A new radical relay approach for the efficient construction of alkylsulfonated indolo[2,1-a]isoquinoline-6(5H)-ones in moderate to good yields under mild conditions via SO2 insertion/Smiles rearrangement/intramolecular cyclization is developed. This approach utilizes 4-alkyl substituted Hantzsch esters as alkyl radical sources and further undergoes in situ SO2 insertion to obtain alkylsulfonyl radical species, which avoids the use of unstable alkyl sulfonyl chlorides and highly toxic sulfonyl hydrazides. Additionally, this method for the direct assembly of alkylsulfonated polycyclic molecule skeletons displays high chemical selectivity, broad substrate scope, and step-economy. [ABSTRACT FROM AUTHOR]

A direct visible‐light‐promoted radical‐triggered tandem cyclization of 1,7‐diene systems with cyclic sulfonium salts by using copper(I)‐based complexes as photoredox catalyst is developed. With this approach, a variety of sulfur‐containing polycyclic derivatives are prepared through alkylation/6‐exo‐trig cyclization/5‐exo‐trig cyclization of 1,7‐diene systems under mild conditions. In addition, the photoinduced copper‐catalyzed ring‐opening of cyclic sulfonium salts provides the alkyl radical intermediates, which prefers addition to the C−C double bond that connected to the aromatic ring rather than the electron‐deficient vinyl in 1,7‐dienes. Moreover, oxidation of the desired sulfur‐containing polycyclic products provides a strategy for the preparation of remote nitrogen‐fused polycyclic substituted sulfoxide or sulfone‐containing derivatives. A novel strategy of visible‐light photoredox catalysis ipso‐annulation of activated alkynes with polychloroalkanes for the synthesis of 3‐polychloroalkyl spiro[4,5]trienones, which used 4‐methoxybenzenediazonium tetrafluoroborate salt as a hydrogen atom transfer reagent, is described. This polychloroalkylation/ spirocyclization of alkynes offers a widely range of 3‐polychloroalkyl‐spiro[4,5]trienone derivatives in high yields with a broad substrate scope via selective C(sp3)‐H homolytic cleavage. The results of controlled experiments showed that this method involves a radical process. A simple and efficient oxidative‐promoted radical cascade annulation of ynone derivatives with ketones for the construction of a series of oxoalkyl‐substituted indenones in moderate to good yields is described. The control experiment results indicate that the oxidative acetmethylation/arylation of ynone derivatives involves a radical cascade cyclization process. 2-Aminobenzothiazoles are commonly encountered in various functional compounds. Herein, we disclose an electro-oxidative three-component reaction for the effective synthesis of 2-aminobenzothiazoles under mild conditions, utilizing non-toxic and abundant elemental sulfur as the sulfur source. Both aliphatic amines and aryl amines demonstrate good compatibility at room temperature, highlighting the broad functional group tolerance of this approach. Additionally, elemental selenium demonstrated reactivities comparable to those of elemental sulfur.

Cyclobutanone oximes and their derivatives are pivotal core structural motifs in organic chemistry. Iminyl-radical-triggered C-C bond cleavage of cyclobutanone oximes delivers an efficient strategy to produce stable distal cyano-substituted alkyl radicals, which can capture SO 2 , CO or O 2 to form cyanoalkylsulfonyl radicals, cyanoalkylcarbonyl radicals or cyanoalkoxyl radicals under mild conditions. In the past several years, cyanoalkylsulfonylation/cyanoalkylcarbonyaltion/cyanoalkoxylation has attracted a lot of interest. In this updated report, the strategies for trapping SO 2 , CO or O 2 via iminyl-radical-triggered ring-opening of cyclobutanone oximes are summarized.

A new protocol is herein described for the direct generation of alkylated indolo/benzoimidazo[2,1- a ]isoquinolin-6(5 H )-one derivatives by using Hantzsch esters as alkylation radical precursors using a photoredox/K 2 S 2 O 8 system. This oxidative alkylation of active alkenes involves a radical cascade cyclization process and a sequence of Hantzsch ester single electron oxidation, C-C bond cleavage, alkylation, arylation and oxidative deprotonation.

A copper-catalyzed remote benzylic C-H functionalization strategy enabling 1,2-difunctionalization of alkenes with 2-methylbenzeneamides and nucleophiles, including alcohols, indoles, pyrroles, and the intrinsic amino groups, is reported, which is characterized by its redox-neutral conditions, exquisite site-selectivity, broad substrate scope, and wide utilizations of late-stage modifying bioactive molecules. This reaction proceeds through nitrogen-centered radical generation, hydrogen atom transfer, benzylic radical addition across the alkenes, single-electron oxidation, and carbocation electrophilic course cascades. While using external nucleophiles manipulates three-component alkene alkylalkoxylation and alkyl-heteroarylation with 2-methylbenzeneamides to access dialkyl ethers, 3-alkylindoles, and 3-alkylpyrroles, omitting the external nucleophiles results in two-component alkylamidation ([5+2] annulation) of alkenes with 2-methylbenzeneamides to benzo-[ f ][1,2]thiazepine 1,1-dioxides.

Aim: To develop a pharmacokinetic model for the enterohepatic circulation of mycophenolic acid (MPA)., Methods: Twenty healthy volunteers were orally given a single dose of 500 mg mycophenolate mofetil. Plasma samples were collected during 48 hours and MPA concentration was measured by HPLC method. Pharmacokinetic (PK) model was established based on physiological and biopharmaceutical consideration and PK parameters were obtained using nonlinear mixed effect model., Results: The proposed model included an intestinal compartment and gall bladder compartment in addition to the central compartment. The predicted time-concentration curve and AUC0-t, Cmax, Tmax estimated by the established model were in agreement with the observations., Conclusion: The established model was well defined for the MPA disposition and could afford a useful approach for the further clinical investigation.
