PurposeThe objective of this study was to investigate alterations in functional connectivity density (FCD) mapping and their impact on functional connectivity (FC) among individuals diagnosed with Type 2 diabetes mellitus (T2DM) across different cognitive states. Moreover, the study sought to explore the potential association between aberrant FCD/FC patterns and clinical or cognitive variables.MethodsA total of 211 participants were recruited for this study, consisting of 75 healthy controls (HCs), 89 T2DM patients with normal cognitive function (DMCN), and 47 T2DM patients with mild cognitive impairment (DMCI). The study employed FCD analysis to pinpoint brain regions exhibiting significant FCD alterations. Subsequently, these regions showing abnormal FCD served as seeds for FC analysis. Exploratory partial correlations were conducted to explore the relationship between clinical biochemical indicators, neuropsychological test scores, and altered FCD or FC.ResultsThe FCD analysis revealed an increased trend in global FCD (gFCD), local FCD (lFCD), and long-range FCD (lrFCD) within the bilateral supramarginal gyrus (SMG) among individuals with DMCN. Additionally, significant lFCD alterations were observed in the right inferior frontal gyrus and left precuneus when comparing DMCN to HCs and DMCI.ConclusionWhen comparing individuals with T2DM and healthy controls (HCs), it was revealed that DMCN exhibited significant improvements in FCD. This suggests that the brain may employ specific compensatory mechanisms to maintain normal cognitive function at this stage. Our findings provide a novel perspective on the neural mechanisms involved in cognitive decline associated with T2DM.

Accurately classifying the quality of apples is crucial for maximizing their commercial value. Deep learning techniques are being widely adopted for apple quality classification tasks, achieving impressive results. While existing research excels at classifying apple variety, size, shape, and defects, color and deformity analysis remain an under-explored area. Therefore, this study investigates the feasibility of utilizing convolutional neural networks (CNN) to classify the color and deformity of apples based on machine vision technology. Firstly, a custom-assembled machine vision system was constructed for collecting apple images. Then, image processing was performed to extract the largest fruit diameter from the 45 images taken for each apple, establishing an image dataset. Three classic CNN models (AlexNet, GoogLeNet, and VGG16) were employed with parameter optimization for a three-category classification task (non-deformed slice–red apple, non-deformed stripe–red apple, and deformed apple) based on apple features. VGG16 achieved the best results with an accuracy of 92.29%. AlexNet and GoogLeNet achieved 91.66% and 88.96% accuracy, respectively. Ablation experiments were performed on the VGG16 model, which found that each convolutional block contributed to the classification task. Finally, prediction using VGG16 was conducted with 150 apples and the prediction accuracy was 90.50%, which was comparable to or better than other existing models. This study provides insights into apple classification based on color and deformity using deep learning methods.

Background: Condyloma acuminatum (CA), which is a highly contagious sexually transmitted illness generated by human papillomavirus (HPV) infection, is characterized by abnormal proliferation of keratinocytes resulting in verrucous lesions. Although solute carrier family 30 member 1 (ZNT1) is highly expressed in CA tissues, the function of ZNT1 in CA remains unclear. Methods: HPV transfection was performed in HaCaT to simulate the CA pathological environment. The mRNA and protein levels were monitored using RT-qPCR and immunoblotting. Cell viability was found using the MTT test. Cell invasion and migration were probed using the transwell and wound healing. Results: ZNT1 expression was up-regulated in CA tissues, and HPV transfection increased the expression of ZNT1. Overexpression of ZNT1 promoted the proliferation, migration and invasion of Human immortalized keratinocyte (HaCaT) transfected with HPV. Meanwhile, ZNT1 knockdown repressed the proliferation, migration and invasion of HaCaT that HPV transfected. Further research displayed that ZNT1 promoted the proliferation, migration and invasion of HaCaT transfected with HPV through the PI3K/Akt pathway. Conclusion: Our research confirmed that ZNT1 regulated the proliferation, migration and invasion of HaCaT transfected with HPV through the PI3K/Akt pathway, providing a new target for the effective remedy of CA.

Background: Systemic lupus erythematosus (SLE) has been correlated with osteomyelitis (OM), yet the underlying causal relationship remains poorly understood. This study aims to investigate the causal association between SLE and OM using Mendelian randomization (MR) analysis. Methods: Genetic instrumental variables (IVs) correlated with SLE were extracted from a comprehensive genome-wide association study (GWAS) summary database (5201 cases and 9066 controls). OM was considered a SLE phenotype, and summary data from the fast GWA data portal were utilized for the analysis. Eligible IVs were extracted following rigorous quality control measures (P 0.001, distance 1 Mb, and F > 10). MR analysis was conducted using the Inverse Variance Weighted (IVW), MR-Egger, and Weighted Median (WM) methods after excluding potential confounders. Cochran's Q was applied for heterogeneity test. Pleiotropy was evaluated through MR-Egger intercept, MR-Pleiotropy Residual Sum and Outlier (MR-PRESSO) method, and Leave-one-SNP-out analysis. Result: A total of 40 eligible IVs were included for MR analysis. IVW results demonstrated a positive causal association between SLE and OM (P = 0.049, OR = 1.167). Heterogeneity analysis reveal no significant heterogeneity in the IVW analysis (P = 0.5503). Pleiotropy tests, including MR-PRESSO global test and MR-Egger intercept, indicated no evidence of pleiotropy in our findings (P > 0.05). Additionally, the Leave-one-SNP-out analysis showed no substantial deviations when removing individual SNPs, thus supporting the robustness of our results. Conclusion: This study establishes a genetic causal relationship between SLE and OM, indicating an increased risk of developing OM in individuals with SLE. Therefore, proactive management of SLE is advised to mitigate the risk of developing OM.

Anticipated permafrost thaw in upcoming decades may exert significant impacts on forest soil nitrogen (N) dynamics. The rate of soil N mineralization (Nmin) plays a crucial role in determining soil N availability. Nevertheless, our understanding remains limited regarding how biotic and abiotic factors influence the Nmin of forest soil in response to permafrost thaw. In this study, we investigated the implications of permafrost thaw on Nmin within a hemiboreal forest based on a field investigation along the degree of permafrost thaw, having monitored permafrost conditions for eight years. The results indicate that permafrost thaw markedly decreased Nmin values. Furthermore, Nmin demonstrated positive associations with soil substrates (namely, soil organic carbon and soil total nitrogen), microbial biomass carbon and nitrogen, and soil moisture content. The decline in Nmin due to permafrost thaw was primarily attributed to the diminished quality and quantity of soil substrates rather than alterations in plant community composition. Collectively, our results underscore the pivotal role of soil substrate and microbial biomass in guiding forest soil N transformations in the face of climate-induced permafrost thaw.

IntroductionType 2 diabetes mellitus (T2DM) can accelerate cognitive decline and even dementia so that the underlying mechanism deserves further exploration. In the resting state, brain function is still changing dynamically. At present, it is still unknown whether the dynamic functional connectivity (dFC) between various brain regions is in a stable state. It is necessary to interpret brain changes from a new perspective, that is, the stability of brain architecture.MethodsIn this study, we used a fixed dynamic time scale to explore the stability of dynamic functional architecture in T2DM, then the dynamic effective connectivity (dEC) was used to further explain how information flows through dynamically fluctuating brain architecture in T2DM.ResultTwo brain regions with decreased stability were found including the right supra-marginal gyrus (SMG) and the right median cingulate gyrus (MCG) in T2DM subjects. The dEC variation has increased between the left inferior frontal gyrus (IFG) and the right MCG. The direction of causal flow is from the right MCG to the left IFG.ConclusionThe combination of stability and dEC can not only show the stability of dynamic functional architecture in brain but also reflect the fluidity of brain information, which is an innovative and interesting attempt in the field of neuroimaging. The changes of dynamic architecture in T2DM patients may present an innovative perspective and explanation for their cognitive decline.

Abstract The comprehensive evaluation of pipeline loess collapsibility risk is a necessary means to control the safety risks of pipelines in the collapsible loess section. It is also one of the critical scientific bases for risk prevention, control, and management. The comprehensive evaluation system of cloud theory consists of quantitative and qualitative indexes, and the evaluation system has the characteristics of randomness and fuzziness. In view of this problem, the standard qualitative and semi-quantitative evaluation methods have intense subjectivity in dealing with the uncertainty problems such as randomness and fuzziness of the system, the cloud theory, which can effectively reflect the randomness and fuzziness of things at the same time, is introduced. The state scale cloud and index importance weight cloud of pipeline loess collapse risk are constructed by the golden section method. The uncertainty cloud reasoning process of the quantitative indexes and the expert scoring method of the qualitative indexes are proposed. The comprehensive evaluation model of loess collapsibility risk of oil and gas pipeline is established, and the engineering example is analyzed. The complete evaluation results of 10 samples to be evaluated are consistent with the results of the semi-quantitative method and are compatible with the actual situation. The evaluation process softens the subjective division of index boundary, simplifies the preprocessing of index data, realizes the organic integration of quantitative and qualitative decisions, and improves the accuracy, rationality, and visualization of the results.

In order to research the special embankment (thermosyphon embankment) damages and the distribution of permafrost under the Qinghai–Tibet Highway (QTH) embankment. The section K2952–K2953, which is a typical representative of the QTH, was chosen for the detection and research of the permafrost and embankment damages in order to determine the sources of the damages. In this study, the performance characteristics of the embankment, the active layer, and the permafrost table found in ground-penetrating radar (GPR) images were researched, combined with multi-source. According to the research findings, the construction of the embankment in this section has stabilized the effect on the permafrost table. Under the embankment of the unemployed thermosyphon section, the permafrost distribution has good structural integrity and continuity, with the permafrost table at a depth of around 5 m. The continuity of the permafrost distribution under the embankment in the thermosyphon section was poor, and there was localized degradation, with the permafrost table being approximately 6 m deep. The main cause of the irregular settlement and other damage in this section is the presence of a loose area at the base of the embankment. Although the thermosyphon on both sides of the embankment also plays a role in lifting the permafrost table, it is not ideal for managing the damage to high embankments where the type of permafrost under the embankment is high-temperature permafrost with a high ice content and where the sunny–shady slope effect is obvious. The research results described in this article can therefore provide a crucial foundation for the detection of highway damage and permafrost under embankments in permafrost regions in the future.

Spatial-frequency domain imaging (SFDI) has been developed as an emerging modality for detecting early-stage bruises of fruits, such as apples, due to its unique advantage of a depth-resolved imaging feature. This paper presents theoretical and experimental analyses to determine the light penetration depth in apple tissues under spatially modulated illumination. Simulation and practical experiments were then carried out to explore the maximum light penetration depths in ‘Golden Delicious’ apples. Then, apple experiments for early-stage bruise detection using the estimated reduced scattering coefficient mapping were conducted to validate the results of light penetration depths. The results showed that the simulations produced comparable or a little larger light penetration depth in apple tissues (~2.2 mm) than the practical experiment (~1.8 mm or ~2.3 mm). Apple peel further decreased the light penetration depth due to the high absorption properties of pigment contents. Apple bruises located beneath the surface peel with the depth of about 0–1.2 mm could be effectively detected by the SFDI technique. This study, to our knowledge, made the first effort to investigate the light penetration depth in apple tissues by SFDI, which would provide useful information for enhanced detection of early-stage apple bruising by selecting the appropriate spatial frequency.

Climate change has caused high salinity in many fields, particularly in the mud flats in coastal regions. The resulting salinity has become one of the most significant abiotic stresses affecting the world’s rice crop productivity. Developing elite cultivars with novel salinity-tolerance traits is regarded as the most cost-effective and environmentally friendly approach for utilizing saline-alkali land. To develop a highly efficient green strategy and create novel rice germplasms for salt-tolerant rice breeding, this study aimed to improve rice salinity tolerance by combining targeted CRISPR/Cas9-mediated editing of the OsRR22 gene with heterosis utilization. The novel alleles of the genic male-sterility (GMS) and elite restorer line (733Srr22-T1447-1 and HZrr22-T1349-3) produced 110 and 1 bp deletions at the third exon of OsRR22 and conferred a high level of salinity tolerance. Homozygous transgene-free progeny were identified via segregation in the T2 generation, with osrr22 showing similar agronomic performance to wild-type (733S and HZ). Furthermore, these two osrr22 lines were used to develop a new promising third-generation hybrid rice line with novel salinity tolerance. Overall, the results demonstrate that combining CRISPR/Cas9 targeted gene editing with the “third-generation hybrid rice system” approach allows for the efficient development of novel hybrid rice varieties that exhibit a high level of salinity tolerance, thereby ensuring improved cultivar stability and enhanced rice productivity.

This paper reports on the measurement of optical property mapping of apples at the wavelengths of 460, 527, 630, and 710 nm using spatial-frequency domain imaging (SFDI) technique, for assessing the soluble solid content (SSC), firmness, and color parameters. A laboratory-based multispectral SFDI system was developed for acquiring SFDI of 140 “Golden Delicious” apples, from which absorption coefficient (μa) and reduced scattering coefficient (μs′) mappings were quantitatively determined using the three-phase demodulation coupled with curve-fitting method. There was no noticeable spatial variation in the optical property mapping based on the resulting effect of different sizes of the region of interest (ROI) on the average optical properties. Support vector machine (SVM), multiple linear regression (MLR), and partial least square (PLS) models were developed based on μa, μs′ and their combinations (μa × μs′ and μeff) for predicting apple qualities, among which SVM outperformed the best. Better prediction results for quality parameters based on the μa were observed than those based on the μs′, and the combinations further improved the prediction performance, compared to the individual μa or μs′. The best prediction models for SSC and firmness parameters [slope, flesh firmness (FF), and maximum force (Max.F)] were achieved based on the μa × μs′, whereas those for color parameters of b* and C* were based on the μeff, with the correlation coefficients of prediction as 0.66, 0.68, 0.73, 0.79, 0.86, and 0.86, respectively.

Characterized by low bearing capacity and high compressibility, warm and ice-rich frozen soil is a kind of problematic soil, which makes the original frozen ground formed by of that unreliable to meet the stability requirements of engineering infrastructures and foundations in permafrost regions. With the design and construction of major projects along the Qinghai-Tibet Engineering Corridor (QTEC), such as expressway and airport runway, it is a great challenge to favor the stability of overlying structures by formulating the proper engineering design principles and developing the valid engineering supporting techniques. The investigations carried out in recent years indicated that warm and ice-rich permafrost foundations were widespread, climate warming was significant, and the stability of existing engineering structures was poor, along the QTEC. When the warm and ice-rich frozen ground is used as the foundation soil, the implementation of ground improvement is an alternative measure to enhance the bearing capacity of foundation soil and eliminate the settlement of structures during operation, in order to guarantee the long-term stability of the structures. Based on the key factors determining the physicomechanical properties of frozen soil, an innovative idea of stabilizing the warm and ice-rich frozen soil based on chemical stabilization is proposed in this study, and then, an in situ ground improvement technique is introduced. This study intends to explore the feasibility of ground improvement in warm and ice-rich permafrost regions along the QTEC based on in situ chemical stabilization and provide the technical support and scientific reference to prevent and mitigate the hazards in the construction of major projects in the future.

Ground collapse is one of the main geological disasters affecting the safe operation of oil and gas pipelines. Studying the stress-strain characteristics of pipe-soil interaction under ground collapse has an important guiding role for the prevention of ground collapse and the safety protection of pipelines. The current results are mostly concentrated in a single theory or the stress of the pipeline itself, which cannot fully consider the pipe-soil interaction. In this paper, ABAQUS is used to establish the finite element geometric model. Considering the pipeline and the surrounding geological environment conditions, the study is carried out from five aspects: the length of the collapse area, the thickness of the cover layer, the buried depth of the pipeline, the diameter of the pipeline, and the thickness of the pipeline. The displacement of the pipeline soil, the deformation of the pipeline, and the characteristics of the pipeline stress are analyzed, and the variation law is determined through the development trend of the pipeline stress and strain. At the same time, by fitting and analyzing the relationship between the span of different subsidence areas, the thickness of the cover layer, the buried depth of the pipeline, the diameter of the pipeline, the thickness of the pipeline, and the maximum deformation of the pipeline, the reference value of the maximum subsidence displacement of the pipeline under the action of ground collapse is proposed. The work has practical application value for pipeline monitoring, early warning, and disaster management.

Hybrid rice cultivars are widely planted around the world. Here, the authors resequence 1,143 indica accessions, focusing on the parents of superior hybrid rice lines in China, and reveal genetic loci that are associated with heterosis via measuring frequency of parental variation difference (FPVD).

Remobilization of pre-anthesis NSCs (non-structural carbohydrates) is significant for effective grain filling in rice (Oryza sativa L.). However, abundant starch particles as an important component of NSCs are still present in the leaf sheath and stem at the late stage of grain filling. There are no studies on how bioengineering techniques can be used to improve the efficiency of NSC remobilization. In this study, RAmy1A was expressed under the senescence-specific promoter of SAG12, which was designed to degrade starch in the leaf sheath and stem during grain filling. RAmy1A mRNA successfully accumulated in the leaf, stem, and sheath of transgenic plants after anthesis. At the same time, the starch and total soluble sugar content in the leaf, stem, and leaf sheath were obviously decreased during the grain-filling period. The photosynthetic rate of transgenic lines was higher than that of the wild types by an average of 4.0 and 9.9%, at 5 and 10 days after flowering, respectively. In addition, the grain-filling rate of transgenic lines was faster than that of the wild types by an average of 26.09%. These results indicate an enhanced transport efficiency of NSCs from source tissues in transgenic rice. Transgenic rice also displayed accelerated leaf senescence, which was hypothesized to contribute to decreased grain weight.

In cold regions, the long-term stability of engineering facilities is unavoidably influenced by the negative temperature, freeze-thaw process, dry-wet process, and dynamic loading conditions induced by earthquakes and traffic loads. In order to investigate the effects of different cyclic stress paths on the evolution of dynamic mechanical properties of frozen silt clay, a series of cyclic triaxial tests with variation confining pressure (VCP) or constant confining pressure (CCP) were performed. Triaxial low-temperature apparatus (MTS-810) was taken advantage of to simulating various cyclic stress paths by changing cyclic loading conditions of axial stress and confining pressure. In this paper, the evolution features of the axial resilient modulus, damping ratios, and the shape of hysteresis loops with an increase in the number of load cycles under different dynamic stress paths are comprehensively studied. The results show that the loading angle of cyclic stress path and the phase difference between cyclic axial stress and confining pressure are the main factors that remarkably affect the development characteristics of the resilient modulus and damping ratio. With increasing of the loading angle and phase difference, the resilient modulus increases, but damping ratio increases with increasing of loading angle and with decreasing of phase difference. With the continuous increase in the number of loading cycles, the samples of frozen soil show compacting and hardening characteristics. With an increase in the number of load cycles, the shape of hysteresis loop becoming narrows, the resilient modulus decreases at the initial stage and then gradually increases, and the damping ratio stably decreases. According to contrastively analyzing the evolution of dynamic parameters and the shape features of hysteresis loops under various cyclic stress paths, it can be clearly discovered that the evolution of sample microstructure and the development of dynamic characteristics of frozen samples have obvious dependence on the cyclic stress path. Therefore, the effects of variable confining pressure (VCP) on dynamic behaviors of frozen soils are nonnegligible in practical cold region engineering.

The gene tms5, which controls thermo-sensitive genic male sterility (TGMS), has been widely used in two-line hybrid rice breeding in China. The tms5 lines have two sources, namely, AnnongS-1 (AnS) and Zhu1S (ZhS) and, interestingly, are commonly subject to an alteration at cds.71. However, whether cds.71 acts as a mutation hotspot is unknown. Herein, another tms5 mutant named T98S (induced from T98B by irradiation) was used to explore this. First, the gene of tms(t) responsible for T98S was fine-mapped on chromosome 2 based on an F2 group of T98S/R893. In T98S, the candidate gene TMS5 (LOC_Os02g12290.1) mutated at cds.71 with a transversion from cytosine (C) to adenine (A), as also observed in AnS and ZhS. Moreover, the entire coding sequence of TMS5 from T98B converted T98S from sterile to fertile by Agrobacterium tumefaciens-mediated transformation, confirming that T98S is controlled by tms5. Next, detection on nearly 40,000 single nucleotide polymorphisms (SNPs) on Rice 56K SNP Array revealed T98S was 99.99% similar to T98B but only 72.84% and 77.47% similar to AnS and ZhS, respectively, demonstrating that T98S originated from T98B rather than from existing tms5 lines. Furthermore, the cds.70 was found to exist as a T/G haplotype, and it was T rather than G that helped to induce a TGMS trait. The T frequency was 67.52% in indica rice but decreased to 1.75% in japonica rice in 2,644 cultivars tested, which partly explains why tms5 mutants were mostly found in indica lines. Our findings provide evidence that cds.71 may act as a mutation hotspot and clues for breeding TGMS lines in a more efficient way.

“Easy-to-shatter” trait is a major cause of rice crop yield losses, emphasizing the economic value of developing elite rice cultivars with reduced seed shattering capable of achieving higher yields. In the present study, we describe the development of new indica rice lines that exhibit lower rates of seed shattering following the targeted CRISPR/Cas9-mediated editing of the qSH1 gene. We were able to identify qSH1 mutant T0 transgenic plants, with transgene-free homozygous mutants being obtained via segregation in the T1 generation. We then utilized two T2 transgene-free homozygous lines in order to assess the degree of seed shattering and major agronomic traits of these mutant lines and of wild-type rice plants (HR1128-WT). This approach revealed that qsh1 homozygous mutant lines exhibited significantly reduced seed shattering relative to HR1128-WT without any significant changes in other analyzed agronomic traits. We then used these mutant lines to develop new promising hybrid rice lines with intermediate seed shattering. Overall our results reveal that combining targeted gene editing via CRISPR/Cas9 with heterosis utilization approach can allow for the efficient development of novel promising hybrid rice cultivars that exhibit a intermediate of seed shattering, thereby ensuring better stability and improved rice yields.

Measurement of optical properties is critical for understanding light-tissue interaction, properly interpreting measurement data, and gaining better knowledge of tissue physicochemical properties. However, conventional optical measuring techniques are limited in point measurement, which partly hinders the applications on characterizing spatial distribution and inhomogeneity of optical properties of biological tissues. Spatial-frequency domain imaging (SFDI), as an emerging non-contact, depth-varying and wide-field optical imaging technique, is capable of measuring the optical properties in a wide field-of-view on a pixel-by-pixel basis. This review first describes the typical SFDI system and the principle for estimating optical properties using the SFDI technique. Then, the applications of SFDI in the fields of biomedicine, as well as food and agriculture, are reviewed, including burn assessment, skin tissue evaluation, tumor tissue detection, brain tissue monitoring, and quality evaluation of agro-products. Finally, a discussion on the challenges and future perspectives of SFDI for optical property estimation is presented.

Panicle traits are among the most important agronomic characters which directly relate to yield in rice. Grain number (GN), panicle length (PL), primary branch number (PBN), and secondary branch number (SBN) are the major components of rice panicle structure, and are all controlled by quantitative trait loci (QTLs). In our research, four advanced backcross overlapping populations (BIL152, BIL196a, BIL196b, and BIL196b-156) carrying introgressed segments from chromosome 6 were derived from an indica/japonica cross that used the super-hybrid rice restorer line HR1128 and the international sequenced japonica cultivar 'Nipponbare' as the donor and recurrent parents, respectively. The four panicle traits, GN, PL, PBN, and SBN, were evaluated for QTL effects using the inclusive composite interval mapping (ICIM) method in populations over two years at two sites. Results showed that a total of twelve QTLs for GN, PL, PBN, and SBN were detected on chromosome 6. Based on marker loci physical positions, the QTLs were found to be tightly linked to three important chromosomal intervals described as RM7213 to RM19962, RM20000 to RM20210, and RM412 to RM20595. Three QTLs identified in this study, PL6-5, PBN6-1, and PBN6-2, were found to be novel compared with previous studies. A major QTL (PL6-5) for panicle length was detected in all four populations at two locations, and its position was narrowed down to a 1.3Mb region on chromosome 6. Near isogenic lines (NILs) carrying PL6-5 will be developed for fine mapping of the QTL, and our results will provide referable information for gene excavation of panicle components in rice.

The development of alternative casting processes is essential for the high demand of light weight magnesium components to be used in the automotive industry, which often contain different section thicknesses. Squeeze casting with its inherent advantages has been approved for the capability of minimizing the gas porosity in magnesium alloys. For advanced engineering design of light magnesium automotive applications, it is critical to understand the effect of section thickness on mechanical properties of squeeze cast magnesium alloys. In this study, magnesium alloy AM60 with different section thicknesses of 6, 10 and 20 mm squeeze cast under an applied pressure of 30 MPa was investigated. The prepared squeeze cast AM60 specimens were tensile tested at room termperature. The results indicate that the mechanical properties including yield strength (YS), ultimate tensile strength (UTS) and elongation (A) decrease with an increase in section thickness of squeeze cast AM60. The microstructure analysis shows that the improvement in the tensile behavior of squeeze cast AM60 is primarily attributed to the low-gas porosity level and fine grain strucuture which result from the variation of cooling rate of different section thickness. The numerical simulation (Magmasoft? was employed to determine the solidification rates of each step, and the simulated results show that the solidification rate of the alloy decreases with an increase in the section thickness. The computed solidification rates support the experimental observation on grain structural development.

The variation and control of soil organic carbon (SOC) and other nutrients in permafrost regions are critical for studying the carbon cycle and its potential feedbacks to climate change; however, they are poorly understood. Soil nutrients samples at depths of 0–10, 10–20, 20–30, and 30–40 cm, were sampled eight times in 2009 in alpine swamp meadow, alpine meadow and alpine steppe in permafrost regions of the central Qinghai-Tibetan Plateau. SOC and total nitrogen (TN) in the alpine swamp meadow and meadow decreased with soil depth, whereas the highest SOC content in the alpine steppe was found at depths of 20–30 cm. The vertical profiles of total and available phosphorus (P) and potassium (K) were relatively uniform for all the three grassland types. Correlation and linear regression analyses showed that soil moisture (SM) was the most important parameter for the vertical variation of SOC and other soil nutrients, and that belowground biomass (BGB) was the main source of SOC and TN. The spatial variations (including seasonal variation) of SOC and TN at plot scale were large. The relative deviation of SOC ranged from 7.18 to 41.50 in the alpine swamp meadow, from 2.88 to 35.91 in the alpine meadow, and from 9.33 to 68.38 in the alpine steppe. The spatial variations in the other soil nutrients varied among different grassland types. The most important factors for spatial variations (including seasonal variation) of SOC, TN, total P, available P, and both total and available K were: SM, SM and temperature, SM, air temperature, and SM and BGB, respectively. The large variation in the three grassland types implies that spatial variation at plot scale should be considered when estimating SOC storage and its dynamics.
