Abstract Differential Evolution (DE) stands as a potent global optimization algorithm, renowned for its application in addressing a myriad of practical engineering issues. The efficacy of DE is profoundly influenced by its control parameters and mutation strategies. In light of this, we introduce a refined DE algorithm characterized by adaptive parameters and dual mutation strategies (APDSDE). APDSDE inaugurates an adaptive switching mechanism that alternates between two innovative mutation strategies: DE/current-to-pBest-w/1 and DE/current-to-Amean-w/1. Furthermore, a novel parameter adaptation technique rooted in cosine similarity is established, with the derivation of explicit calculation formulas for both the scaling factor weight and crossover rate weight. In pursuit of optimizing convergence speed whilst preserving population diversity, a sophisticated nonlinear population size reduction method is proposed. The robustness of each algorithm is rigorously evaluated against the CEC2017 benchmark functions, with empirical evidence underscoring the superior performance of APDSDE in comparison to a host of advanced DE variants.

Abstract Background There is increasing evidence that gut fungi dysbiosis plays a crucial role in the development and progression of colorectal cancer (CRC). It has been reported that gut fungi exacerbate the severity of CRC by regulating tumor immunity. Our previous studies have shown that the opportunistic pathogenic fungal pathogen, Candida tropicalis (C. tropicalis) promotes CRC progression by enhancing the immunosuppressive function of MDSCs and activating the NLRP3 inflammasome of MDSCs. However, the relationship between IL-1β produced by NLRP3 inflammasome activation and the immunosuppressive function of MDSCs enhanced by C. tropicalis in CRC remains unclear. Methods The TCGA database was used to analyze the relationship between IL-1β and genes related to immunosuppressive function of MDSCs in human CRC. The expression of IL-1β in human CRC tissues was detected by immunofluorescence staining. The proteomic analysis was performed on the culture supernatant of C. tropicalis-stimulated MDSCs. The experiments of supplementing and blocking IL-1β as well as inhibiting the NLRP3 inflammasome activation were conducted. A mouse colon cancer xenograft model was established by using MC38 colon cancer cell line. Results Analysis of CRC clinical samples showed that the high expression of IL-1β was closely related to the immunosuppressive function of tumor-infiltrated MDSCs. The results of in vitro experiments revealed that IL-1β was the most secreted cytokine of MDSCs stimulated by C. tropicalis. In vitro supplementation of IL-1β further enhanced the immunosuppressive function of C. tropicalis-stimulated MDSCs and NLRP3-IL-1β axis mediated the immunosuppressive function of MDSCs enhanced by C. tropicalis. Finally, blockade of IL-1β secreted by MDSCs augmented antitumor immunity and mitigated C. tropicalis-associated colon cancer. Conclusions C. tropicalis promotes excessive secretion of IL-1β from MDSCs via the NLRP3 inflammasome. IL-1β further enhances the immunosuppressive function of MDSCs to inhibit antitumor immunity, thus promoting the progression of CRC. Therefore, targeting IL-1β secreted by MDSCs may be a potential immunotherapeutic strategy for the treatment of CRC.

Abstract Hydrochar from waste biomass is a promising material for removing emerging contaminants (e.g., antibiotics) in water/soil environment. Abundant small-sized hydrochar particles (HPs) with a high content of reactive functional groups and high mobility are easily released into ecosystems through hydrochar applications. However, the photodegradation ability and corresponding structures of HPs are largely unknown, which hinder accurate estimation of the remediation effect of hydrochar in ecosystems. Herein, photodegradation performance of HP towards targeted norfloxacin (NOR, a typical antibiotic) under light irradiation (visible and UV light) were investigated after adsorption processes upon release into soil/water, and its reactive species and photoactive structures were clarified and compared with those of residual bulk hydrochar (BH) comprehensively. The results showed that: (1) photodegradation percentages of HPs were 4.02 and 4.48 times higher than those of BHs under UV and visible light, in which reactive species of both HPs and BHs were ·OH and ·O2 −; (2) density functional theory (DFT) results identified that the main photoactive structure of graphitic-N decreased the energy gap (Eg) of HPs, and C=O, COOH groups improved electron donating ability of BHs; (3) well-developed graphitization structure of HP resulted from higher polymerization reaction was an significant photoactive structure involving its superior photodegradation ability relative to that of BH. The distinct heterogeneities of photodegradation ability in HP and BH and underlying photoactive structures provide an in-depth understanding of hydrochar application for removing emerging contaminants in soil/water environment. Identifying photoactive structures is helpful to predict photodegradation ability of hydrochar according to their abundance. Graphical Abstract

Abstract Background It has been proposed that anti-angiogenesis therapy could induce tumor "vascular normalization" and further enhance the efficacy of chemotherapy, radiotherapy, target therapy, and immunotherapy for nearly twenty years. However, the detailed molecular mechanism of this phenomenon is still obscure. Method Overexpression and knockout of CCL28 in human lung adenocarcinoma cell line A549 and murine lung adenocarcinoma cell line LLC, respectively, were utilized to establish mouse models. Single-cell sequencing was performed to analyze the proportion of different cell clusters and metabolic changes in the tumor microenvironment (TME). Immunofluorescence and multiplex immunohistochemistry were conducted in murine tumor tissues and clinical biopsy samples to assess the percentage of pericytes coverage. Primary pericytes were isolated from lung adenocarcinoma tumor tissues using magnetic-activated cell sorting (MACS). These pericytes were then treated with recombinant human CCL28 protein, followed by transwell migration assays and RNA sequencing analysis. Changes in the secretome and metabolome were examined, and verification of retinoic acid metabolism alterations in pericytes was conducted using quantitative real-time PCR, western blotting, and LC–MS technology. Chromatin immunoprecipitation followed by quantitative PCR (ChIP-qPCR) was employed to validate the transcriptional regulatory ability and affinity of RXRα to specific sites at the ANGPT1 promoter. Results Our study showed that after undergoing anti-angiogenesis treatment, the tumor exhibited a state of ischemia and hypoxia, leading to an upregulation in the expression of CCL28 in hypoxic lung adenocarcinoma cells by the hypoxia-sensitive transcription factor CEBPB. Increased CCL28 could promote tumor vascular normalization through recruiting and metabolic reprogramming pericytes in the tumor microenvironment. Mechanistically, CCL28 modified the retinoic acid (RA) metabolism and increased ANGPT1 expression via RXRα in pericytes, thereby enhancing the stability of endothelial cells. Conclusion We reported the details of the molecular mechanisms of "vascular normalization" after anti-angiogenesis therapy for the first time. Our work might provide a prospective molecular marker for guiding the clinical arrangement of combination therapy between anti-angiogenesis treatment and other therapies.

Objective: This study aimed to compare the effects of high-intensity interval training (HIIT) with work-to-rest ratios of 2:1 and 3:1 on physical performance and physiological responses in male college judo athletes. Methods: Forty-eight male college judo athletes were randomly assigned to one of three groups. One used HIIT with a 2:1 work-to-rest ratio (2:1 group), another used HIIT with a 3:1 work-to-rest ratio (3:1 group), while a control group (CG) continued its regular training program as before. Subjects completed two training sessions per week for 6 weeks. The participants completed the Modified Agility T-test, 30-m sprint, medicine ball put, countermovement jump, 20-m multistage shuttle run test, and 30-s Wingate test before and after the training, and blood lactate measurements at 3, 5, 10, and 15 min after the 30-s Wingate test were taken before and after the training. Results: The 2:1 group (p = 0.010, d = 0.88 (large)) and 3:1 group (p = 0.002, d = 0.46 (small)) showed significant improvements in VO2max estimated by the 20-m multistage shuttle run test in comparison with pre-test. The 3:1 group had a significant difference in agility in comparison with the CG post-training (p = 0.004). Peak power (PP) and relative peak power (PP/kg) significantly improved in the 3:1 group (p < 0.05) compared to the pre-test. Blood lactate values at the fifth minute (p = 0.029) after the Wingate test in the 3:1 group were significantly decreased compared to the pre-test. Conclusion: Our findings suggest that HIIT with the 3:1 and 2:1 work-to-rest ratios can improve aerobic power, and HIIT with a 3:1 work-to-rest ratio can improve absolute and relative peak power.

Abstract Objectives The purpose of our study is to further understanding of the depression symptoms of HIV/AIDS patients in Guilin, Guangxi via exploring whether there is a mediating effect of sleep quality on medical-social support and depression symptoms and therefore provide a theoretical basis for application of medical-social support to alleviate depression symptoms of HIV/AIDS patients. Methods A convenience sampling method was used to select 200 HIV/AIDS patients for the study. Depression symptoms, sleep quality, and medical-social support of the study participants were investigated using The Center for Epidemiological Studies Depression Scale (CES-D), The Pittsburg Sleep Quality Index (PSQI), and The Medical Outcomes Study Social Support Survey (MOS-SSS), respectively. Predictors of depression symptoms were explored by multiple linear regression, and Pearson correlation was used to analyze the relationship between sleep quality, medical-social support, and depression symptoms. Mediating effect analysis was performed by nonparametric Bootstrap test. Results In this study, the incidence of depression symptoms was 54.4%. Abstract Background Exposure to heavy metals alone or in combination can promote systemic inflammation. The aim of this study was to investigate potential associations between multiple plasma heavy metals and markers of systemic immune inflammation. Methods Using a cross-sectional study, routine blood tests were performed on 3355 participants in Guangxi, China. Eight heavy metal elements in plasma were determined by inductively coupled plasma mass spectrometry. Immunoinflammatory markers were calculated based on peripheral blood WBC and its subtype counts. A generalised linear regression model was used to analyse the association of each metal with the immunoinflammatory markers, and the association of the metal mixtures with the immunoinflammatory markers was further assessed using weighted quantile sum (WQS) regression. Results In the single-metal model, plasma metal Fe (log10) was significantly negatively correlated with the levels of immune-inflammatory markers SII, NLR and PLR, and plasma metal Cu (log10) was significantly positively correlated with the levels of immune-inflammatory markers SII and PLR. In addition, plasma metal Mn (log10 conversion) was positively correlated with the levels of immune inflammatory markers NLR and PLR. The above associations remained after multiple corrections. In the mixed-metal model, after WQS regression analysis, plasma metal Cu was found to have the greatest weight in the positive effects of metal mixtures on SII and PLR, while plasma metals Mn and Fe had the greatest weight in the positive effects of metal mixtures on NLR and LMR, respectively. In addition, blood Fe had the greatest weight in the negative effects of the metal mixtures for SII, PLR and NLR. Conclusion Plasma metals Cu and Mn were positively correlated with immunoinflammatory markers SII, NLR and PLR. While plasma metal Fe was negatively correlated with immunoinflammatory markers SII, NLR, and PLR.

Abstract Exciton–polaritons (polaritons) resulting from the strong exciton–photon interaction stimulates the development of novel low-threshold coherent light sources to circumvent the ever-increasing energy demands of optical communications1–3. Polaritons from bound states in the continuum (BICs) are promising for Bose–Einstein condensation owing to their theoretically infinite quality factors, which provide prolonged lifetimes and benefit the polariton accumulations4–7. However, BIC polariton condensation remains limited to cryogenic temperatures ascribed to the small exciton binding energies of conventional material platforms. Herein, we demonstrated room-temperature BIC polariton condensation in perovskite photonic crystal lattices. BIC polariton condensation was demonstrated at the vicinity of the saddle point of polariton dispersion that generates directional vortex beam emission with long-range coherence. We also explore the peculiar switching effect among the miniaturized BIC polariton modes through effective polariton−polariton scattering. Our work paves the way for the practical implementation of BIC polariton condensates for integrated photonic and topological circuits.

Abstract Introduction Bipolar disorder (BD) is a chronic psychiatric disorder that combines hypomania or mania and depression. The study aims to investigate the research areas associated with cognitive function in bipolar disorder and identify current research hotspots and frontier areas in this field. Methodology Publications related to cognitive function in BD from 2012 to 2022 were searched on the Web of Science Core Collection (WoSCC) database. VOSviewer, CiteSpace, and Scimago Graphica were used to conduct this bibliometric analysis. Results A total of 989 articles on cognitive function in BD were included in this review. These articles were mainly from the United States, China, Canada, Spain and the United Kingdom. Our results showed that the journal “Journal of Affective Disorders” published the most articles. Apart from “Biploar disorder” and “cognitive function”, the terms “Schizophrenia”, “Meta analysis”, “Rating scale” were also the most frequently used keywords. The research on cognitive function in bipolar disorder primarily focused on the following aspects: subgroup, individual, validation and pathophysiology. Conclusions The current concerns and hotspots in the filed are: “neurocognitive impairment”, “subgroup”, “1st degree relative”, “mania”, “individual” and “validation”. Future research is likely to focus on the following four themes: “Studies of the bipolar disorder and cognitive subgroups”, “intra-individual variability”, “Validation of cognitive function tool” and “Combined with pathology or other fields”.

Abstract Background Caspase Recruitment Domain-containing protein 9 (CARD9) expressed in myeloid cells has been demonstrated to play an antifungal immunity role in protecting against disseminated candidiasis. Hereditary CARD9 ablation leads to fatal disseminated candidiasis. However, the myeloid cell types and molecular mechanisms implicated in CARD9 protecting against disseminated candidiasis remain wholly elusive. Methods The role of CARD9 ablation in exacerbating disseminated candidiasis was determined in vivo and in vitro. The molecular mechanism by which CARD9 ablation promotes acute kidney injury in disseminated candidiasis was identified by RNA-sequencing analysis. The expression of mitochondrial proteins and ferroptosis-associated proteins were measured by Quantitative real-time PCR and western blot. Results CARD9 ablation resulted in a reduced proportion of myeloid-derived suppressor cells (MDSCs) and a substantially lower expression of solute carrier family 7 member 11 (SLC7A11) in the kidneys, which increased susceptibility to acute kidney injury and renal ferroptosis during disseminated Candida tropicalis (C. tropicalis) infection. Moreover, CARD9-deficient MDSCs were susceptible to ferroptosis upon stimulation with C. tropicalis, which was attributed to augmented mitochondrial oxidative phosphorylation (OXPHOS) caused by reduced SLC7A11 expression. Mechanistically, C-type lectin receptors (CLRs)-mediated recognition of C. tropicalis promoted the expression of SLC7A11 which was transcriptionally manipulated by the Syk-PKCδ-CARD9-FosB signaling axis in MDSCs. FosB enhanced SLC7A11 transcription by binding to the promoter of SLC7A11 in MDSCs stimulated with C. tropicalis. Mitochondrial OXPHOS, which was negatively regulated by SLC7A11, was responsible for inducing ferroptosis of MDSCs upon C. tropicalis stimulation. Finally, pharmacological inhibition of mitochondrial OXPHOS or ferroptosis significantly increased the number of MDSCs in the kidneys to augment host antifungal immunity, thereby attenuating ferroptosis and acute kidney injury exacerbated by CARD9 ablation during disseminated candidiasis. Conclusions Collectively, our findings show that CARD9 ablation enhances mitochondria-mediated ferroptosis in MDSCs, which negatively regulates antifungal immunity. We also identify mitochondria-mediated ferroptosis in MDSCs as a new molecular mechanism of CARD9 ablation-exacerbated acute kidney injury during disseminated candidiasis, thus targeting mitochondria-mediated ferroptosis is a novel therapeutic strategy for acute kidney injury in disseminated candidiasis.

Abstract Image inpainting aims to repair the damaged region according to the known content in the damaged image. Recently, image inpainting methods have poor effects on high‐resolution damaged images, and the research on the inpainting of large‐area damaged images is limited. Therefore, this paper proposes an image inpainting method based on Generative Adversarial Networks (GAN) inversion and autoencoder. This work consists of two phases: first, the authors design an autoencoder‐based GAN, which learns the mapping from noise to low‐dimensional feature maps by training a generator, and then converts the generated feature maps into high‐resolution images. Thus, the difficulty of learning the mapping relationship is reduced. Second, the authors adopt the learning‐based GAN inversion to infer the closest latent code. The trained GAN is then used to reconstruct the complete image. Finally, the authors compare their method with other classical methods on the CelebAMask‐HQ, Flickr‐Faces‐HQ, and ImageNet datasets. According to the quantitative comparison, when the mask range is large, in other words, when the image has a large area of damage, the authors’ method is superior to the comparison methods. According to the qualitative comparison, the structure of the high‐resolution image inpainted by the authors’ method is more reasonable and the texture details are more realistic.

Underground small indoor gymnasiums (USIG) are important public places, it is vital to design and build a very economical and efficient ventilation system for effective closed-loop regulation of temperature and gases concentration at prescribed levels. In the article, the model-based prototype design, establishment and operation were proposed and applied to closed-loop control system of the underground small indoor gymnasiums’ ventilation system (USIGVS). First of all, the extended Multiphysics model was developed through feedback connecting the 3D Multiphysics model of air flow rate, temperature, O2 and CO2 concentration with a 0D proportional-integral-derivative (PID) controller via Neumann boundary condition, hence a close-loop USIGVS was constructed for feedback control of temperature and gases concentration in ping-pong USIG. Simultaneously, a cost function sufficiently representing the design requirement was formulated. Then global parameter sensitivity analysis (GPSA) was applied for sensitivity ranking of parameters including geometric parameters of USIGVS and tunable parameters of PID controller. The GPSA proved that sensitivity ordering of the cost function to each parameter was proportional gain (kp) > derivative gain (kd) > distance from left inlet to bottom (r) > distance from outlet pipe to bottom (d) > integrative gain (ki) > distance from upper inlet pipe to left (h), respectively, and the kp, kd and r was the parameter influencing the cost function the most. The optimal parameters determined by both GPSA and response optimization were kp = 3.17 m4 mol−1 s−1, kd = 1.49 m4 mol−1, r = 2.04 m, d = 3.12 m, ki = 0.37 m4 mol−1 s−2 and h = 3.85 m. Finally, the closed-loop USIGVS prototype with optimal parameters was designed and established through real-time simulation. The real-time operation confirmed that the temperature and gases concentrations were robust maintained at prescribed levels with desired dynamic response characteristics and lower power consumption, and the expected requirements were achieved for the design, establishment and operation of closed-loop USIGVS control system prototype.

Summary: Natural silks are renewable proteins with impressive mechanical properties and biocompatibility that are useful in various fields. However, the cellular and spatial organization of silk-secreting organs remains unclear. Here, we combined single-nucleus and spatially resolved transcriptomics to systematically map the cellular and spatial composition of the silk glands (SGs) of mulberry silkworms late in larval development. This approach allowed us to profile SG cell types and cell state dynamics and identify regulatory networks and cell-cell communication related to efficient silk protein synthesis; key markers were validated via transgenic approaches. Notably, we demonstrated the indispensable role of the ecdysone receptor (ultraspiracle) in regulating endoreplication in SG cells. Our atlas presents the results of spatiotemporal analysis of silk-secreting organ architecture late in larval development; this atlas provides a valuable reference for elucidating the mechanism of efficient silk protein synthesis and developing sustainable products made from natural silk.

Background: For adolescent soccer players, good sprinting and jumping abilities are crucial for their athletic performance. The application of plyometric training on boosting explosive strength in adolescent soccer players is contingent upon the maturation phase, which can mediate the training-induced adaptations. Purpose: This systematic review and meta-analysis aim to explore the maturation effect of plyometric training on the lower limb explosive power of adolescent soccer players, with vertical countermovement jump (CMJ) and 20-m sprint as the main outcome indicators. Methods: An extensive search of the literature was carried out on various databases including PubMed, Web of Science, Scopus, ProQuest, and the China National Knowledge Infrastructure (CNKI), covering the time period from the establishment of each database to February 6, 2023. The search was conducted using English keywords such as ‘Plyometric,’ ‘Adolescent,’ ‘football,’ and ‘Explosive strength.’ This study utilized the Cochrane risk of bias assessment tool to conduct a standardized quality evaluation of all the included literature. Additionally, the Review Manager 5.4 software was employed to perform data analysis on all the extracted data. Results: A total of 17 studies involving 681 adolescent soccer players aged 10 to 19 were included. Purpose: To date, the relationship between Growth Differentiation Factor 15 (GDF-15) gene polymorphism and the risk of type 2 diabetes mellitus (T2DM) has not been clarified. Our study aims to explore the association between serum GDF-15 levels and related gene polymorphism with the risk of T2DM in a Chinese rural Yao population. Methods: This was a 1:1 case-control study with 179 T2DM patients and 179 age- and sex-matched control participants. Serum GDF-15 levels were measured by enzyme-linked immunosorbent assay, and polymorphisms (rs1059519, rs1059369, rs1804826 and rs1054564) were genotyped by MassArray mass spectrometry. IntroductionThere exists a noticeable gap between consumers' willingness to purchase green food and their actual purchase behavior. However, the awareness of green development is a crucial factor influencing this purchase behavior and acts as an internal driving force promoting green consumption. Consumers' green development awareness is shaped by various psychological motivations, including environmental concern, health consciousness, knowledge, norms, and price considerations. The existing literature often focuses on specific regions or groups, lacking comprehensive cross-regional and multivariate evaluations, and frequently overlooks the potential impact of moderating variables such as economic development level, product type, and behavior type.MethodsTo clarify the overall effect of each motivational factor on green food purchase behavior, this study conducted a meta-analysis. We selected eight causal variables and three moderating variables that significantly influence consumers' green food purchase behavior. The analysis included 132 independent effect values from 45 research papers.ResultsThe meta-analysis revealed that: Consumers' green food purchase behavior is significantly positively correlated with eight motivational factors: environmental awareness, health awareness, green attitude, green knowledge, subjective norms, price awareness, perceived behavior control, and perceived usefulness. Economic development level, product type, and behavior type significantly affect consumers' green food purchase behavior. The impact of motivational factors on actual purchase behavior is weaker than on purchase intention, suggesting that interventions should focus on converting purchase intentions into actual purchase behavior. The findings indicate that environmental responsibility, government policies, and marketing strategies can influence consumers' psychological motives, guiding them toward more responsible consumption choices.DiscussionEnhancing consumers' environmental and health awareness is essential, and policy support and marketing strategies can effectively promote green food consumption. These insights underscore the importance of targeted interventions to bridge the gap between green purchase intention and behavior.

Perfluorobutanesulfonic acid (PFBS), a short-chain alternative to perfluorooctanesulfonic acid (PFOS), is widely used in various products and is increasingly present in environmental media and human bodies. Recent epidemiological findings have raised concerns about its potential adverse health effects, although the specific toxic mechanism remains unclear. This study aimed to investigate the metabolic toxicity of gestational PFBS exposure in maternal rats. Pregnant Sprague Dawley (SD) rats were randomly assigned to three groups and administered either 3% starch gel (control), 5, or 50 mg/kg bw·d PFBS. Oral glucose tolerance tests (OGTT) and lipid profiles were measured, and integrated omics analysis (transcriptomics and non-targeted metabolomics) was employed to identify changes in genes and metabolites and their relationships with metabolic phenotypes. The results revealed that rats exposed to 50 mg/kg bw·d PFBS exhibited a significant decrease in 1-h glucose levels and the area under the curve (AUC) of OGTT compared with the starch group. Transcriptomics analysis indicated significant alterations in gene expression related to cytochrome P450 exogenous metabolism, glutathione metabolism, bile acid secretion, tumor pathways, and retinol metabolism. Differentially expressed metabolites (DEMs) were enriched in pathways such as pyruvate metabolism, the glucagon signaling pathway, central carbon metabolism in cancer, and the citric acid cycle. Co-enrichment analysis and pairwise correlation analysis among genes, metabolites, and outcomes identified several differentially expressed genes (DEGs), including Gstm1, Kit, Adcy1, Gck, Ppp1r3c, Ppp1r3d, and DEMs such as fumaric acid, L-lactic acid, 4-hydroxynonenal, and acetylvalerenolic acid. These DEGs and DEMs may play a role in the modulation of glucolipid metabolic pathways. In conclusion, our results suggest that gestational exposure to PFBS may induce molecular perturbations in glucose homeostasis. These findings provide insights into the potential mechanisms contributing to the heightened risk of abnormal glucose tolerance associated with PFBS exposure.

Abstract Immunotherapy, including immune checkpoint inhibitors and adoptive cell transfer, has obtained great progress, but their efficiencies vary among patients due to the genetic and epigenetic differences. Human MEX3B (hMEX3B) protein is an RNA-binding protein that contains two KH domains at the N-terminus and a RING domain at its C-terminus, which has the activity of E3 ubiquitin ligase and is essential for RNA degradation. Current evidence suggests that hMEX3B is involved in many important biological processes, including tumor immune evasion and HLA-A regulation, but the sequence of substrate RNA recognized by hMEX3B and the functional molecular mechanisms are unclear. Here, we first screened the optimized hMEX3B binding sequence on the HLA-A mRNA and reported that the two tandem KH domains can bind with their substrate one hundred times more than the individual KH domains. We systematically investigated the binding characteristics between the two KH domains and their RNA substrates by nuclear magnetic resonance (NMR). Based on this information and the small-angle X-ray scattering (SAXS) data, we used molecular dynamics simulations to obtain structural models of KH domains in complex with their corresponding RNAs. By analyzing the models, we noticed that on the KH domains’ variable loops, there were two pairs of threonines and arginines that can disrupt the recognition of the RNA completely, and this influence had also been verified both in vitro and in vivo. Finally, we presented a functional model of the hMEX3B protein, which indicated that hMEX3B regulated the degradation of its substrate mRNAs in many biological processes. Taken together, our research illustrated how the hMEX3B protein played a key role in translation inhibition during the immune response to tumor cells and provided an idea and a lead for the study of the molecular mechanism and function of other MEX3 family proteins.

Abstract Background Renal cell carcinoma (RCC) is one of the most common malignant tumor worldwide. Metastasis is a leading case of cancer-related deaths of RCC. Circular RNAs (circRNAs), a class of noncoding RNAs, have emerged as important regulators in cancer metastasis. However, the functional effects and regulatory mechanisms of circRNAs on RCC metastasis remain largely unknown. Methods High-throughput RNA sequencing techniques were performed to analyze the expression profiles of circRNAs and mRNAs in highly and poorly invasive clear cell renal cell carcinoma (ccRCC) cell lines. Functional experiments were performed to unveil the regulatory role of circPPAP2B in the proliferation and metastatic capabilities of ccRCC cells. RNA pulldown, Mass spectrometry analysis, RNA methylation immunoprecipitation (MeRIP), RNA immunoprecipitation (RIP), co-immunoprecipitation (CoIP), next-generation RNA-sequencing and double luciferase experiments were employed to clarify the molecular mechanisms by which circPPAP2B promotes ccRCC metastasis. Results In this study, we describe a newly identified circular RNA called circPPAP2B, which is overexpressed in highly invasive ccRCC cells, as determined through advanced high-throughput RNA sequencing techniques. Furthermore, we observed elevated circPPAP2B in ccRCC tissues, particularly in metastatic ccRCC tissues, and found it to be associated with poor prognosis. Functional experiments unveiled that circPPAP2B actively stimulates the proliferation and metastatic capabilities of ccRCC cells. Mechanistically, circPPAP2B interacts with HNRNPC in a m6A-dependent manner to facilitate HNRNPC nuclear translocation. Subcellular relocalization was dependent upon nondegradable ubiquitination of HNRNPC and stabilization of an HNRNPC/Vimentin/Importin α7 ternary complex. Moreover, we found that circPPAP2B modulates the interaction between HNRNPC and splicing factors, PTBP1 and HNPNPK, and regulates pre-mRNA alternative splicing. Finally, our studies demonstrate that circPPAP2B functions as a miRNA sponge to directly bind to miR-182-5p and increase CYP1B1 expression in ccRCC. Conclusions Collectively, our study provides comprehensive evidence that circPPAP2B promotes proliferation and metastasis of ccRCC via HNRNPC-dependent alternative splicing and miR-182-5p/CYP1B1 axis and highlights circPPAP2B as a potential therapeutic target for ccRCC intervention.

A real-time image compression method based on field programmable gate array is proposed for the problem of high-frame-rate high-resolution camera image transmission under the limited bandwidth of universal serial bus (USB). This method quantizes image pixels on a per-row basis, taking advantage of the high correlation between adjacent pixels within a row, thus reducing the data volume of a single frame image transmission. The algorithm also aims to minimize the decoding complexity on the central processor of the receiving end. Corresponding hardware circuits and software programs are designed, and tests are conducted on an experimental platform. The experimental results show that this method effectively compresses image data losslessly on the board, improves the transmission frame rate. The maximum frame rate of $1280\times 1280$ images tested in a USB 2.0 environment can reach 25.58 fps, an improvement of 11.67 fps compared to the original data transfer, with a compression rate of up to 55.8%. Furthermore, this method outperforms PNG decoding in terms of decoding speed, supports multi-core decoding, and achieves the highest decoding speed of 61fps when tested on the $1920\times 1080$ image with 16 threads. This method provides a feasible transfer solution for real-time compressed transmission of high-speed and high-definition cameras in the industrial field.

In light of rapid economic and urban growth, the proliferation of structures including transmission towers, signal poles, and wind generators has become evident. Consequently, precise object detection of these structures emerges as a pivotal approach to enhance infrastructure management. This technique establishes a robust foundation for achieving elevated efficiency, precision, optimized energy management, and heightened safety monitoring. This article introduces a novel model for detecting structure based on channel and large-scale selective kernel (CLSK) model using high-resolution images. The method is rooted in a two-stage target detection network, enabling simultaneous identification of both primary structures and their associated shadows. The incorporation of deformable convolutions augments the model's ability to extract intricate features. Moreover, the introduction of the innovative LSK attention module, along with the CLSK attention module, enhances the optimization of features gleaned from the network's core architecture. Simultaneously, the complete intersection over union loss function refines the network's focus by considering parameters such as center point distance and aspect ratio in addition to the conventional overlapping area. This comprehensive approach facilitates improved feedback on detection outcomes. Empirical evaluation of the proposed network underscores its superior performance when juxtaposed with both conventional network models and the rotating detection box network.

Abstract Background In solid-predominantly invasive lung adenocarcinoma (SPILAC), occult lymph node metastasis (OLNM) is pivotal for determining treatment strategies. This study seeks to develop and validate a fusion model combining radiomics and deep learning to predict OLNM preoperatively in SPILAC patients across multiple centers. Methods In this study, 1325 cT1a-bN0M0 SPILAC patients from six hospitals were retrospectively analyzed and divided into pathological nodal positive (pN+) and negative (pN-) groups. Three predictive models for OLNM were developed: a radiomics model employing decision trees and support vector machines; a deep learning model using ResNet-18, ResNet-34, ResNet-50, DenseNet-121, and Swin Transformer, initialized randomly or pre-trained on large-scale medical data; and a fusion model integrating both approaches using addition and concatenation techniques. The model performance was evaluated by the area under the receiver operating characteristic (ROC) curve (AUC). Results All patients were assigned to four groups: training set (n = 470), internal validation set (n = 202), and independent test set 1 (n = 227) and 2 (n = 426). Among the 1325 patients, 478 (36%) had OLNM (pN+). The fusion model, combining radiomics with pre-trained ResNet-18 features via concatenation, outperformed others with an average AUC (aAUC) of 0.754 across validation and test sets, compared to aAUCs of 0.715 for the radiomics model and 0.676 for the deep learning model. Conclusion The radiomics-deep learning fusion model showed promising ability to generalize in predicting OLNM from CT scans, potentially aiding personalized treatment for SPILAC patients across multiple centers.

Abstract Background The Hengduan Mountains (HDM) are one of the major global biodiversity hotspots in the world. Several evolutionary scenarios, especially in-situ diversification, have been proposed to account for the high species richness of temperate plants. However, peripatric speciation, an important mode of allopatric speciation, has seldom been reported in this region. Results Here, two chloroplast DNA regions and 14 nuclear loci were sequenced for 112 individuals from 10 populations of Torreya fargesii var. fargesii and 63 individuals from 6 populations of T. fargesii var. yunnanensis. Population genetic analyses revealed that the two varieties are well differentiated genetically (F ST, 0.5765) and have uneven genetic diversity (π, 0.00221 vs. 0.00073 on an average of nuclear loci). The gene genealogical relationship showed that T. fargesii var. yunnanensis is inferred as derived from T. fargesii var. fargesii, which was further supported by the coalescent simulations (DIYABC, fastsimcoal2 and IMa2). By the coalescent simulations, the divergence time (~ 2.50–3.65 Ma) and the weak gene flow between the two varieties were detected. The gene flow was asymmetrical and only occurred in later stages of divergence, which is caused by second contact due to the population expansion (~ 0.61 Ma) in T. fargesii var. fargesii. In addition, niche modeling indicated that the two varieties are differentiated geographically and ecologically and have unbalanced distribution range. Conclusions Overall, T. fargesii var. fargesii is always parapatric with respect to T. fargesii var. yunnanensis, and the latter derived from the former in peripatry of the HDM following a colonization from central China during the late Pliocene. Our findings demonstrate that peripatric speciation following dispersal events may be an important evolutionary scenario for the formation of biodiversity hotspot of the HDM.

IntroductionAccording to the PRISMA criteria, a systematic review has been conducted to investigate the clinical relevance between patients with severe congenital neutropenia (SCN) and cyclic congenital neutropenia (CyN) induced by ELANE mutations.MethodsWe have searched PubMed, EMBASE, Web of Science, Scopus, Cochrane, CNKI, Wanfang Medicine, and VIP for ELANE mutation related literature published from 1997 to 2022. Using Microsoft Excel collect and organize data, SPSS 25, GraphPad Prism 8.0.1, and Omap analyze and plot statistical. Compare the gender, age, geography, mutation sites, infection characteristics, treatment, and other factors of SCN and CyN patients induced by ELANE mutations, with a focus on exploring the relationship between genotype and clinical characteristics, genotype and prognosis.ResultsThis study has included a total of 467 patients with SCN and 90 patients with CyN. The onset age of SCN and CyN are both less than 1 year old, and the onset and diagnosis age of SCN are both younger than CyN. The mutation of ELANE gene is mainly missense mutation, and hot spot mutations include S126L, P139L, G214R, c.597+1G>A. The high-frequency mutations with severe outcomes are A57V, L121H, L121P, c.597+1G>A, c.597+1G>T, S126L, C151Y, C151S, G214R, C223X. Respiratory tract, skin and mucosa are the most common infection sites, Staphylococcus aureus, Pseudomonas aeruginosa and Escherichia coli are the most common.DiscussionPatients with refractory G-CSF are more likely to develop severe outcomes. The commonly used pre-treatment schemes for transplantation are Bu-Cy-ATG and Flu-Bu-ATG. Cytospora canker caused by Cytospora chrysosperma has a wide range of hazards in Xinjiang, seriously affecting the development of Xinjiang’s characteristic forest and fruit industries. Climate change affects the distribution of fungal species and may exacerbate the risk of forest diseases such as cytospora canker. The present study was conducted on C. chrysosperma and makes predictions of potentially suitable area based on 133 records of C. chrysosperma distribution points and 47 environmental factors. We applied the MaxEnt model adjusted by the Kuenm package for feature class parameters (FC) and regularization multipliers (RM) to explore the main environmental factors affecting the geographical distribution of C. chrysosperma. And then we predicted its spatial distribution pattern and centroid change trend in potentially suitable area under three different Representative Concentration Pathways in the current scenario, 2041–2070, and 2071–2,100. The results showed that the optimal model with parameters FC = LQ and RM = 0.3 had the lowest model complexity and overfitting, and the model predicted with very high accuracy, AUC = 0.971 ± 0.0019. Considering the percent contribution, permutation importance of jackknife and single-factor response curve, the main environmental factors affecting its geographical distribution are precipitation seasonality (bio15), aspect cosine (aspectcosine), monthly variability in potential evapotranspiration (PETseasonality), and mean monthly potential evapotranspiration of the coldest quarter (PETColdestQuarter), with a cumulative contribution rate reaching 70.8%. Under the current climate scenarios, the total suitable area for C. chrysosperma is 82.35 × 104 km2, and the highly suitable area is 5.49 × 104 km2, accounting for 6.67% of the total suitable area, primarily located in the Tacheng, Yili, and Changji regions. Meanwhile, centroid transfer analysis indicates a tendency for its distribution to migrate towards lower latitudes under future climatic conditions. The MaxEnt model proposed in this study can be used to predict the distribution and risk of C. chrysosperma in Xinjiang and provide guidance for the prevention and control of cytospora canker.

Earthquakes generally consist of one mainshock and subsequent aftershocks. Although effects of aftershocks following the mainshock on surface structures has been studied extensively, similar studies on underground structures are rarely reported in the literature. With the fast advances in underground space development, robustness of remediation measures against underground structures uplift induced by soil liquefaction shall be examined to ensure their functioning subject to not only the mainshock but also the subsequent aftershocks. This paper studies the uplift behaviour of a conventional manhole subjected to the mainshock-aftershock sequence. It was found that, in the ground that becomes liquefied during the mainshock, manholes become more vulnerable when faced with aftershocks. Due to this reason, some of the previously proposed remediation measures, such as increasing the manholes’ self-weight, roughening the sidewalls, were examined using centrifuge modeling in this study. It was found that such measures had little effect in the aftershocks despite their effectiveness in the precedent mainshock. In contrast, the methods that mitigates manhole uplift by enhancing the manhole’s base permeability, demonstrated better performance in the aftershock than in the mainshock, indicating its promising application potential in future mitigation design.

Abstract Background The duplicated middle cerebral artery (DMCA), a rare anatomical variant of the middle cerebral artery, arises between the anterior choroidal artery and the distal end of the internal carotid artery. We present the case of a patient who had an acute progressive stroke caused by severe stenosis in the initial segment of the DMCA and was successfully treated with endovascular stenting. Case presentation A 57-year-old man was admitted to our hospital with sudden left extremity weakness for three days. Cranial magnetic resonance imaging revealed multiple fresh infarcts in the right basal ganglia and temporal lobe. Cerebrovascular imaging revealed severe stenosis of the right DMCA’s initial segment. However, despite standard medical therapy, the patient’s limb weakness worsened. Based on the clinical and imaging findings, we speculated that severely stenotic DMCA is responsible for the acute progressive stroke. On the basis of the best medical treatment, the patient successfully underwent endovascular stent implantation under general anesthesia two weeks after the onset. The patient’s condition was stable after interventional therapy, and his postoperative follow-up prognosis was favorable. Conclusions Endovascular stenting may be a feasible treatment for symptomatic severe stenosis of the DMCA in cases of poor control with standard medications.

Abstract Background Ulcerative colitis (UC) represents a clinically challenging condition characterized by persistent damage to the colonic epithelial mucosa as the principal pathological feature. Polyvinyl alcohol (PVA) solution, primarily composed of glue, is a biodegradable polymer material that has found utility in the medical field. This research endeavors to investigate the therapeutic potential of PVA water solution in ameliorating UC in mice. Methods UC was induced in 48 C57BL/6 mice by administering 2.5% DSS in their diet for 6 days. Mice were treated with different concentrations of PVA (0.1 mg/ml PVA, 0.3 mg/ml PVA, 1 mg/ml PVA, 3 mg/ml PVA, 10 mg/ml PVA) enemas (n = 6). Disease Activity Index (DAI) and histologic score were evaluated for inflammation degree. Furthermore, mouse colon organoids were cultured, which were used to assess the effects of PVA on expansion in vitro. Results PVA aqueous solutions (1 mg/ml and 3 mg/ml) were able to alleviate the DAI in mice. By DAY 6, there was a significant 3/5-fold decrease in DAI within the 1 mg/ml PVA group (p = 0.02). Histopathology scores demonstrated improvements, while the levels of inflammatory factors in the intestinal mucosal tissue were reduced. Additionally, it was confirmed that PVA could promote the expansion of colonic organoids in vitro. Conclusions In summary, our investigation has yielded findings indicating that PVA holds the potential to ameliorate symptoms associated with colitis in murine subjects afflicted by DSS-induced colitis, primarily through its facilitation of intestinal stem cell expansion. This study might provide a new candidate for the clinical treatment of ulcerative colitis.

Introduction Nursing staff’s assistance for smokers to quit smoking can increase the rate of quitting. The smoking cessation help can be affected by many factors. This study surveyed the use of the 5As (Asking, Advising, Assessing, Assisting, Arranging) approach to support smoking cessation by the nursing staff in Chongqing, China, and analyzed the corresponding influencing factors. Methods A stratified random cluster sampling method was used to select nursing staff from 8 tertiary hospitals, 5 secondary hospitals, 12 community health centers, and 35 township health centers in different geographical regions of Chongqing. A questionnaire survey was conducted among the nursing staff to investigate their participation in smoking cessation. Binary logistic regression analysis was employed to analyze the influencing factors of smoking cessation 5As behavior of the nursing staff. Results The 1669 participants were 44 males (2.6%) and 1625 females (97.4%), with an average age of 37.00 ± 10.89 years. Among the participants, 55.2% were from tertiary hospitals, 23.2% from secondary hospitals, 14.2% from township health centers, and 7.4 from community health centers. The often or always used behaviors were: Asking, 69.2%; Advising, 53.0%; Assessing, 39.5%; Assisting, 33.7%; and Arranging, 25.1%. The factors that affected all the 5As were: smoking cessation training (AOR=1.60; 95% CI: 1.22–2.11), knowledge of smoking cessation guidelines (AOR=1.75; 95% CI: 1.32–2.32) and the use of smoking cessation Apps (AOR=1.50; 95% CI: 1.09–2.06), and smokers’ willingness to quit (AOR=2.20; 95% CI: 1.60–3.02). Conclusions Smoker’s motivation to quit smoking and nurses’ knowledge of tobacco cessation resources affected nurses’ participation in smoking control behavior. While encouraging smokers to quit smoking, clinical nursing staff should be provided with related resources to advocate smoking cessation. Guided by the Chinese Clinical Tobacco Cessation Guidelines, smoking control knowledge and skills training should be provided for nursing staff to increase their positive attitude towards smoking control, so as to promote their active participation.

Abstract Colorectal cancer (CRC) is a major cause of cancer-related deaths in humans, and effective treatments are still needed in clinical practice. Despite significant developments in anticancer drugs and inhibitors, their poor stability, water solubility, and cellular membrane permeability limit their therapeutic efficacy. To address these issues, multifunctional CaCO3 nanoparticles loaded with Curcumin (Cur) and protein deacetylase (HDAC) inhibitor QTX125, and coated with hyaluronic acid (HA) (CaCO3@Cur@QTX125@HA), were prepared through a one-step gas diffusion strategy. Dynamic light scattering (DLS), transmission electron microscopy (TEM), and scanning electron microscopy (SEM) showed that CaCO3@Cur@QTX125@HA nanoparticles have uniform spherical morphology and elemental distribution, with diameters around 450 nm and a Zeta potential of − 8.11 mV. The controlled release of Cur from the nanoparticles was observed over time periods of 48 h. Cellular uptake showed that CaCO3@Cur@QTX125@HA nanoparticles were efficiently taken up by cancer cells and significantly inhibited their growth. Importantly, CaCO3@Cur@QTX125@HA nanoparticles showed specific inhibitory effects on CRC cell growth. Encouragingly, CaCO3@Cur@QTX125@HA nanoparticles successfully internalized into CRC patient-derived organoid (PDO) models and induced apoptosis of tumor cells. The multifunctional CaCO3@Cur@QTX125@HA nanoparticles hold promise for the treatment of CRC.

Abstract Background This study aims to investigate the relationship between different dietary patterns and non-alcoholic fatty liver disease (NAFLD). Methods Residents over 30 years old in the ecological longevity cohort in Gongcheng Yao Autonomous County, Guangxi Province were the research objects selected from 2018 to 2019. Physical examination, baseline population survey, and food frequency questionnaire (FFQ) survey were conducted. Dietary patterns were analyzed by factor analysis. Influencing factors of NAFLD were analyzed by multiple logistic regression. Results NAFLD was diagnosed in 241 of 2664 participants based on ultrasonography, and the detection rate was 9.0%. Factor analysis yielded a total of three dietary patterns, namely, traditional Chinese, Western, and cereal-potato dietary patterns. Results of multivariate logistic regression analysis showed that after adjusting for confounding factors, participants in the highest quartile of the Western dietary pattern exhibited a higher prevalence of NAFLD (OR = 2.799; 95% CI: 1.620–4.837; p

This study evaluates the effectiveness of artificial Kandelia obovata forests in wave attenuation through physical model experiments conducted in a wave flume. The experiments meticulously replicated real-world hydrodynamic conditions and mangrove movement responses using the principles of gravitational and motion similarity, with a scaled 1:10 model of Kandelia obovata. Our approach included comparative experiments against a 1:100 gradient concrete slope to isolate the effects of seabed friction and flume wall reflections. The wave height was measured using strategically placed wave gauges. The findings indicated that the artificial Kandelia obovata forests significantly attenuated waves, with a decrease in the total attenuation capacity as the water depth increased from 2.75 m to 3.28 m under both regular and irregular waves. The elastic mangrove model with a canopy effect led to a 15% increase in wave attenuation over cylindrical models. Predictive models using multivariate nonlinear regression and back propagation neural networks showed that the latter provided a superior accuracy in estimating wave transmission coefficients

As a typical component of remote sensing signals, remote sensing image (RSI) information plays a strong role in showing macro, dynamic and accurate information on the earth’s surface and environment, which is critical to many application fields. One of the core technologies is the object detection (OD) of RSI signals (RSISs). The majority of existing OD algorithms only consider medium and large objects, regardless of small-object detection, resulting in an unsatisfactory performance in detection precision and the miss rate of small objects. To boost the overall OD performance of RSISs, an improved detection framework, I-YOLO-V5, was proposed for OD in high-altitude RSISs. Firstly, the idea of a residual network is employed to construct a new residual unit to achieve the purpose of improving the network feature extraction. Then, to avoid the gradient fading of the network, densely connected networks are integrated into the structure of the algorithm. Meanwhile, a fourth detection layer is employed in the algorithm structure in order to reduce the deficiency of small-object detection in RSISs in complex environments, and its effectiveness is verified. The experimental results confirm that, compared with existing advanced OD algorithms, the average accuracy of the proposed I-YOLO-V5 is improved by 15.4%, and the miss rate is reduced by 46.8% on the RSOD dataset.

Using physical vapor deposition (PVD) technology, GeSe nanowires were successfully fabricated by heating GeSe powder at temperatures of 500 °C, 530 °C, 560 °C, 590 °C, and 620 °C. The microstructure, crystal morphology, and chemical composition of the resulting materials were thoroughly analyzed employing methods like Scanning Electron Microscopy (SEM), X-ray Diffraction (XRD), plus Raman Spectroscopy. Through a series of photoelectric performance tests, it was discovered that the GeSe nanowires prepared at 560 °C exhibited superior properties. These nanowires not only possessed high crystalline quality but also featured uniform diameters, demonstrating excellent consistency. Under illumination at 780 nm, the GeSe nanowires prepared at this temperature showed higher dark current, photocurrent, and photoresponsivity compared to samples prepared at other temperatures. These results indicate that GeSe nanomaterials hold substantial potential in the field of photodetection. Particularly in the visible light spectrum, GeSe nanomaterials exhibit outstanding light absorption capabilities and photoresponse.

The annotation of magnetic resonance imaging (MRI) images plays an important role in deep learning-based MRI segmentation tasks. Semi-automatic annotation algorithms are helpful for improving the efficiency and reducing the difficulty of MRI image annotation. However, the existing semi-automatic annotation algorithms based on deep learning have poor pre-annotation performance in the case of insufficient segmentation labels. In this paper, we propose a semi-automatic MRI annotation algorithm based on semi-weakly supervised learning. In order to achieve a better pre-annotation performance in the case of insufficient segmentation labels, semi-supervised and weakly supervised learning were introduced, and a semi-weakly supervised learning segmentation algorithm based on sparse labels was proposed. In addition, in order to improve the contribution rate of a single segmentation label to the performance of the pre-annotation model, an iterative annotation strategy based on active learning was designed. The experimental results on public MRI datasets show that the proposed algorithm achieved an equivalent pre-annotation performance when the number of segmentation labels was much less than that of the fully supervised learning algorithm, which proves the effectiveness of the proposed algorithm.

The objective of this study is to investigate the effects of functional corrective training and static stretching on the quality of movement patterns and physical fitness in college students. The study was conducted with 30 male college students from a university in Guangzhou, China. The participants were randomly assigned to either the functional corrective training group (FCT, n = 15, age = 20.93 ± 0.85, BMI = 22.07 ± 2.33) or the static stretching group (SS, n = 13, age = 20.85 ± 0.86, BMI = 21.98 ± 1.80). Two participants from the SS group dropped out due to personal reasons, leaving 13 subjects in that group. Both groups underwent a 6-week training intervention, with sessions held twice a week. The FCT group participated in flexibility training, and/or static motor control training, and/or dynamic motor control training for 10–15 min. The SS group performed static stretching exercises targeting five specific muscles, with 30 s per side and two sets. The Functional Movement Screen (FMS), body composition, sit-and-reach, standing long jump, and pull-ups were assessed before and after the intervention. Differences in FMS outcomes were analyzed using two samples of the Mann-Whitney U test. Physical fitness outcomes were analyzed using a repeated measures analysis of variance (ANOVA) with a 2 (group) × 2 (time) design. After 6 weeks of intervention, the FCT group showed statistically significant improvements in the hurdle step (Z = −2.449, p = 0.014), inline lunge (Z = −2.000, p = 0.046), rotary stability (Z = −2.309, p = 0.021), and composite scores (Z = −3.316, p = 0.001). Comparisons between groups indicated that BMI (FCT, ES = 0.04; SS, ES = −0.11), 30-m sprint (FCT, ES = 0.12; SS, ES = 0.28), body fat percentage (BF%) (FCT, ES = −0.25; SS, ES = −0.07), and sit-and-reach (FCT, ES = 0.17; SS, ES = 0.06) were not statistically significant in both the pre- and post-tests. The effect sizes of all physical fitness indicators were greater in the FCT group than in the SS group. The FCT, consisting of two sessions per week for 6 weeks, has been proven to be effective in improving the quality of movement patterns by improved stability and advanced movements. However, the improvements in physical fitness did not reach statistical significance. FMS and FCT are generally affordable and accessible for college students. College students have the opportunity to employ the FMS tool to assess potential injury risks and address them, thereby reducing the risk of injuries.

Summary: Immune escape is identified as one of the reasons for the poor prognosis of colorectal cancer (CRC) patients. Circular RNAs are considered to promote tumor progression by mediating tumor immune escape. We discovered that higher expression of circYAP1 was associated with a worse prognosis of CRC patients. Functional experiments in vitro and in vivo showed that circYAP1 upregulation inhibited the cytotoxicity of CD8+ T cells by upregulating programmed death ligand-1 (PD-L1). Mechanistically, we found that circYAP1 directly binds to the YAP1 protein to prevent its phosphorylation, enhancing proportion of YAP1 protein in the nucleus, and that YAP1 interacts with TCF4 to target the PD-L1 promoter and initiate PD-L1 transcription in CRC cells. Taken together, circYAP1 promotes CRC immune escape and tumor progression by activating the YAP1/TCF4-PD-L1 axis and may provide a new strategy for combination immunotherapy of CRC patients.

The Beishan orogen, a significant component of the southern Altaids, presents an opportunity for investigating the intracontinental deformation and exhumation history of the Altaids during the Mesozoic era. Although previous studies indicated that the Beishan orogen has experienced multiple reactivation since the late Mesozoic, the precise extent of these events remains poorly constrained. Here, we provide a comprehensive synthesis of field observations and apatite fission track (AFT) thermochronological dating throughout the Beishan orogen. Detailed field observations confirmed four major E-W trending thrusts in our study area. Based on the youngest truncated strata associated with the thrusts and previous dating results from neighboring regions, we propose that these thrust sheets likely developed in the late Middle Jurassic. AFT dating results from seven pre-Mesozoic granitoid samples and associated with thermal history modeling demonstrate that the Beishan orogen experienced a rapid basement cooling during the mid-Cretaceous (~115–80 Ma). Moreover, a compilation of previously published and newly gained AFT data reveals a comparable mid-Cretaceous cooling event in other parts of Central Asia, such as Qilian Shan, Eastern Tianshan, and Altai-Sayan. This observation suggests that the mid-Cretaceous cooling event is more likely to be regional rather than localized. This mid-Cretaceous cooling pulse is interpreted as a tectonic exhumation controlled by boundary faults and related to the rotation of the Junggar and Tarim basins. These processes are linked to distant plate-margin events along the Eurasian continent.

Abstract Due to their atomically dispersed active centers, single‐atom nanozymes (SAzymes) have unparalleled advantages in cancer catalytic therapy. Here, loaded with chlorin e6 (Ce6), a hydrothermally mass‐produced bimetallic silicate‐based nanoplatforms with atomically dispersed manganese/gadolinium (Mn/Gd) dual sites and oxygen vacancies (OVs) (PMnSAGMSNs‐V@Ce6) is constructed for tumor glutathione (GSH)‐triggered chemodynamic therapy (CDT) and O2‐alleviated photodynamic therapy. The band gaps of silica are significantly reduced from 2.78 to 1.88 eV by doping with metal ions, which enables it to be excited by a 650 nm laser to produce electron‐hole pairs, thereby facilitating the generation of reactive oxygen species. The Gd sites can modulate the local electrons of the atom‐catalyzed Mn sites, which contribute to the generation of superoxide and hydroxyl radicals (•OH). Tumor GSH‐triggered Mn2+ release can convert endogenous H2O2 to •OH and realize GSH‐depletion‐enhanced CDT. Significantly, the hydrothermally generated OVs can not only capture Mn and Gd atoms to form atomic sites but also can elongate and weaken the O‐O bonds of H2O2, thereby improving the efficacy of Fenton reactions. The degraded Mn2+/Gd3+ ions can be used as tumor‐specific magnetic resonance imaging contrast agents. All the experimental results demonstrate the great potential of PMnSAGMSNs‐V@Ce6 as cancer theranostic agent.

The increasing atmospheric [CO2] poses great challenges to wheat production. Currently, the response of starch characteristics in different specialized wheat cultivars to elevated [CO2], as well as the underlying physiological and molecular mechanisms remains unclear. Therefore, an experiment was conducted with open-top chambers to study the effects of ambient [CO2] [a(CO2)] and elevated [CO2] [e(CO2)] on photosynthetic performance, yield and starch characteristics of bread wheat (Zhengmai 369, ZM369) and biscuit wheat (Yangmai 15, YM15) from 2020 to 2022. The results demonstrated a significant improvement in photosynthetic performance, yield, amylose and amylopectin content, volume ratio of large granules under e[CO2]. Moreover, e[CO2] upregulated the gene expression and enzyme activities of GBSS (Granule-bound starch synthase) and SSS (Soluble starch synthase), increased starch pasting viscosity, gelatinization enthalpy and crystallinity. Compared to YM15, ZM369 exhibited a higher upregulation of GBSSI, greater increase in amylose content and volume ratio of large granules, as well as higher gelatinization enthalpy and crystallinity. However, ZM369 showed a lower increase in amylopectin content and a lower upregulation of SSSI and SSSII. Correlation analysis revealed amylose and amylopectin content had a positive correlation with GBSS and SSS, respectively, a significant positively correlation among the amylose and amylopectin content, starch granule volume, and pasting properties. In conclusion, these changes may enhance the utilization value of biscuit wheat but exhibit an opposite effect on bread wheat. The results provide a basis for selecting suitable wheat cultivars and ensuring food security under future climate change conditions.

The rise of quantum computing has prompted the interest in the field of cryogenic electronics. Carbon-based materials hold great promise in the area of cryogenic electronics due to their excellent material properties and emergent quantum effects. This paper introduces the advantages of carbon-based materials for cryogenic applications and reviews recent progress in carbon nanotubes and graphene for logic devices, sensors and novel quantum devices at cryogenic temperatures. Finally, the main challenges and extensive prospects for the further development of carbon-based cryoelectronics are summarized.

Small round cell undifferentiated sarcoma is a rare and highly invasive group of malignant bone and soft tissue tumors, often associated with a high misdiagnosis rate. The patient in this case was a 34-year-old male who presented with a two-month history of abdominal pain that worsened over the past two weeks. Elevated levels of tumor markers CA19-9 and CA72-4 were observed. Imaging revealed a substantial, well-vascularized mass in the lower left abdomen, located in the posterior abdominal cavity, invading the descending colon and the root of the small mesentery, and infiltrating the serous layer. The lesion was extensively resected without any postoperative complications. Microscopic examination indicated a combination of mucinous adenocarcinoma (approximately 30%) and small round cell undifferentiated sarcoma (approximately 70%). The patient was followed up for six months, and one month after surgery, a recurrence of the tumor was observed in the left paracolonic sulcus area, with metastases to the abdominal wall, peritoneum, and medial iliac muscles. Chemotherapy and targeted therapy were administered, and the patient currently survives with the presence of tumors. Small round cell undifferentiated sarcoma is an uncommon and highly invasive tumor, and clinical surgeons need to raise their awareness and realize to the maximum extent possible that this disease can be described through a multi-modal combination of immunohistochemistry and genetic test to improve diagnostic accuracy and reduce missed diagnoses. Further research in the field of biology is necessary to explore targeted drugs specifically suitable for this disease.

Abstract Objective To analyze the relationship between plasma metal elements, ApoE gene polymorphisms and the interaction between the two and impaired cognitive function in elderly population. Method A stratified sample was drawn according to the age of the study population, and 911 subjects were included. Baseline information and health indicators were obtained, and cognitive function status was assessed by health examination, a general questionnaire and Mini-Mental Status Examination. Plasma metal elements were measured, and SNP typing was performed. Abstract Background Colorectal cancer is one of the most serious malignant tumors, and lymph node metastasis (LNM) from colorectal cancer is a major factor for patient management and prognosis. Accurate image detection of LNM is an important task to help clinicians diagnose cancer. Recently, the U-Net architecture based on convolutional neural networks (CNNs) has been widely used to segment image to accomplish more precise cancer diagnosis. However, the accurate segmentation of important regions with high diagnostic value is still a great challenge due to the insufficient capability of CNN and codec structure in aggregating the detailed and non-local contextual information. In this work, we propose a high performance and low computation solution. Methods Inspired by the working principle of Fovea in visual neuroscience, a novel network framework based on U-Net for cancer segmentation named Fovea-UNet is proposed to adaptively adjust the resolution according to the importance-aware of information and selectively focuses on the region most relevant to colorectal LNM. Specifically, we design an effective adaptively optimized pooling operation called Fovea Pooling (FP), which dynamically aggregate the detailed and non-local contextual information according to the pixel-level feature importance. In addition, the improved lightweight backbone network based on GhostNet is adopted to reduce the computational cost caused by FP. Results Experimental results show that our proposed framework can achieve higher performance than other state-of-the-art segmentation networks with 79.38% IoU, 88.51% DSC, 92.82% sensitivity and 84.57% precision on the LNM dataset, and the parameter amount is reduced to 23.23 MB. Conclusions The proposed framework can provide a valid tool for cancer diagnosis, especially for LNM of colorectal cancer.

Abstract Small molecule direct BAK activators can potentially be used for the development of anti-cancer drugs or as tools to study BAK activation. The thrombopoietin receptor agonist eltrombopag (Eltro) inhibits BAX activation and BAX-mediated apoptosis. Here we report that, in contrast to its function as a BAX inhibitor, Eltro directly binds BAK but induces its activation in vitro. Moreover, Eltro induces or sensitizes BAK-dependent cell death in mouse embryonic fibroblasts (MEFs) and Jurkat cells. Chemical shift perturbation analysis by NMR indicates that Eltro binds to the BAK α4/α6/α7 groove to initiate BAK activation. Further molecular docking by HADDOCK suggests that several BAK residues, including R156, F157, and H164, play an important role in the interaction with Eltro. The introduction of an R156E mutation in the BAK α4/α6/α7 groove not only decreases Eltro binding and Eltro-induced BAK activation in vitro but also diminishes Eltro-induced apoptosis. Thus, our data suggest that Eltro directly induces BAK activation and BAK-dependent apoptosis, providing a starting point for the future development of more potent and selective direct BAK activators.

Abstract Hyperuricemia (HUA) endangers human health, and its prevalence has increased rapidly in recent decades. The current study investigated HUA's prevalence and influencing factors in Gongcheng, southern China. A cross-sectional investigation was conducted; 2128 participants aged 30–93 years were included from 2018 to 2019. Univariate and multivariate logistic regression models were used to screen HUA variables. A Bayesian network model was constructed using the PC algorithm to evaluate the association between influencing factors and HUA. The prevalence of HUA was 15.6% (23.2% in men, 10.7% in women). After screening the variables using a logistic regression analysis model, fatty liver disease (FLD), dyslipidemia, abdominal obesity, creatinine (CREA), somatotype, bone mass, drinking, and physical activity level at work were included in the Bayesian network model. The model results showed that dyslipidemia, somatotype, CREA, and drinking were directly related to HUA. Bone mass and FLD were indirectly associated with HUA by affecting the somatotype. The prevalence of HUA in Gongcheng was high in China. The prevalence of HUA was related to somatotype, drinking, bone mass, physical activity level at work, and other metabolic diseases. A good diet and moderate exercise are recommended to maintain a healthy somatotype and reduce the prevalence rate of HUA.
