25. ​Studies on Cefquinome Sulfate Intramammary Infusion Composited with Varied Excipients for the Prevention of Dairy Cows Mastitis in the Dry Cow Period

Abstract

Background: The prevention and treatment of dairy cow mastitis during the dry period are crucial for the dairy industry. In this work, 200 dairy cows without mastitis and normal entering dry period were selected and randomly divided into 4 groups, for the first time, to explore the prevention effects using the different formulations of Cefquinome Sulfate Breast Injection with varied excipients. Methods: Three formulations with various excipients were further evaluated based on the drug residue analysis, somatic cell reduction in the milk and clinical symptoms of mastitis. In the control group, cloxacillin benzathine was utilized for comparisons. Result: All three Cefquinome test groups showed a better effect on prevention than the benzathine cloxacillin group in terms of drug release time, post-partum drug residue, reducing somatic cell count and preventing mastitis in the dry period. Various excipients showed slightly different impacts on the drug residues, the changes in somatic cell counts of dairy cows before and after medication. The prevention effects on mastitis from different excipients were similar after 30 d.

It would be desirable to develop safe and efficient adjuvants with the aim of improving the cellular immune response against the target antigen. In this study, the targeting and sustained release of simple nanoliposomes containing Lycium barbarum polysaccharides (LBP) as an efficacious immune adjuvant to improve immune responses were explored. LBP liposome (LBPL) with high entrapment efficiency (86%) were obtained using a reverse-phase evaporation method and then used to encapsulate the model antigen, ovalbumin (OVA). We demonstrated that the as-synthesized liposome loaded with OVA and LBP (LBPL-OVA) was stable for 45days and determined the encapsulation stability of OVA at 4°C and 37°C and the release profile of OVA from LBPL-OVA was investigated in pH 7.4 and pH 5.0. Further in vivo investigation showed that the antigen-specific humoral response was correlated with antigen delivery to the draining lymph nodes. The LBPL-OVA were also associated with high levels of uptake by key dendritic cells in the draining lymph nodes and they efficiently stimulated CD4+ and CD8+ T cell proliferation in vivo, further promoting antibody production. These features together elicited a significant humoral and celluar immune response, which was superior to that produced by free antigen alone. Abstract

Shanghai is the largest industrial and commercial city in China. The urban turbidity island effect is remarkable. The comparison of air quality between urban and rural areas shows that the concentration of urban air pollutants, such as SO2, NOx, total suspended particulates and falling dust, is much higher than that of rural air pollutants. Owing to the more hygroscopic nuclei in the urban atmosphere and to the thermal and mechanical turbulence in an urban area being stronger than in a rural area, the low cloud amount in an urban area is greater than that in rural areas. Taking the ratio of solar direct radiation (S) to diffuse radiation (D) as the measure of turbidity factor (D/S), it can be seen that the turbidity factor in an urban area is obviously greater than that of a suburban area. With the urban district as a center, a distinct turbidity island is formed. This kind of turbidity island effect is gradually increasing with the development of Shanghai city.

Abstract

The deposition of TiO2-P25 on Pyrex glass is studied in order to propose a procedure for obtaining uniform and adherent thin films with a controlled thickness in the range between fractions of micrometer to a few tens of micrometers. Different deposition methods such as spray-coating, dip-coating, and drop-coating of liquid suspensions of TiO2-P25 powder are considered. The effects of pretreatment of the glass support, as well as postdeposition steps such as drying condition and thermal treatment, are also reported. A basic glass pretreatment and an acidic TiO2suspension are chosen for an optimized adherence of the film. Drop-coating, associated with a slow drying under 55% relative humidity and a high-temperature thermal treatment is the method of choice to prepare coated glass slides in a large range of film thicknesses.
