no signs of poisoning or mortality were observed in the acute oral toxicity test (intragastric dose of 11700 mg/kg) conducted on mice. Furthermore, in the 28 d and 90 d oral toxicity tests conducted on rats, no abnormal changes were observed in terms of food utilization, organ weight ratios, and various physiological activities, when compared to the control group (1500, 3000, 6000 mg/kg). Additionally, there were no significant differences (P>0.05) in blood routine and blood biochemical indices between the treatment groups and the control group. Histopathological examination of organ tissues revealed intact morphology without any toxic lesions. These findings provided solid evidence that peony protein was non-toxic in the dose range of less than 11700 mg/kg and hold promising potential for application in the food industry.

In this study, a high-strength carbon fiber reinforced composite laminate for rail vehicles was machined as a perforated sample and repaired by bonding a patch of the same material. The mechanical response of the repaired laminate to a low-velocity impact with energies of 25J and 30J was investigated through experiments and simulations with variations in patch thickness, size radius, and off-axis angle. The finite element simulation model was established in Abaqus/Explicit. The model integrated the progressive damage model based on the 3D-Hashin failure criterion, which can simulate the intralaminar damage of the fiber and the matrix. The cohesive zone model was used to simulate the delamination damage. According to the finite element analysis results, the mechanism and process of impact damage of the repaired laminates were analyzed. Based on the validated finite element model, the effect of patch size, thickness and off-axis angle on impact response was investigated. Patch radius reduced to 15 mm or thickness reduced to 2.1 mm can still resist low-velocity impacts with an energy of 30 J. Although the off-axis angle of the patch had little effect on the impact response of the repaired laminate, an off-axis angle of 90° was effective in suppressing delamination damage within the patch.

Abstract Objective The current body of evidence on the association between the ratio of triglycerides to high-density lipoprotein cholesterol (TG/HDL-c) and the reversal of prediabetes to normoglycemia remains limited. The aim of this study is to investigate the association between TG/HDL-c and the reversion to normoglycemia in patients with prediabetes. Methods This retrospective cohort study included 15,107 individuals with prediabetes from 32 Chinese districts and 11 cities who completed health checks from 2010 to 2016. The Cox proportional-hazards regression model examined baseline TG/HDL-c and reversion to normoglycemia from prediabetes. Cox proportional hazards regression with cubic spline functions and smooth curve fitting determined the non-linear connection between TG/HDL-c and reversion to normoglycemia. We also ran sensitivity and subgroup analysis. By characterizing progression to diabetes as a competing risk for the reversal of prediabetes to normoglycemic event, a multivariate Cox proportional hazards regression model with competing risks was created. Results Upon adjusting for covariates, the findings indicate a negative association between TG/HDL-c and the likelihood of returning to normoglycemia (HR = 0.869, 95%CI:0.842–0.897). Additionally, a non-linear relationship between TG/HDL-c and the probability of reversion to normoglycemia was observed, with an inflection point of 1.675. The HR on the left side of the inflection point was 0.748 (95%CI:0.699, 0.801). The robustness of our results was confirmed through competing risks multivariate Cox's regression and a series of sensitivity analyses. Conclusion The present study reveals a negative and non-linear correlation between TG/HDL-c and the reversion to normoglycemia among Chinese individuals with prediabetes. The findings of this study are anticipated to serve as a valuable resource for clinicians in managing dyslipidemia in prediabetic patients. Interventions aimed at reducing the TG/HDL-c ratio through the reduction of TG or elevation of HDL-c levels may substantially enhance the likelihood of achieving normoglycemia in individuals with prediabetes.

Abstract Psoriasis is a chronic inflammatory skin disease that affects millions of people worldwide. Sulforaphane (SFN) has been shown to have anti-inflammatory and antioxidant properties. In this study, we investigated the effects of SFN on a mouse model of psoriasis induced by imiquimod (IMQ) and its underlying molecular mechanism. Mice treated with SFN showed significant improvement in psoriatic symptoms, including reduced erythema, scales, and cutaneous thickness. Histopathological analysis and immunohistochemical staining revealed decreased expression of K16, K17, and Ki67 in SFN-treated mice, indicating reduced abnormal differentiation of keratinocytes and cutaneous inflammation. SFN treatment also reduced the activation of STAT3 and NF-κB pathways and downregulated pro-inflammatory cytokines IL-1β, IL-6, and CCL2. In vitro experiments using HaCaT cells demonstrated that SFN inhibited IL-22 and TNF-α-induced activation of inflammatory pathways and keratinocyte proliferation. Network pharmacology analysis suggested that the KEAP1-NRF2 pathway might be involved in the protective effects of SFN on psoriasis. We observed reduced NRF2 expression in human psoriatic lesions, and subsequent experiments showed that SFN activated KEAP1-NRF2 pathway in vivo and in vitro. Importantly, NRF2-deficient mice exhibited aggravated psoriasis-like symptoms and reduced response to SFN treatment. Our findings indicate that SFN ameliorates psoriasis symptoms and inflammation through the KEAP1-NRF2 pathway, suggesting a potential therapeutic role for SFN in the treatment of psoriasis.

Most earthen sites are located in open environments eroded by wind and rain, resulting in spalling and cracking caused by shrinkage due to constant water absorption and loss. Together, these issues seriously affect the stability of such sites. Gypsum–lime-modified soil offers relatively strong mechanical properties but poor water resistance. If such soil becomes damp or immersed in water, its strength is significantly reduced, making it unviable for use as a material in the preparation of earthen sites. In this study, we achieved the composite addition of a certain amount of sodium methyl silicate (SMS), titanium dioxide (TiO2), and graphene oxide (GO) into gypsum–lime-modified soil and analyzed the microstructural evolution of the composite-modified soil using characterization methods such as XRD, SEM, and EDS. A comparative study was conducted on changes in the mechanical properties of the composite-modified soil and original soil before and after immersion using water erosion, unconfined compression (UCS), and unconsolidated undrained (UU) triaxial compression tests. These analyses revealed the micro-mechanisms for improving the waterproof performance of the composite-modified soil. The results showed that the addition of SMS, TiO2, and GO did not change the crystal structure or composition of the original soil. In addition, TiO2 and GO were evenly distributed between the modified soil particles, playing a positive role in filling and stabilizing the structure of the modified soil. After being immersed in water for one hour, the original soil experienced structural instability leading to collapse. While the water absorption rate of the composite-modified soil was only 0.84%, its unconfined compressive strength was 4.88 MPa (the strength retention rate before and after immersion was as high as 93.1%), and the shear strength was 614 kPa (the strength retention rate before and after immersion was as high as 96.7%).

Fecal scores are crucial for assessing the digestive and gastrointestinal status of animals. The Bristol fecal scoring system is a commonly used method for the subjective evaluation of host feces, there is limited research on fecal scoring standards for fattening Hu sheep. In this study, Hu sheep were collected for rumen, rectum, and colon contents for 16S rDNA sequencing. 514 Hu sheep feces were scored based on the Bristol fecal scoring system, and production performance at each stage was measured. Finally, we developed the scoring standard of the manure of Hu sheep in the fattening period (a total of five grades). The result shows that moisture content significantly increased with higher grades (p < 0.05). We analyzed the relationship between fecal scores and production traits, blood indices, muscle nutrients, and digestive tract microorganisms. The growth traits (body weight, body height, body length, average daily gain (ADG), and average daily feed intake (ADFI) during 80–180 days), body composition traits of the F3 group, and the carcass traits were found to be significantly higher (p < 0.05) than those of the F1 and F2 groups. There was no significant difference in gastrointestinal microflora diversity among all groups (p > 0.05). Significant differences were observed in Aspartate aminotransferase, Glucose, Total bilirubin, and Red Blood Cell Count between groups (p < 0.05). The mutton moisture content in group F4 was significantly higher than in the other groups, and the protein content was also the lowest (p < 0.05). The results of the correlation analysis demonstrated that Actinobacteria, Peptostreptococcaceae, Acidaminococcales, Gammaproteobacteria, and Proteobacteria were the significant bacteria affecting fecal scores. In addition, Muribaculaceae and Oscillospiraceae were identified as the noteworthy flora affecting growth performance and immunity. This study highlights the differences in production traits and blood indicators between fecal assessment groups and the complex relationship between intestinal microbiota and fecal characteristics in Hu sheep, suggesting potential impacts on animal performance and health, which suggest strategies for improved management.

Monkeypox virus (MPXV) can induce systemic skin lesions after infection. This research focused on studying MPXV proliferation and the response of keratinocytes. Using transmission electron microscopy (TEM), we visualized different stages of MPXV development in human immortalized keratinocytes (HaCaT). We identified exocytosis of enveloped viruses as the exit mechanism for MPXV in HaCaT cells. Infected keratinocytes showed submicroscopic changes, such as the formation of vesicle-like structures through the recombination of rough endoplasmic reticulum membranes and alterations in mitochondrial morphology. Transcriptome analysis revealed the suppressed genes related to interferon pathway activation and the reduced expression of antimicrobial peptides and chemokines, which may facilitate viral immune evasion. In addition, pathway enrichment analysis highlighted systemic lupus erythematosus pathway activation and the inhibition of the Toll-like receptor signaling and retinol metabolism pathways, providing insights into the mechanisms underlying MPXV-induced skin lesions. This study advances our understanding of MPXV’s interaction with keratinocytes and the complex mechanisms leading to skin lesions.

Livelihood strategies are an effective response to survival risks and stress shocks. Agricultural residents engaged in agriculture, forestry, animal husbandry, and fishery, who are extremely dependent on natural resources, are vulnerable to various livelihood risks. Therefore, the livelihood strategies of agricultural residents are increasingly receiving attention from researchers around the world. However, research on the livelihood strategies of agricultural residents has not yet been systematically analyzed through bibliometrics. Our study was based on 1424 publications in the Web of Science Core Collection database (WoSCC) from 2014 to 2023 to analyze the development history, research hotspots, and trends in the field. Bibliometric analysis was conducted on publications, countries, institutions, authors, journals, and keywords, as well as cited journals and cited references using Excel and CiteSpace software. The number of publications has steadily increased and showed an upward trend. The United States and China were the countries with the most contributions, and Chinese institutions were more active. Cooperation between authors was relatively weak. Sustainability was one of the most productive and highly cited journals. The research hotspots mainly included the relationship between climate change and the livelihood strategies of agricultural residents, the relationship between the land and the livelihood strategies of agricultural residents, the sustainable development of livelihood strategies of agricultural residents, and the characteristics of livelihood strategies of agricultural residents. This is the first time that bibliometric and visual analyses have been conducted on the livelihood strategies of agricultural residents, which may further promote development of the field and lay a foundation for future research.

The synergistic effect of sodium dodecyl benzene sulfonate (SDBS) on the flotation separation of magnesite and dolomite using sodium oleate (NaOL) as a collector has been studied through flotation experiments, zeta potential measurements, contact angle measurements, Fourier transformation infrared spectroscopy analysis (FT-IR), particle size measurements and transmittance measurements. The flotation experiments show that when the synergist, SDBS, is added to the collector, NaOL, the collecting ability and ion resistance of NaOL can be improved so that the flotation separation of magnesite and dolomite can be realized. Zeta potential measurements, contact angle measurements and FT-IR analysis indicate that SDBS and NaOL can co-adsorb on the surface of magnesite. Particle size measurements and transmittance measurements show that SDBS can also improve the dispersion and solubility of NaOL in an aqueous solution, so as to achieve a synergistic effect.

In this paper, using methyl cinnamate as raw material, the new cinnamic hydroxamic acid collector (CIHA) was synthesized by the hydroxylamine method. The collector performance of hydroxamic acid was investigated for scheelite and gangue calcite, and the flotation separation test of scheelite and calcite was carried out with CIHA as the collector. The interaction mechanism between hydroxamic acid and scheelite minerals has also been investigated through zeta potential, Fourier transform infrared spectroscopy (FTIR) experiments, X-ray photoelectron spectroscopy (XPS) experiments, and density functional theory (DFT) calculation. The single mineral flotation test and artificially mixed ore showed that CIHA had an excellent collection effect and selectivity. Zeta potential, FTIR, and XPS showed that CIHA was adsorbed on the scheelite surface by strong chemical adsorption. The active group of CIHA was analyzed through quantum chemical calculation. It was speculated that CO and NO bonds could synthesize a five-membered chelated hydroxamic acid group with Ca element chelate on scheelite surface, changing hydrophobicity and making it more likely to emerge from the pulp.

The fixed-total-pressure-based air-blowing method in free-jet wind tunnels only produces uniformity in a limited area for supersonic jets. To address the issue, we investigated the effect of total pressure on static pressures at the nozzle exit and test chamber. We proposed an air-blowing method for pressure matching and established corresponding equations while suggesting ways to determine its parameters. The pressure-matching method was experimentally verified to analyze and compare the effects of different air-blowing methods and pressure-matching parameters on jet uniformity. The results of the study are as follows: (1) The pressure-matching-based air-blowing method improved the uniformity of the flow field at Mach numbers (Ma) of 1.5, 2.0, and 3.0 by 445.5%, 275%, and 215.8%, respectively, when compared to the air-blowing method that maintains the total pressure above the designed value. (2) The pressure-matching-based air-blowing method reduced the root-mean-square deviation in the Ma of the flow field at a Ma of 2.0 by 47.1% compared to the air-blowing method that maintains the total pressure at the designed value. The findings indicate that the proposed air-blowing technique based on pressure matching can enhance the uniformity of the supersonic jet and the area of the uniform region in the flow field. This method is highly significant in improving the capabilities of free-jet wind tunnel facilities.

The epidemic of alcohol abuse affects millions of people worldwide. Relevant evidence supports the notion that the gut microbiota (GM) plays a crucial role in central nervous system (CNS) function, and its composition undergoes changes following alcohol consumption. Therefore, the purpose of this study was to investigate the effect of reconstructing the gut microbiota by fecal microbiota transplantation (FMT) on alcohol dependence. Here, we established an alcohol dependence model with C57BL/6J mice and proved that FMT treatment improved anxiety-like behavior and alcohol-seeking behavior in alcohol-dependent mice. Additionally, we found that the expression of the intestinal intercellular tight junction structure proteins ZO-1 and occludin was significantly increased after FMT. FMT repaired intestinal permeability in alcohol-dependent mice and decreased the levels of lipopolysaccharide (LPS) and proinflammatory factors. Moreover, the serotonin (5-hydroxytryptamine, 5-HT) content was significantly increased in alcohol-dependent mouse intestinal and brain tissues after receiving the fecal microbiome from healthy mice. 16S rRNA sequencing demonstrated that FMT markedly reshaped the composition of the gut microbiota and elicited changes in the intestinal barrier and 5-HT levels. Collectively, our results revealed that FMT has a palliative effect on alcohol dependence and explored the underlying mechanisms, which provides new strategies for the treatment of alcohol dependence.

Salmonella is a prevalent foodborne and waterborne pathogens threating global public health and food safety. Given the diversity of Salmonella serotypes and the emergence of antibiotic-resistant strains, there is an urgent need for the development of broadly protective therapies. This study aims to prepare monoclonal antibodies (Mabs) with broad reactivity against multi-serotype Salmonella strains, potentially offering cross-protection. We prepared two Mabs F1D4 and B7D4 against protein FliK and BcsZ, two potential vaccine candidates against multi-serotype Salmonella. The two Mabs belonging to IgG1 isotype exhibited high titers of 1:256,000 and 1:512,000 respectively, as well as broad cross-reactivity against 28 different serotypes of Salmonella strains with percentages of 89.29% and 92.86%, correspondingly. Neutralizing effects of the two Mabs on Salmonella growth, adhesion, invasion and motility was evaluated in vitro using bacteriostatic and bactericidal activity with and without complement and bacterial invasion inhibition assay. Additionally, cytotoxicity assays, animal toxicity analyses, and pharmacokinetic evaluations demonstrated the safety and sustained effectiveness of both Mabs. Furthermore, F1D4 or B7D4-therapy in mice challenged with S. Typhimurium LT2 exhibited milder organs damage and lower Salmonella colonization, as well as the higher relative survival of 86.67% and 93.33% respectively. This study produced two broadly reactive and potential cross protective Mabs F1D4 and B7D4, which offered new possibilities for immunotherapy of salmonellosis.

Since wolframite is usually associated with calcite, the separation and enrichment of wolframite by froth flotation remains a great challenge. Herein, a novel trisiloxane surfactant N-(2-aminoethyl)-3-aminopropyltrisiloxane (AATS) was successful synthesized, which was used for the separation of wolframite from calcite for the first time. The flotation separation performance of AATS was studied by flotation test, and its adsorption mechanism was explored based on contact angle, infrared spectrum analysis (FTIR), zeta potential and density functional theory (DFT) calculation. The results of micro-flotation test and binary mixed ore flotation test pointed that AATS had excellent selectivity and more prominent collection capacity for the flotation of wolframite when compared with industrial reagent sodium oleate (NaOL). The measurement results of contact angle proved that AATS improved the hydrophobicity of the wolframite surface. The highly selective adsorption mechanism of AATS surfactant on mineral surfaces were further researched and analyzed by FTIR and zeta potential. The results revealed that AATS surfactant had significant adsorption effect on wolframite, yet almost no adsorption on calcite. DFT calculation indicated that AATS produced electrostatic adsorption with wolframite surface through —N+H3 group.

This study aims to comprehensively investigate the effects of hot-air dehydration on the quality of blue honeysuckle berries (Lonicera caerulea L.). The results demonstrated that drying with hot air at 40–65 °C for 7–72 h resulted in blue honeysuckle berries with a moisture content of 0.21–1.10 g H2O/g dry weight. Generally, low to medium temperatures (40–55 °C) showed a better effect on the quality than high temperatures (60–65 °C). Specifically, drying at 40 °C exclusively resulted in better retention of cuticular wax, the best sensory appearance, and the highest total phenolic content. Drying at 45 °C and 50 °C resulted in the highest antioxidant capacity and the optimal sensory flavor. Drying at 55 °C led to the highest soluble solid/acid ratio, ascorbic acid concentration, total flavonoid, and total anthocyanin. The work introduces an innovative raw berry product and provides a comprehensive practical and theoretical framework for convective dehydration of blue honeysuckle berries.

The problem of wellbore leakage is a key challenge in the petroleum industry, limiting drilling progress and increasing drilling costs. Plugging agents play a role in repairing leaks and fractures; however, traditional plugging materials generally have low mechanical strength, poor adaptability to permeable strata, limited water absorption and expansion capabilities, and poor temperature and salt resistance. To address these limitations, a pioneering polyacrylic acid-polyacrylamide (PAA/PAM) double-network hydrogel was synthesized through aqueous solution polymerization in this study. Its strength, water absorption, expansion, temperature resistance, salt resistance, and plugging effectiveness were comprehensively evaluated. The results demonstrate that good mechanical performance is exhibited by the synthesized hydrogel, capable of withstanding a maximum stress of approximately 3.5 MPa at a 90% strain. Excellent water absorption and expansion are observed in the synthesized double-network hydrogel, with a maximum expansion of 6 times within 30 min and 8 times after 2 h. Test results show that the hydrogel had good temperature resistance and salt resistance, maintaining a strength grade E within the experimental range. The simulated evaluation of the plugging experiment indicates that, under conditions of 130 °C and 6 MPa, the leakage rate of the drilling fluid is maintained below 5 mL/min when the double-network hydrogel is utilized. From the above experimental results, it can be illustrated that excellent mechanical properties, impressive water absorption, and expansion capabilities are exhibited by the synthesized double-network hydrogel. Furthermore, the high-temperature resistance and salt resistance of the double-network hydrogel were also demonstrated. Therefore, In comparison to traditional plugging materials, significant promise is held by this newly synthesized double-network hydrogel material as a plugging agent in drilling fluids.

Lung cancer progression relies on angiogenesis, which is a response to hypoxia typically coordinated by hypoxia-inducible transcription factors (HIFs), but growing evidence indicates that transcriptional programs beyond HIFs control tumor angiogenesis. Here, we show that the redox-sensitive transcription factor BTB and CNC homology 1 (BACH1) controls the transcription of a broad range of angiogenesis genes. BACH1 is stabilized by lowering ROS levels; consequently, angiogenesis gene expression in lung cancer cells, tumor organoids, and xenograft tumors increased substantially following administration of vitamins C and E and N-acetylcysteine in a BACH1-dependent fashion under normoxia. Moreover, angiogenesis gene expression increased in endogenous BACH1–overexpressing cells and decreased in BACH1-knockout cells in the absence of antioxidants. BACH1 levels also increased upon hypoxia and following administration of prolyl hydroxylase inhibitors in both HIF1A-knockout and WT cells. BACH1 was found to be a transcriptional target of HIF1α, but BACH1’s ability to stimulate angiogenesis gene expression was HIF1α independent. Antioxidants increased tumor vascularity in vivo in a BACH1-dependent fashion, and overexpressing BACH1 rendered tumors sensitive to antiangiogenesis therapy. BACH1 expression in tumor sections from patients with lung cancer correlated with angiogenesis gene and protein expression. We conclude that BACH1 is an oxygen- and redox-sensitive angiogenesis transcription factor.

Prediction of the performance of the pressure-regulating valves in a large high-speed jet wind tunnel is important for obtaining and adjusting a high-quality flow field in the test section. In this work, we proposed a prediction method that corrects the theoretical characteristics of the pressure-regulation of the valve based on the experimental testing data of the ratio between the high- and low-pressure of the valve. The current method was validated for a given Mach number of 0.8, and the results showed that compared with the theoretical characteristics, the displacement deviation of the spool was reduced by 9.84%, and the total pressure deviation of the settling chamber was reduced by 43.61%. The results also show that compared with the single low-pressure ratio correction characteristic, the displacement deviation of the spool was reduced by 0.94%, and the total pressure deviation of the settling chamber was reduced by 3.4%. This method can consider the variation of the pressure loss for different pressure ratios and improve the pressure prediction of the valve.

The main source of potassium fertilizers is sylvinite ores consisting primarily of halite (NaCl), silicate and clay-carbonate slurries (clay-salt slurries). Processing of natural potash ores is mainly carried out by the flotation method, which separates KCl, NaCl, and clay-salt slurry. The research is aimed at revealing the effect of sonochemical pretreatment of the depressor reagents, CMC and starch, on dynamic viscosity, aggregate size, electrokinetic potential of these reagent solutions and sylvin flotation performance. It has been established that sonochemical treatment of depressor solutions decreases the size of aggregates of starch molecules by more than 133 times and that of aggregates of CMC molecules from 6 to 4 nm. It has been revealed that sonochemical treatment of anionic CMC solution shifts the electrokinetic potential towards the area of negative values with an increase in acoustic power, while sonochemical treatment of any acoustic power has no effect on the zeta potential of nonionic starch. It has been found that the sonochemical treatment lowers the dynamic viscosity of CMC and starch solutions: the viscosity of CMC solution at a maximum acoustic power of 420 W decreases by 44 % and the viscosity of starch solution at the same acoustic (ultrasonic) power decreases by 70 %. Furthermore, sonochemical pretreatment of sylvin flotation depressors contributes to an increase in KCl recovery and a decrease in the slurry content in the flotation concentrate. The possibility of reducing the consumption of ultrasonic treated depressor is also demonstrated. It is expedient to test the obtained findings in pilot-plant conditions.

This paper focuses on the influence of road weight captured by vibration on vibratory vibrator energy excitation and the optimization of vibratory vibrator baseplate when there is road instead of direct contact between baseplate and the ground. For this purpose, based on the vibration attenuation formula of the foundation surface, the variation law of the captured road weight is obtained. The excitation dynamics model considering the captured road weight is constructed and verified. The energy excitation effect of vibration system is analyzed. By using topology optimization method, the baseplate is optimized, and the comparative study of vibratory vibrator excitation effect before and after optimization is carried out. In the process of energy transmission of the vibrator, the reaction mass will consume most of the energy, the actual output vibratory wave energy is only 5.5% of the total energy. The octagonal I-steel baseplate can reduce its weight by 442 kg while ensuring its rigidity, and the energy excitation effect is relatively improved by 18.2%. The research will provide a reference for improving the energy excitation effect of vibratory vibrator considering the weight of the captured road.

Blue honeysuckle (Lonicera caerulea L.) is an emerging fruit crop; however, determining its proper harvest time in commercial cultivation remains challenging due to its rapid fruit development characteristics. In this study, we investigated 17 agronomic traits of three blue honeysuckle cultivars harvested on 5 successive dates within their respective harvest windows. ‘Lanjingling’, ‘Wulan’, and ‘Berel’ showed solid–acid ratios (SS:TA) ranging from 10.00 to 16.01, 8.13 to 10.23, and 5.77 to 7.11, respectively; anthocyanin contents ranged from 233.85 to 276.83 mg/100 g, 236.38 to 312.23 mg/100 g, and 235.71 to 334.98 mg/100 g, respectively; vitamin C contents ranged from 88.43 to 99.68 mg/100 g, 108.13 to 191.23 mg/100 g, and 89.71 to 120.40 mg/100 g, respectively; phenolic contents ranged from 25.22 to 37.59 mg/g, 25.40 to 36.52 mg/g, and 37.66 to 50.00 mg/g, respectively. The results revealed the SS:TA value consistently increased with delayed harvesting and were significantly negatively correlated with fruit firmness, total acidity, shelf life, and respiration intensity, suggesting it is an ideal maturity indicator for blue honeysuckle berries. The factor analysis suggests that the suitable harvest date for ‘Lanjingling’ could be either 47 days after flowering (DAF) with an SS:TA value of approximately 10.0, characterized by high firmness, extended shelf life, and elevated levels of anthocyanins and phenolics; or 67 DAF (SS:TA ≈ 16.0), characterized by high vitamin C content and sweetness, and larger size and weight. For ‘Wulan’, it suggests the suitable harvest date is either 54 DAF (SS:TA ≈ 9.0), yielding fruit with high levels of anthocyanins and vitamin C; or 62 DAF (SS:TA > 10.0), yielding fruit with high sweetness and large size and weight. For ‘Berel’, it is suggested to be either 52 DAF (SS:TA ≈ 6.5), resulting in fruit with high levels of anthocyanins and vitamin C; or 62 DAF (SS:TA > 7.0), resulting in balanced levels of the fruit quality traits.

To study the perforation performance of CFRP laminates for rail vehicles under high-velocity impact from foreign objects, impact tests on CFRP laminates at a velocity of 163 m/s were carried out, and a corresponding finite element model was established using ABAQUS and verified. The user-defined material subroutine combined the material strain rate hardening effect and the 3D-Hashin damage criterion. The effects of impact velocity, impact object shape, and oblique angle on the perforation performance of CFRP laminates are discussed. Results show that impact velocity positively correlates with impact peak force and residual velocity. Laminates can be perforated by projectiles with a velocity above 120 m/s, and impact velocity greatly influences delamination below 140 m/s. Three shapes of projectile impacting laminates are considered: spherical, cylindrical, and conical. The conical projectile penetrates the laminate most easily, with the largest delamination area. The cylindrical projectile with a flat end suffers the most resistance, and the delaminated area is between the impact conditions of the conical and spherical projectiles. Increasing the angle of inclination increases the impacted area of the laminate and the extent of damage, thus dissipating more energy. The projectile fails to penetrate the laminate when the oblique angle reaches 60°. CFRP composite structures penetrated by high-speed impacts pose a significant threat to the safety of train operations, providing an opportunity for the application of bio-inspired composite structures.

Abstract Objective Evidence regarding the relationship between serum triglyceride-to-high density lipoprotein cholesterol (TG/HDL-c) ratio and outcomes in acute ischemic stroke (AIS) patients is still mixed. Therefore, the present study was undertaken to explore the link between the TG/HDL-c ratio and unfavorable outcomes in patients with AIS. Methods This was a second analysis based on a cohort study. The study population was 1764 patients with AIS collected from January 2010 to December 2016 at a hospital in South Korea. We used a binary logistic regression model to assess the linear association between the TG/HDL-c ratio and unfavorable outcomes for AIS patients. A generalized additive model (GAM) and smooth curve fitting (penalized spline method) was conducted to explore the nonlinear relationship between TG/HDL-c ratio and unfavorable outcomes for AIS patients. Additionally, we compute the inflection point using a recursive algorithm and then build a two-piece binary logistic regression model on both sides of the inflection point. A log-likelihood ratio test was used to determine the most appropriate model describing the association of TG/HDL-c ratio and unfavorable outcomes in patients with AIS. Results The incidence rate of unfavorable outcomes was 28.2%, and the median TG/HDL-c ratio was 2.130. After adjusting covariates, the results of the binary logistic regression model suggested that the relationship between the TG/HDL-c ratio and the risk of unfavorable outcomes for AIS patients was not statistically significant. However, there was a nonlinear relationship between them, and the inflection point of the TG/HDL-c ratio was 3.515. On the left side of the inflection point, each 1-unit increase in the TG/HDL-c ratio was associated with a 22.6% lower risk of unfavorable outcomes (OR = 0.774, 95%CI:0.656 to 0.914, p = 0.002). On the right side of the inflection point, the effect size (OR) was 1.195 (95%CI:1.004 to1.423, p = 0.003). Conclusion There is a nonlinear relationship and threshold effect between the TG/HDL-c ratio and 3-month unfavorable outcomes in AIS patients. When the TG/HDL-c ratio is lower than 3.515, the TG/HDL-c ratio is significantly negatively related to the risk of unfavorable outcomes. When the TG/HDL-c ratio is greater than 3.515, the TG/HDL-c ratio was positively associated with the risk of unfavorable outcomes in AIS patients. This provides a reference for optimizing lipidemia intervention and promoting clinical communication in patients with AIS.

ObjectiveEvidence regarding the relationship between body mass index (BMI) and reversion to normoglycemia from prediabetes is still limited. The purpose of our study is to survey the link of BMI on reversion to normoglycemia among patients with impaired fasting glucose (IFG).MethodsThis study, a retrospective cohort, covered 32 regions and 11 cities in China and collected 258,74 IFG patients who underwent a health check from 2010 to 2016. We investigated the association between baseline BMI and reversion to normoglycemia in patients with IFG using the Cox proportional-hazards regression model. The nonlinear relationship between BMI and reversion to normoglycemia was determined using a Cox proportional hazards regression with cubic spline functions and smooth curve fitting. In addition, we also performed a series of sensitivity analyses and subgroup analyses. A competing risk multivariate Cox regression was performed using progression to diabetes as a competing risk for reversal of normoglycemic events.ResultsAfter adjusting covariates, the results showed that BMI was negatively related to the probability of reversion to normoglycemia (HR=0.977, 95%CI:0.971-0.984). Seasons are the important influencing factor for gut microbiota, which in turn affects the ecology and evolution of the host. The seasonal variation in gut microbiota has increasingly attracted the attention of researchers and professionals worldwide. However, studies of seasonal variations in gut microbiota have not been systematically analyzed by bibliometrics or visual analysis. This study is based on 271 publications from 2012 to 2022 in the Web of Science Core Collection database (WOSCC) to analyze hot spots and trends in this field. The collaborations between different countries, institutions, authors, journals, and keywords were bibliometrically analyzed using Excel, CiteSpace (Version 6.2. R4), and VOSviewer (version 1.6.19) software. The number of publications has been increasing rapidly and shows a general upward trend. China and the Chinese Academy of Sciences are the country and institution contributing the most, respectively. The research hotspots and trends mainly include the diversity of gut microbiota communities in different seasons, the relationship between diet and gut microbiota in seasonal changes, and the relationship between gut microbiota and evolutionary adaptation in seasonal changes. This is the first bibliometric and visualization analysis of seasonal variations in gut microbiota, which may advance this field and lay the foundation for future research.

There has been a noticeable rise in the construction of lightweight stress-ribbon pedestrian bridges. In regions with a high risk of seismic activity, it is crucial to employ advanced seismic control technology to mitigate the impact of earthquakes and improve the bridge’s performance and durability. The objective of this study is to investigate the effect of tuned mass dampers (TMDs) on a steel-plated stress-ribbon bridge using the finite element method. The study aims to analyze the performance of various TMD designs focusing on both vertical and torsional modes. Multiple TMD configurations are considered and numerically compared. The results indicate that TMDs offer both a tuning effect and a static mass effect for stress-ribbon bridges. The tuning effect is most pronounced when the mass ratio falls between 2% and 4%. Installing a single TMD with a mass ratio of 0.05 can decrease vertical displacement along the entire span by up to 36%. Furthermore, a torsional TMD effectively reduces both the torsion of stress ribbons and the pier forces. As a recommendation, the combination of a single vertical displacement TMD and a torsional TMD can be considered the most appropriate scheme for earthquake response control. Moreover, the addition of TMDs reduces the bridge’s sensitivity to the direction of earthquake excitation. These findings contribute to a broader understanding of the earthquake performance of stress-ribbon bridges and assist designers in selecting appropriate control schemes to address vibrational issues.

How to sustainably produce bauxite by effective reverse froth flotation of kaolinite at low temperature is an urgent problem to be solved in the field of mineral processing. In this work, a novel amino-based Gemini surfactant butadiyl-1, 4-bis (dimethyl dodecylammonium bromide) (BBDB) was prepared and first utilized as a novel collector for kaolinite flotation. Its flotation performance for kaolinite was compared with that of the common monomolecular surfactant 1-dodecylamine (DDA) by micro-flotation tests. The tests results indicated that 95% kaolinite recovery was obtained using 2.0 × 10−4 mol/L BBDB at 25 ℃, which was half of the dosage when DDA obtained the maximum kaolinite recovery of 81%. At extremely low temperature (0 ℃), 3.0 × 10−4 mol/L BBDB could still collect 91% kaolinite, while DDA showed a frustrating ability. The contact angle tests indicated that BBDB could still significantly improve the hydrophobicity of the kaolinite surface (contact angle 71.7°) than DDA (contact angle only 25.8°) at 0 ℃. The Krafft point comparison tests indicated that BBDB had a much lower Krafft point (below 0 ℃) than DDA. Fourier transform infrared spectroscopy (FTIR)-spectrum analysis and zeta potential measurements showed that BBDB was physically adsorbed on the surface of kaolinite through electrostatic interaction.

BackgroundThere are trillions of microbiota in our intestinal tract, and they play a significant role in health and disease via interacting with the host in metabolic, immune, neural, and endocrine pathways. Over the past decades, numerous studies have been published in the field of gut microbiome and disease. Although there are narrative reviews of gut microbiome and certain diseases, the whole field is lack of systematic and quantitative analysis. Therefore, we outline research status of the gut microbiome and disease, and present insights into developments and characteristics of this field to provide a holistic grasp and future research directions.MethodsAn advanced search was carried out in the Web of Science Core Collection (WoSCC), basing on the term “gut microbiome” and its synonyms. The current status and developing trends of this scientific domain were evaluated by bibliometric methodology. CiteSpace was used to perform collaboration network analysis, co-citation analysis and citation burst detection.ResultsA total of 29,870 articles and 13,311 reviews were retrieved from the database, which involve 42,900 keywords, 176 countries/regions, 19,065 institutions, 147,225 authors and 4,251 journals. The gut microbiome and disease research is active and has received increasing attention. Co-cited reference analysis revealed the landmark articles in the field. The United States had the largest number of publications and close cooperation with other countries. The current research mainly focuses on gastrointestinal diseases, such as inflammatory bowel disease (IBD), ulcerative colitis (UC) and Crohn’s disease (CD), while extra-intestinal diseases are also rising, such as obesity, diabetes, cardiovascular disease, Alzheimer’s disease, Parkinson’s disease. Omics technologies, fecal microbiota transplantation (FMT) and metabolites linked to mechanism would be more concerned in the future.ConclusionThe gut microbiome and disease has been a booming field of research, and the trend is expected to continue. Overall, this research field shows a multitude of challenges and great opportunities.

Due to the increasing cases of bone damage and bone graft demand, bone-repair technology has great social and economic benefits and the manufacturing of artificial bone implants has become a focus in the domain of regenerative therapy. Considering that the traditional manufacturing process cannot effectively control the overall size of the scaffold, the diameter and shape of micropores, and the interoperability of micropores, 3D printing technology has emerged as a focal point of research within the realm of bone tissue engineering. However, the printing accuracy of extrusion-based biological 3D printing techniques is low. In this research, we utilized three-dimensional printing technology to develop high-precision magnesium-containing silicate (CSi-Mg) scaffolds. The precision of this innovative method was scrutinized and the influence of pore size on scaffold strength was systematically analyzed. Furthermore, the influence of the pore architecture on the sidewalls of these 3D-printed scaffolds was evaluated in terms of mechanical properties. The CSi-Mg scaffold, post a 3-week immersion in a simulated body of fluid, demonstrated a high modulus of elasticity (exceeding 404 MPa) and significant compressive strength (beyond 47 MPa). Furthermore, it exhibited commendable bioactivity and biodegradability. These results suggest that the high-precision 3D-printed CSi-Mg scaffolds hold great promise for addressing challenging bone defect cases.

Vibroseis trucks are in danger of rollover during the steering process, which severely threatens the safety of the drivers and the equipment. Therefore, it is important to examine approaches to increase the anti-rollover ability of vibroseis trucks. According to the structural characteristics and working field of the vibroseis truck, an analysis of the influence of different factors on its rollover stability was carried out. Then, a rollover warning index and an anti-rollover warning system were established, which can achieve four levels of judgment of different driving states. A simulation analysis of the anti-rollover warning system was conducted with MATLAB/Simulink software. The results showed that this warning system could accurately determine the driving state on a flat road and a sloping road and produce four types of sound–light alarm according to different rollover states. At the same time, the vibroseis truck showed poor roll stability in the slope steering process which is more prone to rollover accidents. This study establishes a foundation for further research on the design of warning systems not only for vibroseis trucks but also for other articulated trucks.

The two estrogen receptors ERα and ERβ are nuclear receptors that bind estrogen (E2) and function as ligand-inducible transcription factors. They are homologues and can form dimers with each other and bind to the same estrogen-response element motifs in the DNA. ERα drives breast cancer growth whereas ERβ has been reported to be anti-proliferative. However, they are rarely expressed in the same cells, and it is not fully investigated to which extent their functions are different because of inherent differences or because of different cellular context. To dissect their similarities and differences, we here generated a novel estrogen-dependent cell model where ERα homodimers can be directly compared to ERβ homodimers within the identical cellular context. By using CRISPR-cas9 to delete ERα in breast cancer MCF7 cells with Tet-Off-inducible ERβ expression, we generated MCF7 cells that express ERβ but not ERα. MCF7 (ERβ only) cells exhibited regulation of estrogen-responsive targets in a ligand-dependent manner. We demonstrated that either ER was required for MCF7 proliferation, but while E2 increased proliferation via ERα, it reduced proliferation through a G2/M arrest via ERβ. The two ERs also impacted migration differently. In absence of ligand, ERβ increased migration, but upon E2 treatment, ERβ reduced migration. E2 via ERα, on the other hand, had no significant impact on migration. RNA sequencing revealed that E2 regulated a transcriptome of around 800 genes via each receptor, but over half were specific for either ERα or ERβ (417 and 503 genes, respectively). Functional gene ontology enrichment analysis reinforced that E2 regulated cell proliferation in opposite directions depending on the ER, and that ERβ specifically impacted extracellular matrix organization. We corroborated that ERβ bound to cis-regulatory chromatin of its unique proposed migration-related direct targets ANXA9 and TFAP2C. In conclusion, we demonstrate that within the same cellular context, the two ERs regulate cell proliferation in the opposite manner, impact migration differently, and each receptor also regulates a distinct set of target genes in response to E2. The developed cell model provides a novel and valuable resource to further complement the mechanistic understanding of the two different ER isoforms.

ObjectiveEvidence regarding the relationship between the triglyceride-to-high-density lipoprotein cholesterol (TG/HDL-c) ratio and the risk of progression from prediabetes to diabetes remains limited. The purpose of our study was to investigate the relationship between the TG/HDL-C ratio and incident diabetes in prediabetic patients.MethodsThis retrospective cohort study covered 32 regions and 11 cities in China and consecutively and non-selectively collected data from 15,017 patients with prediabetes who had received a health check from 2010 to 2016. Data were obtained from the DATADRYAD database (www.datadryad.org). The Cox proportional-hazards regression model with cubic spline functions and smooth curve fitting (cubic spline smoothing) was used to explore the non-linear relationship between the baseline TG/HDL-c ratio and the risk of diabetes in patients with prediabetes. In addition, we performed a series of sensitivity and subgroup analyses.ResultsThe mean age of the included individuals was 50.95 ± 13.48 years, and 9,745 (64.51%) were men. The median (interquartile range) TG/HDL-c ratio was 1.09 (0.69–1.72). During a median follow-up time of 3.05 years, 1,731 (11.46%) patients had a final diagnosis of diabetes. The analysis after adjusting for covariates showed that the TG/HDL-c ratio was positively related to incident diabetes in patients with prediabetes (HR = 1.111, 95% CI 1.061–1.164). Participants with the highest TG/HDL-c ratio (Q4) had higher diabetes incidence rates than those with the lowest TG/HDL-c ratio (Q1) (P < 0.001 for the trend). There was a non-linear relationship between the TG/HDL-c ratio and the risk of diabetes, and the inflection point of the TG/HDL-c ratio was 1.415. The effect sizes (HR) on the left and right sides of the inflection point were 1.336 (95% CI: 1.134–1.573) and 1.055 (95% CI: 0.988–1.126), respectively. The sensitivity analysis demonstrated the robustness of these results.ConclusionThis study demonstrates a positive, non-linear relationship between the TG/HDL-c ratio and the risk of diabetes in Chinese patients with prediabetes. Aggressive intervention from a treatment perspective is required to lower the TG/HDL-c ratio below the inflection point (1.415) by lowering TG or increasing HDL-c levels.

Summary: Chromosome conformation capture combined with high-throughput sequencing (4C-seq) is a powerful tool to map genomic DNA regions that communicate with a specific locus of interest such as functional single-nucleotide polymorphism (SNPs)-containing regions. This protocol describes detailed steps to perform 4C-seq in mouse macrophage RAW264.7 cells, starting from the primer design based on cistrome and epigenome data, sample processing, and to the bioinformatics analysis.For complete details on the use and execution of this protocol, please refer to Huang et al. BackgroundAlthough congestive heart failure (CHF) is considered a risk factor for postoperative mortality, reliable quantification of the relationship between CHF and postoperative mortality risk is limited. We aimed to investigate the association between CHF and 1-year mortality after surgery in a large cohort of the Singaporean population.MethodsIn this retrospective cohort study, the study population included 69,032 adult patients who underwent surgery at Singapore General Hospital between 1 January 2012 and 31 October 2016. The target independent and dependent variables were CHF and 1-year mortality after surgery, respectively. Propensity score was estimated using a non-parsimonious multivariable logistic regression model. Multivariable adjustment, propensity score matching, propensity score adjustment, and propensity score-based weighting Cox proportional-hazards regression were performed to investigate the association between CHF and 1-year mortality after surgery.ResultsThe multivariate-adjusted hazard ratio (HR) in the original cohort was 1.39 (95% confidence interval (CI): 1.20–1.61, P < 0.001). In additional propensity score adjustment, the HR between CHF and 1-year mortality after surgery was 1.34 (95% CI: 1.15–1.56, P < 0.001). In the propensity score-matched cohort, the multivariate-adjusted Cox proportional hazard regression model analysis showed participants with CHF had a 54% increased risk of 1-year mortality after surgery (HR 1.54, 95% CI: 1.19–1.98, P < 0.001). The multivariate-adjusted HR of the inverse probability of treatment-weighted and standardised mortality ratio-weighted cohorts was 1.34 (95% CI: 1.10–1.62, P = 0.004) and 1.24 (95% CI: 1.17–1.32, P < 0.001), respectively.ConclusionCHF is an independent risk factor for 1-year mortality after surgery in patients undergoing surgery. Depending on the statistical method, patients with CHF had a 24–54% increased risk of 1-year all-cause mortality after surgery. This provides a reference for optimising clinical decision-making, improving preoperative consultation, and promoting clinical communication.

Circular RNAs (circRNAs), which are produced post-splicing of pre-mRNAs, are strongly linked to the emergence of several tumor types. The initial stage in conducting follow-up studies involves identifying circRNAs. Currently, animals are the primary target of most established circRNA recognition technologies. However, the sequence features of plant circRNAs differ from those of animal circRNAs, making it impossible to detect plant circRNAs. For example, there are non-GT/AG splicing signals at circRNA junction sites and few reverse complementary sequences and repetitive elements in the flanking intron sequences of plant circRNAs. In addition, there have been few studies on circRNAs in plants, and thus it is urgent to create a plant-specific method for identifying circRNAs. In this study, we propose CircPCBL, a deep-learning approach that only uses raw sequences to distinguish between circRNAs found in plants and other lncRNAs. CircPCBL comprises two separate detectors: a CNN-BiGRU detector and a GLT detector. The CNN-BiGRU detector takes in the one-hot encoding of the RNA sequence as the input, while the GLT detector uses k-mer (k = 1 − 4) features. The output matrices of the two submodels are then concatenated and ultimately pass through a fully connected layer to produce the final output. To verify the generalization performance of the model, we evaluated CircPCBL using several datasets, and the results revealed that it had an F1 of 85.40% on the validation dataset composed of six different plants species and 85.88%, 75.87%, and 86.83% on the three cross-species independent test sets composed of Cucumis sativus, Populus trichocarpa, and Gossypium raimondii, respectively. With an accuracy of 90.9% and 90%, respectively, CircPCBL successfully predicted ten of the eleven circRNAs of experimentally reported Poncirus trifoliata and nine of the ten lncRNAs of rice on the real set. CircPCBL could potentially contribute to the identification of circRNAs in plants. In addition, it is remarkable that CircPCBL also achieved an average accuracy of 94.08% on the human datasets, which is also an excellent result, implying its potential application in animal datasets. Ultimately, CircPCBL is available as a web server, from which the data and source code can also be downloaded free of charge.

In order to achieve solid-liquid separation and recycling of drilling fluid during petroleum drilling, a new type of solid-liquid separation device was designed and developed to achieve high-efficiency solid-liquid separation and low cost. The working principle of the equipment was introduced, the mechanical model was established, and the sieve flow of drilling fluid and the movement of wet particles in the real situation were analyzed. The relationship between the processing capacity of the solid-liquid separator and the speed of the solid phase in the drilling fluid and the speed of the excitation motor and the rotary drum were given. Multi-objective evolutionary algorithm was used to solve the multi-objective optimization model. Two typical evolutionary algorithms NSGA-II and MOEA/D were analyzed. An algorithm that solves this problem was selected and improved. A series of optimal solution sets for this problem were obtained by using the improved multi-objective evolutionary algorithm. Finally, based on the experimental prototype of the solid-liquid separator, a functional experiment was performed on the basis of the calculated optimal solution set. The experimental results show the feasibility of the multi-objective optimization model and algorithm.

Spatiotemporal fusion is an effective and cost-effective method to obtain both high temporal resolution and high spatial resolution images. However, existing methods do not sufficiently extract the deeper features of the image, resulting in fused images which do not recover good topographic detail and poor fusion quality. In order to obtain higher quality spatiotemporal fusion images, a novel spatiotemporal fusion method based on deep learning is proposed in this paper. The method combines an attention mechanism and a multiscale feature fusion network to design a network that more scientifically explores deeper features of the image for different input image characteristics. Specifically, a multiscale feature fusion module is introduced into the spatiotemporal fusion task and combined with an efficient spatial-channel attention module to improve the capture of spatial and channel information while obtaining more effective information. In addition, we design a new edge loss function and incorporate it into the compound loss function, which helps to generate fused images with richer edge information. In terms of both index performance and image details, our proposed model has excellent results on both datasets compared with the current mainstream spatiotemporal fusion methods.

Resilient manufacturing is a vision in the Industry 5.0 blueprint for satisfying sustainable development goals under pandemics or the rising individualized product needs. A resilient manufacturing strategy based on the Industrial Internet of Things (IIoT) networks plays an essential role in facilitating production and supply chain recovery. IIoT contains confidential data and private information, and many security issues arise through vulnerabilities in the infrastructure. The traditional centralized IIoT framework is not only of high cost for system configuration but also vulnerable to cyber-attacks and single-point failure, which is not suitable for achieving the resilient manufacturing vision in Industry 5.0. Recently, researchers are seeking a secure solution of middleware based on blockchain technology integration for decentralized IIoT, which can effectively protect the consistency, integrity, and availability of IIoT data by utilizing the auditing and tamper-proof features of the blockchain. This paper presented a review of secure blockchain middleware for decentralized IIoT towards Industry 5.0. Firstly, the security issues of conventional IIoT solutions and the advantages of blockchain middleware are analyzed. Secondly, an architecture of secure blockchain middleware for decentralized IIoT is proposed. Finally, enabling technologies, challenges, and future directions are reviewed. The innovation of this paper is to study and discuss the distributed blockchain middleware, investigating its ability to eliminate the risk of a single point of failure via a distributed feature in the context of resilient manufacturing in Industry 5.0 and to solve the security issues from traditional centralized IIoT. Also, the four-layer architecture of blockchain middleware presented based on the IIoT application framework is a novel aspect of this review. It is expected that the paper lays a solid foundation for making IIoT blockchain middleware a new venue for Industry 5.0 research.

Kick is one of the most important drilling problems, and because its occurrence makes drilling engineering extremely complex, it is essential to predict the possibility of kick as soon as possible. In this study, k-means clustering was combined with four artificial neural networks: regularized RBFNN, generalized RBFNN, GRNN, and PNN, to estimate the kick risk. To reduce data redundancy and normalize the drilling data, which contain kick conditions, k-means clustering was introduced. The output layer weights were then determined using a brute-force search with different Gaussian function widths, resulting in a series of artificial neural networks composed of different clustering samples and different Gaussian function widths. The results showed that the prediction accuracy of regularized RBFNN + k-means model was the highest, that of the GRNN + k-means model was the lowest. The kick prediction accuracy for regularized RBFNN, generalized RBFNN, GRNN, and PNN were 75.90%, 65.20%, 51.70%, and 70.16%, respectively. This method can be used to enhance the speed and accuracy of kick risk prediction in the field while facilitating the use of and advances in risk warning technology for deep and high-temperature and high-pressure wells.

Accurate information about the distribution of natural fractures is a key factor for the success of the exploration and development of oil and gas in carbonate rock formations. Forward modeling of natural fractures generated by tectonic movement within carbonate rock formations was investigated by jointly using the continuum damage model and finite element numerical technology. Geological analysis of natural fractures was used as the basis of the geomechanical finite element calculation. A workflow of numerical calculations for natural fractures was proposed. These achievements were applied to investigate natural fractures’ distribution within Ordovician carbonate rock formations of the Fuman Oilfield, Xinjiang, in the west of China. Finite element sub-modeling technology was used to further investigate natural fractures within key target reservoir formations with a finer mesh. The contour of natural fractures represented by the localization band of continuum damage variables was obtained. A comparison of the numerical results of the natural fractures’ distribution represented by continuum damage variables with those of natural fractures interpreted from seismic data shows that: (1) the numerical solution of natural fractures matches the measured data, and their orientations are in good accordance; (2) their distribution and locations are basically the same, with some small differences in local details; (3) the numerical results indicate that the maximum value of the damage variable SDEG within the zones of natural fractures is 0.2686, and the widths of the bands of natural fractures/faults are in the range of 500 m to 1000 m. Validation of the results of the distribution of natural fractures was performed indirectly via the distribution of the minimum horizontal stress gradient ShG.

Salmonella is a leading cause of foodborne pathogen which causes intestinal and systemic diseases across the world. Vaccination is the most effective protection against Salmonella, but the identification and design of an effective broad-spectrum vaccine is still a great challenge, because of the multi-serotypes of Salmonella. Reverse vaccinology is a new tool to discovery and design vaccine antigens combining human immunology, structural biology and computational biology with microbial genomics. In this study, reverse vaccinology, an in-silico approach was established to screen appropriate immunogen targets by calculating the immunogenicity score of 583 non-redundant outer membrane and secreted proteins of Salmonella. Herein among 100 proteins identified with top-ranked scores, 15 representative antigens were selected randomly. Applying the sequence conservation test, four proteins (FliK, BcsZ, FhuA and FepA) remained as potential vaccine candidates for in vivo evaluation of immunogenicity and immunoprotection. All four candidates were capable to trigger the immune response and stimulate the production of antiserum in mice. Furthermore, top-ranked proteins including FliK and BcsZ provided wide antigenic coverage among the multi-serotype of Salmonella. The S. Typhimurium LT2 challenge model used in mice immunized with FliK and BcsZ showed a high relative percentage survival (RPS) of 52.74 % and 64.71 % respectively. In conclusion, this study constructed an in-silico pipeline able to successfully pre-screen the vaccine targets characterized by high immunogenicity and protective immunity. We show that reverse vaccinology allowed screening of appropriate broad-spectrum vaccines for Salmonella.
